A peroxidase (EC 1.11.1.7) has been isolated and purified from Senna angustifolia. The enzyme was purified by ion-exchange chromatography on high Q and high S columns. SDS-PAGE electrophoresis showed that the protein has a molecular mass of approximately 70 kDa. Hydroxy-anthraquinones and hydroxy-anthracenones were evaluated as substrate of S. angustifolia and horseradish peroxidases. Both peroxidases catalyzed the oxidation of alizarin and purpurin anthraquinones to the corresponding 3,3'-bializarin and the new compound 3,3'-bipurpurin, respectively, as well as the formation of 2,2'-biquinizarin from quinizarin anthracenone. The K(Mapp) and V(max) values for alizarin and purpurin were 97 and 95 microM, and 1.5 and 2.1 microM min(-1) mg prot(-1), respectively. The results suggest that peroxidase may participate in the biogenesis of anthraquinones.  相似文献

Seven 9,10-seco-9,19-cyclolanostane arabinosides, named podocarpasides A-G (1-7), were isolated from the roots of Actaea podocarpa DC., a species closely related to black cohosh (a well known dietary supplement). Their structures were determined with the help of spectroscopic data including extensive 2D NMR spectroscopy. The isolates were found inactive, when tested for cytotoxic, estrogenic, and antioxidant activities in cell based assays. They were also tested for anticomplement activity against the classical pathway of complement system and only podocarpaside C (3) inhibited modest complement activity with an IC50 value of 200 microM.  相似文献

Phytochemical studies on the branches and leaves of Lyonia ovalifolia yielded a new grayanane diterpenoid, lyonin A ( 1 ), together with two known compounds, secorhodomollolides A and D ( 2 and 3 , resp.). The structure of 1 was elucidated by combination of 1D‐ and 2D‐NMR, and MS analyses. Compound 1 turned out to be a new, highly O‐acylated grayanane diterpenoid, of which ring B has undergone an oxidative cleavage between C(9) and C(10), yielding a system differing from the previously reported grayanane type with a 5/7/6/5 ring system. Results of the cAMP regulation activity assay showed that compounds 2 and 3 at 50 μM induced a significantly decreased cAMP level in N1E‐115 neuroblastoma cells (p<0.001), indicating neuropharmacological potential.  相似文献

The donor-free 9,10-dialkyl-9,10-dihydro-9,10-digallaanthracenes 1 (alkyl: methyl) and 2 (alkyl: ethyl) were prepared by reaction of 1,2-di(chloromercurio)benzene with the corresponding trialkylgallium and isolated as colourless air- and moisture sensitive crystalline compounds. In the solid-state structure of 1, two slightly different monomers 1A and 1B are found, which form a dimer 1A?1B held together by “medium” strong gallium arene π-interactions. Further weak π-interactions between 1A and 1A and 1B and 1B constitute a one-dimensional coordination polymer containing strands of the composition [?(1A?1B)?(1B?1A)?]n. In contrast, compound 2 crystallizes in the form of distinct molecular units without any further intermolecular π-interactions. The molecular units possess D2d symmetry and are built by strong π-interactions between two digallaanthracene monomers. Two symmetrical aryl group bridges between two gallium atoms are observed for the first time in the subunits of 2. By addition of a Lewis-base (THF, Pyridine) to 2, a monomeric planar digallaanthracene framework is restored, as proven by an X-ray crystal structure analysis of 2 · 2Py. The different structures of 1 and 2 are explained on the basis of steric effects.  相似文献

The Australian Thrasorinae are revised and Mikeius is transferred to Mikeiinae Paretas-Martínez & Pujade-Villar, subfam. n., and Mikeius clavatus Pujade-Villar & Restrepo-Ortiz, sp. n., is described. Two new genera of Thrasorinae are erected: Cicatrix Paretas-Martínez, gen. n., including Cicatrix pilosiscutum(Girault), comb. n. from Amblynotus, Cicatrix schauffi (Buffington), comb. n. from Mikeius, and Cicatrix neumannoides Paretas-Martínez & Restrepo-Ortiz, sp. n.; and Palmiriella Pujade-Villar & Paretas-Martínez, gen. n., including Palmiriella neumanni (Buffington), comb. n. from Mikeius, Thrasorus rieki Paretas-Martínez & Pujade-Villar, sp. n., is also described. A phylogenetic analysis of 176 morphological and biological characters, including all these new taxa and all genera previously included in Thrasorinae, was conducted. All subfamilies were recovered as monophyletic, with the following relationships: Parnipinae (Euceroptrinae (Mikeiinae (Plectocynipinae (Thrasorinae)))). A worldwide key to the subfamilies of Figitidae is provided that includes the new subfamily, as well as a key to genera Thrasorinae.  相似文献

A new synthesized acridine derivative, 3,6-diamino-10-methyl-9, 10-dihydroacridine (AcrH), was tested for in vitro reverse mutations with Salmonella TA strains, chromosome aberrations and sister chromatid exchanges (SCE) in human lymphocytes, and for in vivo chromosome aberrations in bone marrow of mice. Using the classic plate incorporation method, mutagenicity of AcrH in bacterial cells (TA97a, TA98, TA100 and TA102) was observed in the experiments performed with, and without, rat liver S9 metabolic activation. The reverse mutation assay showed no difference in mutagenic activity between AcrH and acriflavine (Acr(+)) in the test with TA97. The results of in vitro chromosome aberrations assay revealed potential clastogenicity. The test using macroculture of human lymphocytes induced mainly chromatid gaps. The experiments with human lymphocytes revealed SCE-inducing effect of AcrH and Acr(+). In an in vivo study, AcrH given intraperitoneally to Balb/c mice did not cause any significant increase in the percentage of cells with aberrations compared to the negative control.  相似文献

Tetraspanins are animal proteins involved in membrane complexes that are involved in cell adhesion, differentiation, and motility. The PLS1 gene from rice blast fungus Magnaporthe grisea encodes a protein (Pls1p) structurally related to tetraspanins that is required for pathogenicity. In Botrytis cinerea public sequences, we identified an EST homologous to PLS1. Using degenerated oligonucleotides, we amplified sequences homologous to PLS1 in fungi Colletotrichum lindemuthianum and Neurospora crassa. Analysis of N. crassa and M. grisea genome sequences revealed the presence of a single tetraspanin gene. Thus, fungi differ from animals, which contain between 20 and 37 paralogous tetraspanin genes. Fungal proteins encoded by BcPLS1, ClPLS1, and NcPLS1 display all the structural hallmarks of tetraspanins (predicted topology with four transmembrane domains, extra- and intracellular loops; conserved cysteine-based patterns in second extracellular loop). Phylogenetic analysis suggests that these genes define a new family of orthologous genes encoding fungal-specific tetraspanins.  相似文献

Three new species of Aizoaceae from the Western Cape are described. Octopoma tanquanum Klak and Vlokia montana Klak are dwarf shrubby or mat-forming succulents which belong to the Ruschieae in the Ruschioideae. Mesembryanthemum knolfonteinense Klak of the Mesembryanthemoideae is a geophyte. Octopoma tanquanum inhabits relatively low lying arid areas within the Tanqua Karoo and the Little Karoo and is thought to be closely allied to the two species of Octopoma found in the Little Karoo. Both V. montana and M. knolfonteinense grow at relatively high altitudes within the fynbos biome. In addition, Ruschia littlewoodii L.Bolus is transferred to Phiambolia, and two new combinations are made in Antimima for Ruschia hexamera L.Bolus and Ruschia radicans L.Bolus.  相似文献

A thermal retro-Diels-Alder decomposition of N-hydroxyurea-derived acyl nitroso compounds and 9,10-dimethylanthracene cycloadducts followed by acyl nitroso compound hydrolysis produces nitrous oxide, evidence for the formation of nitroxyl, the one-electron reduced form of nitric oxide that has drawn considerable attention for its potential roles in biological systems. EPR and NMR spectroscopy provide further evidence for nitroxyl formation and kinetic information, respectively. Such compounds may prove to be useful alternative nitroxyl donors.  相似文献

Molecular phylogenetic evidence indicates that the octocoral family Alcyoniidae is highly polyphyletic, with genera distributed across Octocorallia in more than 10 separate clades. Most alcyoniid taxa belong to the large and poorly resolved Holaxonia–Alcyoniina clade of octocorals, but members of at least four genera of Alcyoniidae fall outside of that group. As a first step towards revision of the family, we describe a new genus, Parasphaerasclera gen. n., and family, Parasphaerascleridae fam. n., of Alcyonacea to accommodate species of Eleutherobia Pütter, 1900 and Alcyonium Linnaeus, 1758 that have digitiform to digitate or lobate growth forms, completely lack sclerites in the polyps, and have radiates or spheroidal sclerites in the colony surface and interior. Parasphaerascleridae fam. n. constitutes a well-supported clade that is phylogenetically distinct from all other octocoral taxa. We also describe a new genus of Alcyoniidae, Sphaerasclera gen. n., for a species of Eleutherobia with a unique capitate growth form. Sphaerasclera gen. n. is a member of the Anthomastus–Corallium clade of octocorals, but is morphologically and genetically distinct from Anthomastus Verrill, 1878 and Paraminabea Williams & Alderslade, 1999, two similar but dimorphic genera of Alcyoniidae that are its sister taxa. In addition, we have re-assigned two species of Eleutherobia that have clavate to capitate growth forms, polyp sclerites arranged to form a collaret and points, and spindles in the colony interior to Alcyonium, a move that is supported by both morphological and molecular phylogenetic evidence.  相似文献

Coccidia provide a rich hunting ground for drug-designers, as there are significant biochemical differences between the parasites and their hosts. Recent years have brought the discovery of the plastid and its possible metabolic machinery, characterisation of acidocalcisomes, reports on the apparent absence from some coccidia of a typical mitochondrion, and the discovery of the mannitol cycle and shikimate pathway in the parasites. Moreover, modern technologies such as genomics and proteomics are bringing new insights into the biochemistry of coccidia and highlighting possible drug targets in abundance. A major issue for would-be drug discoverers is to decide upon the targets to prioritise. This review provides an update on recent findings on how coccidia differ biochemically from vertebrates. It includes discoveries within coccidian parasites themselves but also uses findings in Plasmodium to provide an overview of biochemical features that may be characteristics of many apicomplexan parasites and so potential targets for broad-spectrum drugs.  相似文献

From the roots of various Stemona species four new dehydrotocopherols (chromenols) were isolated and their structures and stereochemistry elucidated by spectroscopic methods. The double bond between C-3 and C-4 proved to be a typical chemical character of the genus found in most of the species. Various C-methylations of the aromatic ring reflect differences in methyltransferase activities and agreed with the current species delimitations showing an exclusive accumulation of dehydro-delta-tocopherol for the Stemona tuberosa group, whereas different provenances of Stemona curtisii were characterized by dehydro-gamma-tocopherol accompanied by small amounts of dehydro-alpha-tocopherol. From Stemona collinsae all four tocopherols were isolated with a clear preponderance of dehydro-delta-tocopherol accompanied by smaller amounts of the rare dehydro-beta-tocopherol. Stemona burkillii and a group of unidentified species showed a weak accumulation trend towards dehydro-alpha-tocopherol, whereas Stemona cochinchinensis and especially Stemona kerrii clearly differed by a preponderance of chromanol derivatives. In Stemona cf. pierrei no tocopherols could be detected at all. Based on TLC tests and microplate assays with the free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH*) the antioxidant capacities of all chromenol derivatives were comparable with that of alpha-tocopherol showing no significant differences among each other, except for a more rapid kinetic behaviour of the 5,7,8-methylated dehydro-alpha-tocopherol.  相似文献

Two new species and two new genera (Pachyphasma, Striatophasma) of Mantophasmatodea are described from Namibia. Pachyphasma brandbergense is endemic to the Brandberg massif; Striatophasma occupies an extensive area south of the region inhabited by Mantophasma. Phylogenetic analyses (see Predel et al. in press) suggest a sistergroup relationship of Striatophasma and the South African Austrophasmatidae.  相似文献

1. The main products of the metabolism of 7,12-dimethylbenz[a]anthracene by rat-liver homogenates are the isomeric monohydroxymethyl derivatives. The syntheses of these compounds are described. 2. Two phenolic products and two dihydrodihydroxy compounds were formed, but none of these appeared to have been formed by hydroxylation at the `K region'. There was little evidence for the formation of a glutathione conjugate of the hydrocarbon. 3. The monohydroxymethyl derivatives are products of the hydroxylation of the hydrocarbon in the ascorbic acid–Fe2+–oxygen model hydroxylating system.  相似文献

The availability of the dnaJ1 gene for identifying Mycobacterium species was examined by analyzing the complete dnaJ1 sequences (approximately 1200 bp) of 56 species (54 of them were type strains) and comparing sequence homologies with those of the 16S rRNA gene and other housekeeping genes (rpoB, hsp65). Among the 56 Mycobacterium species, the mean sequence similarity of the dnaJ1 gene (80.4%) was significantly less than that of the 16S rRNA, rpoB and hsp65 genes (96.6%, 91.3% and 91.1%, respectively), indicating a high discriminatory power of the dnaJ1 gene. Seventy-one clinical isolates were correctly clustered to the corresponding type strains, showing isolates belonging to the same species. In order to propose a method for strain identification, we identified an area with a high degree of polymorphism, bordered by conserved sequences, that can be used as universal primers for PCR amplification and sequencing. The sequence of this fragment (approximately 350 bp) allows accurate species identification and may be used as a new tool for the identification of Mycobacterium species.  相似文献
