It is proposed that the current nomenclature by which individual antimicrobial peptides from the skins of frogs belonging to the genus Leptodactylus are named from the species of frog from which they were isolated should be replaced by one that emphasizes the fact that these peptides are evolutionarily related. As the ocellatins from Leptodactylus ocellatus were the first such peptides to be characterized, it is suggested that all orthologous peptides should be described as "ocellatins". Consistent with accepted terminology for other families of antimicrobial peptides, the upper case initial letter of the species is used to indicate their origin and isoforms are designated by numbers. When two species begin with the same initial letter, a second distinguishing letter shall be employed. Thus, the terms ocellatin-1, -2, -3, and -4 are retained for the parent peptides. Fallaxin is replaced by ocellatin-F1, pentadactylin by ocellatin-P1, laticeptin by ocellatin-L1, syphaxin by ocellatin-S1, and the paralogs from L. validus are termed ocellatin-V1, -V2, and -V3. 相似文献

Chromosomes from all 17 species of native Jamaican Eleutherodactylus as well as introduced E. johnstonei were subjected to computer-assisted analyses. Diploid chromosome numbers of 24, 26, 28, 30 and 32 were found and no two species had identical karyotypes. Karyotypic data were superimposed on a phylogeny derived from allozyme and immunological data in order to assess karyotypic changes that occurred in lineages of Jamaican Eleutherodactylus. Chromosome number changes have occurred at least nine times on the island and have involved both fission and fusion mutational events. C-bands and the sites of secondary constrictions varied and provide very little phylogenetic information. In most instances, karyotypically determined interspecific evolutionary relationships corresponded with the molecular data. The combination of karyological analyses and molecular data clarified lineages which involved convergent chromosome numbers or extremely divergent karyotypes. Karyotypic changes in Jamaican Eleutherodactylus are best explained by chromosome fission, fusion, translocations and inversions which arose in isolated demes and have been fixed through inbreeding and genetic drift. Rates of karyotypic evolution among Jamaican Eleutherodactylus are much faster than previous published rates for frogs. Karyotypic evolution appears to be dictated by behavioural factors and effective population sizes irrespective of taxonomic groupings. 相似文献

Water loss, cutaneous resistance, and the effects of dehydration on jumping ability were measured in two neotropical frogs,
the common coquí (Eleutherodactylus coqui) and the cave coquí (Eleutherodactylus cooki). In both species jumping performance declined with an increase in water loss and a greater duration of exposure to dehydrating
conditions. The arboreal species, E. coqui, had a slightly higher rate of water loss and lower cutaneous resistance than the non-arboreal species, E. cooki. However, differences in cutaneous resistance and water loss were too small to explain differences in geographic distributions
of these species. In both species, a decline in boundary layer resistance contributed to an increased rate of water loss at
higher temperature.

2. | Using a dorsal approach we recorded from the VIIIth nerve near its entry in the brainstem and analyzed responses to both sound and vibration stimuli for 65 fibers in R. pipiens pipiens and 25 fibers in Leptodactylus albilabris. The fibers were classified as amphibian papilla (AP), basilar papilla (BP), saccular or vestibular fibers based on their location in the nerve. Only AP and saccular fibers responded to vibrations. The AP-fibers responded to vibrations from 0.01 cm/s2 and to sound from 40 dB SPL by increasing their spike rate. Best frequencies (BFs) ranged from 60 to 900 Hz, and only fibers with BFs below 500 Hz responded to vibrations. The fibers had identical BF's for sound and vibration. The saccular fibers had BFs ranging from 10 to 80 Hz with 22 fibers having BFs at 40–50 Hz. The fibers responded to sound from 70 dB SPL and'to vibrations from 0.01 cm/s2. |

Early development in the frog model, Xenopus laevis, is governed by RNAs, localized to the vegetal cortex of the oocyte. These RNAs include Xdazl RNA, which is involved in primordial germ cell formation, and VegT RNA, which specifies the mesoderm and endoderm. In order to determine whether orthologues of these RNAs are localized and have similar functions in other frogs, we cloned RpDazl and RpVegT from Rana pipiens, a frog that is phylogenetically distant from X. laevis. RNAs from both genes are localized to the vegetal cortex of the R. pipiens oocyte, indicating that the vegetal localization is likely the basal state. The animal location of EcVegT RNA in Eleutherodactylus coqui that we found previously (Beckham et al., 2003) is then a derived state, probably due to the great increase in egg size required for direct development of this species. To answer the question of function, we injected RpVegT or EcVegT RNAs into X. laevis embryos, and assayed animal caps for gene expression. Both of these RNAs induced the expression of endodermal, mesodermal, and organizer genes, showing that the function of RpVegT and EcVegT as meso-endodermal determinants is conserved in frogs. The RNA localizations and the function of VegT orthologues in germ layer specification may be synapomorphies for anuran amphibians. 相似文献

Clutch effects explain heart rate variation in embryonic frogs (Cave Coqui, Eleutherodactylus cooki)

Few physiological studies to date have focused on whether variation among sibling groups during development can account for often large, intraspecific physiological variation. In this study, we measured heart rate in the direct-developing frog Eleutherodactylus cooki throughout its embryonic development and examined heart rate variation among egg clutches comprising from 10 to 40 eggs. Clutches were collected in the wild in Yubucoa, Puerto Rico, and individual eggs were maintained under equivalent conditions in the lab. Heart rate showed large increases during development, rising from about 40 beats min(-1) in the earliest stages to about 110 beats min(-1) at hatching. The effect of stage (averaged across clutches) was highly significant (P<0.001). However, repeated-measures MANOVA also revealed that there were highly significant effects on heart rate associated with both clutch (variation among clutches averaged across development; P<0.001) and clutch-stage interactions (differences among clutches in the developmental change in heart rate; P<0.0001). These effects and interactions reveal that throughout development, heart rate in siblings is much more similar than in nonsiblings and that sib groups follow different heart rate trajectories during their development. Collectively, these data indicate that "clutch effects" caused by genetic and/or maternal influences can strongly affect patterns of heart function during development within cave coqui populations. This phenomenon also occurs in bird eggs and armadillo neonates, suggesting that physiological variation attributable to clutch effects might be a widespread phenomenon in vertebrates. 相似文献

Three endemic species of the aquatic-breeding frog genus Leptodactylus are recognized from the West Indies: Leptodactylus albilabris (Puerto Rico and the Virgin Islands), Leptodactylus dominicensis (Hispaniola), and Leptodactylus fallax (Lesser Antilles). DNA sequences were obtained from several mitochondrial genes to resolve taxonomic questions involving these species and to provide insights into their origin and distribution in the islands. We found low levels of sequence divergence between L. dominicensis and L. albilabris, supporting morphological evidence that the former species is a junior synonym of the latter species. Phylogenetic analysis supported previous species-group allocations, finding that L. albilabris is a member of the fuscus group and L. fallax is a member of the pentadactylus group. Molecular time estimates for the divergence of L. albilabris from its closest relative in South America (24-58 million years ago, Ma) and for L. fallax from its closest relative in South America (23-34Ma) indicate that they colonized the West Indies independently by over-water dispersal in the mid-Cenozoic. The absence of detectable sequence divergence between the two extant populations of L. fallax (Dominica and Montserrat), a species used for human food and now critically endangered, suggests that one or both arose by human introduction from an island or islands where that species originated. The relatively minor genetic differentiation of populations of L. albilabris can be explained by vicariance and dispersal in the Pleistocene and Holocene, although human introduction of some populations cannot be ruled out. 相似文献

We studied phylogenetic relationships of the Eleutherodactylus auriculatus species group to infer colonization and diversification patterns in this endemic radiation of terrestrial frogs of the genus Eleutherodactylus in the largest of the Greater Antilles, Cuba. An initial screening of genetic diversity based on partial sequences of the 16S rRNA gene in almost 100 individuals of all species of the group and covering the complete known geographic range of their occurrence found most species endemic to small ranges in the eastern Cuban mountains while a single species was widespread over most of Cuba. Our molecular phylogeny, based on 3731 bp of four mitochondrial and one nuclear gene, suggests that most cladogenetic events within the group occurred among clades restricted to the eastern mountains, which acted as refugia and facilitated the diversification in this group. Our results reveal two separate colonization events of Central and Western Cuba and allow inferring the timing of the subsequent diversification events that occurred between 11 and 2 Mya. Because populations previously assigned to E. auriculatus represent four genetically strongly divergent lineages that also differ in their advertisement calls, we propose that E. auriculatus as currently recognized comprises four species. The difficulties in assigning the name auriculatus to any of these four species, and the fact that E. principalis is nested within one of them, stress the need for a thorough taxonomic revision of this group. 相似文献

During the breeding season of the mountain chicken frog Leptodactylus fallax, fighting between males results in the emergence of dominant animals that subsequently attract females to nesting sites. A peptide, termed Leptodactylus aggression-stimulating peptide (LASP), was isolated from norepinephrine-stimulated skin secretions from male specimens of L. fallax that was not present in skin secretions obtained from females. The primary structure of the peptide was established as: Gly-Leu-Trp-Asp-Asp-Leu-Lys-Ala-Ala-Ala-Lys-Lys-Val-Val-Ser-Ser-Leu-Ala-Ser-Ala-Ala-Ile-Glu-Lys-Leu NH2. LASP had no pheromone-like action on females but had a chemoattractive effect on males and stimulated aggressive behaviors, such as rearing and leaping. It is suggested that this peptide may play an important role in initiating the competitive male-male interactions that are associated with the onset of reproductive behavior in L. fallax. 相似文献

Antisera to serum albumins from five West Indian species of the frog genus Eleutheroductylus were prepared, and the reciprocal immunological distances (IDs) obtained were used to provide a time frame for the evolution of this group in the West Indies. One-way IDS were obtained to 25 additional species within the genus, with emphasis on those from the West Indies. These immunological data support both a recent classification of Eleutheroductylus based on an analysis of slow-evolving allozyme loci, and the monophyly of the 17 native Jamaican species as indicated by a more comprehensive electrophoretic study. This is in contrast to the results of morphological studies supporting multiple invasions of Jamaica by Eleutherodactylus . Within the subgenus Euhyas , IDS ranged from6–27 between Jamaican species, whereas between species on different islands the range was29–67. The subgenus Syrrhophus in southern North America was found to be the sister group to the subgenus Euhyus , a western Caribbean clade. Pelorius , a subgenus restricted to Hispaniola, was found to be the sister group of the subgenus Eleutheroductylus in the West Indies. The largest IDs obtained for West Indian species were those between the two major groups, the subgenera Eleutheroductylus and Euhyas .

Morphologically cryptic species act as a wild card when it comes to biodiversity assessments and conservation, with the capacity to dramatically alter our understanding of the biological landscape at the taxonomic, ecological, biogeographic, evolutionary, and conservation levels. We discuss the potential effects that cryptic species may have on biodiversity assessments and conservation, as well as some of the current issues involving the treatment of cryptic species both at taxonomic and conservation levels. In addition, using a combination of advertisement call and morphological data, we describe a new species of the Leptodactylus marmoratus group from the upper Amazon basin, and we assess how cryptic species can affect conservation assessments of species in the Leptodactylus marmoratus group by examining how recent findings affect our understanding of the distribution of what is assumed to be a widespread Amazonian species, Leptodactylus andreae. 相似文献

We report the first phylogenetic analysis of DNA sequence data for the Central American component of the genus Eleutherodactylus (Anura: Leptodactylidae: Eleutherodactylinae), one of the most ubiquitous, diverse, and abundant components of the Neotropical amphibian fauna. We obtained DNA sequence data from 55 specimens representing 45 species. Sampling was focused on Central America, but also included Bolivia, Brazil, Jamaica, and the USA. We sequenced 1460 contiguous base pairs (bp) of the mitochondrial genome containing ND2 and five neighboring tRNA genes, plus 1300 bp of the c-myc nuclear gene. The resulting phylogenetic inferences were broadly concordant between data sets and among analytical methods. The subgenus Craugastor is monophyletic and its initial radiation was potentially rapid and adaptive. Within Craugastor, the earliest splits separate three northern Central American species groups, milesi, augusti, and alfredi, from a clade comprising the rest of Craugastor. Within the latter clade, the rhodopis group as formerly recognized comprises three deeply divergent clades that do not form a monophyletic group; we therefore restrict the content of the rhodopis group to one of two northern clades, and use new names for the other northern (mexicanus group) and one southern clade (bransfordii group). The new rhodopis and bransfordii groups together form the sister taxon to a clade comprising the biporcatus, fitzingeri, mexicanus, and rugulosus groups. We used a Bayesian MCMC approach together with geological and biogeographic assumptions to estimate divergence times from the combined DNA sequence data. Our results corroborated three independent dispersal events for the origins of Central American Eleutherodactylus: (1) an ancestor of Craugastor entered northern Central America from South American in the early Paleocene, (2) an ancestor of the subgenus Syrrhophus entered northern Central America from the Caribbean at the end of the Eocene, and (3) a wave of independent dispersal events from South America coincided with formation of the Isthmus of Panama during the Pliocene. Summary Rate of evaporation, resistance to drying, temperature selection, and critical thermal maximum were measured for Jamaican Eleutherodactylus acclimated to 20° C and a 12:12 L:D photoperiod. Two introduced species, E. planirostris and E. johnstonei, selected higher temperatures and had higher CTMs than two native species, E. cundalli and E. gossei. The introduced species lost water as rapidly as the native species, but tolerated 30–73% greater water loss before losing their righting response. The physiological differences are reflected in the microhabitat selection and activity patterns of the four species. 相似文献

Methylthioadenosine phosphorylase (MTAP) is a key enzyme in the methionine and adenine salvage pathways. In mammals, the liver plays a central role in methionine metabolism, and this essential function is lost in the progression from liver cirrhosis to hepatocarcinoma. Deficient MTAP gene expression has been recognized in many transformed cell lines and tissues. In the present work, we have studied the expression of MTAP in human and experimental liver cirrhosis and hepatocarcinoma. We observe that MTAP gene expression is significantly reduced in human hepatocarcinoma tissues and cell lines. Interestingly, MTAP gene expression was also impaired in the liver of CCl4-cirrhotic rats and cirrhotic patients. We provide evidence indicating that epigenetic mechanisms, involving DNA methylation and histone deacetylation, may play a role in the silencing of MTAP gene expression in hepatocarcinoma. Given the recently proposed tumor suppressor activity of MTAP, our observations can be relevant to the elucidation of the molecular mechanisms of multistep hepatocarcinogenesis. 相似文献
