The duration of diapause in the stem borers Busseola fusca (Fuller) and Chilo partellus (Swinhoe) was studied in South Africa by collecting diapausing larvae from the field throughout winter (April–August). B. fusca larvae emerged as moth around the middle of October regardless of the date of collection and the length of time they were kept in the laboratory under constant 21 °C. C. partellus larvae collected in April–June emerged in November, those collected in July emerged in October, and those collected in August emerged in September. Regardless of the collection date C. partellus started to emerge from diapause earlier and moth emergence lasted up to twice as long as in B. fusca. Under laboratory conditions at 60% RH both borer larvae lost about 50% of their body mass during diapause. When provided with water B. fusca larvae lost about 30% of their body mass and adults emerged 20 days earlier than when kept dry. C. partellus, on the other hand, lost only 13% of their body weight and emerged 34 days earlier. The differences between the two species are discussed in light of different types of diapause; i.e., obligatory diapause in B. fusca and facultative diapause in C. partellus.  相似文献

A biogeochemical study of a polluted wetland site in Kitwe, Zambia shows high concentration of trace metals (e.g., > 25 and ≈ 2 fold higher than the Eco-toxic threshold values of copper and cobalt, respectively) with many sequestered with the sediment organic phase. Depth profiles in surface sediments suggest trace metal cycling between porewater and solid phases, including that of cobalt. This study documents a bacterium displaying resistance to, and accumulation of cobalt, and that cobalt has a positive effect on growth. The isolate was enriched from the microbial community and identified using 16S rRNA gene sequence analysis as a strain of Comamonas testosteroni (designated C. testosteroni TDKW). Improved growth of C. testosteroni TDKW was seen with the addition of up to 200 μM cobalt (optimal growth ca. 100 μM), while concentrations above 4 mM completely inhibited growth. C. testosteroni TDKW also exhibited resistance to high concentrations of iron and manganese, but showed limited resistance to copper or nickel. Further analysis revealed cellular cobalt accumulation and the presence of heavy-metal resistance genes, tentatively suggesting that this organism could contribute to in situ biological cycling of cobalt in mineral contaminated aquatic systems.  相似文献

Cadmium (Cd) is believed to be one of the most abundant and ubiquitously distributed toxins in the aquatic system. This metal is released to the aquatic environment from both anthropogenic sources, such as industrial, agricultural and urban effluents as well as natural sources, such as rocks and soils. Otherwise, the temperature increase of water bodies, which has been observed due to global climatic changes, has been shown to increase Cd toxicity for several aquatic animal species including fish. In the present study, Nile tilapia, Oreochromis niloticus (L.), (26.0±0.38 g) were reared at 20, 24, 28, or 32 °C and exposed to 0.0 or 0.5 mg Cd/L for 8 weeks to investigate effects of water temperature, Cd toxicity and their interaction on fish performance as well as metallothionein (MT) and Cd distribution in different fish organs. It was found that fish reared in Cd-free group at 28 °C showed the optimum growth and feed intake, while Cd-exposed fish showed low growth and feed intake irrespective to water temperature. A synergetic relationship between water temperature and Cd toxicity was observed where Cd toxicity increased as water temperature increased and the worse growth was obtained in Cd-exposed fish reared at 32 °C. Additionally, the highest Cd residues in different fish organs were detected in Cd-exposed fish reared at 32 °C. Similarly, MT concentrations in different fish organs increased as water temperature increased especially in Cd-exposed fish groups. A high positive correlation between MT and Cd concentrations in fish organs was detected. The distribution of MT and Cd levels was in the order of liver>kidney>gills>muscles. The present study revealed that the optimum water temperature suitable for Nile tilapia growth is 28 °C. Additionally, Cd exposure had a deteriorate effect on the growth and health of Nile tilapia. This hazardous effect increased as water temperature increased. Further, liver and kidney were the prime sites of Cd accumulation, while Cd load in the muscles was the lowest as compared to the other investigated organs.  相似文献

Concentration of heavy metals (lead (Pb), cadmium (Cd), chromium (Cr), and copper (Cu)) was determined in the liver, gills, kidneys, and muscles of eight edible fish species (Channa punctata, Cirrhinus reba, Labeo rohita, Heteropneustes fossilis, Mystus cavasius, Oreochromis niloticus, Puntius sophore, and Wallago attu) from upstream and downstream zones of the Nullah Aik and Palkhu tributaries of the River Chenab located in the Sialkot district known for its tanning industries worldwide. The pattern of metal accumulation in studied organs was in the order: Cr > Pb > Cu > Cd. Liver showed greater metal accumulation, followed by gills, kidneys, and muscles. Accumulation of Pb and Cr was significantly different in organs between upstream and downstream zones. Accumulation was greater in fish species dwelling downstream, indicating impairment of ambient stream water due to untreated discharge of industrial and municipal effluents into studied streams. Highest concentration of Pb and Cr and lowest of Cd was detected in H. fossilis whereas Cu showed higher concentration and Cr lowest concentration in P. sophore. In contrast, lower concentration of Pb and Cu was recorded in M. cavasius, O. niloticus, and W. attu. Mean concentrations of Cd, Cr, and Cu were higher in pre-monsoon compared to post-monsoon season. Measured concentrations of Pb, Cd, and Cr in muscles of species such as C. punctata, W. attu, L. rohita, P. sophore, and O. niloticus were above permissible limits of heavy metals for human consumption, indicating potential health risks. Therefore, these fish species from studied locations should be avoided for human diet.  相似文献

Abstract

The distribution and relative importance of lepidopteran stem borers attacking maize and sorghum were investigated in farmers’ fields during the main cropping seasons of 2015 and 2016 across three main agro-climatic zones (ACZs) of eastern Ethiopia. Three stem borer species, namely Busseola fusca Fuller, Chilo partellus Swinhoe and Sesamia calamistis Hampson were found attacking these crops with different levels of distribution and abundance among ACZs. Busseola fusca was the only borer species in the highlands and dominant (67%) in the midhighland zones, while C. partellus was predominant (75%) in the lowlands. The abundance of S. calamistis was low across its distribution range. Across both years, mean plant infestation by B. fusca ranged from 20.38 in lowlands to 42.97% in highlands. Chilo partellus resulted in a mean infestation of 27.38% in the midaltitude to 68.24% in lowlands. Mean density of larvae per plant was higher for C. partellus (2.85), followed by B. fusca (1.41) and S. calamistis (0.46). Moreover, within each ACZs, variation in plant damage variables was observed between the study periods. These spatio-temporal differences in community structures of stem borers are discussed particularly in relation to agro-climatic requirements.  相似文献

The effect of exposure duration to pre-storage (representing fall conditions) and storage (representing winter conditions) temperatures on the emergence rate in sexual and asexual Trichogramma brassicae Bezdenko (Hymenoptera: Trichogrammatidae) was studied under controlled laboratory conditions. Asexual T. brassicae was infected by Wolbachia, an endosymbiont bacterium. We considered 0, 30, 40, 50 and 60 days as exposure duration at 10°C and 0, 30, 60, 90 followed by 180 days as exposure duration at 4°C for both T. brassicae strains. Emergence rate was significantly affected by the presence of Wolbachia, the duration of exposure to 10°C, the duration of storage at 4°C and their interactions, but not by the interaction between strain × storage duration at 4°C. Emergence rate of adults was positively correlated with exposure duration at 10°C, but it gradually decreased as the cold storage duration (4°C) increased in both strains. In conclusion, we are able to store both asexual and sexual strains for longer periods of time at 4°C, but the emergence rate of sexual T. brassicae was greater than asexual in all the treatments. The presence of Wolbachia could have a negative effect on fitness of T. brassicae and reduces its emergence rate after long-term storage.  相似文献

DNA hybridization techniques showed Chlorella fusca var. vacuolata and C. kessleri to be homogeneous species with DNA homologies of 90–100% C. fusca var. fusca and var. rubescens, however, have only about 15% DNA homology with C. fusca var. vacuolata and should no longer be regarded as varieties. A good correlation was found so far between biochemical and physiological characters used in the taxonomy of Chlorella and DNA relatedness. Mutant strains of Chlorella were tested for DNA homologies to prove the reliability of the taxonomical interpretation.  相似文献

ABSTRACT

The combined effects of pH and salinity were investigated on the bioaccumulation of inorganic mercury in the Turkish crayfish Astacus leptodactylus, at the whole organism and organ levels. Effects on the ionic balance were also analysed during the acclimatization phase and after 3 and 15 days of exposure, via the direct route. The experimental protocol, based on a complete factorial design, took into account nine experimental conditions, resulting from the combination of three levels for pH (6, 7.5 and 9) and for salinity (1, 10 and 100 mM Cl-). The results showed an important Hg accumulation in the organs or tissues in direct contact with the surrounding medium, e.g. gills and carapace. The effects of the nine pH and salinity conditions were complex and variable from one organ to another. Correlations were studied between chemical species concentrations in the water (HgCl2, HgOHCl, HgCl3-,HgCl42, Hg(OH)2) and amounts of metal accumulated in the different organs. A positive correlation was found between the neutral species HgCl2 and metal accumulated in the gills and in the carapace. Perturbations of the haemolymph ionic concentrations were no significant, except for Na+ after 3 days of exposure.  相似文献

Toxicokinetics of cobalt was studied on rats by the method of radioactive isotopes using single or repeated administrations. Co was used in the form of sulphate in doses of 0.1 and 0.2 mg/kg administered by 4 different routes (intragastric, intraperitoneal, intratracheal, subcutaneous). Good absorption was established in the parenteral routes of administration, the regularities of absorption, retention in the internal organs and elimination of cobalt were determined and mathematically expressed. Clear dependence was established between the doses administered and the content in biomedia. The liver and the kidneys were found to be the organs of highest accumulation of cobalt. Elimination half-lives and the time of establishment of a constant level during long-term exposure were determined. It is proposed to carry out diagnostic examination in the course of a few hours after the work shift. Urine and blood are the test objects in which direct correlation has been established. Delayed elimination of cobalt from the organism under protracted exposure may be the essential cause of increased hazard for man, which conditions the necessity of determining maximum acceptable concentration for this substance.  相似文献

Abstract

This study monitors and assesses the effect of battery-manufacturing effluent containing metals Pb, Zn and Cd on endogenous antioxidants. Malonaldehyde (MDA), reduced glutathione sulfyhydryl (GSH) and catalase (CAT) which are known biomarkers of effluent were exposed to 0%, 25%, 50%, 75% and 90% amendments for 74h on the gills, liver and kidneys of C. punctata. There was more metal Zn accumulation in the gills and GSH contents increased significantly in the gills (P<0.01), liver accumulation of Pb was found to be more (P<0.05), whereas lowest accumulation of Pb was found in kidneys and the highest accumulation of Cd (P<0.05). Over all amendments of the effluents, MDA contents were increased in the gills, liver and kidneys (P<0.01). GSH levels were decreased among the liver and kidneys compared to the gills (P<0.01) at 90% amendment. Effluent exposure caused a significant decrease in the activities of CAT in the gills, liver and kidneys (P<0.01, 0.05 and 0.05) of fish. Increased MDA activity was indicative of the formation of free radicals in the fish with exposure to amendments of battery manufacturing effluent, while increased levels of GSH pointed to the occurrence of a scavenging mechanism of free radicals.  相似文献

Lepidopteran stemborers are the most destructive insect pests of cereal crops in sub‐Saharan Africa. In nature, these insects are often exposed to multiple environmental stressors, resulting in potent impact on their thermal tolerance. Such environmental stressors may influence their activity, survival, abundance and biogeography. In the present study, we investigate the effects of acclimation to temperature, starvation and desiccation on thermal tolerance, measured as critical thermal limits [critical thermal minima (CTmin) and maxima (CTmax)] on laboratory‐reared economic pest species Chilo partellus Swinhoe (Lepidoptera: Crambidae), Busseola fusca (Fuller) and Sesamia calamistis Hampson (Lepidoptera: Noctuidae) using established protocols. Low temperature acclimation results in improved CTmin for B. fusca and C. partellus, whereas high temperature acclimation enhances the same trait for B. fusca and S. calamistis. Similarly, high temperature and starvation pretreatment improve CTmax for C. partellus relative to S. calamistis and B. fusca. In addition, starvation and desiccation pretreatments improve CTmin for all stemborer species. Furthermore, rapid cold‐hardening (RCH) enhancs CTmin for B. fusca and C. partellus, whereas rapid heat‐hardening (RHH) improves the same trait for C. partellus. However, RCH and RHH impair CTmax for all stemborer species. These findings show differential thermal tolerances after exposure to heterogeneous environmental stress habitats. Chilo partellus, of exotic origin, shows a higher magnitude of basal thermal tolerance plasticity relative to the indigenous African species S. calamistis and B. fusca. This indicates that C. partellus may have a fitness and survival advantage under climate‐induced heterogeneous environments, and also have a greater chance for geographical range expansion and invasion success compared with the indigenous B. fusca and S. calamistis.  相似文献

The Nile catfish, Clarias lazera was found to concentrate radioactive cesium-134 and cobalt-6o from the aquatic environment. For cesium-134 the rate of uptake increased by increase of exposure time, while for cobalt-6o maximum uptake occurred after one day of exposure. The corresponding concentration factors at maximum uptake levels were 0.37 and 0.36 for cesium and cobalt respectively.The internal distribution of these radionuclides in the different tissues and organs of the fish due to uptake from the aquatic environment followed the decreasing order:For 134Cs: muscle, bone, gills, stomach, kidneys, intestine and liver.For 60Co: bone, muscle, gills, intestine, kidneys, stomach and liver.The internal distribution due to ingestion of these radionuclides followed nearly the same order as was found in case of uptake from the aquatic environment.  相似文献

Abstract

In a previous work temporal succession of two sheathed, filamentous methanotrophic γ-Proteobacteria, Crenothrix polyspora and Clonothrix fusca, was observed by analyzing the microbial community of an artesian well. Although a correlation between physical-chemical environmental parameters and growth of these microorganisms was suggested, the link with the microbial physiology remained elusive. In this study we demonstrate that phosphatidylethanolamine and cardiolipin, generally abundant in most γ-Proteobacteria, could not be detected in C. fusca membranes by thin-layer chromatography. The absence of cardiolipin was supported by results of enzymatic assay of cardiolipin synthase activity and by Southern blot analysis that failed to detect cls homologous sequences in the genomic DNA of this microorganism. The absence of phosphatidylethanolamine and cardiolipin might account for requirement of millimolar concentrations of magnesium and calcium for growth, and for massive over-growth of this microorganism during the winter season when the water levels of magnesium and calcium increased in the artesian well.  相似文献

In this study, concentration of mercury was determined in the trophic levels of benthic, benthopelagic, pelagic fish species, and river birds from Arvand River, located in the Khuzestan province in the lowlands of southwestern Iran at the head of the Persian Gulf. The order of mercury concentrations in tissues of the fish species was as follows: liver>gill>muscle and in tissues of the kingfisher species was as follows: feather>liver>kidney>muscle. Therefore, liver in fish and feather in kingfisher exhibited higher mercury concentration than the other tissues. There was a positive correlation between mercury concentrations in fish and kingfisher species with size of its food items. We expected to see higher mercury levels in tissues of female species because they are larger and can eat larger food items. The results of this study show that the highest mean mercury level were found in the kingfisher (Anas crecca), followed by benthic (Epinephelus diacanthus), benthopelagic (Chanos chanos), and pelagic fish (Strongylura strongylura). Mean value of mercury in fish species, S. strongylura were (0.61 μg g?1 dry weight), C. chanos (0.45 μg g?1 dry weight), E. diacanthus (0.87 μg g?1 dry weight), and in kingfisher species A. crecca was (2.64 μg g?1 dry weight). Significant correlation between mercury concentration in fish and kingfisher may be related to high variability of mercury in the fish.  相似文献

Copper (Cu) is one of the most commonly reported metal pollutants in African water bodies, but there are few studies on African freshwater fish species of copper accumulation and copper toxicity. Adult O. mossambicus were exposed to 0mg l?1 (control) and 0.75mg l?1 Cu for 96h and 0 (control), 0.11, 0.29 and 0.47mg l?1 copper for 64 days. Samples of liver and gills were collected after 96h, and after 1, 32 and 64 days, respectively. There were significant differences in the mean Cu accumulation values in the liver and gills between the control and the Cu-exposed fish after the 96-h exposure. In fish exposed to 0.11 and 0.29mg l?1 Cu for 64 days there was an increase in copper level in the tissues. In fish exposed to 0.47mg l?1 Cu the concentration in the gill and liver tissue did not increase between Day 1 and Day 32. At this time, Cu accumulation in the liver was higher than for fish exposed to 0.11 and 0.29mg l?1 Cu for 64 days. Exposure to approximately 0.47mg l?1 Cu for more than 32 days induced mortality.  相似文献

The effects of 4 or 8 drought cycles on four grass species,Cenchrus pennisetiformis, Leptochloa fusca, Panicum turgidum, andPennisetum divisum were assessed in a pot experiment. There were significant differences between the species in biomass production under water stress.C. pennisetiformis andP. turgidum produced significantly greater fresh and dry matter thanP. divisum and especially thanL. fusca. L. fusca had the lowest andP. divisum highest osmotic potentials compared with the other species after the completion of 4 or 8 drought cycles. Osmotic adjustment (difference between osmotic potential of droughted/rehydrated plants and control plants) was highest inL. fusca. The stomatal conductance was significantly decreased with increased drought stress inC. pennisetiformis. The elasticity ofC. pennisetiformis, P. turgidum andP. divisum increased with increase in number of drought cycles, whereas that ofL. fusca remained unchanged.L. fusca andP. turgidum had the lowest leaf hydration of all species after 8 drought cycles. The chlorophyllsa andb in all species remained unaffected by drought treatments. The proline content ofC. pennisetiformis andL. fusca increased significantly with increased drought stress, whereas that ofP. turgidum remained unaffected after 4 or 8 drought cycles.L. fusca synthesized great amount of leaf soluble proteins during 8 drought cycles, whereasP. divisum had low protein content after 4 drought cycles. The protein contents ofC. pennisetiformis andP. turgidum remained unaffected after 8 drought cycles. The leaf epicuticular wax ofL. fusca increased consistently with increased drought stress, but leaf wax ofP. divisum increased only at the highest drought stress and that ofC. pennisetiformis andP. turgidum increased after 4 drought cycles. On the basis of these results it was established thatC. pennisetiformis andP. turgidum were the most tolerant,P. divisum intermediate, andL. fusca the most sensitive to drought stress. The osmotic adjustment did not positively correlate with the degree of drought resistance.  相似文献

Abstract

The order Actinomycetales includes a number of genera that contain species that actively degrade cellulose and these include both mesophilic and facultative thermophilic species. Cellulases produced by strains from two of the genera containing thermophilic organisms have been studied extensively: Microbispora bispora and Thermomonospora fusca. Fractionation of M. bispora cellulases has identified six different enzymes, all of which were purified to near homogeneity and partially characterized. Two of these enzymes appear to be exocellulases and gave synergism with each other and with the endocellulases. The structural genes of five M. bispora cellulases have been cloned and one was sequenced. Fractionation of T. fusca cellulases has identified five different enzymes, all of which were purified to near homogeneity and partially characterized. One of the T. fusca enzymes gives synergism in the hydrolysis of crystalline cellulose with several T. fusca endocellulases and with Trichoderma reesei CBHI but not with T. reesei CBHII. Each T. fusca cellulase contains distinct catalytic and cellulose binding domains. The structural genes of four of the T. fusca endoglucanases have been cloned and sequenced, while three cellulase genes have been cloned from “T. curvata”. The T. fusca cellulase genes are expressed at a low level in Escherichia coli, but at a high level in Streptomyces lividans. Sequence comparisons have shown that there are no significant amino acid homologies between any of the catalytic domains of the four T. fusca cellulases, but each of them shows extensive homology to several other cellulases and fits in one of the five existing cellulase gene families. There have been extensive studies of the regulation of the synthesis of these cellulases and a number of regulatory mutants have been isolated. This work has shown that the different T. fusca cellulases are coordinately regulated over a 100-fold range by two independent controls; induction by cellobiose and repression by any good carbon source.  相似文献

Cotesia sesamiae (Cameron) (Hymenoptera: Braconidae) is an indigenous larval endoparasitoid of Busseola fusca (Fuller) (Lepidoptera: Noctuidae) in sub-Saharan Africa. In Kenya, reports suggest that C. sesamiae occurs as two biotypes. Biotype avirulent to B. fusca gets encapsulated by haemocytes in this host and is unable to complete development. Biotype virulent to B. fusca is able to overcome immune defences. Factors present in the calyx fluid such as the PolyDNAviruses (PDV), venom and calyx fluid proteins have been implicated in the variation of C. sesamiae virulence against B. fusca. In the present study, calyx fluid proteins of the two C. sesamiae biotypes were compared using 2-D gel electrophoresis. More protein spots were observed in the virulent parasitoid calyx fluid, but some proteins were specifically observed in the avirulent parasitoid calyx fluid while others were observed in both. To study changes in proteins due to parasitism of B. fusca larvae by the two strains, SDS-PAGE gel were performed on fat body tissues and the haemolymph at three time points. Differences between the two strains were observed in both the fat body and haemolymph tissues. Parasitism-specific protein bands were detectable in fat body tissues of B. fusca larvae parasitized by the two C. sesamiae strains. These proteins were absent in unparasitized larvae. Implications for using C. sesamiae as a biocontrol agent of B. fusca in Africa are discussed.  相似文献

The number and distribution of chemosensilla located on different organs of Busseola fusca (Fuller) (Lepidoptera: Noctuidae) males and females are described based on observations using scanning electron microscopy, selective staining with silver nitrate, and gustatory electrophysiological recording. The antennae and the fifth tarsomere of the prothoracic legs of both sexes bear chemosensilla: uniporous chaetica and multiporous trichoidea sensilla. However, there is a sexual dimorphism in the number and size of sensilla on these organs. The distal part of the ovipositor has uniporous gustatory chemosensilla of the chaetica type. The involvement of these sensilla in oviposition site selection by B. fusca is discussed.  相似文献
