Polypyrrole multi-walled carbon nanotube composite layers were used to modify the gold layer to measure heavy metal ions using the surface plasmon resonance technique. The new sensor was fabricated to detect trace amounts of mercury (Hg), lead (Pb), and iron (Fe) ions. In the present research, the sensitivity of a polypyrrole multi-walled carbon nanotube composite layer and a polypyrrole layer were compared. The application of polypyrrole multi-walled carbon nanotubes enhanced the sensitivity and accuracy of the sensor for detecting ions in an aqueous solution due to the binding of mercury, lead, and iron ions to the sensing layer. The Hg ion bonded to the sensing layer more strongly than did the Pb and Fe ions. The limitation of the sensor was calculated to be about 0.1 ppm, which produced an angle shift in the region of 0.3° to 0.6°.  相似文献

The retina is the light-sensitive tissue of the eye that facilitates vision. Mutations within genes affecting eye development and retinal function cause a host of degenerative visual diseases, including retinitis pigmentosa and anophthalmia/microphthalmia. The characin fish Astyanax mexicanus includes both eyed (surface fish) and eyeless (cavefish) morphs that initially develop eyes with normal retina; however, early in development, the eyes of cavefish degenerate. Since both surface and cave morphs are members of the same species, they serve as excellent evolutionary mutant models with which to identify genes causing retinal degeneration. In this study, we crossed the eyed and eyeless forms of A. mexicanus and quantified the thickness of individual retinal layers among 115 F2 hybrid progeny. We used next generation sequencing (RAD-seq) and microsatellite mapping to construct a dense genetic map of the Astyanax genome, scan for quantitative trait loci (QTL) affecting retinal thickness, and identify candidate genes within these QTL regions. The map we constructed for Astyanax includes nearly 700 markers assembled into 25 linkage groups. Based on our scans with this map, we identified four QTL, one each associated with the thickness of the ganglion, inner nuclear, outer plexiform, and outer nuclear layers of the retina. For all but one QTL, cavefish alleles resulted in a clear reduction in the thickness of the affected layer. Comparative mapping of genetic markers within each QTL revealed that each QTL corresponds to an approximately 35 Mb region of the zebrafish genome. Within each region, we identified several candidate genes associated with the function of each affected retinal layer. Our study is the first to examine Astyanax retinal degeneration in the context of QTL mapping. The regions we identify serve as a starting point for future studies on the genetics of retinal degeneration and eye disease using the evolutionary mutant model Astyanax.  相似文献

Packing of ribosomes in crystals from the lizard Lacerta sicula   总被引:2,自引:0,他引:2  
The packing of ribosomes in the large crystalline sheets found in the lizard Lacerta sicula has been investigated by electron microscopy. The ribosomes in each of the two layers composing a sheet are organised as tetramers on a P4 space group lattice. The two layers face in opposite directions and tend to be related to one another crystallographically, generating a family of P422 crystals of different unit cell dimensions. The projected structure of one layer was determined from negatively stained, isolated sheets by separating the contributions from each layer in Fourier transforms computed from electron micrographs. Comparison of the projection map with other, low resolution, analyses of images of isolated eukaryotic ribosomes indicates that the large subunit- small subunit axis lies approximately parallel to the plane of the sheet.  相似文献

SLAM is a popular task used by robots and autonomous vehicles to build a map of an unknown environment and, at the same time, to determine their location within the map. This paper describes a SLAM-based, probabilistic robotic system able to learn the essential features of different parts of its environment. Some previous SLAM implementations had computational complexities ranging from O(Nlog(N)) to O(N2), where N is the number of map features. Unlike these methods, our approach reduces the computational complexity to O(N) by using a model to fuse the information from the sensors after applying the Bayesian paradigm. Once the training process is completed, the robot identifies and locates those areas that potentially match the sections that have been previously learned. After the training, the robot navigates and extracts a three-dimensional map of the environment using a single laser sensor. Thus, it perceives different sections of its world. In addition, in order to make our system able to be used in a low-cost robot, low-complexity algorithms that can be easily implemented on embedded processors or microcontrollers are used.  相似文献

提出一种使用生长、分级的自组织映射(growing hierarchical self-organizing map,GHSOM)模型进行基于EEG信号的意识任务分类来实现脑机接口技术的方法。GHSOM模型是自组织映射(self-organizing map,SOM)的一种变体,由多层的SOM组成,具有一定的分级结构,能够表达数据中不同层次的信息。同时研究了使用平均量化误差(mean quantization error,mqe)和量化误差(quantization error,qe)两种方法实现的GHSOM模型对意识任务分类的作用。结果表明,GHSOM模型对于意识任务的可分性能够提供可视化的信息,并且发现使用量化误差方法实现的GHSOM模型提供较多的数据信息和较高的分类精度。使用GHSOM模型进行了5类意识任务的分类,平均分类精度可达80%。  相似文献

A rice (Oryza sativa L.) genomic clone encoding the gene for a form of soluble starch synthase (SSS1) and its 5'- and 3'-flanking regions has been isolated and sequenced. The SSS1 gene contained 15 exons interrupted by 14 introns. The exon/intron organization of the SSS1 gene was divergent from that of the rice Waxy gene coding for granule-bound starch synthase, thus suggesting that the SSS1 and granule-bound starch synthase genes have evolved from an ancestral gene in a different way or that the two genes are products of different ancestral genes that have converged during evolution. However, these two genes were closely located to each other on rice chromosome 6 at an approximate map distance of 5 centimorgans. The nucleotide sequence of the 5'-end region of the gene is unique because of the presence of some repetitive sequences.  相似文献

This paper presents a Spiking Neural Network (SNN) architecture for mobile robot navigation. The SNN contains 4 layers where dynamic synapses route information to the appropriate neurons in each layer and the neurons are modeled using the Leaky Integrate and Fire (LIF) model. The SNN learns by self-organizing its connectivity as new environmental conditions are experienced and consequently knowledge about its environment is stored in the connectivity. Also a novel feature of the proposed SNN architecture is that it uses working memory, where present and previous sensor states are stored. The water-binding properties of bovine lens α-crystallin, collagen from calf skin and bovine serum albumin (BSA), were investigated with various techniques. The water absorptive capacity was obtained in high vacuum desorption experiments volumetrically, and also gravimetrically in controlled atmosphere experiments. NMR spin–echo technique was used to study the hydration of protein samples and to determine the spin–spin relaxation times (T2) from the protons of water, absorbed on the proteins. Isolated bovine lenses were sectioned into 11–12 morphological layers (from anterior cortex through nucleus to posterior cortex). Crystallin profiles were obtained for each lens layer using thin-layer isoelectric focusing in polyacrylamide gel (IEF). The water content in relation to dry weight of proteins was measured in individual morphological lens layers. During the water vapor uptake P/P0=0.75, α-crystallin did not absorb water, suggesting that hydrophobic regions of the protein are exposed to the aqueous solvent. At P/P0=1.0, the absorption of water by α-crystallin was 17% with a single component decay character of spin–echo (T2=3 ms). Addition of water to α-crystallin to about 50% of its w/w in the protein sample showed T2=8 ms with only one single component decay of the spin–echo signal. The single component decay character of the spin–echo indicates at the tightly bound water by α-crystallin. Under a relative humidity P/P0=1.0, collagen and BSA absorbed correspondingly 19.3% and 28% of water and showed a two-component decay curve with T2 of about 5 and 40 ms. The findings demonstrate the presence of two water fractions in collagen and BSA which are separated in space. The IEF data suggest a tight binding of water with α-crystallin with similar distribution patterns in the lens layers. The IEF data demonstrate a possible chaperone-like function for α-crystallin in the nucleus and inner cortex of the lens, but not in the outer cortex. To conclude, it was found that α-crystallin can immobilize and bind water to a greater extent than other proteins such as collagen and BSA. These results shed new light on structural properties of α-crystallin and have important implications for understanding the mechanism of the chaperone-like action of this protein in the lens and non-ocular tissues.  相似文献

Acoustoelastic (AE) ultrasound image analysis is a promising non-invasive approach that uses load-dependent echo intensity changes to characterize stiffness of tendinous tissue. The purpose of this study was to investigate whether AE can detect localized changes in tendon stiffness due to partial and full-thickness tendon tears. Ovine infraspinatus tendons with different levels of damage (Intact, 33%, 66% and full thickness cuts initiated on the articular and bursal sides) were cyclically loaded in a mechanical testing system while cine ultrasound images were recorded. The load-induced changes in echo intensity on the bursal and articular side of the tendon were determined. Consistent with AE theory, the undamaged tendons exhibited an increase in echo intensity with tendon loading, reflecting the strain-stiffening behavior of the tissue. In the intact condition, the articular region demonstrated a significantly greater increase in echo intensity during loading than the bursal region. Cuts initiated on the bursal side resulted in a progressive decrease in echo intensity of the adjacent tissue, likely reflecting the reduced load transmission through that region. However, image intensity information was less sensitive for identifying load transmission changes that result from partial thickness cuts initiated on the articular side. We conclude that AE approaches may be useful to quantitatively assess load-dependent changes in tendon stiffness, and that disruption of AE behavior may be indicative of substantial tendon damage.  相似文献

We describe two Java applets which are useful for insightful presentation of intermediate experimental data in gene discovery projects involving large scale sequencing. One of these applets provides a physical map of a genomic region and provides easy access to the second applet, which furnishes a detailed map of sequence contigs associated with clones on the physical map. In particular, the second applet displays all the known information about each contig, including the presence of exons, database homology `hits', repetitive elements and other features; the graphics are linked to other World Wide Web pages, providing detailed information on each feature. A multi-layer mathematical model of oxygen supply and consumption in the rat retina is described. The model takes advantage of the highly layered structure of the retina and the compartmentalisation of the available oxygen sources. The retina is divided into eight layers, each with a distinct oxygen consumption or supply rate. When applied to the available data from intraretinal oxygen measurements in the rat under normal physiological conditions, a close fit between the model and the data was achieved (r(2)=0.98+0.005, n=6). The model was then used to investigate recent evidence of oxygen regulating mechanisms in the rat retina during systemic hyperoxia. Fitting our model to the experimental data (r(2)=0.988+0.004, n=25) allowed the relative oxygen delivery or consumption of the key retinal layers to be determined. Two factors combine to produce the relative stability of inner retinal oxygen levels in hyperoxia. The retinal layer containing the outer plexiform layer/deep retinal capillaries, switches from a net source to a net consumer of oxygen, and the oxygen consumption of the outer region of the inner plexiform layer increases significantly. The model provides a useful tool for examining oxygen consumption and supply in all retinal layers, including for the first time, those layers within the normally perfused inner retina.  相似文献

Three types of imaging, namely layer structure, electrochemical reaction, and enzyme sensor response, were achieved by applying surface plasmon resonance (SPR) measurement to an electrochemical biosensor. We constructed glucose oxidase based mediator type sensors on a gold electrode by spotting the mediator that contained horseradish peroxidase and spin coating the glucose oxidase film. The layer structure of the sensor was imaged by means of angle scanning SPR measurement. The single sensor spot (about 1 mm in diameter) consisted of about 100 x 100 pixels and its spatial structure was imaged. The multilayer structure of the enzyme sensor had a complex reflectance-incident angle curve and this required us to choose a suitable incident angle for mapping the redox state. We chose an incident angle that provided the most significant reflection intensity difference by using data obtained from two angle scanning SPR measurements at different electrode potentials. At this incident angle, we controlled the electrochemical states of the spotted mediator in cyclic voltammetry and imaged the degree to which the charged site density changed. Finally, we mapped the enzymatic activity around the mediator spot by the enzymatic reoxidation of pre-reduced mediator in the presence of glucose.  相似文献

Direct determination of crystallographic phases based on probabilistic of sigma 1 and sigma 2 "triplet" structure invariants has been found to be an effective technique for structure analysis with lamellar x-ray or electron diffraction intensity data from phospholipids. In many cases, nearly all phase values are determined, permitting a structure density (electron density for x-ray diffraction; electrostatic potential for electron diffraction) map to be calculated, which is directly interpretable in terms of known bilayer lipid structure. The major source of error is found to be due to the distortion of observed electron diffraction intensity data by incoherent multiple scattering, which can significantly affect the appearance of the electrostatic potential map, but not the success of the phase determination, as long as the observed Patterson function can be interpreted.  相似文献

The mstacercarial cyst of Cloacitrema narrabeenensis which is formed in the open is composed of four layers: an outermost layer of acid mucopolysaccharide, a layer of protein which is presumed to be tanned, a layer of neutral mucopolysaccharide and an innermost layer of keratinized protein. The two layers which together form the outer cyst wall can be split off by slight pressure from the two remaining layers which together form the inner cyst wall. In the centre of the ventral side of the inner cyst wall, the keratinized layer is incomplete and this ventral plug region is composed of neutral mucopolysaccharide. The cyst wall is therefore very similar to that of Fasciola hepatica, the main difference being that the order of the two layers of the outer cyst is reversed. General evolutionary and functional relationships of metacercarial cysts are discussed.  相似文献

Chikungunya virus has been discovered in about 60 countries of the world. It leads to joint pain, joint swelling, headache, muscle pain, and fatigue of the human body. In this work, a surface plasmon resonance (SPR)based sensor is developed to detect chikungunya virus through normal and infected platelets and plasma blood cells. The proposed SPR-based sensor uses silicon and graphene layers coated over the base of a glass prism sputtered with a silver layer. The graphene layer has the advantage of enhancing the biomolecules adsorption on the metal layer. The silicon layer between silver and graphene enhances the sensor performance. The number of graphene layers along with the thicknesses of silicon and silver layers is optimized to get the highest sensitivity of the detector. To investigate the effect of the light source wavelength, simulations are performed for four different wavelengths. The highest sensitivities exhibited by the SPR-based sensor are 393 and 160 deg/RIU for the platelets and plasma cells, respectively.

Combined small-angle x-ray scattering and transmission electron microscopy studies of intramuscular fish bone (shad and herring) indicate that the lateral packing of nanoscale calcium-phosphate crystals in collagen fibrils can be represented by irregular stacks of platelet-shaped crystals, intercalated with organic layers of collagen molecules. The scattering intensity distribution in this system can be described by a modified Zernike-Prins model, taking preferred orientation effects into account. Using the model, the diffuse fan-shaped small-angle x-ray scattering intensity profile, dominating the equatorial region of the scattering pattern, could be quantitatively analyzed as a function of the degree of mineralization. The mineral platelets were found to be very thin (1.5 nm ∼ 2.0 nm), having a narrow thickness distribution. The thickness of the organic layers between adjacent mineral platelets within a stack is more broadly distributed with the average value varying from 6 nm to 10 nm, depending on the extent of mineralization. The two-dimensional analytical scheme also leads to quantitative information about the preferred orientation of mineral stacks and the average height of crystals along the crystallographic c axis.  相似文献

Jelly coats surrounding the eggs of the South African clawed toad, Xenopus laevis, consist of three transparent, gelatinous layers: the innermost layer (J1), the middle layer (J2), and the outer layer (J3). The distribution of N-acetylglucosamine within these jelly coats, as probed with FITC-conjugated wheat germ agglutinin (WGA-FITC), and the matrix ultrastructure of each layer, as visualized in platinum replicas produced by the quick-freeze, deep-etch, and rotary-shadowing technique, suggests that each layer has a unique fiber and glycoprotein composition. J1 extends nearly 200 μm from the egg surface and exhibits no WGA-FITC staining. Stereo images of platinum replicas indicates that J1 consists of a tightly knit network of 5–10 nm fibers decorated with 10–20 nm particulate components. In contrast, J2 is a relatively thin layer, extending only 25–40 μm from the outer aspect of J1. When visualized by confocal microscopy, J2 displays a multilayered WGA-FITC staining pattern. The ultrastructure of J2 consists of sheets of fine fibers that run parallel to one another and that can be identified by their ability to bind WGA-colloidal gold. The fibers of each sheet run at an oblique angle to fibers in neighboring layers. J3 extends 100 μm or more from J2. The WGA-FITC staining pattern shows high intensity in its outer region and less intensity in regions closer to J2. Like J1, the J3 ultrastructure consists of a network of 5–10 nm fibers, decorated with 10–20 nm particulate components. The results of these studies add to a growing body of information that suggests the jelly coats surrounding the eggs of many animals consist of a fibrous glycoprotein superstructure that acts as a scaffold to which globular glycoproteins are bound. © 1996 Wiley-Liss, Inc.  相似文献

Based on the high-resolution Mg/Ca and oxygen isotope ratio (δ18O) of planktonic foraminifera Globigerinoides ruber, sea surface temperature (SST) and salinity (SSS) were reconstructed in Core PC-1 from the northern Okinawa Trough during the last 24 kyr. From the last glacial maximum to Holocene, SST varied from 20.2°C to 24.6°C. Millennial-scale climatic events of the SST during the last glacial, such as the Heinrich events, Bølling-Allerød warming, and Younger Dryas, have been identified, which are synchronous with the climate changes in the North Atlantic, suggesting a teleconnection between the northwestern Pacific and the North Atlantic. During the last 24 kyr, the SSS variation can be divided into three parts: (1) during 21–15.5 ka, there was a significantly low SSS; (2) during 15.5–11.7 ka, the SSS increased obviously; (3) since 11.7 ka, the SSS is relatively stable. In order to discuss the main factors influencing the SSS in the northern Okinawa Trough, the relative abundance of Pulleniatina obliquiloculata and the oxygen isotope difference between the northern and middle Okinawa Trough (Δδ18Osw) have been used to indicate the intensity of the Kuroshio Current and Changjiang freshwater discharge, respectively. The Δδ18Osw result shows that there is a large amount of Changjiang freshwater emptied into the northern Okinawa Trough during 18–15.5 ka, which is caused by the subtropical monsoon rain band lingering in the lower reaches of the Changjiang River drainage, supporting the Jet Transition Hypothesis. As the Kuroshio Current strengthened since 15.5 ka as indicated by the increased relative abundance of P. obliquiloculata, the variation of Kuroshio Current became the more important contribution to the SSS in the study region. The results of our study indicate that the key factor influencing the SSS in the northern Okinawa Trough is variable during the last 24 kyr.  相似文献

The structure of the polyoma virus defective species D74 (74% the size of full-length polyoma virus DNA) has been determined and compared with that of polyoma virus A2 DNA. D74 appears to be composed entirely of viral DNA sequences. (No host DNA sequences have been detected.) It is made up of three DNA segments, each about 24, 24 and 27% in size. The two 24% segments appear to be identical and the 27% segment contains one copy of all the sequences found in the 24% fragments as well as a duplication of some of the sequences. When related to the physical map of A2 DNA, each segment is found to be composed of viral sequences from 1 to about 19 map units, 67 to 69 map units and 70 to 72 map units.Three features found in other polyoma virus defective species (Lund et al., 1977) are also present in D74. (1) Sequences from the region around 67 map units are linked to other (non-contiguous) viral sequences. (2) Sequences at about 72 map units are linked to sequences at 1 map unit. (3) Multiple copies of sequences from around the origin of viral DNA replication are present. From studies on other polyoma defective molecules (Griffin &; Fried, 1975; Lund et al., 1977), the origin of DNA replication for polyoma virus has been defined to lie within the sequences from 67 to 72 map units. Since D74 replicates efficiently but lacks the sequences between 69 to 70 map units, the origin of DNA replication appears to be further defined as lying within 67 and 69 map units and/or 70 to 72 map units.  相似文献

Identifying influences of earthworm invasion and distribution in the northern Great Lakes is an important step in predicting the potential extent and impact of earthworms across the region. The occurrence of earthworm signs, indicating presence in general, and middens, indicating presence of Lumbricus terrestris exclusively, in the Huron Mountains located in the Upper Peninsula of Michigan were modeled using generalized linear models and stepwise regression to identify important environmental variables. Models were then applied to earthworm occurrence data from Seney National Wildlife Refuge, also located in the Upper Peninsula of Michigan to validate results. Occurrence of earthworm signs was associated with high soil pH, high basal area of earthworm preferred overstory species, and north facing aspects. Middens of L. terrestris were associated with high soil pH, high basal area of preferred species, and close proximity to roads. The resulting model for L. terrestris was incorporated into a geographic information system (GIS) to map the expected distribution, both current and potential, across the study area. Results indicate that L. terrestris has not yet fully saturated its potential habitat, as it is currently found close to roads and has yet to establish in most interior forests sampled. Comparing field measured data to GIS layers revealed limitations in the precision of publicly available spatial data layers that should be addressed in future attempts to predict the extent of earthworm invasion across the larger Great Lakes region. However, within the Huron Mountains, it is predicted that the distribution of L. terrestris will cover, at minimum, 41 % of the area.  相似文献
