Maan[1] and Endo[2] et al. first reported that some chromosomes from Ae. longgissima, Ae. sharonensis and Ae. triuncialis showed preferential transmission when introduced into wheat background. Specific chromosomes of certain Aegilops species introduced into wheat genome background may often facilitate chromosome breakage and refusion, and finally result in a variety of chromosome restructuring. Such a phenomenon is commonly called gametocidal effect of the chromosomes. The chromosome 2C of Ae. cylindrica is one of such chromosomes. In the present study, scab resistant wheat-L. racemosus addition lines involving chromosomes Lr.2 and Lr.7 were crossed to wheat-Ae. cylindrica disomic addition line Add2C. Then F1 hybrids were subsequently backcrossed with wheat cv “Chinese Spring”. BC1 plants with chromosome structural aberration were identified by C-banding. In the self-pollinated progenies of these plants, three translocation lines were developed and characterized by mitotic and meiotic analysis combined with C-banding and fluorescent in situ hybridization (FISH) using biotin-labeled genomic DNA of L. racemosus as probe. Some other putative translocation lines to be further characterized were also found. The practicability and efficiency of the translocation between wheat and alien chromosomes induced by gametocidal chromosomes, as well as the potential use of the developed alien translocation lines were also discussed.  相似文献

通过醋酸洋红压片和荧光原位杂交技术(包括基因组原位杂交技术),确定在我国广西西南部地区广泛分布着的水生薏苡(Coix aquatica Roxb.)属于一种新的六倍体细胞类型.这种水生薏苡与已报道的几种水生薏苡细胞类型的染色体数目均不相同,它的染色体数目是2n=30,在减数分裂前期Ⅰ和中期Ⅰ的细胞中形成10个二价体和10个单价体.基因组原位杂交结果表明,这种水生薏苡的20条染色体与四倍体的薏苡(C.lacryma-jobi,2n=20)的基因组DNA是高度同源的.45S和5S rDNA分别杂交到这种水生薏苡的两条染色体上,其中各有一条染色体与薏苡中携带45S和5S rDNA杂交信号的染色体具有相同的形状和信号的分布状态.据此推测:四倍体的薏苡可能是这种新的水生薏苡细胞类型的一个亲本,它的另一个亲本可能是八倍体的水生薏苡(C.aquatica,2n=40),因为这种八倍体的水生薏苡在核型、植株形态及生长环境等方面与新的六倍体细胞类型的水生薏苡相似.  相似文献

用改良的Giemsa C-带技术对10个经辐射处理的带有黑麦某些性状的普通小麦品系进行鉴定。结果鉴定出1个小麦—黑麦易位系及2个小麦—黑麦异代换系,探索出冬季和夏季染色体分带的不同条件。因此,只要条件控制得当,C-分带不失为一种快速、精确而经济的鉴定外源染色体的方法。  相似文献

Chromosome aberration frequency and lipid peroxidation levels were analysed to investigate their efficacy as biological markers for monitoring the genotoxicity and oxidative damage in Korean chromium (Cr)-exposed workers. Fifty-one Cr-exposed workers and 31 age-matched controls in ten chrome-plating plants were sampled. The Cr level was measured in the workers' blood and urine, and in the ambient air at the workplaces. The conventional Giemsa staining method and fluorescence in situ hybridization (FISH) technique were used for chromosome aberration analysis. Spectrum green whole chromosome paint specific for chromosome 4 was used in the FISH procedure. As for lipid peroxidation, malondialdehyde (MDA) was measured in the blood plasma as thiobarbituric acid-reactive substances (TBARS). The blood Cr concentration was statistically correlated with both the frequency of chromatid exchange and the total frequency of chromosome/chromatid breaks and exchanges, as detected by the Giemsa staining. Meanwhile, the frequency of translocation, as detected by the FISH technique, was significantly higher in the Cr-exposed workers than in the controls and it correlated with the blood Cr concentration. Although the concentration of MDA, the metabolite of lipid peroxidation, in the exposed workers was higher than that in the controls, no statistically significant correlation between the MDA level and the blood or urine Cr levels was observed. Accordingly, the genotoxicity and oxidative damage (plasma lipid peroxidation) in the Korean Cr-exposed workers were consequential at quite low exposure levels, plus chromosome rearrangement, especially translocation, was clearly evident as a biological response marker for Cr exposure based on a significant positive correlation between the translocations detected by FISH and the Cr in the blood.  相似文献

Fluorescence in situ hybridization (FISH) was carried out in somatic cells of racemosus (lam.) Tzvel. and Thinopyrum junceum ( Savul. & Rayss) A. Love using Th. bessarabicum ( Savul. & Rayss) A. Love genomic DNA as probe. Fourteen pairs of chromosomes in L. racemosus gave positive signal, and only seven of fourteen pairs of chromosomes in Th. junceum showed signal. In FISH probed with PHv62, the chromosomes in Th. bessarabicum and L. racemosus hybridized with PHv62, whereas Th. Bessarabicum showed positive signal in four pairs of chromosomes, and the latter in thirteen pairs of chromosomes. No positive signal was observed in chromosomes of Psathyrostachys juncea (Fisch.) Nevski and Th. junceum. The results of Southern hybridization probed with PHv62 were similar to those of in situ hybridization. Twelve alien chromosome lines of T. aestivum-L, racemosus were detected by PHv62 probing. In most of the alien chromosome lines, PHv62 hybridized in fragment of Leymus chromosomes, except for the line containing chromosomes 5Lr # 1 and 1 1Lr # 1. It is inferred that Th. bessarabicum may involve in the formation of species. Non the less, significant changes have occurred during the evolution of Leymus genomes.  相似文献

用Thinopyrumbesarabicum(Savul.&Rays)A.Lve的基因组DNA作探针,分别与大赖草Leymusracemosus(Lam.)Tzvel.和脆轴偃麦草Th.junceum(Savul.&Rays)A.Lve的体细胞杂交,大赖草的14对染色体均出现杂交信号,脆轴偃麦草只有7对染色体有杂交信号。在用重复DNA序列PHv62作探针的原位杂交中,Th.besarabicum有4对染色体有杂交信号,大赖草有13对染色体显示杂交信号,新麦草Psathyrostachysjuncea(Fisch.)Nevski和脆轴偃麦草无杂交信号。用PHv62作探针的Southern杂交结果与原位杂交相似。在被检测的12个普通小麦大赖草异源染色体系中,除二体附加系中5Lr#1和双二体附加系1Lr#1+5Lr#1没有杂交信号外,其余的异染色体系与PHv62都有特异杂交信号。据此推测Th.besarabicum有可能参予了赖草属物种的形成过程。但是,大赖草的染色体组在进化过程中显然已发生过变异。  相似文献

A standard karyotype and a generalized idiogram of Triticum umbellulatum (syn. Aegilops umbellulata, 2n = 2x = 14) was established based on C-banding analysis of ten accessions of different geographic origin and individual T. umbellulatum chromosomes in T. aestivumT. umbellulatum chromosome addition lines. Monosomic (MA) and disomic (DA) T. aestivumT. umbellulatum chromosome addition lines (DA1U = B, DA2U = D, MA4U = F, DA5U = C, DA6U = A, DA7U = E = G) and telosomic addition lines (DA1US, DA1UL, DA2US, DA2UL, DA4UL, MA5US, (+ iso 5US), DA5UL, DA7US, DA7UL) were analyzed. Line H was established as a disomic addition line for the translocated wheat — T. umbellulatum chromosome T2DS·4US. Radiation-induced wheat — T. umbellulatum translocation lines resistant to leaf rust (Lr9) were identified as T40 = T6BL·6BS-6UL, T41 = T4BL·4BS-6UL, T44 = T2DS·2DL-6UL, T47 = Transfer = T6BS·6BL-6UL and T52 = T7BL·7BS-6UL. 45SrDNA和5SrDNA是水稻中与核糖体RNA合成有关的2个功能片段,有关这2个序列在水稻染色体上的位置,不同研究者的研究结果不尽相同,在获得水稻染色体清晰制片的基础上,通过FISH确定了45SrDNA序列位于水稻的第9号和第10号染色体的短臂末端,并且第9号染色体上的拷贝数多于第10号染色体,5SrDNA序列位于第11号染色体短臂靠近着丝点处。  相似文献

本实验室已经通过基因芯片技术筛选到一个白粉菌诱导后上调表达的抗病相关基因Hv-S/TPK, 并获得了它的全长cDNA序列。利用Hv-S/TPK的特异引物筛选小麦-簇毛麦6VS/6AL易位系基因组可转化人工染色体(Transformation-competent artificial chromsome, TAC)文库, 获得了阳性TAC单克隆, 并进一步获得了含有Hv-S/TPK cDNA序列的5160 bp(GenBank Accession No. EU153366)的亚克隆。对亚克隆的序列分析结果表明, Hv-S/TPK基因在起始密码子和终止密码子之间有3个内含子和4个外显子, 4个外显子序列与簇毛麦上已得到的Hv-S/TPK的cDNA序列100%同源。对起始密码子上游序列分析结果表明, 该基因的调控序列中, 含有W-Box、OCS-element等与抗病相关的元件。以TAC克隆为探针与小麦-簇毛麦6VS/6AL易位系有丝分裂中期染色体进行荧光原位杂交(Fluorescence in situ hybridization, FISH), 结果表明含有Hv-S/TPK基因的TAC克隆来自于簇毛麦。  相似文献

By chromosome C-banding and bi-color fluorescence in situ hybridization (FISH) using digoxigenin-labelled total genomic DNA of Leymus racemosus (Lam.) Tzvel. and biotinylated total genomic DNA of Haynaldia villosa (L.) Schur as probes, three wheat-alien lines with L. racemosus Lr.7 addition and H. villosa 6VS/6AL translocated chromosomes, and eight lines with L. racemosus Lr.14 addition and H.villosa 6VS/6AL translocated chromosomes were respectively identified from DALr.7×T6VS/6AL (93G51-4×P64) and DALr.14×T6VS/6AL (94G15×P64)F2 or F3 hybrids. Fluorescein-isothiocyanate-conjugated avidin and rhodamine-conjugated sheep anti-digoxigenin Fab fragment were used in bi-color FISH detection. The chromosomes of L.racemosus and 6VS fragment of H. villosa were simultaneously detected by their red and green fluorescence. Powdery mildew and scab resistance were also evaluated. The result showed that the obtained plants had high resistance to these two diseases. The potential usage of bi-color FISH in identifying chromatin of L.racemosus and H.villosa was discussed.  相似文献

Aegilops speltoides Tausch (2n = 2x = 14, SS) is considered as the closest living relative of the B and G genomes of polyploid wheats. A complete set of Triticum aestivum L. cv Chinese Spring-Ae. speltoides whole chromosomes and seven telosomic addition lines was established. A low pairing accession was selected for the isolation of the chromosome addition lines. Except for chromosomes 3S and 6S, which are presently only available as monosomic additions, all other lines were recovered as disomic or ditelosomic additions. The individual Ae. speltoides chromosomes isolated in the wheat background were assayed for their genetic effects on plant phenotype and cytologically characterized in terms of chromosome length, arm ratio, distribution of marker C-bands, and FISH sites using a Ae. speltoides-specific repetitive element, Gc1R-1, as a probe. The homoeology of the added Ae. speltoides chromosomes was established by using a standard set of RFLP probes. No chromosomal rearrangements relative to wheat were detected. The cytogenetics of the backcross generations and self-bred progenies (BC2F1, BC3F1, BC2F2, BC3F2 and BC2F3) of intergenetic hybrid of Triticum aestivum L. x Eremopyrum orientale (kedeb) Jaub. Et Spach were studied. The results showed that the plants BC3F1 (2n = 43 ) isolated from the backcross generations of the plants (2n = 44 ) accounted for 41.09%, but the plants (2n = 44) isolated was only 4. 11%, and the plants BC2F2 (2n = 44) isolated from self-cross generations of the plants BC2F1 (2n = 44) accounted for 13.21%. The number of univalents in pollen mother cells was higher in some plants (BC2F1) and the averages of univalents were negatively related to the backcross seed-setting and self-cross seed-setting with a related coefficient of - 0.6766* and -0.7429* respectively. The results of genomic in situ hybridization (GISH) showed that some plants of BC2Fs (2n=44) contained different number of alien chromosomes. All those indicated that alien chromosomes caused unusual pairing and segregation of wheat homologous chromosomes and also lowed the hereditary stability of wheat chromosomes.  相似文献

Two substitution lines, designated as 930498 and 930483, and one addition line, designated as 930029, via Fo immature embryo culture of Triticum aestivum x octoploid triticale ( x Triti-cosecale Wittmack) were identified. Fluorescence in situ hybridization (FISH) using total genomic DNA of rye ( Secale cereale L. ) as probe corroborated the existence of rye chromosomes, further confirmed through chromosome paring at meiotic metaphase 1, C-banding and glutenin SDS- PAGE. The results demonstrated that the two substitution lines are ID/IR, and the addition line is also IR addition. Rye chromosomes that are distinct to the red-colored wheat chromosomes appear yellow-green at mitotic metaphase after FISH.  相似文献

利用染色体荧光原位杂交技术(FISH),将3种重复序列5S rDNA、45S rDNA和C0t-1 DNA用不同荧光物进行标记,对我国10个不同地理来源的苜蓿种质(Medicago sativa L.;2n=4X=32)进行了染色体多态性检测。结果表明,利用以上重复序列可以较好地将苜蓿32条染色体区分为16对特征不同的染色体,10份不同种质材料FISH带纹特征表现高度相似,比较不同种质间同源染色体重复序列杂交特征,揭示出种质群体内和群体间多态性染色体的存在,其中不同的同源染色体多态性表现不尽一致,1号染色体(随体染色体)多态性最高,10份材料中检出7个多态型,3、4、15号染色体保守性较强,在不同种质间表现为单态,其他染色体多态性居中。对在地理分布上自西向东的10个材料进行染色体多态性比较,结果显示分布于西藏、新疆以及分布在辽宁的材料部分染色体多态型显著区别于其他材料。  相似文献
