The twenty-four hour activity cycle of captive coypus was investigated by direct visual observation, automatic recording and by measuring the amount of faeces eliminated at hourly intervals. Coypus were active from just before sunset and the "Zeitgeber" or cue for activity onset may be declining light. The activity period usually ended before sunrise and the interval between the end of the period and dawn was inversely related to temperature. The animals fed and defaecated throughout the active period and 80–86 % of the faecal pellets were produced in water. Refection started shortly after the end of the feeding period, when the animals had returned to the nest, and continued until an hour or two after midday.
A model is suggested which involves two passages of food through the gut and a cycle length of 24 hours from ingestion to defaecation. The feeding and defaecation periods were equal in duration, but the refection period was 40% shorter. Hypothetically this may be due to a caecum by-pass mechanism during the second passage of food through the gut so that the defaecation period, like the feeding period, is controlled by the rate at which food passes through the pyloric sphincter. In contrast the period of refection may be controlled by the rate at which food remains are voided from the caecum. Coypus spent the pre-dawn and daylight hours on nests and in contact with familiar conspecifics. These behaviours may have the function of temperature maintenance in the coldest part of the night and of avoiding diurnal predators.  相似文献

1. Readings were made on the rates of oxygen consumption and on the activities of the succinoxidase system of eggs of the mealworm for each day of embryonic development at 30°C. 2. The rate of oxygen consumption, expressed as microliters/50 eggs/hour, was low (4.89) in newly laid eggs. It rose to 7.41 during the next 24 hours, remained at this level for the next 2 days, and then increased during the remainder of the embryonic period reaching a high value of 14.79 at the time of hatching. 3. The activity of cytochrome oxidase in eggs from newly emerged beetles, expressed as Δ log [Cy Fe++]/minute, remained at a value of 0.042 during the first half of the embryonic period, increasing to 0.233 during the latter half of this period. 4. The activity of succinic dehydrogenase showed the same series of changes except at much lower values. Expressed as Δ log [Cy Fe+++]/minute, they ranged from 0.010 in the newly laid egg to 0.034 at the end of the embryonic period. 5. The activity of cytochrome oxidase of the egg was found to decrease with parental age. Eggs from newly emerged beetles had activity values considerably higher than those of beetles 6 or 8 weeks after emergence. However, no comparable changes were noted in the activity of succinic dehydrogenase or in the rate of oxygen consumption. These observations suggest that cytochrome oxidase is not a rate-limiting enzyme in the respiratory metabolism of the mealworm egg.  相似文献

Malate dehydrogenase activity and soluble protein content in testes from rats exposed to a 14:00 h light:10:00 h dark photoperiod, have been determined every two or four hours over a 24 hour period in 5, 15, 25 and 120 day-old rats. By using the Cosinor method, the ontogeny of an unimodal rhythm was studied for MDH activity and soluble protein content in testis. In 5 and 15 day-old rats, the MDH acrophases were recorded around 19:00 h and 17:00 h, respectively. Rats aged 25 and 110 days showed the MDH acrophases during the dark period. An inversion of the MDH circadian rhythms was detected in 25 day-old compared to those of 5 and 15 day-old rats. An inversion of the protein circadian rhythm was also detected at 15 days compared to that at 5 days. These inversions persist in the adult rats. The amplitude of the MDH and protein rhythms reached the lowest value in adulthood. The mean daily value of testicular MDH increased between day 5 and 15, decreasing at day 35 and remaining unchanged until adulthood. The variation of malate dehydrogenase activity, soluble protein content levels, and the circadian rhythm parameters during the maturation process may be related to gonad development.  相似文献

Feeding high protein diet for 5 days caused a 3,5-fold and 2-fold increase of the activity of xanthine dehydrogenase (EC 1.2.1.37) and 5-nucleotidase (EC 3.1.3.31) respectively, in chicken liver. Six hours after feeding the high protein diet there was no change in either enzyme activity although a 3-fold increase in the level of serum uric acid was observed. High protein diet considerably decreased the activity of AMP deaminase at low, but not at high substrate concentration. The activity ratio, measured at 10.0 and 0.16 mM AMP increased from 14:1 (low protein diet) to 23:1 and 24:1 after 6 h and 5 days of high protein diet, respectively. It has been suggested that feeding birds a high protein diet may cause transformation of liver AMP deaminase (EC 3.5.4.6) from a low Km form toward a high Km form.  相似文献

用鱼粉和黄豆饼粉为主要蛋白源的试验饲料饲养草鱼两周,测定摄食前、后肠组织和肝胰脏蛋白酶、淀粉酶活性的变化以及不同时间前、中、后肠组织蛋白酶活性。结果表明:随摄食后时间的推移,鱼粉组肠组织蛋白酶活性迅速增高,到10h 达最大值,黄豆饼组则在15h 以后。两组肠组织淀粉酶活性均在10h 达到最大值。肝胰脏蛋白酶活性均在摄食后立即下降,到5h 降到最低值,随后缓缓升高,淀粉酶活性变化较平缓。肝胰脏蛋白酶与肠组织蛋白酶活性之间存在一种消长关系。总的看来,鱼粉组的蛋白酶、淀粉酶活性均高于黄豆饼组。前肠组织蛋白酶活性最高,中肠次之,后肠最低。    相似文献

A culture of Skeletonema costatum grown at a light intensity of 3 klux and at 20°C was synchronized in diurnally intermittent illumination of 12 hour light and 12 hour dark. The culture was hardly fully synchronous as the cell division period lasted about 9 hours. The cell division started in the middle of the light period. The concentration of the pigments: chlorophyll a, chlorophyll 6 and fucoxanthin and the rate of light-saturated photosynthesis were followed every hour during the 24 hour period. Both the concentration of pigments and the photosynthetic activity showed a rhythmical variation. The concentration per cell of all three pigments examined increased during the development of the cells and decreased automatically during the period of cell division. An increase in the pigment concentration was found only in the light period. The rate of light-saturated photosynthesis calculated per unit of cell number increased during the cell development and decreased during the division period. The increase in the photosynthetic activity at light-saturation started about 4 hours after the end of cell division, which was 4 hours before the light was turned on while the increase in the concentration of chlorophyll a first started 1–2 hours after this moment. The variation in photosynthetic activity was compared with that found by other workers. The results found with Chlorella ellipsoidea by Japanese scientists (Nihci et al.) was explained as an inhibition phenomenon because the cells were not adapted to the experimental conditions.  相似文献

Roots of Vicia faba were treated with solutions of colchicine or IAA or both. Mitotic indices and the frequencies of the different stages of mitosis were determined immediately after a three hour treatment or following a 24 hour period of recovery. Roots scored after treatment with colchicine for three hours showed several effects, none of which were reversed by simultaneous treatment with IAA. Treatment with IAA for three hours had little detectable effect on mitotic index (MI) on the frequencies of the various stages of mitosis. After a recovery period, following a three hour treatment, of 24 hours, colchicine treated roots showed a significant increase in their MI; this was due largely to an increase in the number of metaphases but it was also due in part to the presence of tetraploid cells in division. IAA treated roots revealed an inhibition of mitotic activity, which was most marked at 3.13–6.26×10–4 M IAA. The results from roots treated with mixtures of colchicine and IAA for three hours and fixed 24 hours later showed: 1) the increase in MI induced by colchicine is reversed by IAA, the intensity of the reversal increasing with increasing concentrations of IAA; 2) reductions in the total numbers of cells in prophase or in metaphase occur after treatment with different concentrations of IAA; 3) IAA leads to a reduction in the number of tetraploid cells seen in division.It appears that colchicine induces a change in the pattern of mitotic activity 24 hours after the end of treatment and its effects are reversed by IAA. At 4.2×10–4 M IAA a balance occurs between the opposing effects of colchicine and IAA and the MI is not significantly different from that of the controls. It is suggested that one result of a treatment with colchicine is a change in the level of growth factors in root meristems. This change, which appears to result in a temporary increase in MI is reversed by the addition of IAA. Thus one of the growth factors, the level of which has been affected, is replaceable by exogenous IAA.  相似文献

We assessed the daily patterns of parameters involved in energy metabolism in plasma and brain of rainbow trout. Where daily rhythms were found, we analyzed the potential influence of feeding. Immature rainbow trout were randomly distributed in 3 groups: fish fed for 7 days, fish fasted for 7 days, and fish fasted for 7 days and refed for 4 days. On sampling day, fish of fed and refed groups were fed at 11.00 h, and all fish were sampled from each treatment group using the following time schedule: 14.00, 18.00, 21.00, 00.00, 04.00, 07.00, 10.00 and 14.00 h. The results obtained from metabolic parameters assessed in plasma and brain can be grouped into three different categories, such as (i) those displaying no 24 h changes in fed fish such as plasma lactate, protein or acetoacetate levels, as well as brain amino acid and protein levels, and lowKm(glucose) hexokinase, and aspartate aminotransferase activities, (ii) those displaying 24 h changes that were apparently dependent on feeding since they disappeared in fasted fish such as the case of plasma cortisol, glucose and triglyceride levels, as well as brain glycogen, glucose, and lactate levels, and pyruvate kinase and hexokinase IV activities, and (iii) those parameters displaying 24 h changes apparently not dependent on feeding such as plasma amino acids, brain acetoacetate levels as well as several enzyme activities measured in brain such as glucose 6-phosphate dehydrogenase, alpha-glycerophosphate dehydrogenase, glutamate dehydrogenase, and lactate dehydrogenase-oxidase. In general, 24 h changes dependent on feeding indicate an increased use of glucose in brain several hours post-feeding whereas those changes not dependent on feeding were characterized by reduced levels/activity at the night period suggesting a metabolic depression in brain during darkness.  相似文献

This study aimed to elucidate the role of urea synthesis in the slender African lungfish Protopterus dolloi in detoxifying ammonia after feeding. There were significant increases in the rate of ammonia excretion in P. dolloi between hours 6 and 15 after feeding. Simultaneously, there were significant increases in urea excretion rates between hours 3 and 18. Consequently, the percentage of total nitrogen (N) excreted as urea N increased to ~60% between hours 12 and 21 post-feeding. Hence, after feeding, the normally ammonotelic P. dolloi became ureotelic. Approximately 41% of the N intake from food was excreted within 24 h by P. dolloi, 55% of which was in the form of urea N. At hour 12 post-feeding, the accumulation of urea N was greater than the accumulation of ammonia N in various tissues, which indicates that feeding led to an increase in the rate of urea synthesis. This is contrary to results reported previously on the infusion of ammonia into the peritoneal cavity of the marine dogfish shark, in which a significant portion of the exogenous ammonia was excreted as ammonia. In contrast, feeding is more likely to induce urea synthesis, which is energy intensive, because feeding provides an ample supply of energy resources and leads to the production of ammonia intracellularly in the liver. The capacity of P. dolloi to synthesize urea effectively prevented a postprandial surge in the plasma ammonia level as reported elsewhere for other non-ureogenic teleosts. However, there was a significant increase in the glutamine content in the brain at hour 24, indicating that the brain had to defend against ammonia toxicity after feeding.Communicated by I.D. The objective of this study was to determine the effects of feeding on the excretory nitrogen (N) metabolism of the aquatic Chinese soft-shelled turtle, Pelodiscus sinensis, with a special emphasis on the role of urea synthesis in ammonia detoxification. P. sinensis is ureogenic and possesses a full complement of ornithine-urea cycle enzymes in its liver. It is primarily ureotelic in water, and the estimated rate of urea synthesis in unfed animals was equivalent to only 1.5% of the maximal capacity of carbamoyl phosphate synthetase I (CPS I) in its liver. Approximately 72 h was required for P. sinensis to completely digest a meal of prawn meat. During this period, there were significant increases in ammonia contents in the stomach at hour 24 and in the intestine between hours 12 and 36, which could be a result of bacterial activities in the intestinal tract. However, ammonia contents in the liver, muscle, brain and plasma remained unchanged throughout the 72-h post-feeding. In contrast, at hour 24, urea contents in the stomach, intestine, liver, muscle, brain and plasma increased significantly by 2.9−, 3.5−, 2.6−, 2.9−, 3.4 and 3.0-fold, respectively. In addition, there was a 3.3- to 8.0−fold increase in the urea excretion rate between hours 0 and 36 post-feeding, which preceded the increase in ammonia excretion between hours 12 and 48. By hour 48, 68% of the assimilated N from the feed was excreted, 54% of which was excreted as urea-N. The rate of urea synthesis apparently increased sevenfold during the initial 24 h after feeding, which demanded only 10% of the maximal CPS I capacity in P. sinensis. The postprandial detoxification of ammonia to urea in P. sinensis effectively prevented postprandial surges in ammonia contents in the plasma and other tissues, as observed in other animals, during the 72-h period post-feeding. In addition, postprandial ammonia toxicity was ameliorated by increased transamination and synthesis of certain amino acids in the liver and muscle of P. sinensis. After feeding, a slight but significant increase in the glutamine content occurred in the brain at hour 24, indicating that the brain might experience a transient increase in ammonia and ammonia was detoxified to glutamine.  相似文献

目的探讨相同剂量双歧杆菌完整肽聚糖(WPG)在不同时间对小鼠骨髓来源树突状细胞分泌IL-12的影响。方法将双歧杆菌WPG与小鼠骨髓来源树突状细胞共培养,分别在加入WPG后0、12、24、36、48和60h收集培养上清液,用ELISA法测定不同时间点上清液中IL-12的量。结果双歧杆菌WPG与树突状细胞共培养后,上清液中IL-12的量在24h内逐渐升高,至24h达高峰,24h后呈下降趋势;同0h组比较,12、24和36h组IL-12分泌量明显增加(P〈0.05),而48、60h组同0h组比较差异无显著性(P〉0.05)。结论双歧杆菌WPG能够刺激小鼠骨髓来源的树突状细胞分泌IL-12;双歧杆菌WPG对树突状细胞的免疫刺激存在时间效应关系。  相似文献

The digestibility of different forms of starch was examined in an ileostomy model. Six otherwise healthy ileostomists were fed a controlled polysaccharide-free diet for four days, on three of which a test starch was added at breakfast. 50 g starch was fed as either whole or homogenized chick peas or as a retrograded starch gel. A readily digestible wheat starch biscuit was used as a control. Ileostomy effluent was collected every 2 hours over a 16 hour period and a final collection made at 24 hours after the test meal. The monosaccharide composition and glycosyl linkages of the residual carbohydrate in the 2 hour peak period following the test meal was determined. Following consumption of the starch gel, poly- and oligo-saccharides from mucin and starch were identified in the effluent. At the peak of effluent production following the test meal, the average ratio of starch polysaccharide to mucin was 1:0.4. Of the 50 g of starch consumed, 7% of the starch escaped digestion in this fraction. Following consumption of cooked, cooled chick peas, which were fed whole or homogenized, polysaccharides deriving from starch, mucin and the cell wall were detected in the effluent. It was estimated from comparison of the composition of the food and effluent that 14% and 16% of the ingested starch in the form of homogenized and whole chick peas had escaped digestion in the small intestine. Linkage analysis showed the chemical structure of the starch escaping digestion after feeding the whole and homogenized chick peas was similar to that obtained after feeding the starch gel.  相似文献

The experiments on (CBA X C57BL/6)F1 mice have shown that regular corazol injections in subliminal doses stimulated seizure susceptibility (pharmacological kindling). Cytophotometric assay of the activity of oxidative metabolism enzymes (glutamate dehydrogenase, malate dehydrogenase, succinate dehydrogenase, alpha-oxoglutarate dehydrogenase, lactate dehydrogenase) and GABA-transaminase in the sensorimotor cortex of kindled mice in post-convulsive period, and 24 hours or 30 days after corazol injections were discontinued, has revealed some specific alterations of the enzymes under study, that suggest the existence of two phases of energy metabolism disturbances. The first phase (24 hours after corazol injections were discontinued) is characterized by intensified succinic acid oxidation, while the second phase (30 days after the last injection) is characterized by anaerobic glycolysis in neuronal and glial cells. Inhibition of GABA-transaminase activity was particularly marked in postconvulsive period. From a molecular point of view these data may be considered as enzyme disturbances during stimulation of seizure susceptability or seizure activity and as a compensation component ensuring anticonvulsive mechanisms and reparative processes (antagonistic principle of molecular mechanism regulation) during activation of antiepileptic system.  相似文献

A study of the puffing pattern of the salivary gland autosomes of D. melanogaster was performed through the last 24 hours of larval development and 0-hour prepupae. Since both prominent and small puffs were taken into account, the total puff number amounted to 275. Of these, 116 are almost constant in size during the 24 hours observation period, 106 increase in size or appear before pupation. 37 puffs are active in 96 hour larvae and disappear or decrease sharply in size by 115–118 hours. 12 biphasic puffs have been found with higher activity in 96 hour larvae and 0-hour prepupae and lower activity by 115–118 hours. Three extremely irregular puffs have been detected in chromosome 4. The data obtained evidence that a larger number of D. melanogaster polytene chromosome loci are active during larval development than it has been thought earlier. It has also been shown that only 38% of autosomal puffs change before the beginning of metamorphosis. The functional significance of small puffs and strain specificity of puffs are discussed.  相似文献

The activity of tyrosine aminotransferase (TAT) was measured in the livers of rats which were entrained to eat for the first 2 hours of a daily 12 hour dark period (‘2+22’ schedule) and were treated with the synthetic glucocorticoid dexamethasone and with glucagon at several times of day. TAT activity in untreated animals varies diurnally with a maximum 4 to 6 hours after the beginning of feeding. In both fed and fasted rats there was a small diurnal variation in inducibility by dexamethasone: in fed rats induction was greatest near the beginning of the dark period, shortly after feeding; in fasted rats induction increased towards the end of the dark period. Glucagon induction showed a marked diurnal variation in fed rats with a decrease coincident with the decline in control TAT activity after its food-induced peak. This variation did not appear to be depemdent on food intake, however, since the decline in inducibility occurred in fasted rats at the same time as in fed rats. Co-treatment with dexamethasone did not affect the decrease in glucagon inducibility. The diurnal variation in TAT induction may reflect a diurnal rhythm in the components of the enzyme synthesizing system (e.g. in the availability of mRNA or in enzyme degradation).  相似文献

昆虫取食作为一种关键的生物胁迫因子对棉花防御机制产生了重要影响。植物对昆虫取食产生的防御响应,在昆虫与植物的生态关系中具有重要作用。为了明确棉铃虫Helicoverpa armigera取食与棉花防御性之间的动态互作关系,本文研究了棉铃虫持续取食下及停止取食后,棉花中3种防御相关酶活性变化的时间效应。在明确了棉花受损程度与棉铃虫取食时间关系的基础上,分别考察了棉铃虫持续取食2、6、12、18和24h,对棉花中苯丙氨酸解氨酶(PAL)、脂氧合酶(LOX)和多酚氧化酶(PPO)活性的影响。针对棉铃虫持续取食棉叶12h后停止取食,研究了去除虫害胁迫后0、6、12、24和36h,棉花体内PAL、LOX、PPO活性的变化。结果表明:在棉铃虫持续取食棉叶24h内,棉花中3种防御酶的活性响应有所不同,其中,棉铃虫持续取食2和6h对棉花体内PAL活性没有产生显著影响,而持续取食12h显著诱导了PAL活性,持续取食24和36h,均极显著诱导了PAL活性;棉铃虫持续取食2、6、12、和18h均显著诱导了棉花体内LOX活性,持续取食24h极显著诱导了LOX活性;棉铃虫持续取食6h极显著诱导了棉花体内PPO活性,持续取食24h显著诱导了PPO活性。棉铃虫取食12h后停止取食,在去除虫害除胁迫后0、6、12、24和36h,棉花体内PAL活性均显著升高;而LOX活性则呈现出先升高后恢复正常的现象;PPO活性开始无变化,但在胁迫去除后12和24h显著增高,到36h恢复正常。可见,棉花体内PAL、LOX和PPO活性对棉铃虫取食产生的防御响应,与其受虫害持续取食胁迫时间的增长呈正相关,随着取食时间和受危害的程度加大而升高。并且,在虫害胁迫去除后的一定时间内,棉花体内PAL、LOX和PPO活性依然会保持较高的活性状态,而同等程度机械损伤后的棉叶内PAL、LOX、PPO活性均没有发生显著性变化。说明棉花对于棉铃虫取食胁迫的防御与棉花生理生化性质的改变有关,且具有持续性。  相似文献

Waterbuck activity was recorded at four-minute intervals for continuous periods of 12 hours on two or three consecutive days, and for single nocturnal periods. Feeding patterns of the male and female were similar and during the day there was no hour during which some feeding did not take place. Only male activity was recorded at night. This showed more intensive feeding but with periods of no feeding just after dark and just before dawn. The use of a four-minute recording interval was found to work well in practice.  相似文献

In rat liver following white phorphorus poisoning a biphasic increase in cyclic AMP concentration was observed. After a lag period of 1 hour the cyclic AMP content rose to a first peak at 4 hours and to a second peak at 12 hours of intoxication. The cyclic AMP level fell to normal after 24 hours, by which time the cyclic nucleotide concentration was approaching control values. On the contrary, cyclic GMP content was found to the normal level during the different stages of intoxication. Only at 36 hours the cyclic GMP amount appeared significantly increased above the control values. Serum activity of alanine- and aspartate-amino transferases was found changed from 8 hours to 24 hours after poisoning. The serum level of the two enzymes was overlapping the control values after 36 hours. These results are discussed in relation to hepatocyte necrosis following white phosphorus intoxication.  相似文献

Mammary explants from midpregnant mice were cultured for 4, 24, 48, and 72 hours in the presence and absence of insulin. Changes in the activities of phosphoglucose isomerase (an enzyme of the glycolytic pathway for glucose metabolism) and of glucose-6-phosphate and 6-phosphogluconate dehydrogenases (enzymes of the pentose phosphate pathway) were assessed at each culture interval. During the first four hours of culture, no significant effect could be attributed to insulin on the activity of these enzymes. Moreover, insulin had no detectable stimulatory effect on phophoglucose isomerase until 48 hours, at which time the hormone caused a marked increase in the activity of this enzyme over the next 24 hours. In contrast, insulin stimulated only a small increase in dehydrogenase activity at 24 hours, after which this hormone acted mainly to maintain the activity that was present initially. These results indicate differential actions of insulin on two groups of enzymes catalyzing the same substrate.  相似文献
