Allozyme electrophoresis was used to assess temporal genetic variation in three successive generations of the Mediterranean killifish, Aphanius fasciatus. Samplings were carried out in 1995, 1996 and 1997 in a brackish-water habitat at Elba Island, Italy and a total of 212 specimens were collected. The five loci for which polymorphism has been detected in a previous study were assayed. Mean expected heterozygosity values [H=0.397 (SE 0.077), H=0.336 (SE 0.092) and H=0.313 (SE 0.092) in 1995, 1996 and 1997, respectively] were not significantly different by ANOVA test. Deviations from Hardy–Weinberg equilibrium were minimal, with only one out of the 15 probability tests showing a significant departure from the equilibrium; whereas genotypic linkage disequilibrium was not detected. Values of Nei's genetic distance were lower than 0.04. Temporal genetic variation in the A. fasciatus population at Elba Island was observed, with F-statistics indicating significant genetic divergence among samples (=0.035, SE 0.027, p<0.001). Genetic drift acting on two loci (GPD-1 * and LDH-3 *) is presumably the main force determining the temporal genetic heterogeneity observed; however, the occurrence of selection on individual loci and/or sampling error cannot be excluded. The observed allelic variation among generations in a single population of A. fasciatus is much less than levels observed among geographically discrete samples in previous studies.  相似文献

Second chromosome inversions and genotypic frequencies at seven allozyme loci were determined in a natural population of the cactophilic species Drosophila buzzatii that uses as breeding sites the necrotic cladodes of the prickly pear Opuntia quimilo and the rotting stems of cardón, Trichocereus terschekii. Different processes govern the evolutionary fate of inversion and allozyme polymorphisms. A pattern of heterotic balance for inversions seems to be acting uniformly in each breeding site and could depend on different regimes of density‐dependent selection within cactus hosts. Patterns of variation of allozymes revealed significant heterogeneity in allele frequencies for Esterase‐1 (Est‐1) among O. quimilo rots and Aldehyde oxidase (Aldox) and Xanthine dehydrogenase (Xdh) among T. terschekii substrates and showed gene‐cactus effects only for Esterase‐2 (Est‐2). Consistent and significant excesses of homozygotes were detected at both the within‐rot and in the total population levels that could be accounted for by diversifying selection among individual breeding sites.  相似文献

Size-related sexual selection (SRSS) was examined on four traits (thorax and wing length and head and face width) inDrosophila buzzatii, by scoring male copulatory status in two mass-mating experiments. Using axenic females, experiment 1 was carried out with axenic males, and experiment 2 with yeast-supplemented males. While there was no indication of SRSS in experiment 1, such selection was substantial in yeast-supplemented males, which transmitted yeasts to mating females. Multivariate analyses of selection indicated that face width is the measured trait on which directional SRSS essentially acted in yeast-supplemented males, resulting in indirect selection on body size. Because this selection was affected by yeast diet in males, its possible interaction with the yeast transmission from males to females during the courtship is discussed.  相似文献

Drosophila antonietae is an endemic South American cactophilic species that uses Cereus hildmaniannus rotting cladodes as breeding sites. We assessed temporal and spatial intrapopulational allozyme variation of two natural populations. Our results suggest that environmental variation (rain precipitation) is probably influencing allozyme temporal variation. Moreover, it seems that D. antonietae does not have intrapopulation structure and has N ev (variance effective size) 83 and N ec (number of adult flies that colonize each rotting cladode) = 21. The deficiency of heterozygotes found must be due to null alleles, a temporal Wahlund effect, or selection against heterozygotes. Assortative mating and inbreeding are discarded. This is the first report on allozyme variation in D. antonietae. It gives some insight on intrapopulational genetics through space and time for this species. This is important to understand its general genetic variability and will be essential to future works on the natural history and evolution of this species.  相似文献

In this study, samples of Ancherythroculter nigrocauda, an endemic fish in the upper Yangtze River, were collected above and below dams in the Longxi River, a tributary of the upper Yangtze River, China, to investigate the genetic impacts of dams. The mitochondrial cytochrome b gene (cyt b) and 13 microsatellite (SSR) loci were used to analyze whether dams have resulted in loss of genetic diversity of the two fragmented populations or caused genetic differentiation between them. The results showed that the haplotype diversity (0.488; 0.486), nucleotide diversity (0.084%; 0.082%) and average expected heterozygosity (0.652; 0.676) of the two populations were all at a low level, and recent bottlenecks were detected. However, there was no genetic differentiation detected by the low genetic differentiation index (Fst, cyt b: ?0.1677, p = 0.99707; SSR: 0.00259, p = 0.81427). Besides, 11 pairs of half‐sibling relationship were found between the two populations indicating that there were individual movements and gene flow between them. This could be the larvae moving from upstream to downstream when water spilled over dams in flooding season. Therefore, our analysis showed that the dams have caused a loss of genetic diversity of the populations of A. nigrocauda in the Longxi River, blocked the active upstream movement but allowing passive downstream drift of larvae.  相似文献

Results of intrapopulation studies of sexual selection and genetic variation and covariation underlying elements of the sex comb of Drosophila bipectinata are presented. The magnitude of the sex comb, a sexual ornament, varies significantly among Australasian populations, motivating research into the evolutionary mechanisms responsible for its incipient diversification. The comb is composed of stout black teeth on the front legs of males arranged in three distinct segments: C1, C2, and C3. Significant sexual selection in field populations in northeastern Queensland, Australia, was detected for increasing C2 and body size, and simultaneously for reducing comb positional fluctuating asymmetry. In contrast, sexual selection was not detected for other comb segments, nor for sternopleural bristle number or symmetry. Selection intensities for C2 and comb positional fluctuating asymmetry were similar in magnitude, and although they were opposite in sign, values across twelve sampling dates, or selection episodes, were uncorrelated. Heritability estimates for C2 were high and significant across years, whereas heritability estimates for comb positional asymmetry were small, and generally nonsignificant. The major sex comb segments (C1 and C2) were significantly and positively correlated genetically, indicating the potential for correlated evolution of these components of the comb under sexual selection. The original finding of a significant positive genetic correlation between the magnitude of this sex trait and its positional asymmetry indicates that the counteracting and independent selection pressures detected could contribute to the maintenance of genetic variation sustaining sexual selection. The study documents the simultaneous presence of sexual selection in nature and of heritable genetic variation underlying expression of the sex comb, fundamental conditions necessary for its adaptive diversification. Drosophila bipectinata may be a valuable model for studies of adaptive diversification and incipient speciation by sexual selection.  相似文献

Using wild-reared flies, we examined sexual selection on five phenotypic traits (thorax length, wing length, wing width, head width, and face width) inDrosophila buzzatii, by scoring copulatory status in nine mass mating cages. Only male face width was identified as a direct target of sexual selection in an analysis of selection gradient, while indirect selection was present on all other studied traits, as expected from their correlations with face width. In contrast to males, there was no indication of selection in females. Nor was there evidence of assortative mating. The suggested direct selection on face width seems to take place during licking behavior of the courtship and might be related to courtship feeding. This study suggests that courtship success gives rise to indirect selection on body size.  相似文献

The Corsica–Sardinia archipelago is a hotspot of Mediterranean biodiversity. Although tempo and mode of arrival of species to this archipelago and phylogenetic relationships with continental species have been investigated in many taxa, very little is known about the current genetic structure and evolutionary history subsequent to arrival. In the present study, we investigated genetic variation within and among populations of the Tyrrhenian treefrog Hyla sarda, a species endemic to the Corsica–Sardinia microplate and the surrounding islands, by means of allozyme electrophoresis. Low genetic divergence (mean Dnei = 0.01) and no appreciable differences in the levels and distribution of genetic variability (HE: 0.06–0.09) were observed among all but one populations (Elba). Historical demographic and isolation‐by‐distance analyses indicated that this diffused genetic homogeneity could be the result of recent demographic expansion. Along with paleoenvironmental data, such expansion could have occurred during the last glacial phase, when wide and persistent land bridges connected the main islands and a widening of lowland areas occurred. This scenario is unprecedented among Corsica–Sardinia species. Together with the lack of concordance even among the few previously studied species, this suggests either that species had largely independent responses to paleoenvironmental changes, or that most of the history of assembly of the Corsica–Sardinia biota is yet to be written. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 159–167.  相似文献

Gaps in the large-scale distribution of the tephritid fly Urophora cardui in Europe have been explained as the results of an ongoing re-immigration from Pleistocene refugia due to a very low dispersal capacity. Following evidence of a much greater dispersal capacity of U. cardui than previously assumed, the pattern of genetic differentiation of 41 populations from 16 European regions was studied using allozyme electrophoresis. In these analyses 18 enzyme systems were scored consistently providing 27 alleles. Allozyme variation indicated high gene flow and low levels of genetic differentiation within and between sampling regions as well as in recently colonized areas. No geographical pattern of heterozygosity or allozyme differentiation could be found matching the previously suggested recent immigration pattern. An observed south-north gradient in allozyme frequencies was interpreted as a geographical cline due to environmental factors. The results corroborate evidence from more recent studies that U. cardui is a highly mobile species which is likely to have repeatedly colonized some suboptimal European regions since the Pleistocene after retreats during 'little ice ages'. Patterns resulting from postglacial immigration processes are likely to have been long wiped out through high exchange rates.  相似文献

Sexual selection and sexual signaling have been prominent topics in recent behavioral studies, but limited data have led to controversy regarding these topics. For example, the Hawaiian Drosophila are often cited as examples in which female choice has resulted in the evolution of elaborate male courtship signals, but relatively few data exist to test these claims adequately. We studied D. grimshawi, a lek-forming Hawaiian Drosophila, to determine whether there was evidence for female choice without male competition and to elucidate the possible cues females use to discriminate. Male mating success was found to be nonrandom and males that courted females intensely and deposited many pheromone-containing streaks on the substrate were the most successful. Hence, multiple cues seem to be involved in male mating success in this species. Some males performed only one display, however, and may represent an alternate male mating tactic. The protein content of the adult male diet significantly influenced the level of pheromone streak deposition, and thus, foraging environment may affect the outcome of sexual selection.  相似文献

The Drosophila melanogaster species complex consists of four species: D. melanogaster, D. simulans, D. sechellia and D. mauritiana. To identify these closely related species, researchers often examine the male genitalia, especially species‐specific shapes of the posterior process, as the most reliable and easily observable character. However, compared to genetic aspects, the evolutionary significance of the posterior process and other genital parts remains largely unexplained. By comparing genital coupling among these species, we revealed that the posterior processes, which are hidden under the female abdominal tergite VII when genital coupling is established, mesh with different parts of the intersegmental membrane between the tergite VIII and the oviscapts and that this membrane region broadens in a species‐specific manner. Furthermore, in D. simulans and D. sechellia, this membrane region is likely to incur wounds from the sharply pointed tip of the posterior process. On the basis of the use and functions of these and other genital parts, we discuss possible evolutionary forces underlying the diversification of genitalia in this group.  相似文献

Islands are generally colonized by few individuals which could lead to a founder effect causing loss of genetic diversity and rapid divergence by strong genetic drift. Insular conditions can also induce new selective pressures on populations. Here, we investigated the extent of genetic differentiation within a white‐tailed deer (Odocoileus virginianus) population introduced on an island and its differentiation with its source mainland population. In response to their novel environmental conditions, introduced deer changed phenotypically from mainland individuals, therefore we investigated the genetic bases of the morphological differentiation. The study was conducted on Anticosti Island (Québec, Canada) where 220 individuals were introduced 120 years ago, resulting in a population size over 160,000 individuals. We used genotyping‐by‐sequencing (GBS) to generate 8,518 filtered high‐quality SNPs and compared patterns of genetic diversity and differentiation between the continental and Anticosti Island populations. Clustering analyses indicated a single panmictic island population and no sign of isolation by distance. Our results revealed a weak, albeit highly significant, genetic differentiation between the Anticosti Island population and its source population (mean FST = 0.005), which allowed a population assignment success of 93%. Also, the high genetic diversity maintained in the introduced population supports the absence of a strong founder effect due to the large number of founders followed by rapid population growth. We further used a polygenic approach to assess the genetic bases of the divergent phenotypical traits between insular and continental populations. We found loci related to muscular function and lipid metabolism, which suggested that these could be involved in local adaptation on Anticosti Island. We discuss these results in a harvest management context.  相似文献

A 2012–13 survey on Penang Island, Malaysia, revealed the existence of both Drosophila ananassae and Drosophila parapallidosa, the latter of which carries chromosomes Y and 4 from D. ananassae and thus is of hybrid origin. We collected the flies again from the same location in 2018. The hybrid population remained present, which suggests that the D. parapallidosa of hybrid origin does not represent a mere transient population but is stable. Why do these two species coexist irrespective of gene flow? We realized that body size is generally larger in D. ananassae than in D. parapallidosa, which constitutes a new character with which to discriminate these species; previously the number of sex comb teeth was the only diagnostic trait. Character displacement was not detected, however, for those traits. We crossed these two species, which resulted in offspring that had an altered genomic constitution. The body size of D. ananassae was dominant, and the presence of chromosomes Y and 4 did not have a significant effect on body size. By contrast, the presence of chromosome 4 from D. ananassae significantly affected the number of sex comb teeth. Even flies having a genomic constitution similar to that of the Penang D. parapallidosa exhibited a number of sex comb teeth that was intermediate between the two species. We propose that the D. parapallidosa sex comb character underwent selection during evolution of the Penang Island population. Reproductive interference between the species, presumably caused by signal jamming, was detected.  相似文献

To investigate the relative importance of homogenizing factors, such as gene flow, and diversifying factors, such as drift, genetic variation in pikeperch ( Sander lucioperca ) in two Fennoscandian regions (North and South) was analysed with microsatellites. Allelic richness and the degree of differentiation were significantly higher in the North ( F ST= 0·20) than in the South ( F ST= 0·064). In northern areas, assignments of genotypes were almost exclusively to the population of origin, but in southern areas, the proportion of correct assignments was significantly lower. Most samples exhibited significant heterozygote deficits, and the level of relatedness was higher than expected from randomness. These combined results suggest that there has been more gene flow between populations in southern areas than in northern areas, where the importance of genetic drift has been greater. Effective population sizes were small ( c. 100) and did not differ between areas. The effect of a common history appears minor, and thus processes such as genetic drift and gene flow have been more influential in shaping the patterns of genetic diversity in this species.  相似文献

A new polymerase chain reaction-based screening method for microsatellites is presented. Using this method, we isolated 12 microsatellite loci from Drosophila buzzatii, two of which were X-linked. We applied the other 10 microsatellite loci to the analysis of genetic variation in five natural populations of D. buzzatii. Two populations were from the species' original distribution in Argentina, whereas the other three were from Europe (two) and Australia that were colonized 200 and 65 years ago, respectively. Allelic variation was much larger in the original populations than in the colonizing ones and there was a tendency to decreased heterozygosity in the colonizing populations. We used three different statistical procedures for detecting population bottlenecks. All procedures suggested that the low variability in the populations in the Old World was not the result of the recent population decline, but was due to a founder effect followed by a population expansion. In fact, one procedure which detects population expansions and declines based on the genealogical history of microsatellite data suggested that an expansion had taken place in all the colonized populations.  相似文献

1. The recent arrival and explosive spread of the zebra mussel, Dreissena polymorpha (Pallas), in Ireland provided a rare opportunity to study the population genetics of an invasive species.
2. Eight polymorphic allozyme loci ( ACO-1, ACO-2 , EST-D, GPI, IDH-2, MDH, OPDH and PGM ) were used to investigate genetic diversity and population structure in five Irish populations, and the results were compared with those from a previous microsatellite study on the same samples.
3. The mean number of alleles per locus (2.7 ± 0.1) was similar to the mean for the same loci in European populations, suggesting that Irish founder populations were large and/or multiple colonization events took place after foundation. A deficiency of heterozygotes was observed in all populations, but was uneven across loci.
4. Pairwise comparisons, using Fisher's exact tests and F ST values, revealed significant genetic differentiation among populations. The overall multilocus F ST estimate was 0.118 ± 0.045, which contrasted with an estimate of 0.015 ± 0.007 from five microsatellite loci on the same samples in a previous study.
5. Assuming that microsatellites can be used as a neutral baseline, the discordant results from allozymes and microsatellites suggest that selection may be acting on some allozyme loci, specifically ACO-1, ACO-2 , IDH-2 and MDH, which contributed most to the significant differentiation between samples.  相似文献

Adult resistance to DDT conferred by various regions of the second chromosome of a DDT-resistant strain of Drosophila melanogaster (Meigen) was analyzed using the quantitative genetic method of Thoday and a topical test procedure. Resistance was localized in a small region near and to the left of cn. Both the genetic method and the topical test technique were effective. Results are consistent with the idea that the number of loci responsible for insecticide resistance in D. melanogaster is not large.  相似文献

雌性对雄性表饰的偏好性有利于性别选择。目前尚不清楚这一偏好性是否只限于雄性表饰或这一偏好性实际上是源于影响后代适合度的基因。对于雄性可直接有利于雌性或其后代适合度的交配系统而言,答案是肯定的--雌性偏好于与对气候胁迫具有更强生理抗性的雄性交配。对果蝇Drosophila biarmipes 的室内研究已经证明了求偶过程中翅斑的作用,但是其生态学意义仍然不清楚。我们检验了有翅斑与无翅斑雄性果蝇D. biarmipes 及雌性偏好的雄性所产生的后代对环境胁迫的抗性是否不同。结果表明:在干燥或冷胁迫条件下,有翅斑的雄性果蝇比无翅斑的雄性果蝇的交配成功率明显要高。相反,在高湿条件下,无翅斑雄性果蝇的交配频率更高。我们也发现在较为干旱的条件下,与有翅斑雄性交配的雌性果蝇的生殖力以及所得后代从卵至成虫的存活率更高。我们的结果与优良基因性选择假说一致,说明交配选择能给雌性带来间接好处。这是对热带物种D. biarmipes翅色二型性生态学意义的首次报道。  相似文献
