探讨凤梨释迦Annona×atemoya套袋果实发育过程中不同类型果袋袋内温度、光参数变化,以及果袋类型对果实品质的影响。结果表明,不同类型果袋内温光环境差异大,果袋类型对果实外观及固形物含量存在显著影响。内黑外黄单层袋不透光,夜间袋温较高,果实固形物含量高于白色袋及半边透明白色袋,但果色较差,呈黄白色。白色袋具有一定透光性,白天袋内温度高于内黑外黄单层袋,果实呈浅绿色,果色较好。半边透明白色袋,袋内光强最大,其果实颜色、固形物含量与白色袋未见明显差异。不同类型果袋对果实大小,单果重,以及果柄的长度和粗度均未见显著影响。  相似文献

The present study was designed to determine whether a bag made from ethylene-vinyl acetate copolymer (EV) with superior flexibility at subzero temperature is suitable for a storage container of single-donor apheresis platelets. Apheresis platelets were stored with 100 ml plasma in 1-liter bags made of EV or standard polyvinyl chloride (PVC) plastic at 22 degrees C with constant agitation. The oxygen permeability of the 1-liter EV bag averaged 1447 nmol/min/atm, which was about 1.5 times higher than that of PVC bags. The partial oxygen tension (PO2) of platelet concentrates (PC) has linearly decreased to 16 mm Hg with increasing platelet counts. The level of the partial carbon dioxide was always higher in EV bags than in PVC bags. Oxygen consumption rates of platelets stored in EV and PVC bags with a sufficient oxygen supply averaged 1.25 and 1.20 nmol/min/10(9) platelets, respectively. The rates of glucose consumption and lactate production were not changed in two bags. Ninety percent of the total ATP production of about 8 nmol/min/10(9) platelets were generated through the aerobic metabolism. The platelet counts in the 1-liter EV and PVC bags, at which PO2 is 16 mm Hg, were 2.2 and 1.5 x 10(11) platelets, respectively. The study indicates that apheresis platelets stored in EV bags at 22 degrees C have no different metabolic changes when compared with those of PVC bags. In addition, the number of platelets maintaining the aerobic metabolism is 1.5 times higher than that of PVC bags.  相似文献

Correlated traits are important from an evolutionary perspective as natural selection acting on one trait may indirectly affect other traits. Further, the response to selection can be constrained or hastened as a result of correlations. Because mating behaviour and body colour can dramatically affect fitness, a correlation between them can have important fitness ramifications. In this work, melanic (black) male mosquitofishes (Gambusia holbrooki) with temperature-sensitive body-colour expression are bred in captivity. Half of the sons of each melanic sire are reared at 19 degrees C (and express a black body colour) and half are reared at 31 degrees C (and express a silver body colour). The two colour morphs are placed in the same social setting and monitored for behavioural differences. Mating behaviour and colour are correlated traits. Mating behaviour differs markedly between the two phenotypes, despite high genetic relatedness. Melanic (black) phenotypes are more aggressive towards females, chasing them and attempting more matings than their silver siblings. Females avoid melanic-male mating attempts more than silver-male mating attempts. When males with temperature-sensitive colour expression are melanic and aggressive, they probably experience a very different selective regime in nature from when they are silver and less aggressive. Under some conditions (e.g. predation), melanic coloration and/or aggression is advantageous compared with silver coloration and/or less aggressive behaviour. However, under different conditions (e.g. high-frequency melanism), melanism and/or aggression appears to be disadvantageous and melanic males have reduced survival and reproduction. Selective advantages to each morph under different conditions may enable the long-term persistence of this temperature-sensitive genotype.  相似文献

The use of disposable bags for cell culture media storage has grown significantly in the past decade. Some of the key advantages of using disposable bags relative to non‐disposable containers include increased product throughput, decreased cleaning validation costs, reduced risk of cross contamination and lower facility costs. As the scope of use of disposable bags for cell culture applications increases, problematic bags and scenarios should be identified and addressed to continue improving disposables technologies and meet the biotech industry's needs. In this article, we examine a cell culture application wherein media stored in disposable bags is warmed at 37°C before use for cell culture operations. A problematic bag film was identified through a prospective and retrospective cell culture investigation. The investigation provided information on the scope and variation of the issue with respect to different Chinese hamster ovary (CHO) cell lines, cell culture media, and application‐specific parameters. It also led to the development of application‐specific test methods and enabled a strategy for disposable bag film testing. The strategy was implemented for qualifying an alternative bag film for use in our processes. In this test strategy, multiple lots of 13 bag film types, encompassing eight vendors were evaluated using a three round, cell culture‐based test strategy. The test strategy resulted in the determination of four viable bag film options based on the technical data. The results of this evaluation were used to conclude that a volatile or air‐quenched compound, likely generated by gamma irradiation of the problematic bag film, negatively impacted cell culture performance. © 2013 American Institute of Chemical Engineers Biotechnol. This study was carried out to investigate leakage/transport across the bag material of six outer cryopreservation bags in common use within NHS Blood and Transplant. In order to do this two different leak testing procedures; coloured dye and hydrogen tracer gas, were used. The data obtained show that a coloured dye cannot permeate through the materials both at room temperature and following storage at liquid nitrogen temperature (? 196 °C). In addition, when filled with the smallest elemental molecule, hydrogen, in the form of a tracer gas, all of the bags only allowed trace amounts of hydrogen to escape, either through the seal or the bag material. The data indicated that each of the bag materials tested would be capable of preventing bacterial or viral cross-contamination as long as the material remained intact.  相似文献

The production of red blood cell concentrates from human donors is a very expensive procedure and human resources are in short supply. Under perfect storage conditions at a temperature of 2-6 degrees C, a blood bag must be used within 35-49 days (in Germany). Visual inspection of the bag for apparent hemolysis by a blood bank physician is a crucial but subjective quality control assessment. Since an interruption of the cold chain cannot be definitely ruled out, bags are often disposed of prematurely for safety reasons. There is currently no method of testing a closed blood bag with respect to hemolysis for its suitability to be used in a transfusion. The proposed optical measuring device is a hemoglobin sensor which determines the free hemoglobin in standard erythrocyte concentrates without opening the bag. The optical measurements are done on the flexible tube connected to the main bag. The optical measurements were evaluated using standard hemoglobin solutions with an accuracy of 0.005 g/dL. These investigations show that in the future each blood bag can be tested non-invasively for its content of free hemoglobin. This will contribute to decreasing the wastage rate of red blood cell concentrates.  相似文献

Softness and flexibility of PVC are due to the addition of plasticizers in high concentration; the most used of them for blood storage bags is DEHP. In this work, a method for labelling DEHP with 14C from 14C phtalic anhydrid is given. A piece of PVC from a commercial blood bag is labelled, in our laboratory, with 14C DEHP and used to follow the kinetics of DEHP leaching in blood during storage. It is also used to study the influence of some parameters such as lipids amount of blood, shaking, and plastic sterilisation on this leaching. DEHP leaching is a three steps phenomenon and the level is not correlated to lipids content of blood. Thermal treatment of PVC and shaking have an influence on leaching. DEHP is not metabolised during blood storage at 4 degrees C and can not be detected as free molecule; it is absorbed on plasmatic proteins.  相似文献

Rapid, reversible colour change is unusual in animals, but is a feature of male chameleon grasshoppers (Kosciuscola tristis). Understanding what triggers this colour change is paramount to developing hypotheses explaining its evolutionary significance. In a series of manipulative experiments the author quantified the effects of temperature, and time of day, as well as internal body temperature, on the colour of male K. tristis. The results suggest that male chameleon grasshoppers change colour primarily in response to temperature and that the rate of colour change varies considerably, with the change from black to turquoise occurring up to 10 times faster than the reverse. Body temperature changed quickly (within 10 min) in response to changes in ambient temperature, but colour change did not match this speed and thus colour is decoupled from internal temperature. This indicates that male colour change is driven primarily by ambient temperature but that their colour does not necessarily reflect current internal temperature. I propose several functional hypotheses for male colour change in K. tristis.  相似文献

An entirely new mixing device, particularly suitable for preparing bacterial suspensions from foods, fabrics, swabs, and other fairly soft materials, has been developed. With this technique the sample and diluent are put into an inexpensive, sterile plastic bag which is vigorously pounded on its outer surfaces by paddles when placed inside the machine. The resulting compression and shearing forces effectively remove even deep-seated bacteria. After samples are taken for analysis the bag and its remaining contents are thrown away. Labor involved in cleaning and sterilizing reusable homogenizer cups or probes is eliminated, and the device is immediately ready for reuse. Running costs are thus drastically reduced, compared with conventional homogenizers. Additional advantages of this device, which is simple and inexpensive to manufacture, are low noise level, negligible temperature rise, and the small storage space required for bags.  相似文献

The temperature profile of infectious laboratory waste being autoclaved was examined relative to the type of containers used in the process. A standardized waste load (1,750 +/- 4 g) placed in the container was evaluated by using a direct readout thermocouple. The sensor of the thermocouple was placed within an unused and outdated agar plate, centrally located about 5 cm from the bottom of the container. The gravity displacement autoclave tested reached 121 degrees C within 3 min. Waste within a steel container (plus 1 liter of water) reached 108, 120, and 122 degrees C at 12, 30, and 50 min, respectively. Without the addition of water, the corresponding temperatures were 60, 110, and 120 degrees C, respectively. With a steel container, "autoclavable" plastic bags, and no additional water, the temperatures were 36, 71, and 105 degrees C, respectively. When 1 liter of water was placed in the autoclavable bag, the temperatures were 98, 115, and 121 degrees C, respectively. Waste within a polypropylene container (dimensions similar to those of the steel container) with and without the addition of 1 liter of water, reached a maximum temperature of 108 degrees C at 50 min. With a polypropylene container, autoclavable plastic bag, and 1 liter of water, the corresponding temperature was 99 degrees C. Without the addition of water, the temperature was 92 degrees C. The importance of container, moisture, and material in autoclaving was demonstrated.  相似文献

OBJECTIVE--To use data from the fourth national survey of morbidity in general practice to investigate the association between home visiting rates and patients'' characteristics. DESIGN--Survey of diagnostic data on all home visits by general practitioners. SETTING--60 general practices in England and Wales. SUBJECTS--502 493 patients visited at home between September 1991 and August 1992. MAIN OUTCOME MEASURES--Home visiting rates per 1000 patient years and home visiting ratios standardised for age and sex. RESULTS--10.1% (139 801/1 378 510) of contacts with general practitioners took place in patients'' homes. The average annual home visiting rate was 299/1000 patient years. Rates showed a J shaped relation with age and were lowest in people aged 16-24 years (103/1000) and highest in people aged > or = 85 years (3009/1000). 1.3% of patients were visited five or more times and received 39% of visits. Age and sex standardised home visiting ratios increased from 69 (95% confidence interval 68 to 70) in social class I to 129 (128 to 130) in social class V. The commonest diagnostic group was diseases of the respiratory system. In older age groups, diseases of the circulatory system was also a common diagnostic group. Standardised home visiting ratios for the 60 practices in the study varied nearly eightfold, from 28 to 218 (interquartile range 67 to 126). CONCLUSIONS--Home visits remain an important component of general practitioners'' workload. As well as the strong associations between home visiting rates and patient characteristics, there were also large differences between practices in home visiting rates. A small number of patients received a disproportionately high number of home visits. Further investigation of patients with high home visiting rates may help to explain the large differences in workload between general practices and help in allocation of resources to practices.  相似文献

During the past five years the incidence of imported malaria increased among patients seen in East Birmingham Hospital and in St Thomas''s Hospital, London. Plasmodium vivax was the predominant species in Birmingham, and was almost always acquired by Asian immigrants visiting the Indian subcontinent. In St Thomas''s P falciparum was most commonly imported, usually by African immigrants visiting Nigeria and Ghana. Two patients (one Irish, one Japanese) died of falciparum malaria after visiting tropical Africa. In both hospitals the immigrant patients had seldom taken prophylactic drugs, and the few who had, ceased to do so on arrival in the UK and sometimes before leaving the malarious country. Apparently immigrants who visit their homeland do not consult their general practitioners before travelling, are given inappropriate advice, or do not take appropriate advice when given. Since the incidence of imported falciparum malaria in the UK is rising, the following points should be considered: the infection may be lethal, particularly in patients lacking immunity; it can mimic other diseases, which may lead to delayed diagnosis; severe disease may be associated with few parasites on a blood film, and even if the result is negative further tests should be performed; clinicians and hospital pharmacists should be aware of the need to keep permanent stocks of parenteral chloroquine and quinine preparations.  相似文献

Three hundred and seventy-five families replied to a questionnaire about the use of their disabled children''s wheelchairs. Many problems were found, such as difficulty in folding the chair and placing it in the car boot and difficulty in using pulbic transport. These families need adequate guidance to anticipate and deal with the problems. Provision of a wheelchair does not solve the mobility problems. The parents are faced with the task of transporting both the disabled child and the wheelchair whenever they want to use their own car or public transport. Ease of folding and lightness are important criteria for wheelchairs carried by cars. Parents need careful guidance about selecting suitable cars and also need to be shown how to lift the chairs without endangering their backs. Such advice should be provided by every wheelchair clinic and assessment and rehabilitation certre.  相似文献

Millions of blood products are transfused each year, and many lives are directly affected by transfusion. Platelet concentrate (PC) is one of the main products derived from blood. Even under good storage conditions, PC is likely to suffer cell damage. The shape of platelets changes after 5 to 7 days of storage at 22°C. Taking into consideration that some platelet proteins undergo changes in their shape and functionality during PC storage. Sixteen PC bags were collected and each PC bag tube was cut into six equal pieces to perform experiments with platelets from six different days of storage. Thus, on the first day of storage, 1/6 of the tube was used for miRNA extraction, and the remaining 5/6 was stored under the same conditions until extraction of miRNAs on each the following five days. Samples were sequenced on an Illumina Platform to demonstrate the most highly expressed miRNAs. Three miRNAs, mir127, mir191 and mir320a were validated by real-time quantitative PCR (RQ-PCR) in 100 PC bags tubes. Our method suggests, the use of the miRNAs mir127 and mir320a as biomarkers to assess the "validity period" of PC bags stored in blood banks for long periods. Thus, bags can be tested on the 5th day of storage for the relative expression levels of mir127 and mir320a. Thus, we highlight candidate miRNAs as biomarkers of storage damage that can be used as tools to evaluate the quality of stored PC. The use of miRNAs as biomarkers of damage is unprecedented and will contribute to improved quality of blood products for transfusions.  相似文献

By the end of June 1969 home nurses in Bristol were attached to 18 general practices caring for about 137,000 patients, or about one-third of the city''s population. Attachment was associated with an increase by about one-third in the number of patients referred by general practitioners for home nursing. Additional benefits derived from attachment during the nine months from January to September 1969 were 2,047 items of service performed by nurses in general practitioners'' surgeries, 65 home visits to patients who were not receiving domiciliary nursing care, improved communications between general practitioners and nurses, and opportunities for both doctors and nurses to widen their fields of work. The travelling expenses paid to Bristol''s nurses increased by 9·5%.It is suggested that the benefits to patients, doctors, and nurses of attachment far outweigh the costs and that there is scope for extending the role of the attached nurse in the surgery and in home visiting.  相似文献

The effect of within-day delays of 0.5, 2.5, 4.5 and 6.5 h between collection of rumen fluid from a cow and initiation of in vitro fermentation, as well as storage of rumen fluid for 48 h at either −24°C or 6, 22 and 39°C, on in vitro digestion of neutral detergent fibre (NDF) at 48 h was determined. In addition, the 48 h in vitro digestion of NDF, determined with a minimum time delay (i.e., 0.5 h) between collection from the cow and initiation of incubation, was compared to NDF digestion determined in sacco at 48 h. Rumen inoculum from a single cow was utilized in a thrice replicated incubation with whole crop alfalfa, corn, cereal and sudangrass forages of a lower and higher quality. The same cow was used as the host for the in sacco bags. The in vitro procedure used a bulk procedure with 5.0 cm × 5.5 cm multi-weave polyethylene polyester polymer bags that retained particles of 25 mm and larger. The in sacco procedure used the same bags retained in a large mesh bag. A within-day time delay of up to 6.5 h between collection of rumen fluid from the cow and initiation of in vitro fermentation had no impact on measured 48 h in vitro digestion of NDF. In contrast, no temperature dependent storage procedure maintained 48 h in vitro digestion of NDF at levels determined with no 48 h storage, although high quality alfalfa was least affected by any storage procedure. The 48 h in vitro digestion of NDF, determined using the minimum time delay between collection from the cow and initiation of the incubation, was higher than values obtained in sacco. Results show that this bulk in vitro procedure resulted in higher 48 h digestion of NDF than those determined with a similar in sacco procedure, thereby suggesting that laboratories located some distance from the donor animal can utilize in vitro procedures to accurately estimate 48 h digestion of NDF. However, storage of rumen fluid for 48 h, by any temperature dependent procedure examined, in order to facilitate fewer trips to the donor animal, or trips of substantially longer duration, will underestimate 48 h digestion of NDF to an extent that depends upon the forage incubated.  相似文献

AIMS: Recoveries of spiked standard suspensions are used to evaluate method performance. For many applications, gamma-irradiated Cryptosporidium oocysts are appropriate. In contrast, methods that determine viability, such as Cryptosporidium cell culture, require the use of live oocysts. Oocyst standards are usually prepared at a flow cytometry laboratory for use at another laboratory, and thus the samples are shipped. The goal of this study was to evaluate the shipping and storage stability of flow cytometry enumerated live oocysts over time at three temperatures: 4 degrees C, room temperature and 37 degrees C. METHODS AND RESULTS: Replicate samples containing 100 live C. parvum oocysts were prepared by flow cytometry and stored at 4 degrees C, room temperature and 37 degrees C. These samples were counted at various time points. Significant oocyst losses were observed after storage for 1 day at 37 degrees C, 7 days at room temperature and 21 days at 4 degrees C. CONCLUSIONS: Live C. parvum oocysts internal standards should be used within 10 days of preparation, and stored and shipped at 4 degrees C. SIGNIFICANCE AND IMPACT OF THE STUDY: When evaluating method performance with live oocysts, both the storage temperature and time are critical factors for obtaining reliable and accurate results.  相似文献

The effect of whole lupin seeds (Lupinus albus cv Lublanc) at 120, 150 and 195 degrees C on in situ nitrogen degradability (Dg.N) was measured by the nylon bag technique using fistulated non-lactating Holstein cows. The N degradation was evaluated in nylon bags suspended in the rumen; heating the seeds at 120, 150 and 195 degrees C decreased the Dg.N value: 83.9, 72.9 and 53.0 respectively vs 95.3% (rumen outflow rate of 0.06/h). To estimate the total N disappearing in the digestive tract, bags were incubated in the rumen for 16 h, then in a pepsin bath for 2 h and then introduced into the duodenum for subsequently recovery in feces. The whole tract degradability of N was always high, approximately 98.3%. The amounts of N which disappeared in the intestine increased from 3.1 (untreated seeds) to 15.1, 26.3 and 44.7% as the temperature rose to 120, 150 and 195 degrees C respectively. The PDIN and PDIE contents (g/kg of DM) of the raw whole lupin seeds were 224 and 84 respectively; extrusion elevated these values by 10-32% for PDIN and 57-194% for PDIE. The augmentation in the supply of dietary proteins to the postruminal parts as a result of extrusion could rapidly benefit high yielding cows.  相似文献

Corn stover has great potential as a biomass feedstock due its widespread availability. However, storage characteristics of moist corn stover harvested from single-pass harvesters have not been well quantified. In 2007, whole-plant corn stover at 19.1–40.3% (w.b.) moisture content was stored for 237 days in aerobic piles, one covered and one uncovered, as well an anaerobic silo bag. In 2008, two stover materials—whole plant and cob/husk from 31.7% to 58.1% (w.b.) moisture—were stored for 183 or 204 days in covered and uncovered anaerobic piles, ventilated bags, or anaerobic silo bags. Stover stored in uncovered piles was rehydrated by precipitation, which increased biological activity resulting in dry matter (DM) losses from 8.2% to 39.1% with an average of 21.5%. Stover in covered piles was successfully conserved when the average moisture was less than 25% (w.b.) with DM losses of 3.3%. Stover above 36% (w.b.) moisture and piled under a plastic cover had DM losses from 6.4% to 20.2% with an average of 11.9%. Localized heating occurred in the aerobic piles when moisture was above 45% (w.b.) which lead to temperatures where spontaneous combustion might be a concern (i.e., >70°C). Ambient air blown through a center tube in the ventilated storage bag dried stover near the tube to an average of 24.2% (w.b.), but the remainder of the bag averaged 46.8% (w.b.) at removal. Loss of DM ranged from 7.4% to 22.0% with an average of 11.8% with this storage method. Stover was most successfully conserved in the bags where anaerobic conditions were maintained. Under anaerobic conditions, DM losses ranged from 0.2% to 0.9%. When anaerobic conditions were not maintained in the silo bag, DM losses averaged 6.1% of DM. Anaerobic storage is the best solution for conserving the value of moist corn stover.  相似文献
