The contributions made by photosynthesis in the first leaf toseedling growth have been examined by a variety of methods includinginfra-red gas analysis and the use of 14CO2. The first leafis fully expanded by day 8 and maximal rates of photosynthesisare achieved about I day later. Up to day 8 growth of the seedlingsresults from the redistribution of seed reserves and once theseare exhausted growth is dependent upon the first leaf, beingreduced to very low levels if this is shaded. The second leafwhich begins to expand rapidly after day 10 is contributingto growth by day 14, and the contribution from the first leafbegins to decline after day 12. Apart from greatly reducing photosynthesis in treated leaves,shade also affects the development of photosynthetic capacity.When applied for 2 days or more from day 6, shade reduces thepeak level of carbon fixation achieved on days 9 and 10 by upto 35 per cent. It is shown that development of the first leafin terms of increased dry weight and photosynthetic capacity,is dependent on photosynthesis in the developing leaf itself.The mechanisms by which shading affects development are discussed.  相似文献

The rate of carbon transport from an old tomato leaf (54 days),grown at 80 W m–2, was measured under light flux densitiesbetween 7 and 90 W m–2. Under low light, the rate of carbontransport over a 6 h period was about 1 mg C dm–2 h–1,well in excess of the concurrent photosynthetic rate. The lossfrom these leaves of 14C-leaf assimilate which was fixed beforethe experimental period amounted to 62 per cent of the totalinitial uptake and was higher than that from leaves with higherconcurrent photosynthetic rates. The higher loss of 14C fromleaves with low photosynthetic rates was due to a greater contributionof 14C from the starch and residue fractions. The rate of transportappeared to be determined by the concentration of the labilesucrose, not the total sucrose concentration. In comparisonwith young fully-expanded tomato leaves (Ho, 1976) the sizeof the labile sucrose pool appeared to decrease with age. Thephotosynthesistranslocation coefficient was low (k1k2=0•21)for an old tomato leaf. Based on these results a scheme of carbonpartitioning in relation to translocation is proposed. Criteriafor assessing the efficiency of translocation in leaves arediscussed.  相似文献

The growth of white clover (Trifolium repens L.) in conditionstypical of April in Southern England (8 °C day/4 °Cnight, 12 h photoperiod of 90 J m–2 s–1 visibleradiation) was extremely slow, whether the plants were dependentfor nitrogen on fixation by their root nodules or were suppliedwith abundant nitrate; although growth was slower in the nodulatedplants. The reasons for slow growth were a large root: shootratio and a small leaf area, particularly in the nodulated plants,and a low photosynthetic rate in all plants. The probable effectsof these characteristics on the growth of white clover withgrasses in mixed pastures are discussed. The contribution of carbon from various sources to leaves 2,3 and 4 of Proctor barley as they expanded, was determined usingdata from photosynthetic measurements and 14CO2 feeding. Bythe time that it was half expanded, the growing leaf providedabout half its carbon requirement from current photosynthesis,and this proportion increased further during the later phasesof expansion. In all, about half of the carbon in a fully expandedleaf was derived from local photosynthesis. The pattern of supplyfrom older leaves changed with time, depending on their ageand photosynthetic activity. Shading the third or expandingfourth leaf led to enhanced photosynthetic activity in the unshadedleaf, but this was not sufficient to compensate fully for thereduction due to shade treatment, and carbon supply for thegrowing leaf was reduced. To examine 14CO2 fixation, potential translocation, and carbonflow among leaf chemical fractions of young developing leaves,the shoot tip of 24-leaf cottonwood (Populus deltoides Bartr.ex. Marsh) plants were cut off under water, placed in artificialxylem sap, and treated with 14CO2 in continuous and pulse-chaseexperiments. Additional leaves on whole plants were spot treatedon the lamina tip to follow export from the tip only. The analysedleaves ranged in age from leaf plastochron index(LPI) –5to 3, the spot treated leaves from LPI 2 to 5. After 30 minfixation, the specific activity in the lamina tip increasedlinearly with leaf age from LPI –5 to 1 (0.5 to 4.5 kBqmg–1). Specific activity in the lower lamina increasedslowly with leaf age and did not reach 500 kBq mg–1 untilLPI –1. Total 14CO2 fixed in the lower lamina exceededthat fixed in the tip by LPI –2 because of the large amountof tissue present in the lower lamina. Although the lamina tipfixed high levels of 14CO2, pulse-chase studies coupled withautoradiography indicated no vein loading or translocation fromthe tip until about LPI 4 or 5. The 14C fixed in both tip andlower lamina was incorporated at the site of fixation and wasnot distributed to younger tissue or translocated from the lamina.Although the percentage distribution (14C in each chemical fractioncompared with the total in all fractions) of 14C among certainchemical fractions, e.g. sugars, amino acids and proteins, indicatedthat the mesophyll of the tip was more mature than the lowerlamina, physiologically both leaf sectors were immature basedon the expected 14C distribution in mature tissue. Informationfrom this and other studies indicates that the extreme tip ofa developing cottonwood leaf first begins to export photosynthateabout LPI 4 or 5 on a 24-leaf plant. The first photosynthatetranslocated may be incorporated into the vascular tissues andmesophyll directly below the tip. During vegetative growth in controlled environments, the patternof distribution of 14C-labelled assimilates to shoot and root,and to the meristems of the shoot, was measured in red and whiteclover plants either wholly dependent on N2 fixation in rootnodules or receiving abundant nitrate nitrogen but lacking nodules. In experiments where single leaves on the primary shoot wereexposed to 14CO2, nodulated plants of both clovers generallyexported more of their labelled assimilates to root (+nodules),than equivalent plants utilizing nitrate nitrogen, and thiswas offset by reduced export to branches (red clover) or stolons(white clover). The intensity of these effects varied with experiment.The export of labelled assimilate to growing leaves at the terminalmeristem of the donor shoot was not influenced by source ofnitrogen. Internode elongation in the donor shoot utilized nolabelled assimilate. Whole plants of white clover exposed to 14CO2 on seven occasionsover 32 days exhibited the same effect on export to root (+nodules),which increased slightly in intensity with increasing plantage. Nitrate Accumulation and its Relation to Leaf Elongation in Spinach Leaves   总被引:6,自引:0,他引:6  
The leaf elongation rate (LER) of spinach leaves during theday was twice that during the night when grown at a photon fluxdensity of 145 µmol m–2 s–1. All leaves showedthe same LER-pattern over 24 h. Due to low turgor, LER was lowin the afternoon and in the first hours of the night until wateruptake restored full turgor. Osmotic potential remained constantdue to increased nitrate uptake and starch degradation in thisperiod. LER increased to high rates in the second part of thenight and in the morning. The lower rate in the dark comparedto the light was not caused by the lower night temperatures,as increased photon flux density during growth resulted in equalrates in the light and the dark. Increased relative humiditydecreased LER and afternoon rates were most sensitive to waterstress. A ‘low light’ night period did not changeLER-pattern during the night or on the following day. We concludethat nitrate is not an obligatory osmoticum during the nightand can be exchanged for organic osmotica without decreasingLER. During the night the turgor is first restored by increasingwater uptake, nitrate uptake and starch degradation. This resultedin increased leaf fresh weight in this period. Thereafter, elongationincreased by simultaneous uptake of nitrate and water. Nitrateconcentration was, therefore, constant in the older leaves.In the younger leaves nitrate concentration increased to replacesoluble carbohydrates. The single rooted leaf of soybean (Glycine max L. Merr.) wasused to study source-sink relationships in photosynthesis. Whenthe leaves were kept under a regime of 10 h light (410–480µmol photons m–2, 400–700 nm)–14 h dark,they did not expand, the increase in leaf dry weight almoststopped, and photosynthetic activity remained at a high andconstant level for 8 d while the dry weight of the roots increasedat a constant rate throughout the period. Thus, under this conditionthe leaf and the root system served as the only source and sinkorgans, respectively. When leaves grown for 7 d under this conditionwere placed under continuous light to alter the source/sinkbalance in photosynthate, the root dry weight increased at aconstant rate equal to that found under the 10 h light–14h dark condition. The leaf dry weight markedly increased andby day 5 of continuous light had increased 1.6-fold, mainlyas a consequence of accumulation of starch and sucrose, whichwere not translocated for root growth. The continuous lightcaused an abrupt decrease in the photosynthetic activity (40%of initial value by day 5). However, the activity recoveredalmost completely after a 32-h transfer to darkness. Significantnegative correlations existed between photosynthetic activityand the sucrose and starch contents in the rooted leaves placedunder continuous light. When the plants were treated with variouslight conditions, there was no significant difference (p<0.01)among the regression line slopes for photosynthetic activityon the sucrose content, but there was some deviation among thosefor the photosynthetic activity on the starch content. Thisresult suggests that sucrose accumulated in the leaf has a moredirect influence on photosynthetic activity when the source/sinkbalance was altered. Activities of photosynthetic and photorespiratory enzymes viz.,ribulose bisphosphate carboxylase, phosphoenol pyruvate carboxylaseand glycolate oxidase from jute (Corchorus olitorius L.; cv.JRO 632) leaves were compared with those from maize (C4) andsunflower (C3) leaves. The photosynthetic CO2 fixation products,the release of 14CO2 in light and dark following photosynthesisin 14CO2, chlorophyll a: b ratio, gross leaf photosyntheticrate and dry matter production rate were also studied. Leaves of different ages from B. calycinum were exposed to 14CO2in light during day and night. The labelling pattern on thechromatogram differed with leaf age. Young leaves had similarpatterns to those of C3 plants during both day and night. Matureleaves showed high incorporation of 14C into C4 acids, especiallyat night. In contrast, no significant difference with leaf agewas observed in the pattern of dark 14CO2 fixation products.Study of the enzyme activity and the content of titratable acidat each leaf age suggested that high incorporation of 14C inC4 acids during the night was due to the simultaneous absorptionof CO2 by both enzymes RuDPcarboxylase and PEPcarboxylase. Four co-existing species (Deschampsia flexuosa, Festuca ovina,Juncus squarrosus and Nardus stricta) were subjected to clippingand the net photosynthetic and dark respiration rates were followedafter this treatment for 50 d. Concurrently carbon partitioningin F. ovina plants clipped initially and again at 50 and 100d was examined. An expansion of new leaf lamina was observed with F. ovina,which had a greater net photsynthetic rate per unit leaf areathan unclipped lamina. The remaining leaf lamina (stubble) afterclipping also showed net photosynthetic and dark respirationrates greater than unclipped lamina; these responses were uniqueto F. ovina plants. N. stricta was the only other species toattain a pre-clipping photosynthetic rate within 6 d. Clipped F. ovina plants showed a change in carbon allocationpattern, with a reduction in carbon allocated to roots. 14Caccumulated in roots and stubble was shown to have a role inregrowth, as was current assimilate via the production of newleaf lamina. Plants initially clipped before exposure to 14C,redistributed less 14C to new shoot growth and, therefore, lostless when subsequently clipped. Further redistribution of 14Ccame from leaf stubble tissue and not at the expense of roots.The variation between species in clipping response are discussedin terms of the implications for coexistence. The relative assimilatory activity of the inflorescence, itsindividual components, and the leaves of flowering tillers ofPoa annua L. and Lolium perenneL. was determined over the periodfrom inflorescence emergence to seed shedding. The pattern of14CO2 fixation was similar for both species and the inflorescencewas by far the most important assimilatory organ of the reproductivetiller, particularly over the latter period of seed developmentas leaf senescence progressed. With the exception of the seedsall parts of the inflorescence showed significant assimilatoryactivity and the lemmas and paleas accounted for 40–50per cent of the total 14C fixed by the inflorescence in bothspecies. The importance of the grass inflorescence as a photosyntheticstructure is discussed in relation to similar studies on cereals. The photosynthetic capacity of newly expanded leaves of vernalizedor non-vernalized plants of S24 perennial ryegrass (Lolium perenneL.), grown in long or short photoperiods, was measured in twoexperiments. In the first, leaves were protected from shadingduring development, while in the second, the natural shade ofneighbouring tillers in a sward was allowed. In the first experiment there was little effect of vernalization,day length or flowering, and leaves in all treatments had photosyntheticrates at 250 W m–2 of between 28 and 32 mg CO2 dm–2h–1.In the second experiment the photosynthetic rate of successiveleaves fell as sward leaf area increased. This downward trendwas reversed, however, in flowering tillers in the vernalizedlong-day treatment, while in the other treatments, which didnot flower, photosynthetic capacity continued to fall. It isconcluded that the leaves of reproductive tillers have highphotosynthetic capacities because stem extension carries themto the top of the canopy where they are well illuminated duringexpansion. The rates of gross photosynthesis of the flag leaf and the nextleaf below (second leaf) in crops of winter wheat were estimatedfrom the 14C uptake of the leaves after exposure to short pulsesof 14CO2. The photosynthetic rates of both leaves during thegrain-filling period decreased with increase in nitrogen fertilizerbecause the intensity of photosynthetically active radiationwas less at the surface of the leaves in the dense crops withadditional nitrogen. In addition, the rate of photosynthesisat saturating light intensity was slightly decreased by nitrogen.The effects of nitrogen, in decreasing the rate of photosynthesisper unit area of leaf and in increasing the leaf-area indexof the top two leaves, were such that the photosynthetic productivityper unit area of land of the flag leaf was increased by nitrogenbut the productivity of the second leaf was unaffected. Applying180 kg N ha–1 increased the productivity of the top twoleaves by a factor of 2.3 but increased grain yield by only1.8. The photosynthetic productivity of the second leaf duringthe grain-filling period was about half that of the flag leaf. There was no difference in photosynthetic rate per unit areaof leaves of Cappelle-Desprez and Maris Huntsman which couldaccount for the larger yield of the latter cultivar. There wasa slight indication that the leaves of the semi-dwarf cultivarsMaris Fundin and Hobbit photosynthesized faster than those ofMaris Huntsman. The respiratory losses and the pattern of carbon supply froma leaf of unicuim barley were examined during a complete diurnalperiod using a steady state 14C-labelling technique. After a delay of c. 1 h a portion of the 14C exported from acontinuously assimilating leaf was lost in respiration in thelight. This respiratory loss amounted to c. 20% of the total14C fixed. A further 28% of the total 14C fixed was respiredduring the dark period. In the light, carbon was fixed at a rate of c. 8·9 mgC dm–2 h–1 and exported from the leaf at c. 5·3mg C dm–2 h–1. Dark export averaged c. 31% of theday-time rate. Carbohydrate was stored in the leaf during the day and was almostcompletely remobilized during the dark. Sucrose, the major reservecarbohydrate, was exported first whilst the starch level remainedconstant. After some 9 h of darkness, sucrose declined to alow level and starch remobilization began.  相似文献

Nitrate reductase activity (in vivo method, substrate non-limiting)in unshaded leaves from the top of the canopy has been determinedfor field-grown potato plants over the course of the growingseason. The pattern of change was almost identical for plantsreceiving no added fertilizer and those receiving 24 g N m–2.Activity increased to a peak at about 90 days after plantingand declined thereafter. On a fresh weight basis activity wasalways higher in fertilized plants. Nitrate reductase activitywas positively and significantly correlated with leaf proteincontent in high N plants (r2 = 0.71; P = 0.05), but poorly correlatedwith both the nitrate content of the leaf lamina and the nitrateconcentration in petiole sap. Up until 90 days after planting(mid-July) there appeared to be a positive relationship betweenincreased activity of nitrate reductase and solar radiation.However, results obtained over two seasons showed that the declinein activity after this time was not consistently linked witha fall in the level of solar radiation. Remobilization of reduced-Nand stored nitrate from leaves and stems accompanied this declinein nitrate reductase activity and in the latter part of theseason appeared to account for all of the N gained by growingtubers. In unfertilized plants nitrate-N accounted for 5 per cent orless of total plant N. Fertilized plants contained up to 25per cent nitrate-N. While nitrate availability limited growthin unfertilized plants, sub-optimal rates of nitrate assimilationin fertilized plants, particularly during the early stages ofpost-emergence growth, may contribute to inefficient use ofacquired nitrate. The carbohydrate status of leaf lamina and petiole sap weremodified by N supply. The soluble sugar and starch contentsof low N leaves were higher than in their high N counterparts.By contrast, the concentration of soluble sugars in petiolesap increased to a higher value in high N samples. Althoughsap sugar levels declined in both treatments towards the endof the season, N application delayed this decline for severalweeks. Petiole heat-girdle treatments (followed by a 5 min 14CO2 assimilation)were performed on mature leaves of Vicia faba, in order to assesstheir effect on the partitioning of photo-assimilates to theminor vein phloem. Whole leaf autoradiographic evidence indicateda high leaf-to-leaf variation in the image intensity over theminor veins (relative to the mesophyll/epidermal background)in both control and heat-girdled groups of leaves. The averagedegree of minor vein labelling in heat-girdled leaves, however,was found to be significantly lower than that in controls. Comparativeassessment of vein labelling was based on microscopic densityreadings of silver grains over veinal and interveinal regionsin autoradiographic images. Investigations into the cause ofthis alteration in vein labelling indicated no involvement ofan inhibition of apoplasmic phloem loading, as both heat-girdledand control leaves of Vicia were shown to have comparable minorvein uptake of exogenously supplied 14C-sucrose. Heat-girdlingwas shown, however, to increase significantly the partitioningof recently fixed carbon into the insoluble (mainly starch)fraction relative to the ethanol-soluble fraction, within 12min of the treatment. We suggest that this carbon partitioningchange can primarily account for the change in vein labelling,since an increase in the insoluble fraction would result in(1) more 14C-activity remaining in the leaf mesophyll and (2)less 14C-activity going into the mesophyll export pool, andthus, less 14C-sucrose being transferred to the minor vein region.Additionally, although leaf export was completely halted inheat-girdled leaves, 14C-activity was found within the majorveins as far as the point of petiole heat-girdling (followinga 5 min assimilation and 4 h chase). Apparently, continued (butlimited) solution flow within the sieve elements is maintainedby transport pathway unloading within the treated leaves. An investigation was made to study the assimilation and transportof 15N-labelled nitrate nitrogen in rice plant (Oryza sativaL.). Nitrogen from labelled nitrate at the end of plant feedingwas found mainly in nitrate form, and was more prevalent inroots, stem and leaf sheaths. The nitrite fraction had the nextlargest 15N enrichment. The 15NO3 assimilation in the newlyemerged panicle was mainly in amide and amino acid. The 15N-incorporation at day 0 was greatest in amino acid andnitrate of roots and decreased towards the stem and leaves.Incorporation in these fractions considerably decreased fromday 0 to day 10. Probably most of the nitrogen from the nitratesource was transported from the roots to the shoot in nitrateand amino acid forms. A decrease of 15N-incorporation in the soluble N fraction andincrease in the insoluble N fraction from day 0 to day 10 inplant parts, particularly the blades, suggested that proteinsynthesis occurred mostly in young parts of the shoot duringthis period. The marked variation in 15N distribution in differentparts of the plant during the 10 days indicated that the nitrogenin roots and tillers was probably remobilized and transportedto other parts, particularly the upper leaf blades. The rates of 14CO2 fixation and translocation of labelled assimilatesin the leaf below the topmost fully expanded leaf of four sugarcaneclones with widely differing weights of leaf per unit weightof cane (L/J ratio) and storage cell volumes, were measuredin the field. Differences in the rate of fixation and rate andvelocity of translocation were found between the clones and,within species groups, the former only was related to the L/Jratio and to storage cell volume A possible restriction of translocationthrough the Lamina/sheath junction (dewlap), indicated by abuild up of radioactivity just distal to the dewlap, occurredin two clones and it is suggested that the efficiency of translocationthrough the dewlap is another factor which may influence thephotosynthetic rate of sugarcane leaves. C4 model plants composed of single-rooted Amarantus cruentusL. leaves were developed to study source-sink relationships.The photosynthetic activity of CO4 fixation in the rooted leavescultured under the control condition (10-h light/14-h darkness,730 µmol photons m–2s–1) remained constantand high. When the leaves kept for 8 or 9 d under the controlcondition were exposed to continuous light (CL) for 4 d, theactivity decreased steeply. The starch content increased significantlyduring the first 2 d in CL whereas the sucrose content increasedalmost linearly during the 4 d in CL. The close relationshipbetween the decrease in photosynthetic activity and the increasein sucrose indicates that the activity was subjected to feedbackinhibition when the end product was forced to accumulate inthe leaves. In the treated leaves, the initial activity of ribulose-1,5-bisphoshatecarboxylase decreased significantly. A large increase in ribulose-1,5-bisphosphateand decrease in 3-phosphoglycerate occurred following the deactivationin RuBPcase, which was fully restored by the preincubation withCO2 and Mg2+ and/or P1 and Mg2+. The activity of phosphoenolpyruvatecarboxylase also decreased but not as rapidly as RuBPcase. Thealanine and pyruvate levels decreased markedly but the oxaloacetate,glutamine and asparagine levels significantly increased whilephosphoenolpyruvate and glutamate were kept at almost constantlevels. The malate level first increased slightly and then decreasedsignificantly. The mechanism of coordinated decrease in carboxylationreactions between phtosynthetic carbon reduction cycle and C4metabolism in response to change in the source-sink balanceis discussed.
