We obtained quantitative estimates of the effect exerted by a blocker of GABAA receptors, bicuculline, on GABA-activated transmembrane ion currents in isolated neurons from the rat spinal ganglia. The currents were evoked by short (20 msec) local GABA applications. Increases in the bicuculline concentration from 10 nM to 2 M progressively decreased the amplitude of GABA-activated ion currents; 5 M of the blocker completely suppressed these currents, while washing out recovered them. The dependence between the level of bicuculline-induced blockade and concentration of the agent was nonlinear and could be satisfactorily fitted by a reverse logistic function. Within the studied concentration range, bicuculline exerted no influence on the kinetic parameters of the current rising and falling phases.  相似文献

In cultured pyramidal neurons of the rat brain cortex, we recorded (in the whole-cell configuration) postsynaptic currents (PSC) evoked by direct electrical microstimulation of an axon of the interneuron adjacent to the pyramidal cell. Application of 5 M bicuculline rapidly, entirely, and reversibly blocked these currents. Linear changes in the holding potential on the membrane of the postsynaptic cell resulted in linear changes in the amplitude of averaged currents. The currents underwent reversion when the holding potential was –16 mV, which was close to the reversal potential for Cl- ions at their respective concentrations in the extra- and intracellular solutions. We conclude that the recorded currents are inhibitory PSC (IPSC) mediated by GABA release. The amplitudes of the recorded currents varied from a measurable minimum (8 pA) to more than 150 pA at a holding potential on the postsynaptic cell membrane of –80 mV. Times to peak of the high- and low-amplitude currents showed no significant differences, being about 6.4 msec on average. Decays of the current could be satisfactorily approximated by a monoexponential function with a mean time constant of 17 msec. The time constants of IPSC decay were distributed accordingly to the Gaussian law. In some cases, the amplitude distributions of IPSC were unimodal ((with a rightward asymmetry), but in most cases they were clearly polymodal. The amplitude distribution can be described by the sum of several Gaussian distributions; the distance between modes of the Gaussians was 25 ± 6 pA, on average. The obtained estimates of the amplitude of monoquantal GABA-induced IPSC in neurons of the brain cortex allow us to conclude that in various CNS regions the dimension of the vesicles in GABA-ergic synapses formed by inhibitory interneurons is identical.  相似文献

By intracellular dialysis of isolated neurons of the mollusksHelix pomatia andLimnaea stagnalis and by a voltage clamp technique the characteristics of transmembrane ionic currents were studied during controlled changes in the ionic composition of the extracellular and intracellular medium. By replacing the intracellular potassium ions by Tris ions, functional blocking of the outward potassium currents was achieved and the inward current distinguished in a pure form. Replacement of Ringer's solution in the extracellular medium with sodium-free or calcium-free solution enabled the inward current to be separated into two additive components, one carried by sodium ions, the other by calcium ions. Sodium and calcium inward currents were found to have different kinetics and different potential-dependence: mNa=1±0.5 msec, mCa=3±1 msec, hNa=8±2 msec, hCa=115±10 msec (Vm=0), GNa=0.5 (Vm=–21±2 mV), GCa=0.5 (Vm=–8±2 mV). Both currents remained unchanged by tetrodotoxin, but the calcium current was specifically blocked by cadmium ions (2·10–3 M), verapamil, and D=600, and also by fluorine ions if injected intracellularly. All these results are regarded as evidence that the soma membrane of the neurons tested possesses separate systems of sodium and calcium ion-conducting channels. In co-culture of spinal cord and dorsal root ganglion (DRG) neurons, we studied at different terms of culturing postsynaptic currents in DRG neurons evoked by direct electrical stimulation of single spinal neurons using a voltage-clamp technique in the whole-cell configuration. According to the reversal potential and sensitivity to bicuculline, these currents were classified as inhibitory postsynaptic currents (IPSC) carried by Cl- ions through GABAA receptors. During neuronal development in dissociated co-culture, the amplitude of evoked IPSC and their time to peak significantly increased. The time to peak of spontaneous IPSC (sIPSC) in DRG neurons remained unchanged, while the frequency of these currents increased with increasing culturing time. It is concluded that under culturing conditions spinal neurons establish inhibitory synaptic contacts with the somata of DRG neurons, and the number of such functional contacts increases in the course of culturing. Our findings show that in dissociated co-culture the process of formation of inhibitory synapses on the axon terminals of primary afferent neurons is akin to that realized in vivo, but with dissimilar topography of distribution of such synapses.  相似文献

Synaptoneurosomes isolated from cerebral cortices of male Sprague-Dawley rats were used for studying GABAA receptor-regulated chloride influx. The in vitro effects of GABA antagonists, SR 95531 (a pyridazinyl GABA derivative) and bicuculline, on pentobarbital-stimulated, muscimol-stimulated or flunitrazepam-enhanced, muscimol-stimulated chloride uptake were studied. The chloride uptake was determined at 30°C, for 5 sec. Pentobarbital and muscimol produced a maximal stimulation of chloride uptake in cortical synaptoneurosomes at 500 M and 50M, respectively. SR 95531 as well as bicuculline had no effect on the basal uptake of chloride. Whereas, SR 95531 (0.3–30 M) and bicuculline (0.1–100 M), when added 5 min before muscimol (50 M), produced a significant concentration-dependent inhibition of muscimol (50 M)-stimulated chloride uptake (IC50 s of 0.89±0.11 M and 13.45±2.10M, respectively). In studies of the inhibitory effects of SR 95531 and bicuculline on pentobarbital (500 M)-stimulated chloride uptake, the IC50 s were 0.81±0.12 M and 3.86±1.14 M, respectively. SR 95531 exhibited a more potent inhibitory effect than bicuculline on flunitrazepam-enhanced, muscimol-stimulated chloride uptake. The results revealed that SR 95531 has a more potent antagonistic effect than bicuculline on GABAA-regulated chloride flux.  相似文献

Acutely dissociated and cultured chick and rat dorsal root ganglion (DGR) neurons were studied by means of whole-cell patch-clamp technique. The high voltage-activated calcium current in DRG neurons is a result of mixture of N-type (-conotoxin-sensitive), P-type (-aga-IVA-sensitive), and L-type (dihydropyridine-sensitive) calcium currents. The modulation of these calcium channel currents in DRG neurons involves the GTP binding proteins and GABA B receptors. FLRFa, FIRFa, YMRFa, and some other peptides are found to slow down the kinetics of desensitization of proton-gated currents in rat trigeminal neurons but not in HEK-293T cell culture transfected with ASIC. The observed heterogeneity of peptide action in sensory neurons leads to a suggestion that FMRFa-related peptides may selectively affect transient current components of ASIC and DRASIC/DRASIC-MDEG2 heteromers.  相似文献

The percentages of the -chain variant Hb G-Philadelphia (Hb G) or 2 68 AsnLys2 were evaluated in 84 adult and 18 newborn heterozygotes. These included members of three families who were studied in more detail by nucleic acid hybridization techniques. The adult heterozygotes fell in two categories, one with a higher proportion of Hb G [46.5±1.0% (SD), N=21] and another with lower values (33.9±3.4%, N=63). Among the newborn heterozygotes, two babies fell in the category with the higher proportion of Hb G while 16 babies gave values between 25 and 34%. Studies of -chain gene organization on the parents of one neonate with a Hb G level of 27% at birth and 37% at 8 months excluded the presence of chromosomes with triplicated -chain genes which could lead to the 0G/ genotype. Rather, these studies on five Hb G heterozygotes from three families confirmed the linkage between Hb G and a specific type of -thalassemia-2 associated with the presence of a 16-kbp Bgl II fragment which most probably carries the G locus since it has been found in 19 Hb G heterozygotes studied to date. The presence of an -thal-2 heterozygosity and three -chain genes (0G/) was confirmed among Hb G heterozygotes with lower proportions of this variant. It is likely that the even lower values found in some newborn could arise through defective assembly of G- dimers. The presence of an -thal-2 homozygosity and two active -chain genes, one on each chromosome (0G/0), was confirmed among heterozygotes with the higher proportion of Hb G. One of each of these categories was present in each of the three families investigated. This type of variability in the number of active -chain genes due to a heterozygosity or a homozygosity for -thalassemia-2 explains the trimodality of Hb S percentages among heterozygotes and the atypical hematological or biosynthetic features among patients with -thalassemia and sickle-cell syndromes.This research was supported by USPHS Research Grants HLB-05168 and HLB-15158 and by designated research funds of the Veterans Administration. Summary The formation and development of synaptic contacts between dissociated chick spinal cord neurons has been investigated. By the 6th day in vitro immature profiles with few vesicles were observed. By 14–18 days mature types with numerous vesicles were found, indistinguishable from those of newly hatched chick spinal cord. After this period degeneration occurred, and was especially marked in the post-synaptic element. Such degeneration could be postponed by the addition of small numbers of somatic muscle cells. The Kanaseki and Kadota (1969) technique was applied to the study of coated vesicles at various stages of synaptic development.  相似文献

Voltage dependence of the deactivation kinetics of calcium inward currents was investigated in the somatic membrane of murine spinal ganglia neurons. It was found that deactivation of high threshold calcium current has a slower component (=0.80–0.85 msec at a repolarizing potential of –80 mV) as well as the principal transient exponential component (130 sec at the same potential repolarizing level). A dissimilar relationship exists between amplitudes of the transient and slower exponential components, describing deactivation of high threshold calcium current and degree of activation of the depolarizing shift in membrane potential; the former dependence is expressed by a sigmoid and the latter by a V-shaped curve. The slower component of deactivation of high threshold current was inhibited substantially by perfusing the cell with a Tris-PO4-containing solution. Low-threshold calcium tail current undergoes slower deactivation (=1.1–1.2 msec) at a repolarizing potential of –160 mV. A relationship between the time constant of low threshold current deactivation and the type of penetrating cation used was observed. A kinetic model of calcium current deactivation is suggested, taking account of the three different types of calcium channels, (one low and two high threshold) present in the somatic membrane.A. A. The mangrove killifish Rivulus marmoratus was reared at 25°±1°C and 17ppt salinity from 0 to 100 days after hatching (DAH), and its early development was described by examining growth and morphometric parameters, meristic characters (vertebral and fin-ray counts), bone-cartilage development, and pigmentation. Growth was isometric for preanal length, head length, snout length, body depth, pectoral-fin length, dorsal-fin length, anal-fin length, and caudal-peduncle depth. Negative allometric growth was observed in eye diameter and gape size. Meristic counts (mean±SD) for vertebrae (34.2±0.4) and dorsal- (8.6±0.5), anal- (11.4±0.5), and caudal-fin rays (30.2±0.8) were complete at 0 DAH (n=5), whereas pectoral-fin rays and pelvic-fin rays were complete by 30 DAH (14.5±0.4, n=5) and 60 DAH (4.2±0.8, n=5). Full ossification of meristic elements proceeded in the following sequence: vertebrae (by 30 DAH), caudal-, dorsal-, and anal-fin rays (by 60 DAH), pectoral-fin rays (between 60 DAH and 100 DAH), and pelvic-fin rays (by 100 DAH). Both morphological characters and meristic counts indicate that this species can be considered to be a juvenile after 9.8mm in standard length (20 DAH).  相似文献

Summary Ultrastructural localization of acid phosphatase activity was investigated in ultrathin (0.05 m) and semithin (0.5 and 0.75 m) sections of the small intestinal epithelial cells of adult rats. The results showed that the enzyme activity was localized on the membrane of microvilli, lateral cell membranes, lysosomes, the Golgi complex, and the GERL of Novikoff (a part of the smooth-surfaced endoplasmic reticulum located in close proximity to the inner Golgi saccules) of duodenal absorptive cells. The lysosomes contained within the duodenal and jejunal absorptive cells appeared to be mainly heterolysosomes rather than autolysosomes. The enzyme activity of absorptive cells was lower in the jejunum than in the duodenum, and was barely detectable except in the GERL and lysosomes of the ileum. The average numbers of lysosomes having a diameter of 0.21.0 m, per cell profile in sections of 214 duodenal, 226 jejunal and 318 ileal epithelial cells were 8.9±0.189, 6.4±0.155 and 3.5±0.027 (mean±SE), respectively. From these results, it was assumed that both the Golgi apparatus and GERL produce some lysosomes in the duodenal and jejunal absorptive cells, but only GERL does so in the ileum. It was considered also that because of an unexpectedly high number of lysosomes contained within the epithelial absorptive cells of the proximal intestine of adult rats, these cells may possess the strong heterophagic, as well as absorptive capacity.  相似文献

In experiments on the somata of sensory neurons isolated from the spinal and trigeminal ganglia of rats, we characterized three subclasses of proton-gated currents differing from each other in their kinetics of desensitization and characteristics of stationary desensitization (but not in the characteristics of stationary activation). A voltage clamp technique in the whole cell configuration and intracellular perfusion were used. Expression of the channels providing currents of each subclass depended on the soma diameter but not on anatomical localization of the neuron. Proton-gated channels of type I were characterized by mono- or biexponential kinetics of current desensitization with the duration of complete decay within a 1 to 15 sec range; the mean pH50 of the curve of stationary desensitization was 7.21 ± 0.02. Channels of type II possessed mostly monoexponential desensitization kinetics with the duration of decay within a 1 to 3 sec range; their pH50 of the stationary desensitization curve was 7.11 ± 0.02. Channels of type III showed mostly biexponential desensitization kinetics; the complete current decay lasted about 5 sec, while the mean pH50 was about 6.78 ± 0.02. Channels of type I were typical of small neurons (soma diameter 10-20 m), while those of types II and III were found mostly in large cells (35-60 m).  相似文献

The dynamics of coupled biological oscillators can be modeled by averaging the effects of coupling over each oscillatory cycle so that the coupling depends on the phase difference between the two oscillators and not on their specific states. Average phase difference theory claims that mode locking phenomena can be predicted by the average effects of the coupling influences. As a starting point for both empirical and theoretical investigations, Rand et al. (1988) have proposed d/dt= — K sin ), with phase-locked solutions =arcsin( /K), where is the difference between the uncoupled frequencies and K is the coupling strength. Phase-locking was evaluated in three experiments using an interlimb coordination paradigm in which a person oscillates hand-held pendulums. was controlled through length differences in the left and right pendulums. The coupled frequency c was varied by a metronome, and scaled to the eigenfrequency v of the coupled system K was assumed to vary inversely with c. The results indicate that: (1) and K contribute multiplicatively to (2) =0 or = regardless of K when =0; (3) 0 or regardless of when K is large (relative to ); (4) results (1) to (3) hold identically for both in phase and antiphase coordination. The results also indicate that the relevant frequency is c/v rather than c. Discussion high-lighted the significance of confirming =arcsin(/K) for more general treatments of phase-locking, such as circle map dynamics, and for the 11 phase-entrainment which characterizes biological movement systems.  相似文献

Three lindane (-1,2,3,4,5,6-hexachlorocyclohexane) treated soils were studied under laboratory conditions to determine the interaction between lindane and the soil microorganisms. Microbial populations and respiration were monitored to study insecticide effects. Formation of lindane degradation products and chloride content were examined to determine effects of the microorganisms. Some populations in lindane treated soils showed temporary declines but all ultimately recovered to at least the level of the controls in 16 weeks. Respiration was stimulated over a 9-week period especially in the sandy and clay loams, suggesting the possibility of microbial degradation of the insecticide. Lindane degradation products separated and identified by TLC included -2,3,4,5,6-pentachloro-1-cyclohexene (-PCCH), -3,4,5,6-tetrachloro-1-cyclohexene (-TCCH), -3,4,5,6-tetrachloro-1-cyclohexene (-TCCH), and pentachlorobenzene (PCB). Stable isotopes (13C, D) and radiocarbon weremeasured in methane bubbles emitted from rice paddies and swamps in southernThailand. Methane emitted from the Thai rice paddies was enriched in13C (mean 13C; –51.5 ±7.1 and–56.5 ± 4.6 for mineral soil and peat soil paddies,respectively)relative to the reported mean value of methane from temperate rice paddies(– 63 ± 5). Large seasonal variation was observed in13C(32) in the rice paddies, whereas variationinD was much more smaller (20), indicating that variation in13C is due mainly to changes in methane production pathways.Values of 13C were lower in swamps (–66.1 ±5.1)than in rice paddies. The calculated contribution of acetate fermentation from13C value was greater in rice paddies (mineral soils:62–81%, peat soils: 57–73%) than in swamps (27–42%). Din methane from Thai rice paddies (–324± 7 (n=46)) isrelativelyhigher than those from 14 stations in Japanese rice paddies ranging from–362 ± 5 (Mito: n=2) to –322 ± 8(Okinawa: n=3), due tohigher D in floodwaters. 14C content in methane produced fromThai rice paddies (127±1 pMC) show higher 14Cactivity compared with previous work in paddy fields and those from Thai swamps(110±2 pMC).  相似文献

1. The first part of this study looks at spontaneously active neurons located in the rostral ventrolateral medulla (RVLM) with projections to the thoracic spinal cord. Sixteen neurons were intracellularly recorded in vivo. Four out of 16 neurons were antidromically activated from the thoracic spinal cord (axonal conduction velocities varied from 1.8 m/s to 9.5 m/s).2. The simultaneous averages of the neuronal membrane potential and arterial blood pressure triggered by the pulsatile arterial wave or the EKG-R wave demonstrated changes in membrane potential (hyperpolarization or depolarization) locked to the cardiac cycle in four neurons in this group. These neurons (three of them bulbospinal) were further tested for barosensitivity by characterizing the responses to electrical stimulation of the aortic depressor nerve. Four neurons responded with inhibitory hyperpolarizing responses characterized as inhibitory postsynaptic potentials (IPSP) to aortic nerve stimulation (onset latency: 32.3 ± 5.0 ms; mean ± SEM).3. In two neurons in the RVLM, one of them characterized as barosensitive, electrical stimulation of the opposite RVLM (0.5 Hz, 1.0 ms pulse duration, 25–100 A) elicited excitatory postsynaptic potentials (EPSPs) with latencies of 9.07 and 10.5 ms. At resting membrane potential, the onset latency of the evoked EPSPs did not change with increasing stimulus intensities. Some of the recorded neurons were intracellularly labelled with biocytin for visualization. They were found in the RVLM.4. These experiments in vivo would support the idea of a functional commissural pathway between the RVLM of both sides.5. Anatomical data have shown that some of those commissural bundle fibers originate in the C1 adrenergic neuronal group in the RVLM. In the second part of this study, we used an intracellular recording technique in vitro to investigate the effects of the indirect adrenergic agonist tyramine on neurons in the RVLM with electrophysiological properties similar to premotor sympathetic neurons in vivo.6. Tyramine (0.5–1 mM) produced a pronounced inhibitory effect with hyperpolarization and increase in membrane input resistance on two neurons characterized as regularly firing (R), and on one neuron characterized as irregularly firing (I). This effect was preceded by a transient depolarization with increases in firing rate.7. These results would indicate that neurons in the RVLM recorded in vitro and with properties similar to premotor sympathetic neurons can be modulated by catecholamines released from terminals probably making synaptic contacts.  相似文献

Embryogenic avocado cultures were exposed to ionizing irradiation in order to determine its effect on proliferation and subsequent somatic embryo development. The approximate PD50 as determined by linear regression is 35 Gy 2 weeks after irradiation for Fuerte 2.11.1 and 4 weeks after irradiation for T362 2.11.1. Irradiation of embryogenic cultures did not significantly affect the number of early stage Fuerte 2.11.1 somatic embryos that developed directly from irradiated cultures; however, 10–50 Gy inhibited somatic embryo development. Irradiation of T362 2.11.1 embryogenic cultures at 25–50 Gy inhibited the number of intermediate and mature stages of somatic embryos that developed directly from irradiated cultures, and 50 Gy inhibited somatic embryo maturation. Inhibition of somatic embryo development could be partially offset by proliferation of irradiated embryogenic cultures as suspensions. Irradiation up to 10 Gy significantly increased the number of mature Fuerte 2.11.1 somatic embryos that developed from suspension cultures. Irradiation with doses up to 25 Gy stimulated development of heart stage T362 2.11.1 somatic embryos; however, mature somatic embryo development was suppressed at dosages of 10 Gy and greater.  相似文献

Summary Dissociated chick embryo spinal ganglia neurons, cultivated without direct contact with glial cells maintain some enzymatic activities, for example: carboxylic esterases, succinic-dehydrogenase (SDH), glutamic-dehydrogenase (GDH), monoamine oxidase (MAO), lactico-dehydrogenase (LDH) and alcoholic-dehydrogenase (ADH) for several days periods.Nerve growth factor (NGF) prolongs the maintenance of the mitochondrial enzymes, carboxylic esterases, LDH and ADH in cultures of isolated neurons. Changes in body temperature (Tb) and feeding were characterized in unrestrained rats following the micro-injection into the anterior hypothalamic preoptic area (AH/POA) of macrophage inflammatory protein-1 (MIP-1), MIP-1 or MIP-1. After the rats recovered from the stereotaxic implantation of a single guide tube placed in the AH/POA, either one of the MIP-1 compounds or control CSF was micro-injected in a volume of 1.0 l into this area. Changes in body temperature (Tb) and food and water intakes were monitored throughout each experiment. When micro-injected into the AH/POA in a dose of 28 or 280 pg, doublet MIP-1 and MIP-1 evoked a monophasic fever which increased above baseline to a mean maximum of 2.17±0.14°C and 2.1±0.24°C, respectively. MIP-1 micro-injected similarly evoked a biphasic fever, with the Tb declining transiently at the 30 min point 0.4°C lower than the congruent rises in Tb evoked by doublet MIP-1 or MIP-1. The secondary rise in Tb induced by MIP-1 had a latency of 1.5–2.0 hrs and reached a maximum of 1.56±0.16°C. Although all three cytokines significantly attentuated the rats' mean intake of food during the 24 hr interval after their micro-injection into the AH/POA, doublet MIP-1 exerted the most potent anorexic effect in comparison to that of the saline control rats. However, neither body weight nor intake of water was altered significantly by the three cytokines. These results demonstrate a functional distinction between the febrile actions of MIP-1 and MIP-1 on cells of the AH/POA. It is envisaged that MIP-1 underlies the initial phase of a pyrogen-induced fever, whereas MIP-1 could mediate the secondary phase of a biphasic fever. Clearly, the localized elevation of either MIP-1 or MIP-1 in the AH/POA perturbs the neuronal mechanism underlying the set-point for body temperature. Thus, both MIP-1 and MIP-1 could play a functional role in the pathological events occurring in neurons of the AH/POA in response to an endotoxin or other pyrogen challenge.  相似文献
