Summary This paper describes variations of a mode of stomatal development already described in a species (E. orbifolia) ofEucalyptus L'Herit. (Carr andCarr, Protoplasma 96, 127, 1978) in which the outer part of the stomatal pore (ostiole) is formed by the creation of a break in the thin layer of cuticle lying over the stomatal chamber. In a number of species with a thick cuticle (e.g., E. cooperana) the process of breakthrough is different: additions to the guard cell upper thickenings extend from them as ridges, pressing the leaf cuticle outwards. Breakthrough of the cuticle occurs above the tips of these extensions. The anterior chamber is lined throughout by the extensions, which become heavily cutinized. This mode of stomatal development is typical of many other species of eucalypts, including those dealt with in this paper.In addition, inE. halophila thickenings develop on the end walls of the anterior chamber above the unusual upturned poles of the guard cells. Cutinized thickenings, pseudo-outer stomatal ledges are also formed on the upper guard cell walls. All these wall thickenings occlude the anterior chamber, leaving only a narrow passage in the form of a letter H.Similar occlusions are found inE. balladoniensis but here the thickenings are developed into the chamber from its lateral walls. InE. gracilis, and the related speciesE. celastroides andE. calycogona, less regular occluding thickenings develop principally from the lateral walls of the chamber. In addition, large pseudo-outer stomatal ledges may be formed.These phenomena are discussed in terms of the mechanism underlying the formation of the occluding thickenings and the possibility of their adaptive significance.  相似文献

The structure of the stomata of the floating leaves ofSalvinia herzogii has been analyzed with light and scanning electron microscopy. Peculiar extensions (cuticle?) of the outer and inner tangential sides of the walls of the guard-cells are described. Both extensions, partially joined at their ends, are encircling a pore. This structure is used to propose an alternative explanation for the risk of the occlusion of the ostioles, different from that given byHaberlandt. The conductivity of these stomata was checked with a porometer and the ostioles proved to be permanently open. Their type of stomatogenesis is discussed. The stomata ofSalvinia are considered to be vestigial structures and their sunken position is taken as evidence in support of a xeric hypothetic ancestor of the genus.  相似文献

The polytrophic ovarioles of three insect species, the fruit fly Drosophila melanogaster, the fungus gnat Bradysia tritici, and the honeybee Apis mellifera, were compared morphologically and with respect to the cytological organization of the peripheral somatic layers. Staining with rhodaminyl-phalloidin revealed differences in the organization of the muscle strands of the epithelial sheath and the microfilament pattern in the basal part of the follicular epithelium (mid-vitellogenic stages). Also, the size of the intercellular space between the follicle cells differed considerably in the three analyzed species. The basement membrane of Drosophila and Bradysia follicles was partially digested using purified collagenase. The observed morphological changes indicated that in both species the basement membrane of the follicular epithelium plays an important role in shaping the follicles. The possible functional significance of the species-specific structural differences is discussed.  相似文献

Summary A mature stomate of the water fernAzolla consists of a single apparently unspecialized annular guard cell (GC) with two nuclei surrounding an elongated pore aligned longitudinally in the leaf. During development, the guard mother cell develops a preprophase band (PPB) of microtubules (MTs) oriented transverse to the leaf axis. This is followed by a cell plate which fuses with the parental walls at the PPB site. Subsequently only the central part of the cell plate is consolidated, while the parts to either side become perforated and tenuous and may disperse completely, forming a single composite GC.Meanwhile, a dense array of MTs appears along both faces of the central part of the new wall, oriented normal to the leaf surface. Further MT arrays radiate out across the periclinal walls from the region of the consolidated cell plate. Putative MT nucleating sites are seen along the cell edges between these anticlinal and periclinal arrays. Polarized light microscopy reveals cellulose deposition parallel to the periclinal MT arrays. At the same time lamellar material is deposited within the new anticlinal wall. As the GC complex elongates, a split appears in these lamellae creating an initially transverse slit which then opens up to become first circular and ultimately an elongated pore aligned in the long axis of the leaf,i.e., at right angles to the wall in which it originated. The radiating pattern of cellulose microfibrils in the periclinal walls contributes to the shaping of the pore. Elongation at the apical and basal ends of the GC is restricted by longitudinal microfibril orientation, while that at the sides is facilitated by transverse alignment.  相似文献

Stomata are epidermal bi-celled structures that differentiate within special cell lineages initiated by a subset of protodermal cells. Recently, we showed that the Arabidopsis photomorphogenic repressor COP10 controls specific cell-lineage and cell-signaling developmental mechanisms in stomatal lineages. Loss-of-function cop10-1 mutant cotyledons and leaves produced (in the light and in the dark) abundant stomatal clusters, but nonlineage epidermal cells were not affected. Here we examine COP10 role in hypocotyls, cylindrical organs displaying a distinct epidermal organization with alternate files of protruding and non-protruding cells, with the latter producing a limited number of stomata. COP10 prevents stomatal clusters and restricts stomata production in hypocotyls; these roles are specific to lineage cells as in cotyledons, since COP10 loss of function does not elicit stomatal fate in nonlineage cells; COP10 also sustains the directional cell expansion of all hypocotyl epidermal cell types, and seems necessary for the differentiation between protruding and non-protruding cell files.  相似文献

The influence of far-red (FR; 700–800 nm) radiation on steady-state stomatal conductance and net photosynthesis in P. vulgaris has been studied. Whereas FR radiation alone was relatively ineffective, addition of FR to a background of white light (WL; predominantly 400–700 nm) resulted in increased stomatal conductance. Stomata exhibited a marked diurnal sensitivity to FR. The action maximum for enhancing stomatal conductance was near 714 nm. A combination of FR and infra-red (IR; >800 nm) enhanced net photosynthesis when added to a background of WL. When IR alone was added to WL, there was a net decrease in photosynthesis, indicating that it is the FR waveband which is responsible for the observed photosynthetic effects. Naturally occurring levels of FR radiation (235 mol·m-2·s-1) in vegetation-canopy shade enhanced net photosynthetic CO2 gain by 28% when added to a background of 55 mol·m-2·s-1 WL.Abbreviations BL blue - FR far-red - IR infra-red - PAR photosynthetically active radiation - R red - WL white light  相似文献

Exposure of barley plants (Hordeum vulgare L.) to soil flooding for 2 to 24 h reduced the net photosynthetic rate and transpiration rate. Stomatal conductance also decreased in flooded plants. Stomatal closure started within 2 – 6 h and stomata remained closed up to 24 h of treatment.  相似文献

Chloride ions are necessary to compensate for the positively charged potassium ions imported into guard cells of Allium cepa L. during stomatal opening. Therefore an external Cl- supply of intact Allium plants is important. But high levels of chloride have been found to reduce the sensitivity of the starch-lacking stomata and isolated guard cell protoplasts (GCPs) from Allium to potassium ions, fusicoccin and abscisic acid. Furthermore, with high levels of chloride, malate anions disappear from the guard cells of Allium, a finding which contrasts with situation in Vicia where the stomatal sensitivity to K+ ions, fusicoccin and ABA is not influenced by Cl- ions and malate levels are unaffected. Straight-chain saturated fatty acids (C6-C11) and abscisic acid (ABA) accumulate in the leaves of Phaseolus vulgaris L. and Hordeum vulgare L. under water stress. ABA and certain of the fatty acids, particularly decanoic and undecanoic acid, can inhibit stomatal opening and cause stomatal closure in epidermal strips of Commelina communis L. depending on the incubating medium used. 10-4 M (±)-ABA inhibits opening in media containing either high or relatively low concentrations of KCl but causes closure only in the latter medium. The fatty acids (at 10-4 M) prevent opening in both media while significant closure of open stomata was caused only by undecanoic acid in both media and, additionally, by decanoic acid in the low-KCl medium. 10-4 M formic acid also caused stomatal closure and prevented opening to significant extents in the low-KCl medium (it was not tested in the high-KCl medium). The efficacy of undecanoic acid in causing 50% inhibition of opening is about three orders of magnitude lower than that of ABA. At a concentration of 10-3 M, nonanoic, decanoic and particularly undecanoic acid and all-trans-farnesol cause increased cell leakage in Beta vulgaris L. root tissue. Undecanoic acid (10-4 M) also causes some loss of guard cell integrity in C. communis within 1.5 h of treatment. ABA (10-4 M) reduces transpiration rates in barley and C. communis leaves when applied via the transpiration stream but decanoic and undecanoic acids did not have this effect. Observations made with primary leaves of Phaseolus vulgaris L. demonstrated that phytochrome modulates light-induced stomatal movement. Removal of the far-red-absorbing form of the pigment (Pfr) with far-red (FR) radiation decreased the time required by the stomata to reach maximal opening following a dark-to-light transition; this effect of FR was fully reversible with red. Removal of Pfr with FR also decreased the time required to reach maximal closure following a light-to-dark transition, and the rate of closure was dependent on the final irradiation treatment before darkness. The physiological and biochemical changes during the adaptation of Nostoc muscorum to salt are accompanied by specific structural changes. Cells of Nostoc muscorum exposed to saline medium vary in size and envelope organization. There are also drastic changes in the intracellular organization of the thylakoidal assembly. The heterocysts exhibit a preferential tolerance to NaCl rather than mannitol. These findings suggest that Nostoc muscorum is equipped with a specific physiological capacity for NaCl tolerance.  相似文献

Plastocyanin is one of the best characterized of the photosynthetic electron transfer proteins. Since the determination of the structure of poplar plastocyanin in 1978, the structure of algal (Scenedesmus, Enteromorpha, Chlamydomonas) and plant (French bean) plastocyanins has been determined either by crystallographic or NMR methods, and the poplar structure has been refined to 1.33 Å resolution. Despite the sequence divergence among plastocyanins of algae and vascular plants (e.g., 62% sequence identity between theChlamydomonas and poplar proteins), the three-dimensional structures are remarkably conserved (e.g., 0.76 Å rms deviation in the C positions between theChlamydomonas and poplar proteins). Structural features include a distorted tetrahedral copper binding site at one end of an eight-stranded antiparallel -barrel, a pronounced negative patch, and a flat hydrophobic surface. The copper site is optimized for its electron transfer function, and the negative and hydrophobic patches are proposed to be involved in recognition of physiological reaction partners. Chemical modification, cross-linking, and site-directed mutagenesis experiments have confirmed the importance of the negative and hydrophobic patches in binding interactions with cytochromef and Photosystem I, and validated the model of two functionally significant electron transfer paths in plastocyanin. One putative electron transfer path is relatively short (4 Å) and involves the solvent-exposed copper ligand His-87 in the hydrophobic patch, while the other is more lengthy (12–15 Å) and involves the nearly conserved residue Tyr-83 in the negative patch.  相似文献

Structure and ontogeny of stomata and trichomes have been studied in 23 species and 3 varieties of theUrticales. Stomata are anomocytic, more rarely paracytic; anisocytic and sometimes helicocytic and transitorial types are found inUrticaceae andDorstenia, rarely inArtocarpus. The ontogeny of anomocytic and actinocytic stomata is perigenous, of paracytic either mesogenous or perigenous, of anisocytic either mesogenous or mesoperigenous, and of helicocytic and transitional types mesogenous. Among trichomes eglandular unicellular (wide spread), bicellular or uniseriate filiform (Cannabis); glandular capitate with uni- or bicellular (Moraceae, Ulmaceae, Cannabaceae), uniseriate filiform (Ulmaceae) or multiseriate stalk (Cannabis); sunken glands (Artocarpus); uniseriate glandular with uniseriate stalk (Celtis), and stinging emergences (Urticaceae) have been observed. It is concluded that theUrticales represent a natural order with four families:Ulmaceae, Moraceae, Urticaceae andCannabaceae which are distinct but interrelated with each other.  相似文献

Summary ResA is a positive regulator of genes for extracellular polysaccharide biosynthesis in the Enterobacteriaceae. The nucleotide sequence of the rcsA gene from Erwinia stewartii was determined and compared to rcsA sequences from E. amylovora, Escherichia coli, and Klebsiella pneumoniae. Three highly conserved regions of the gene were identified. The C-terminal end of the open reading frame (ORF) shared significant amino acid homology to the LuxR class of bacterial activator proteins. Insertion and deletion mutagenesis of the 5 non-coding region indicated that maximal expression of rcsA was dependent upon cis-acting regulatory sequences located more than 300 by upstream of the translational start site.  相似文献

The development and structure of stomata on the succulent leaves of six species ofSenecio are reported. It was found that in all the species investigated the development of stomata was of the mesogenous type and the structure of the stomata was anisocytic. The constancy in the development and structure of stomata in all the species of the genus investigated agrees with the view that a stomatal type and its developmental modes are constant at the genus level. Abnormalities such as contiguous stomata, degenerated guard cells, abortive and single guard cells are also recorded as natural phenomena.  相似文献

Streptococcus pneumoniae expresses on its surface adhesive pili, involved in bacterial attachment to epithelial cells and virulence. The pneumococcal pilus is composed of three proteins, RrgA, RrgB, and RrgC, each stabilized by intramolecular isopeptide bonds and covalently polymerized by means of intermolecular isopeptide bonds to form an extended fiber. RrgB is the pilus scaffold subunit and is protective in vivo in mouse models of sepsis and pneumonia, thus representing a potential vaccine candidate. The crystal structure of a major RrgB C-terminal portion featured an organization into three independently folded protein domains (D2-D4), whereas the N-terminal D1 domain (D1) remained unsolved. We have tested the four single recombinant RrgB domains in active and passive immunization studies and show that D1 is the most effective, providing a level of protection comparable with that of the full-length protein. To elucidate the structural features of D1, we solved the solution structure of the recombinant domain by NMR spectroscopy. The spectra analysis revealed that D1 has many flexible regions, does not contain any intramolecular isopeptide bond, and shares with the other domains an Ig-like fold. In addition, we demonstrated, by site-directed mutagenesis and complementation in S. pneumoniae, that the D1 domain contains the Lys residue (Lys-183) involved in the formation of the intermolecular isopeptide bonds and pilus polymerization. Finally, we present a model of the RrgB protein architecture along with the mapping of two surface-exposed linear epitopes recognized by protective antisera.  相似文献

In the regeneration of a shoot from a leaf of the succulent, Graptopetalum paraguayense E. Walther the first new organs are leaf primordia. The original arrangement of cellulose microfibrils and of microtubules (MTs) in the epidermis of the leaf-forming site is one of parallel, straight lines. In the new primordium both structures still have a congruent arrangement but it is roughly in the form of concentric circles that surround the new cylindrical organ. The regions which undergo the greatest shift in orientation (90°) were studied in detail. Departures from the original cellulose alignment are detected in changes in the polarized-light image. Departures from the original cortical MT arrangement are detected using electron microscopy. The over-all reorganization of the MT pattern is followed by the tally of MT profiles, the various regions being studied in two perpendicular planes of section. This corrects for the difference in efficiency in counting transverse versus longitudinal profiles of MTs. Reorientation takes place sporadically, cell by cell, for both the cellulose microfibrils and the MTs, indicating a coordinated reorientation of the two structures. That MTs and cellulose microfibrils reorient jointly in individual cells was shown by reconstruction of the arrays of cortical MTs in paradermal sections of individual cells whose recent change in the orientation of cellulose deposition had been detected with polarized light. Closeness of the two alignments was also indicated by images where the MT and microfibril alignments co-varied within a single cell. The change-over in alignment of the MTs appears to involve stages where arrays of contrasting orientation co-exist to give a criss-cross image. During this critical reorganization, the frequency of the MTs is high. It falls during subsequent enlargement of the organ. It was found that the rearrangement of the cortical MTs to approximate a series of concentric circles on the residual meristem occurred before the emergence of leaf primordia. Through their apparent influence on microfibril alignments, the changes in MT disposition, described here, have the potential to generate major biophysical changes that accompany organogenesis.Abbreviation MT(s) microtubule(s)  相似文献

The effect of broad band green, blue and red light on stomatal opening of Vicia faba L. (broad bean) leaves was examined. In air, blue light caused greater stomatal opening than red light. In air with green light stomata were only slightly opened. In a nitrogen atmosphere red light caused greater opening than blue light, and green light caused only slight opening. Opening in air or nitrogen atmosphere in red or blue light was inhibited by the uncoupler CCCP, while the photosynthetic inhibitor DCMU inhibited opening in air but not in nitrogen atmosphere. We concluded that more than one light activated metabolic pathway can supply the energy needed to effect stomatal opening and that different pathways are operative under different conditions.  相似文献

Penaeid crustins were described in Litopenaeus vannamei and L. setiferus as proteins belonging to an antibacterial peptide family with similar sequences but different sizes. Six crustin-coding clones were isolated from a cDNA library from L. vannamei hemocytes, sequenced and compared. Two different isoforms (named I and P) were found, based on two nucleotide differences that produce one change in amino acid sequence (Ile/Pro). Other single differences in nucleotide sequences were also noted, but they did not change the translated product. The mRNA steady state levels of crustin I, but not of crustin P, were down regulated by Vibrio alginolyticus inoculation. Thus, the differences among penaeid crustins seem to be associated with one amino acid substitution, which affects their expression after bacterial inoculation. By structural similarity, shrimp crustins seem to belong to an antibacterial WAP-domain containing protein family.  相似文献
