A survey of entomopathogenic fungi associated with Hylastes ater and Hylurgus ligniperda was undertaken in six Pinus radiata forest sites in New Zealand. In each site soil, bark and frass associated with beetle galleries were sampled, as well as a selection of live beetles, larvae and mycosed cadavers. The density of Beauveria spp. (as measured by colony forming units) varied both within and between the sites and substrates sampled. The highest CFU density detected was 4 × 106 CFU/g dry frass, which was collected from a stump at the Mahurangi site in the North Island. A number of isolates were selected and characterized using PCR amplification of a terminal region of the EF1-α fragment. This analysis revealed that two Beauveria species, Beauveria caledonica and Beauveria bassiana, were commonly associated with bark beetle habitat. B. caledonica was the only species isolated from mycosed beetles, and was also recovered from soils, bark and frass, though not live insects. B. bassiana was also found in soils, bark, frass and live insects; however, it was not isolated from mycosed cadavers in this study. It is speculated that B. caledonica is the common ‘natural’ pathogen of bark beetles in their breeding habitat. A third species, Beauveria malawiensis, was isolated once from soil. This is the first report of this species from New Zealand. Thirteen isolates (selected from all substrates sampled and live beetles) were selected for laboratory bioassays. Under laboratory conditions all isolates were pathogenic to H. ater and H. ligniperda adults; there was little difference among the isolates in terms of efficacy despite the paucity of B. bassiana infection observed in the field. The mechanisms behind the lack of field infection observed for B. bassiana are not known, but are likely to be linked to limiting environmental factors in the breeding sites. These results indicate potential risks associated with the selection of strains for use in biological control programmes solely on the basis of laboratory bioassays, especially if the mode of action and biology of the species used are not fully understood. The importance of selecting strains which are environmentally competent is evident. Overall, however, this study demonstrates that entomopathogenic fungi can play a significant role in the regulation of H. ater and H. ligniperda populations in pine plantations.  相似文献

Hylastes ater and Hylurgus ligniperda are introduced pests of re-established Pinus radiata in New Zealand. Both species breed under the bark of stumps in recently harvested areas. Adult maturation feeding on pine seedlings planted in adjacent areas can significantly impact seedling growth, and in severe cases seedlings will die. Entomopathogenic fungi are important natural mortality factors in bark beetle populations, and Beauveria spp. are predominant. Here, we report on the isolation of other fungal species from H. ater in New Zealand. Based on morphological characteristics and sequencing data, two species, Metarhizium flavoviride var. pemphigi and Hirsutella guignardii, were recovered from H. ater. Both are new records for New Zealand and appear to be the first records of these species from bark beetles worldwide.  相似文献

Flower visitors on 12 species of New CaledonianWinteraceae were studied. The visitors were two species of ancestral moths (Sabatinca; Micropterigidae), three species of weevils (Palontus; Curculionidae), and a species of thrips. Behavior observations and pollen records suggest that the beetles and occasionally the moths serve as pollinators ofZygogynum and Exospermum, andBelliolum is pollinated primarily by thrips. The floral volatiles are simple in composition, usually dominated by short esters. Preliminary experiments showed that ethyl acetate elicited alighting, and a distinctive huddling behavior was elicited by artificially mixed fragrance. The host associations of otherSabatinca andPalontus spp. do not support the hypothesis that theWinteraceae have radiated in association with their pollinators. The available evidence supports the notion of opportunistic isolated host colonizations at some point in the radiation of the pollinator groups.  相似文献

Twenty isolates of four species of entomopathogenic hypocrealean fungi (Beauveria bassiana, Beauveria brongniartii, Isaria farinosa, and Metarhizium anisopliae) were found to be pathogenic to adults of the Asian longhorned beetle, Anoplophora glabripennis. Survival times for 50% of the beetles tested (ST50) ranged from 5.0 (M. anisopliae ARSEF 7234 and B. brongniartii ARSEF 6827) to 24.5 (I. farinosa ARSEF 8411) days. Screening studies initially included strains of B. brongniartii, which is registered as a microbial control agent in Europe, Asia and South America but not in North America. At that time, we could not confirm that this fungal species is native to North America which added uncertainty regarding future registration of this species for pest control in the USA. Therefore, subsequent bioassays documented median survival times for three M. anisopliae isolates (5–6 days to death) and two of these isolates are suggested for further development because they are already registered for pest control in the USA. Summary Feeding relationships of adultEuparia castanea Serville andMartinezia dutertrei Chalumeau with their ant hosts were studied in the laboratory using the radioactive tracer32P.Euparia castanea was tested withSolenopsis geminata (F.),Martinezia dutertrei Chalumeau was tested withS. invicta Buren,S. richteri Forel, andS. geminata. Unlabeled beetles were exposed to various radioisotope labeled conditions for 24 hr and then checked for acquired radioactivity. In whole colony tests, both species of beetles acquired radioactivity.M. dutertrei obtained food from live ants, butE. castanea did not. Both species of beetles ate ant larvae.E. castanea also obtained food from ant larvae by strigilation. Neither species of beetle fed on ant feces or other secretions on the substrate. Both species of beetles obtained food by strigilation from fresh and decomposed worker ant cadavers.M. dutertrei also ate both kinds of ant cadavers. Both species of beetles also ate dead house flies, indicative of scavenging or feeding on ant booty.Martinezia dutertrei showed no preference for any particular ant species. Ants did not obtain food by trophallaxis or glandular secretion from either species of beetle. A number of species in the plant pathogen genus Armillaria are known from South America where they cause root rot disease on a wide variety of hosts. Knowledge pertaining to phylogenetic relationships of these species with those of other Armillaria species is almost non-existent. In addition, very few cultures representing these species are available, making DNA-based phylogenetic analyses impossible. The aim of this study was to characterise a collection of Armillaria isolates from the Patagonian Andes using DNA sequences and to determine their phylogenetic relationships with other Armillaria species. DNA sequences were obtained from the internal transcribed regions (ITS1, 5.8S and ITS4) and ribosomal large subunit (LSU) gene and used in phylogenetic analyses. Phylogenetic trees generated from the sequences separated the Armillaria isolates into four lineages. Lineages I and II represented A. novae-zelandiae and A. luteobubalina, respectively. Isolates belonging to A. novae-zelandiae from Malaysia, New Zealand, Australia and South America showed considerable intra-clade sub-structure. Lineages III and IV are probably distinct species and are most closely related to A. hinnulea and an unnamed species isolated from New Zealand and Kenya. This is the first comprehensive study of the phylogenetic relationships of Armillaria species from Patagonia and it provides a foundation for future research in this region.  相似文献

The cassava green mite (CGM), Mononychellus tanajoa, a native of South America was accidentally introduced into Africa where it causes serious crop losses. The possibility of introducing classical biological agents from the native home of CGM into Africa was investigated. Thus, we conducted a series of laboratory assays of the native fungal pathogens, Neozygites tanajoae from Brazil and Neozygites floridana from Colombia and Brazil, and compared them with N. tanajoae isolates from Benin. Infectivity of both fungal species, was assayed against the twospotted spider mite, Tetranychus urticae, and against the red mite, Oligonychus gossypii. Pathogenicity against CGM and host range studies were conducted by transferring adult females of each mite species to leaf discs containing sporulated cadavers with a halo of conidia of each fungal isolate. All isolates caused some degree of infectivity to CGM. None of the isolates of N. floridana and N. tanajoae tested were pathogenic to O. gossypii, and only two isolates infected T. urticae. Most isolates from Brazil were highly virulent and infected only CGM. Sixteen N. tanajoae isolates caused more than 89% mortality and more than 62% of the CGM became mummified. A mummified CGM is characteristically a swollen, brown fungus-killed mite that has great potential to produce conidia. However, high mortality was not always associated with high mummification. The median mummification time ranged from 4.4 to 6.7 days. Five Brazilian isolates caused >75% mummification with a median mummification time <5 days. Isolates that cause high mummification in a short period of time would be more likely to cause epizootics and to establish in the new environment. Species in the fungal genus Beauveria are pathogens of invertebrates and have been commonly used as the active agent in biopesticides. After many decades with few species described, recent molecular approaches to classification have led to over 25 species now delimited. Little attention has been given to the mitochondrial genomes of Beauveria but better understanding may led to insights into the nature of species and evolution in this important genus. In this study, we sequenced the mitochondrial genomes of four new strains belonging to Beauveria bassiana, Beauveria caledonica and Beauveria malawiensis, and compared them to existing mitochondrial sequences of related fungi. The mitochondrial genomes of Beauveria ranged widely from 28,806 to 44,135 base pairs, with intron insertions accounting for most size variation and up to 39% (B. malawiensis) of the mitochondrial length due to introns in genes. Gene order of the common mitochondrial genes did not vary among the Beauveria sequences, but variation was observed in the number of transfer ribonucleic acid genes. Although phylogenetic analysis using whole mitochondrial genomes showed, unsurprisingly, that B. bassiana isolates were the most closely related to each other, mitochondrial codon usage suggested that some B. bassiana isolates were more similar to B. malawiensis and B. caledonica than the other B. bassiana isolates analyzed.  相似文献

Invasive species are a serious threat to biodiversity worldwide. The relatively simple ecological systems of the subantarctic have the potential to be significantly damaged by predatory species that invade. Two species of exotic, predatory, terrestrial flatworms were first collected in 1997 from two localities only 2 km apart, in the southeast of subantarctic Macquarie Island. The species were later identified as Kontikia andersoni and Arthurdendyus vegrandis. We report here the results of fieldwork in 2004 that established that both species now occupy about a seventh of the southeast of the island which has a total area of only 170 km2 and that there seem to be no barriers to further expansion. The island was first discovered in 1810 and so it is likely the species were introduced by means of human intervention within the last 200 years. We provide evidence to show that both species originated in New Zealand and have probably been on the island for ∼100 years giving an average rate of spread of about 10 m per year. Other species of Arthurdendyus have been introduced from New Zealand to the United Kingdom where they prey on earthworms. The quarantine significance of A. vegrandis for Australia is discussed and recommendations made to reduce the probability of it entering Tasmania where it has the potential to become an agricultural pest.  相似文献

The best-known interaction between bacteria and plants is the Rhizobium-legume symbiosis, but other bacteria–plant interactions exist, such as between Burkholderia and Rubiaceae (coffee family). A number of bacterial endophytes in Rubiaceae are closely related to the soil bacterium Burkholderia caledonica. This intriguing observation is explored by investigating isolates from different geographic regions (Western Europe vs. sub-Saharan Africa) and from different niches (free-living bacteria in soil vs. endophytic bacteria in host plants). The multilocus sequence analysis shows five clades, of which clade 1 with two basal isolates deviates from the rest and is therefore not considered further. All other isolates belong to the species B. caledonica, but two genetically different groups are identified. Group A holds only European isolates and group B holds isolates from Africa, with the exception of one European isolate. Although the European and African isolates are considered one species, some degree of genetic differentiation is evident. Endophytic isolates of B. caledonica are found in certain members of African Rubiaceae, but only in group B. Within this group, the endophytes cannot be distinguished from the soil isolates, which indicates a possible exchange of bacteria between soil and host plant.  相似文献

An overview of carabid beetles in the New Zealand conservation context is provided. Processes for threatened species recovery within the New Zealand Department of Conservation are outlined, and the Department’s two major beetle-related recovery documents (The Conservation Requirements for New Zealand’s Nationally Threatened Invertebrates, and The Carabid Beetle Recovery Plan) are discussed. A subjective consideration of the merits and drawbacks of both the documents, and the systems that support them, is presented, along with an option for the way forward for threatened species conservation in New Zealand.  相似文献

The peacock moraeas are South African geophytes withIris-like flowers that are characterized by broad outer tepals with iridescent spots. Three of the seven species were studied and found to be exclusively visited and pollinated by beetles. The primary pollinators were hopliine (Scarabaeidae) beetles in the generaPeritrichia, Monochelus, Anisonyx andAnisochelus. These beetles visit flowers for nectar, pollen and mating.Peritrichia rufotibialis was the primary pollinator ofMoraea villosa, P. abdominalis was the primary pollinator ofM. tulbaghensis, whileMonochelus sp. nov. was the primary pollinator of the third species,M. neopavonia. High levels of fruit set attest to the effectiveness of these beetles as pollinators. Average fruit set ranged from 83.5 ± 34.8% (M. villosa) to 97.2 ± 11.6% (M. tulbaghensis). Average seed set in the various populations was more variable, but was also quite high, ranging from 77.3 ± 40.8 (M. villosa) to 342.2 ± 115.0 (M. neopavonia) seeds per capsule. Although hopliine beetles are abundant and diverse in southern Africa and commonly visit flowers, this is the first well-documented report of pollination by these beetles.  相似文献

The New Zealand distributions of three species of Boeckella (Copepoda, Calanoida), B. triarticulata, B. dilatata and B. hamata are mapped. B. triarticulata is primarily a pond dweller but is also found in reservoirs and shallow lakes. B. dilatata is mainly found in the deeper glacial lakes and ponds in the central region of the South Island and B. hamata has a more widespread distribution in lakes and ponds in the South Island and lower half of the North Island. Differences in temperature optima, food requirements and dispersal ability among the three Boeckella species are related to vicariant events to explain their distribution in New Zealand.  相似文献

Polystichum, one of the largest genera of ferns, occurs worldwide with the greatest diversity in southwest China and adjacent regions. Although there have been studies of Chinese Polystichum on its traditional classification, geographic distributions, and even a few on its molecular systematics, its relationships to other species outside China remain little known. Here, we investigated the phylogeny and biogeography of the Polystichum species from China and Australasia. The evolutionary relationships among 42 Polystichum species found in China (29 taxa) and Australasia (13 taxa) were inferred from phylogenetic analyses of two chloroplast DNA sequence data sets: rps4-trnS and trnL-F intergenic spacers. The divergence time between Chinese and Australasian Polystichum was estimated. The results indicated that the Australasian species comprise a monophyletic group that is nested within the Chinese diversity, and that the New Zealand species are likewise a monophyletic group nested within the Australasian species. The divergence time estimates suggested that Chinese Polystichum migrated into Australasia from around 40 Ma ago, and from there to New Zealand from about 14 Ma. The diversification of the New Zealand Polystichum species began about 10 Ma. These results indicated that Polystichum probably originated in eastern Asia and migrated into Australasia: first into Australia and then into New Zealand.  相似文献

The geographic range and bloom frequency of the toxic dinoflagellate Alexandrium minutum and other members of the A. minutum group have been increasing over the past few decades. Some of these species are responsible for paralytic shellfish poisoning (PSP) outbreaks throughout the world. The origins of new toxic populations found in previously unaffected areas are typically not known due to a lack of reliable plankton records with sound species identifications and to the lack of a global genetic database. This paper provides the first comprehensive study of minutum-group morphology and phylogeny on a global scale, including 45 isolates from northern Europe, the Mediterranean, Asia, Australia and New Zealand.Neither the morphospecies Alexandrium lusitanicum nor A. angustitabulatum was recoverable morphologically, due to large variation within and among all minutum-group clonal strains in characters previously used to distinguish these species: the length:width of the anterior sulcal plate, shape of the 1′ plate, connection between the 1′ plate and the apical pore complex, and the presence of a ventral pore. DNA sequence data from the D1 to D2 region of the LSU rDNA also fail to recognize these species. Therefore, we recommend that all isolates previously designated as A. lusitanicum or A. angustitabulatum be redesignated as A. minutum. A. tamutum, A. insuetum, and A. andersonii are clearly different from A. minutum on the basis of both genetic and morphological data.A. minutum strains from Europe and Australia are closely related to one another, which may indicate an introduction from Europe to Australia given the long history of PSP in Europe and its recent occurrence in Australia. A minutum from New Zealand and Taiwan form a separate phylogenetic group. Most strains of A. minutum fit into one of these two groups, although there are a few outlying strains that merit further study and may represent new species. The results of this paper have greatly improved our ability to track the spread of A. minutum species and to understand the evolutionary relationships within the A. minutum group by correcting inaccurate taxonomy and providing a global genetic database.  相似文献

Summary With the idea to develop a selection system for asymmetric somatic hybrids between oilseed rape (Brassica napus) and black mustard (B. nigra), the marker gene hygromycin resistance was introduced in this last species by protoplast transformation with the disarmed Agrobacterium tumefaciens strain C58 pGV 3850 HPT. The B. nigra lines used for transformation had been previously selected for resistance to two important rape pathogens (Phoma lingam, Plasmodiophora brassicae). Asymmetric somatic hybrids were obtained through fusion of X-ray irradiated (mitotically inactivated) B. nigra protoplasts from transformed lines as donor with intact protoplasts of B. napus, using the hygromycin resistance as selection marker for fusion products. The somatic hybrids hitherto obtained expressed both hygromycin phosphotransferase and nopaline synthase genes. Previous experience with other plant species had demonstrated that besides the T-DNA, other genes of the donor genome can be co-transferred. In this way, the produced hybrids constitute a valuable material for studying the possibility to transfer agronomically relevant characters — in our case, diseases resistances — through asymmetric protoplast fusion.  相似文献

Leptographium species have predominantly been described from North America, Canada and Europe. These fungi generally occur on conifers and many cause blue-stain of lumber. MostLeptographium species are also associated with insects and in particular, bark beetles (Coleoptera: Scolytidae). Recently, an unknown species ofLeptographium was isolated from pine infested with anIps sp. in Indonesia. In addition, two unknown species have been collected from red spruce (Picea rubra) and balsam fir (Abies balsamea) roots from high elevation sites in Eastern North America. The latter isolates are unusual in that they are associated with the feeding wounds made by the conifer swift mothKorscheltellus gracilus (Lepidoptera: Hepialidae), which is a habitat unique for species ofLeptographium. Comparison with knownLeptographium species has revealed that the isolates from Indonesia and those from Eastern North America represent three previously undescribed taxa. They are, therefore, described in this study asL. pineti sp. nov,L. abieticolens sp. nov. andL. peucophilum sp. nov.  相似文献
