Summary A study of the thermal responses of Na-ATPase and NaK-ATPase activities in microsomes prepared from gill tissue of rainbow trout (Salmo gairdneri) revealed further evidence that the two activities are distinct from one another. Arrhenius plots of the NaK-ATPase from sea water-adapted fish and the Na-ATPase from fresh water-adapted fish were linear (Fig. 4) with estimated activation energies of 19.5 and 7.7 kcal/mole, respectively. The Na-ATPase and NaK-ATPase both showed optimum activity at 45°C (Figs. 2 and 3). The Mg-ATPase from fresh water fish showed a distinct temperature optimum at 24°C (Fig. 1) while Mg-ATPase activity from sea water fish was optimum at temperatures of about 15–24 °C (Fig. 3). The Na+ dependence of the Na-ATPase and the NaK-ATPase was examined at an assay temperature of 37 °C (Fig. 5) and the results compared with those obtained at 13 °C. No apparent differences were noted for the Na-ATPase, but with the NaK-ATPase both theK 0.5 for Na+ and optimum Na+ concentration increased at the higher assay temperature. 1. Two Na(+)-stimulated ATPase activities were determined in gill homogenates from squid, shrimp and teleost fish; in kidney slice homogenates from teleost fish, bullfrog, toad, iguana, chicken, duck, rat, pig and cow, as well as in homogenates from rat small intestinal cells, brain cortex and liver slices. The two Na(+)-stimulated ATPase activities, the Na- and the Na,K-ATPase, showed a different behavior toward K+ and ouabain. 2. The ouabain-insensitive, K(+)-independent, Na-ATPase activity for all the studied homogenates was completely inhibited by 2 mM furosemide. 3. An increase in cell volume of the kidney, brain cortex and liver slice preparations, as well as of the rat small intestinal cells, produced a concomitant increase of the ouabain-insensitive Na-ATPase.  相似文献

Removal of phospholipids from brain microsomes using a purified, protease-free phospholipase C preparation led to proportional losses of net Na+,K+-stimulated adenosine triphosphatase, K+-stimulated p-nitrophenylphosphatase, and Na+-stimulated ADP-ATP exchange activities. These enzymatic activities were restored to 60–100% of control values by the addition of a variety of purified phospholipids, but not by detergents or EGTA. These findings support the concept of a general phospholipid requirement for this enzyme system. This work further suggests that phospholipids are important both for formation and decomposition of the phosphorylated intermediate (s) which probably participate in the net reaction.  相似文献

Homogenates of gills from the freshwater shrimp M. amazonicum exhibit the following ATPase activities: (i) a basal, Mg2+-dependent ATPase; (ii) an ouabain-sensitive, Na+ + K+-stimulated ATPase; (iii) an ouabain-insensitive, Na+-stimulated ATPase; and (iv) an ouabain-insensitive, K+-stimulated ATPase.
  • 2.2. K+ suppresses the Na+-stimulated ATPase activity in a mixed-type kind of inhibition, whereas Na+ does not exert any noticeable effect on the K+-stimulated ATPase activity.
  • 3.3. The Na+- and the K+-stimulated ATPase activities are totally inhibited by 5 mM ethacrynic acid in the incubation medium.
  • 4.4. The Na+- and the K+-stimulated ATPase activities are not expressions of the activation of a Ca-ATPase.
  • 5.5. The possible localization and roles of the described ATPases within the gill epithelium are briefly discussed and evaluated.

The present work evaluates the effect of lipid peroxidation on the ouabain-insensitive Na-ATPase of basolateral plasma membranes from rat kidney proximal tubular cells as an indirect way to study the lipid dependence of this enzyme. An inverse relationship between lipid peroxidation and Na-ATPase activity was found. This effect was due neither to a change in the optimalK m of the system for Na+ nor for the substrate Mg : ATP, nor the optimal pH value of the medium. The optimal temperature value, however, was shifted toward a higher value. There was also an increase of the apparent energy of activation in the region of temperatures above the transition point (20°C) with increase in lipid peroxidation. Peroxidized membranes incubated with phosphatidylcholine from soybean restored their Na-ATPase activity. On the other hand, the Na-ATPase activity was sensitive to oleoly lysophosphatidylcholine. These results suggest that lipid peroxidation might be affecting the Na-ATPase activity through either an increase of peroxidized phospholipids, which might change the membrane fluidity of the lipid microenvironment of the ATPase molecules, or through a direct effect of lysophospholipids released during the lipid peroxidation.  相似文献

It was found that a facultatively anaerobic alkaliphile, Exiguobacterium aurantiacum, possesses a membrane-bound ATPase, which was activated specifically by Na+. The Na+-stimulated ATPase activity reached a maximum value at 200 mM NaCl. In the presence of 200 mM NaCl, the activity was drastically reduced by vanadate, a potent inhibitor of P-type ATPase, with a half-maximal inhibition at 1 μM. Incubation of the membranes with [γ-32P]ATP followed by acidic lithium dodecyl sulfate–polyacrylamide gel electrophoresis demonstrated the existence of two phosphorylated intermediates with apparent molecular masses of 60 and 100 kDa. Only phosphorylation of the 100-kDa polypeptide was inhibited by vanadate. The membrane extract containing Na+-stimulated ATPase, when reconstituted into soybean phospholipid vesicles, exhibited 22Na+ transport by the addition of ATP, which was inhibited by vanadate and gramicidin. The classical E2-P intermediate of (Na+ + K+)-ATPase dephosphorylates readily in the presence of K+ and is not affected by the addition of ADP. To determine the significane in the reaction cycle of (Na+ + K+)-ATPase of kinetically atypical phosphorylations of rat brain (Na+ + K+)-ATPase we compared these phosphorylated components with the classical E2-P intermediate of this enzyme by gel electrophoresis. When rat brain (Na+ + K+)-ATPase was phosphorylated in the presence of high concentrations of Na+ a proportion of the phosphorylated material formed was sensitive to ADP but resistant to K+. Similarly, if phosphorylation was carried out in the presence of Na+ and Ca2+ up to 300 pmol/mg protein of a K+-resistant, ADP-sensitive material were formed. If phosphorylation was from [γ-32P]CTP up to 800 pmol 32P/mg protein of an ADP-resistant, K+-sensitive phosphorylated matterial were formed. On gel electrophoresis these phosphorylated materials co-migrated with authentic Na+-stimulated, K+-sensitive, E2-P-phosphorylated intermediate of (Na+ + K+)-ATPase, supporting suggestions that they represent phosphorylated intermediates in the reaction sequence of this enzyme.  相似文献

In a previous study, evidence was presented for an external Na+-dependent, ouabain-insensitive component of Na+ efflux and an external K+-dependent component of K+ efflux in the Ehrlich ascites tumor cell. Evidence is now presented that these components are inhibited by the diuretic furosemide and that under conditions of normal extracellular Na+ and K+ they represent Na+-for-Na+ and K-+for-K+ exchange mechanisms. Using 86Rb to monitor K+ movements, furosemide is shown to inhibit an ouabain-insensitive component of Rb+ influx and a component of Rb+ efflux, both representing approx. 30% of the total fux. Inhibition of Rb+ efflux is greatly reduced by removal of extracellular K+. Furosemide does not alter steady-state levels of intracellular K+ and it does not prevent cells depleted of K+ by incubation in the cold from regaining K+ upon warming. Using 22Na to monitor Na+ movements, furosemide is shown to inhibit an ouabain-insensitive component of unidirectional Na+ efflux which represents approx. 22% of total Na+ efflux. Furosemide does not alter steady-state levels of intracellular Na+ and does not prevent removal of intracellular Na+ upon warming from cells loaded with Na+ by preincubation in the cold. The ability of furosemide to affect unidirectional Na+ and K+ fluxes but not net fluxes is consistent with the conclusion that these components of cation movement across the cell membrane represent one-for-one exchange mechanisms. Data are also presented which demonstrate that the uptake of α-aminoisobutyrate is not affected by furosemide. 1. The adenosine-triphosphatase activity of rat-brain microsomes was measured between 0° and 37°. The stimulatory effect of Na+ plus K+ on the Mg2+-dependent adenosine-triphosphatase activity decreased sharply with decreasing temperature and became negligible at 0°. An Arrhenius plot drawn from the experimental data showed two discontinuities: one at about 6° and the other at about 20°. 2. The increment in activity induced by Na+ plus K+ was more sensitive to oligomycin at lower than at higher temperatures, but the opposite was observed for ouabain. The action of oligomycin showed a biphasic character, since below a certain concentration it caused slight activation of Na+-plus-K+-activated adenosine triphosphatase. 3. Where oligomycin increased the activity of the enzyme, it also enhanced the accumulation of an acid-precipitable phosphorylated compound formed through the transfer of the γ-phosphate group of [32P]ATP to the enzyme system. Stimulatory concentrations of oligomycin did not interfere with K+-mediated dephosphorylation of the intermediate, though high concentrations of oligomycin counteracted the effect of K+. 4. The temperature profile of K+-stimulated microsomal phosphatase qualitatively resembled that of microsomal adenosine triphosphatase.  相似文献

Summary The effect of the loop diuretic furosemide (4-chloro-N-furfuryl-5-sulfamoyl-anthranilic acid) on the thiol-dependent, ouabain-insensitive K(Rb)/Cl transport in low K+ sheep red cells was studied at various concentrations of extracellular Rb+, Na+ and Cl. In Rb+-free NaCl media, 2×10–3 m furosemide inhibited only one-half of thiol-dependent K+ efflux. In the presence of 23mm RbCl, however, the concentration of furosemide to produce 50% K+ efflux inhibition (IC50) was 5×10–5 m. In Rb+ containing NaCl media, the inhibitory effect of 10–3 m furosemide was equal to that caused by NO 3 replacement of Cl in the medium. The apparent synergistic action of furosemide and external Rb+ on K+ efflux was also seen in the ouabain-insensitive Rb+ influx. A preliminary kinetic analysis suggests that furosemide binding alters both maximal K+(Rb+) transport and apparent external Rb+ affinity. In the presence of external Rb+, Na+ (as compared to choline) exerted a small but significant augmentation of the furosemide inhibition of K+(Rb+) fluxes. There was no effect of Cl on the IC50 value of furosemide. As there is no evidence for coupled Na+K+ cotransport in low K+ sheep red cells, furosemide may modify thiol-dependent K+(Rb+/Cl flux or Rb+ (and to a slight degree Na+) modulate the effect of furosemide.  相似文献

Ouabain, aspecific inhibitor ofNa+-K+-ATPase,was coupled to epoxy agarose via a 13-atom spacer to make an affinitycolumn that specifically bindsNa+-K+-ATPase.Na+-K+-ATPasefrom rat and dog kidney was bound to the column and was eluted as afunction of enzyme conformation, altered by adding specificcombinations of ligands.Na+-K+-ATPasefrom both sources bound to the column in the presence of Na + ATP + Mgand in solutions containing 30 mM K. No binding was observed in thepresence of Na or Na + ATP. These experiments suggest thatNa+-K+-ATPasebinds to the column under the same conditions that it binds tountethered ouabain.Na+-K+-ATPasealready bound to the column was competitively eluted with excess freeNa + ouabain or with Na + ATP. The latter eluted active enzyme. Forcomparable amounts of boundNa+-K+-ATPase,Na + ouabain and Na + ATP eluted more rat than dogNa+-K+-ATPase,consistent with the lower affinity of the ratNa+-K+-ATPasefor ouabain. The ouabain-affinity column was used to purify activeNa+-K+-ATPasefrom rat kidney microsomes and rat adrenal glomerulosa cells. Thespecific activity of the kidney enzyme was increased from ~2 to 15 µmolPi · mg1 · min1.Na+-K+-ATPasepurified from glomerulosa cells that were prelabeled with [32P]orthophosphatewas phosphorylated on the -subunit, suggesting that these cellscontain a kinase that phosphorylatesNa+-K+-ATPase.

We investigated the effect of salinity on the relationship between Na+-K+-ATPase and sulfogalactosyl ceramide (SGC) in the basolateral membrane of rainbow trout (Oncorhynchus mykiss) gill epithelium. SGC has been implicated as a cofactor in Na+-K+-ATPase activity, especially in Na+-K+-ATPase rich tissues. However, whole-tissue studies have questioned this role in the fish gill. We re-examined SGC cofactor function from a gill basolateral membrane perspective. Nine SGC fatty acid species were quantified by tandem mass spectrometry (MS/MS) and related to Na+-K+-ATPase activity in trout acclimated to freshwater or brackish water (20 ppt). While Na+-K+-ATPase activity increased, the total concentration and relative proportion of SGC isoforms remained constant between salinities. However, we noted a negative correlation between SGC concentration and Na+-K+-ATPase activity in fish exposed to brackish water, whereas no correlation existed in fish acclimated to freshwater. Differential Na+-K+-ATPase/SGC sensitivity is discussed in relation to enzyme isoform switching, the SGC cofactor site model and saltwater adaptation.This revised version was published online in June 2005 with a corrected cover date.  相似文献

—Microsomal fractions prepared from guinea pig cerebral cortex manifested ADP-ATP exchange activity, 40–99 per cent of which was extractable by dilute salt solutions. All of the (Na+, K+)-ATPase activity remained in the particulate material. The unextracted ADP-ATP exchange activity was stimulated six to seven fold by a non-ionic detergent (Lubrol W). When pre-extracted microsomes were sedimented in a sucrose density gradient, the ADP-ATP exchange activity was more widely distributed than (Na+, K+)-ATPase or adenylate kinase activities. The ADP-ATP exchange activity of microsomes extracted with NaI was stimulated by Na+ ions when the Mg2+ concentration in the reaction mixture was low (0·2 mm ). The Na+ stimulation of exchange activity was more variable than was the stimulation of phosphate formation by Na+ plus K+. The Na+-stimulated ADP-ATP exchange reaction of extracted microsomes may be a component of the (Na+, K+)-ATPase system, which has not been freed from adenylate kinase or possibly other contributing enzyme systems.  相似文献

The kidney is an organ playing an important role in ion regulation in both freshwater (FW) and seawater (SW) fish. The mechanisms of ion regulation in the fish kidney are less well studied than that of their gills, especially at the level of transporter proteins. We have found striking differences in the pattern of Na+/K+/2Cl- cotransporter (NKCC) expression between species. In the killifish kidney, NKCC is apically localized in the distal and collecting tubules and basolaterally localized in the proximal tubules. However, in the SW killifish gill, NKCC is basolaterally co-localized with Na+/K+-ATPase, whereas in FW, NKCC immunoreactivity is primarily apical, although still colocalized within the same mitochondria-rich cell with basolateral Na+/K+-ATPase. Rainbow trout kidney has NKCC only in the apical membrane of the distal and collecting tubules in both environments, with no signal being detected in the proximal tubule. On the other hand, in the trout gill, NKCC is found basolaterally in both FW and SW environments. An important observation is that, in the gills of rainbow trout, the trailing edge of the filament possesses mostly Na+/K+-ATPase-positive but NKCC-negative mitochondria-rich cells, whereas in the region between and at the roots of the gill lamellae, most mitochondria-rich cells exhibit both Na+/K+-ATPase- and NKCC-positive immunoreactivity. These results suggest that the differential localization of transporters between the two species represents differences in function between these two euryhaline fishes with different life histories and strategies. Summary Experimental evidence is given that the hallucinogen harmaline (HME) behaves as an inhibitor of the (Na++K+)-ATPase system, specifically in the Na+-dependent phosphorylation reaction. HME at 0.3 to 3mm inhibited several membrane ATPase preparations such as those from human erythrocytes, rat brain and squid retinal axons. The same concentration blocked Na+ outflow from squid giant axons. The behavior of several harmane derivatives such as harmine, harmalol and harmaline demonstrated that certain groups influenced the concentration for 50% inhibition of the ATPase system. The following evidence demonstrated that HME blocked the formation of the phosphorylated intermediate by competition with Na ions in the (Na++K+)-ATPase reaction in rat brain. (1) The HME effect on the overall (Na++K+)-ATPase reaction showed a fully competitive inhibition with respect to Na ion concentration. (2) The inhibition of the Na+-stimulated phosphorylation by HME was fully competitive with respect to Na ions, with or without oligomycin present. (3) HME inhibited the effect of ADP on the phosphorylation reaction using32P-ATP. (4) HME did not accelerate the rate of membrane dephosphorylation by means of32P-ATP and cold ATP.From the behavior of HME as a competitive inhibitor at Na ion sites of the (Na++K+)-ATPase reactions one may gain information about (a) The chemical nature of Na+ sites which may be responsible for the selectivity of this cation, and (b) The sequence of Na+ and ATP entrance into the Na+-dependent phosphorylation reaction. The experimental evidence supports the hypothesis that the entrance of Na+ into the enzyme system may precede the formation of the phosphorylated intermediate.  相似文献

The ouabain-insensitive, Mg2+-dependent, Na+-stimulated ATPase activity present in fresh basolateral plasma membranes from guinea-pig kidney cortex cells (prepared at pH 7.2) can be increased by the addition of micromolar concentrations of Ca2+ to the assay medium. The Ca2+ involved in this effect seems to be associated with the membranes in two different ways: as a labile component, which can be quickly and easily ‘deactivated’ by reducing the free Ca2+ concentration of the assay medium to values lower than 1 μM; and as a stable component, which can be ‘deactivated’ by preincubating the membranes for periods of 3–4 h with 2 mM EDTA or EGTA. Both components are easily activated by micromolar concentrations of Ca2+. The Ka of the system for Na+ is the same, 8 mM, whether only the stable component or both components, stable and labile, are working. In other words, the activating effect of Ca2+ on the Na+-stimulated ATPase is on the Vmax, and not on the Ka of the system for Na+. The activating effect of Ca2+ may be related to some conformational change produced by the interaction of this ion with the membranes, since it can also be obtained by resuspending the membranes at pH 7.8 or by ageing the preparations. Changes in the Ca2+ concentration may modulate the ouabain-insensitive, Na+-stimulated ATPase activity. This modulation could regulate the magnitude of the extrusion of Na+ accompanied by Cl? and water that these cells show, and to which the Na+-ATPase has been associated as being responsible for the energy supply of this mode of Na+ extrusion.  相似文献

The primary sequence of non-gastric H,K-ATPase differs much more between species than that of Na,K-ATPase or gastric H,K-ATPase. To investigate whether this causes species-dependent differences in enzymatic properties, we co-expressed the catalytic subunit of human non-gastric H,K-ATPase in Sf9 cells with the β1 subunit of rat Na,K-ATPase and compared its properties with those of the rat enzyme (Swarts et al., J. Biol. Chem. 280, 33115-33122, 2005). Maximal ATPase activity was obtained with NH4+ as activating cation. The enzyme was also stimulated by Na+, but in contrast to the rat enzyme, hardly by K+. SCH 28080 inhibited the NH4+-stimulated activity of the human enzyme much more potently than that of the rat enzyme. The steady-state phosphorylation level of the human enzyme decreased with increasing pH, [K+], and [Na+] and nearly doubled in the presence of oligomycin. Oligomycin increased the sensitivity of the phosphorylated intermediate to ADP, demonstrating that it inhibited the conversion of E1P to E2P. All three cations stimulated the dephosphorylation rate dose-dependently. Our studies support a role of the human enzyme in H+/Na+ and/or H+/NH4+ transport but not in Na+/K+ transport.  相似文献

The K+-stimulated phosphatase activity of microsomes from rat kidney was not inhibited by l-phenylalanine, but the HCO3?-stimulated phosphatase activity was markedly inhibited by l-phenylalanine. Valinomycin enhanced the HCO3?-stimulated phosphatase activity, but did not enhance the K+-stimulated phosphatase activity. Ouabain did not inhibit the HCO3?-stimulated phosphatase activity, but inhibited the K+-stimulated phosphatase activity.The renal K+-stimulated phosphatase activity was suppressed to 40% of the control values by adrenalectomy, but the renal HCO3?-stimulated phosphatase activity was little suppressed by adrenalectomy. The renal K+-stimulated phosphatase activity in intact and adrenalectomized rats was found to be significantly elevated, in a manner similar to the elevation of the renal (Na+ + K+)-ATPase activity by aldosterone treatment (P < 0.02).  相似文献

A furosemide-sensitive, ouabain-insensitive [86Rb+] uptake is described in glioma cells in culture which is dependent upon external Na+, K+, and Cl? concentrations. This transport activity was also inhibited by bumetanide at 100-fold lower concentrations than furosemide. Furosemide-sensitive swelling of glioma cells is demonstrated and this activity is dependent upon external Na+ and K+ in a manner similar to [86Rb+] uptake. This transport activity was not detected in neuroblastoma cells and the possible relevance of these findings to extracellular K+ buffering by glia is discussed.  相似文献

The change of the enzyme activation kinetics by Mg2+, ATP, Na+ and K+ from simple saturation in FW to cooperativity in BW and other habitat-dependent variations including the pH alkaline shift in BW are hypothetically related to an adaptive significance to the different environmental salinity.
  • 4.4. Gill total lipids and phospholipids are 30% lower in BW than in FW while their ratio is constant; some differences in gill total lipid fatty acid composition between FW and BW do not significantly affect the unsaturation parameters.
