The growth of a mixed methane-utilizing culture in a continuous flow fermenter has been studied under both methane and oxygen limitation. Small additions of methanol have been shown to inhibit the methane-utilizing moiety in the culture and it has been shown that the Hyphomicrobium sp. in the mixed culture removes any inhibitory methanol. The interaction between the methane-utilizing Pseudomonas sp., and the Hyphomicrobium sp. has been explained and a model of the continuous mixed culture under oxygen limitation has been formulated. Qualitative predictions of transient phenomena by the model have been verified experimentally.  相似文献

Cytoskeletal proteins are important mediators of cellular organization in both eukaryotes and bacteria. In the past, cytoskeletal studies have largely focused on three major cytoskeletal families, namely the eukaryotic actin, tubulin, and intermediate filament (IF) proteins and their bacterial homologs MreB, FtsZ, and crescentin. However, mounting evidence suggests that these proteins represent only the tip of the iceberg, as the cellular cytoskeletal network is far more complex. In bacteria, each of MreB, FtsZ, and crescentin represents only one member of large families of diverse homologs. There are also newly identified bacterial cytoskeletal proteins with no eukaryotic homologs, such as WACA proteins and bactofilins. Furthermore, there are universally conserved proteins, such as the metabolic enzyme CtpS, that assemble into filamentous structures that can be repurposed for structural cytoskeletal functions. Recent studies have also identified an increasing number of eukaryotic cytoskeletal proteins that are unrelated to actin, tubulin, and IFs, such that expanding our understanding of cytoskeletal proteins is advancing the understanding of the cell biology of all organisms. Here, we summarize the recent explosion in the identification of new members of the bacterial cytoskeleton and describe a hypothesis for the evolution of the cytoskeleton from self-assembling enzymes.  相似文献

In study of the oxygen effect in relation to the growth curve ofEscherichia coli B, its size was found to be correlated to the stage of growth at which the bacteria were irradiated. The lowest dose reduction factor value (DRF = 1.5) was found with bacteria irradiated at the outset of the logarithmic phase and the highest value (DRF = 2.7) with bacteria in the stationary phase. This phenomenon was observed both in a culture cultivated under aerobic conditions prior to irradiation and in a culture cultivated under anaerobic conditions. The cultivation conditions influenced only the radiosensitivity of the bacteria.  相似文献

This article analyses the Surface-Limited Growth Model put forward to explain the very tight synchrony, over more than ten division cycles, obtained experimentally by subjecting a growing bacterial culture to alternating periods of starvation and dilution, using inorganic phosphate as the limiting substrate. The Model states that when an essential nutrient is in limited supply, the rate of growth of an individual cell will be proportional to its surface area (and the current concentration of the limiting substance) rather than to its volume. This decrease in dimensionality from volume to surface is expected to favor the smaller cells and so result ultimately in a narrower size distribution. The Surface-Limited Growth Model deals with cell growth under unusual nutritional conditions, and its predictions depend on how the cell replication cycle is assumed to behave under these same circumstances. Two alternatives are considered: the volume at which cells divide is the same during the starvation phase as during steady-state exponential growth, and the cells adjust immediately to the changing growth rate. In the latter case, we have tested both C + D constant with time and C + D variable (where C + D is the time between initiation of chromosome replication and the corresponding cell division), the incremental value at any instant being computed separately for each individual cell from its current effective growth rate. The simulation results are of two sorts depending on the auxiliary assumptions used. Either the dilution-starvation cycles have no effect whatsoever on the cell volume distribution, or the width of the distribution decreases gradually with time, approaching zero slowly and asymptotically, but the mean cell volume decreases as well--directly contradicting experimental observations. We conclude that the Surface-Limited Growth Model is incapable of explaining the synchronization of cells by periodic starvation of a growing bacterial culture.  相似文献

Mathematical studies for ecosystems involving 2 predators competing for a growing prey population have shown that the 2 competitors can coexist in a state of sustained oscillations for a range of values of the system parameters. For the case of 1 suspension-feeding protozoan population, recent experimental observations suggest that the predator-prey interaction is complicated by the ability of the bacteria to grow on products produced by the lysis of protozoan cells. This situation is studied here for the case where 2 suspension-feeding protozoan populations compete for a growing bacterial population in a chemostat. Computer simulations show that the 2 protozoan populations can coexist over a range of the operating parameters. Some necessary conditions for coexistence are presented as are some speculations regarding the possible physical explanations of results.  相似文献

An approach to the optimization of product yield in an inducible inclusion body-producing system is presented. Following induction by indoleacrylic acid (IAA) of a trpLE-HIVgp41 fusion protein, we found a large increase in culture turbidity and single-cell dry weight. After an initial transition phase, new and constant values for specific growth rate, single-cell light turbidity, and single cell dry weight were achieved, allowing for the determination of optimal conditions for product formation.  相似文献

Summary Soil was taken from the top 10 cm of a soil column that removed halogenated aliphatic hydrocarbons in the presence of natural gas. This soil was used as an enrichment inoculum to determine that the removals seen in the soil column were in fact of a microbiological nature. Methane served as the source of carbon and energy and was consumed immediately by the enrichments. After several transfers of the enrichments, a stable consortium of at least three bacterial types was obtained. The predominant bacterium was a non-motile, gram-negative coccus. This stable consortium was able to remove chlorinated methanes, ethanes, and ethylenes when grown with methane and oxygen in the headspace. Methane was required for the removals to be observed. Acetylene inhibited the removals, which further suggests the involvement of methanotrophs. Benzene and toluene were removed by the mixed culture with or without methane in the headspace. Fatty acid analysis of the mixed culture resulted in a profile that indicated that the predominant organism was a type II methanotroph. This study provides further evidence that methanotrophic bacteria are capable of cometabolizing a wide range of chlorinated methanes, ethanes, and ethylenes.  相似文献

Summary The studies reported here were undertaken to determine if Erysipelothrix crude extract (CE) could elicit consistent, detectable alterations in synovial cells in vitro. Synovial cells of rabbits were cultured in vitro and data about normal synovial cells in vitro were presented. Such cells were consistently and characteristically affected by exposure to small amounts of CE; an effect that was neutralized in the presence of specific anti-CE antiserum. With an increase in the amount of CE added to the cells in culture, a decreasing number of cells was formed but protein content remained relatively constant. A modified membrane-Bovis agar, containing a reduced quantity of sodium azide, for the isolation and enumeration of Streptococcus bovis is described and evaluated.
