A new technique for measuring the size distribution of anaerobic granular sludge, which involves the use of digital image analysis, is presented. Sludge samples are embedded in gelatin, spread over glass dishes, which are then placed over a flat-bed scanner where an image is captured. The images are processed with an image analysis software. Environmental deterioration together with the need for water reuse and the increasingly restrictive legislation of water quality standards have led to a demand for compact, efficient and less energy consuming technologies for wastewater treatment. Aerobic granular sludge and membrane bioreactors (MBRs) are two technologies with several advantages, such as small footprint, high-microbial density and activity, ability to operate at high organic- and nitrogen-loading rates, and tolerance to toxicity. However, they also have some disadvantages. The aerobic granular sludge process generally requires post-treatment in order to fulfill effluent standards and MBRs suffer from fouling of the membranes. Integrating the two technologies could be a way of combining the advantages and addressing the main problems associated with both processes. The use of membranes to separate the aerobic granules from the treated water would ensure high-quality effluents suitable for reuse. Moreover, the use of granular sludge in MBRs has been shown to reduce fouling. Several recent studies have shown that the aerobic granular membrane bioreactor (AGMBR) is a promising hybrid process with many attractive features. However, major challenges that have to be addressed include how to achieve granulation and maintain granular stability during continuous operation of reactors. This paper aims to review the current state of research on AGMBR technology while drawing attention to relevant findings and highlight current limitations.  相似文献

The present study investigates the formation of aerobic granular sludge in sequencing batch reactor (SBR) fed with palm oil mill effluent (POME). Stable granules were observed in the reactor with diameters between 2.0 and 4.0 mm at a chemical oxygen demand (COD) loading rate of 2.5 kg COD m−3 d−1. The biomass concentration was 7600 mg L−1 while the sludge volume index (SVI) was 31.3 mL g SS−1 indicating good biomass accumulation in the reactor and good settling properties of granular sludge, respectively. COD and ammonia removals were achieved at a maximum of 91.1% and 97.6%, respectively while color removal averaged at only 38%. This study provides insights on the development and the capabilities of aerobic granular sludge in POME treatment.  相似文献

A model has been developed and experimentally checked for the physical behavior of sludge in the blanket in upflow reactors. The model is based on the mass balance for the sludge in the blanket, and can be used to predict the distribution of sludge in an upflow reactor in relation with the gas production, sludge settling characteristics, and the linear fluid velocity in the reactor. The quantitative values of the transport factors that are a measure of the efficiency of the transport of sludge by the fluid streams occurring in the reactor were determined experimentally in reactors of 30- and 200-m(3) volumes. As this was done for wastewater containing lower fatty acids as the main organic pollutants and for sludge with good settling characteristics, the predictive value of the model is limited. It may be used for the second (methane forming) step of anaerobic treatment of wastewater.  相似文献

The application of the expanded granular sludge bed (EGSB) reactor for the anaerobic treatment of low-strength soluble wastewaters using ethanol as a model substrate was investigated in laboratory-scale reactors at 30oC. Chemical oxygen demand (COD) removal efficiency was above 80% at organic loading rates up to12 g COD/L . d with influent concentrations as low as 100 to 200 mg COD/L. These results demonstrate the suitability of the EGBS reactor for the anaerobic treatment of low-strength wastewaters. The high treatment performance can be attributed to the intense mixing regime obtained by high hydraulic and organic loads. Good mixing of the bulk liquid phase for the substrate-biomass contact and adequate expansion of the substrate-biomass contact and adequate expansion of the sludge bed for the degassing were obtained when the liquid upflow velocity (V(up)) was greater than 2.5 m/h. Under such conditions, an extremely low apparent K(s) value for acetoclastic methanogenesis of 9.8 mg COD/L was observed. The presence of dissolved oxygen in the wastewater had no detrimental effect on the treatment performance. Sludge piston flotation from pockets of biogas accumulating under the sludge bed occurred at V(up) lower than 2.5 m/h due to poor bed expansion. This problem is expected only in small diameter laboratory-scale reactors. A. more important restriction of the EGSB reactor was the sludge washout occurring at V(up) higher than 5.5 m/h and which was intensified at organic loads higher than 7 g COD/L. d due to buoyancy forces from the gas production. Thermal extraction was used to quantify extracellular polymers (ECP) in granules from anaerobic upflow reactors. The optimal time for extraction was determined as the time needed before the intracellular material gives a significant contribution to the extracted extracellular material due to cell lysis. ECP contents of 41 to 92 mg · g–1 volatile suspended solids of granules were found depending on the type of granular sludge examined. The content of polysaccharides, protein and lipids in the extracted ECP was quantified. Furthermore, the different methyl esters of the lipids were determined and quantified. Lower amounts of polysaccharides and proteins were found in the extracellular material from granules grown on methanogenic and acetogenic substrates compared to granules grown on more complex substrates. In contrast, the lipid content was lower on complex substrates. Changing the feed of an upflow anaerobic sludge blanket reactor from a sugar-containing waste-water to a synthetic waste-water containing acetate, propionate and butyrate resulted in a decrease in both the protein and polysaccharide content and an increase in the lipid content of the extracellular material. Furthermore, the amount of protein and polysaccharides in the ECP found under mesophilic conditions was significantly higher than under thermophilic conditions, while the lipid content was lower.  相似文献

Aerobic granular sludge sequencing batch reactors (SBR) are a promising technology for treating wastewater. Increasing evidence suggests that aerobic granulation in SBRs is driven by selection pressures exerted on microorganisms. Three major selection pressures have been identified as follows: settling time, volume exchange ratio and discharge time. This review demonstrates that these three major selection pressures can all be unified to one, the minimal settling velocity of bio-particles, that determines aerobic granulation in SBRs. The unified selection pressure theory is a useful guide for manipulating and optimizing the formation and characteristics of aerobic granules in SBRs. Furthermore, the unified theory provides a single engineering basis for scale up of aerobic granular sludge SBRs.  相似文献

Activated sludge is one of the most widely implemented technologies for municipal wastewater treatment. Yet, more restrictive environmental standards demand for more efficient technologies. Aerobic granular sludge (AGS) is a promising alternative in this context since this technology has shown potential for simultaneous organic matter and nutrient removal using smaller bioreactors and consuming less energy. However, despite such engaging claims, only ca. 40 full-scale AGS systems have been installed worldwide after 30 years of development. This reduced implementation suggests the existence of significant bottlenecks for this technology, which currently only have partially been overcome. This overview aims to analyze the recent progress in R&D concerning aerobic sludge granulation for municipal wastewater treatment via the analysis of research articles and invention patents as well as to elucidate exiting technological gaps and development opportunities. Culturing methods aiming at fast granulation, long-term stability and excellent process performance are of utmost interest for promoting massive implementation of full-scale AGS systems. Moreover, the recovery of biomaterials from waste sludge could contribute to the implementation of the biorefinery paradigm in wastewater treatment plants.

Poor long-term stability of aerobic granules developed in sequencing batch reactors (SBRs) remains a limitation to widespread use of aerobic granulation in treating wastewater. Filamentous growth has been commonly reported in aerobic granular sludge SBR. This review attempts to address the instability problem of aerobic granular sludge SBR from the perspective of filamentous growth in the system. The possible causes of filamentous growth are identified, including long retention times of solids, low substrate concentration in the liquid phase, high substrate gradient within the granule, dissolved oxygen deficiency in the granule, nutrient deficiency inside granule, temperature shift and flow patterns. Because of cyclic operation of aerobic granular sludge SBR and peculiarities of aerobic granules, various stresses can be present simultaneously and can result in progressive development of filamentous growth in aerobic granular sludge SBR. Overgrowth of filamentous bacteria under stress conditions appears to be a major cause of instability of aerobic granular sludge SBR. Specific recommendations are made for controlling filamentous growth.  相似文献

The biological removal of nitrogen and phosphorus from nutrient-rich abattoir wastewater using granular sludge has been investigated. A lab-scale sequencing batch reactor, seeded with granular sludge developed using synthetic wastewater, was operated for 13 months under alternating anaerobic and aerobic conditions. It is demonstrated that the granules could be sustained and indeed further developed with the use of abattoir wastewater. The organic, nitrogen, and phosphorus loading rates applied were 2.7 gCOD L(-1) day(-1), 0.43 gN L(-1) day(-1), and 0.06 gP L(-1) day(-1), respectively. The removal efficiency of soluble COD, soluble nitrogen and soluble phosphorus were 85%, 93%, and 89%, respectively. However, the high suspended solids in the effluent limited the overall removal efficiency to 68%, 86%, and 74% for total COD, TN, and TP, respectively. This good nutrient removal was achieved through the process known as simultaneous nitrification, denitrification, and phosphorus removal, likely facilitated by the presence of large anoxic zones in the center of the granules. The removal of nitrogen was likely via nitrite optimizing the use of the limited COD available in the wastewater. Accumulibacter spp. were found to be responsible for most of the denitrification, further reducing the COD requirement for nitrogen and phosphorus removal. Mineral precipitation was evaluated and was not found to significantly contribute to the overall nutrient removal. It is also shown that the minimum HRT in a granular sludge system is not governed by the sludge settleability, as is the case with floccular sludge systems, but likely by the limitations associated with the transfer of substrates in granules.  相似文献

Sterile granular sludge was inoculated with either Methanosarcina mazeii S-6, Methanosaeta concilii GP-6, or both species in acetate-fed upflow anaerobic sludge blanket (UASB) reactors to investigate the immobilization patterns and dynamics of aceticlastic methanogens in granular sludge. After several months of reactor operation, the methanogens were immobilized, either separately or together. The fastest immobilization was observed in the reactor containing M. mazeii S-6. The highest effluent concentration of acetate was observed in the reactor with only M. mazeii S-6 immobilized, while the lowest effluent concentration of acetate was observed in the reactor where both types of methanogens were immobilized together. No changes were observed in the kinetic parameters (Ks and mumax) of immobilized M. concilii GP-6 or M. mazeii S-6 compared with suspended cultures, indicating that immobilization does not affect the growth kinetics of these methanogens. An enzyme-linked immunosorbent assay using polyclonal antibodies against either M. concilii GP-6 or M. mazeii S-6 showed significant variations in the two methanogenic populations in the different reactors. Polyclonal antibodies were further used to study the spatial distribution of the two methanogens. M. concilii GP-6 was immobilized only on existing support material without any specific pattern. M. mazeii S-6, however, showed a different immobilization pattern: large clumps were formed when the concentration of acetate was high, but where the acetate concentration was low this strain was immobilized on support material as single cells or small clumps. The data clearly show that the two aceticlastic methanogens immobilize differently in UASB systems, depending on the conditions found throughout the UASB reactor.  相似文献

Anaerobic sludge granules were obtained from laboratory-scale anaerobic bioreactors used to treat pharmaceutical-like (methanol-, acetone- and propanol-contaminated) wastewater under low-temperature conditions (15 degrees C). The microbial diversity and diversity changes of the sludge samples were ascertained by applying 16S rRNA gene cloning and terminal restriction fragment length polymorphism (TRFLP) analyses, respectively, and using sludge samples from the inoculum, throughout and at the conclusion of the bioreactor trial. Data from genetic fingerprinting correlated well with those from physiological activity assays of the reactor biomass. Specifically, for example, TRFLP profiles indicated the dominance of hydrogenotrophic methanogens within the archaeal community, thus supporting the findings of specific methanogenic activity measurements. TRFLP data supported the hypothesis that the deviation between the replicated reactors, in terms of treatment efficiency, was associated with succession within the microbial communities present, and indicated that community development was linked to both operating temperature and wastewater composition. Fluorescence in situ hybridization (FISH) was also applied, to quantitatively assess the abundance of selected microbial groups, and revealed the underestimation of the abundance Methanosarcina by gene cloning analysis and demonstrated the spatial arrangement of these organisms within the architecture of the low-temperature solvent-degrading anaerobic biofilms.  相似文献

Solutions of sodium caprate and sodium laurate were digested in upflow anaerobic sludge bed (UASB) reactors inoculated with granular sludge and in expanded granular sludge bed (EGSB) reactors. UASB reactors are unsuitable if lipids contribute 50% or more to the COD of waste water: the gas production rate required to obtain sufficient mixing and contact cannot be achieved. At lipid loading rates exceeding 2–3 kg COD m−3 day−1, total sludge wash-out occurred. At lower loading rates the system was unreliable, due to unpredictable sludge flotation. EGSB reactors do fulfil the requirements of mixing and contact. They accommodate space loading rates up to 30 kg COD m−3 day−1 during digestion of caprate or laurate as sole substrate, at COD removal efficiencies of 83–91%, and can be operated at hydraulic residence times of 2 h without any problems. Augmentation of granular sludge in lab-scale EGSB reactors was demonstrated. The new granules had excellent settling properties. Floating layer formation, as well as mixing characteristics in full-scale EGSB reactors require further research.  相似文献

New denitrifying strains of phototrophic bacteria isolated from photosynthetic sludge reactors for wastewater treatment were characterized. All of the new isolates were mesophilic, nonhalophilic, facultative photoheterotrophs that were able to grow by anaerobic photosynthesis, aerobic respiration, or nitrate respiration. They had ovoid cells that were motile by single polar flagella, formed vesicular photosynthetic membranes together with bacteriochlorophyll a and carotenoids of the spheroidene series, required biotin, thiamine, and biotin as growth factors, and utilized a wide variety of organic compounds as electron donor and carbon sources. In these respects, the isolates most closely resembled Rhodobacter sphaeroides. However, they differed from this species in utilizing malonate and dulcitol but not tartrate as carbon sources and in their inability to grow anaerobically in darkness with trimethylamine N-oxide or dimethylsulfoxide as a terminal oxidant. Partial sequencing of 16S rRNA genes provided evidence for genetic differences between the new isolates and R. sphaeroides or other members of the genus Rhodobacter. Activities of nitrate reductase, nitrite reductase, and nitrous oxide reductase were detected in intact cells of one of the new isolates. All these enzyme activities were induced by cultivation with nitrate.  相似文献
