Transmission of chromosomes 5R via gametes and its effect on somatic embryoidogenesis have been studied with the use of the model 5R(5A) substitution line L2837 = L503/Secale cereale L., cultivar Saratovskaya 5//L503, where L503 is a cultivar of spring bread wheat. It has been found that the frequencies of transmission of univalent chromosomes 5R and 5A determined in experiments on F1 reciprocal hybrids with cultivar Saratovskaya 29 do not reflect their frequencies in the self-pollinated offspring of F1 hybrids; the frequency of transmission of chromosomes 5R and 5A depends on the genotypes of both the recipient cultivar and the donor rye cultivar; and the 5R(5A) substitution in cultivar L503 significantly increases the parameters of somatic embryoidogenesis in vitro in explants from inflorescences.  相似文献

The effect of individual rye chromosomes on the induction of callus and the character of its regenerating capacity was studied with cultured immature embryos of wheat-rye (Triticum aestivum L. cv. Saratovskaya 29-Secale cereale L. cv. Onokhoiskaya) substitution lines. The genotypic diversity of the substitution lines proved to significantly affect variation of parameters characterizing the major types of callus cultures, that is, frequencies of embryogenic calli, which are capable of shoot regeneration, and of morphogenic calli, which produce root structures. Functioning in the genotypic background of common wheat cultivar Saratovskaya, chromosomes 2R and 3R of rye cultivar Onokhoiskaya stimulated significantly the induction of embryogenic callus highly capable of shoot regeneration. Rye chromosome 2R present in place of chromosome 2D in the common wheat genome suppressed the induction of callus producing root structures. Rye chromosomes 1R and 6R suppressed the induction of embryogenic callus capable of shoot regeneration.  相似文献

Leaf-rust resistance and androgenesis were studied in the anther cultures of Triticum aestivum L., which included Saratovskaya 29 cultivar, the isogenic line Ps29, and three F1 hybrids (L503/S55, L504/S58, ATS7/L1063) with 7DS-7DL-7Ae#1L translocation of Lr19 gene (Lr19 translocation) from Agropyron elongatum (Host) P.B. The Lr19 translocation was shown to affect the induction of embryogenesis and green plant regeneration. The frequencies of Lr19 translocation differed in F2 hybrids obtained by traditional hybridization and in sets of DH lines obtained in F1 anther cultures derived from the same combinations of T. aestivum parental forms. The number of leaf-rust resistant genotypes tended to decrease. The frequency of Lr19 translocation in the set of DH3 lines derived from F1 L504/S58 was significantly lower than in other sets of DH3 lines and F2 hybrid populations.  相似文献

The effect of rye chromosomes on polyembryony was studied for reciprocal hybrid combinations between (Hordeum vulgare L.)-Triticum aestivum L. alloplasmic recombinant lines and five wheat T. aestivum L. (cultivar Saratovskaya 29)-rye Secale cereale L. (cultivar Onokhoiskaya) substitution lines: IR(1D), 2R(2D), 3R(3B), 5R(5A), and 6R(6A), and for direct hybrid combinations between the [H. marinum ssp. gussoneanum (H. geniculatum All.)]-T. aestivum alloplasmic recombinant line and the wheat-rye substitution lines 1R (1A), 1R (1D), and 3R(3B). Chromosomes 1R and 3R of rye cultivar Onokhoiskaya proved to affect the expression of polyembryony in the hybrid combinations that involved the alloplasmic recombinant lines of common wheat as maternal genotypes. Based on this finding, polyembryony was regarded as a phenotypic expression of nuclear-cytoplasmic interactions where an important role is played by rye chromosomes 1R and 3R and the H. vulgare cytoplasm. Consideration is given to the association between the effect of rye chromosomes 1R and 3R on polyembryony in the [(Hordeum)-T. aestivum x wheat-rye substitution lines] hybrid combinations and their stimulating effect on the development on angrogenic embryoids in isolated anther cultures of the wheat-rye substitution lines.  相似文献

The role of individual chromosomes of rye in the manifestation of crossability and seedling development in hybrid combinations between common barley Hordeum vulgare L., cultivar Nepolegayushchii (2n = 14) and five wheat-rye substitution lines Triticum aestivum L., cultivar Saratovskaya 29/Secale cereale L., cultivar Onokhoiskaya (2n = 40 wheat + 2 rye chromosomes). Crossability, which was measured by two parameters--frequency of set grains and frequency of grains with embryos--was shown to be significantly affected by each of the five rye chromosomes examined: 1R, 2R, 3R, 5R, and 6R; the development of barley haploids was affected by rye chromosomes 1 R, 3R, and 5R. We were the first to demonstrate that polyembryony could be induced by mutual effects of barley cytoplasm and rye chromosome 1R. Possible mechanisms controlling the development of haploids and twins in hybrid combinations H. vulgare x T. aestivum/S. cereale are discussed. The conclusion is drawn that hybrid combinations between common barley and wheat-rye substitution lines can serve as new models for studying incompatibility mechanisms in distant crosses and genetic control of parthenogenesis.  相似文献

Genetic regulation of grain hardness and protein content in intervarietal substitution lines for chromosomes of homeologous group 5 was examined. Common wheat cultivar Saratovskaya 29 with high bread-backing properties served as the recipient. Donors of chromosomes 5A and 5D were 18 cultivars with variable traits examined, including high-protein cultivars (Atlas 66 and Diamant 2), and soft-grain cultivars (Ul’yanovka and Chinese Spring). Analysis of substitution lines pointed to a substantial effect of chromosome 5D on the regulation of both traits. It was demonstrated that as a result of intervarietal substitution for chromosome 5D from donor cultivars Ul’yanovka and Chinese Spring, the endosperm softness was increased compared to the recipient cultivar Saratovskaya 29. Substitution lines Saratovskaya 29/Atlas 66 5D and Saratovskaya 29/Diamant 2 5D were characterized by high grain protein content, as well as by high endosperm hardness. In addition, the line Saratovskaya 29/Novosibirskaya 67 5D, characterized by grain hardness higher than in Saratovskaya 29, was isolated. In the lines with intervarietal substitution of chromosome 5A, grain protein content was found to be lower than in recipient cultivar Saratovskaya 29.  相似文献

The characteristic features of androgenesis in six wheat–rye substitution lines Triticum aestivumL. (cv. Saratovskaya 29)/Secale cerealeL. (cv. Onokhoiskaya) and triticale (2n= 56) using anther culture at different concentrations of 2,4-D in the growth medium were studied. Under variable cultivation conditions, the significant effect of genotypic diversity on the variability of such androgenesis parameters as the frequency of productive anthers, the frequency of embryoid formation, and the frequency of total regenerated plantlets, was shown. It was demonstrated that chromosomes 1R, 3R, and 7R stimulated the formation of androgenous embryoids, while chromosome 5R produced an opposite effect. In triticale and substitution lines, the regeneration ability of androgenous embryoids induced by elevated 2.4-D concentrations was inhibited. Chromosome 1R of the Onokhoiskaya cultivar was suggested to contain genes suppressing regeneration of green plantlets, while chromosome 3R, conversely, stimulated their formation. Chromosomes 1R, 2R, 3R, and 7R of the Onokhoiskaya cultivar did not inhibit the spontaneous formation of androgenous hexaploids in the substitution lines.  相似文献

Regulation of meiotic restitution in androgenic haploids generated by cultivation of isolated anthers of three wheat-rye substitution lines 2R(2D)1, 2R(2D)3, and 6R(6A) (Triticum aestivum L., cultivar Saratovskaya 29/Secale cereale L., cultivar Onokhoiskaya) was studied. The presence of rye chromosomes and the absence of homeologous wheat chromosomes in the haploid plant genome was shown to cause meiotic restitution, as observed in the case of androgenic haploids 6R(6A), or to inhibit it—in meiosis of haploids 2R(2D)1 and 2R(2D)3. In haploids of lines 2R(2D)1 and 2R(2D)3, the reductional type of division of univalent chromosomes was observed, leading to preferential formation of tetrads. In haploids of line 6R(6A), the equational type of division of univalents into sister chromatids, resulting in the block of the second division and formation of diads in approximately 50% of cells, was detected. These results confirm data on the effect of the genotype of line 2R(2D)1 on the induction of reductional type division of univalents and two-phase meiosis, which were earlier obtained in studies of meiosis in polyhaploids 2R(2D)1 × S. cereale L., cultivar Onokhoiskaya.  相似文献

It was demonstrated that the substitution of individual chromosomes of the recipient cultivar Saratovskaya 29 with the homologous chromosomes of the donor cultivar Janetzkis Probat caused significant changes in lipoxygenase activity and several other quality characteristics. The correlations between the lipoxygenase activity and the parameters of physical dough properties were determined. Three molecular forms of lipoxygenase (Lpx-1, Lpx-2, and Lpx-3), differing in the value of surface charge and enzymatic activity, were detected by native PAGE.  相似文献

Polymorphism of heterochromatin C-blocks in chromosomes of rye genome has been studied in the F1BC1 hybrids and the D/A substitution lines of rye-wheat amphidiploids (Verasen x L374, Novosibirskaya x x L246)--secalotriticum for revealing cytogenetic markers of rye chromosomes. An increase in polymorphism for the presence and value of heterochromatin blocks in chromosomes of rye genome was shown in the F1BC1 hybrids ((Verasen x L374) x L145 x L145) and unstable genotypes of the F6BC1 chromosome substitution lines (line 118 (Novosibirskaya x L246) x Reso x Reso), line 104 ((Verasen x x L374) x Garmoniya x Garmoniya) that was related to activation of mobile genetic elements present in cereals. Heterochromatin markers of all seven chromosomes in rye genome were revealed. The polymorphism system of heterochromatin blocks may serve as a marker for specificity of the linear chromosome structure during reconstruction of synthetic cereal genomes, and as a test for cytological and morphogenetic stability of hybrid polygenomes in a series of generations.  相似文献

A gene determining reduced height, Rht10, from the wheat cultivar Ai-Bian 1 was introgressed into the triticale genotype. Initially, Ai-Bian 1 was crossed with the wheat cultivar Chinese Spring (CS), a carrier of Kr genes, to overcome the uncrossability of this cultivar with rye. Amphidiploids were produced by hybridizing the F2 (CS x Ai-Bian 1) plants displaying reduced height (at the level of Ai-Bian 1) with rye. Free pollination of F1 (F2 of CS x Ai-Bian 1) x Saratovskaya 7 with triticale pollen gave fertile viable hybrids; the majority of hybrids were phenotypically closer to octoploid triticale; however, the variants intermediate between octo- and hexaploids were also present. The height of amphidiploids varied from 40 to 90 cm, and the grain yield per spike amounted on the average to 11.7--24.7 grains, which exceeded essentially this value in F1 plants.  相似文献

The effects of rye chromosomes 1R and 5R on androgenesis in cultured anthers of wheat–rye substitution lines was studied as dependent on the cultivar origin of the rye chromosomes and on the wheat genome (A or D) subjected to substitution. Chromosome 1R stimulated embryogenesis in anther cultures, while chromosome 5R suppressed it regardless of whether the corresponding wheat chromosomes were substituted in the A or D genome. The effect of chromosome 1R on embryogenesis proved to depend on its cultivar origin. Along with rye chromosome 1R, wheat chromosome 1A was shown to substantially affect total seedling regeneration. Regeneration of green seedlings was dramatically affected both by rye chromosome 1R and by wheat chromosome 1D. The results supported the published data that individual androgenesis parameters (embryogenesis, total plant regeneration, green plant regeneration) are controlled by different genetic mechanisms.  相似文献

Summary The substitution patterns of rye chromosomes in hexaploid triticale × wheat F2 hybrids, along with the transmission patterns of rye chromosomes through egg cells and pollen when several of the F1 hybrids were test crossed to triticale and wheat were investigated. The data indicated that the rye chromosome transmission through both the egg and pollen was random in number and in composition. The test crosses suggested that it was best to use wheat pollen for the transmission of rye chromosomes through the egg cells of the F1 hybrids and triticale egg cells for the transmission of rye chromosomes through F1 hybrid pollen. A deviation from random segregation in the F2 and the transmission rate was observed for rye chromosomes 1R, 4R/7R, and 6R. The transmission rates of 1R and 6R varied depending on the direction in which the cross was made. The results also indicated that there was little or no compensation between the R- and D-genomes and that the chromosomes of these two genomes appeared to be transmitted independently of each other.  相似文献

Summary Two somatic hybrid plants generated from a single fusion event between Lycopersicon esculentum and irradiated L. pennellii protoplasts have been analyzed at the molecular level. Over 30 loci have been analyzed using isozymes and RFLPs. All loci tested on chromosomes 2–10 were heterozygous, while those loci on chromosome 12 were homozygous L. pennellii in both somatic hybrids. In one of the somatic hybrids, 2850, loci on chromosome 1 were also homozygous L. pennellii. The other somatic hybrid, 28F5, was heterozygous at all chromosome 1 loci tested, but exhibited altered stoichiometry of parental bands as compared to the sexual hybrid. Loci on chromosome 2 from both somatic hybrids have altered stoichiometry, with L. pennellii alleles being four times more abundant than expected. Both somatic hybrids contain the L. esculentum chloroplast genome, while only L. pennellii polymorphisms have been detected in the mitochondrial genome.  相似文献
