Compatible and incompatible reactions in rice plants (Oryza sativa L. cv. Shenxianggen No.4) were resulted from inoculation with two different virulent races of rice blast fungus (Magnaporthe grisea (Hebert) Barr), and thus an effective infecting system was established between rice plants and the rice blast pathogen. Two cDNA clones that showed induced and temporal patterns in expression in the very early stage in response to infection of the fungus were obtained from the plants by use of differential display. Of the two cDNA clones, Fastresp-a was induced to express in both compatible and incompatible interactions although it was expressed earlier in the former reaction. The second one, Fastresp-b, was only expressed in incompatible interaction. Southern blot analysis of the rice genomic DNA indicated that both of the two clones were from genome of the plant. No significant homology to the two genes was found from the rice gene database. This suggested that they were novel genes in rice and may play important roles in rice resistant response to infection of rice blast fungus.  相似文献

Rice breeding and crop research predominantly emphasize adaptation to ecological conditions. Based on qualitative and quantitative research conducted between 2000 and 2012 we show how ecological factors, combined with socioeconomic variables, cultural norms and values, shape the use and development of local technologies related to the cultivation of African rice (Oryza glaberrima Steud.) in seven West African countries (Ghana, Guinea, Guinea-Bissau, Senegal, Sierra Leone, The Gambia and Togo). In this region the role of African rice is diverse across ethnic groups. Findings suggest that farmers, through various pathways, are active in the development of promising new varieties based on genetic resources of Asian rice, African rice, or both, as well as in the adoption of modern varieties. These findings require further research into interactions among ecological, genetic, socioeconomic and cultural factors within farmers?? innovation systems and recognition of emergent knowledge and technologies resulting from such interactions.  相似文献

Many factors produced by the pathogen Magnaporthe oryzae enhance its ability to infect rice. We found a novel infection-promoting activity in the supernatant of a conidia suspension (SCS) of M . oryzae . The addition of SCS promoted the invasion of excised rice leaf sheaths by infectious hyphae. The activity was heat-stable and was found in SCSs from five virulent and three avirulent isolates of M . oryzae on the rice cv. Nipponbare ( Pia ). The effect was exclusively detected in compatible interactions. The infection of rice plants by non-rice blast fungi was not enhanced by SCS. These results suggest that SCS includes a heat-stable factor(s) that promotes M . oryzae infection during compatible interactions.  相似文献

Joint effects of Cd and other heavy metals (Pb, Cu, Zn and As) on the growth and development of rice plants and the uptake of these heavy metals by rice were studied using the pot-culture method combined with chemical and statistical analyses. The results showed that the growth and development of rice plants were strongly influenced by the double-element combined pollution. There was an average decrease in the height of rice plants of 4.0–5.0 cm, and grain yield was decreased by 20.0–30.0%, compared with the control. The uptake of Cd by rice plants was promoted due to the interactions between Cd and the other heavy metals added to the soil. The Cd concentration in roots, stems/leaves and seeds increased 31.6–47.7, 16.7–61.5 and 19.6–78.6%, respectively. Due to interactions, uptake of Pb, Cu and Zn by roots and stems/leaves was inhibited, accumulation of Pb, Cu and Zn in seeds was increased, uptake of As by roots was promoted and uptake of As by stems/leaves was inhibited. In particular, the upward transporting ability of the heavy metals absorbed by rice plants was significantly increased.  相似文献

穗长是影响水稻(Oryza sativa)产量的重要因子之一, 研究水稻穗长QTL间的上位性效应对于发掘水稻产量潜力具有重要意义。该研究以16个单片段代换系(single segment substitution lines, SSSLs)和15个双片段聚合系(double segment pyramiding lines, DSPLs)为材料研究了水稻穗长QTL的加性及上位性效应。以P<0.01为阈值, 共检测到6个穗长QTL和9对基因互作座位。其中2个(Pl-2和Pl-10)是尚未报道的穗长QTL。穗长QTL互作后, 一些互作对的上位性效应与单个QTL的作用方式及效应大小各不相同, 预示着基因聚合后会产生不同的互作效应。该研究结果对于通过分子聚合育种手段改良穗长具有重要意义。  相似文献

以2个粳型光温敏核不育系和4个籼稻品种为材料,配制籼粳交组合,用包括基因型×环境互作效应的胚乳性状遗传模型对3个蒸煮品质性状(直链淀粉含量、胶稠度、碱消值)进行了遗传研究,结果表明:直接加性和母体加性效应对三个性状的遗传变异起主要作用.基因型×环境互作主要表现为显性×环境以及细胞质×环境互作.直链淀粉含量的普通遗传率都不显著,只有较高的互作母体遗传率;胶稠度具有显著的普通直接遗传率和互作细胞质遗传率;碱消值的普通直接遗传率和普通母体遗传率都极显著.根据遗传效应预测值对供试亲本的利用价值作了评价。  相似文献

病程相关(PR)蛋白质经常被用作抗病反应的分子标记。利用免疫印迹(WB)技术检测了7个PR1家族蛋白质在水稻(Oryza sativa)叶片生长及与白叶枯病菌互作反应过程中的表达,发现6个PR1家族蛋白质在叶片生长中有表达。检测PR1蛋白质在Xa21介导的抗白叶枯病过程中的表达,结果显示PR1#052、PR1#072、PR1#073和PR1#121四个蛋白质在抗病反应后期呈上调或诱导表达,PR1#071则表达下调。进一步比较它们在抗病、感病和对照(Mock)反应中的表达丰度,发现在抗病和感病反应中的变化幅度均明显大于对照反应,推测这些PR蛋白质在水稻-白叶枯病菌互作反应中发挥作用。另外,对PR1基因上游启动子区的cis元件进行了分析。该研究初步揭示了水稻PR1家族蛋白质的表达谱,为进一步了解PR1蛋白质的功能提供了线索。  相似文献

将稻瘟菌细胞壁来源的专化性糖蛋白激发子接种于一套水稻抗稻瘟病近等基因系后,非亲和性互作水稻超氧阴离子(O-.2)积累在互作早期明显高于亲和性互作水稻;超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性均趋于下降,不同亲和性互作水稻间的差异不明显;脂氧合酶(LOX)活性在水稻/激发子非亲和互作早期增加明显、速度快;这些指标的变化进而导致非亲和性互作水稻的膜脂过氧化,其相对电导率及丙二醛(MDA)含量的高峰期和强度也明显早于和高于亲和性互作水稻.非亲和性互作水稻过氧化物酶(POD)活性在互作早期明显高于亲和性互作水稻,可能与其参与其它抗性有关.研究同时表明,激发子可专化性诱导完全和高度非亲和性互作水稻的过敏性坏死反应;而中度非亲和性互作和亲和性互作水稻则未发生过敏性(HR)坏死反应.这些结果表明,膜脂过氧化和HR反应的发生是激发子诱导水稻抗性的主要生理机制之一.  相似文献

稻瘟病是世界上影响水稻(Oryza sativa)粮食生产的主要病害之一, 抗病基因的发掘与利用是抗病育种的基础和核心。随着寄主水稻和病原菌稻瘟病菌(Magnaporthe oryzae)基因组测序和基因注释的完成, 水稻和稻瘟病菌的互作体系成为研究植物与真菌互作的模式系统。该文对稻瘟病抗病基因的遗传、定位、克隆及育种利用进行概述, 并通过生物信息学分析方法, 探讨了水稻全基因组中NBS-LRR类抗病基因在水稻12条染色体上的分布情况, 同时对稻瘟病菌无毒基因的鉴定及无毒蛋白与抗病蛋白的互作进行初步分析。最后对稻瘟病抗病基因研究存在的问题进行分析并展望了未来的研究方向, 以期为水稻抗稻瘟病育种发展和抗病机制的深入理解提供参考。  相似文献

用由247个株系组成的珍汕97B/密阳46重组自交系群体及其含207个分子标记的连锁图谱,在2002年和2003年分别测定亲本和重组自交系群体开花后10 d和20 d籽粒的淀粉分支酶的活性,检测到3个控制开花后10 d Q酶活性的主效应QTL(qnantitative trait loci),联合贡献率为10%,其中qQ10-6与环境发生显著的互作;分别检测到5对和2对染色体区间对开花后10 d、20 d Q酶活性的影响具有加性×加性上位性作用,其中开花后10 d的3对染色体区间具有显著的上位性×环境互作效应.由此可见,水稻籽粒Q酶活性相关基因的表达,受到环境因子的极大影响.  相似文献

Specific effects of food processing on food digestion remain to be clearly established. The objective of this study was to determine the effect of rice processing (cooked brown and white rice) on the rheological properties, chemical composition (water, starch, fiber, protein) and pH of gastric chyme in pigs over a two hour post-ingestion period in the proximal and distal regions of the stomach. Results showed the distal region having a higher moisture content, lower pH, and lower viscosity compared to the proximal region for both brown and white rice. Starch content was significantly affected by the region x time x rice interaction (p?=?0.0259), protein content by both the rice x time (p?=?0.0143) and the region x time (p?=?0.0181) interactions, and total dietary fiber by rice type (p?=?0.005). Rice digesta behaved as a gel, with G’ (42288–1911 Pa) greater than G” (6307–360 Pa). The extent of particle breakdown for brown and white rice was different. Overall, the results indicated a connection between food processing and food breakdown during gastric digestion. These findings have implications in the management of nutrient absorption and satiety control that can be used in future food design.  相似文献

Gall midges are insects specialized in maneuvering plant growth, metabolic and defense pathways for their benefit. The Asian rice gall midge and rice share such an intimate relationship that there is a constant battle for survival by either partner. Diverse responses by the rice host against the midge include necrotic hypersensitive resistance reaction, non-hypersensitive resistance reaction and gall-forming compatible interaction. Genetic studies have revealed that major R (resistance) genes confer resistance to gall midge in rice. Eleven gall midge R genes have been characterized so far in different rice varieties in India. In addition, no single R gene confers resistance against all the seven biotypes of the Asian rice gall midge, and none of the biotypes is virulent against all the resistance genes. Further, the interaction of the plant resistance gene with the insect avirulence gene is on a gene-for-gene basis. Our recent investigations involving suppressive subtraction hybridization cDNA libraries, microarray analyses, gene expression assays and metabolic profiling have revealed several molecular mechanisms, metabolite markers and pathways that are induced, down-regulated or altered in the rice host during incompatible or compatible interactions with the pest. This is also true for some of the pathways studied in the gall midge. Next generation sequencing technology, gene expression studies and conventional screening of gall midge cDNA libraries highlighted molecular approaches adopted by the insect to feed, survive and reproduce. This constant struggle by the midge to overcome the host defenses and the host to resist the pest has provided us with an opportunity to observe this battle for survival at the molecular level.  相似文献

Rice proteins that bind single-stranded G-rich telomere DNA   总被引:4,自引:0,他引:4  
In this work, we have identified and characterized proteins in rice nuclear extracts that specifically bind the single-stranded G-rich telomere sequence. Three types of specific DNA-protein complexes (I, II, and III) were identified by gel retardation assays using synthetic telomere substrates consisting of two or more single-stranded TTTAGGG repeats and rice nuclear extracts. Since each complex has a unique biochemical property and differs in electrophoretic mobility, at least three different proteins interact with the G-rich telomere sequences. These proteins are called rice G-rich telomere binding protein (RGBP) and none of them show binding affinity to double-stranded telomere repeats or single-stranded C-rich sequence. Changing one or two G's to C's in the TTTAGGG repeats abolishes binding activity. RGBPs have a greatly reduced affinity for human and Tetrahymena telomeric sequence and do not efficiently bind the cognate G-rich telomere RNA sequence UUUAGGG. Like other telomere binding proteins, RGBPs are resistant to high salt concentrations. RNase sensitivity of the DNA-protein interactions was tested to investigate whether an RNA component mediates the telomeric DNA-protein interaction. In this assay, we observed a novel complex (complex III) in gel retardation assays which did not alter the mobilities or the band intensities of the two pre-existing complexes (I and II). The complex III, in addition to binding to telomeric sequences, has a binding affinity to rice nuclear RNA, whereas two other complexes have a binding affinity to only single-stranded G-rich telomere DNA. Taken together, these studies suggest that RGBPs are new types of telomere-binding proteins that bind in vitro to single-stranded G-rich telomere DNA in the angiosperms.  相似文献
