This paper is concerned with optimization of the fermentation conditions for preparation of alkaline pectin lyase (poly-galacturonic acid trans-eliminase, PATE) by Erwinia carotovora IFO3830 (E.C.IFO3830). The orthogonal matrix method was adopted as the experimental design method for media. The effects of media composition on the growth rate of E.C.IFO3830 are in the order of pectin > KH(2)PO(4)/Na(2)HPO(4) > MgSO(4) > (NH(4))(2)SO(4) > glucose > L-glutamate sodium, and those on PATE activity are in the order of glucose > pectin > L-glutamate sodium > KH(2)PO(4)/Na(2)HPO(4) > MgSO(4) > (NH(4))(2)SO(4). The strain of E.C.IFO3830 grows quickly and easily near an initial pH of 7.0, and especially at the alkaline range of pH 7.0-8.0. The growth rate profiles indicate that E.C.IFO3830 grows very quickly; its generation time t(1/2) is about 0.2 h. The appropriate fermentation period is about 10-20 h for a high level productivity of biomass and 25-35 h for high productivity of PATE enzyme.  相似文献

对新霉素产生菌费氏链霉菌进行中子辐射诱变和突变株筛选, 获得不产新霉素的突变株。以突变株为转化菌种, 以新霉素为底物, 对转化发酵液进行高效液相色谱分析, 研究了不同转化条件对新霉素转化的影响。结果表明, 底物浓度、底物添加时间、底物添加方式、接种量、培养基装量、转化时间、碳源、氮源、pH、温度对新霉素转化具有不同程度的影响。以转化条件优化参数进行转化大量培养, 转化液经4步离子交换层析进行分离纯化, 薄层层析检测纯化样品为单一斑点。采用薄层生物自显影对获得的4个转化产物分离样品做生物活性检测, 发现4个样品对金黄色葡萄球菌和姜青枯假单孢杆菌都具有抑制活性, 1个样品对大白菜软腐样品具有明显的抑制活性。  相似文献

The optimum conditions (pH and initial sugar concentration) of fermentation for the production of ethanol by 4 strains ofZymomonas mobilis (ATCC 10988, ATCC 12526, NRRL B 4286 and IFO 13756) were studied. An initial sugar concentration of 15 % (w/v) at pH 7.0 was found to be optimal for the first two strains and 20 % (w/v) initial sugar at pH 7.0 was found to be optimal for the last two strains. The fermentation pattern of these strains on synthetic medium, cane juice and molasses were compared. Strain NRRL B 4286 showed maximum ethanol production on synthetic medium while on cane juice ATCC 10988 and ATCC 12526 performed well. However, all the strains fermented molasses poorly.  相似文献

The optimal fermentation medium and conditions for mycelial growth and water-soluble exo-polysaccharides production by Isaria farinosa B05 were investigated. The medium components and fermentation conditions were optimized according to the one at a time method, while the concentration of medium components was determined by the orthogonal matrix method. The results showed that the optimal fermentation medium was as follows: sucrose 3.5% (w/v), peptone 0.5%, yeast extract 0.2%, K(2)HPO(4) 0.1%, and MgSO(4) 0.05%. The suitable fermentation conditions were as follows: initial pH 7.0, temperature 25 degrees C, medium volume 75 mL/250 mL, inoculum volume 5% (v/v), time 5d. In such optimal nutrition and environmental conditions, the maximal mycelial yield was 2.124 g/100 mL after 4 day's fermentation, while maximal water-soluble exo-polysaccharides production reached 2.144 g/L after 5 day's fermentation.  相似文献

Summary The reaction conditions for the enzymatic production of l-cystathionine were optimized, using the two kinds of cystathionine -synthase, types I and II, which are abundant in cell-free extracts of Erwinia carotovora (IFO 3830) and Bacillus sphaericus (IFO 3526), respectively. Under the optimal conditions, 178 and 184 mM l-cystathionine (40 and 41 g per liter of the reaction mixture) were synthesized with conversion ratios of 89 and 92% with the Erwinia and Bacillus enzymes, respectively.Recipient of a JSPS Fellowship for Japanese Junior Scientists  相似文献

This study was carried out to determine the possibility of the psychrophilic methane fermentation of the excess sludge produced from fish-processing wastewater treatment. Three different kinds of seed sludge, named M seed sludge, K seed sludge and KL seed sludge were used for batch methane fermentation in Erlenmeyer flasks. M seed sludge was mesophilic, while the other two were prepared in our laboratory by soil collection to obtain a psychrophilic biological community. The effects of temperature on the total volume of biogas generated and on the rate of biogas generation were examined. The activity of K seen sludge with respect to gas yield at 15°C was found to be 1.6-fold higher than that of M seed sludge. Enrichment techniques were then used to accumulate psychrophilic bacteria in KL seed sludge, and the activity at 15°C increased by about two times due to these techniques. Comparison of the three kinds of seed sludge indicates that methane fermentation at low temperature may be feasible, and therefore further study to isolate the psychrophilic bacteria in anaerobic digestion is needed.  相似文献

目的:研究酵母产γ-氨基丁酸的发酵条件,提高其产γ-氨基丁酸的能力。方法:以高产γ-氨基丁酸的酵母突变株为材料,通过单因素实验研究培养温度、摇床转速、接种量、种龄和培养时间等条件对菌株发酵生产γ-氨基丁酸的影响。结果:最适发酵条件为:培养温度30℃,摇床转速220 rpm,接种量4%,种龄为2d的种子菌,培养时间4d。在此发酵条件下,变异菌发酵液中γ-氨基丁酸含量高达2.588 g.L-1,较优化前提高了53%。结论:发酵条件的优化,提高了菌株产γ-氨基丁酸的能力。  相似文献

The soft rot coliform bacteria Erwinia carotovora subsp. carotovora and E. carotovora subsp. atroseptica were isolated by an enrichment method from the rhizosphere of many weed species and crop plants, collected in commercial potato fields either currently in potatoes or in a different crop as part of the rotation. Erwinia carotovora was isolated from 24 plant species in Colorado and 47 species in Scotland. Weeds contaminated with E. carotovora were found in fields growing other crops in which potatoes had not been grown for 1–2 and sometimes much longer. Weeds collected from virgin land in Colorado were not contaminated with E. carotovora but in Scotland virgin soils containing weed roots yielded E. carotovora subsp. carotovora . In general, the numbers of contaminated weeds rose from nil or low levels in spring and early summer to considerably higher levels during mid-season, and fell to progressively lower levels later. Erwinia carotovora subsp. carotovora was the predominant organism recovered from the rhizosphere, but E. carotovora subsp. atroseptica was less common, especially in Scotland, and its incidence varied in different seasons depending on factors such as temperature and moisture conditions. The bacteria could apparently persist in the root zone for an extended period of time and may be a source of inoculum to contaminate soft rot erwinia-free seed potato stocks; the origin of the bacteria was uncertain.  相似文献

目的对产类细菌素嗜酸乳杆菌P302的发酵条件进行研究。方法采用琼脂扩散法测定发酵上清液对大肠埃希菌O139的抑菌活性。结果通过正交试验确定P302产类细菌素的最佳培养基组分为牛肉膏4%、胰蛋白胨0.2%、酵母粉0.8%、碳酸钙0.4%和蔗糖1%,0.4%复合维生素,0.3%Tween-80有利于类细菌素的产生。发酵工艺条件为最适初始pH7.0,最适温度37℃,最佳接种量0.3%,种龄14h,厌氧培养22h。结论通过优化发酵条件和发酵工艺,类细菌素的产量提高了35%。  相似文献

混合菌群发酵秸秆可有效提高秸秆纤维的降解率及菌体蛋白的转化率,对拓广蛋白饲料来源、减少环境污染起到积极的作用。本研究以小麦秸秆为原料,在纤维素酶水解预处理的基础上,以米曲霉作为先导菌,进一步分解残留的粗纤维,为后期发酵提供充足的碳源。根据不同微生物的代谢特征和协同机理,试验确定了发酵阶段混合菌群的组成为:米曲霉、产朊假丝酵母和枯草芽胞杆菌;接种顺序为:先接种米曲霉,再接种产朊假丝酵母,最后接种枯草芽胞杆菌。正交试验表明,影响发酵主要因素的主次顺序为:秸秆与麸皮配比>接种比例>发酵时间>接种量>发酵温度;发酵的最适条件为:米曲霉的接种量2.5%,发酵12h后接入5%的产朊假丝酵母,继续发酵8h后接入2.5%的枯草芽胞杆菌,发酵温度为28℃,秸秆与麸皮的配比为4∶1,尿素添加量为1.2%;结合动力学分析,将混合菌群的发酵时间优化为35h,发酵产物中粗蛋白含量由原来的5.47%提高到25%左右。对最适发酵条件下的动力学过程进行了探讨,建立了以Logistic方程为基础的数学模型和动力学方程。本研究表明,混合菌群发酵秸秆提高了发酵产物中的粗蛋白含量。动力学分析对于了解发酵机理、掌握整个发酵过程中混合菌群生长的动态变化、优化发酵工艺具有重要的指导意义。  相似文献

对培养条件,如氮源种类、C/N比率、pH、种子种龄与接种量对被孢霉(Mortierellasp.)M14菌株的细胞生长和油脂形成的影响作了研究。结果表明,二级发酵优于一级发酵。M14菌株二级发酵时,种子种龄以49h为适,接种量宜大(30%)。无机氮源有利于不饱和脂肪酸的产生,有机氮源有利于细胞的增殖。低C/N比率有利于菌丝体产量的提高,提高C/N比率则能促进菌体细胞内的油脂合成。pH对菌体生长、油脂产生以及γ-亚麻酸含量都有显著的影响。  相似文献

【目的】以柚皮为原料,优化棘孢曲霉利用柑橘加工副产物固态发酵柚苷酶的条件。【方法】采用高效液相色谱法检测酶活力,通过单因素试验考察固水比、装样量、接种量、温度对柚苷酶发酵的影响,用正交试验优化发酵条件。【结果】单因素试验结果的显著性分析表明培养基的固水比、装样量和培养温度对柚苷酶产量有显著性影响,而接种量影响不显著;经正交试验确定的优化条件是:固水比1:1 (质量体积比),装样量5 g/250 mL三角瓶,温度为30 °C,接种1 mL孢子悬浮液,发酵8 d。在此优化条件下,柚苷酶酶活力为8.19 IU/g干物质,比初始培养基产柚苷酶活力提高7.38倍。【结论】通过对固水比、装样量和发酵温度进行优化,大幅度提高了棘孢曲霉固态发酵柑橘加工副产物的柚苷酶产量,为柚苷酶的生产提供了一种高产发酵工艺。  相似文献

Poly(glutamic acid) (PGA) is a water soluble, biodegradable biopolymer that is produced by microbial fermentation. Recent research has shown that poly(glutamic acid) can be used in drug delivery applications for the controlled release of paclitaxel (Taxol) in cancer treatment. The molecular weight of microbial poly(glutamic acid) is generally larger than what is required for drug delivery. As such, molecular weight reduction is a necessary step in producing poly(glutamic acid) for this application. Poly(glutamic acid) produced by Bacillus subtilis IFO 3335 was subjected to in situ depolymerization in the cell-free fermentation broth. Molecular weight reduction was measured, and an empirical kinetic model was used to correlate the experimental data. The kinetic rate constant, k, was found to be 6.92 × 10−6 h−1 at pH 7.0 and 37 °C, which were the optimum depolymerization conditions.  相似文献

葵花籽是一种优质的油料资源,是世界第四大油料作物。葵花籽不但含油量高,葵花籽油以其高达90%的不饱和脂肪酸和富含维生素E、胡萝卜素等营养成分的特点,被称为"保健油",有延缓衰老、调节新陈代谢和降低胆固醇等功能。采用酶辅助压榨法制备葵花籽油,在提高出油率的同时保留油脂的天然风味。通过单因素实验法研究了酶的种类、酶添加量、pH值、液固比、温度和时间对压榨葵花籽油出油率的影响,以确定最佳的工艺条件。结果表明,最佳工艺条件为:选用纤维素酶,酶添加量为0.7%,pH为4.5,液固比为25%,酶解温度为55℃,酶解时间为2.5 h。在此最佳条件下,出油率是传统冷榨法的3.48倍,油脂提取率达85%。  相似文献

为实现优势菌种的工程应用,由焦化废水处理站污泥筛选优势好氧菌A和兼性厌氧菌F,使用谷壳g、豆皮d为载体,采用固体发酵法进行固定化产品制备。初步研究表明固体发酵法固定菌种具有可行性,主要调控因素有温度、初始含水率、工业废水比例、pH、通风量和发酵时间。以pH8.0、温度好氧菌30℃(兼性厌氧菌35℃)、接种量1mL茴液儋田定化载体,初始含水量80%,固体发酵时间5d,低于60℃鼓风干燥,制备固定化产品。保存三个月,谷壳固定化产品好氧菌八的硝化能力和兼性厌氧菌R的反硝化能力不随保存时间的延长而改变;豆皮固定化产品氏、Fd降解能力随保存时问延长下降12%左右。以实验室岔/O工艺系统验证固定化产品处理焦化废水效果,缺氧池中无NO3-N积累,反硝化作用明显。氐和气对氰化物的去除率均达99%以上;对苯酚的去除效果气(98.84%)优于氏(79.6%);但对NH4+-N的去除率氏(75.46%)优于Ag(62.55%)。  相似文献

A study was taken up to evaluate the role of some fermentation parameters like inoculum concentration, temperature, incubation period and agitation time on ethanol production from kinnow waste and banana peels by simultaneous saccharification and fermentation using cellulase and co-culture of Saccharomyces cerevisiae G and Pachysolen tannophilus MTCC 1077. Steam pretreated kinnow waste and banana peels were used as substrate for ethanol production in the ratio 4:6 (kinnow waste: banana peels). Temperature of 30°C, inoculum size of S. cerevisiae G 6% and (v/v) Pachysolen tannophilus MTCC 1077 4% (v/v), incubation period of 48 h and agitation for the first 24 h were found to be best for ethanol production using the combination of two wastes. The pretreated steam exploded biomass after enzymatic saccharification containing 63 gL−1 reducing sugars was fermented with both hexose and pentose fermenting yeast strains under optimized conditions resulting in ethanol production, yield and fermentation efficiency of 26.84 gL−1, 0.426 gg −1 and 83.52 % respectively. This study could establish the effective utilization of kinnow waste and banana peels for bioethanol production using optimized fermentation parameters.  相似文献

Yeast flocculation is an important trait in the brewing industry as well as in ethanol production, through which biomass can be recovered by cost-effective sedimentation. However, mass transfer limitation may affect yeast growth and ethanol fermentation if the flocculation occurs earlier before fermentation is completed. In this article, a novel type of cell-cell flocculation induced by trehalose-6-phosphate synthase 1 (TPS1) promoter was presented. The linear cassette HO-P(TPS1)-FLO1(SPSC01)-KanMX4-HO was constructed to transform the non-flocculating industrial yeast S. cerevisiae 4126 by chromosome integration to obtain a new flocculating yeast strain, ZLH01, whose flocculation was induced by ethanol produced during fermentation. The experimental results illustrated that flocculation of ZLH01 was triggered by 3% (v/v) ethanol and enhanced as ethanol concentration increased till complete flocculation was achieved at ethanol concentration of 8% (v/v). Real time PCR analysis confirmed that the expression of FLO1(SPSC01) was dependent on ethanol concentration. The growth and ethanol fermentation of ZLH01 were improved significantly, compared with the constitutive flocculating yeast BHL01 engineered with the same FLO gene but directed by the constitutive 3-phosphoglycerate kinase promoter PGK1, particularly under high temperature conditions. These characteristics make the engineered yeast more suitable for ethanol production from industrial substrates under high gravity and temperature conditions. In addition, this strategy offers advantage in inducing differential expression of other genes for metabolic engineering applications of S. cerevisiae.  相似文献
