Sequences were determined for the nuclear-encoded small subunit (SSU) rRNA and 5.8S rRNA genes as well as the internal transcribed spacers ITS1 and ITS2 of the parasitic dinoflagellate genus Syndinium from two different marine copepod hosts. Syndinium developed a multicellular plasmodium inside its host and at maturity free-swimming zoospores were released. Syndinium plasmodia in the copepod Paracalanus parvus produced zoospores of three different morphological types. However, full SSU rDNA sequences for the three morphotypes were 100% identical and also their ITS1-ITS2 sequences were identical except for four base pairs. It was concluded that the three morphotypes belong to a single species that was identified as Syndinium turbo, the type species of the dinoflagellate subdivision Syndinea. The SSU rDNA sequence of another Syndinium species infecting Corycaeus sp. was similar to Syndinium turbo except for three base pairs and the ITS1-ITS2 sequences of the two species differed at 34-35 positions. Phylogenetic analyses placed Syndinium as a sister taxon to the blue crab parasite Hematodinium sp. and both parasites were affiliated with the so-called marine alveolate Group II. This corroborates the hypothesis that marine alveolate Group II is Syndinea.  相似文献

Anilocra pilchardi n. sp. is described from several specimens collected mainly from clupeiform fishes from off Lebanon. Various stages of the parasite are considered: ovigerous and non-ovigerous females, male and intramarsupial larvae. Taxonomic comments on the relationship of A. pilchardi to other congeneric species are given. A. pilchardi belongs to a particular group of Anilocra species previously known only from the Indo-Pacific Region.  相似文献

Neoechinorhynchus (Neoechinorhynchus) mexicoensis sp. n. is described from the intestine of Dormitator maculatus (Bloch 1792) collected in 5 coastal localities from the Gulf of Mexico. The new species is mainly distinguished from the other 33 described species of Neoechinorhynchus from the Americas associated with freshwater, marine and brackish fishes by having smaller middle and posterior hooks and possessing a small proboscis with three rows of six hooks each, apical hooks longer than other hooks and extending to the same level as the posterior hooks, 1 giant nucleus in the ventral body wall and females with eggs longer than other congeneric species. Sequences of the internal transcribed spacer (ITS) and the large subunit (LSU) of ribosomal DNA including the domain D2 + D3 were used independently to corroborate the morphological distinction among the new species and other congeneric species associated with freshwater and brackish water fish from Mexico. The genetic divergence estimated among congeneric species ranged from 7.34 to 44% for ITS and from 1.65 to 32.9% for LSU. Maximum likelihood and Bayesian inference analyses with each dataset showed that the 25 specimens analyzed from 5 localities of the coast of the Gulf of Mexico parasitizing D. maculatus represent an independent clade with strong bootstrap support and posterior probabilities. The morphological evidence, plus the monophyly in the phylogenetic analyses, indicates that the acanthocephalans collected from intestine of D. maculatus from the Gulf of Mexico represent a new species, herein named N. (N.) mexicoensis sp. n.  相似文献

The systematic relationships of acanthocephalans, including Leptorhynchoides and Pseudoleptorhynchoides that occur in freshwater and marine fishes in Neartic and Neotropical regions, are enigmatic. Leptorhynchoides (3 species) and Pseudoleptorhynchoides (1 species) are presently classified in the Rhadinorhynchidae. However, recent molecular and morphological phylogenies have challenged the monophyly of this family. Sequences of nuclear ribosomal DNA (large subunit, small subunit regions) and the mitochondrial cytochrome c oxidase subunit I gene of Leptorhynchoides thecatus and Pseudoleptorhynchoides lamothei were used in phylogenetic analyses with available sequences of 26 other acanthocephalans. Maximum parsimony and maximum likelihood analyses were identical in placing both genera in the Illiosentidae. Bootstrap analyses also indicate that placement of these genera with members of Illiosentidae is reliably supported.  相似文献

To better understand the phylogenetic status of the snakehead, Channa argus, we determined its complete mitogenome sequence using long-polymerase chain reaction and the direct sequencing method. The complete mitogenome sequence was 16,559?bp in length and contained 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and 1 control region (D-loop), the gene composition/order of which was identical to that observed in most other vertebrates. This was the first report of the mitogenome sequence in suborder Channoidei. Phylogenetic relationships of 14 perciform suborders based on mitogenome sequences were reconstructed using Bayesian inference and maximum likelihood methods. The results strongly supported the monophyly of Perciformes and the snakehead, as a representative species of suborder Channoidei, formed the most basal branch having sister relationship with the clade containing all other analyzed perciform fishes. The further phylogenetic analyses of six channid species, based on cytochrome b gene, suggested that two channid genera constituted reciprocally monophyletic clades. In addition, the relaxed molecular clock method was used to estimate divergence dates among major suborders of Perciformes and major species in Channoidei.  相似文献

Pomphorhynchus purhepechus n. sp. is described from the intestine of the Mexican redhorse Moxostoma austrinum Bean (Catostomidae) in central Mexico. The new species can be distinguished from the other seven described species of Pomphorhynchus Monticelli, 1905 in the Americas by a subspherical proboscis and 14 longitudinal rows with 16–18 hooks each; the third and the fourth row of hooks are alternately longest. Sequences of the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene and the large subunit (LSU) rDNA (including the domains D2-D3) were used to corroborate the morphological distinction between the new species and Pomphorhynchus bulbocolli Linkins in Van Cleave, 1919, a species widely distributed in several freshwater fish species across Canada, USA, and Mexico. The genetic divergence estimated between the new species and the isolates of P. bulbocolli ranged between 13 and 14% for cox1, and between 0.6 and 0.8% for LSU. Maximum likelihood and Bayesian inference analyses of each dataset showed that the isolates of P. bulbocolli parasitising freshwater fishes from three families, the Catostomidae, Cyprinidae and Centrarchidae, represent a separate lineage, and that the acanthocephalans collected from two localities in central Mexico comprise an independent lineage. In addition, our analysis of the genetic variation of P. bulbocolli demonstrates that individuals of this acanthocephalan from different host species are conspecific. Finally, the distribution, host-association, and phylogenetic relationship of the new species, when placed in the context of the region’s geological history, suggest that both host and parasite underwent speciation after their ancestors became isolated in Central Mexico.  相似文献

Juvenile parasitic cymothoid isopods (mancae) can injure or kill fishes, yet few studies have investigated their biology. While the definitive host of the adult cymothoids is usually a single host from a particular fish species, mancae may use so-called optional intermediate hosts before settling on the definitive host. Little, however, is known about these early interactions. The cymothoid isopod, Anilocra apogonae, infests the definitive host, Cheilodipterus quinquelineatus. This study examined their host preference among potential optional intermediate hosts. Their effect on the growth and mortality of the young of three apogonid fishes, including the definitive host, was investigated. The number of mancae produced per brood was positively correlated with female length. When given a choice of intermediate hosts, significantly more mancae attached to Apogon trimaculatus (Apogonidae) than to Apogon nigrofasciatus. When presented with Ap. trimaculatus and Pomacentrus amboinensis (Pomacentridae), mancae only attached to Ap. trimaculatus suggesting that mancae may show a taxonomic affiliation with preferred hosts. Mancae fed on all three apogonid species, with C. quinquelineatus being fed on earlier than Ap. trimaculatus and Ap. nigrofasciatus. Mancae feeding frequency, adjusted for fish survival, was lowest on C. quinquelineatus and highest on Ap. trimaculatus. Infested apogonids had reduced growth and increased mortality compared with uninfested fish. A. apogonae mancae can use several species of young apogonid fishes as optional intermediate hosts. Via reduced growth and increased mortality, mancae have the potential to negatively influence definitive host populations and also other young species of apogonid fishes.  相似文献

Nematodes belonging to the genus Hysterothylacium (family Raphidascarididae) infect various species of marine fish in both the larval and adult stages. Humans can be accidentally infected upon eating infected seafood. In spite of their importance, relatively little is known of their occurrence and systematics in Australia. An examination of various species of marine teleosts in Australian waters revealed a high prevalence of Hysterothylacium larval types. In the present study, seven previously undescribed Hysterothylacium larval morphotypes (V to VII and IX to XII) were discovered. In total we found 10 different morphotypes and we genetically characterised nine morphotypes identified. A morphological dichotomous identification key has been established to differentiate these morphotypes. Since some larvae of Hysterothylacium from marine fishes cannot be differentiated morphologically from other nematode larvae, such as Paraheterotyphlum, Heterotyphlum, Iheringascaris and Lapetascaris, the first and second internal transcribed spacers (ITS-1 and ITS-2) of nuclear ribosomal DNA (rDNA) of these larvae were characterised to confirm their taxonomic status. This genetic characterisation implied that some distinct morphotypes belong to different developmental stages of the same species. In addition, it revealed that some morphotypes can comprise distinct genotypes. No match was found between ITS-1 and ITS-2 sequences obtained from larvae in the present study and those from adults available in the GenBank, highlighting the lack of knowledge on occurrence of adult nematodes infecting Australian fish.  相似文献

The porgies (Sparidae) comprise a diverse group of neritic fishes with a broad geographic distribution. We used mitochondrial DNA sequences from partial 16S ribosomal RNA and cytochrome b genes to reconstruct the phylogenetic history of these fishes. Sequences from 38 sparid species, 10 species in outgroups closely related to sparids, seven basal percoid species, and a non-perciform outgroup species were analyzed with parsimony and maximum likelihood. The Sparidae were monophyletic with the inclusion of Spicara, which is currently placed in the Centracanthidae. The genera Spicara, Pagrus, and Pagellus, were not monophyletic indicating a need for revision. Two main sparid lineages were recovered in all analyses, but the previously proposed six sparid subfamilies (Boopsinae, Denticinae, Diplodinae, Pagellinae, Pagrinae, and Sparinae) were not monophyletic. This suggests that dentition and feeding modes, upon which these subfamilies are based, were independently derived multiple times within sparid fishes. There was no evidence from the 16S or combined analyses for a monophyletic Sparoidea.  相似文献

A new tick species belonging to the African subgenus Afrixodes Morel, 1966, namely, Ixodes ( Afrixodes ) fynbosensis n. sp., is described. The female of I. fynbosensis is easily differentiated from the other African Ixodes species by a large, tapering triangular ventrolateral spur on palpal segment I. Nymph and larva of I. fynbosensis can be distinguished from those of other members of Afrixodes by a combination of the following characters: pointed hypostome, long auriculae, long and acute ventrolateral projections of basis capituli of nymph, only 2 pairs of central dorsal setae, and a straight posterior margin of scutum of the larva. Cytochrome oxidase I mtDNA sequence comparisons between I. fynbosensis and 10 other Ixodes Latreille, 1795, species support the recognition of this taxon as genetically distinct (>13% corrected sequence divergences separate it from the remainder of the 10 recognized species used in this study), and preliminary phylogenetic analyses reveal that this taxon is most closely related to the southern African Ixodes pilosus Koch, 1844, and Ixodes rubicundus Neumann, 1904. Ixodes fynbosensis is known only from South Africa, where females have been collected from a domestic dog and a rodent, Rhabdomys pumilio (Sparrman), and nymphs and larvae have been collected from R. pumilio and unidentified shrews belonging to the Soricidae. Sequences generated for both nymphs and adult individuals were identical, confirming the correlation between the described life stages.  相似文献

选择线粒体COⅠ基因作为分子标记,进行沙鳅亚科鱼类(Botiinae)DNA条形码及其分子系统发育研究。研究获得了沙鳅亚科7属19种共131个个体的COⅠ基因序列,利用MEGA5.0软件分析了沙鳅亚科鱼类COⅠ基因的序列特征,计算了种内及种间遗传距离。沙鳅亚科鱼类的分子系统发育关系的重建分别采用NJ法和Bayesian法。研究发现,沙鳅亚科COⅠ基因的碱基组成为: A 24.4%、T 29.5%、G 18.0%、C 28.1%。沙鳅亚科鱼类种内平均遗传距离为0.0020.000,种间平均遗传距离为0.1480.008。DNA条形码研究结果显示,所分析的19种沙鳅鱼类各自分别聚成单系分支,表明COⅠ基因在本研究中具有100%的物种鉴别率。同时,系统发育分析支持各属的单系性,并且结果显示沙鳅亚科鱼类聚为两个分支,其中一支由薄鳅属和副沙鳅属构成,另一分支则包括: (沙鳅属、色鳅属)和 中华沙鳅属、(缨须鳅属、安彦鳅属)。因此,COⅠ基因可以作为有效的分子标记对沙鳅亚科进行DNA条形码研究以及分子系统发育研究。    相似文献

Cichlid fishes have played an important role in evolutionary biology and aquaculture industry. Nile tilapia (Oreochromis niloticus), blue tilapia (Oreochromis aureus) and Mozambique tilapia (Oreochromis mossambicus), the useful models in studying evolutionary biology within Cichlid fishes, are also mainly cultured species in aquaculture with great economic importance. In this paper, the complete nucleotide sequence of the mitochondrial genome for O. niloticus and O. aureus were determined and phylogenetic analyses from mitochondrial protein-coding genes were conducted to explore their phylogenetic relationship within Cichlids. The mitogenome is 16,625 bp for O. niloticus and 16,628 bp for O. aureus, containing the same gene order and an identical number of genes or regions with the other Cichlid fishes, including 13 protein-coding genes, two rRNA genes, 22 tRNA genes and one putative control region. Phylogenetic analyses using three different computational algorithms (maximum parsimony, maximum likelihood and Bayesian method) show O. niloticus and O. mossambicus are closely related, and O. aureus has remotely phylogenetic relationship from above two fishes.  相似文献

草兔(Lepus capensis)在我国数量多,分布广,但人们对草兔的系统地理学、草兔与其他兔类的系统发育关系了解不多,其亚种水平的分类也长期存在争议。本研究通过LA-PCR 技术对草兔线粒体基因组全序列进行PCR 扩增、序列测定和分析,并基于线粒体基因组12 个蛋白质编码基因10 776 bp的核苷酸序列,采用贝叶斯法(BI)和最大似然法(ML)构建哺乳纲(Mammalia) 灵长总目(Euarchontoglires)5 个目12 种动物的系统发育关系,结果支持兔形目(Lagomorpha)的单系起源,其中草兔与欧洲野兔(L. europaeus)亲缘关系最近,两者互为姐妹群,进而与家兔(Oryctolagus cuniculus)构成姐妹群关系。此外,基于线粒体细胞色素b 基因全序列,构建31 个兔类个体的系统发育关系,结果显示,草兔与雪兔(L. timidus),云南兔(L. comus) 与高原兔(L.oiostolus),东北兔(L. mandschuricus)与塔里木兔(L. yarkandensis) 有较近的亲缘关系,而在草兔内部,中国的草兔与南非草兔亲缘关系较远,采自江苏盐城的草兔与来自山东、陕西、四川的草兔聚为一支,具有明显较近的亲缘关系。该研究为更好地探讨草兔系统发育关系提供了进一步的分子生物学资料.  相似文献

The tubificid clitellates are a common component in the freshwater bottom fauna and are also the most abundant oligochaete group in marine habitats. There are over 800 described species classified in six subfamilies; Tubificinae, Limnodriloidinae, Rhyacodrilinae, Telmatodrilinae, Phallodrilinae, and Naidinae. In this study we examine the phylogenetic relationships in Tubificidae using a combination of mitochondrial 16S rDNA and nuclear 18S rDNA sequence data. Sequences were obtained from five outgroup and 56 ingroup taxa, including five of the six subfamilies of Tubificidae. The data were analysed by maximum parsimony and Bayesian inference. The resulting tree topologies are virtually without conflict. Several associations traditionally recognized within the family Tubificidae are supported, in the Bayesian analysis including a sister group relationship between Tubificinae and Limnodriloidinae. The results also indicate that Rhyacodrilinae is polyphyletic--some of its members (Heterodrilus spp.) fall into a clade with Phallodrilinae, all other groups with Naidinae. Naidinae is also polyphyletic with two rhyacodriline genera, Monopylephorus and Ainudrilus, nested within. Most of the tubificid genera included in the study are supported as monophyletic; however, Tubifex and Limnodriloides are refuted, and Tubificoides is unresolved from other tubificine taxa.  相似文献

Adult tetraphyllidean tapeworms (Platyhelminthes: Eucestoda) from the spiral intestines of 3 species of potamotrygonid stingrays (Paratrygon aiereba, Potamotrygon castexi, and Portamotrygon motoro) in the Madre de Dios river in Peru were found to host numerous cysts embedded in their parenchymal tissues. Histological sections of the cysts revealed the presence of a scolex bearing 4 suckers and an unarmed apical organ consistent with larval stages of both Cyclophyllidea and Proteocephalidea. To further elucidate their identities, partial 28S ribosomal DNA (rDNA) sequences were characterized from 3 cysts and 4 adult Rhinebothrium spp. 'host' worms and screened against all available cestode 28S rDNA data. Initial BLAST screening and subsequent alignment ruled out the possibility that the cysts were cyclophyllidean, and the cyst and adult sequences were thus aligned together with all available lecanicephalidean, litobothriidean, proteocephalidean, and tetraphyllidean sequences. Sequences from all 3 cysts were identical, and phylogenetic analysis clearly placed them among derived members of the Proteocephalidea, although no exact match was found. Sequences from the adult host worms formed 2 identical pairs and grouped together with other tetraphyllidean species from rays. These results are compared with records of hyperparasites of South American catfish cestodes. This is the first confirmed record of a proteocephalidean cestode parasitizing a tetraphyllidean cestode.  相似文献

We describe 11 new species of cryptogonimids belonging to Retrovarium n. gen., from eight species of Lutjanidae and one species of Haemulidae, from the Great Barrier Reef, French Polynesia and the Maldives. We also transfer Neoparacryptogonimus saccatus (Manter, 1963) and Neoparacryptogonimus sphericus Nahhas, Sey & Nishimoto, 1998 to Retrovarium. The morphologically based taxonomic approach was augmented with DNA sequence data from three nuclear ribosomal DNA regions (28S, ITS1 and ITS2) to explore the species integrity, biogeographic distribution and evolution of the species recognised here. Sequencing included multiple replicates and revealed 11 distinct genotypes which corroborated our morphologically based hypotheses of putative species present in the system. There was no intraspecific variation and all three rDNA regions differed between every combination of species. Two species exhibited wide geographic ranges, having identical rDNA sequences between the Great Barrier Reef and the Maldives, localities separated by over 9600 km. One host species, Symphorus nematophorus, proved to be exceptionally rich, harbouring six species. Minimum evolution analyses were conducted on each of the rDNA datasets independently; minimum evolution, maximum likelihood and Bayesian inference analyses were conducted on a combined (28S, ITS1 and ITS2) dataset for sequence comparison purposes and to explore the evolutionary history of these parasites. To examine the coevolutionary history of this complex, assessment of phylogenetic relationships between the 23 species of Lutjanidae and two species of Haemulidae collected during this survey was performed with data from 16S and cytochrome b mtDNA using Bayesian inference analysis. Despite the high host specificity observed in most of the species, mapping of the parasites on the host phylogeny revealed an absence of strict coevolution or co-descent within this complex. Overall, Retrovarium appears to have had an exceptionally patchy radiation, failing to infect many taxa, infecting species with no readily discernible pattern, and radiating dramatically within one species.  相似文献

Populations of Trichuris spp. isolated from six species of sigmodontine rodents from Argentina were analyzed based on morphological characteristics and ITS2 (rDNA) region sequences. Molecular data provided an opportunity to discuss the phylogenetic relationships among the Trichuris spp. from Noth and South America (mainly from Argentina). Trichuris specimens were identified morphologically as Trichuris pardinasi, T. navonae, Trichuris sp. and Trichuris new species, described in this paper. Sequences analyzed by Maximum Parsimony, Maximum Likelihood and Bayesian inference methods showed four main clades corresponding with the four different species regardless of geographical origin and host species. These four species from sigmodontine rodents clustered together and separated from Trichuris species isolated from murine and arvicoline rodents (outgroup). Different genetic lineages observed among Trichuris species from sigmodontine rodents which supported the proposal of a new species. Moreover, host distribution showed correspondence with the different tribes within the subfamily Sigmodontinae.  相似文献

During independent surveys of freshwater ascomycetes in Japan and USA two new species of Lindgomyces were collected from submerged wood in freshwater. These species are described and illustrated based on morphological data and phylogenetic relationships based on analyses of nuclear ribosomal sequence data (partial SSU and LSU, and ITS). Lindgomyces apiculatus, collected in Japan, is characterized by immersed to erumpent, globose to subglobose ascomata; fissitunicate, cylindrical to clavate asci; and fusiform, one-septate ascospores with acute ends and short terminal appendages. Lindgomyces lemonweirensis, collected in Wisconsin, USA, differs from L. apiculatus in having clavate to cymbiform asci and oblong to fusiform ascospores that are distinctively multiguttulate and surrounded by an oval, ephemeral gelatinous sheath. The new species formed a strongly supported clade within the family Lindgomycetaceae (Pleosporales, Dothideomycetes) based on analyses of combined SSU and LSU sequence data. In addition phylogenetic analyses with ITS sequence data support the establishment of the new taxa as separate species within Lindgomyces because they were separated from each other and other Lindgomyces species based on maximum likelihood bootstrap and Bayesian analyses.  相似文献

This work is a prospective study to estimate the potential species diversity of terrestrial planarians in the Iberian Peninsula. Live specimens were collected from several Iberian localities and assigned to different morphotypes on the basis of their external morphological characteristics. From the same specimens, sequences from the mitochondrial Cytochrome Oxidasa subunit I (COI) and from the nuclear ribosomal gene 18S rRNA were obtained. Sequences from GenBank of the families Dendrocoelidae, Planariidae, Bipaliidae, Geoplanidae and Rhynchodemidae have been used as outgroups in the phylogenetic analysis. The results showed that terrestrial planarians have a wide distribution in the Peninsula, with all individuals found belonging to the Rhynchodemidae family. Morphological observations indicated the presence of 10 morphospecies confirmed by the molecular analyses. At the same time, COI sequences were successfully used as a molecular marker for species identification in the barcoding mode, which is of great use in groups like this with few external morphological characteristics. The combined data strongly suggest the presence of at least 15 species in the Iberian Peninsula, a number that nearly doubles previous estimates, indicating that terrestrial planarians are more diverse than expected in the region and, as proposed, may be a good biodiversity indicator and model for biogeographical studies.  相似文献
