The sciarid fly Lycoriella auripila is the major pest of mushrooms cultivated in the UK. Its larvae, which are capable of damaging the crop at all stages of production, may cause severe yield losses and can only be controlled with chemical pesticides. An indigenous isolate of the insect‐parasitic nematode Steinemema feltiae was tested as a biological control agent and its effects compared with two commonly used insecticides, diazinon and diflubenzuron. The timing of application of nematodes was found to affect their efficacy. When applied to compost during spawning, nematodes did not significantly reduce fly emergence, but they did if applied at casing when they were almost as effective as diflubenzuron. Diazinon incorporated into compost did not reduce fly emergence and was also the only treatment that did not lower the incidence of mushrooms spoiled by tunnelling of the larvae of L. auripila. When compared with untreated control plots those treated both with diazinon and diflubenzuron showed significant mean losses in yield of 10% in total weight and 17% in total numbers of mushrooms picked. In contrast, when S. feltiae was applied at casing significant mean increases in yield of 7% and 19%, respectively, were attained. Infective nematodes persisted well in casing, very few were found on sporophores.  相似文献

Thirteen species of saprobic rhabditid nematodes (11 genera) were identified from samples of compost and casing material collected from mushroom farms in the British Isles. Caenorhabditis elegans, the most frequently found saprobe, was mass-produced monoxenically and its effects on the cultivated mushroom, Agaricus bisporus (strain U3) were studied. C. elegans did not multiply in well-prepared, pasteurised, spawned compost, whereas casing material proved to be a highly suitable environment for its reproduction. An initial casing inoculum of 106 nematodes/crate of compost (7.5 kg), caused a significant reduction in mushroom yield. Losses in total mushroom yields of 11%, 20% and 26% were caused by initial inoculum rates of 106, 107and 2 × 107 nematodes/crate, respectively. Yields were negatively correlated with the initial nematode inoculation level and regression equations were derived. The nematode treatments caused fewer mushrooms to be produced and an absence of the usual distinctive flushing patterns. C. elegans caused considerable deterioration in mushroom quality and characteristic distortion of mushrooms. Individual sporophores were mis-shapen, notched and had brown or violet coloured grills. Up to 3.8%, 6.7% and 10.8% of total weight and 3.5%, 5.4% and 8% of total numbers of mushrooms were distorted at the three highest nematode inoculum rates tested. Weights and numbers of distorted mushrooms were positively correlated with the initial nematode population. C. elegans commonly colonised sporophores.  相似文献

The life-history of L. auripila, as a pest of mushrooms, is outlined and an explanation is given of the different types of damage caused. The most satisfactory control is based on the incorporation of insecticides in compost at spawning. Emulsions of chlorfenvinphos and diazinon at 50 ppm are lethal to young larvae for at least 7 weeks but granular formulations of the former must be applied at 100 ppm to achieve comparable results. Drenches of 0–01% malathion, applied at about 40 gal/1000 ft2, are effective against larvae in the casing.  相似文献

Sugar mill by-products compost may be a good soil amendment to promote tomato (Lycopersicon esculentum L.) growth. In addition, the compost may further promote plant growth by inoculation with N2-fixing bacteria. Compost from sugar-mill waste was prepared with and without the N2-fixing bacteria, Azotobacter vinelandii, Beijerinckia derxii and Azospirillum sp. and incubated for 50 days. Each compost type was added to 10 kg of soil in pots at rates of 0, 15, and 45 g with and without fertilizer N at rates of 0, 0.75, and 1.54 g. A blanket application of P and K was applied to all pots. Shoot and root dry weights and N content of the whole plant was measured at 55 days. Dry weight of tomato shoots was increased by 40% by addition of fertilizer N and root weight was increased by 66%. Without fertilizer N the high rate of inoculated compost increased shoot growth 180% and uninoculated compost increased shoot growth 112%. For most treatments with and without fertilizer N, inoculated compost enhanced shoot growth and nitrogen content more than uninoculated compost. Root weights were nearly doubled by addition of either compost in comparison to the 0 N treatment. At the low rate of compost addition without fertilizer N, root weight was the same for uninoculated and inoculated compost but at the high rate of compost addition root weight was 32% higher for inoculated compost. The N2-fixing bacteria colonized roots when inoculated compost was used. Sugar mill by-products compost proved to be an effective soil amendment for promoting the growth of tomato plants.  相似文献

When added to spawned mushroom compost the microphagous saprobic nematodes Pelodera cylindrica, Mesorhabditis spiculigera and Acrobeloides bütschlii did not affect mycelial growth or compost pH. The eelworms multiplied at first but their numbers declined as the mycelium colonized the compost. In contrast, the fungal-feeding Aphelenchoides composticola increased rapidly and destroyed the mycelium. Excess water, and the presence of the fungal competitor of mushroom, Chaetomium olivaceum, allowed some increase of the microphagous forms. Different numbers of Mesorhabditis spiculigera added to the ‘casing’ had no effect on mushroom yield but the mushroom ‘flushes’ seemed less pronounced. Reasons for the failure of the saprobic eelworms to affect mushroom, and the possibility of synergistic pathogenicity by these eelworms and bacteria, are discussed.  相似文献

Three crops of Agaricus bisporus were grown on non-composted substrate (NCS), spent mushroom compost (SMC), a 50/50 mixture of NSC/SMC, or pasteurized Phase II compost. NCS consisted of oak sawdust (28% oven dry wt), millet (29%), rye (8%), peat (8%), ground alfalfa (4%), ground soybean (4%), wheat bran (9%) and CaCO3 (10%). Substrates were non-supplemented or supplemented with Target® (a commercial delayed release nutrient for mushroom culture) or soybean meal at spawning or casing, or with Micromax® (a mixture of nine micronutrients) at spawning. Mushroom yield (27.2 kg/m2) was greatest on a 50/50 mixture of NCS/SMC supplemented with 10% (dry wt) Target® at casing. The same substrate supplemented with Target® at spawning yielded 20.1 kg/m2. By comparison, mushroom yield on Phase II compost supplemented at casing or at spawning with Target® was 21.6 kg/m2 and 20.6 kg/m2, respectively. On NCS amended with 0.74% or 0.9% Micromax® at spawning, yields increased by 51.8% (12.9 kg/m2) and 71.8% (14.6 kg/m2), respectively, over non-amended NCS (8.5 kg/m2). Conversely, mushroom yields were not affected when Micromax® was added to a 50/50 mixture of NCS/SMC. Mushroom solids content was higher in mushrooms harvested from NCS amended with 0.74% Micromax® (9.6%) compared to non-amended NCS (8.3%).  相似文献

Aim: The pathogen growth in dairy compost was studied in a greenhouse setting under different seasons. Methods and Results: The five‐strain mixtures of each Escherichia coli O157:H7, Salmonella spp. and Listeria monocytogenes were inoculated separately into dry compost to yield c. 1 log CFU g?1. After acclimation at room temperature, the inoculated compost was initially adjusted to moisture levels of 10–50% and then kept in a greenhouse under different seasons. The populations of all three pathogens increased by 2·1–3·9 log CFU g?1 within 3 days in autoclaved compost with initial moisture content of at least 40%. Listeria monocytogenes multiplied up to 2·4 log CFU g?1 in compost with initial moisture content of 30% and was detected up to 28 days for all seasons, whereas populations of both E. coli O157:H7 and Salmonella increased by c. 1 log in compost with initial moisture content of 30% during winter months only. No pathogen growth in nonautoclaved compost was detected. Conclusion: Bacterial species, temperature, light intensity and moisture content affected the growth potential and survival of pathogens in compost when the population of background microflora was low. Significance and Impact of the Study: Keeping compost as dry as possible and maintaining certain levels of background microflora may be critical to prevent the growth of pathogens.  相似文献

Three insect growth regulator insecticides and an entomopathogenic strain of Bacillus thuringiensis (GC327), products effective against the mushroom sciarid, Lycoriella auripila, were compared for their effect on mushroom cropping. Cyromazine and diflubenzuron were applied as a surface drench to mushroom compost before or after pasteurisation (at filling or spawning, respectively); admixed into casing material (at casing); or at a combination of these times. Hexaflumuron and GC327 were applied only at filling and casing, respectively. The presence of the target pest, L. auripila, had no effect on treatment trends, although it was accounted for in the analysis by use of a yield model. The trial was notable for the disparate effects that cyromazine and diflubenzuron casing treatments had on mushroom cropping. Cyromazine treatments that included application at casing resulted in increases in yield, compared to the untreated control whereas, with diflubenzuron, the opposite was true, with treatment at casing alone causing the greatest reduction overall (10%). GC327 applied at casing was also conspicuous for giving a 13% increase in yield. Treating the crop at casing with either cyromazine or GC327, therefore, resulted in a 15% or 24% increase in yield, respectively, compared to a similar treatment with diflubenzuron. Hexaflumuron applied at filling caused increases in yield compared to application of cyromazine at filling and cyromazine or diflubenzuron at spawning. There were also effects on crop timing. The addition of a cyromazine casing treatment normally caused the distinct flushes of mushrooms to be produced significantly earlier than the untreated control (up to 2.5 days), as did GC327. With diflubenzuron, the earlier flushes were only produced by those treatments that did not include a casing application. The combinations that included a casing treatment with diflubenzuron initially produced mushroom flushes earlier than the untreated control. They became either synchronous with the control or they were delayed. From the crop tolerance perspective, therefore, cyromazine and GC327 would be the sciarid control products of choice for a commercial mushroom grower.  相似文献

Aims

To assess, in a cleaned and disinfected barn environment, the efficacy of an animal disinfection strategy to reduce the livestock‐associated methicillin‐resistant Staphylococcus aureus (LA‐MRSA) prevalence in sows, their offspring and the barn environment.

Methods and Results

On each farm, six sow rounds were sampled; sows were divided into either a test or control group. Per round, 20 sows and 40 of their piglets were sampled at different time points together with the barn environment. The disinfection strategy of the test groups consisted of washing the sows with a shampoo followed by disinfection of the skin with a solution containing chlorhexidine digluconate and isopropanol. On the first day of disinfection and 6 days after stopping the disinfection, a significant decrease (P < 0·01) of on average 68 and 66% in sow MRSA prevalence was observed on both farms, whereas no decrease was seen in the control groups. Just before weaning, 21–28 days after the end of the disinfection strategy, the difference in MRSA prevalence between both groups was reduced to 4% and no longer significant (P = 0·20). The MRSA prevalence of the piglets in the test groups was significantly lower (26%; P < 0·01) 6 days after the end of disinfection. Just before weaning, this difference was reduced to 5% but still significant (P < 0·01). In the swine nursery unit, no significant difference (P = 0·99) was seen between both groups. Based on semi‐quantitative counts, a relationship (r2 > 0·6; P < 0·01) was seen between MRSA contamination in the barn environment and the MRSA prevalence in pigs.

Conclusion

Results show that the tested disinfection strategy reduces temporarily the sow and piglet MRSA status, but does not result in a final reduction in MRSA at weaning or in the nursery unit.

Laboratory tests of bacteria isolated from the body surface, or from the gut, of a saprophagous rhabditid nematode Caenorhabditis elegans infesting mushrooms (Agaricus bisporus) showed that some bacteria enhanced nematode reproduction and that others inhibited it. As some bacteria were shown to inhibit mycelial growth of the mushroom, the effects of Acinetobacter calcoaceticus var. anitratus, Enterobacter cloacae and Serratia liquefaciens, either alone or in combination with C. elegans, on the flushing patterns, quality and yield of A. bisporus (strain Horst U3) were studied. Bacteria alone had little effect on flushing patterns whereas C. elegans delayed the onset of mushroom production and significantly disrupted the growth pattern of crops, with mushrooms appearing more regularly and not within obvious flushes. Inoculation with bacteria resulted in ‘browning’ of mushrooms that was even more pronounced in C. elegans treatments. Characteristic distortion of sporophores was observed only in the presence of C. elegans. Nematodes commonly colonised sporophores. Bacteria affected the size of nematode populations both on the sporophores and in the casing. Significant yield loss occurred; up to 10% when bacteria were inoculated, up to 27.8% when C. elegans was inoculated, and up to 35% with both bacteria and nematodes. Synergism between C. elegans and A. calcoaceticus var. anitratus was observed; the combination resulted in significantly greater reduction in mushroom yield than any other treatment. It is concluded that bacteria contribute to yield loss and quality deterioration in A. bisporus but that the effects are far greater in the presence of C. elegans.  相似文献

Abstract

Growth, nutrient content and nodulation response of cowpea plants (Vigna unguiculata L. Walp) inoculated with a Arbuscular Mycorrhiza (AM) fungus (Glomus etunicatum) and Bradyrhizobium (BR) strain IRC 25B peat-based were assessed on an alfisol in a two-cropping cycle experiment conducted in the greenhouse. A total of 5 kg sieved unsterilized topsoil plastic pots was amended with compost consisting of 2.4% N, 1.7% P, 2.7% Ca, 0.4% Mg and 0.7% Fe. Analysis from this first cropping cycle showed that all cowpea plants were infected with mycorrhiza in both AM inoculated and uninoculated treatments. However, all the AM inoculated plants had higher infections than the uninoculated cowpea plants. Nevertheless, nodule number and nodule weight of cowpea plants generally increased in response to compost application when used alone, or when combined with BR or AM; except for nodule weight of BR + Compost treatment. At 13 weeks after planting, the plants were harvested for a second cropping cycle experimental analysis. Results showed higher mycorrhizal infections in all the treatments inoculated with AM. However, infection was highest in cowpea plants treated with AM + BR + Compost, followed by those treated with AM + BR. This shows an increase in the number of AM propagules during the period of cropping. All other parameters measured were found generally lower in their mean values compared to the first cropping cycle. It was observed in this study that compost applications with AM inoculation could substitute for inorganic fertilizer. Thus, tropical countries should direct their efforts towards making the best use of AM to improve conditions for the peasant farmers that account for over 70% agricultural productivity in the region.  相似文献

One of the main impacts of urban sprawl in rapidly growing countries has been contamination of coastal environments by waterborne pathogens, posing a critical risk to ecosystem and human health. Microbial source tracking (MST) has been a robust tool to identify the origin of these pathogens globally. This study compared the occurrence of a human-associated Bacteroides marker (BT-α) with faecal indicator bacteria (FIB) in an urban estuary (Golden Horn, Istanbul, Turkey). Faecal coliform (culture method), enterococci (both culture and qPCR method) concentrations and physicochemical variables were compared with the BT-α concentrations in monthly collected samples for a year (n = 108). Enterococci concentrations detected by culture and qPCR were positively correlated (r = 0·86, P < 0·01) suggesting that qPCR can be an alternative method for monitoring. BT-α marker was positive for 30% of the samples and positively correlated with enterococci (r = 0·61 and r = 0·64 for culture and qPCR methods respectively, P < 0·01). Rainfall had a moderate positive correlation with all faecal/MST indicators suggesting combined sewer overflows also severely impacted estuarine water quality. The high FIB and BT-α concentrations at upper estuary suggested that faecal pollution mainly originated from the peri-urban settlements around two creeks entering the estuary.  相似文献

Legionella longbeachae has been frequently identified in composted plant material and can cause Legionnaires’ disease (LD). We wanted to determine how frequently L. longbeachae DNA was present on gardeners’ gloves, and how long L. longbeachae could persist on inoculated gloves and masks. Volunteers completed a survey of gardening practices and their gardening gloves were tested for L. longbeachae DNA by qPCR. The persistence of viable L. longbeachae was assessed by timed subcultures after inoculation of gardening gloves and masks. Gloves but not masks were used regularly. L. longbeachae was detected on 11 (14%; 95% CI 8–24%) gloves. Viable organisms were recovered from 25–50% of inoculated cotton, leather and PU coated gloves but not rubber gloves after 8 h incubation. There was a difference in dose-response curve slopes by glove material (P = 0·001) and time to 50% sterility (P = 0·036). There were differences in persistence of L. longbeachae between mask types from analysis of the slopes and 50% sterility on the decay curves (P = 0·042, P < 0·001 respectively). Gardening gloves and masks may act as a vector for transmission of L. longbeachae during gardening. Washing gardening gloves and prompt disposal of masks could reduce risk of LD.  相似文献

After grass carps Ctenopharyngodon idellus were injected with cortisol, with (CBC) and without (C) a cocoa butter carrier, the effects of both slowly and rapidly acting exogenous cortisol on their non‐specific immune functions were investigated. On the one hand, after injection with CBC, the cortisol concentration and lysozyme activity in fish serum were enhanced and were sustained at high levels for a long period (30 days). The killing activity in the serum declined with time, and phagocytosis of head kidney macrophages diminished significantly (P < 0·05 or P < 0·01). The leukocrit values in the high dose group (31·8 mg cortisol fish?1) increased over time, however, with the maximum average being 5·6% at day 30. The spleen mass index in the high dose group was 0·93 × 10?3 after 30 days, notably lower (P < 0·05) than that in the control group. In addition, a decrease in resistance to Aeromonas hydrophila infection in cortisol‐treated fish was shown, with the final cumulative mortalities being 54·5 and 66·7% in the low and high dose groups, respectively. On the other hand, there was a decrease in both serum cortisol concentration and lysozyme activity of the experimental fish within 2 weeks after injection with C, where plasma bactericidal activities in the high dose group (31·8 mg cortisol fish?1) were remarkably lower (P < 0·01) than those in the control group at each sampling, but were increased slightly over time. The results of which were different from those in the CBC trial. Phagocytic activity of head kidney macrophages and spleen mass index decreased significantly (P < 0·05), while there were increases in leukocrit value and cumulative mortality due to A. hydrophila. The results of which were similar to those in the CBC trial. This study indicated that the injection of cortisol depressed the non‐specific immune functions of the grass carp and increased its susceptibility to disease.  相似文献

Supplementation of mushroom compost at spawning and at casing with various refined and crude seed oils resulted in 1 to 1.5 lb/ft2 increases in mushroom yield. Supplementation at casing with ground seeds or protein-oil combinations caused 2 to 2.5 lb/ft2 increases in mushroom yield. Further evidence is presented for a relationship between lipid metabolism and the initiation of fruiting in the cultivated mushroom, Agaricus bisporus (Lange) Sing. Preliminary results suggest the possible involvement of sterols in the fruiting stimulation.  相似文献

Cleome viscosa is an emerging weed with the potential of interfering with okra and influencing pests of okra. Screen house studies were conducted on the phenology of C. viscosa, its interference with okra and its interaction with root-knot nematode-infected okra. Seedlings of C. viscosa were monitored in pots for growth, yield and dry matter accumulation for 14 weeks. C. viscosa was planted with okra at densities 0, 2, 4, 6, 8 and 10 weeds per okra plant and observed for 11 weeks. Data were collected on growth, yield and dry matter of okra. Plants were inoculated with 2,500 M. incognita eggs per pot while control plants were not inoculated. C. viscosa attained 91.7 cm height and accumulated 7.8 g/plant biomass at 14 weeks after planting. The percentage reduction in okra plant height, plant dry weight and fruit yield due to interference at lowest cleome density (2 plants/pot) was 33.7, 83.6 and 82.1%, respectively. Nematode reproductive factor was significantly lower for okra alone (4.9) compared to okra with cleome (7.6). This study shows that C. viscosa is a fast-growing weed that suppressed the performance of okra even at low density, is a good host to M. incognita and increased the population of the nematode in soil.  相似文献

In a mushroom crop (Agaricus bisporus) affected by a very low level of sciarid fly (Lycoriella auripila) infestation, the effects of an indigenous isolate of insect-parasitic nematode (Steinernema feltiae) and of two commonly used insecticides (diazinon and diflubenzuron) were studied. When compared with untreated plots, nematodes applied to the casing had no adverse effects on mushroom yields whereas insecticides decreased yields. At a rate of 3 × 106 infective juveniles per tray (surface area = 0.56 m2), S. feltiae elicited increases of 28.5% and 19% in the mean total numbers and weights of mushrooms respectively. Treatment only with diflubenzuron resulted in 14.6% and 6% reductions in mean total numbers and weights of mushrooms, respectively; treatment with both diazinon and diflubenzuron caused 18.5% and 9.4% losses. Application of nematodes generally reduced the mean weight per mushroom whereas insecticides increased it; nematodes delayed the onset of mushroom production (first flush) whereas diflubenzuron delayed the third and fourth flushes. Nematode contamination of sporophores was minimal when S. feltiae was applied at casing. Although their numbers declined with time, the nematodes persisted, in the casing layer, throughout the cropping period of seven weeks. It is concluded that yield benefits associated with nematode application can result mainly from nematode effects on A. bisporus and not solely from suppression of a damaging pest population.  相似文献

Effects of combinations of insecticides and herbicides on the growth and yield of cauliflower plants (cvs No. 110, Garant and Strong) were determined in two field experiments with seed drilled to give a stand of c. 300000 plants/ha. Granular formulations of chlorfenvinphos or fonofos were applied by the bow-wave method at 2 kg a.i./ha to control cabbage root fly (Delia brassicae). Weeds were successfully controlled with combinations of trifluralin, incorporated into the soil at 0·6 or 1·2 kg a.i./ha before drilling, and propachlor, applied pre-emergence at 2·2 or 4·4 kg a.i./ha. Fonofos, with and without herbicides, significantly lessened (P < 0·01) numbers of seedlings that would survive to harvest. Herbicides and chlorfenvinphos generally did not significantly affect the emergence of healthy seedlings of any cultivar. Combinations of herbicides and chlorfenvinphos were compatible for control of target species. Chlorfenvinphos reduced root damage by at least 50% but did not significantly increase (P > 0·05) the total or marketable weight of curds. Numbers of curds were not usually affected and consistent effects of treatments on maturation periods of the three cultivars were not detected.  相似文献

This study investigated the effects of dietary Bifidobacterium bifidum (BFD) and mannan-oligosaccharide (MOS), as a synbiotic, on the production performance, gut microbiology, serum biochemistry, antioxidant profile and health indices of broiler chicken. Six dietary treatments were T1 (negative control), T2 (positive control-20 mg antibiotic BMD kg−1 diet; BMD: bacitracin methylene disalicylate), T3 (0·1% MOS + 106 CFU BFD per g feed), T4 (0·1% MOS + 107 CFU BFD per g feed), T5 (0·2% MOS + 106 CFU BFD per g feed) and T6 (0·2% MOS + 107 CFU BFD per g feed). Significantly (P < 0·01) better growth performance and efficiency was observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to BMD and control birds. Supplementation with 0·2% MOS along with either 106 or 107 CFU BFD per g feed reduced (P < 0·01) the gut coliform, Escherichia coli, total plate count, and Clostridium perfringens count and increased the Lactobacillus and Bifidobacterium count. Significantly (P < 0·01) higher serum and liver antioxidant enzyme pool, serum HDL cholesterol and lower serum glucose, triglyceride, total cholesterol, cardiac risk ratio, atherogenic coefficient and atherogenic index of plasma were observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to control or BMD supplemented birds. Better production performance, gut microbial composition, serum biochemistry, antioxidant profile and health indices were depicted by broiler chicken supplemented with 0·2% MOS and 106 CFU BFD per g of feed.  相似文献

Diflubenzuron and bendiocarb treatments of compost and casing soil at a rate of 1 g a.i./m2 each, resulted in comparable or even improved compost and casing soil colonisation over endosulfan treatments (at a rate of 1.5 g a.i./m2). Alternating diflubenzuron/bendiocarb treatments were preferable to the reverse treatments, which tended to diminish total yield of mushrooms significantly by 1.4–3.9 kg/m2. This effect was due to the casing soil treatment with diflubenzuron and not due to the compost treatment with bendiocarb after spawning. The effectiveness of these substitutes for endosulfan was tested on five commercial mushroom farms, where a similar or improved control of Megaselia halterata (Diptera: Phoridae) was obtained over control (endosulfan) treatments. If present, Lycoriella auripila (Diptera: Lycoriidae) was very effectively controlled by the substitute insecticides compared with endosulfan. Relatively high numbers of M. halterata were occasionally observed in endosulfan-treated houses, indicating that a certain level of resistance towards this insecticide may already have developed. This assessment of tolerance in M. halterata emphasised the need for substitute insecticides with different modes of action, in addition to environmental reasons.
