Abstract

Cd(II) adsorption of root exudates from sunflower (Helianthus annuus L.) seedling was investigated by Cd ion-selective electrode, Fourier Transform Infrared spectroscopy, and fluorescence spectroscopy. Root exudates from Helianthus annuus L. had strong adsorption ability toward Cd(II). The adsorption process was pH-dependent and the maximum adsorption capacity, 150.8 mg g?1, was observed at pH 7.0. Root exudates had pK a1 at 4.7 for carboxyl and pK a2 at 9.2 for phenolic, and amino groups. The aliphatic and aromatic (C?H) groups, amide III group, and the C (=O)?O and sulfonate groups were responsible for Cd(II) adsorption. The excitation emission matrix fluorescence spectroscopy showed protein-like substances participated in Cd adsorption and formed strong complexes, with conditional stability constants of 4.70 and 4.32, which is a little lower than that determined by potentiometric methods, 5.13. The strong Cd complexing ability of root exudates implies that root exudates may significantly affect mobility, toxicity, and phytoavailability of Cd. Cd binding of root exudates may be attributed to its interaction with the proteins, polysaccharides, and phenolic compounds in root exudates.  相似文献

This study investigated the characteristics of exudates from mangrove plant Avicennia marina seedling roots under 0, 200 and 600?mM NaCl treatments and their complexation behavior with trace metals using excitation emission matrix (EEM) fluorescence spectrometry. Two fulvic-like fluorescence peaks, namely peak A (Em = 440?nm, Ex = 250?nm, UV fulvic-like compounds) and peak B (Em = 440?nm, Ex = 340?nm, visible fulvic-like compounds) were identified. The fluorescence intensities of peak A and peak B were enhanced by increasing salinity. Furthermore, the fluorescence of both peaks could be quenched by the ions of copper (Cu2+), manganese (Mn2+) and cadmium (Cd2+). Conditional stability constant (logKa) exhibited that binding capacity of both peak A and peak B with trace metals are Cu2+?>?Mn2+?>?Cd2+ in the range from 2.21 to 4.01. Besides, Hill coefficient (n) >1 for Cu2+ but n?<?1 for Mn2+ and Cd2+. The results of high n and high logKa for Cu2+ rather than Mn2+ and Cd2+ indicate that the fulvic-like compounds in root exudates of A. marina have maximum potential for Cu2+ complexation compared to Mn2+ and Cd2+, suggesting the fulvic acids in root exudates of A. marina have strong complexation with Cu2+ rather than Mn2+ and Cd2+.  相似文献

Microorganisms are important for phytoremediation of soil contaminated with heavy metals. In the present study, bacteria Bacillus sp., Pseudomonas sp., Alcaligenes sp., and Flavobacterium sp. isolated from the Zhangshi Irrigation Area were applied to bioadsorbed Cd and Pb in liquid cultures with root exudates of sunflower as a sole carbon source. The experimental data demonstrated that these bacteria had a high potential of enrichment of Cd and Pb, and Bacillus sp. and Alcaligenes sp. had better ability to accumulate Cd or Pb than the others; the distinct bioadsorption of Cd and Pb by bacteria might depend on the physiology of bacteria, categories of heavy metals, and environmental factors (such as pH). In addition, root exudates of sunflower could not only support the growth of these bacteria, but also influence the toxicity and bioavailability of Cd and Pb. Our results indicated that amendment with bacteria isolated from heavy-metal-polluted soil and root exudates could be considered as a potential approach to enhance the phytoremediation of Cd- or Pb-contaminated soil.  相似文献

Pb(II) ion serves as a quencher of anthracene fluorescence both intermolecularly and in intracomplex systems reported to date. The advantages of intensimetric analyses showing increasing signal require the design of mechanistically novel fluorescent chemosensors capable of yielding enhanced fluorescence upon chelation of inherently quenching metals. We synthesized both 2- and 9-derivatives of anthracene bearing the N-methylthiohydroxamate ligand, which is capable of quenching fluorescence in the uncomplexed form and which shows some selectivity for Pb(II). Complexation of the 2-derivative to Pb(II) results in rapid metal ion-catalyzed hydrolysis, rendering this compound useless as a sensor with real-time response. However, complexation of the 9-derivative with Pb(II) results in a 13-fold fluorescence increase, reversible upon dissociation of the metal ion. While blood lead analysis is a major potential application for fluorescent chemosensors, the present compound is insufficiently selective for this use.  相似文献

ABSTRACT Soil heavy metal contamination, a major threat due to industrialization, can be tackled by an efficient and economical process called bioremediation. Mushrooms are employed to accumulate heavy metals from soil due to their high metal accumulation potential and better adaptability. The bioaccumulation potential of Galerina vittiformis was already reported for individual metals. At natural conditions, since soil consists of more than one polluting metal, more focus has to be given to multimetal systems. In this study, multimetal accumulation potential was analyzed using central composite design, and the responses obtained were analyzed using response surface methodology. Heavy metals such as Cu(II), Cd(II), Cr(VI), Pb(II), and Zn(II) were subjected to biosorption at 10–250 mg/kg concentrations along with pH 5–8. The results showed that the preference of the organism for the five metals under study was in the order Pb(II) > Zn(II) > Cd(II) > Cu(II) > Cr(VI) at pH 6.5 under multimetal condition. The study also indicates that the metal interaction pattern in multimetal interaction is a property of their ionic radii. The response surface methodology clearly explains the effect of interaction of heavy metals on the accumulation potential of the organism using three-dimensional response plots. The present work suggests that the fungus Galerina vittiformis could be employed as a low-cost metal removal agent from heavy metal–polluted soil.  相似文献

Opportunity costs seriously limit the large-scale production of bio-organic fertilizers (BOFs) both in China and internationally. This study addresses the utilization of amino acids resulting from the acidic hydrolysis of pig corpses as organic nitrogen sources to increase the density of TrichodermaharzianumT-E5 (a typical plant growth-promoting fungi, PGPF). This results in a novel, economical, highly efficient and environmentally friendly BOF product. Fluorescence excitation-emission matrix (EEM) spectroscopy combined with fluorescence regional integration (FRI) was employed to monitor compost maturity levels, while pot experiments were utilized to test the effects of this novel BOF on plant growth. An optimization experiment, based on response surface methodologies (RSMs), showed that a maximum T-E5 population (3.72 × 108 ITS copies g−1) was obtained from a mixture of 65.17% cattle manure compost (W/W), 19.33% maggot manure (W/W), 15.50% (V/W)hydrolytic amino acid solution and 4.69% (V/W) inoculum at 28.7°C after a 14 day secondary solid fermentation. Spectroscopy analysis revealed that the compost transformation process involved the degradation of protein-like substances and the formation of fulvic-like and humic-like substances. FRI parameters (PI, n, PII, n, PIII, n and PV, n) were used to characterize the degree of compost maturity. The BOF resulted in significantly higher increased chlorophyll content, shoot length, and shoot and root dry weights of three vegetables (cucumber, tomato and pepper) by 9.9%~22.4%, 22.9%~58.5%, 31.0%~84.9%, and 24.2%~34.1%, respectively. In summary, this study presents an operational means of increasing PGPF T-E5 populations in BOF to promote plant growth with a concomitant reduction in production cost. In addition, a BOF compost maturity assessment using fluorescence EEM spectroscopy and FRI ensured its safe field application.  相似文献

In Arabidopsis thaliana, the expression of two genes encoding acyl‐CoA‐binding proteins (ACBPs) AtACBP1 and AtACBP4, were observed to be induced by lead [Pb(II)] in shoots and roots in qRT‐PCR analyses. Quantitative GUS (β‐glucuronidase) activity assays confirmed induction of AtACBP1pro::GUS by Pb(II). Electrophoretic mobility shift assays (EMSAs) revealed that Pas elements in the 5′‐flanking region of AtACBP1 were responsive to Pb(II) treatment. AtACBP1 and AtACBP4 were further compared in Pb(II) uptake using Brassica juncea, a potential candidate for phytoremediation given its rapid growth, large roots, high biomass and good capacity to accumulate heavy metals. Results from atomic absorption analyses on transgenic B. juncea expressing AtACBP1 or AtACBP4 indicated Pb(II) accumulation in roots. Subsequent Pb(II)‐tracing assays demonstrated Pb(II) accumulation in the cytosol of root tips and vascular tissues of transgenic B. juncea AtACBP1‐overexpressors (OXs) and AtACBP4‐OXs and transgenic Arabidopsis AtACBP1‐OXs. Transgenic Arabidopsis AtACBP1‐OXs sequestered Pb(II) in the trichomes and displayed tolerance to hydrogen peroxide (H2O2) treatment. In addition, AtACBP1 and AtACBP4 were H2O2‐induced in the roots of wild‐type Arabidopsis, while lipid hydroperoxide (LOOH) measurements of B. juncea AtACBP1‐OX and AtACBP4‐OX roots suggested that AtACBP1 and AtACBP4 can protect lipids against Pb(II)‐induced lipid peroxidation.  相似文献

Biosorption is the process of removal of any chemical molecules by the treatment of biological material. Industrialization resulted in the discharge of various toxic heavy metals into water bodies, which poses serious health hazards to humans and animals. In the present study, live Spirulina platensis was used as a biosorbent for the removal of the heavy metals chromium (Cr(VI)) and lead (Pb(II)) from the aqueous samples. S. platensis were cultured in the presence of different concentrations of heavy metals. The growth of the algal cells was found to be decreased by 59% and 36% in media containing 50 ppm Cr(VI) and Pb(II), respectively. To assess the biosorption of heavy metals, at different time intervals, the spent culture media were used to detect Cr(VI) by atomic absorption spectroscopy method and Pb(II) by 4-(2-pyridylazo)resorcinol indicator method. Results suggested that there was a significant uptake of Cr(VI) and Pb(II) from the medium by S. platensis, with corresponding decrease of metals in the medium. When metal salt solutions or industrial effluent samples were passed through the column containing immobilized live S. platensis in calcium alginate beads, the concentration of Cr(VI) was found to be reduced drastically. The present study indicates the application of S. platensis for the bioremediation of heavy metals from the samples obtained from industrial effluents.  相似文献

植物的根系分泌物是植物根系与周围环境之间的化学媒介,通过传递特定的信息,调节根际微环境,影响周围植物的生长。玉米(Zea mays L.)和荞麦(Fagopyrum esculentum Moench)是农作物间套作体系中典型的不能搭配的组合,其障碍因素尚不清楚。以玉米为受体植物,采用根悬空培养的方法,研究了荞麦、高粱(Sorghum bicolor(L.) Moench)根系分泌物对玉米根边缘细胞和根生长的影响。结果发现,玉米根边缘细胞离体培养条件下,用荞麦根系分泌物中的小分子物质处理4、8 h显著诱导边缘细胞凋亡、死亡,细胞活率分别比对照降低了71.6%和72.3%;荞麦根系分泌物中的小分子物质对玉米根产生氧化胁迫,诱导根SOD、POD和CAT活性分别比对照高22.6%、33.9%和107.2%,根中超氧阴离子(O2)和脯氨酸含量分别比对照高33.9%和49.8%;荞麦根系分泌物中小分子物质的胁迫使根细胞膜透性增大,与对照相比升高80.0%,丙二醛(MDA)含量比对照升高31.5%;荞麦根系分泌物中小分子物质诱导根内源激素(IAA)含...  相似文献

Biosorptive capacity of Pb(II), Cd(II) and Cu(II) by lyophilized cells of Pseudomonas stutzeri was investigated based on Langmuir and Freundlich isotherms. Biosorptive capacity for Pb(II), Cd(II) and Cu(II) decreased with an increase of metal concentration, reaching 142, 43.5 and 36.2 mg/g at initial concentration of 300 mg/l, respectively. Biosorption capacity for metal ions increased with increasing pH. The optimum pH for biosorption rate of Cd(II) and Cu(II) were 5.0, and 6.0 for Pb(II) biosorption. The experimental data showed a better fit with the Langmuir model over the Freundlich model for metal ions throughout the range of initial concentrations. The maximum sorptive capacity (q max) obtained from the Langmuir equation for Pb(II), Cd(II) and Cu(II) were 153.3 (r 2  = 0.998), 43.86 (r 2  = 0.995), and 33.16 (r 2  = 0.997) for metal ions, respectively. The selectivity order for metal ions towards the biomass of P. stutzeri was Pb(II) > Cd(II) > Cu(II) for a given initial metal ions concentration. The interactions between heavy metals and functional groups on the cell wall surface of bacterial biomass were confirmed by FTIR analysis. The results of this study indicate the possible removal of heavy metals from the environment by using lyophilized cells of P. stutzeri.  相似文献

Fungal inoculation and elevated CO2 may mediate plant growth and uptake of heavy metals, but little evidence from Diffusive Gradients in Thin-films (DGT) measurement has been obtained to characterize the process. Lolium mutiforum and Phytolacca americana were grown at ambient and elevated CO2 on naturally Cd and Pb contaminated soils inoculated with and without Trichoderma asperellum strain C3 or Penicillium chrysogenum strain D4, to investigate plant growth, metal uptake, and metal bioavailability responses. Fungal inoculation increased plant biomass and shoot/root Cd and Pb concentrations. Elevated CO2 significantly increased plants biomass, but decreased Cd and Pb concentrations in shoot/root to various extents, leading to a metal dilution phenomenon. Total Cd and Pb uptake by plants, and DGT-measured Cd and Pb concentrations in rhizosphere soils, were higher in all fungal inoculation and elevated CO2 treatments than control treatments, with the combined treatments having more influence than either treatment alone. Metal dilution phenomenon occurred because the increase in DGT-measured bioavailable metal pools in plant rhizosphere due to elevated CO2 was unable to match the increase in requirement for plant uptake of metals due to plant biomass increase.  相似文献

Abstract

Heavy metal bioaccumulation and translocation properties of aquatic plants are interesting because of their potential use in phytoextraction. However, there is not enough knowledge about the seasonal changes of the metal distribution properties of aquatic plants. Our study focused on seasonal variation of some heavy metals in relation to their bioaccumulation and translocation in Nuphar lutea, a floating leaved, widespread plant that is important to wildlife. In this study, N. lutea, corresponding sediment and water samples were collected at different seasons from Lake Abant (Turkey) and analysed for their heavy metal content (Pb, Cr, Cu, Mn, Ni, Zn and Cd). Accumulation and translocation of heavy metal ratios were calculated seasonally. It was found that Cr and Zn were actively transported from sediment to the root, where they accumulated especially in summer; it was also shown that Cu, Mn and Zn were not only taken up from the sediment but also from the surrounding water. The investigations suggested that translocation ratios for leaf/root of Pb, Cr, Mn and Zn reached their highest levels in spring. It was found that the bioaccumulation and translocation of heavy metals at different parts of N. lutea changes with respect to season and the type of heavy metal.  相似文献

DNA-templated silver nanoclusters (AgNC@DNA) are a novel type of nanomaterial with advantageous optical properties. Only a few atoms in size, the fluorescence of nanoclusters can be tuned using DNA overhangs. In this study, we explored the properties of AgNCs manufactured on a short single-stranded (dC)12 when adjacent G-rich sequences (dGN, with N = 3–15) were added. The ‘red’ emission of AgNC@dC12 with λMAX = 660 nm dramatically changed upon the addition of a G-rich overhang with NG = 15. The pattern of the emission–excitation matrix (EEM) suggested the emergence of two new emissive states at λMAX = 575 nm and λMAX = 710 nm. The appearance of these peaks provides an effective way to design biosensors capable of detecting specific nucleic acid sequences with low fluorescence backgrounds. We used this property to construct an NA-based switch that brings AgNC and the G overhang near one another, turning ‘ON’ the new fluorescence peaks only when a specific miRNA sequence is present. Next, we tested this detection switch on miR-371, which is overexpressed in prostate cancer. The results presented provide evidence that this novel fluorescent switch is both sensitive and specific with a limit of detection close to 22 picomoles of the target miR-371 molecule.  相似文献

Four flat-sheet membrane modules, which were operated under four different filtration modes but with the same treatment capacity, were used to treat synthetic wastewater in a submerged membrane bioreactor (MBR). Particle size distribution (PSD), gel filtration chromatography (GFC), capillary suction time (CST), and three-dimensional excitation–emission matrix (EEM) fluorescence spectroscopy were used to characterize membrane fouling properties. The high instantaneous flux induced faster fouling rate and continuous filtration mode was the most applicable filtration mode in this study. The average particle size of all foulants was smaller than that of bulk sludge; and the higher the instantaneous flux was adopted, the larger the average particle size of foulants would be. Only macromolecule substances were detected in all the foulants. The macromolecule substances in the influent were degraded by microorganism and retained by membrane, and small molecular substances could pass through membrane pores to enter the effluent. The membrane foulants had poorer dewaterability compared to the mixed liquor confirmed by CST measurement. Although there were several peaks associated with protein-like fluorophores, fulvic acid-like substances and humic acid-like organics in soluble microbial products (SMP) and extracellular polymeric substances (EPS) sample, it was found that the dominant fluorescence substances in membrane foulants were protein-like substances.  相似文献

To evaluate the role of exogenous application of a phytochelating agent glutathione in increasing resistance against different heavy metals stress, nodal explants excised from 28-day-old in vitro seedlings of Spilanthes calva L. were cultured on Murashige and Skoog’s medium supplemented with 10 μM benzyl adenine and five different concentrations (1, 5, 50, 100, or 200 mg/l) of four heavy metals: As2O3, CuSO4, ZnSO4, or Pb(NO3)2. Data were recorded for percent survival, shoot number, and shoot length after 28 d of heavy metal treatment. All four heavy metals severely inhibited growth and morphogenesis. Pb proved most inhibitory whereas Zn was least effective. Pb was further selected to study the reversal effect of glutathione on morphogenesis. The addition of different concentrations (1, 5, 10, or 25 mg/l) of glutathione to media containing the Pb resulted in a significant improvement in almost all growth parameters. Inclusion of glutathione at 10 mg/l was optimum for maximum reversal of the negative effects of heavy metals on morphogenesis.  相似文献

【目的】探讨耐重金属细菌对铅(lead, Pb)和镉(cadmium, Cd)胁迫下菊苣(Cichorium intybus L.)幼苗Pb、Cd抗性和黄酮生物合成的调控作用。【方法】在不同浓度Pb和Cd [(200+20) mg/kg、(400+40) mg/kg、(800+80) mg/kg]处理下,接种菌株JB19并测定菊苣幼苗生长指标、Pb和Cd含量、抗氧化酶活性、总黄酮含量和黄酮生物合成相关基因表达量。【结果】菌株JB19可显著提高不同浓度Pb和Cd处理下菊苣的生物量和叶绿素含量;减少氧化损伤,地上部和根部Pb、Cd含量均降低,其中在(Pb200+Cd20) mg/kg处理下H2O2和丙二醛(malondialdehyde, MDA)含量分别比对照降低了25.7%和26.1%,地上部Pb、Cd含量降低的幅度最大,分别降低了53.2%和54.1%;加强了菊苣幼苗的抗氧化酶防御系统,黄酮生物合成相关基因均显著上调,其中在(Pb400+Cd40) mg/kg处理下黄酮类化合物的含量增加了105.2%,查尔酮异构酶基因上调最显著,达458....  相似文献

Heavy metal tolerance of two marine strains of Yarrowia lipolytica was tested on solid yeast extract peptone dextrose agar plates. Based on minimum inhibitory concentration esteems, it is inferred that the two strains of Y. lipolytica were tolerant to heavy metals such as Pb(II), Cr(III), Zn(II), Cu(II), As(V), and Ni(II) ions. The impact of various heavy metal concentrations on the growth kinetics of Y. lipolytica was likewise assessed. With increased heavy metal concentration, the specific growth rate was reduced with delayed doubling time. Furthermore, biofilm development of both yeasts on the glass surfaces and in microtitre plates was assessed in presence of different heavy metals. In microtitre plates, a short lag phase of biofilm formation was noticed without the addition of heavy metals in yeast nitrogen base liquid media. A lag phase was extended over increasing metal concentrations of media. Heavy metals like Cr(VI), Cd(II), and As(V) are contrastingly influenced on biofilms’ formation of microtitre plates. Other heavy metals did not much influence on biofilms development. Thus, biofilm formation is a strategy of Y. lipolytica under stress of heavy metals has significance in bioremediation process for recovery of heavy metals from contaminated environment.

Abstract

The purpose of this study was to investigate the distribution of total and bioavailable metals (Cd, Cu, Cr, Fe, Mn, Pb and Zn) in sediments of the Abelardo L. Rodríguez (ALR) dam located in the eastern part of the city of Hermosillo, Sonora, Mexico. Seventy two sediment samples were collected in 2009 during four sampling campaigns in February (spring), May (summer), September (end of summer) and December (winter) in five different areas within the dam surface (Zones I, II, III, IV and V), including the Gate Station. Determination of heavy metals was carried out by flame atomic absorption spectrophotometry (FAAS). The results indicate high levels of total heavy metals in the following order: Fe>Mn>Zn>Pb>Cu>Cr>Cd. This is indicative of the impact of human activities located in areas surrounding the reservoir, specifically urban and industrial. The distribution and state of accumulation of trace metals in the sediment is largely dominated by the residual and Fe/Mn oxides geochemical phases. Fraction I (exchangeable) also presented high concentrations of metals (Cu, Fe, Mn and Zn). From enrichment factor analysis, the study area is prevalently enriched in Cd, Cu and Pb in Zones I, II, III, IV and Gate Station. This indicates that the sediments are impacted by anthropogenic activities such as downloads, domestic and industrial wastewater. Geoaccumulation index (Igeo) indicates that Zones I, II, III, IV and V (including Gate Station) do not show contamination by Cr, Fe, Mn and Zn. However, there is a moderate to heavy contamination by Cd, Cu and Pb (Igeo: 2–4) in all areas of study. The comparison between the results obtained with the sediment quality criteria (LEL and SEL), indicate that Zones I, II, III, IV, V and Gate Station, are contaminated with Cd, Cu and Pb, and severely contaminated by Fe.

The elevated levels of heavy metals detected in the sediment of the ALR Dam require special attention, since in the exchangeable fraction, the metals are specifically adsorbed on the sediment and can be released when the ionic composition of water changes. However, additional studies are required in this reservoir on the chemistry and toxicology of metals for a full assessment of potential risks posed to biota and man.  相似文献

This study was conducted to assess the pollutant uptake capability of water lettuce (Pistia stratiotes L.) in terms of bioaccumulation, enrichment, and translocation of heavy metals grown in sugar mill effluent. Results showed that the maximum fresh weight (328.48 ± 2.04 gm kg?1), total chlorophyll content (2.13 ± 2.03 mg g?1 fwt), and relative growth rate, RGR (11.89 gg?1 d?1) of P. stratiotes were observed at 75% concentration of the sugar mill effluent after 60 days of phytoremediation experiment. The bioaccumulation factor (BF) of different heavy metals was greater than 1 with 50% and 75% concentrations of sugar mill effluent and this indicated that P. stratiotes was hyperaccumulator or phytoremediator of these metals. The enrichment factor (EF < 2 for Cu, Fe, Cr, Pb, Zn, and Mn) and (EF > 2 for Cd) indicated that P. stratiotes mineral enrichment deficient and it moderately enriched the different heavy metals. Moreover, translocation factor (TF) was less than 1 which indicated the low mobility of metals in different parts (root and leaves) of P. stratiotes after phytoremediation. Therefore, P. stratiotes can be used for phytotreatment of sugar mill effluent up to 50% to 75% concentrations and considered as hyperaccumulator aquatic plant for different heavy metals and other pollutants from the contaminated effluents.  相似文献

The concentration and bioavailability of Ni, Cu, Cd, Zn, and Pb in the sediments and leaves of grey mangrove, Avicennia marina, were studied throughout Sirik Azini creek (Iran) with a view to determine heavy metals bioavailability, and two methods were used. Results show that Zn and Ni had the highest concentrations in the sediments, while Cd and Cu were found to have the lowest concentrations in the sediments. Compared to the mean concentrations of heavy metals in sedimentary rock (shales), Zn and Cu showed lower concentration, possibly indicating that the origin of these heavy metals is natural. A geo-accumulation index (I geo) was used to determine the degree of contamination in the sediments. I geo values for Zn, Cu, Pb, and Ni showed that there is no pollution from these metals in the study area. As heavy metal concentrations in leaves were higher than the bioavailable fraction of metals in sediments, it follows that bioconcentration factors (leaf/bioavailable sediment) for some metals were higher than 1.  相似文献
