A portable recording system was developed for analysis of more than three analog signals collected in field works. Stereo audio recorder, available as consumer products, was made use for a core cornponent of the system. For the two tracks of recording, a multiplexed analog signal is stored on one track, and reference code on the other track. The reference code indicates the start of one cycle for multiplexing and swiching point of each channel. Multiplexed signal is playbacked and decoded with a reference of the code to reconstruct original profiles of the signal. Since commercial stereo recorders have cut DC component off, a fixed reference voltage is inserted in the sequence of multiplexing. Change of voltage at switching from the reference to the data channel is measured from playbacked signal to get the original data with its DC component. Movement of vehicles and human head were analyzed by the system. It was verified to be capable to record and analyze multi-channel signal at a sampling rate more than 10Hz.  相似文献

Summary Treatment of 1-day-old worker honey bees with the juvenile hormone analog methoprene prematurely reduced their behavioral threshold sensitivity to alarm pheromones. Electroantennogram assays indicated that peripheral perception was not affected; the effects apparently occurred in the central nervous system. These results support a model (Robinson 1987) of division of labor based on the hormonal regulation of response thresholds to task-associated stimuli. Coupled with other studies (Breed 1983; Robinson 1985), they also suggest that: 1) aggression in the honey bee is hormonally mediated and 2) a single hormone is involved in both the production and perception of an olfactory communication signal, the first report of this phenomenon in an invertebrate.Abbreviations JH juvenile hormone - CNS central nervous system - EAG electroantennogram  相似文献

An early warning system for biological monitoring of aquatic environment is proposed, which is based on an analysis of mussel valve movements. It consists of an analog monitor of an original design (analog recorder and analyzer of opening/closing of valves in a group of mussels) and outside devices (multimeters, computers, and mobile phones). In the monitor, an analysis of the state of valve position sensors, summing/averaging of voltage values, and generation of a warning signal are performed in the analog mode. The proposed system (a) diagnoses the state of valve position sensors, (b) determines the voltage representing the average valve opening in a group of mussels (6–12 individuals), (c) generates a warning signal; and, by means of peripheral devices, (d) registers the average voltage of sensors and (e) transmits a warning signal via a mobile communication net. The system can provide an addition (and, sometimes, an alternative) to complicated digital systems analogous to MosselMonitor®.  相似文献

RNA interactions with protein and small molecule ligands serve a wide variety of biochemical functions in the cell. To best understand the specificity and affinity of these interactions, the free energy contribution made by individual function groups in the RNA must be determined. As an efficient method for obtaining such energetic profiles, we report quantitative nucleotide analog interference mapping (QNAIM). This extension of the NAIM methodology uses the magnitude of analog interference as a function of ligand concentration to calculate binding constants for RNA with individual analog substitutions. In this way, QNAIM not only defines which functional groups are important to an interaction but simultaneously determines the energetic contribution made by each occurrence of that functional group within the RNA polymer. To establish the utility of this approach, QNAIM was used to quantify functional group interactions within the signal recognition particle (SRP), specifically the 4.5S RNA with the M domain of Ffh. In each of the cases in which energetic data were available from previous site-specific substitution analyses, QNAIM provided nearly equivalent results. These experiments on a model system demonstrate that QNAIM is an efficient method to establish a chemically detailed free energy profile for a wide variety of RNA-ligand interactions.  相似文献

In the third part of this series, a data acquisition system is described which digitizes the analog signal from a commercial infrared spectrometer and records the data on paper tape. The paper tape is then processed off-line with a microcomputer and an infrared spectrum is printed. The correction routine for absorbance due to the O-H stretching band is discussed as well as the integration limits for calculation of integrated intensities of the C-H stretching band. The computer program is briefly described and statistical data on correlation between integrated intensity and lipid concentration given.  相似文献

A telemetric system is described which allows the wireless registration of extracellular neuronal activity and vocalization-associated skull vibrations in freely moving, socially living squirrel monkeys (Saimiri sciureus). The system consists of a carrier platform with numerous guiding tubes implanted on the skull. Custom-made microdrives are mounted on the platform, allowing the exploration of two electrode tracks at the same time. Commercially available quartz-insulated platinum-tungsten microelectrodes are used. The electrodes can be moved over a distance of 8-10 mm by turning a screw on the microdrive. Vocalization-associated skull vibrations are recorded with a piezo-ceramic element. Skull vibration signal and the signals from the two microelectrodes are fed into separate transmitters having different carrier frequencies. The signals are picked up by an antenna in the animal cage and are sent to three receivers in the central laboratory. Here, the signals are transferred via an analog/digital interface to a personal computer for data analysis and to a video recorder for long-term storage. The total weight of the head mount including carrier platform, microdrive, electrodes, skull vibration sensor, three transmitters, and protection cap is 32 g. The transmitters are powered with two rechargeable lithium batteries, allowing about 8 h of continuous recording. Reliable signal transmission is obtained over a distance of about 2 m. Recording stability allows to follow the activity of specific neurons up to several hours, with no movement artefacts during locomotion.  相似文献

A seven-channel telemetry system for collection and display of biological data is presented. The system can amplify bioelectrical signals in the range of 2 μV to 200 mV and has a bandwidth of 0.1–80 Hz. After multiplexing, the signals are digitized with a resolution of 8 bits. The data are frequency modulated directly on a VHF transmitter. After receiving the data on a VHF receiver, they are routed directly to the RS232 input connector on the PC. Thereby the advantage of direct communication between the transmitter and the PC can be utilized. Expensive analog equipment is avoided and display of the signals on the PC screen as well as signal analysis can be performed. The system has been tested and was found to be stable and highly reliable.  相似文献

Fluorosulfonylbenzoyl 5'-adenosine (FSBA) bound to one site in simian virus 40 large T antigen (T) and covalently modified greater than 95% of the molecules in a complete reaction. This analog for ATP specifically cross-links to the Mg-phosphate pocket in ATP-binding sites. Cyanogen bromide cleavage and tryptic digestion of [14C]FSBA-labeled protein, paired with T-specific monoclonal antibody analyses, were used to map the site in T to a tryptic peptide just C terminal to the PAb204 epitope. The location of the FSBA linkage was consistent with the predicted tertiary structure of the ATP-binding region in T described previously (M. K. Bradley, T. F. Smith, R. H. Lathrop, D. M. Livingston, and T. A. Webster, Proc. Natl. Acad. Sci. USA 84:4026-4030, 1987). Binding of FSBA to T was cooperative, implying an interaction between two binding sites. This could occur if the protein formed a dimer, and it is known that the ATPase activity is associated with a dimeric T. Most interesting was the activation of the ATPase when up to 50% of T was bound by the analog. The effect was also produced by preincubation with millimolar concentrations of ATP or the nonhydrolyzable analog gamma beta-methylene 5'-adenosine diphosphate at elevated temperatures. When greater than 50% of T was modified by FSBA, the ATPase was inhibited as the analog cross-linked to the second, previously activated, binding site. These data support a dual function for the one ATP-binding site in T as both regulatory and catalytic.  相似文献

In this work we have used the extension of the Gibbs ensemble simulation technique to inhomogeneous fluids [Panagiotopoulos, A.Z. (1987) "Adsorption and capillary condensation of fluid in cylindrical pores by Monte Carlo simulation in the Gibbs ensemble", Mol. Phys. , 62 (3), 701-719], which has been applied to adsorption phenomena of confined fluids. Fluid molecules are described by spherical particles interacting via a square-well potential. The fluid is confined in two types of walls: symmetrical (two hard walls) and non-symmetrical (one square-well wall and one hard wall). In order to analyze the behavior of the confined fluid by varying the potential parameters, we evaluated the bulk and confined densities, the internal energies and the density profiles for different supercritical temperatures. A variety of adsorption profiles can be obtained by using this model. The simulation data reported here complements the available simulation data for this system and can be useful in the development of inhomogeneous fluid theories. Since the square-well parameters can be related to real molecules this system can also be used to understand real adsorption systems.  相似文献

There is increasing evidence for a major and critical involvement of lipids in signal transduction and cellular trafficking, and this has motivated large-scale studies on lipid pathways. The Lipid Metabolites and Pathways Strategy consortium is actively investigating lipid metabolism in mammalian cells and has made available time-course data on various lipids in response to treatment with KDO2-lipid A (a lipopolysaccharide analog) of macrophage RAW 264.7 cells. The lipids known as eicosanoids play an important role in inflammation. We have reconstructed an integrated network of eicosanoid metabolism and signaling based on the KEGG pathway database and the literature and have developed a kinetic model. A matrix-based approach was used to estimate the rate constants from experimental data and these were further refined using generalized constrained nonlinear optimization. The resulting model fits the experimental data well for all species, and simulated enzyme activities were similar to their literature values. The quantitative model for eicosanoid metabolism that we have developed can be used to design experimental studies utilizing genetic and pharmacological perturbations to probe fluxes in lipid pathways.  相似文献

The biological properties of four chemically synthesized signal peptides were compared in mammalian (rabbit reticulocyte) and plant (wheat germ) cell-free protein secretion systems. The precursor-specific region of bovine pre-proparathyroid hormone (preproPTH), [D-Tyr-(+1)]preproPTH-(-29-+1)amide, and a sulfur-free analog, [Nle-(-25), Nle-(-21), Nle-(-18), Ala-(-14), D-Tyr-(+1)]preproPTH-(-29-+1)amide, inhibit the processing of an unrelated precursor protein (pre-prolactin) to its mature secreted form (prolactin) in the mammalian system. In the plant system supplemented with signal recognition particle, the signal peptides arrest translation of both secretory (preprolactin) and cytoplasmic (globin) proteins. One analog, [Nle-(-25), Nle-(-21), Asp-(-18), Ala-(-14), D-Tyr-(+1)]preproPTH-(-29-+1)amide, inhibits preprotein processing in the mammalian system but fails to induce translation arrest in the plant system. A truncated peptide, [N alpha-AcLeu-(-17), Ala-(-14), D-Tyr-(+1)]preproPTH-(-17-+1)amide, lacking the N-terminal (positively charged) region and a portion of the hydrophobic core region, is inactive in both systems. These studies demonstrate that the chemically synthesized signal region of a precursor protein interacts directly with signal recognition particle and functionally mimics the proposed properties of a native signal sequence linked to a nascent protein as it emerges from the ribosome during biosynthesis, and an analog of the signal peptide reveals fundamental differences between the components involved in the protein secretion apparatus in mammals and plants.  相似文献

Quantifying mechanical output is fundamental to understanding metabolism that fuels muscle contraction and more recent attempts to understand signal transduction and gene regulation. The latter requires long-term application of exercise protocols that result in large amounts of data on muscle performance. The purpose of this study was to develop software for automated quantification of skeletal muscle contractions. An in situ mouse sciatic nerve stimulation model was used to produce contractions over a broad range of frequencies and recorded as both digital and analog signals using a PC analog to digital converter board and chart recorder, respectively. Spectral analysis of the noise components formed the basis for designing a smoothing Chebyshev filter. Algorithms implemented in custom software identified twitches and estimated baseline levels from the smoothed signal. The time to peak force, peak force, tension-time integral, and half-relaxation time were determined for each twitch after baseline correction. The automated results were compared to those obtained from manual measurements of the analog signal. Bland–Altman analysis of the parameters computed from digital signals compared with the corresponding measurements by manual planometry demonstrates the agreement of the digital processing algorithm with planometry over a wide range of twitch characteristics. This program may also be used to study the mechanics of other preparations from isolated muscles, human proximal limb performance, and other digital physiologic signals. Adaptation of the filter function is required to apply the analysis to another experimental apparatus with differing noise characteristics. Phase-lifetime spectroscopy has been recently used to obtain kinetic information on biological photocycles. A simple, general method is presented for deriving the amplitude response function for light-driven processes. These amplitude response functions may be used to analyze the experimental data obtained when driving the photosystem with a mechanically chopped, actinic light source. This analysis allows a comparison of kinetic parameters obtained from modulation methods with those obtained by flash techniques. Typically the experimental data consist of the signal amplitude measured at several chopping frequencies of the actinic light. These amplitude dispersion curves will be dependent on the harmonic sensitivity of the phase-sensitive detector used to measure the signal. This harmonic sensitivity is taken into account by performing a Fourier decomposition on the amplitude response function of the system and weighting each harmonic in a fashion appropriate for the specific amplifier under consideration. The resulting response function obtained for two commonly used amplifiers is derived. In addition to simple photocycles, the analysis of photocycle-coupled processes is also considered. This second relaxation process, which is coupled to a photocycle process, could represent the chemiosmotic coupling of a light-driven ion pump to a second ion transport protein. Conditions are established in which the kinetics of the second process can be resolved from the photocycle process.  相似文献

A FORTRAN IV program is described, which may be run interactively or in batch and which allows a user to obtain the frequency response amplitude ratio and phase resulting from the linear analysis of an eye movement system using sine wave stimuli. The response (eye position) signal may contain components contributed by the saccadic eye movements. The program can digitize analog signals and store data on a magnetic tape. With the aid of digital filters, the program can detect saccades without requiring any input parameters from the user. The program interpolates the saccade interval using a method of least square curve fitting with a sine wave. The interpolation is relatively noise immune and works well regardless of the stimulus frequencies and the width of a saccade interval. Moreover, the program can handle long duration of signals such as 90 min of data which covers about 5 cycles of a 0.001 Hz sine wave signal. Sample runs for the cases of 0.001 and 0.1 Hz are given. The resident driver and the overlayable segments of the program have been implemented on a DEC (Digital Equipment Corp.) LAB-11 minicomputer (PDP 11/20).  相似文献

ObjectiveThe present study aims to simulate an alarm system for online detecting normal electrocardiogram (ECG) signals from abnormal ECG so that an individual's heart condition can be accurately and quickly monitored at any moment, and any possible serious dangers can be prevented.Materials and methodsFirst, the data from Physionet database were used to analyze the ECG signal. The data were collected equally from both males and females, and the data length varied between several seconds to several minutes. The heart rate variability (HRV) signal, which reflects heart fluctuations in different time intervals, was used due to the low spatial accuracy of ECG signal and its time constraint, as well as the similarity of this signal with the normal signal in some diseases. In this study, the proposed algorithm provided a return map as well as extracted nonlinear features of the HRV signal, in addition to the application of the statistical characteristics of the signal. Then, artificial neural networks were used in the field of ECG signal processing such as multilayer perceptron (MLP) and support vector machine (SVM), as well as optimal features, to categorize normal signals from abnormal ones.ResultsIn this paper, the area under the curve (AUC) of the ROC was used to determine the performance level of introduced classifiers. The results of simulation in MATLAB medium showed that AUC for MLP and SVM neural networks was 89.3% and 94.7%, respectively. Also, the results of the proposed method indicated that the more nonlinear features extracted from the ECG signal could classify normal signals from the patient.ConclusionThe ECG signal representing the electrical activity of the heart at different time intervals involves some important information. The signal is considered as one of the common tools used by physicians to diagnose various cardiovascular diseases, but unfortunately the proper diagnosis of disease in many cases is accompanied by an error due to limited time accuracy and hiding some important information related to this signal from the physicians' vision leading to the risks of irreparable harm for patients. Based on the results, designing the proposed alarm system can help physicians with higher speed and accuracy in the field of diagnosing normal people from patients and can be used as a complementary system in hospitals.  相似文献

A homogeneous time-resolved fluorescence (HTRF)-based binding assay has been established to measure the binding of the histone methyltransferase (HMT) G9a to its inhibitor CJP702 (a biotin analog of the known peptide-pocket inhibitor, BIX-01294). This assay was used to characterize G9a inhibitors. As expected, the peptide-pocket inhibitors decreased the G9a-CJP702 binding signal in a concentration-dependent manner. In contrast, the S-adenosyl-L-methionine (SAM)-pocket compounds, SAM and sinefungin, significantly increased the G9a-CJP702 binding signal, whereas S-adenosyl-L-homocysteine (SAH) showed minimal effect. Enzyme kinetic studies showed that CJP702 is an uncompetitive inhibitor (vs. SAM) that has a strong preference for the E:SAM form of the enzyme. Other data presented suggest that the SAM/sinefungin-induced increase in the HTRF signal is secondary to an increased E:SAM or E:sinefungin concentration. Thus, the G9a-CJP702 binding assay not only can be used to characterize the peptide-pocket inhibitors but also can detect the subtle conformational differences induced by the binding of different SAM-pocket compounds. To our knowledge, this is the first demonstration of using an uncompetitive inhibitor as a probe to monitor the conformational change induced by compound binding with an HTRF assay.  相似文献
