Two different theoretical models are used to represent the propulsive mechanisms of Opalina. One model uses the concept of an envelope over all the cilia, while the other considers an array of elongated rods, similar to the model used in part 1. The envelope model shows a correlation between the motion of the cilium tip and the type of metachronism exhibited by the organism but under-predicts the velocities of propulsion. Calculations of the velocity profile, force and bending moment are carried out on the three-dimensional beat of a cilium of Opalina ranarum using the cilia sublayer model. The mean velocity profile is found to be twisted in form: in a clockwise direction at the top of the cilia sublayer relative to the effective stroke. Calculations of the force and rate of working emphasize the approximately equal duration of the effective and recovery strokes. Overall the sublayer model is found to be a more informative and useful approach than the envelope model which is limited to small amplitude motions.  相似文献

The α and β subunits of highly potent ovine follitropin have been isolated by dissociation in 8 m urea, pH 7.5, and chromatography on DEAE-Sephadex A25. The isolated subunits display microheterogeneity on polyacrylamide gel electrophoresis and have very low activity in follitropin-specific radioreceptor and radioimmunoassays. The tryptophan fluorescence spectra of native follitropin and the isolated β subunit are different. The recombinant of follitropin α + β subunit had the same activity as the native hormone in the radioimmunoassay, but its activity in the radioreceptor and in vivo bioassay was about 65% of the intact hormone. Substitution of the follitropin α by ovine lutropin α subunit (prepared by a method not involving urea) to form the recombinant restored full activity in all the three assays investigated. The formation of recombined hormone proceeds at a rapid rate and is almost complete by 6 h. The α and β subunits of ovine follitropin differ from each other in amino acid composition. No significant differences were apparent in their carbohydrate composition. The amino acid composition of the ovine follitropin α and lutropin α subunits are very similar. The oxidized α subunit has phenylalanine at its NH2-terminus while aspartic acid is present at this position in the oxidized β subunit.  相似文献

Adsorption of lipases (EC 3.1.1.3) and various proteins at the air-water interface has been investigated in relation to the mechanism of lipase reaction. Aqueous solutions of lipases and denaturated proteins show surface activity as strong as that of synthetic detergents. However, ths surface activity of esterases and various other proteins is little or none. By foam fractionation it was shown that lipases were adsorbed at the air-water interface and the adsorption followed the equation of Langmuir's adsorption isotherm. The properties of lipase at the interface are discussed in relation to the mechanism of lipase reaction and the differences from the esterase reaction.  相似文献

Radiation damage induced in polycrystalline peptides and polypeptides in vacuum and at different oxygen partial pressures has been investigated using e.s.r. spectroscopy. For some peptides no alteration in the presence of oxygen was observed. For others, a reduction in the number of stable free radicals was observed as a function of pO2, time and chain length. In some cases, the presence of oxygen during irradiation produces peroxide radicals. The above observations suggest that oxygen can interact with free radicals, inducing an increase in electron delocalization and the possibility of internal damage migration. The interaction occurs between the radicals which are adjacent to the carboxylic terminals and the oxygen located in the interstitial spaces. It may occur with or without the production of paramagnetic peroxides, depending on the nature of the carboxylic adjacent radicals.  相似文献
