Glucose-limited bean cells (Phaseolus vulgaris L.) were grown in a modified bacterial fermentor at a constant pH of 4.8. The cultures were kept in steady state at different specific growth rates varying from 0.00216 h–1 to 0.0106 h–1. Culture conditions are described that are needed to start a continuous culture. First, it was essential to use log-phase cells as starting material. Second, it was important to increase the dilution rate gradually, otherwise cells in the culture aggregated. Cells grown at the highest dilution rate employed contained twice as much protein per gram dry weight as cells grown at the lowest dilution rate. The composition of the cell walls also varied with the dilution rate in contrast to their relatively constant composition when grown in batch culture.  相似文献

Summary Saccharomyces cerevisiae were observed to undergo drastic morphological changes when grown in continuous culture. Peak elongations and minimum cell volumes were found at intermediate dilution rates when it was believed the insitu glucose concentration was at its lowest. Summary Various ion exchange resins were tested for their ability to adsorb cells of Saccharomyces cerivisiae with the ultimate intention of developing a packed bed immobilized cell reactor for the continuous production of ethanol. The resins varied greatly in their ability to adsorb cells - the least effective resins retained less than 1 mg S. cerivisiae cells (dry weight)/g of resin (dry weight), and the most effective, 130–140 mg cells/g of resin. A column reactor packed with adsorbed yeast cells was operated continuously for over 200 hours using a 12% (w/v) glucose medium at dilution rates of 1.1 h-1 and 1.44 h-1 (based on void volume). High ethanol productivities of 53.1 and 62.0 g ethanol/l-h were obtained.  相似文献

Clostridium acetobutylicum was grown in continuous culture under ammonium limitation (15.15 mM NH4 +). At a pH of 6.0 and at various dilution rates only acetate, butyrate and ethanol were formed as non-gaseous products. A decrease of the pH to values between 5.2 and 4.3 resulted in a shift of the fermentation towards acetone-butanol formation.  相似文献

Summary The effects of forced square-wave perturbations in the dilution rate on plasmid maintenance and gene expression of a population ofEscherichia coli K12 strain carrying the vector plasmid pBR322 grown in a chemostat with a non-selective medium were studied. It was observed that in the control experiments, where the dilution rates were kept constant, the percentage of plasmid-containing cells decreased after a period of time. Eventually, the culture was displaced by the plasmid-free cells. However, when the cells were exposed to forced oscillations in the dilution rate, the reactor culture was able to maintain a mixed population of plasmid-free and plasmid-containing cells for a longer period of time. The above observation seems to be independent of the source of the host cells. That is, the same results were obtained when the plasmid-free cells were generated from the culture itself due to defective partitioning of the plasmids or introduced externally.  相似文献

Dioscorea deltoidea cell suspension cultures were grown at initial sucrose concentrations of 35 to 200 g/L. The growth rates were similar (about 0.50 day–1) with all of the initial sugar concentrations examined. The ratio of fresh weight to dry weight of cells was dependent on the initial sugar concentration, however, it remained fairly constant as long as the sugar was present in the growth medium. These results are different from results recently published, claiming that the growth rate of D. deltoidea cells is dependent on sugar concentration and the fresh weight to dry weight ratio increases throughout growth.  相似文献

The objectives of these studies were to determine cell yield and fermentation responses of a Salmonella enterica serovar Typhimurium poultry isolate using various dilution rates in steady state continuous culture incubations. S. enterica Typhimurium cells were propagated in continuous cultures with a total volume of 0.50 l of Luria Bertani medium containing 0.1% glucose. Dilution rates from 0.0125 to 1.44/h were used. Cell protein concentration generally increased linearly with increased dilution rate up to a rate of 0.54/h and declined at the higher dilution rates. Glucose consumption gave a similar pattern to cell protein concentration by declining at the three highest dilution rates. Short chain fatty acid production was inconsistently influenced by dilution rate. Acetate, the most predominant fatty acid produced, declined at the higher dilution rates, as did propionate. Ammonia production remained stable at the lowest dilution rates, but increased significantly at a dilution rates above 0.27/h.  相似文献

Data for disruption of C. utilis, S. cerevisiae and B. subtilis cells by impingement of a high velocity jet of suspended cells against a stationary surface are compared. Differences between organisms were observed, but there were no general differences found between yeast and bacteria. Summary A computer-assisted on-line glucose analyzer was developed for feed-back control of cell growth. Using this system the glucose consumption rate for Escherichia coli was determined to be linear during batch culture at 0.37 g/hr. On-line feed-back control of glucose concentration at 1.5±0.5 g/L was used with fed-batch cultures to produce 31.2 g dry weight of E. coli cells/L in 12 h.  相似文献

Summary Trichoderma reesei Rut-C30 was found to produce extracellular lactase when grown on lactose medium. Maximum enzyme levels in continuous culture were observed at dilution rates (D) between 0.02 and 0.027 hr-1. The enzyme productivity reached 27.3 U/L hr at D = 0.027 hr-1. Lactase synthesis appears to be inducible and subject to catabolite repression. Optimal growth temperature and pH for enzyme production were 28°C and pH 5. Maximum enzyme activity was observed at 63°C and pH 4.6. The apparent Km, based on the nitrophenyl-galactopyranoside assay was estimated as 0.4 mM. The enzyme is suitable for lactose hydrolysis in acid whey.  相似文献

Summary Experiments were conducted with Zymomonas mobilis in an attached film expanded bed (AFEB) fermentor at different dilution rates, using a feed glucose concentration of 100 gm/l. Vermiculite was used as the bed material for attachment of the bacterial film. Ethanol volumetric productivities were maximum at a dilution rate of 3.6hr-1. The productivities were 105 gm/l-hr and 210 gm/l-hr based on total fermentor volume and bed volume, respectively.  相似文献

Selenomonas ruminantium HD4 produced significant quantities of d- and l-lactate from glucose in batch culture. Both isomers also supported growth if fumarate was present. In glucose-limited continuous culture, d-lactate was detected in the medium only at fast dilution rates. In continuous-culture-grown cells, only a cytoplasmic NAD-dependent l-lactate dehydrogenase (LDH) and a membrane-associated NAD-independent l-LDH were detected; activity of the soluble enzyme was twice as high at the fast dilution rate as at the slow dilution rate. Lactate racemase was also detected; its activity was 4-fold higher at the fast dilution rate. The presence of racemase explains why d-lactate was made and used by this organism.  相似文献

In vitro culture ofBrassica alba anthers on a growth medium containing inorganics of KB5 and organics, iron, sucrose and hormones of B5 resulted in a very high response of anthers (93.75%) towards callus induction. All the calli transferred to regeneration media responded favourably even after six months of callus induction. Numerous torpedo-shaped embryoids developed in clusters at many sites from each callus mass. Secondary embryogenesis and multiple shoot formation was also observed in many cases. The number of embryoids and plantlets produced by one embryogenic anther were as high as 169.8 and 17 respectively. 87% of the regenerated plants were haploids.  相似文献

Summary An immobilization technique has been developed for the conversion of both cellobiose and xylose to ethanol, which may be considered as one stage of a process for the conversion of cellulosic biomass to ethanol. Relatively inexpensive charcoal was used as a support material, with 23 mg dry weight of Clostridium saccharolyticum cells per g dry weight of support. Tests were run for 170 h at 0.15 1/h dilution rate. From a 3% (w/v) sugar mixture, 0.7% (w/v) ethanol was obtained with over 97% cellobiose and 62% xylose utilization.  相似文献

Papaver somniferum L. (opium poppy) cells were elicited with a Botrytis sp. homogenate and cultured by a semi-continuous process. Elicitation induced synthesis of sanguinarine and dihydrosanguinarine. Significant release of both alkaloids into the culture medium occurred. Medium exchange at 2-day intervals enabled product recovery from spent medium and maintained culture viability. Culture growth was not inhibited by elicitor treatment necessitating sub-culture prior to re-elicitation. Cell suspension cultures of Nicotiana were initiated using conditions designed to selectively favor stable chromosome number. These conditions included use of leaf explants to initiate cultures, growth of cells in culture medium containing 2,4-D, and transfer of cells with short subculture intervals. Four cell lines derived from Nicotiana tissue with 2n=24, 48, or 72 were established and retain stable chromosome number. Each line could be regenerated to recover plants that retained the somatic chromosome number during culture. Establishment of haploid and diploid cell lines with stable chromosome number is important for mutant isolation and protoplast fusion.  相似文献

Summary Ethanol production byZ.mobilis has been studied in continuous culture with 10, 15 and 20% glucose media. At 10% glucose, steady state conditions were achieved under glucose-limited conditions. At 15 and 20% glucose, the glucose was not fully metabolized even at low dilution rates and oscillatory behavior was evident. It is proposed that ethanol inhibition of growth is responsible for these phenomena. Comparison of kinetic parameters with those from previously published batch data revealed similar values. The maintenance energy coefficient (m) forZ.mobilis was relatively high and was calculated as 1.6 g/g/h for 10% glucose and 3.1 g/g/h for 15% glucose.  相似文献
