【目的】信息素是个体之间传递信息的重要分子,研究性信息素对斜纹夜蛾 Spodoptera litura 嗅觉相关基因表达的影响对于增加性信息素作用机理的认识及其应用有重要的意义。【方法】本研究通过实时定量PCR(qRT-PCR)技术探究在性信息素刺激处理条件下,斜纹夜蛾成虫嗅觉相关基因 abp, pbp 和 or 表达水平的变化;利用性信息素在田间诱捕斜纹夜蛾雄蛾,并通过自动计数器记录每小时诱虫量,从而间接显示其交配行为的节律性。【结果】斜纹夜蛾雄虫触角中嗅觉相关基因abp, pbp 和 or 的表达具有节律特性。经性信息素化合物(Z9, Z11-14:OAc+Z9, Z12-14:OAc)刺激处理后,abp, pbp 和 or 表达量也发生了显著的改变。通过记录田间性信息素诱捕器在一天中不同时间段内诱捕的雄蛾数量,发现诱捕到的斜纹夜蛾也具有节律特性。【结论】基因表达水平上的节律特性可能与雄虫交配活动的节律相关联,说明性信息素处理也在一定程度上改变了其节律及其对性信息素的神经反应。这一结果也首次从基因水平证明性信息素的刺激处理提高了周缘神经系统对性信息素反应的敏感性,有助于我们理解性信息素作用的分子机理,对迷向及性诱和测报应用具有指导意义。  相似文献

In insects and other animals, intraspecific communication between individuals of the opposite sex is mediated in part by chemical signals called sex pheromones. In most moth species, male moths rely heavily on species-specific sex pheromones emitted by female moths to identify and orient towards an appropriate mating partner among a large number of sympatric insect species. The silkmoth, Bombyx mori, utilizes the simplest possible pheromone system, in which a single pheromone component, (E, Z)-10,12-hexadecadienol (bombykol), is sufficient to elicit full sexual behavior. We have previously shown that the sex pheromone receptor BmOR1 mediates specific detection of bombykol in the antennae of male silkmoths. However, it is unclear whether the sex pheromone receptor is the minimally sufficient determination factor that triggers initiation of orientation behavior towards a potential mate. Using transgenic silkmoths expressing the sex pheromone receptor PxOR1 of the diamondback moth Plutella xylostella in BmOR1-expressing neurons, we show that the selectivity of the sex pheromone receptor determines the chemical response specificity of sexual behavior in the silkmoth. Bombykol receptor neurons expressing PxOR1 responded to its specific ligand, (Z)-11-hexadecenal (Z11-16:Ald), in a dose-dependent manner. Male moths expressing PxOR1 exhibited typical pheromone orientation behavior and copulation attempts in response to Z11-16:Ald and to females of P. xylostella. Transformation of the bombykol receptor neurons had no effect on their projections in the antennal lobe. These results indicate that activation of bombykol receptor neurons alone is sufficient to trigger full sexual behavior. Thus, a single gene defines behavioral selectivity in sex pheromone communication in the silkmoth. Our findings show that a single molecular determinant can not only function as a modulator of behavior but also as an all-or-nothing initiator of a complex species-specific behavioral sequence.  相似文献

为提高现有性信息素对雄蛾的引诱活性, 本研究通过大量的田间试验探索植物气味化合物与斜纹夜蛾Spodoptera litura性信息素(顺9, 反11-十四碳二烯乙酸酯∶顺9, 反12-十四碳二烯乙酸酯=10∶1)的协同作用机制。从斜纹夜蛾寄主植物和花的气味化合物中, 选择9种有代表性的化合物, 并以一定剂量分别加入到斜纹夜蛾性信息素诱芯中, 在田间测试对雄蛾的引诱活性。结果表明: 在测试的9种植源性化合物中, 发现一定剂量(每个诱芯加入0.4 mg)的苯乙醛(PAA), 具有显著提高斜纹夜蛾性信息素的引诱作用, 而高剂量的苯乙醛则强烈抑制性信息素的引诱活性; 此外, 其他各种浓度的测试化合物或混合物对性信息素则没有统计上显著的增效作用。不同剂量的苯乙醛单个化合物及各种植物气味化合物组成的混合物对斜纹夜蛾也有微弱的引诱作用。苯乙醛必须要与性信息素的完整组分(以10∶1比例混合的顺9, 反11-十四碳二烯乙酸酯和顺9反, 12-十四碳二烯乙酸酯)混合才能起作用, 缺少顺9, 反12-十四碳二烯乙酸酯则没有引诱活性。本研究证明, 苯乙醛作为理想的性信息素诱芯增效剂, 可应用于建立更理想的斜纹夜蛾性信息素诱杀技术, 对性诱害虫防治和测报具有应用价值。  相似文献

Pheromones play pivotal roles in the reproductive behavior of moths, most prominently for the mate finding of male moths. Accordingly, the molecular basis for the detection of female‐released pheromones by male moths has been studied in great detail. In contrast, little is known about how females can detect pheromone components released by themselves or by conspecifics. In this study, we assessed the antenna of female Heliothis virescens for elements of pheromone detection. In accordance with previous findings that female antennae respond to the sex pheromone component (Z)‐9‐tetradecenal, we identified olfactory sensory neurons that express its cognate receptor, the receptor type HR6. All HR6 cells coexpressed the “sensory neuron membrane protein 1” (SNMP1) and were associated with supporting cells expressing the pheromone‐binding proteins PBP1 and PBP2. These features are reminiscent to male antennae and point to congruent mechanisms for pheromone detection in the two sexes. Further analysis of the SNMP1‐expressing cells revealed a higher number in females compared to males. Moreover, in females, the SNMP1 neurons were arranged in clusters, which project their dendrites into a common sensillum, whereas in males there were only solitary SNMP1‐neurons and only 1 per sensillum. Not all SNMP1 positive cells in female antennae expressed HR6 but instead the putative pheromone receptors HR11 and HR18, respectively. Neurons expressing 1 of the 3 receptor types were assigned to different sensilla. Together the data indicate that on the antenna of females, sensory neurons in a subset of sensilla trichodea are equipped with molecular elements, which render them responsive to pheromones.  相似文献

环境因素影响昆虫两性间的化学通讯,也影响性信息素技术的田间防治效果.本文探讨了温度对二化螟雌蛾性信息素产生以及雄蛾对性信息素触角电位反应的影响,以期为田间二化螟的性信息素防治提供指导.在二化螟蛹期和成虫期进行不同温度处理(15、20、25、30和35 ℃),然后利用气相色谱仪(GC)分析雌蛾性腺内各性信息素组分的含量及比例,同时利用触角电位仪(EAG)测定雄蛾对性信息素组分的电生理反应.结果表明: 25 ℃处理中雌蛾性腺内3个性信息素组分(Z9-16:Ald、Z11-16:Ald和Z13-18:Ald)的含量均显著高于其他温度处理(15、20和30 ℃),且25 ℃处理中Z13-18:Ald的相对比例也显著低于其他温度处理.就雄蛾对性信息素的敏感性而言,对3种性信息素单一组分及特定比例混合物的EAG反应在15~25 ℃间没有显著差异,但在25~35 ℃间(Z13-18:Ald在30~35 ℃间)随温度升高呈下降趋势,且30 ℃较25 ℃显著降低,35 ℃较30 ℃又显著降低.综合分析认为,二化螟性信息素通讯的适宜温度为20~25 ℃,温度过高或过低均不利于二化螟两性间的正常化学通讯.研究结果为二化螟性信息素防治技术的合理应用及极端温度条件下害虫种群发生的预测预报,提供了重要参考.  相似文献

Mating in moths is generally mediated by female-produced sex pheromones. Mating activity, female pheromone production/release and male pheromone responsiveness all show diurnal variations in many species. We found that the response of the male Egyptian cotton leafworm, Spodoptera littoralis, to sex pheromone gland extracts showed a diel rhythm in olfactometer tests, and the variation was persistent for at least 1 day in constant darkness. High male response to sex pheromone was correlated in time with high mating and locomotor activity. Male S. littoralis, maintained in constant darkness and exposed to pheromone gland extracts on a daily basis, showed an induced temporal variation in response after several days, in contrast to unexposed males. This suggests that in the absence of other external zeitgebers, exposure to sex pheromone may function to synchronise circadian behavioural rhythms in male moths. The daily rhythm in mating activity in S. littoralis is also shown to be persistent for at least 2 days in constant darkness. Pairs mated significantly less when either the male or female had been raised in a light:dark cycle 10 h out of phase, indicating that the proposed circadian rhythm in mating activity is composed of rhythmic mating preference/ability in both sexes.  相似文献

Female moths produce blends of odorant chemicals, called pheromones. These precise chemical mixtures both attract males and elicit appropriate mating behaviors. To locate females, male moths must rapidly detect changes in environmental pheromone concentration. Therefore, the regulation of pheromone concentration within antennae, their chief organ of smell, is important. We describe antennal-specific aldehyde oxidases from the moths Antheraea polyphemus and Bombyx mori that are capable of catabolizing long chain, unsaturated aldehydes such as their aldehyde pheromones. These soluble enzymes are associated uniquely with male and female antennae and have molecular masses of 175 and 130 kDa, respectively. The A. polyphemus aldehyde oxidase has been localized to the olfactory sensilla which contain the pheromone receptor cell dendrites. These same sensilla contain a previously described sensilla-specific esterase that degrades the acetate ester component of A. polyphemus pheromone. We propose that sensillar pheromone-degrading enzymes modulate pheromone concentration in the receptor space and hence play a dynamic role in the pheromone-mediated reproductive behaviors of these animals.  相似文献

蛾类昆虫性信息素受体首先从烟芽夜蛾Heliothis virescens和家蚕Bombyx mori中鉴定出来, 到目前为止已经克隆得到了19种蛾类昆虫的几十种性信息素受体基因, 并且这些基因在系统发育树中聚成一个亚群。性信息素受体从蛾类蛹期开始表达, 主要表达在雄性触角的毛形感器中, 少部分受体在雌性触角、 雄性触角其他感器以及身体其他部位中也有表达。大部分蛾类性信息素受体的配体并不是单一的, 而是能够对多种性信息素组分有反应, 部分性信息素受体还能够识别性信息素以外的其他物质, 还有一部分性信息素受体的识别配体目前尚不清楚。另外发现在雌性蛾类触角中也存在一些嗅觉受体能够识别雄性分泌的性信息素。在蛾类性信息素受体与性信息素识别的过程中, 性信息素结合蛋白不仅能够特异性地运送配体到嗅觉神经元树状突上, 还能够提高性信息素与性信息素受体之间的结合效率。另外, OrCo类受体与性信息素受体共表达在嗅觉神经元中, 在蛾类性信息素受体与配体的识别过程中扮演了重要角色。但是蛾类信息素对神经元刺激的终止并非由性信息素受体控制, 而是由细胞中的气味降解酶等其他因子调控。蛾类性信息素受体研究中还有很多疑问需要解答, 其过程可能比我们想象的更为复杂。  相似文献

Background

Female sex pheromones attracting mating partners over long distances are a major determinant of reproductive isolation and speciation in Lepidoptera. Males can also produce sex pheromones but their study, particularly in butterflies, has received little attention. A detailed comparison of sex pheromones in male butterflies with those of female moths would reveal patterns of conservation versus novelty in the associated behaviours, biosynthetic pathways, compounds, scent-releasing structures and receiving systems. Here we assess whether the African butterfly Bicyclus anynana, for which genetic, genomic, phylogenetic, ecological and ethological tools are available, represents a relevant model to contribute to such comparative studies.

Methodology/Principal Findings

Using a multidisciplinary approach, we determined the chemical composition of the male sex pheromone (MSP) in the African butterfly B. anynana, and demonstrated its behavioural activity. First, we identified three compounds forming the presumptive MSP, namely (Z)-9-tetradecenol (Z9-14:OH), hexadecanal (16:Ald ) and 6,10,14-trimethylpentadecan-2-ol (6,10,14-trime-15-2-ol), and produced by the male secondary sexual structures, the androconia. Second, we described the male courtship sequence and found that males with artificially reduced amounts of MSP have a reduced mating success in semi-field conditions. Finally, we could restore the mating success of these males by perfuming them with the synthetic MSP.

Conclusions/Significance

This study provides one of the first integrative analyses of a MSP in butterflies. The toolkit it has developed will enable the investigation of the type of information about male quality that is conveyed by the MSP in intraspecific communication. Interestingly, the chemical structure of B. anynana MSP is similar to some sex pheromones of female moths making a direct comparison of pheromone biosynthesis between male butterflies and female moths relevant to future research. Such a comparison will in turn contribute to understanding the evolution of sex pheromone production and reception in butterflies.  相似文献

Background

In the course of evolution butterflies and moths developed two different reproductive behaviors. Whereas butterflies rely on visual stimuli for mate location, moths use the ‘female calling plus male seduction’ system, in which females release long-range sex pheromones to attract conspecific males. There are few exceptions from this pattern but in all cases known female moths possess sex pheromone glands which apparently have been lost in female butterflies. In the day-flying moth family Castniidae (“butterfly-moths”), which includes some important crop pests, no pheromones have been found so far.

Methodology/Principal Findings

Using a multidisciplinary approach we described the steps involved in the courtship of P. archon, showing that visual cues are the only ones used for mate location; showed that the morphology and fine structure of the antennae of this moth are strikingly similar to those of butterflies, with male sensilla apparently not suited to detect female-released long range pheromones; showed that its females lack pheromone-producing glands, and identified three compounds as putative male sex pheromone (MSP) components of P. archon, released from the proximal halves of male forewings and hindwings.

Conclusions/Significance

This study provides evidence for the first time in Lepidoptera that females of a moth do not produce any pheromone to attract males, and that mate location is achieved only visually by patrolling males, which may release a pheromone at short distance, putatively a mixture of Z,E-farnesal, E,E-farnesal, and (E,Z)-2,13-octadecadienol. The outlined behavior, long thought to be unique to butterflies, is likely to be widespread in Castniidae implying a novel, unparalleled butterfly-like reproductive behavior in moths. This will also have practical implications in applied entomology since it signifies that the monitoring/control of castniid pests should not be based on the use of female-produced pheromones, as it is usually done in many moths.  相似文献

ABSTRACT. The acoustic properties of the clicks emitted in response to male courtship pheromone by female Pyrrharctia Isabella (J. E. Smith) (Lepidoptera: Arctiidae) have been investigated. Extracts of male scent organs or synthetic male pheromones can be applied to a glass rod and used to stimulate females to produce these sounds. Power spectra, sound pressure readings, and oscillographic analyses show that the acoustic signals elicited by male extracts or synthetic male pheromones are not distinguishable from those produced in response to disturbance (handling). This is the first reported example of a sound produced by a female moth in a sexual context, and is the first reported example in moths of an acoustic response to a pheromonal stimulus.  相似文献

Previous research has revealed that natural and synthetic pheromones can enhance ratings of opposite sex attractiveness. The present study investigated the effects of exposure to male axillary secretions on female ratings of the sexual attractiveness of male stimuli. Thirty-two female undergraduates, half of whom were contraceptive pill users, rated male vignette characters and photographs of male faces on aspects of attractiveness. On two separate study days, corresponding to different phases of their menstrual cycle, stimuli were presented while exposed to male axillary pheromones and under a control condition (no pheromone). The order of testing was balanced with respect to pheromone/control condition and menstrual cycle phase. Pheromone exposure resulted in significantly higher attractiveness ratings of vignette characters and faces. Use of the contraceptive pill or menstrual cycle phase had equivocal effects on some vignette items and neither had any influence on female ratings of male facial attractiveness. The results of this study suggest that exposure to natural male axillary pheromones can significantly enhance female perceptions of various aspects of male attractiveness.  相似文献

Electroantennogram (EAG) measurements were recorded from the antennae of male and female codling moth, Cydia pomonella L., to determine whether adult moths exposed to surfaces treated with the ecdysteroid agonist methoxyfenozide experience a decline in their antennal reception and thus olfactory sensitivity. Such a phenomenon would offer a possible mechanism for the previously reported decreased responsiveness from moths treated with methoxyfenozide to pheromone‐ and plant volatile‐based monitoring lures. Mean EAG data revealed that the antennae from methoxyfenozide‐treated male moths appear to be just as sensitive to various doses of synthetic codlemone as the antennae from the control and surfactant‐treated moths, but they appeared to be less sensitive to the pheromone component 12OH (collected from female effluvia) than the control male antennae. Mean male EAG responses to the pheromone components E8,E10‐12Al and codlemone collected from methoxyfenozide‐treated females were significantly less than the responses towards those two pheromone components collected from the control and surfactant‐treated females. Female moth exposure to methoxyfenozide did not negatively impact the sensitivity of female antennae to the plant volatile pear ester, but it did towards the apple volatile butyl hexanoate. Data from this study show that adult C. pomonella exposure to methoxyfenozide‐treated surfaces does appear to negatively impact, in a minor way, the (i) olfactory sensitivity (or detection) of male antennae towards some components of the female sex pheromone, (ii) the female antennal sensitivity towards a key apple volatile and (iii) the attractiveness of female pheromone effluvia.  相似文献

【目的】研究旨在深入探讨中国黄杨绢野Diaphania perspectalis 的雌蛾性信息素组成及诱蛾活性。【方法】利用气质联用仪(GC-MS)对黄杨绢野螟正常型性成熟雌蛾的性腺体提取物和合成标样比较分析,并用反-11-十六碳烯醛(E11-16:Ald) 、顺-11-十六碳烯醛(Z-11-16:Ald)、顺-9-十六碳烯醛(Z-9-16:Ald)、顺-11-十六碳烯醇(Z-11-16:OH)等物质进行触角的电生理测定,最后开展田间诱集比较试验以筛选出最佳性信息素混合物。【结果】Z-11-16:Ald为中国黄杨绢野螟正常型性信息素主要组分,E-11-16:Ald的含量极低,Z-11-16:OH未检测到。正常型雄性黄杨绢野螟触角对Z-11-16:Ald, E-11-16:Ald, Z-9-16:Ald和Z-11-16:OH产生强烈的EAG反应,并随着浓度的提高而显著增加;而对Z-11-16:Ac和E-11-16:Ac的嗅觉反应较弱,低于对植物绿叶气味顺3-己烯乙酸酯(Z-3-6:Ac)的反应。单一Z-1-16:Ald对正常型雄性黄杨绢野螟具有强烈的诱集效果,加入E-11-16:Ald有一定的增效作用,但在统计上则不显著。单一Z-11-16:Ald组分对黑化型雄性黄杨绢野螟无引诱活性,必需加入一定比例的E-11-16:Ald才显示诱蛾活性。Z-11-16:Ald:E-11-16:Ald的比例为250 μg:250 μg时诱集到的黑化型雄性黄杨绢野螟数量最多,而Z-11-16:Ald:E-11-16:Ald的比例为429 μg:71 μg时则诱集到的正常型雄性黄杨绢野螟数量最多。同时,单一Z-11-16:Ald也可引诱大量雄性粘虫Mythimna separata,但E-11-16:Ald抑制其活性。【结论】中国黄杨绢野螟的性信息素主成分是Z-11-16:Ald,单一组分即可在田间强烈引诱雄蛾,E-11-16:Ald的功能只起到微弱的增效作用,但也可能起种的专一性的作用。正常型与黑化型黄杨绢野螟对性信息素的嗅觉反应存在差异,黑化型黄杨绢野螟的性信息素接近日本种,即性信息素组成为Z-11-16:Ald和E-11-16:Ald 的混合物,其比例为1:1,且E-11-16:Ald为必需。  相似文献

Background

Tuning of the olfactory system of male moths to conspecific female sex pheromones is crucial for correct species recognition; however, little is known about the genetic changes that drive speciation in this system. Moths of the genus Ostrinia are good models to elucidate this question, since significant differences in pheromone blends are observed within and among species. Odorant receptors (ORs) play a critical role in recognition of female sex pheromones; eight types of OR genes expressed in male antennae were previously reported in Ostrinia moths.

Methodology/Principal Findings

We screened an O. nubilalis bacterial artificial chromosome (BAC) library by PCR, and constructed three contigs from isolated clones containing the reported OR genes. Fluorescence in situ hybridization (FISH) analysis using these clones as probes demonstrated that the largest contig, which contained eight OR genes, was located on the Z chromosome; two others harboring two and one OR genes were found on two autosomes. Sequence determination of BAC clones revealed the Z-linked OR genes were closely related and tandemly arrayed; moreover, four of them shared 181-bp direct repeats spanning exon 7 and intron 7.

Conclusions/Significance

This is the first report of tandemly arrayed sex pheromone receptor genes in Lepidoptera. The localization of an OR gene cluster on the Z chromosome agrees with previous findings for a Z-linked locus responsible for O. nubilalis male behavioral response to sex pheromone. The 181-bp direct repeats might enhance gene duplications by unequal crossovers. An autosomal locus responsible for male response to sex pheromone in Heliothis virescens and H. subflexa was recently reported to contain at least four OR genes. Taken together, these findings support the hypothesis that generation of additional copies of OR genes can increase the potential for male moths to acquire altered specificity for pheromone components, and accordingly, facilitate differentiation of sex pheromones.  相似文献

Many species of female moths produce sex pheromones to attract conspecific males. To date, sex pheromones from more than 570 moth species have been chemically identified. Most moth species utilize Type I pheromones that consist of straight-chain compounds 10-18 carbons in length with a functional group of a primary alcohol, aldehyde, or acetate ester and usually with several double bonds. In contrast, some moth species use unsaturated hydrocarbons or hydrocarbon epoxides, classified as Type II lepidopteran pheromones, as sex pheromones. Studies over the past three decades have demonstrated that female moths usually produce sex pheromones as multi-component blends where the ratio of the individual components is precisely controlled, thus making it possible to generate species-specific pheromone blends. As for the biosynthesis of Type I pheromones, it is well established that they are de novo synthesized in the pheromone gland (PG) through modifications of fatty acid biosynthetic pathways. However, as many of the molecular components within the PG cells (i.e., enzymes, proteins, and small regulatory molecules) have not been functionally characterized, the molecular mechanisms underlying sex pheromone production in PG cells remain poorly understood. To address this, we have recently characterized some of the molecules involved in the biosynthesis of the sex pheromone bombykol in the silkmoth, Bombyx mori. Characterization of these, and other, key molecules will facilitate our understanding of the precise mechanisms underlying lepidopteran sex pheromone production.  相似文献

Studies were conducted with codling moth, Cydia pomonella L., to evaluate the mating status of male and female moths in apple, Malus domestica (Borkhausen), orchards treated with and without sex pheromone dispensers. Laboratory studies first examined the effect of multiple mating of male and female moths on female fecundity and egg fertility. Females that had mated three times had a significantly higher fecundity than singly mated moths. Sequential mating by male moths had no effect on the fecundity of female moths or egg fertility. However, male moth age did impact female fecundity, with significantly fewer eggs laid after mating with virgin 1- versus 3-d-old males. The mean size of the first spermatophore transferred by males was significantly larger than all subsequent spermatophores. Classifying spermatophores based on size was used in field sampling to categorize the mating status of the female's partner. The proportion of mated females with small spermatophores (partner had previously mated) was significantly higher in treated versus untreated orchards. 蛾类通过产生和识别物种特异性性信息素来引发后续交配行为,因此它在两性交配行为中至关重要.它们具有不同碳链长度、末端官能团、不同双键位置和构型等化学结构特征,本文详细讨论了不同蛾类性信息素的合成途径以及催化每一步反应的相关酶系,列举了15种夜蛾科不同亚科常见物种的性信息素组分及其比例,总结了产生特定比例性信息素的可能原因,查阅了夜蛾科不同物种已经鉴别的性信息素,并按照不同亚科、不同官能团和碳链长度对其进行分类,归纳了同一物种及其亲缘物种性信息素组分和比例的变异,总结了产生变异的分子机理,讨论了性信息素变异和物种进化的关系.最后以生物合成激活神经肽(PBAN)为主, 介绍了其调控途径和机制.本文旨在以不同的蛾类性信息素合成途径为线索,从共有合成途径出发深入了解其规律和共性,从特异合成途径出发探究物种间的进化和变异,展望未来的研究方向及其应用.  相似文献
