Telocytes (TCs) are a new type of interstitial cells, a small cellular body with the presence of 2–5 prolongations named as telopode (Tp)‐very thin (less than 0.2 µm) and extremely long (10–1,000 µm), a moniliform aspect, and caveolae, containing a nucleus surrounded by a small amount of cytoplasm. The nucleus occupies about 25% of TC body volume and contains clusters of heterochromatin attached to the nuclear envelope. The perinuclear cytoplasm is rich in mitochondria and contains a small Golgi complex, rough and smooth endoplasmic reticulum and cytoskeletal elements. TCs have several immunophenotypes such as CD34, c‐kit, and vimentin. TCs were found in many organs of mammals with potential biological functions, even though the exact function remains unclear. Recently, we identified and isolated TCs from the trachea for the first time and confirmed the existence of TC in lung tissues, which could have the potential significance in the pathogenesis of pulmonary diseases. The nucleus rotundus of 21 species of teleosts was studied by a modified Bodian and the Golgi method to clarify the histological organization, with special reference to the cell lamination and the glomerular formation. The common components of the nucleus in all species are as follows: a thick fiber bundle which comes from the commissura horizontalis and enters the nucleus from the dorsal surface, many small cells, large cells, glomeruli, and a surrounding fibrous capsule. The nuclei of all species studied are classified into three types mainly on the distribution of the small cells, and to a lesser degree on the location of the large cells and the glomeruli. The first type of nucleus has small cells, large cells and glomeruli throughout its extent. In the second type of nucleus, many small cells form a peripheral cell layer, while the large cells and glomeruli are found all over the nucleus. The third type of nucleus is clearly laminated. It is composed of four layers arranged concentrically around a central fiber net in the following order: a glomerular layer, a fibrous layer, a small-cell layer, and a peripheral fibrous capsule. In some species, the large cells are located in the fibrous capsule, and all glomeruli contain a star-like structure, which corresponds to the tips of the large cell dendrites.  相似文献

Histochemical study of neuronally isolated area AI of the auditory cortex in cats by the reaction for acetylcholinesterase 3 days and 1, 2, and 3 weeks after undercutting showed that the cholinergic neuropil of this area is formed mainly by incoming fibers and to a lesser degree by processes from a few intrinsic cholinergic neurons. The intrinsic cholinergic neurons include, first, cholinergic long-axon association neurons responding to cortical isolation by retrograde changes and by hyperreaction to acetylcholinesterase (Cajal-Retzius cells of layer I and neurons of layer VI, whose axons run into the subcortical layer of association fibers), and, second, cholinergic short-axon association neurons of layers II–VI, preserving their normal cell structure and moderate acetylcholinesterase activity after isolation. Axon collaterals of similar cells terminate on neighboring neurons. Short-axon neurons are more numerous in the lower layers of the cortex, and exceed in number the long-axon association neurons. Choliniceptive neurons (pyramidal and stellate), on whose bodies and proximal dendrites are located terminals formed by axons of cholinergic association neurons, are found in the isolated cortex. The concentrations of 5-hydroxytryptamine (5HT), noradrenaline, and dopamine were estimated post mortem in brain stem, hypothalamus, and caudate nucleus in 33 patients who had been treated with isocarboxazid, clorgyline, or tranylcypromine and 11 controls. Similar and highly significant increases in 5HT and noradrenaline concentration occurred with all three drugs. The distribution was unimodal, but about a quarter of the patients showed only a small increase in brain amines. Tranylcypromine seemed to have a significantly greater effect on dopamine in caudate nucleus and hypothalamus compared with isocarboxazid and clorgyline. In the doses used chlorpromazine did not reduce the amine concentrations. Four patients with Parkinson''s syndrome had low concentrations of dopamine in caudate nucleus in spite of monoamine oxidase inhibitor administration.  相似文献

Carbon nanotube (CNTs) is a new alternative for efficient drug delivery and it has a great potential to change drug delivery system profile in pharmaceutical industry. One of the important advantage of CNTs is their needle-like, cylindrical shape. This shape provides a high surface area for multiple connections and adsorption onto for millions of therapeutic molecules. CNTs can be internalized by cells via endocytosis, passive diffusion and phagocytosis and release the drug with different effects like pH and temperature. The acidic nature of cancer cells and the susceptibility of CNTs to release the drug in the acidic environment have made it a promising area of research in cancer drug delivery. In this research, we investigated cell viability, cytotoxicity and drug delivery in breast cancer cell line by designing non-covalent single walled carbon nanotubes (SWNT)–doxorubicin (DOX) supramolecular complex that can be developed for cancer therapy. Applied high concentrations of DOX loaded SWNTs changed the actin structure of the cells and prevented the proliferation of the cells. It was showed that doxorubicin loaded SWNTs were more effective than free doxorubicin at relatively small concentrations. Once we applied same procedure for short and long (short: 1–1.3 µm; long: 2.5–4 µm) SWNTs and compared the results, more disrupted cell structure and reduction in cell proliferation were observed for long CNTs. DOX is bounded more to nanotubes in basic medium, less bound in acidic environment. Cancer cells were also examined for concentration at which they were effective by applying DOX and it was seen that 3.68 µM doxorubicin kills more than 55% of the cells.  相似文献

Abstract: Atrophy with ageing of human whole brain, entire temporal lobe, and caudate nucleus was assessed in autopsy specimens, by biochemical techniques. Only the caudate nucleus showed changes. Markers for several neurotransmitter systems were also examined for changes with age. In neocortex and temporal lobe of human brain, small decreases were detected in markers of cholinergic nerve terminals, whereas a large decrease (79%) occurred in the caudate nucleus. Findings were similar in striatum from 3–33-month-old rats. No change occurred in binding of [3H]quinuclidinyl benzilate by human samples. Markers of serotonergic terminals were also unchanged in human and rat brain. By contrast, binding of [3H]lysergic acid diethylamide and [3H]serotonin was decreased (32–81%) in human neocortex and temporal lobe, but not in caudate nucleus. A 43% loss of a marker of γ-aminobutyrate terminals occurred in human neocortex, while [3H]muscimol binding increased (179%). No changes were detected in markers of catecholamine synapses in temporal lobe or rat striatum. Hence, with human ageing there appears to be a loss of markers of γ-aminobutyrate neurones intrinsic to neocortex and acetylcholine cells intrinsic to the caudate nucleus, as well as a change in postsynaptic serotonin receptors in neocortex. These losses are accompanied by relative preservation of markers of ascending projections from basal forebrain and brain stem.  相似文献

Perinuclear organelles are found in the sensory cell of the Grandry cutaneous corpuscle in the duck. They are ovoid, fusiform or conical. They measure up to 5 µ length and l µ in diameter. They are formed by regular alternation of granular layers (mornolayers of RNA-rich granules which can be interpreted as ribosomes) and fibrous layers (generally formed by two sublayers of parallel fibrils). These fibrils (60-80 Å diameter) are in continuity with the intra-cytoplasmic fibrils which are very abundant in the Grandry cell. The central part of the organelles is devoid of RNA-rich granules.The formation of these organelles begins about one week after hatching, in the cytoplasmic perinuclear area where abundant granular endoplasmic reticulum and very numerous ribosomes and fibrils are present. The function of these perinuclear organelles remains unknown.  相似文献

Ultrastructural 3-D analysis of nucleolar architecture and Ag-NOR protein distribution in mouse kidney-cortex proximal-tubule epithelium has been performed. A principal scheme of structural changes of the nucleolus and organization of its components during the intensification of pre-rRNA synthesis (dynamic model of a nucleolus) based on computer spatial modelling has been advanced. According to the nucleolar composition, three groups of cells, which differ from each other by rRNA synthesis, are defined in normal kidney. Most nephron proximal-section cells (about 52%) are characterized by lower activity of RNA synthesis. Such kind of cells are defined as group I (nucleolar diameter 0.7–1.5 µm) and always contain resting, ring-shaped or close to ring-shaped dense nucleoli, which have 2 or 3 fibrillar centers. Nucleoli of group II cells (about 37%, nucleolar diameter 1.5–2.5 µm) have a higher level of activity, contain 4–7 fibrillar centers, and their structural organization is close to reticulated forms due to the first indications of vacuolar network (identified as prereticulated nucleoli). The most active cells of group III (about 11%, nucleolar diameter 2.5–3.5 µm) include cells with typical reticulated nucleoli with a well expressed vacuolar network and numerous fibrillar centers (18–22). Increased functional load of the epithelium caused by unilateral nephrectomy and diuretic (4-chlor-H [2-furylmethyl] 5-sulphamyl-antranic acid) injection changed the proportion of the different cell groups: group I decreased (about 25%), whereas groups II and III increased (about 8% and 17%, respectively). The increase of nucleolar activity first causes a deformation of the individual fibrillar centers as well as complication and growth of their surface. Further, a progressive fragmentation of the fibrillar centers and the growth of their total volume is observed. The complication and growth of the total volume of Ag-positive zones is another indication of the nucleolar activation. The vacuolar system develops by a gradual fusion of small isolated cavities into a united vacuolar network. Nucleoli with 2–7 fibrillar centers are considered to be intermediate forms reflecting successive stages of its activation or inactivation: from the resting ring-shaped nucleolus via transient stages of increasing functional activity to the active reticulated nucleoli and vice versa. The observed differences in the nucleolar ultrastructure are regarded as evidence of the functional heterogeneity of cell populations within one functional segment of nephron.  相似文献

Optic axons were labelled with horseradish peroxidase to establish the presence of side branches and examine their distribution and morphology in the developing optic nerve of the quokka wallaby, Setonix brachyurus, the cat and rat at stages when axon numbers are at their peak. In each species, three quarters of the axons were essentially straight and lacked side branches. The remaining axons took significantly longer paths and bore side branches, mostly at points where axons undulated or changed direction. Side branches occurred at intervals of 28–43 µm, had lengths of 2–3 µm and were usually simple rather than branched. A minority (1%) of the axons crossed diagonally between fascicles and two thirds of these had more side branches (interval: 10–18 µm) on the interfascicular portion than were found on the forward-directed axons. A small number of axons (0.01%) doubled back to grow retrogradely towards the eye, these axons also bore relatively more side branches (interval: 8–22 µm), especially at points where the axons changed direction. Ultrastructural reconstruction showed that side branches resembled small axonal profiles and constituted 2% of the total axon number. It is suggested that side branches are involved in the fine-tuning of growth cone navigation. Most side branches are lost by adulthood, indicating their transient nature. The absence of retrogradely-directed axons from adults suggests that cells with such axons are removed by naturally occurring cell death.  相似文献

When studying the neuronal organization of the large-cell part of the red nucleus in cats by the Golgi and Golgi-Kopsch methods three types of neurons have been revealed: large (50-90 mu), medium-sized (20-50 mu) and small cells (8-20 mu). long axon and short-axon neurons were found as well as long-dendrite cells with few thorns (the length of the dendrites form 600-900 mu) and short-dendrite cells (up to 400 mu). On basis ofanalysis of neuronal groups found in the large-cell part of the red nucleus of the cat a neuronal map of this part of the central nervous sytem is composed.  相似文献

Three species of the red algal genus Herposiphonia (Ceramiales, Rhodomelaceae) found in Japan are described, and taxonomic features of the genus are discussed. Herposiphonia crassa Hollenberg is reported from Japan for the first time and is characterized by thick axes (200–350 µm in diameter) and determinate branches (100–200 µm in diameter), relatively short determinate laterals (400–1200 µm in length) with a large number of periaxial cells (15–19 per segment) and three (occasionally two or four) vigorously developed (1.8–2.5 mm in length by 50–75 µm in diameter basally) trichoblasts on each determinate lateral. Herposiphonia elongata Masuda et Kogame is also reported from Japan for the first time and is characterized by the conspicuous thickening growth of cystocarp‐bearing branches and spermatangial branches with an elongated sterile tip. Some newly found features of Herposiphonia fissidentoides (Holmes) Okamura are presented: the rhizoid production from the central portion of parental periaxial cells in addition to the distal end, virtual absence of vegetative trichoblasts, production of procarpial trichoblasts and spermatangial branches on fertile determinate branches on short indeterminate laterals, cystocarps sometimes with a short spur, and extremely large tetrasporangia.  相似文献

In this study, we have estimated the number of total mortality (T-mortality), cardiovascular morbidity (CV-mortality), respiratory mortality (R-mortality), hospital admissions due to cardiovascular diseases (HA-CVD), respiratory diseases (HA-RD), chronic obstructive pulmonary diseases (COPD) and acute myocardial infarction (AMI) due to exposure to particulate matter less than 10 µm (PM10), 2.5 µm (PM2.5) and sulfur dioxide (SO2) in western Iran in 2016. The World Health Organization (WHO) method was used to assess the mortality and morbidity among the exposed people. The results showed that about 3.9% CM (95% CI: 2.9–7.8%), 3.9% HA-RD (95% CI: 2.4–7.8%) and 4.4% HA-CVD (95% CI: 3.0–6.8%) for ambient PM10 and about 7.3% TM (95% CI: 4.2–9.7%), 12.1% CM (95% CI: 3.5–14.6%) and 3.0% RM (95% CI: 0–6.3%) for PM2.5 are respectively attributed to concentrations exceeding 10 µg/m3. Furthermore, 3.2% HA-COPD (95% CI: 0–5.04%) and 4.2% AMI (95% CI: 1.6–4.3%) can be attributed to SO2 concentrations greater than 10 µg/m3, respectively. To reduce the adverse health effect of PM, health advices provided by health authorities should be given to general population especially vulnerable people such as people with chronic lung and heart pathologies, elderly and children during the dusty days.  相似文献

A desktop incubator with temperature control over the range 1–20°C (±0.5°C) was designed to hold two microtitre plates. Illumination of individual wells in the plate was by a matrix of 96 light-emitting diodes, whose intensity, period and pulsation could be controlled individually in each of 12 rows of eight chambers. The incubator was used to test how the length of light period affected cell length of the planktonic diatom Aulacoseira baicalensis, which only grows below 4°C. Short cells (mean length 26 µm) were formed under a 8-h : 16-h light–dark cycle, intermediate cells (mean length 35 µm) under a 4-h : 20-h light–dark cycle and longer cells (mean 38 µm) under a 2-h : 22-h light–dark cycle. These were similar to changes in mean cell length from 28 to 49 µm found in Lake Baikal during a decrease in light period caused by increased convective mixing from 15 m under-ice to 94 m after ice break-up. The laboratory experiments confirmed that decreasing light period was the environmental cue that initiated the production of long cells that then developed into resting stages.  相似文献

We examined faeces of 76 endangered Tibetan antelopes Pantholops hodgsonii (Abel) in May 2017, from the Hoh Xil Nature Reserve, Qinghai Province, China, and found 62/76 (82%) discharging oöcysts representing five new species of Eimeria Schneider, 1875. Oöcysts of Eimeria pantholopensis n. sp., found in 54/76 (71%) chiru, are subspheroidal/ellipsoidal, 15–22 × 12–19 (18.6 × 16.1) µm, with a length/width (L/W) ratio of 1.0–1.3 (1.2); micropyle cap and 1–3 polar granules are present, but oöcyst residuum is absent. Sporocysts are ovoidal, 7–11 × 4–6 (9.2 × 5.3) µm, with a L/W ratio of 1.6–2.0 (1.7); Stieda body and sporocyst residuum of small, scattered granules are present; each sporozoite contains 2 refractile bodies. Oöcysts of Eimeria wudaoliangensis n. sp. found in 52/76 (68%) chiru, are pyriform, 21–29 × 17–21 (24.9 × 19.0) µm, with a L/W ratio of 1.1–1.5 (1.3); micropyle, micropyle cap and 1–4 polar granules are present, but oöcyst residuum is absent. Sporocysts are ovoidal, 9–13 × 5–8 (11.7 × 6.7) µm, with a L/W ratio of 1.4–2.7 (1.7); Stieda body and sporocyst residuum of disbursed granules are present; sporozoites have a single large refractile body. Oöcysts of Eimeria hodgsonii n. sp. found in 20/76 (26%) chiru, are elongate-ellipsoidal, 25–32 × 18–21 (28.9 × 19.8) µm, with a L/W ratio of 1.2–1.7 (1.5); micropyle, micropyle cap and 1–3 polar granules are present, but oöcyst residuum is absent. Sporocysts are ovoidal, 11–14 × 6–7 (12.3 × 6.8) µm, with a L/W ratio of 1.7–2.1 (1.8); Stieda body and sporocyst residuum as group of large granules lying along the interface between intertwined sporozoites are present; sporozoites have 2 refractile bodies. Oöcysts of Eimeria schalleri n. sp. found in 49/76 (64.5%) chiru, are ellipsoidal, 26–36 × 19–25 (30.4 × 23.2) µm, with a L/W ratio of 1.2–1.5 (1.3); micropyle with micropyle cap and polar granules appearing as many diffuse tiny bodies are present, but oöcyst residuum is absent. Sporocysts are ovoidal, 12–16 × 7–9 (14.2 × 7.8) µm, with a L/W ratio of 1.6–2.1 (1.8); Stieda body and sporocyst residuum are present, the latter as a group of small dispersed granules between intertwined sporozoites; sporozoites with 2 refractile bodies. Oöcysts of Eimeria sui n. sp. found in 4/76 (5%) chiru, are ovoidal, 32–38 × 26–30 (36.6 × 28.6) µm, with a L/W ratio of 1.0–1.4 (1.3); micropyle and micropyle cap and 1–3 polar granules are present, but oöcyst residuum is absent. Sporocysts are ovoidal, 15–18 × 8–10 (16.7 × 8.9) µm, with a L/W ratio of 1.7–2.1 (1.9); Stieda body and sporocyst residuum are present, the latter as a group of dispersed small granules; sporozoites with 2 refractile bodies. Five of 62 faecal samples in which oöcysts were detected (8%) had a single species infection, 13 of 62 (21%) had two species, 28 of 62 (45%) had three species and 16 of 62 (26%) had four species.

Abstract— The specific binding of [3H]spiperone and [3H]domperidone, as defined by 1 μ m -(+)butaclamol, was compared in homogenates of bovine retina and caudate nucleus. Scatchard analyses of saturation data for [3H]spiperone binding yielded dissociation constants ( K d) of 0.35 n m in the retina and 0.64 n m in the caudate nucleus. Comparison of the maximum number of binding sites (Bmax) present in each tissue indicated that the density of sites in bovine caudate nucleus (270 fmol/mg protein) was approximately three times higher than in bovine retina (92 fmol/mg protein). This difference was even more marked in guinea pig tissues, with a ratio of 7:1 between corpus striatum and retina. The pharmacological analysis of [3H]spiperone binding in both the bovine retina and caudate nucleus indicated an interaction with dopaminergic rather than serotonergic sites. However, inhibition curves obtained to dopaminergic agonists in the bovine retina were significantly steeper than those observed in the bovine caudate nucleus, as reflected in the greater Hill coefficients obtained for these agents in the retina. Furthermore, only a small amount of specific [3H]domperidone binding was observed in either the bovine caudate nucleus or the guinea pig striatum, whilst no specific [3H]domperidone binding was detectable in homogenates of either bovine or guinea pig retina. These data suggest that the retina possesses only a small population of dopaminergic D2 sites and that these binding sites may differ from those present in the caudate nucleus.  相似文献

The presence of surface and grain boundary defects in organic–inorganic halide perovskite films can be detrimental to both the performance and operational stability of perovskite solar cells (PSCs). Here, the effect of chloride additives is studied on the bulk and surface defects of the mixed cation and halide PSCs. It is found that using an antisolvent technique, the perovskite film is divided into two layers, i.e., a bottom layer with large grains and a thin capping layer with small grains. The addition of formamidinium chloride (FACl) into the precursor solution removes the small‐grained perovskite capping layer and suppresses the formation of bulk and surface defects, providing a perovskite film with enhanced crystallinity and large grain size of over 1 µm. Time‐resolved photoluminescence measurements show longer lifetimes for perovskite films modified by FACl and subsequently passivated by 1‐adamantylamine hydrochloride as compared to the reference sample. Impedance spectroscopy measurements show that these treatments reduce the recombination in the PSCs, leading to a champion device with power conversion efficiency (PCE) of 21.2%, an open circuit voltage of 1152 mV and negligible hysteresis. The Cl treated PSC also shows improved operational stability with only 12% PCE loss after 700 h under continuous illumination.  相似文献

Feces from a specimen of Tamandua tetradactyla (Linn.) from Portel, Para State, north Brazil, contained two different coccidial oocysts; one identified as Eimeria tamanduae Lainson 1968, and the other as a new species, described here as Eimeria corticulata n. sp. Oocysts of E. corticulata are ellipsoidal, 37.4 × 30.4 (31.2–43.7 × 23.7–35.0) μm, shape index (length/width) 1.2 (1.0–1.5). Oocyst wall 2.5–3.7 μm thick and composed of two layers; an outer thick, brown-yellow one with radial striations, and a thin inner smooth one: no visible micropyle. Oocyst residuum a large globule of about 10.7 × 10.3 μm, usually accompanied by a number of smaller attached globules. Sporocysts ellipsoidal, 21.0 × 11.0 (20.0–22.5 × 10.0–12.5) μm, with a conspicuous Stieda body: shape index 1.9 (1.6–2.2). Sporocyst residuum a small number of scattered granules: sporozoites 18.7 × 5.0 μm, with a large posterior refractile body. Eimeria zygodontomyis n. sp. is described in feces from Zygodontomys lasiurus (Lund) from the Serra dos Carajas, Para. Oocysts ellipsoidal to cylindrical, 16.5 × 12.0 (13.7–18.7 × 11.2–12.3) μm, shape index 1.4 (1.2–1.5). Wall colorless, smooth, single-layered and about 0.6 μm thick: no micropyle. No oocyst residuum, but a polar granule of about 1.8 × 1.0 μm is sometimes present. Sporocysts ellipsoidal, 8.4 × 5.5(7.5–8.7 × 5.0–6.2) μm, shape index 1.5 (1.4–1.7), with a thin colorless wall and a delicate Stieda body. Sporozoites enclose a compact residuum of about 2.5 × 3.7 μm.  相似文献
