It has been shown that intraperitoneal injections of L-DOPA cause an increase in the matrix activity of chromatin and stimulate the incorporation of [3H]uridine into the nuclear fraction of rat brain cells by 35%. In vitro studies have shown that preincubation of brain chromatin with L-DOPA diminishes the inhibiting effect of actinomycin D on RNA synthesis. It has been found that the rate of RNA synthesis in vitro depends on concentrations of catecholamines (L-DOPA, dopamine, norepinephrine) and serotonin.  相似文献

After administration of 14C-thymidine in vivo, the incorporation of the isotope in isolated nucleoli and nuclei of adult rat liver has been demonstrated by track-autoradiography. The advantages of the method used are discussed. The results confirm unambiguously our previous biochemical findings suggesting a synthesis of nucleolar DNA independent of that of the chromosomal extranucleolar DNA in adult rat liver.  相似文献

It has been shown that the induction of D-RNA synthesis in rat liver nuclei by electrostimulation of hypothalamus is accompanied by a decrease in chromatin protein synthesis and an increase in phosphorylation and acetylation of chromatin proteins. The decrease of the histone synthesis is mainly due to the decrease of [14C]lysine and [14C]alanine incorporation into histones H1 and H4. The relationship between H1, H2b-H3, H2a and H4 histone fractions remains unchanged. Electrostimulation of hypothalamus increases acetylation of H2a and H4 histone fractions and phosphorylation of all histones with the exception of histone H1.  相似文献

RNA synthetic activity of human 2-16-cell embryos developing in vitro was studied by [3H]uridine light-microscope autoradiography. Parallelly cut thin sections were examined in the electron microscope. The first extranucleolar RNA synthesis was detected in 4-cell embryos, but nucleoli were never labelled until the 3rd cleavage (6-8-cell embryos). In 6-cell embryos the nucleolar labelling was mostly confined to a narrow peripheral zone. In later cleavage stages most of the blastomeres showed intensive labelling of nucleoli and extranucleolar chromatin. However, rather low levels of extranucleolar RNA synthesis and the absence of nucleolar activity were often seen even in blastomeres of fully compacted morulae. The activation of nucleolar RNA synthesis entailed a noticeable increase in the number of ribosomes (estimated by electron microscope morphometry) that followed a marked drop during the period between the 2-cell and 8-cell stages. The results indicate that the concentration of ribosomes in the preovulatory oocyte is a major factor of its developmental potential.  相似文献

gamma-Irradiation of DNA results in a several-fold decrease of its primer activity measured as one substrate synthesis catalyzed by DNA polymerase beta. However, the combined treatment of injured DNA with 3'----5' exonuclease and endonuclease I from rat liver chromatin almost normalizes primer activity of DNA. Therefore the above-mentioned nucleases are capable of excising the gamma-injured nucleotides from 3'-OH ends of DNA.  相似文献

A nuclear nonhistone protein which decreases in chromatin during growth (Yeoman, L. C., et al. (1975) Cancer Res. 35, 1249) has been isolated in high purity from the chromatin of normal rat liver nuclei by gel electrophoresis and column chromatography. This protein, designated BA (Yeoman, L.C., et al. (1973) Biochem Biophys. Res. Commun. 53, 1067), has a molecular weight of 31 000, an acidic to basic amino acid composition ratio of 0.9, and contains one tryptophan residue per molecule. Hydrazinolysis indicated protein BA has a lysine carboxyl terminus; however, the amino terminal is blocked as no reaction occurred with dansyl chloride. Maps of tryptic peptides of protein BA contained 46 spots. Protein BA binding to various DNAs was examined by the nitrocellulose filter assay. Binding was slightly enhanced by 2mM Mn2+ion; Mg2+, however, decreased binding. Binding was optimal at neutral pH and an ionic strength of 0.2 M [NaCl]. Equilibrium competition binding studies indicated a binding preference of protein BA for dA-dT rich DNA.  相似文献

It was demonstrated that neutral Mn-dependent DNAase from rat liver chromatin stimulates the incorporation of labeled precursors of DNA into high molecular weight fractions of isolated nuclear DNA. The effects of DNA-polymerase inhibitors and the properties of DNA synthesis products suggest that neutral Mn-dependent DNAase can induce replicative synthesis of DNA in the nuclei of normal and regenerating rat liver.  相似文献

Degenerative and regenerative changes induced in rat liver by single exposure to diethylnitrosamine (DEN) were examined by morphological and biochemical approaches. Apoptotic changes were observed in livers of rats exposed to a 'subnecrogenic' dose of DEN (10 mg/kg) as well as in liver parenchyma of those receiving a necrogenic dose (100 mg/kg). Zonal centrilobular necrosis was observed exclusively in the latter group. Regenerative changes, i.e., increases in DNA synthesis, labeling index and mitotic activity, occurred only in animals exposed to the higher dose. The mitogenic effect obtained in these conditions was about half that induced by two-thirds hepatectomy and the maximum response occurred about 24 h later than in partially hepatectomized rats.  相似文献

Using the osmium-ammine DNA tracer (Cogliati, R & Gautier, A, Compt rend acad sci ser D 276 (1973) 3041 [1]), we were able to study the structure of intranucleolar chromatin in thin sections of rat hepatocytes fixed in situ after partial hepatectomy and cortisol treatment. Three levels of chromatin organization were observed: (1) highly compact clumps; (2) fibres with a thickness varying from 11 to 25 nm; (3) loose agglomerates of extended DNA filaments with a thickness of 2–3 nm. Both the clumps and the fibres of chromatin showed nucleosomal configuration which was absent in the loose agglomerates. These latter structures appeared to be peculiar to nucleolar chromatin.  相似文献

The status of DNA methylation, as measured by m5C2 content of nuclear DNA, was examined during corticosteroid mediated TAT induction in rat liver and in dividing and nondividing HTC cells. The m5C content, determined by HPLC, was not significantly altered in HTC cells during TAT induction whether the cells were in the logarithmic or stationary growth phase. In the liver of adrenalectomized rats where the range of corticosteroid effects is greater than in HTC cells, a small but significant decline in genomic levels of m5C was detected between 1 to 8 hr post-induction. The alterations in DNA methylation did not fluctuate during induction by more than 8% in the liver or 7.5% in HTC cells. These results demonstrate that no gross change or elevation in m5C content is detected in two, different, hormonally responsive hepatocellular systems during gene activation.  相似文献

Summary Inhibition of RNA synthesis in Hela cells, evoked by high doses of thymidine, was followed. The author used labelling with 3H-uridine and quantitative autoradiography in two groups of experiments: first, of 4 mM thymidine was being followed for the period of 1, 6, 12 and 24 hours; second, the effect of 0.33, 1, 3, 9 and 27 mM of thymidine for the period of 21 hours. A significant decrease of both the nucleolar and extranucleolar chromatin RNA synthesis was detected as a due to the exposure to 4 mM thymidine for the period of 1–6 hours or to the exposure to 1 mM thymidine for the period of 21 hours. A further deepening of the inhibition of the extranucleolar RNA synthesis appears by protracting the period of action or by increasing the concentration of thymidine in the medium.The author is of the opinion that the inhibition of the nucleolar RNA synthesis is very likely a result of the blockade of the DNA reduplication and of the cell synchronization. The inhibition of the extranucleolar RNA synthesis is on the other hand caused directly, without the intermediate of DNA.  相似文献
