This study investigated factors important to the development of the liquid nitrogen (LN) vapor sperm cryopreservation technique in farmed greenlip abalone Haliotis laevigata, including (1) cryoprotectant agent (CPA) toxicity; (2) cooling temperature (height above LN surface); (3) thawing temperature; (4) sperm to egg ratio; and (5) sugar supplementation, using sperm motility, fertilization rate or integrity/potential of sperm components and organelles as quality assessment indicators. Results suggested that among the single CPAs evaluated 6% dimethyl sulfoxide (Me2SO) would be the most suitable for sperm cryopreservation in this species. The highest post-thaw sperm motility was achieved with the sperm that had been exposed to LN vapor for 10 min at 5.2 cm above the LN surface, thawed and recovered in 60 and 18 °C seawater bathes, respectively after at least 2 h storage in LN. The highest fertilization rates were achieved at a sperm to egg ratio of 10,000:1 or 15,000:1. Addition of 1% glucose or 2% sucrose produced significantly higher post-thaw sperm motility than 6% Me2SO alone. Among the three cryoprotectant solutions further trialled, 6% Me2SO + 1% glucose produced the highest fertilization rate of 83.6 ± 3.7%. Evaluation of sperm has shown that the addition of glucose could significantly improve the sperm plasma membrane integrity and mitochondrial membrane potential. These results demonstrated a positive role of glucose in the improvement of sperm cryopreservation in farmed greenlip abalone.  相似文献

金樱根为三金片的主要成分,但目前对于金樱根的化学成分和药理作用研究甚少。为了阐明金樱根的物质基础和生物活性,该研究采用硅胶、Sephadex LH-20、MCI gel CHP 20P等柱色谱以及HPLC半制备等方法,对金樱根(Rosa laevigata)的化学成分进行研究。结果表明:从中共分离得到9个化合物,经过波谱数据分析结合文献对照分别鉴定为儿茶素(1),表儿茶素(2),rosamultin(3),sericoside(4),2α,3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid-3-O-β-D-glucopyranosyl ester(5),kaji-ichigoside F1(6),β-D-Glucopyranosyl 3β,19α-dihydroxy-2-oxo-urs-12-en-28-oate(7),胡萝卜苷(8),β-谷甾醇(9),其中化合物2、4、5、7为首次从该植物中分离得到。该研究结果为金樱根在功能医药领域的开发利用提供了理论依据。  相似文献

The present phytochemical investigation on Salacia amplifolia led to the isolations of 13 triterpenes, including quinonemethides, friedelanes, oleananes and ursanes triterpenes, three simple phenolics, one polyol and one chromanone. All of them were isolated from this plant for the first time. The presences of quinonemethide triterpenoids might be employed as the common characteristic constituents of both Hippocrateaceae and Celastraceae families.  相似文献

Phytochemical investigation of the methanolic extract from the aerial parts of Agrimonia pilosa led to the isolation of three compounds, (−)-aromadendrin 3-O-β-d-glucopyranoside (1), desmethylagrimonolide 6-O-β-d-glucopyranoside (2), and 5,7-dihydroxy-2-propylchromone 7-O-β-d-glucopyranoside (3), together with nine known compounds, agrimonolide 6-O-glucoside, takanechromone C, astragalin, afzelin, tiliroside, luteolin, quercetin, isoquercetrin, and quercitrin. Their structures were determined by various spectroscopic analysis and chemical transformations.  相似文献

Nine (1–9) and seven (1–6, 10) compounds were isolated from the fruits of Sonneratia caseolaris and Sonneratia ovata, respectively. Their structures were identified by comparing their MS and NMR data as well as the physical properties with the literature. All the isolated compounds were screened against a rat glioma C-6 cell line using the MTT assay method; only compounds (-)-(R)-nyasol (1), (-)-(R)-4′-O-methylnyasol (2) and maslinic acid (6) were found to show moderate cytotoxic activity. Our findings from these two kinds of fruits can be used as a foundation for further chemotaxonomic studies on Sonneratia species. The nor-lignans (1, 2) and 6H-benzo[b,d]pyran-6-one derivatives (3, 4) were isolated from this genus for the first time, indicating that these two classes of compounds may tentatively be considered as taxonomic markers for Sonneratia genus.  相似文献
