Understanding and managing ischemic stroke   总被引:21,自引:0,他引:21
Transient or permanent focal brain injury following acute thromboembolic occlusion develops from a complex cascade of pathophysiological events. The processes of excitotoxicity, peri-infarct depolarisation, inflammation, and apoptosis within the ischemic penumbra are proposed. While the translation of therapeutic agents from the animal models to human clinical trials have been disappointing, there remains an atmosphere of optimism as a result of the development of new diagnostic and therapeutic approaches, which include physiological, as opposed to pharmacological, intervention. This article provides an insight into the understanding of cerebral ischemia, together with current and future treatment strategies.  相似文献

Fisheries enhancements are a set of management approaches involving the use of aquaculture technologies to enhance or restore fisheries in natural ecosystems. Enhancements are widely used in inland and coastal fisheries, but have received limited attention from fisheries scientists. This paper sets out 10 reasons why fisheries scientists should care about understanding and managing enhancements. (1) Enhancements happen, driven mostly by resource users and managers rather than scientists. (2) Enhancements create complex fisheries systems that encompass and integrate everything fisheries stakeholders can practically manage. (3) Enhancements emerge in fisheries where the scope for technical and governance control is high, and they synergistically reinforce both. (4) Successful enhancements expand management options and achievable outcomes. (5) Many enhancements fail or do ecological harm but persist regardless. (6) Effective science engagement is crucial to developing beneficial enhancements and preventing harmful ones. (7) Good scientific guidance is available to aid development or reform of enhancements but is not widely applied. (8) Enhancement research advances, integrates and unifies the fisheries sciences. (9) Enhancements provide unique opportunities for learning about natural fish populations and fisheries. (10) Needs, opportunities and incentives for enhancements are bound to increase.  相似文献

Race was an important topic to the physical anthropologists of 1918, but their views were not monolithic. Multiple perspectives on race are expressed in the first volume of the AJPA, which encompass biological determinism and assumptions about evolutionary processes underlying the race concept. Most importantly, many of the significant alternative approaches to the study of human variation were already expressed in 1918. This paper examines race from the different perspectives of three key contributions to the first volume of the AJPA: papers from Hrdlička, Hooton, and Boas. The meaning of race derived from this work is then discussed. Despite new understandings gained through the neo-Darwinian synthesis and the growth of genetics, the fundamentals of the modern discussions of race were already planted in 1918. Until recently, it was impracticable to identify the genes that are responsible for variation in continuous traits, or to directly observe the effects of their different alleles. Now, the abundance of genetic markers has made it possible to identify quantitative trait loci (QTL)--the regions of a chromosome or, ideally, individual sequence variants that are responsible for trait variation. What kind of QTL do we expect to find and what can our observations of QTL tell us about how organisms evolve? The key to understanding the evolutionary significance of QTL is to understand the nature of inherited variation, not in the immediate mechanistic sense of how genes influence phenotype, but, rather, to know what evolutionary forces maintain genetic variability.  相似文献

Animal cognition experiments frequently reveal striking individual variation but rarely consider its causes and largely ignore its potential consequences. Studies often focus on a subset of high-performing subjects, sometimes viewing evidence from a single individual as sufficient to demonstrate the cognitive capacity of a species. We argue that the emphasis on demonstrating species-level cognitive capacities detracts from the value of individual variation in understanding cognitive development and evolution. We consider developmental and evolutionary interpretations of individual variation and use meta-analyses of data from published studies to examine predictors of individual performance. We show that reliance on small sample sizes precludes robust conclusions about individual abilities as well as inter- and intraspecific differences. We advocate standardization of experimental protocols and pooling of data between laboratories to improve statistical rigour. Our analyses show that cognitive performance is influenced by age, sex, rearing conditions and previous experience. These effects limit the validity of comparative analyses unless developmental histories are taken into account, and complicate attempts to understand how cognitive traits are expressed and selected under natural conditions. Further understanding of cognitive evolution requires efforts to elucidate the heritability of cognitive traits and establish whether elevated cognitive performance confers fitness advantages in nature.  相似文献

Interannual variation of terrestrial carbon cycle: Issues and perspectives   总被引:1,自引:0,他引:1
With accumulation of carbon cycle observations and model developments over the past decades, exploring interannual variation (IAV) of terrestrial carbon cycle offers the opportunity to better understand climate–carbon cycle relationships. However, despite growing research interest, uncertainties remain on some fundamental issues, such as the contributions of different regions, constituent fluxes and climatic factors to carbon cycle IAV. Here we overviewed the literature on carbon cycle IAV about current understanding of these issues. Observations and models of the carbon cycle unanimously show the dominance of tropical land ecosystems to the signal of global carbon cycle IAV, where tropical semiarid ecosystems contribute as much as the combination of all other tropical ecosystems. Vegetation photosynthesis contributes more than ecosystem respiration to IAV of the global net land carbon flux, but large uncertainties remain on the contribution of fires and other disturbance fluxes. Climatic variations are the major drivers to the IAV of net land carbon flux. Although debate remains on whether the dominant driver is temperature or moisture variability, their interaction, that is, the dependence of carbon cycle sensitivity to temperature on moisture conditions, is emerging as key regulators of the carbon cycle IAV. On timescales from the interannual to the centennial, global carbon cycle variability will be increasingly contributed by northern land ecosystems and oceans. Therefore, both improving Earth system models (ESMs) with the progressive understanding on the fast processes manifested at interannual timescale and expanding carbon cycle observations at broader spatial and longer temporal scales are critical to better prediction on evolution of the carbon–climate system.  相似文献

Having evolved as a grazing animal, a horse's digestive physiology is characterized by rapid gastric transit, a rapid but intense enzymatic digestion along the small intestine, and a long and intense microbial fermentation in the large intestine. The process of understanding and describing feed degradation mechanisms in the equine digestive system in general, and in the hindgut ecosystem in particular, is essential. Regardless of its importance for the nutritional status of the host, the significance of the cecum-colon ecosystem has not yet been fully understood, and few reports have focused deeply on the contribution of the hindgut microbial population to the nitrogen and energy requirements of the horse. Compared to ruminal activity, very little is known about hindgut ecosystem activity in the horse. Information concerning the metabolism of this microbial population and its requirements is lacking. The use of internal bacterial markers for quantifying microbial outflow in ruminants is widely reported. These techniques can be applied to cecum-colon microbial quantification, contributing to a better characterization of this ecosystem. It is likely wrong to believe that the optimization strategy in the hindgut is similar to what happens in the rumen - that is, to maximize microbial growth and, therefore, fermentation. If we consider the type of substrate that, in normal conditions, arrives in the hindgut, we can expect it to be nitrogen limiting, providing limited nitrogen-based substrates for microbial fermentation. In this review paper, we intend to gather existing information on the equine ecosystem and to provide future perspectives of research.  相似文献

Quantitative genetics, or the genetics of complex traits, is the study of those characters which are not affected by the action of just a few major genes. Its basis is in statistical models and methodology, albeit based on many strong assumptions. While these are formally unrealistic, methods work. Analyses using dense molecular markers are greatly increasing information about the architecture of these traits, but while some genes of large effect are found, even many dozens of genes do not explain all the variation. Hence, new methods of prediction of merit in breeding programmes are again based on essentially numerical methods, but incorporating genomic information. Long-term selection responses are revealed in laboratory selection experiments, and prospects for continued genetic improvement are high. There is extensive genetic variation in natural populations, but better estimates of covariances among multiple traits and their relation to fitness are needed. Methods based on summary statistics and predictions rather than at the individual gene level seem likely to prevail for some time yet.  相似文献

Over the past century researchers have identified normal genetic variation and studied that variation in diverse human populations to determine the amounts and distributions of that variation. That information is being used to develop an understanding of the demographic histories of the different populations and the species as a whole, among other studies. With the advent of DNA-based markers in the last quarter century, these studies have accelerated. One of the challenges for the next century is to understand that variation. One component of that understanding will be population genetics. We present here examples of many of the ways these new data can be analyzed from a population perspective using results from our laboratory on multiple individual DNA-based polymorphisms, many clustered in haplotypes, studied in multiple populations representing all major geographic regions of the world. These data support an "out of Africa" hypothesis for human dispersal around the world and begin to refine the understanding of population structures and genetic relationships. We are also developing baseline information against which we can compare findings at different loci to aid in the identification of loci subject, now and in the past, to selection (directional or balancing). We do not yet have a comprehensive understanding of the extensive variation in the human genome, but some of that understanding is coming from population genetics.  相似文献

Human height is a highly variable trait, both within and between populations, has a high heritability, and influences the manner in which people behave and are treated in society. Although we know much about human height, this information has rarely been brought together in a comprehensive, systematic fashion. Here, we present a synthetic review of the literature on human height from an explicit evolutionary perspective, addressing its phylogenetic history, development, and environmental and genetic influences on growth and stature. In addition to presenting evidence to suggest the past action of natural selection on human height, we also assess the evidence that natural and sexual selection continues to act on height in contemporary populations. Although there is clear evidence to suggest that selection acts on height, mainly through life‐history processes but perhaps also directly, it is also apparent that methodological factors reduce the confidence with which such inferences can be drawn, and there remain surprising gaps in our knowledge. The inability to draw firm conclusions about the adaptiveness of such a highly visible and easily measured trait suggests we should show an appropriate degree of caution when dealing with other human traits in evolutionary perspective.  相似文献

An area of research that has been recently gaining attention is the relationship between cancer stem cell (CSC) biology and chemo-resistance in colon cancer patients. It is well recognized that tumor initiation, growth, invasion and metastasis are promoted by CSCs. An important reason for the widespread interest in the CSC model is that it can comprehensibly explain essential and poorly understood clinical events, such as therapy resistance, minimal residual disease, and tumor recurrence. This review discusses the recent advances in colon cancer stem cell research, the genes responsible for CSC chemoresistance, and new therapies against CSCs.  相似文献

The genetic management of captive populations to conserve genetic variation is currently based on analyses of individual pedigrees to infer inbreeding and kinship coefficients and values of individuals as breeders. Such analyses require that individual pedigrees are known and individual pairing (mating) can be controlled. Many species in captivity, however, breed in groups due to various reasons, such as space constraints and fertility considerations for species living naturally in social groups, and thus have no pedigrees available for the traditional genetic analyses and management. In the absence of individual pedigree, such group breeding populations can still be genetically monitored, evaluated and managed by suitable population genetics models using population level information (such as census data). This article presents a simple genetic model of group breeding populations to demonstrate how to estimate the genetic variation maintained within and among populations and to optimise management based on these estimates. A numerical example is provided to illustrate the use of the proposed model. Some issues relevant to group breeding, such as the development and robustness evaluation of the population genetics model appropriate for a particular species under specific management and recording systems and the genetic monitoring with markers, are also briefly discussed.  相似文献
