Mono- and disialogangliosides containing glucose, galactose and sialic acids were isolated from the total lipid extract of hepatopancreas of the starfish Aphelasterias japonica. Their structures were elucidated by total and partial acid hydrolysis, trideuteriomethylation analysis, neuraminidase treatment, chromium trioxide oxidation, methanolysis and periodate oxidation. The monosialoganglioside was identified as 8-O-methyl-N-glycolylneuraminosyl-alpha-(2-3)-galactosyl-beta-(1- 4)-glucosyl-beta-(1-1)-ceramide. The disialoganglioside has the additional N-glycolylneuraminic acid or its 8-O-methyl derivative residue at the subterminal position to which the terminal sialic acid residue is linked through the hydroxy group of the glycolic acid unit. The long-chain bases were found to be mixtures of phytosphingosines with both branched and linear chains, and the fatty acids were shown to be mixtures of normal and alpha-hydroxy fatty acids, the latter amounted to about 90% of the fatty-acid mixtures. The composition of the lipid moieties of the gangliosides was determined by GLC and GLC-MS.  相似文献

A method has been developed to determine the distribution of substituents in the glucose units of sulfonylbutylethers of cyclomaltoheptaose (beta-cyclodextrin). This method involves hydrolysis of the glucosidic linkages, permethylation, formation of sulfonylchlorides and subsequent transformation to the permethylated sulfonylfluoride derivatives. The latter were thermostable and could be analyzed by GLC and identified by EI and CIMS. For confirmation, the 2-, 3-, and 6-O-substituted standard compounds were independently synthesized and characterized by NMR and GLC-MS.  相似文献

Mixtures of lipids and phospholipids were separated by centrifugally accelerated thin-layer chromatography on a preparative scale (300-500 mg lipid mixture per run). The isolated lipids and phospholipids were identified by 1H and 13C NMR spectroscopy and their fatty acid composition was determined by GLC and GLC-MS of their methyl esters.  相似文献

The composition of the amino acid and peptide fraction of the protein hydrolyzate and its dependence upon the spray-drying process at elevated temperatures were investigated by GLC and GLC-MS. Spray-drying leads to decrease in the Glu and Met contents, to transformation of Cys into cystine and to formation of lactames (pyroglutamic acid and pyrrolidone) and diketopiperazines. Some dipeptides, acetylproline, paraffines, fatty acid pyridinecarboxylic and lactic acids, ethylene glycol were identified in the fraction. The incidental occurrence of these compounds should be taken into account when investigating the biological activity of protein hydrolyzates.  相似文献

The essential oil of Senecio aegyptius var. discoideus flowers, leaves, stems and roots were isolated by hydrodistillation. Analysis of the oils by capillary GLC and GLC-mass spectrometry were performed and 34 out of 37 compounds were identified. The main component was isolated and characterized as 1,10-epoxyfuranoeremophilane using a combination of GLC, GLC-MS, and NMR analyses. The oils of flowers, leaves and stems were rich in monoterpene hydrocarbons while the root oil mainly contains furanoeremophilanes. Flower and leaf volatile oils showed significant level of antifungal activity against C. albicans, moderate effect against Gram positive bacteria, however, it has weak activity against Gram negative bacteria. The isolated sesquiterpene (1,10-epoxyfuranoeremophilane) exhibited substantial inhibitory activity against Gram negative bacteria.  相似文献

The alkaloid composition of Convolvulus lanatus was investigated by GLC and GLC-MS. Altogether, thirteen tropane and pyrrolidine alkaloids were identified on the basis of mass spectral data and/or Kovats retention indices. Twelve of them have been recorded for the first time in this plant and some of these alkaloids were identified for the first time in the genus Convolvulus. Cuscohygrine is the major constituent. Qualitative and quantitative differences of root and aerial parts are discussed.  相似文献

5-Methylfurfural diethyl acetal, 5-methylfuran acid ester, and 4-hydroxypentanoic acid ethyl ester were first identified in high-sacchariferous corn media subjected to alcoholic fermentation by Mucor X-I culture and in aged sweet wines by means of GLC, GLC-MS, and TLC, as well as by UV and IR spectroscopic examinations. Alkaloid profiles of five Senecio species (Asteraceae), including S. ambiguus subsp. ambiguus, S. ambiguus subsp. nebrodensis, S. gibbosus subsp. bicolor, S. gibbosus subsp. gibbosus, and S. gibbosus subsp. cineraria, were studied. Eleven pyrrolizidine alkaloids were identified and their content was evaluated by GLC-MS and GLC analysis. Otosenine and florosenine were found to be the major alkaloids in all studied species. It is interesting that only S. ambiguus subsp. nebrodensis was characterized by a high content of the alkaloids jacobine, jacoline, jaconine, and jacozine.  相似文献

The long-chain fatty acids of 4 Saccharopolyspora hirsuta strains were examined as their methyl and picolinyl esters using gas liquid chromatography (GLC) and GLC-mass spectrometry (GLC-MS). All the strains had similar fatty acid profiles composed mainly of isoanteiso-, and 10-methyl-branched components. Three new substituted 10-methyl-branched fatty acids were also detected and the major component identified as 10,15-dimethylhexadecanoic acid. The implications of these data for Saccharopolyspora systematics are discussed.  相似文献

Abstract Butyrivibrio fibrisolvens strain X6C61 produces two extracellular polysaccharides (EPS-I and EPS- II) separable by anion-exchange chromatography. The neutral sugar constituents of EPS-I were identified by gas-liquid chromatography (GLC) as the alditol acetates of rhamnose, mannose, galactose, glucose, and an unidentified component. These results were confirmed using thin-layer chromatography (TLC). Neutral sugar analysis of EPS-II, which eluted from DEAE-Sephadex at 0.4 M NaCl, yielded the alditol acetates of rhamnose, galactose, glucose, and idose. However, idose was not found when hydrolysates of EPS-II were analysed by TLC. Further investigations showed that the iditol hexaacetate detected via GLC was an artifact of the commonly-used procedures for neutral sugar analysis. This compound was instead generated from l -iduronic acid, as shown by GLC-MS studies.  相似文献

Alkaloid profiles of root and shoot cultures, seedlings and mature plants were analysed by capillary GLC and GLC-MS. beta-Carboline alkaloids, such as harmine, harmaline dominate in normal and root cultures transformed by Agrobacterium rhizogenes, as well as in roots and fruits of the plant. In shoots, flowers and shoot cultures quinoline alkaloids such as peganine, deoxypeganine, vasicinone and deoxyvasicinone widely replace the beta-carboline alkaloids. In root cultures, the formation of beta-carboline alkaloids can be induced by methyljasmonate and several other elicitors indicating that these alkaloids are part of the reactive chemical defence system of Peganum harmala.  相似文献

Free sterol fractions from the holothurians (sea cucumbers) Synapta maculata, Cladolabes bifurcatus and Cucumaria sp. have been isolated and studied by HPLC, GLC, GLC-MS and NMR methods. Forty seven sterols were identified, including several rare ones. In contrast with previously studied holothurians, the presence of a minor amount of Delta7 sterols was indicated in the sterols of S. maculata. This animal contains predominantly Delta(9(11))sterols as well as an abnormally high concentration of Delta5 sterols. In C. bifurcatus and Cucumaria sp., 14alpha-methyl and 4alpha,14alpha-dimethyl-Delta(9(11))sterols were found to be the main sterol constituents. Peculiarities of sterol distribution and the relationship between sterol compositions and taxonomic positions, ecology and toxicity of the corresponding sea cucumbers were discussed.  相似文献

Periodate oxidation of terminalN-acetyl- andN-glycoloylneuraminic acid residues in the mucins from edible bird nest substance and pig submandibular gland, respectively, can be carried out under conditions which exclusively give rise to the formation of the C-7 analogues of these sialic acids. In contrast, the C-8 compounds can be obtained in a maximum yield of about 40%. Under identical conditions,N-glycoloylneuraminic acid is oxidized about 1.5 times faster than theN-acetylated derivative. After release of the sialic acids by acid hydrolysis, the characterization of the oxidation products was carried out by TLC, by GLC and GLC-MS of the corresponding pertrimethylsilyl derivatives, and by 500-MHz1H-NMR spectroscopy. In addition, molar response factors for GLC analysis and extinction coefficients in the orcinol/Fe3+/HCl assay were determined.  相似文献

Tanacetum santolinoides, Essential Oil Composition, n-Hexane-Ether Extract Composition The essential oil of the aerial parts of Tanacetum santolinoides was analyzed by capillary GLC and GLC-MS. Altogether 30 components were identified. The main constituents were thymol (18%), trans-thujone (17.5%), trans-chrysanthenyl acetate (13.2%), cis-chrysanthenyl acetate (9.2%), umbellulone (9.7%) and 1,8-cineole (4.7%). Similar essential oil pattern in addition to palmitic acid methyl ester, palmitic acid, stigmasterol, sitosterol and two flavonoidal aglycons were found in the n-hexane-ether extract. The oil showed strong in vitro activity against E. coli, Bacillus subtilis and Candida albicans.  相似文献

The alkaloid composition of the aerial parts of two taxa of Teline maderensis was studied by capillary GLC and GLC-MS. N-Methylcytisine was the major alkaloid found in both plants. Contents of cytisine and lupanine were higher in T. maderensis var. paivae while anagyrine content was more pronounced in T. maderensis var. maderensis. The alkaloids dehydrocytisine, N-acetylcytisine and epibaptifoline appeared only in T. maderensis var. maderensis and N-formylcytisine was identified as a minor constituent in T. maderensis var. paivae, and detected only in trace amounts in the other variety of the plant.  相似文献

Aeciospores of Cronartium fusiforme isolated from slash pine (Pinus elliottii) trees were analyzed for volatile terpenoids by GLC and GLC-MS. α-Pinene, β-pinene, Δ3-carene, myrcene, linonene, β-phellandrene, and δ-terpinene were the major monoterpenoid hydrocarbons present with only traces of camphene. A number of monoterpenoid alcohols were also present of which terpinen- 4-ol predominated. Among the various acyclic sesquiterpenes present, β-farnesene and β-citronellol were identified. Summary ABA has been identified by GLC-MS and routinely determined by GLC as one of several inhibitory substances in the testa and pericarp of hazel nuts. Its concentration in newly harvested nuts, which had not developed embryo dormancy, was 19.0 nmoles/g dry weight for the testa, 1.4 nmoles/g for the pericarp and 0.09 nmoles/g for the embryo. Dry storage of the nuts resulted in the development of embryo dormancy together with a slight loss of ABA. On imbibition of dormant nuts at 5° C and 20° C there was a 61% loss of ABA from the testa and pericarp in both cases. However the 5° C imbibition resulted in the breaking of seed dormancy while the 20° C imbibition had no effect on the dormancy. The ABA of the testa and pericarp seems to be concerned with the maintenance of seed dormancy prior to the onset of embryo dormancy. Abstract Butyrivibrio fibrisolvens strain CF3, a stricyly anaerobic bacterial isolate from the ovine cecum, produces extracellular polysaccharides (EPS) when grown on a defined medium containing glucose as the carbon source. EPS were purified from culture supernatants and their monosaccharide composition was determined by two different procedures. Analysis of EPS hydrolysates by thin-layer chromatography (TLC) yielded spots coincident with standard glucose, altrose, and 1,6-anhydroaltrose. These results were corroborated by both gas-liquid chromatography (GLC) and GLC-mass spectroscopy (GLC-MS) of alditol acetates prepared from EPS hydrolysates. Purification of the altrose from EPS hydrolysates was accomplished by preparative paper and column chromatography. Polarimetry demonstrated the isolated altrose to have the L -configuration. The occurrence of this hexose in nature has not yet been reported.  相似文献

The occurrence of hopanoid lipids in Bradyrhizobium bacteria   总被引:2,自引:0,他引:2  
Abstract Lipid extraction procedures followed by GLC and GLC-MS analysis were used to investigate the triterpenoid content in Bradyrhizobium and Rhizobium bacteria. Unlike the tested strains of Rhizobium bacteria, a range of triterpenoids e.g., squalene and different classes of hopanoid derivatives were detected in bacteria from all Bradyrhizobium strains investigated (different strains from Bradyrhizobium japonicum, Bradyrhizobium elkanii as well as Bradyrhizobium sp.). Furthermore, related compounds were identified from some hopanoid lipids (e.g., diplopterol) that carried an additional methyl group in their molecular structure. The hopanoid content was high in some strains and accounted for more than 40% of the total lipid fraction (e.g., in strains Bradyrhizobium japonicum USDA 110 and USDA 31), while other strains contained only about a tenth of that amount (e.g., Bradyrhizobium japonicum ATCC 10324 and Bradyrhizobium sp. ( Lupinus ) ATCC 10319).  相似文献
