为探索属间杂种的遗传特点以及改良甘蓝型油菜油分品质,进行了甘蓝型油菜和紫罗兰的属间杂交.杂交母本为甘蓝型油菜奥罗(Brassica napus L. cv. oro),父本为紫罗兰(Matthiola incana (L.) R. Br.).将授粉7 d后的油菜子房切下,消毒后,培养于添加适当植物激素的MS培养基.从750个离体培养的油菜授粉子房中,获得了2粒成熟胚胎,其结籽率为0.26%.将胚胎取出,转接于MS培养基(添加2.0 mg/L 6-BA和0.1 mg/L NAA),获得了丛生芽.将丛生芽分割为许多单芽,转接到新鲜培养基中,长成了22株小苗.杂种一代植株呈中间性,它的许多性状介于两个亲本之间,一些性状倾向母本,少数性状表现显著的超亲杂种优势,植株结实性差.杂种后代(F2)植株表现多样性,多数植株的性状倾向母本,能育.部分植株表现中间性、育性差,少数植株发育不良、不育.染色体研究表明,杂种一代植株为混倍体.在杂种体细胞中,许多细胞的染色体数为2n=26,为两个亲本的配子染色体数之和.杂种后代(F2)中,倾母植株的染色体数为2n=38,矮小植株的许多细胞具非整倍染色体数,如2n-1=37、2n+1=39.从杂种后代中获得了种子油分品质较好的植株,有可能用于油菜的品质育种.  相似文献

以不同类型的大白菜与紫甘蓝为材料,运用蕾期授粉结合胚挽救技术获得大白菜与紫甘蓝的种间杂种,并对其进行细胞学鉴定.结果表明,大白菜与紫甘蓝杂交,获得了57株F1代幼苗;对根尖染色体数量进行观察和杂种花粉特性调查发现,其中47株具有预期的染色体数目,2n =19,鉴定为真杂种,其花粉败育;另有6株具有38条染色体,鉴定为种间异源双二倍体,应该是发生了染色体自然加倍,其花粉可育.可育的杂种F1代与大白菜回交,获得了大白菜-紫甘蓝BC1代材料.田间观测结果显示,杂种F1代植株综合性状均介于双亲之间,BC1代植株包球明显,综合性状偏向母本大白菜.  相似文献

小麦×冰草属间杂种F_1的植株再生及其变异   总被引:2,自引:0,他引:2  
在对0.5—40.cm长幼穗培养4周后诱导出愈伤组织的基础上,获得了88株普通小麦(Triticum aestivum cv.Chinese Spring,2n=6x=42,AABBDD)×沙生冰草(Agropyron desertorum Fisch.> Schult.,2n=4x=28,PPPP)杂种F_1的再生植株。不同长度的幼穗在培养时,其愈伤组织发生的部位及其增殖速度不同。再生植株的产生主要是通过直接器官发生途径。所有的再生植株染色体数目全与杂种F_1相同,为2n=35。与杂种F_1相比,再生植株的减数分裂行为是相当复杂的,证明有染色体结构变异的发生。每茎穗数、叶片失绿斑等形态上的变异是由环境效应引起的;而株高、每穗轴节上小穗数和育性是由遗传效应决定的。特别值得注意的是,再生植株的自交结实率高达5.49%,共获得自交种子484粒,这对利用P染色体组中的期望基因有着极为重要的意义。  相似文献

在离体培养条件下,比较不同浓度、不同处理时间的秋水仙素对田埂报春进行染色体加倍的诱导效果。结果表明:0.08%秋水仙素处理48h的诱变效果最佳,诱变率高达56%。经秋水仙素诱导后形成的多倍体植株与原二倍体植株比较,在形态上,四倍体植株表现出多倍体特征,叶片变厚,叶形指数减小,保卫细胞增大,单位面积气孔数减少,叶绿体数明显增多。对变异植株进行细胞学研究发现,体细胞中期染色体数目为2n=4x=36,而原二倍体的染色体数目为2n=2x=18,基数x=9,因此,变异植株(2n=4x=36)为四倍体。前者的核型公式为2n=4x=8L+12M2+4M1+12S,核型属于1A;后者的核型公式为2n=2x=4L+6M2+2M1+6S,核型也属于1A。检测发现少数个体有非整倍体变异。  相似文献

通过细胞学方法和染色体特异分子标记鉴定六倍体小偃麦(AABBEE)与硬粒小麦(AABB)杂交的自交后代F2和F3植株,探讨长穗偃麦草染色体在硬粒小麦背景中世代间的传递特征,并筛选硬粒小麦-长穗偃麦草E染色体附加系。对218个F2单株染色体数检测表明,2n=28植株占41.7%,2n=29植株占18.3%,其余40.0%植株的染色体数在2n=31~42范围内。分子标记鉴定表明,在F2代2n=29单体附加植株中,不同的长穗偃麦草染色体传递率之间存在明显差异,1E传递率最高,3E和6E传递率最低。在F2代2n=30单株中,1E、4E、7E和5E染色体相互组合产生的双单体多,6E参与组合较少,未检测到2E或3E与其他染色体的组合单株。在1E~7E单体附加株自交后代F3中,E染色体传递率变化范围为9.1%~27.5%,1E传递率最高,6E传递率最低,与F2的传递率一致。从F3代中选育出1E~7E单体附加及少数二体附加,所有单体附加均可育。这些附加E染色体材料将对小麦代换系和易位系的创制提供有益的中间材料。  相似文献

用不同浓度秋水仙素处理野生南荻×芒(Miscanthus lutarioriparia×Miscanthus sinensis)远缘杂交后代以诱导产生多倍体,并对变异株进行形态学和细胞学鉴定,以期获得稳定的四倍体植株并分析其生理特性。结果表明:(1)采用秋水仙素加入培养基处理法和秋水仙素溶液浸泡处理法都可获得一定频率的多倍体植株;胚性愈伤组织以0.2%秋水仙素浸泡处理48h的诱变效果较好,四倍体诱导率达8.7%;芽在0.05%秋水仙素培养基中处理15d较好,四倍体诱导率达10.6%;生根苗在0.1%秋水仙素培养基中处理10d较好,四倍体诱导率达11.1%。(2)经体细胞染色体计数,加倍植株染色体数为2n=4x=76,对照植株的染色体数目为2n=2x=38。(3)生长2年的多倍体植株形态、叶片大小、茎粗、茎壁厚、节间等性状表现出巨大性和超亲优势。  相似文献

节节麦×大麦杂种胚再生植株的细胞遗传学研究   总被引:2,自引:1,他引:1  
以节节麦(2n=14)为母本和大麦(2n=14)进行杂交,两组合平均结实率为31 .11%。对31个幼胚进行愈伤组织诱导培养,其中2个形成全能性愈伤组织进而分化出再生杂种植株。细胞学观察表明,杂种胚再生植株均是染色体加倍的节节麦-大麦双二倍体(2n=28)或染色体再加倍的多倍体。节节麦-大麦双二倍体在减数分裂中期Ⅰ染色体配对平均构型为14.79Ⅰ+1.95Ⅱ+ 4.53Ⅱ′+0.01Ⅲ,表现为自交不育。杂种胚再生植株染色体数目再加倍的多倍体有少数产生了自交种子。 Abstract:Hybridizations between Aegilops tauschii(2n=14)and Hordeum vulgare(2n=14)were made using Ae.tauschii as female.The mean frequency of seed set was 31.11% in two cross combinations,13 hybrid embryos were cultured for inducing callus,of which 2 produced totipotent callus,and plants were regenerated from them.These plants were Ae.tauschii-H.vulgare amphidiploids with doubling chromosome number(2n=28)and octoploids with repeatedly doubling chromosome number(2n=50~56).The Ae.tauschii-H.vulgare amphidiploids were sterile and their average chromosome pairing at MI were 14.79I+1.95II+4.53II′+0.01III.Some octoploid(2n=56)plants produced seeds after selfing.  相似文献

在组织培养条件下,对库拉索芦荟(Aloe vera L.)用秋水仙素进行染色体加倍的诱导处理,结果表明:用含0.06%秋水仙素处理12h后诱变频率可达50%,其效果最佳。经秋水仙素诱导的加倍群体与正常二倍体植株比较,植株的大多数叶片变厚,叶色变深,叶片变大,气孔增大而单位叶面积气孔数减少。对变异材料进行细胞学研究所发现,体细胞中期染色体为2n=4x=28,为4倍体。未加倍前的二倍体为2n=2x=14。检测中也发现有少数植株有2n=14和2n=28两种细胞型的情况。  相似文献

首次报道了产于广西的苦苣苔科(Gesneriaceae)4属4种植物的染色体数目。其中,(1)单座苣苔属(Metabriggsia):单座苣苔(M.ovalifolia)的染色体数目2n=24;(2)异裂苣苔属(Pseudochirita):异裂苣苔(P.guangxiensis)的染色体数目为2n=28;(3)长蒴苣苔属(Didymocarpus):东南长蒴苣苔(D.hancei)的染色体数目为2n=20;(4)唇柱苣苔属(Chirita):弄岗唇柱苣苔(Ch.longgangensis)的染色体数目为2n=28。4种植物的染色体大多数为小型染色体。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
