The examination of macrophyte, water and sediment samples, collected at depths less than 1.5 m from 50 different sites along the North Aegean coasts, has revealed, for the first time in Greek coastal waters, the presence of two Ostreopsis species (O. ovata and O. cf. siamensis) and Coolia monotis in the majority of the sampling sites (94% and 100%, respectively). Other epiphytic dinoflagellates of the genera Prorocentrum and Amphidinium and diatoms were accompanying species in this epiphytic community. Morphometric features, plate formula and thecal ornamentation were used for species identification. O. ovata cells were smaller in dorsoventral (DV) diameter and width (W) (26.18–61.88 μm and 13.09–47.60 μm, respectively) in comparison with O. cf. siamensis (35.70–65.45 μm and 23.80–49.98 μm, respectively). In contrast, the anterioposterior (AP) diameter of O. cf. siamensis was smaller (14.28–26.18 μm) resulting in DV/AP ≈ 3, whereas the above ratio for O. ovata was less than 2 (AP ranging between 14.28–35.70 μm). Moreover, the theca of O. ovata cells was ornamented with scattered pores, which fluctuated in a wider range (0.07–0.32 μm) than those of O. cf. siamensis (0.23–0.29 μm). Coolia monotis cells were almost round with average DV diameter 26.88 μm, AP 25.66 μm and width 26.76 μm. Small and large cells were recorded in both field and culture populations of Ostreopsis spp. and C. monotis, while hyaline cysts were observed for O. ovata. The presence of O. ovata and O. cf. siamensis exhibited a clear seasonal pattern dominating (maximum abundance up to 4.05 × 105 cells gr−1 fwm) the period from midsummer to late autumn in years 2003 and 2004, while C. monotis was found also in winter and spring months.  相似文献

Critical thermal maximum (CTmax) and body water losses were measured in first instar larvae of Gnorimus nobilis, Osmoderma eremita (Trichiinae) and Cetonischema aeruginosa (Cetoniinae) when air temperature was increased gradually (0.5 °C/min) from 20 °C to the critical thermal maximum (CTmax), in dry air (near 0% R.H.).

The CTmax was significantly lower in O. eremita (45.6±0.7 °C) than in G. nobilis (48.5±0.6) and C. aeruginosa (51.4±0.9 °C).

An increase of 10 °C (30–40 °C) induced a 2-fold increase of the water loss in C. aeruginosa and O. eremita (Q10=2.10±0.12 and 2.13±0.20, respectively). In the range from 40 to 45 °C to CTmax a strong increase of the water loss was observed in O. eremita and C. aeruginosa, respectively. Body water losses were significantly lower in C. aeruginosa than in O. eremita and G. nobilis over the range 20 °C—CTmax; no significant difference occurred between G. nobilis and O. eremita.  相似文献

The effect of temperature regimes on conidia germination, radial growth and virulence of Beauveriabassiana and Metarhizium anisopliae against the spotted stalk borer Chilo partellus was investigated with Ethiopian isolates. Conidia germination, radial growth and sporulation of all isolates were retarded at 15 and 35°C. A suitable temperature range for the isolates was between 20 to 30°C. Conidia germination was less tolerant of low temperature (15°C) than radial growth. Radial growth and sporulation reacted differently to temperature. At both 25 and 30°C, all isolates induced 100% mortality to C. partellus larvae in six days. The LT50 decreased for the isolates with increasing temperature.  相似文献

The enthalpy of unfolding (ΔuH) of carbonic anhydrase II was determined by titrating the protein with acid and measuring the heat using isothermal titration calorimetry (ITC) in the temperature range of 5 to 59 °C. By combining the ITC results with our previous findings by differential scanning calorimetry (DSC) in the temperature range of 39 to 72 °C, the ΔuH dependence over a wide temperature range was obtained. The temperature dependence of the enthalpy displays significant curvature indicating that the heat capacity of unfolding (ΔuCp) is dependent on temperature. The T-derivative of ΔuCp was equal to 100 ± 30 J/(mol × K2), with the result that the ΔuCp is equal to 15.8 kJ/(mol × K) at 5 °C, 19.0 kJ/(mol × K) at 37 °C and 21.8 kJ/(mol × K) at 64 °C. The enthalpy of unfolding is zero at 17 °C. At lower temperatures, the ΔuH becomes exothermic.

This method of determining protein unfolding thermodynamics using acid-ITC, significantly widens the accessible T-range, provides direct estimate of the thermodynamic parameters at physiological temperature, and gives further insight into the third T-derivative of the Gibbs free energy of unfolding.  相似文献

An extracellular protease from the marine bacterium Sphingomonas paucimobilis, strain 116, isolated from the stomach of Antarctic krill, Euphausia superba Dana, was purified and characterized. The excretion of protease was maximal at temperatures from 5 to 10°C, i.e. below the temperature optimum for the strain growth (15°C). The highly purified enzyme was a metalloprotease [sensivity to ethylenediaminetetraacetic acid (EDTA)] and showed maximal activity against proteins at 20–30°C and pH 6.5–7.0, and towards N-benzoyl-tyrosine ethyl ester (BzTyrOEt) at pH 8.0. At 0°C the enzyme retained as much as 47% of maximal activity in hydrolysis of urea denatured haemoglobin (Hb) (at pH 7.0), and at −5 and −10°C, 37 and 30%, respectively. The metalloprotease was stable up to 30°C for 15 min and up to 20°C for 60 min. These results indicate that the proteinase from S. paucimobilis 116 is a cold-adapted enzyme.  相似文献

Resistance in Cicer bijugum Rech. f. a wild relative of chickpea, to Botrytis grey mould (BGM), caused by Botrytis cinerea Pers., was shown to be associated with high concentrations of maackiain when compared to three susceptible species. The two BGM resistant accessions of C. bijugum contained between 200 and 300 μg maackiain g−1 of foliage whereas the BGM susceptible species C. arietinum, C. echinospermum and C. reticulatum contained less than 70 μg g−1. Furthermore, the concentration of maackiain increased to more than 400 μg g−1 in the resistant wild species after being inoculated with the pathogen whereas no significant increase was recorded in the susceptible species. The germination of spores of Botrytis cinerea, treated with maackiain, was inhibited in a dose dependent manner; less than 10% of spores germinated when treated with 500 μg ml−1. The data indicate that maackiain may be an important component in BGM resistance in the wild chickpea C. bijugum and that the resistance is enhanced in the presence of the pathogen.  相似文献

Patterns of seed germination of the perennial spurge Euphorbia nicaeensis were studied in three populations in south-western Spain. We investigated the variation in seed viability and germination among individuals, and among populations over 2 consecutive years. We also studied if diverse factors such as temperature, acid scarification, darkness or caruncle loss affect the germination of the seeds.

Interindividual and interpopulation variability in seed viability was found. E. nicaeensis seeds are nondormant, so differences in interpopulation viability translate into differences in final germination rates. The germination percentage of the seeds from each population was similar in the 2 years studied. The effects of diverse factors were homogeneous in the two populations studied. Darkness has no effect on seed germination, and ecarunculate seeds germinated in the same proportions as carunculate seeds; this could allow the seeds to germinate in the chambers of deserted anthills. Acid scarification significantly reduced the germination percentage in only one of the populations, but over 50% of the seeds germinated, which could allow herbivores to act as occasional dispersing agents. The seeds that were preheated at 100 °C for 1 and 5 min germinated in the same proportions as the control group. The seeds that were preheated at 120 °C for 5 min displayed a significant decrease in germination, but the percentage was over 40% for both populations, indicating that the seeds could still germinate after the passage of a fire.  相似文献

It has been documented in the many-lined sun skink (Mabuya multifasciata) that pregnant females select lower body temperatures (27.6–30.8 °C) than do nonpregnant females and adult males (29.7–35.7 °C). We therefore used the skink to test the hypothesis that the maximization of reproductive benefits should be achieved in pregnant females by shifting thermal preferences towards the levels optimal for embryonic development but entailing relatively small costs of reproduction. Data on adult males showed that temperatures maximizing swimming stamina (indicative of locomotor endurance) fell within the range of body temperatures selected by nonpregnant females and adult males. Data on swimming stamina and feeding performance of pregnant females, nonpregnant females and adult males measured at 26 and 30 °C showed that: (1) pregnancy impaired locomotor and feeding performances, but such impairments did not persist after parturition; (2) the degree of locomotor impairment during pregnancy was greater at 26 °C than at 30 °C, but the degree of feeding impairment during pregnancy was greater at 30 °C than at 26 °C. Pregnant females of M. multifasciata selecting body temperatures at about 29 °C could not only produce good-quality offspring in a relatively short gestation length but also reduce reproductive costs associated with locomotor and feeding impairments to some extent. Solid-state culture of the white-rot fungus Phanerochaete chrysosporium BKMF-1767 (ATCC 24725) has been carried out, using an inert support, polystyrene foam. Suitable medium and culture conditions have been chosen to favor the secretion of manganese peroxidase (MnP). The enzyme was isolated and purified from immobilized P. chrysosporium and partially characterized. Partial protein precipitation in crude enzyme was affected using ammonium sulphate, polyethylene glycol, methanol, and ethanol methods. Fractionation of MnP was performed by DEAE-Sepharose ion exchange chromatography followed by Ultragel AcA 54 gel filtration chromatography. This purification attained 23.08% activity yield with a purification factor of 5.8. According to data on gel filtration chromatography and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), the molecular weight of the enzyme was 45 000±1000 Da. The optimum pH and temperature of purified MnP were 4.5 and 30 °C, respectively. This enzyme was stable in the pH range 4.5–6.0, at 25 °C and also up to 35 °C at pH 4.5 for 1 h incubation period. MnP activity was inhibited by 2 mM NaN3, ascorbic acid, β-mercaptoethanol and dithreitol. The Km values of MnP for hydrogen peroxide and 2.6-dimetoxyphenol were 71.4 and 28.57 μM at pH 4.5, respectively. The effects of possible inhibitors and activators of enzyme activity were investigated.  相似文献

The effects of temperature, salinity, inorganic and organic nutrients on the growth and physiological performance of Fibrocapsa japonica have been investigated in strains isolated from the northern Adriatic Sea, where intense and regular blooms have occurred since 1997 in coastal waters during summer. Strains isolated at different times and from different locations appeared homogeneous in terms of both physiological responses and molecular (ITS-5.8S rDNA) characteristics. Growth rates were higher at temperatures between 20 and 26 °C and in a salinity range of 30–35 (0.7 div day−1). The temperature of 16 °C inhibited growth, more markedly at the lowest and highest salinity values, a result also confirmed by a lower photosynthetic efficiency and by an increase in cell volume due to impaired division. Higher cell concentrations were obtained with macronutrients at f/2 levels than in a fivefold diluted medium. Comparing the utilization of 200 μM nitrate to that of different N sources (inorganic and organic), F. japonica showed an efficient growth with equivalent amounts of ammonia, urea and amino acids, such as glycine and tryptophan; glutamate was less effective, while methionine had toxic effects. Organic phosphate, administered as glycerophosphate, could also sustain F. japonica growth, probably on account of an alkaline phosphatase whose activity was enhanced in the presence of the organic form. Vitamins were necessary for growth, though no further stimulation was observed when a surplus of vitamin B12 (3 nM) was added. The addition of 11.7 μM iron instead of 2.3 μM, as well as that of humic acid, with or without macronutrients, did not enhance algal growth either. These results led us to hypothesise that F. japonica blooms became more frequent due to a general seawater temperature increase and to the availability of organic forms which, in coastal anthropized areas, are especially abundant in summer periods.  相似文献

The seeds of Cassia occidentalis, an annual weed occurring throughout India, is a rich source of galactomannan gum. The gum derived from seed endosperm can be potentially utilized in a number of industries to replace the conventional gums. With a view to utilize the gum for broader applications, carbamoylethylation of C. occidentalis seed gum was carried out with acrylamide in presence of sodium hydroxide under different reaction conditions. Variables studied were concentration of sodium hydroxide, acrylamide, gum–solvent ratio, reaction time and temperature. The nitrogen content, carboxyl content and total ether content were determined. The optimum condition for preparing carbamoylethyl C. occidentalis seed gum (%N=2.57) comprised concentration of acrylamide (0.070 mol), sodium hydroxide (0.125 mol), C. occidentalis seed gum (0.03 mol) at 30 °C for 3 h. Rheological properties of carbamoylethyl C. occidentalis seed gum solution showed non-Newtonian pseudo-plastic behavior, relatively high viscosity, cold water solubility and solution clarity vis-à-vis unmodified C. occidentalis seed gum.  相似文献

Lipases from Candida rugosa, Mucor javanicus and Rhizopus oryzae were respectively adsorbed on Amberlite XAD-7 followed by incubation in 2-propanol and then lyophilization. The activities of the immobilized enzymes were 1.6–3.4 times higher than those of the immobilized enzymes without incubation in the organic solvent before lyophilization for esterification of lauric acid (0.1 M) and 1-propanol (0.1 M) in isooctane at 37 °C. The immobilized C. rugosa lipase (Sigma) without the incubation did not show any activity but displayed considerable activity (19.8 μmol h−1 mg−1) after the incubation before lyophilization. Besides 2-propanol, acetone, 1-propanol and ethyl acetate were also found to be good solvents for treating M. javanicus lipase immobilized on Amberlite XAD-7 and acetone was the best among them. When incubated in isooctane at 25 °C for 120 h, the immobilized M. javanicus lipase prepared by incubation in acetone for 1 h before lyophilization retained 70% of its initial activity while the immobilized enzyme without the solvent treatment kept only 50% of its initial activity.  相似文献

Optimised immunomagnetic separation methods to detect Cryptosporidium parvum and Escherichia coli O157 in UK shellfish are described. Whole tissue homogenates gave the best recoveries for C. parvum oocysts compared with gill or haemolymph extracts. The sensitivity of recovery from spiked samples was comparable to that achieved when processing water and varied from 12–34% in mussels, 48–69.5% in oysters and 30–65% in scallops. Maximum recovery of E. coli O157 was achieved by enriching in buffered peptone water supplemented with vancomycin at 42 °C. Increasing enrichment temperatures from 37 to 42 °C gave a significant increase in target number recovery. Implementation of these methods into monitoring programmes and end-product testing will enable shellfish producers to better assess product safety.  相似文献

Differential scanning calorimetry, circular dichroism, and visible absorption spectrophotometry were employed to elucidate the structural stability of thermophilic phycocyanin derived from Cyanidium caldarium, a eucaryotic organism which contains a nucleus, grown in acidic conditions (pH 3.4) at 54°C. The obtained results were compared with those previously reported for thermophilic phycocyanin derived from Synechococcus lividus, a procaryote containing no organized nucleus, grown in alkaline conditions (pH 8.5) at 52°C. The temperature of thermal unfolding (td) was found to be comparable between C. caldarium (73°C) and S. lividus (74°C) phycocyanins. The apparent free energy of unfolding (ΔG[urea]=0) at zero denaturant (urea) concentration was also comparable: 9.1 and 8.7 kcal/mole for unfolding the chromophore part of the protein, and 5.0 and 4.3 kcal/mole for unfolding the apoprotein part of the protein, respectively. These values of td and ΔG[urea]=0 were significantly higher than those previously reported for mesophilic Phormidium luridum phycocyanin (grown at 25°C). These findings revealed that relatively higher values of td and ΔG[urea]=0 were characteristics of thermophilic proteins. In contrast, the enthalpies of completed unfolding (ΔHd) and the half-completed unfolding (ΔHd)1/2 for C. caldarium phycocyanin were much lower than those for S. lividus protein (89 versus 180 kcal/mole and 62 versus 115 kcal/mole, respectively). Factors contributing to a lower ΔHd in C. caldarium protein and the role of charged groups in enhancing the stability of thermophilic proteins were discusse.  相似文献

Bacillus cereus sphingomyelinase activity was assayed on large unilamellar vesicles composed of sphingomyelin (SM)/cholesterol (Ch) mixtures at varying proportions. Natural (egg) SM was used with a gel–fluid transition temperature at ca. 40 °C. When the enzyme was assayed at 37 °C, the activity on pure SM was exceedingly low, but a small increase was observed as soon as some Ch was added, and a large enhancement of activity occurred with Ch proportions above 25 mol%. The data were interpreted in terms of sphingomyelinase activity being higher in the cholesterol-induced liquid-ordered phase than in the gel phase. The abrupt increase in activity above 25 mol% Ch would occur as a result of a change in domain connectivity, when the Ch-rich liquid-ordered domains coalesced. In equimolar SM/Ch mixtures, that were in the liquid-ordered state in a wide range of temperatures, sphingomyelinase activity was virtually constant in the 30–70 °C range. The results demonstrate that at the mammalian and bird physiological temperatures Ch modulates sphingomyelinase activity, and that this can occur precisely because most SM have a gel–fluid transition temperature above the physiological temperature range. In addition, Ch activation of sphingomyelinase and the strong affinity of Ch for SM allow the rapid, localised and self-contained production of the metabolic signal ceramide in specific microdomains (rafts).  相似文献

Controlled laboratory culture of Alexandrium catenella was used to determine the effects of a range of temperatures between 10 and 16 °C on the growth and saxitoxin content of this dinoflagellate, using strain ACC02 isolated from seawater at Aysen, XI Region, Southern Chile. Cell cultures were made using L1 culture medium at 30‰ salinity, and a photon flux density of 59.53 μmol m2 s−1. The results showed that the duration of the exponential growth phase was determined by the experimental temperature, with maximum cell concentrations obtained at 12 °C; significantly lower cell concentrations and growth rates were obtained at 16 °C. Cell dry weight and chlorophyll a values followed cell growth trends. The toxicity of A. catenella was variable at all the experimental temperatures, with a tendency towards having an inverse relation to temperature, with the highest values occurring at 10 °C and the lowest at 16 °C. The optimal range of temperature for the growth of the Chilean strain of A. catenella differed from rates reported for this species isolated at other latitudes, and was correlated with natural temperature conditions predominant in the environment from which it was isolated. The inverse relation of toxicity with temperature in the laboratory was broadly reflected in observations on the toxicity of this dinoflagellate in the field, and coincided with results from the literature.  相似文献

The rate of development of Lymnaea auricularia eggs was studied at various constant temperatures between 10° and 36°C. Development was accelerated as the temperature increased and at 36°C the eggs failed to develop. Spring eggs showed differences in their rate of development when compared with summer eggs when measured at similar tempertures.

Both spring and summer eggs were more than 90% fertile. Hatching success was high at temperatures between 10° and 30° (100%–82/9%); while at 34°C it was reduced to 60.6% for spring eggs. It was above 87% at temperatures between 10° and 34°C but it dropped to 62.3% at 36°C for summer eggs.

In one regularly changing temperature experiment a significant acceleration (P < .05) was found. In two others there was no significant difference beween predicted and observed egg durations. In one suddenly changing temperature regime (1 day at 20°, 1 day at 30° and so on) a huge retardation of development was found. In the other suddenly changing experiment (1 day at 15°, 1 day at 25°) no significant difference was found.

The exposure of eggs to extreme temperature (4°C, freezing and 4°C caused a retardation in the race of subsequent development of eggs at 25°C.  相似文献

Thermostable β-glucosidase from Sulfolobus shibatae was immobilized on silica gel modified or not modified with 3-aminopropyl-triethoxysilane using transglutaminase as a cross-linking factor. Obtained preparations had specific activity of 3883 U/g of the support, when measured at 70 °C using o-nitrophenyl β-d-galactopyranoside (GalβoNp) as substrate. The highest immobilization yield of the enzyme was achieved at pH 5.0 in reaction media. The most active preparations of immobilized β-glucosidase were obtained at a transglutaminase concentration of 40 mg/ml at 50 °C. The immobilization was almost completely terminated after 100 min of the reaction and prolonged time of this process did not cause considerable changes of the activity of the preparations. The immobilization did not influence considerably on optimum pH and temperature of GalβoNp hydrolysis catalyzed by the investigated enzyme (98 °C, pH 5.5). The broad substrate specifity and properties of the thermostable β-glucosidase from S. shibatae immobilized on silica-gel indicate its suitability for hydrolysis of lactose during whey processing.  相似文献

In controlled environment experiments, the bioherbicidal fungus Myrothecium verrucaria (Alb. & Schwein.) Ditmar:Fr. was tested alone, in combination with, prior to, and following treatment with glyphosate [N-(phosphonomethyl)glycine] for control of kudzu [Pueraria lobata (Willd.) Ohwi], redvine [Brunnichia ovata (Walt.) Shinners], and trumpetcreeper [Campsis radicans (L.) Seem. ex Bureau] at temperatures of 20, 30, and 40°C. At all temperatures, kudzu was most adversely affected by the fungus, followed by trumpetcreeper and redvine, as indicated by greater mortality and dry weight reductions. Trumpetcreeper and redvine mortalities and dry weight reductions significantly increased when the fungus was applied 2 days after the glyphosate treatment. Application of the fungus combined with or prior to glyphosate treatment resulted in reduced weed control. Although pathogenesis and mortality also occurred at 20°C, disease development was favored by higher temperatures (30 and 40°C). Infected weeds of each species exhibited similar disease symptomatology within 12 h following treatment at incubation temperatures of 30 and 40°C. Disease symptomatology was characterized by necrotic flecking on leaves that coalesced into large lesions. Symptoms progressed, initially infecting cotyledons and leaves, and later (within 48 h) producing stem lesions. The fungus sporulated profusely on infected tissue and was easily reisolated. These results suggest that timing of glyphosate application in relation to combined treatment with the bioherbicide M. verrucaria can improve the control of kudzu, redvine, and trumpetcreeper.  相似文献
