A novel method for the preparation of amino-substituted hypocrellin B.   总被引:2,自引:0,他引:2  
A series of amino-substituted hypocrellins derived from hypocrellin B (HB) were synthesized by a novel mild method, in which the peri-hydroxylated perylenequinone structure of hypocrellin was preserved by the reaction of HB with an amine. The red absorption of the resulting products was significantly enhanced relative to the parent hypocrellins, which will significantly improve its photodynamic therapy effectiveness.  相似文献

Five new derivatives of hypocrellin B were obtained from the reactions of hypocrellin B with ammonia and ethanolamine. Their photophysical and photochemical properties were investigated. The phototoxicity of one compound on AH cells irradiated with red light (lambda = 600-700 nm) was also studied. Their significantly enhanced red absorptivities at wavelengths longer than 600 nm and singlet oxygen-generating function qualify them as promising photodynamic therapy agents.  相似文献

Mono-substituted hypocrellin B (MHB) and di-substituted hypocrellin B (DHB) by mercaptoacetic acid are new photosensitizers synthesized to improve the red absorption and water solubility of the parent hypocrellin B (HB). The photochemistries (Type I and/or Type II) of MHB and DHB have been studied in homogeneous solutions using electron paramagnetic resonance (EPR) and spectrophotometric methods. In anaerobic homogeneous DMSO solution, DHB*- (or MHB*-) was predominantly photoproduced via self-electron transfer between the excited- and ground-state species. The presence of an electron donor significantly promotes the formation of the reduced form of DHB (or MHB). As compared with hypocrellin B, the efficiencies of DHB*- and MHB*- generation was enhanced obviously. When oxygen-saturated solutions of DHB (or MHB) were illuminated with 532 nm light, singlet oxygen (1O2), superoxide anion radical (O2*-), hydroxyl radical (*OH) and hydrogen peroxide (H2O2) were formed. DHB and MHB generate 1O2 with quantum yields of 0.18 and 0.22, respectively, which are much lower than that of HB (0.76) in chloroform. The superoxide anion radical was significantly enhanced by the presence of electron donors. The rate of O2*- production was also dependent on the concentration of DHB or MHB. Moreover, O2*- upon disproportionation can generate H2O2 and ultimately the highly reactive *OH via the Fenton reaction and other pathway with the involvement of DHB*- (or MHB*-). As in the case of DHB*- (or MHB*-), the efficiencies of O2*- and *OH generation by DHB and MHB were also enhanced obviously, consistent with the fact that DHB*- (or MHB*-) acts as the precursor of O2* and thus *OH. These findings suggest that the photodynamic actions of DHB and MHB may proceed via enhanced Type I mechanism and reduced Type II mechanism as compared with that of HB.  相似文献

An amorphous formulation of hypocrellin A for photodynamic therapy is reported which can provide stable aqueous dispersion of such hydrophobic photosensitizers. In vitro studies have demonstrated the active uptake of amorphous formulation of hypocrellin A into the mitochondria of tumor cells. Compared with Tween-80 micelle embedded hypocrellin A, low dark-toxicity but similar light-toxicity of the amorphous one to drug impregnated tumor cells was observed. Thus, the potential of using amorphous formulation of hypocrellin A as drug delivery system for photodynamic therapy has been demonstrated.  相似文献

先对不同产地采集的竹黄菌进行筛选,得到优产竹红菌素的菌株,然后采用单因子和3因素3水平正交试验法对竹红菌素液体发酵条件进行优化,在优化培养基的基础上,选用不同浓度的Cr3+、Fe3+、Cu2+和Ca2+对竹红菌素进行离子调控研究。结果表明:从休宁所采集的菌株不仅生长速度最快,发酵所产的竹红菌素含量也最高;竹红菌素最佳发酵碳源是葡萄糖,最佳发酵氮源是硝酸钠,最佳培养基组合为2%葡萄糖,0.2%硝酸钠,pH7.5;Cr3+和Fe3+浓度为0.005%时竹红菌素含量均最高;0.05%的Ca2+最有利于竹红菌素的分泌;Cu2+为0.03%时竹红菌素含量达到最大值。  相似文献

Hypericin and hypocrellin are potential antiviral and antineoplastic agents with multiple modes of light-induced biological activity connected with a production of singlet oxygen and/or excited-state proton transfer and consequent pH drop formation in the drugs environment. In present work light-induced cytotoxicity of hypericin (1 x 10(-5) - 10(-9) mol) and hypocrellin (1 x 10(-5) - 10(-9) mol) and potentiating effect of omeprazole on human leukemic cell line HL-60 was studied. Under dark condition cultivation none cytotoxicity was observed. The only one exception was hypocrellin in concentration 1 x 10(-5) mol which displayed full cytotoxic effect. However, illumination increased cytotoxic effect of hypericin and hypocrellin, both. Omeprazole, an inhibitor of H+K+-ATPase, has been used for testing the hypothetical pH decreasing effect of hypericin and hypocrellin in their cytotoxic mechanism of action. The results of our experiments have shown that in HL-60 cell line the effect of hypericin and hypocrellin at 1 x 10(-6) mol (both) was significantly potentiated by omeprazole in concentrations 1 x 10(-6) - 10(-9) mol. Our results support the hypothesis that the excited-state proton transfer and the consequent acidification of hypericin and hypocrellin environment could play a role in the biological activity of both agents.  相似文献

15-Deacetyl-13-glycine substituted hypocrellin B (GDHB) is a new type of hypocrellin derivative with enhanced red absorption longer than 600 nm and water solubility. When an anaerobic DMSO or DMSO-buffer (pH 7.4) solution of GDHB was illuminated with >470 nm light, a strong electron spin resonance (ESR) signal was formed. The ESR signal was assigned to the semiquinone anion radical of GDHB (GDHB*-) based on a series of experiments. GDHB*- was predominantly photoproducted via the self-electron transfer between the excited- and ground-state species. Decay of this species, both in the presence and absence of electron donor, was consistent with second-order kinetics. In aqueous solution, the TEMPO counter-spin experiment indicated the formation of GDHB*- that could not be detected by ESR method directly. The formation of GDHB*- and hydroquinone of GDHB (GDHBH*-) was also confirmed by spectrometric method. These findings suggested that GDHB was at least a favorable type I phototherapeutic agent.  相似文献

Synthesis and EPR investigations of new aminated hypocrellin derivatives   总被引:2,自引:0,他引:2  
Hypocrellins are novel photodynamic agents. A recent advance in the synthesis of hypocrellin congeners results in the production of two amino-substituted hypocrellin B derivatives in high yield via photochemical reaction. Both compounds exhibit similar photodynamic activity as hypocrellin B in terms of type I and type II mechanisms. In anaerobic media, semiquinone anion radicals can be detected by electron paramagnetic resonance (EPR) under irradiation; while superoxide anion radical, hydroxyl radical and singlet oxygen are photoproduced when oxygen was present. The quantum yields of singlet oxygen by these two new compounds are determined to be 0.72 and 0.64, respectively, similar to that of hypocrellin B. The comparison of the photosensitization chemistry of compounds 1 and 2 in liposomes with that in homogeneous solution has also been made. In liposomes, the type II photoprocess was favored and predominant over the type I photoprocess due to the decreased interactions between dye molecules. Both compounds exhibit much stronger red light absorption than the parent hypocrellin and therefore, merit investigation as photosensitizers.  相似文献

To improve water solubility and specific affinity for malignant tumors, glycoconjugated hypocrellin B (GHB) has been synthesized. Illumination of deoxygenated DMSO solution containing GHB generates a strong electron paramagnetic resonance (EPR) signal. The EPR signal is assigned to the semiquinone anion radical of GHB (GHB*-) based on a series of experimental results. Spectrophotometric measurements show that the absorption bands at 645 nm and 502 nm (pH 8.0) or 505 nm (pH 11.0) arise from the semiquinone anion radical (GHB*-) and hydroquinone (GHBH2) of GHB, respectively. GHBH2 is readily formed via the decay of GHB*- in water-contained solution. The increase of pH value of the reaction media promotes this process. When oxygen is present, superoxide anion radical (O2*-) is formed, via the electron transfer from GHB*-, the precursor, to ground state molecular oxygen. Hydroxyl radical can be readily detected by DMPO spin trapping when aerobic aqueous solution containing GHB is irradiated. As compared with the parent compound, hypocrellin B (HB), the efficiency of O2* and *OH generation by GHB photosensitization is enhanced significantly. Singlet oxygen (1O2) can be produced via the energy transfer from triplet GHB to ground state oxygen molecules, with a decreased quantum yield, i.e., 0.19. These findings suggest that the new GHB possesses an enhanced type I process and a decreased type II process as compared with hypocrellin B.  相似文献

Absorption, resonance Raman, surface-enhanced Raman spectroscopy and differential scanning microcalorimetry were employed to study the interaction of hypocrellin A with human serum albumin. The identification of the binding place for hypocrellin A as well as the model for the albumin-hypocrellin A complex are proposed. In this model hypocrellin A interacts with albumin through more than one binding site placed on the protein surface. This model of non-specific interaction could explain why the absorption spectrum of hypocrellin A does not change in the presence of albumin and why the presence of the drug does not change significantly the thermodynamic parameters of the protein, while the Raman spectra show evident changes concerning both the protein and the drug structure. Even if hypocrellin A does not interact with an interior binding site, it can affect deeply the general albumin structure.  相似文献

以紫外(UV)与亚硝基胍诱变的竹黄菌(Shiraia bambusicola)菌株NU12和UV4为出发菌株,60℃处理5 min、紫外(距离30 cm,30 W)照射90s进行双亲原生质体灭活,通过30%聚乙二醇PEG6000介导原生质体融合20 min.结合平板初筛和高效液相色谱( HPLC)分析进行复筛,通过3轮重组融合操作,筛选出6株高产竹红菌甲素的融合株.其中融合菌株FIII - 21的竹红菌甲素产量达到80.4 mg/L,比原始出发菌株提高了58.9%~167.1%,且遗传稳定,具有较高的医药及工业应用价值.  相似文献

Visible light (> 470 nm) irradiation of an anaerobic aqueous solution of GDHB, the formation of GDHB*- was detected by an ESR method in the absence or presence of electron donor. When exposed to oxygen, superoxide anion radical and singlet oxygen were formed. The superoxide anion radical was generated by GDHB*- via electron transfer to oxygen and this process was significantly enhanced by the presence of electron donors. Singlet oxygen ((1)O2) was also formed in the photosensitization of GDHB in aerobic solution and 1,4-diazabicyclo [2,2,2] octane (DABCO), sodium azide (NaN3) and histidine inhibited the generation of (1)O2. A 9,10-diphenyl antracene (DPA)-bleaching method was used to determine the quantum yield of (1)O2 generated from GDHB photosensitization. The (1)O2 quantum yield was estimated to be 0.65. With the depletion of oxygen, the accumulation of GDHB*- would replace that of (1)O2. Evidence accumulated that the photodynamic action of GDHB may proceed via both type I and type II mechanisms and that a type II mechanism will be transformed into a type I mechanism as oxygen gets depleted.  相似文献

In present work light-induced cytotoxicity of hypericin and hypocrellin and mechansim of cell death (apoptosis or necrosis) on human leukemic cell line HL-60 was studied. As a mean for apoptosis detection we used poly (ADP-ribose) polymerase (PARP) as a sensitive marker of early stages of apoptosis. Our results show that exposition of HL-60 cells to hypericin (1 x 10(-5) mol x l(-1)) for 4 hours has no effect on PARP cleavage. However, after 24 and 48 hours of illumination there is evident that hypericin in this concentration cleaved PARP (116 kDa) into two fragments (85 and 25 kDa). Contrary to hypericin, hypocrellin in concentration 1 x 10(-5) mol x l(-1) after 4 hours of illumination cleaved PARP into two fragments typical for apoptosis. In lower concentration (1 x 10(-6) mol x l(-1)) hypocrellin possess also significant cytotoxic activity. Because we detected no fragmentation of PARP in all observed time periods we suggest that cytotoxic effect of hypocrellin in this concentration is due to induction of necrosis. Our results support the hypotesis that the hypericin and hypocrellin has similar mechanism of action and illumination increases cytotoxic effect of both agents.  相似文献

采用液体浸泡方法首次测定了菌寄生菌属Hypomyces sp.的次级代谢物苝醌类化合物竹红菌甲素和痂囊羟菌素对松材线虫的致病率。结果表明竹红菌甲素对松材线虫的半致病率为18小时50g/mL,痂囊羟菌素对松材线虫的半致病率为18小时15g/mL,对比(啊维菌素)对松材线虫的半致病率为18小时0.8g/mL。在红外光照射下竹红菌甲素和痂囊羟菌素具有良好的光敏杀伤松材线虫的作用。  相似文献

首次对安徽省三种产地的竹黄菌进行了组织分离培养和液体发酵,比较了该菌固体生长状况和液体发酵形成的竹红菌素产量。结果显示,广德卢村的菌种平皿生长速度为0.31cm/d,液体发酵产生的竹红菌素吸光度为0.21;宁国板桥菌种平皿生长速度为0.30cm/d,液体发酵产生的竹红菌素吸光度为0.30;休宁武城的竹黄菌平皿生长速度为1.85cm/d,菌株液体发酵产生的竹红菌素吸光度为O.39。结论表明,休宁武城菌种无论是平皿生长速度还是发酵形成的竹红菌素量都明显优于其它两地的菌株。  相似文献

Some endophyte isolates were isolated in a bamboo pole sample parasitized the fungus Shiraia bambusicola from Zhejiang Province. After screening through hypocrellin bacteriostatic effect and fermentation test, we got the isolate TX4 of bacterial elicitor and GZUIFR-TT1 of fungal elicitor which had certain effect to promote S. bambusicola to produce hypocrellin. The Plackett–Burman design was introduced to evaluate the effects of nine factors based on single-factor test. Yeast extract, glucose, and isolate GZUIFR-TT1 elicitor were found to be the critical activity factors for increasing the total hypocrellin production. So we identified the isolate GZUIFR-TT1 as Trametes sp. Through response surface methodology, we obtained the optimum production conditions as follows: yeast extract, 2.99 g/L; glucose, 32.45 g/L; and Trametes sp. elicitor, 81.40 μg/mL. Under the above conditions, the experimental value of hypocrellin production was 102.60 mg/L, compared with the control it increased about 7.90 times.  相似文献

为研究竹黄菌与竹红菌化学成分及细胞毒活性的差异,本研究通过高效液相色谱(HPLC)分析结合常规色谱方法,分离鉴定了两种真菌的6个相同成分,分别为3个主要成分竹红菌甲素(1)、竹红菌乙素(2)和竹红菌丙素(3),以及3,6,8-三羟基-1-甲基口山酮(7)、3,8-二羟基-6-甲氧基-1-甲基口山酮(8)和过氧麦角甾醇(9)。另外,从竹黄菌中还分离得到11,11′-二去氧沃替西林(5)、麦角甾-7,22E-二烯-3β,5α,6β-三醇(10)和麦角甾-7,22E-二烯-2β,3α,9α-三醇(11),并首次从竹红菌中分离得到竹红菌丁素(4)、灰黄霉素(6)、化合物7和8。活性筛选发现,化合物5对三株肿瘤细胞NCI-H1975、HepG2和MCF-7有很强细胞毒活性,化合物1有较强细胞毒活性,而化合物6活性较弱。  相似文献
