Chemical speciation of binary complexes of Pb(II) and Cd(II) ions with maleic acid have been studied pH metrically in the concentration range of 0–50% v/v ethylene glycol (EG)–water mixtures maintaining an ionic strength of 0.16 molL?1 at 303 K. Alkalimetric titrations were carried out in different relative concentrations of metal and maleic acid. Stability constants of various models of binary complexes were refined with MINIQUAD75. The best-fit chemical models were selected based on statistical parameters and residual analysis. The species detected are ML2, ML3, and ML2H for Pb(II) and Cd(II). The chemical speciation, metal bioavailability, and transportation are explained based on the distribution diagrams.  相似文献

Derivatives of Co(II), Ni(II), Cu(II) and Zn(II) with 3′AMP and 2′AMP were synthesized and characterized by IR UV-Vis and fluorescence spectroscopy. There seems to be bonding of the metal ion to the base in all cases. The activation test, using the complexes as allosteric labels, was carried out with rabbit muscle glycogen phosphorylase b, but the enzyme was not activated, confirming that the phosphate group must necessarily be bonded to position 5′ of the ribose in order to activate this enzyme.  相似文献

N-Salicyloyl-N-p-hydroxythiobenzohydrazide (H2STPH) and N-benzoyl-N-thiobenzohydrazide (H2BTBH) and their Co(II), Ni(II), Cu(II) and Zn(II) complexes were prepared and characterized by physicochemical studies. IR and NMR spectral studies imply dibasic tetradentate behaviour of the ligands bonding through `thiolato' sulfur, enolic oxygen and the two hydrazinic nitrogens in a polymeric fashion. The electronic spectra indicate [Ni(STPH)(H2O)2], [Co(STPH)(H2O)2] to be distorted octahedral while [Cu(BTBH)] has a square-planar geometry. In vitro antitumor results of the ligand and the complexes on P-815 (murine mastocytoma) and L-929 (murine fibroblast) indicate that these compounds show significant inhibition of 3H-thymidine and 3H-uridine incorporation in DNA and RNA, respectively, in these tumor cells at dose levels of 1, 2.5 and 5 g cm–3. Antitumor studies suggest that [Cu(BTBH)] has significant dose dependent inhibitory effect on DNA synthesis. In vivo administration of [Cu(BTBH)] and [Ni(STPH)(H2O)2] resulted into prolongation of life span of Dalton's Lymphoma (DL) bearing mice.  相似文献

A new ligand, L, bearing four cyanoethyl pendant groups has been synthesized by reaction of the precursor ligand L1 with acrylonitrile. The X-ray crystal structure of ligand L reveals the presence of a nanotubular structure in the solid state connected by intermolecular π,π-stacking interactions between adjacent pyridine rings. The coordination capability towards transition metal ions [Co(II), Ni(II) and Cu(II)] has been investigated starting from the hydrated nitrate and perchlorate salts of the metals. The new ligand L and the metal complexes obtained were characterized by elemental analysis, FAB MS, conductivity measurements, magnetic studies, IR and UV-vis spectroscopy. Furthermore, the crystal structure of ligand L and of the complexes [CoL][Co(NO3)4] · CH3CN (1), [NiL](NO3)2 (3), [NiL](ClO4)2 · CH3CN · 3H2O (4), [CuL][Cu(NO3)3(H2O)2](NO3) · H2O (5) and [CuL](ClO4)2 · 2CH3CN (6) were determined. The nitrate ions in the complexes are located near the pyridine rings and π,π-stacking interactions between pyridine rings, nitrate ions and nitrile groups have been found.  相似文献

A new ligand N-Nicotinoyl-N-o-hydroxythiobenzhydrazide (H2Notbh) forms complexes [Mn(Notbh)(H2O)], [M(Notbh)] [M=Ni(II) Cu(II) and Zn(II)] which were characterized by various physico-chemical techniques. All the metal complexes were observed to inhibit the growth of tumor in vitro, whereas, ligand did not. In vivo administration of these complexes resulted in prolongation of survival of tumor bearing mice. Tumor bearing mice administered with metal complexes showed reversal of tumor growth associated induction of apoptosis in lymphocytes. The paper discusses the possible mechanisms and therapeutic implication of the H2Notbh and its metal complexes in tumor regression and tumor growth associated immunosuppression.  相似文献

Abstract

The solute–solvent interactions of glycine, 1,10-phenanthroline and 2,2-bipyridyl have been studied in 0–60% v/v ethylene glycol–water media by a pH metric method. The protonation constants were estimated with the computer program MINIQUAD75. Selection of the best fit chemical model of the protonation equilibria is based on the standard deviation in protonation constants and residual analysis using a sum of squares of residuals in all mass-balance equations. The observed linear variation of protonation constants with the inverse of dielectric constant of the solvent mixture can be attributed to the dominance of the electrostatic forces. The distribution of species, protonation equilibria and effects of influential parameters on the protonation constants are also presented.  相似文献

Abstract

Chemical speciation of Co(II), Ni(II), Cu(II) and Zn(II) complexes of 2,3-dihydroxybenzoic acid in 0.0-60.0% v/v 1, 2-propanediol-water mixtures maintaining an ionic strength of 0.16 mol dm-3 at 303±0.1 K has been studied pH metrically. The predominant complexes formed are ML, ML2 and ML2H2 for Co(II), Ni(II) and Zn(II) and ML, ML2, ML2H and ML2H2 for Cu(II). Models containing different numbers of species were refined by using the computer program MINIQUAD75. Selection of the best fit chemical models was based on statistical parameters and residual analysis. The trend in variation of complex stability constants with dielectric constant of the medium is explained on the basis of electrostatic and non-electrostatic forces. Distributions of species, formation equilibria and effect of influential parameters on the stability constants have been presented. The possible structures of the various species are elucidated on the basis of the analysis of the pH-metric data.  相似文献

A rather widespread idea on the functional importance of sphingolipids in cell membranes refers to the occurrence of ordered domains enriched in sphingomyelin and ceramide that are largely assumed to exist irrespective of the type of N-acyl chain in the sphingolipid. Ceramides and sphingomyelins are the simplest kind of two-chained sphingolipids and show a variety of species, depending on the fatty acyl chain length, hydroxylation, and unsaturation. Abundant evidences have shown that variations of the N-acyl chain length in ceramides and sphingomyelins markedly affect their phase state, interfacial elasticity, surface topography, electrostatics, and miscibility, and that even the usually conceived “condensed” sphingolipids and many of their mixtures may exhibit liquid-like expanded states. Their lateral miscibility properties are subtlety regulated by those chemical differences. Even between ceramides with different acyl chain length, their partial miscibility is responsible for a rich two-dimensional structural variety that impacts on the membrane properties at the mesoscale level. In this review, we will discuss the miscibility properties of ceramide, sphingomyelin, and glycosphingolipids that differ in their N-acyl or oligosaccharide chains. This work is a second part that accompanies a previous overview of the properties of membranes formed by pure ceramides or sphingomyelins, which is also included in this Special Issue.  相似文献

The chief motive behind this research is the interest provoked by the presence of metal ions as necessary stabilizers of the negative charges of phosphate groups in nucleic acids. The effect that the presence of different metal ions produces on the band principally assigned to the nu(s) PO(3)(2-) mode has been studied using FT-IR and FT-Raman spectroscopy. The results obtained reveal the diagnostic capacity of these techniques in determining the type of metal ion interaction with respect to the mononucleotides that form DNA and RNA, providing a tool for improving the knowledge of the stabilizing or destabilizing effects of these ions on such macromolecules. The metal complexes of the ribonucleotides 5'-CMP and 5'-GMP with Mg(II), Ca(II), Sr(II), Ba(II), Cr(III), Co(II), Cu(II), Zn(II), Cd(II), Al(III) and Ga(III) were obtained in this study. After studying and analyzing the IR and Raman spectra of all these complexes and comparing them with the spectra of the corresponding disodium salts, it was verified that, independently of the type of nucleotide involved, the presence of the metal in the vicinity of the phosphate group produces an alteration in the aforementioned nu(s) PO(3)(2-) band. This effect is related to the type of interaction that the phosphate group has with the metal. Three components are observed: (1) one near 983-975 cm(-1) (detectable in IR and Raman), associated with phosphate groups in an electrostatic type of interaction with the metal ion, separated by two or more water molecules; (2) another near 989-985 cm(-1) (only in IR), associated with phosphate groups in indirect interaction through the water molecules of the coordination sphere of the metal ions; and (3) the IR and Raman bands near 1014-1001 cm(-1), which represent phosphate groups directly bonded to the metal ion. These results are supported by the behavior of 5'-CMP in aqueous solution in the presence of Mg(II) ions.  相似文献

We have performed 20?ns of fully atomistic molecular dynamics simulations of Hen Egg-White Lysozyme in 0, 10, 20, 30, and 100% by weight of glycerol in water to better understand the microscopic physics behind the bioprotection offered by glycerol to naturally occuring biological systems. The solvent exposure of protein surface residues changes when glycerol is introduced. The dynamic behavior of the protein, as quantified by the incoherent intermediate scattering function, shows a nonmonotonic dependence on glycerol content. The fluctuations of the protein residues with respect to each other were found to be similar in all water-containing solvents, but different from the pure glycerol case. The increase in the number of protein–glycerol hydrogen bonds in glycerol–water binary mixtures explains the slowing down of protein dynamics as the glycerol content increases. We also explored the dynamic behavior of the hydration layer. We show that the short length scale dynamics of this layer are insensitive to glycerol concentration. However, the long length scale behavior shows a significant dependence on glycerol content. We also provide insights into the behavior of bound and mobile water molecules.  相似文献

The oxovanadium(IV), acetatomanganese(III), chloroiron(III), nickel(II), copper(II), zinc(II) and palladium(II) of 3,3′-(1,2-phenylenediimino)diacrolein were prepared and investigated by means of mass, electronic, vibrational, NMR and ESR spectroscopy as well as magnetic susceptibility measurements. The acetatomanganese(III) and chloroiron(III) complexes were confirmed to be of high spin type. The absorption bands appearing in the energy range greater than 23 000 cm−1 were attributed to π→π* transitions within a ligand molecule and charge- transfer transitions from metal to ligand. The metal complexes assume the square-planar configuration type since the ligand-field bands were detected in the 12 700–18 500 cm−1 region. Strong bands appearing at 1601 and 1627 cm−1 were assigned to the CC and CO stretching vibrational modes, respectively, and were shifted to lower frequency upon metal-coordination. A VO stretching band was observed at 982 cm−1 for the oxovanadium(IV) complex and a CO stretching band was observed at 1547 cm−1 for the acetatomanganese(III) complex. Upon complex formation the amine proton signal is found to vanish and the aldehydic methine proton signal in the lowest field is shifted upfield for the nickel(II), zinc(II) and palladium(II) complexes. 13C NMR spectra support the coordination structure of the complexes which is revealed by 1H NMR spectra. As judged by the spin Hamiltonian parameters, the oxovanadium(IV) complex is of a square- planar type with an unpaired electron in the dxy orbital and the copper(II) complex assumes a distorted square-planar coordination due to the presence of five- and six-membered chelate rings with an unpaired electron in the dx2−y2 orbital.  相似文献

The one pot aqueous reaction of M(ClO4)2 (M = Cu2+ or Ni2+) with N-methylbis[2-(2-pyridylethyl)]amine (MeDEPA) and N,N′-dimethyl-N,N′-bis(2-pyridylmethyl)ethylenediamine (bpmen) and 1,4,7,10-tetraazacyclododecane (cyclen) in presence of sodium dicyanamide (Nadca) yielded dicyanamido-bridged polynuclear complex {[Cu(MeDEPA)(μ-1,5-dca)]ClO4}n (1), and two dinuclear complexes [Cu2(bpmen)2(μ-1,5-dca)]2(ClO4)5dca (2) and [Ni(cyclen)(μ-1,5-dca)]2(ClO4)2 (3). These complexes were characterized by IR and UV-Vis spectroscopy. Room temperature single-crystal X-ray studies have confirmed that the Cu(II) centers in 1 and 2 adopt geometries that are more close to trigonal bipyramidal (TBP) in 1 and close to square pyramidal (SP) in 2, whereas in 3, the Ni(II) centers are located in octahedral environment with doubly bridged μ-1,5-dca bonding mode. The intermolecular M···M distances in these complexes are in the range of 7.3-8.6 Å. Variable temperature magnetic susceptibility studies have confirmed that the dca-bridges mediate very weak antiferromagnetic interaction between the M(II) centers with J values of −0.35, −0.18 and −0.43 cm−1 for 1, 2 and 3, respectively. The results are compared and discussed in the light of other related bridged μ-1,5-dca Cu(II) and Ni(II) complexes.  相似文献

The interaction of two structurally close flavanones: taxifolin and naringenin with copper(I) ions and its effect on the distribution of flavonoids and the corresponding ions in a biphasic system octanol–water have been studied. It has been shown that these polyphenols form complexes with copper ions of different stoichiometric ratio depending on the pH of medium (5.4, 7.4, and 9.0). The interaction of the flavonoids with copper ions leads to an increase in the fraction of polyphenols in the water phase at all pH values examined. The fraction of metal ions in octanol in the presence of both taxifolin and naringenin is maximal in the range of neutral pH values. The parameters obtained in the study, such as the partition coefficient and the coefficient of distribution in a biphasic system octanol–water (logP and logD) form the physicochemical basis necessary for the estimation of the bioavailability of flavonoids and the corresponding metal ions upon their combined consumption.  相似文献

Abstract

The X-ray structures of the isomorphous Mg2+, Ca2+, Mn2+ and Co2+ complexes of ATP have been determined. The metal ions are wrapped in hexa-coordination by the α, β and γ phosphate groups of two ATP molecules thus blocking the interaction of the metal ions with the adenine base. A second metal ion which is fully hydrated, M(H2O)2+ 6, is engaged in a strong hydrogen bond with the γ phosphate group of ATP and suggests a possible step in facilitating the cleavage between the β and γ phosphates in phosphoryl transfer reactions.  相似文献

The complexes [Cu(samen)Cu(L)] and [Cu(samen)Ni(L)2] (Lbpy, phen) have been synthesized by the reaction of sodium N,N′-ethylenedisalicylamidatocuprate(II) pentahydrate (Na2- [Cu(samen)]·5H2O), a divalent metal ion, and 2,2′- dipyridyl or 1,10-phenanthroline. Cryomagnetic data for the CuCu complexes did not fit the Bleaney- Bowers equation; but the data did fit a modified Bleaney-Bowers equation
with a large negative J and a significant negative θ, suggesting that a considerable magnetic interaction operates between essentially planar [Cu(samen)Cu(L)] molecules. The magnetisms of the CuNi complexes were well interpreted in terms of the susceptibility equation based on the Heisenberg model. An antiferromagnetic spin-exchange interaction (J= −13∼−14 cm−1) was suggested between the metal ions.  相似文献

Abstract

The aluminium complexes of acetic acid (ACT) have been studied using Potentiometric titrations under physiological conditions of temperature (37°C) and ionic strength (0.15 dm?3 dm?3 NaCI) and at different ligand to metal ratios. The variations of pH were measured with the help of a glass electrode calibrated daily in hydrogen ion concentrations. Results obtained within the pH range of 2.6–4.2 were analysed to determine stability constants using the SUPERQUAD program. Different complex combinations were considered during the calculation procedure, and evidence was found for ML2 mononuclear species beside binuclear hydroxo-complexes M2L(OH)2 and M2L(OH)3 and metal ion hydroxides. Speciation calculations based on the corresponding constants were then used to simulate species distributions.  相似文献

Potentiometric, visible, and infrared studies of the complexation of N-(2-acetamido)-2-aminoethanesulfonic acid (ACESH) by Ca(II), Mg(II), Mn(II), Co(II), Zn(II), Ni(II), and Cu(II) are reported. Ca(II), Mg(II), and Mn(II) were found not to complex with ACES?, while Co(II), Zn(II), Ni(II), and Cu(II) were found to form 2:1, ACES? to M2+, complexes, and [Cu(ACES)2] was found to undergo stepwise deprotonation of the amide groups to form [Cu(H?1ACES)22?]. Formation (affinity) constants for the various metal complexes are reported, and the probable structures of the various metal chelates in solution are discussed.  相似文献

Parkinson’s disease (PD) is hallmarked by the abnormal intracellular inclusions (Lewy bodies or LBs) in dopaminergic cells. Amyloidogenic protein α-synuclein (α-syn) and iron (including both Fe(III) and Fe(II)) are both found to be present in LBs. The interaction between iron and α-syn might have important biological relevance to PD etiology. Previously, a moderate binding affinity between α-syn and Fe(II) (5.8 × 103 M−1) has been measured, but studies on the binding between α-syn and Fe(III) have not been reported. In this work, electrospray mass spectrometry (ES-MS), cyclic voltammetry (CV), and fluorescence spectroscopy were used to study the binding between α-syn and Fe(II) and the redox property of the resultant α-syn-Fe(II) complex. The complex is of a 1:1 stoichiometry and can be readily oxidized electrochemically and chemically (by O2) to the putative α-syn-Fe(III) complex, with H2O2 as a co-product. The reduction potential was estimated to be 0.025 V vs. Ag/AgCl, which represents a shift by −0.550 V vs. the standard reduction potential of the free Fe(III)/Fe(II) couple. Such a shift allows a binding constant between α-syn and Fe(III), 1.2 × 1013 M−1, to be deduced. Despite the relatively high binding affinity, α-syn-Fe(III) generated from the oxidation of α-syn-Fe(II) still dissociates due to the stronger tendency of Fe(III) to hydrolyze to Fe(OH)3 and/or ferrihydrite gel. The roles of α-syn and its interaction with Fe(III) and/or Fe(II) are discussed in the context of oxidative stress, metal-catalyzed α-syn aggregation, and iron transfer processes.  相似文献

A phenylthiophenyl-bearing Ru(II) complex of [Ru(bpy)2(Hbptip)](PF6)2 {bpy?=?2,2′-bipyridine, Hbptip?=?2-(4-phenylthiophen-2-yl)-1H-imidazo[4,5-f][1,10]phenanthroline} was synthesized and characterized by elemental analysis, 1H NMR spectroscopy, and electrospray ionization mass spectrometry. The ground- and excited-state acid–base properties of the complex were studied by UV–visible absorption and photoluminescence spectrophotometric pH titrations and the negative logarithm values of the ground-state acid ionization constants were derived to be pK a1?=?1.31?±?0.09 and pK a2?=?5.71?±?0.11 with the pK a2 associated deprotonation/protonation process occurring over 3 pK a units more acidic than thiophenyl-free parent complex of [Ru(bpy)2(Hpip)]2+ {Hpip?=?2-phenyl-1H-imidazo[4,5-f][1,10]phenanthroline}. The calf thymus DNA-binding properties of [Ru(bpy)2(Hbptip)]2+ in Tris–HCl buffer (pH 7.1 and 50?mM NaCl) were investigated by DNA viscosities and density functional theoretical calculations as well as UV–visible and emission spectroscopy techniques of UV–visible and luminescence titrations, steady-state emission quenching by [Fe(CN)6]4?, DNA competitive binding with ethidium bromide, DNA melting experiments, and reverse salt effects. The complex was evidenced to bind to the DNA intercalatively with binding affinity being greater than those for previously reported analogs of [Ru(bpy)2(Hip)]2+, [Ru(bpy)2(Htip)]2+, and [Ru(bpy)2(Haptip)]2+ {Hip?=?1H-imidazo[4,5-f][1,10]phenanthroline, Htip?=?2-thiophenimidazo[4,5-f][1,10]phenanthroline, Haptip?=?2-(5-phenylthiophen-2-yl)-1H-imidazo[4,5-f][1,10]phenanthroline}.  相似文献
