Diabetes mellitus is a group of complicated metabolic disorders characterized by high blood glucose level and inappropriate insulin secreting capacity due to decreased glucose metabolism and pancreatic β cell mass or dysfunction of β cells. Thus, improving glucose metabolism and preserving β cell mass and function might be useful for the treatment of diabetes. In this study, a novel acidic polysaccharide LBP-s-1 extracted from Lycium barbarum L. was obtained by purification using macroporous resin and ion-exchanged column. Monosaccharide composition analysis indicated that LBP-s-1 was comprised of rhamnose, arabinose, xylose, mannose, glucose, galactose, galacturonic acid in the molar ratio of 1.00:8.34:1.25:1.26:1.91:7.05:15.28. The preliminary structure features of LBP-s-1 were investigated by FT-IR, 1H NMR and 13C NMR. In vitro and in vivo hypoglycemic experiments showed that LBP-s-1 had significant hypoglycemic effects and insulin-sensitizing activity through increasing glucose metabolism and insulin secretion and promoting pancreatic β cell proliferation. Preliminary mechanisms were also elucidated.  相似文献

The aim of this study was to ascertain whether the Lycium barbarum polysaccharides (LBP) and healthy Qigong exercise had an effect on the prevention of atherosclerosis in the elderly. LBP was prepared and determined using High-performance liquid chromatography (HPLC). Results showed that the LBP mainly consisted of rhamnose, arabinose, ribose, mannose, galactose, fructose, xylose and glucose with a molar ratio of 1:6.78:0.21:0.63:9.34:0.51:3.11:2.82. LBP was administrated to male subjects for 3 months. Pharmacological function of LBP was further compared with Healthy Qigong exercise. On the basis of experimental result, LBP administration and healthy Qigong exercise is believed to aid in preventing atherosclerosis by the modulation of blood pressure, and by reducing plasma triglycerides, total cholesterol, and low density lipoprotein (LDL) cholesterol, elevating HDL cholesterol in the elderly. The health-promotion effect healthy Qigong exercise can be comparable to that of LBP.  相似文献

Nectar is a solution of mainly three sugars: sucrose, glucose and fructose. Studies have demonstrated that pollinators have preferences according to the sugar composition presented in their diet, and these preferences may be caused by sugar assimilation capacities. However, sugar flavor could also play an important role for sugar preferences of nectar-feeding animals. We evaluated the sugar gustatory thresholds of the broad-billed hummingbird Cynanthus latirostris for sucrose, glucose, fructose and a 1:1 mixture of glucose-fructose. We presented eight C. latirostris to paired feeders containing either a sugar solution or pure water. Additionally, we conducted sugar preference tests at three different concentrations (146, 730 and 1022 mmol L− 1), to relate sugar preferences with sugar gustatory thresholds. C. latirostris had different gustatory thresholds for the three different sugars tested. At low sugar concentrations (146 mmol L− 1), sugar selection followed the gustatory thresholds. Hummingbird sugar preference patterns can be affected by different mechanisms, both pre- and post-ingestive. At low concentrations gustatory thresholds may play an important role to determine sugar selection. However, at intermediate and high concentrations, sugar assimilation rates, and velocity of food processing generated by osmotic constraints, can be the mechanisms that explain the sugar selection of these animals.  相似文献

【目的】为了解不同枸杞产区枸杞根际真菌多样性的差异,从枸杞根际土壤真菌群落的角度试图解释宁夏枸杞优良品质与其土壤微生物菌群的联系。【方法】采用Mi Seq高通量测序方法,分别将我国4个不同地区枸杞根际土壤样品进行ITS区的高通量测序,并分析了物种组成和丰度、Alpha多样性、Beta多样性和菌群结构,测定不同产区枸杞主要有效成分,同时对枸杞品质和其土壤理化因子及枸杞根际真菌种群多样性的相关性做了分析。【结果】多糖含量和甜菜碱含量均是中宁枸杞高于兴仁和精河,格尔木含量最低;宁夏兴仁、宁夏中宁、新疆精河3个地区菌群结构类似,子囊菌门和结合菌门占总菌群的80%左右,而青海格尔木除了子囊菌门占58%外,壶菌门和新丽鞭毛菌门远远高于其他3个地区,比例占总菌群的近25%;4个地区枸杞根际土壤真菌菌群结构相似性依次为宁夏兴仁、新疆精河、宁夏中宁、青海格尔木。【结论】枸杞根际土壤样品真菌物种组成丰富,不同地区枸杞根际土壤真菌种群结构有一定差异;枸杞主要有效成分含量与枸杞根际土壤真菌种群结构具有一定相关性。  相似文献

Escherichia coli can uptake and utilize many common natural sugars to form biomass or valuable target bio-products. Carbon catabolite repression (CCR) will occur and hamper the efficient production of bio-products if E. coli strains are cultivated in a mixture of sugars containing some preferred sugar, such as glucose. Understanding the transport and metabolism mechanisms of the common and inexpensive sugars in E. coli is important for further improving the efficiency of sugar bioconversion and for reducing industrial fermentation costs using the methods of metabolic engineering, synthetic biology and systems biology. In this review, the transport and mediation mechanisms of glucose, fructose, sucrose, xylose and arabinose are discussed and summarized, and the hierarchical utilization principles of these sugars are elucidated.  相似文献

As a vital flavor compound, acetoin is extensively used in dairy products and drinks industry. In this study, Bacillus subtilis was engineered to metabolize glucose and xylose as substrates for acetoin production. Initially, gene araE from B. subtilis, encoding the xylose transport protein AraE, was placed under the control of the constitutive promoter P43 for over-expression. Batch cultures showed that 10 g/L xylose was depleted completely in 32 h. Subsequently, genes xylA and xylB from Escherichia coli, encoding xylose isomerase and xylulokinase respectively, were introduced into B. subtilis, and the recombinant turned out to assimilate glucose and xylose without preference. In shake-flask fermentations, 5.5 g/L acetoin with a yield of 0.70 mol (mol sugar)−1 was obtained by the optimum strain BSUL13 under microaerobic conditions, which offered a metabolic engineering strategy on engineering microbe as cell factory for the production of high-valued chemicals from renewable resource.  相似文献

利用13个经筛选在样品间具多态性的10碱基随机引物,对宁夏枸杞(Lycium barbarum L.)4个主要栽培品种及3个宁夏野生枸杞的总DNA进行PCR扩增。在样品间共产生123条RAPD标记带,其中多态性带68条,多态性百分比为55.28%;以Ntsyspc2.1软件计算样品间的遗传相似系数,并进行UPGMA聚类分析。结果表明宁夏枸杞栽培品种间遗传关系较近,特别是宁杞1号和宁杞2号,相似性系数达81.82%;而野生枸杞与栽培品种间遗传关系较远。  相似文献

In an outdoor mesocosm experiment of 80 weeks, the effect of nitrogen and phosphorus addition was tested on growth of Typha domingensis Presl. rhizomes in a matrix of Schoenoplectus americanus (Pers.) Volkart ex Schinz and Keller, under loading rates of 0.23 gm−2 d−1 of nitrogen, 0.17 gm−2 d−1 of P, both nutrient together and control conditions, to assess the potential expansion of T. domingensis in response to nutrient inputs.  相似文献

The seasonal dynamics of total phenolics and phenolic acids in the stems of the global invader Conyza canadensis, from March (young plants in the form of rosettes) to September (fruit abscission and the beginning of plant decline), and in sandy soil were monitored monthly in non-native areas. The highest amount of total free phenolics was found in its tissues (31,000 μg g−1) during the flowering and fruiting time (August). Bound phenolics peaked (up to 8443 μg g−1) during shoot elongation and intensive plant growth (May–June) and in September. In the stems, bound phenolic acids (p-coumaric, ferulic, p-hydroxybenzoic, vanillic and syringic) have a maximum twice, in May and in August, with ferulic acid predominating (up to 951.6 μg g−1). Free phenolic acids in the plant's tissue peaked in May (plant elongation). In the soil under C. canadensis, the amount of bound phenolics decreased between March and June, before increasing up to the full bloom phase of the plants (August). The amount of bound phenolic acids was several times greater than that of free ones, with maximum values in August. C. canadensis is a highly important source of phenolics in the ruderal phytocoenosis in new areas. In order to better explain the mechanisms of the spread and domination of invasive plants in non-native areas, in which allelopathy plays a decisive role, it is necessary to measure the production of allelochemicals in tissue and their accumulation in soil at the shortest possible intervals and link this with the phases of plant development.  相似文献

Two isomers (R,S,R,S- and R,R,S,S-) of five coordinate complex [Cu(L)Cl]+ have been separated and characterised. These two isomers have significantly different spectrochemical and electrochemical properties. Absorption maximum of R,S,R,S-[Cu(L)Cl]+ shifts to longer wavelength and its reduction potential shifts to more positive direction comparing those of R,R,S,S-[Cu(L)Cl]+. R,S,R,S-[Cu(L)Cl]+ is significantly distorted to trigonal-bipyramidal structure, whereas R,R,S,S-[Cu(L)Cl]+ retains almost square-planar geometry. The average bond distance of Cu-N in basal plane of R,S,R,S-[Cu(L)Cl]+ is longer by 0.024 Å than that of R,R,S,S-[Cu(L)Cl]+, whereas the bond distance of Cu-Cl in former is shorter by 0.200 Å than that in latter. The isolated square-planar complexes of R,R,S,S- and R,S,R,S-[Cu(L)](ClO4)2 are converted to the R,R,S,S- and R,S,R,S-[Cu(L)Cl]+ by the addition of Cl in nitromethane solution with the rate constants, k=1.70 (±0.02) and 8.31 (±0.07) M−1 s−1, respectively.  相似文献

The use of a low-intensity ultrasonic technique (noninvasive, nondestructive, on-line, and able to assess opaque samples) to monitor the kinetics of invertase hydrolysis is presented. Adiabatic compressibility has been shown to be sensitive to sugar species: ultrasonic velocity increasing as saccharose is transformed into glucose and fructose. The influence of initial sucrose mass concentration (2−60%), temperature (25−55 °C), pH (3.5−6.5), and number of microorganisms (105−109 yeasts/ml) on the reaction rate, catalyzed by the extracellular invertases of intact Saccharomyces cerevisiae cells, has been measured. The results were proven to be in strict agreement with the optimal kinetic parameters of the enzyme. Ultrasonic velocity variations are explained in terms of changes of the solute concentrations in the mixture water-saccharose-glucose/fructose and calculated from the velocity of ultrasound in the corresponding pure sugar solutions. A linear relationship between the initial rate of ultrasonic velocity and the number of yeasts (enzymes) is pointed out.  相似文献

The ability to cope with NH4+-N was studied in the littoral helophytes Phragmites australis and Glyceria maxima, species commonly occupying fertile habitats rich in NH4+ and often used in artificial wetlands. In the present study, Glyceria growth rate was reduced by 16% at 179 μM NH4+-N, and the biomass production was reduced by 47% at 3700 μM NH4+-N compared to NO3-N. Similar responses were not found in Phragmites. The amounts (mg g−1 dry wt) of starch and total non-structural carbohydrates (TNC) in rhizomes were significantly lower in NH4+ (8.9; 12.2 starch; 20.1; 41.9 TNC) compared to NO3 treated plants (28.0; 15.6 starch; 58.5; 56.3 TNC) in Phragmites and Glyceria, respectively. In addition, Glyceria showed lower amounts (mg g−1 dry wt) of soluble sugars, TNC, K+, and Mg2+ in roots under NH4+ (5.6; 14.3; 20.6; 1.9) compared to NO3 nutrition (11.6; 19.9; 37.9; 2.9, for soluble sugars, TNC, K+, and Mg2+, respectively), while root internal levels of NH4+ and Ca2+ (0.29; 4.6 mg g−1 dry wt, mean of both treatments) were only slightly affected. In Phragmites, no changes in soluble sugars, TNC, Ca2+, K+, and Mg2+ contents of roots (7.3; 14.9; 5.1; 17.3; 2.6 mg g−1 dry wt, means of both treatments) were found in response to treatments. The results, therefore, indicate a more pronounced tolerance towards high NH4+ supply in Phragmites compared to Glyceria, although the former may be susceptible to starch exhaustion in NH4+-N nutrition. In contrast, Glyceria's ability to colonize fertile habitats rich in NH4+ is probably related to the avoidance strategy due to shallow rooting or to the previously described ability to cope with high NH4+ levels when P availability is high and NO3 is also provided.  相似文献

We investigated the differential responses of invasive alien Lemna minuta and native Lemna minor to nutrient loading as well as the mechanism of competition between the species. The role of nutrients, species identity, species influence in determining the outcome of competition between the species was estimated using the Relative Growth Rate Difference (RGRD) model. The two species differed in their response to nutrient loading. The native L. minor responded indifferently to nutrient loading. The species Relative Growth Rate (RGR) was 0.10 d−1, 0.11 d−1 and 0.09 d−1 in high, medium and low nutrients, respectively. On the other hand, the invasive L. minuta responded opportunistically to high nutrient availability and had an RGR of 0.13 d−1, 0.10 d−1 and 0.08 d−1 in high, medium and low nutrients, respectively. As a result, the invasive species was dominant in high nutrient availability but lost to the native species at low nutrient availability. The invader formed approximately 60% and less than 50% of the stand final total dry biomass in high and low nutrient availability, respectively. Species RGR were reduced by both intra- and interspecific competition but intraspecific effects were stronger than interspecific effects. On the overall, the species significantly differed in their constant RGR. These differences in RGR between the species (species identity) and the differential response to nutrient loading were the main determinant of change in final biomass composition of these species in mixture. Species influence (competition) only had a small influence on the outcome of competition between the species. The observed species response to nutrient loading could be targeted in management of the invasive species. Lowering nutrients can be proposed to reduce the impact of the invasive L. minuta.  相似文献

An improved approach for simultaneous detection of Cryptosporidium parvum and Giardia lamblia (oo)cysts in soil is described. Recoveries > 70% were obtained for concentrations > 55 and 21 (oo)cysts g −1 for C. parvum and G. lamblia, respectively. The potential allelopathic effects of 14 stilbenoids and five flavonoids, isolated from leaves of Carex distachya Desf., were evaluated on the seed germination and seedling growth of three coexisting Mediterranean species (Dactylis hispanica, Petrorhagia velutina, and Phleum subulatum). The structures of the metabolites have been elucidated on the basis of their spectroscopic features (1D and 2D NMR experiments and EI–MS and ESI–MS data). The bioassays showed species-specific effects of the metabolites from C. distachya, specially on the plant growth (root and shoot elongation) which resulted significantly stimulated or inhibited at 10−4 M concentration. The effects on root elongation is generally greater than the shoot growth at all the tested concentrations (10−4–10−8 M). Cluster of biological data showed interesting relationships between the chemical structures of the compounds and their biological effects.  相似文献

Two extracellular chitinases (designated as Chi-56 and Chi-64) produced by Massilia timonae were purified by ion-exchange chromatography, ammonium sulfate precipitation, and gel-filtration chromatography. The molecular mass of Chi-56 was 56 kDa as determined by both SDS-PAGE and gel-filtration chromatography. On the other hand, Chi-64 showed a molecular mass of 64 kDa by SDS-PAGE and 28 kDa by gel-filtration chromatography suggesting that its properties may be different from those of Chi-56. The optimum temperature, optimum pH, pI, Km, and Vmax of Chi-56 were 55 °C, pH 5.0, pH 8.5, 1.1 mg mL−1, and 0.59 μmol μg−1 h−1, respectively. For Chi-64, these values were 60 °C, pH 5.0, pH 8.5, 1.3 mg mL−1, and 1.36 μmol μg−1 h−1, respectively. Both enzymes were stimulated by Mn2+ and inhibited by Hg2+, and neither showed exochitinase activity. The N-terminal sequences of Chi-56 and Chi-64 were determined to be Q-T-P-T-Y-T-A-T-L and Q-A-D-F-P-A-P-A-E, respectively.  相似文献

This study focused on effects from Monoporeia affinis reworking and ventilation activities on benthic fluxes and mineralization processes during a simulated bloom event. The importance of M. affinis density for benthic solute (O2, ΣNO2 + NO3, NH4+ and HPO42−) fluxes and sediment reactivity (mobilization of NH4+ and HPO42−) following additions of organic material to the sediment surface was experimentally investigated using sediment-water and closed sediment (jar) incubations. Three different densities of M. affinis were used to resemble a low, medium and high density situation (1300, 2500 and 6400 ind. m− 2, respectively) of a natural amphipod community. The degradation of phytodetritus (Tetraselmis sp., 5 g C m− 2) added to the sediment surface was followed over a period of 20 days. Benthic solute fluxes of O2, ΣNO2 + NO3 and NH4+ were generally progressively stimulated with increasing number of M. affinis, while no such correlation was found for HPO42−. Solute fluxes were initially enhanced 1 to 2 days after the addition of phytodetritius, caused by mineralization of the most labile organic material and a food-stimulated irrigation by the amphipods. There was no effect from the activity of M. affinis on total denitrification (Dtot = Dn + Dw) or denitrification utilizing nitrate from coupled nitrification/denitrification (Dn) for any of the densities examined. Denitrification utilizing overlying water nitrate (Dw) was only about 10% of Dtot. Dw was significantly enhanced for the highest M. affinis density investigated. The reactivity of the sediment decreased progressively with increasing density of M. affinis and with time of the experiment. However, enhanced ammonium production at least 6 days after the organic addition indicated excretion of N-containing organic compounds by M. affinis. In conclusion, large spatial and temporal variations in density of M. affinis may be of significant importance for benthic solute fluxes and overall mineralization of organic material in Baltic Sea sediments.  相似文献

Apical control is defined as the inhibition of basal axillary bud outgrowth by an upper actively growing axillary axis, whose regulation is poorly understood yet differs markedly from the better-known apical dominance. We studied the regulation of apical control by environmental factors in decapitated Rosa hybrida in order to remove the apical hormonal influence and nutrient sink. In this plant model, all the buds along the main axis have a similar morphology and are able to burst in vitro. We concentrated on the involvement of light intensity and nitrate nutrition on bud break and axillary bud elongation in the primary axis pruned above the fifth leaf of each rose bush. We observed that apical control took place in low light (92 μmol m−2 s−1), where only the 2-apical buds grew out, both in low (0.25 mM) and high (12.25 mM) nitrate. In contrast, in high light (453 μmol m−2 s−1), the apical control only operates in low nitrate while all the buds along the stem grew out when the plant was supplied with a high level of nitrate. We found a decreasing photosynthetic activity from the top to the base of the plant concomitant with a light gradient along the stem. The quantity of sucrose, fructose, glucose and starch are higher in high light conditions in leaves and stem. The expression of the sucrose transporter RhSUC2 was higher in internodes and buds in this lighting condition, suggesting an increased capacity for sucrose transport. We propose that light intensity and nitrogen availability both contribute to the establishment of apical control.  相似文献
