Our society is highly depending on petroleum for its activities. About 90% is used as an energy source for transportation and for generation of heat and electricity and the remaining as feedstocks in the chemical industry. However, petroleum is a finite source as well as causing several environmental problems such as rising carbon dioxide levels in the atmosphere. Petroleum therefore needs to be replaced by alternative and sustainable sources. Plant oils and oleochemicals derived from them represent such alternative sources, which can deliver a substantial part of what is needed to replace the petroleum used as feedstocks.  相似文献

Light control over enzyme function represents a novel and exciting field of biocatalysis research. Blue-light photoreceptors of the L ight, O xygen, V oltage (LOV) family have recently been investigated for their applicability as photoactive switches. We discuss here the primary photochemical events leading to light activation of LOV domains as well as the proposed signal propagation mechanism to the respective effector domain. Furthermore, we describe the construction of LOV fusions to different effector domains, namely a dihydrofolate reductase from Escherichia coli and a lipase from Bacillus subtilis. Both fusion partners retained functionality, and alteration of enzyme activity by light was also demonstrated. Hence, it appears that fusion of LOV photoreceptors to functional enzyme target sites via appropriate linker structures may represent a straightforward strategy to design light controllable biocatalysts.  相似文献

ESC derivation, use and SCNT have raised many moral and ethical issues. In this opinion piece I have focused on the argument that morally less ambiguous alternatives to ESC derived from the ICM of blastocysts exist. These possibilities range from using multiple adult stem cell populations each of which is uniquely suited for a particular disease target or identifying adult ESC-like populations, using transdifferentiated ESC-like cells or alternate methods of deriving ESC. I suggest that while it is important to support such efforts, current results do not provide sufficient compelling data to allow one to stop the use of ESC and perhaps adult cells will never be a reliable alternative. All options need to be fully explored and decisions need to be made with scientific rigor and respect for each individual's moral compass.  相似文献

Loss of natural wetlands is a global phenomenon that has been a serious threat to the wildlife. A common practice is to construct artificial wetlands to compensate for the loss of natural wetlands. To test whether artificial wetlands as habitats for waterbirds are good alternatives to natural wetlands, we compared species richness, abundance, and seasonal dynamics of waterbird communities of natural (here tidelands) and artificial wetlands (here aquacultural ponds) on Chongming Island, China. Our results indicate that habitat preference of waterbirds showed seasonal difference: most of the shorebirds were found on tidelands in spring, whereas most of the natatorial birds were recorded in aquacultural ponds in winter. Waterbirds preferred the tidelands rather than aquacultural ponds in both spring and autumn, whereas they showed no preference for either the tidelands or the ponds in summer and winter. It is concluded that natural wetlands are better habitats for waterbirds than artificial wetlands on Chongming Island, while the artificial ones are also suitable habitats for waterbirds in winter. The waterbirds might use artificial wetlands only when natural wetlands are unavailable or of poor quality. An over-emphasis that artificial wetlands are suitable habitats for waterbirds might encourage land managers to convert natural wetlands into the artificial ones, resulting in considerable loss of bird diversity. Therefore, for the purpose of bird conservation, it would be a better practice to conserve natural wetlands rather than to construct artificial ones after destruction of natural wetlands.  相似文献

The purpose of this study was to contribute to filling the knowledge gap in public opinion and knowledge about forest and its certification in Japan, as well as to identify key elements and the possible role of public opinion within integrated bottom-up policies, bridging the sectors of forest, environment and energy. For the study 1930 questionnaires were disseminated in a small town in early 2007. Results from the statistical analysis indicated that forest was perceived as an ecosystem with a protective function against e.g. soil erosion or flooding, rather than a place that might serve for wood production and providing jobs. Forest certification and bioenergy from forest were identified as key elements for future integrated bottom-up policies that need to concentrate on facilitating the linkage between forestry and renewable energy as well as on promoting environmentally sound management and forest certification.  相似文献

Hundreds of studies have assessed variation in the degree to which people experience disgust toward substances associated with pathogens, but little is known about the mechanistic sources of this variation. The current investigation uses olfactory perception and threshold methods to test whether it is apparent at the cue-detection level, at the cue-interpretation level, or both. It further tests whether relations between disgust sensitivity and olfactory perception are specific to odors associated with pathogens. Two studies (N's = 119 and 160) of individuals sampled from a Dutch university each revealed that pathogen disgust sensitivity relates to valence perceptions of odors found in pathogen sources, but not to valence perceptions of odors not associated with pathogens, nor to intensity perceptions of odors of either type. Study 2, which also assessed olfactory thresholds via a three-alternative forced-choice staircase method, did not reveal a relation between pathogen disgust sensitivity and the ability to detect an odor associated with pathogens, nor an odor not associated with pathogens. In total, results are consistent with the idea that pathogen disgust sensitivity relates to how olfactory pathogen cues are interpreted after detection, but not necessarily to the ability to detect such cues.  相似文献

Ultrasound scanning is a noninvasive, accurate, and cost-effective method to create images of the female reproductive tract clinically and in research. Ultrasonography is particularly valuable for studying the dynamic relationships among mother, placenta, and fetus during pregnancy because this modality does not disturb the ongoing course of gestation. Importantly, the complex vascular changes in the mother induced by pregnancy and the vascular system generated to support placental function can be assessed quantitatively and functionally by ultrasonography. Many mouse models are available that address aspects of human placental function and dysfunction, but high-quality microultrasound technology suitable for use in pregnant mice has become widely available only recently. This technical advance now enables real-time recording of maternal–fetal interactions in pregnant rodents. The ability to perform microultrasonic analyses of parameters such as uterine arterial remodeling, hemodynamic changes, placental development, and fetal growth in mice now permits research that uses the same imaging platform as that for human patients. This capability will enhance the translation of information derived from rodent studies to the clinic.Abbreviations: gd, gestation day; IUGR, intrauterine growth restrictionHuman pregnancy is a complex, dynamic progress accompanied by both inspiring and amazing events. Clinically, disorders of pregnancy are among the least understood areas in health science because of ethical and cultural challenges and the involvement of 2 (or more) subjects (mother and fetus) rather than 1. Because continuation and successful conclusion of pregnancy typically are desired, advances in clinical and research investigations of human pregnancy are highly dependent upon technological advances in noninvasive imaging. The most common human gestational complications are preeclampsia (acute onset of hypertension during the second half of pregnancy), intrauterine growth restriction (IUGR), and diabetes. These pathologies are thought to share many risk factors, particularly an inadequate response of the maternal vasculature to the state of pregnancy.59 In all mammals, maternal vascular adaptation is essential to meet the requirements of blastocyst implantation and placental and fetal development to ensure a successful pregnancy. In species with hemochorial placentation (including human, rat, and mouse), endometrial decidualization is associated with maternal vascular remodeling at implantation sites, and the remodeling process has become a diagnostic and therapeutic target.39,47Ultrasonography was introduced in 1958 and has become the diagnostic tool of choice to study human gestational health,18 not only because this modality is noninvasive with very low risk but also because it provides real-time images with positional information.24 Ultrasonography is relatively inexpensive and portable, compared with other modalities such as MRI and computed tomography. Modern clinical ultrasonography usually is performed by using a pulse–echo approach with a brightness-mode (B-mode) output that can be augmented by using advanced functions such as color Doppler mapping, 3D or 4D imaging, and spectral measurements. Application of these ultrasound techniques has enabled exploration and understanding of human maternal–fetal relationships that previously had been virtually inaccessible. Most studies of the physiology and pathology of pregnancy in animal models still rely on postmortem approaches rather than serial examinations, which would decrease experimental animal numbers. A key advance for studies of pregnancy in mice has been the development of microultrasonography. This technology now provides in vivo morphometric quantification of embryonic and placental growth for mice.21,33 Here we review the application of microultrasonography to hemodynamic monitoring, vascular assessment, and placental development in mice and address its limitations briefly.  相似文献

When conducting field studies, it is common for ecologists to choose the locations of sampling units arbitrarily at the time sampling occurs, rather than using a properly randomised sampling design. Unfortunately, this ‘haphazard’ sampling approach cannot provide formal statistical inference from the sample to the population without making untestable assumptions. Here, we argue that two recent technological developments remove the need for haphazard sampling in many situations. A general approach to simple randomised sampling designs is outlined, and some examples demonstrate that even complicated designs can be implemented easily using software that is widely used among ecologists. We consider that more rigorous, randomised sampling designs would strengthen the validity of the conclusions drawn from ecological studies, to the benefit of the discipline as a whole.  相似文献

Intentional moving of species threatened by climate change is actively being discussed as a conservation approach. The debate, empirical studies, and policy development, however, are impeded by an inconsistent articulation of the idea. The discrepancy is demonstrated by the varying use of terms, such as assisted migration, assisted colonisation, or managed relocation, and their multiple definitions. Since this conservation approach is novel, and may for instance lead to legislative changes, it is important to aim for terminological consistency. The objective of this study is to analyse the suitability of terms and definitions used when discussing the moving of organisms as a response to climate change. An extensive literature search and review of the material (868 scientific publications) was conducted for finding hitherto used terms (N = 40) and definitions (N = 75), and these were analysed for their suitability. Based on the findings, it is argued that an appropriate term for a conservation approach relating to aiding the movement of organisms harmed by climate change is assisted migration defined as follows: Assisted migration means safeguarding biological diversity through the translocation of representatives of a species or population harmed by climate change to an area outside the indigenous range of that unit where it would be predicted to move as climate changes, were it not for anthropogenic dispersal barriers or lack of time. The differences between assisted migration and other conservation translocations are also discussed. A wide adoption of the clear and distinctive term and definition provided would allow more focused research on the topic and enable consistent implementation as practitioners could have the same understanding of the concept.  相似文献

Farmland birds are important indicators of the state of biodiversity in rural landscapes, and the occurrence and abundance of birds contribute to their importance as bioindicators. However, the measurement of farmland bird abundance can be difficult. The rapid growth of crop plants in the spring combined with disturbances related to farming practices, such as weed and pest control and other measures, can profoundly change the habitat suitability of arable fields for birds within a short period of time. Consequently, the existing dynamics must be incorporated into the applied methods, and a single value of bird abundance during the breeding season is insufficient to characterise the habitat functions of arable land. The abundance of farmland bird species is influenced by crop specific features, which profoundly change within the breeding season and which we have described using a novel concept called Moving Window Abundance. Based on field surveys in 29 observation areas within arable landscapes, each 1 km2, Skylark with territorial behaviour were counted using a mapping method related to the habitats and growth of field crops, such as wheat and maize. To describe the dynamic characteristics of the abundance during the breeding season over the time period from 16 March to 18 July, three methods of Moving Window Abundance were tested: patchy, adjacent and overlapping. These methods differ in the time-space continuity of the considered time window throughout the breeding season. In the Patchy Moving Window Abundance method, we used the exact days in which field surveys were conducted throughout the entire time period of the field surveys. However, with the Adjacent Moving Window Abundance method, the time window included five-day, ten-day and semi-monthly adjacent windows; in the Overlapping Moving Window Abundance method, the time window shifted daily, with five-day, ten-day and semi-monthly overlapping windows used in the calculation procedure. The results indicated that the dynamic nature of Skylark abundance (i) reached a maximum level in the first breeding period in the agricultural landscape and (ii) exhibited large variations in level and time within the various field crops. Therefore, abundance was not described conventionally using a single numerical value but rather using mathematical functions based on the spatial scale of the landscape and habitat. We conclude with recommendations for further research to standardise farmland bird monitoring.  相似文献

The spread of anthelmintic resistance in equine strongyle nematodes has become a major problem, advocating for the development of alternative control for strongyles. Our study consisted of both in vivo and in vitro experiments. We investigate for the first time the efficacy of a short-term consumption of tannin-rich sainfoin (Onobrychis viciifolia) or extra proteins in naturally infected horses. We used 30 horses allocated into three groups of 10 individuals that received for 18 days either (i) a tannin-rich diet with 70% DM sainfoin pellets (Sd), (ii) a protein-rich diet with 52% DM Italian rye-grass pellets and 18% DM grinded linseed expeller (Pd), or (iii) a control diet with 45% DM barley and 25% DM cereal-based pellets (Cd). The three diets were isoenergetic, covering 94% of animal energy requirements on average, and the Sd and Pd diets were isoproteic and provided extra proteins (227% of protein requirements v. 93% for the Cd diet). Pd and Cd were compared to test for benefits of receiving extra proteins, while Sd and Pd were compared to account for the effect of sainfoin secondary metabolites. There were no between-diet differences in faecal egg counts (FEC) or in worm burden evaluated from worm counts in faeces of drenched horses at the end of the experiment. However, coprocultures from the faeces collected in each group at the beginning and at the end of the experiment suggested a lower rate of strongyle larval development in the Sd group at the end of the experiment (Sd=8.1%, Pd=30.5%, Cd=22.6%). In vitro tests using sainfoin solutions evidenced the influence of sainfoin on strongyle larval development: adding 29% of sainfoin pellets to faeces reduced the strongyle egg development into infective larvae by 82% (P<0.001) and using solutions with sainfoin concentrations higher than 7.5 mg/ml reduced egg hatching by 37% (P<0.05). The short-term use of tannin-rich plants in horse diet could thus constitute a promising strategy to reduce the risk of infection by strongyles at pasture.  相似文献

Angiomotins were originally identified as angiostatin binding proteins and implicated in the regulation of endothelial cell migration. Recent studies have shed light on the role of Angiomotins and other members of the Motin protein family in epithelial cells and in pathways directly linked to the pathogenesis of cancer. In particular, Motins have been shown to play a role in signaling pathways regulated by small G-proteins and the Hippo–YAP pathway. In this review the role of the Motin protein family in these signaling pathways will be described and open questions will be discussed.  相似文献

There is no lack of pretext for reviewing historically how ecologists have striven to gain the respect of scholars in their own and other fields, and to demonstrate the wider public-utility of their science. If self-serving, in terms of securing the scope and resources required to advance their studies in ecology, such activities have also been encouraged, and indeed commissioned, in the belief that a greater awareness and understanding of the natural world is essential for human wellbeing. The concept of flow, a state of complete absorption in an intrinsically rewarding activity, has played a pivotal role in advancing notions of human well-being beyond minimising suffering towards promoting flourishing and thriving. While flow has played a fundamental role in human positive psychology, it has not yet been explored in non-human animals, leaving an enormous void in our understanding of intrinsic motivation in animals. As ethology and related fields keep progressing in uncovering complex cognitive and affective capacities of non-human animals, we propose the time is ripe to translate the concept of flow to animals. We start by embedding flow in the topic of intrinsic motivation and describe its impact on positive human psychology and potentially positive animal welfare. We then disambiguate flow from related concepts discussed in the animal literature. Next, we derive experimental approaches in animals from the canonical characteristics of flow in humans and provide guidelines for both inducing and assessing flow by focusing on two characteristics that do not necessarily depend on self-report, namely resistance to distraction and time distortion. Not all aspects of the human flow experience are (yet) translatable, but those that are may improve quality of life in captive non-human animals.  相似文献
