作者用激光喇曼光谱法分析半乳糖导致大白鼠晶状体混浊过程中构象的变化。通过SPEX 1403型激光喇曼光谱仪得到了正常及不同混浊度晶状体的喇曼光谱。结果表明晶状体可溶性蛋白质二级结构的光谱未见异常,其残基酪氨酸及色氨酸微环境起了变化。随着晶状体混浊度的增加,SH谱峰强度变小而S-S键谱峰增强,同时观察到荧光背景逐渐加强。经分析认为晶状体混浊是与蛋白质分子的聚集有关。  相似文献

激光喇曼光谱技术是一种非侵入、非弹性的光散射技术,它能够无损地提供丰富的分子结构和物质成分的信息。近来它在食品工业领域表现出很大的应用潜力。本文综述了激光喇曼光谱技术在食品科学中的应用及其新进展。主要包括果蔬农药残留的检测、肉类产品质量检测、伪劣食品鉴定、食物蛋白的研究以及食品加工监控等方面的应用。并对喇曼光谱技术在这些方面的应用前景作了进一步的展望。  相似文献

喇曼光谱技术是一种非侵入、非弹性散射技术,能够在分子层次上探测物质的临床医学特征和结构特征。本文综述了近十年来喇曼光谱技术在生物医学中的最新发展,归纳出了四种目前在生物医学中最为活跃的喇曼光谱技术:近红外喇曼光谱、紫外共振喇曼光谱、表面增强喇曼光谱和多维喇曼成像技术。详细阐述了这四种技术的特点和适用范围,并且列举了丰富的成功范例。  相似文献

目的:探讨共振喇曼光谱技术用于早期恶性肿瘤诊断的研究。方法:利用氩离子激光作为线偏振光的特点,采集偏振荧光光谱,对荧光光谱的偏振态进行分析。利用不同荧光物质的荧光可能具有不同偏振态的特点减少其它荧光物质的荧光对光谱分析的影响。血清样品产生的荧光也具有确定的偏振性。对所检测病人血清经激光分析仪进行喇曼光谱技术分析,光谱数据经计算机软件处理,自动显示图谱和数据,并直接给出各项指标及诊断提示。本结果与细胞病理学结果进行了对照研究。结果:恶性肿瘤样本176例,检测出阳性病例141例,阳性符合率为80.1%;良性肿瘤样本52例,4例阳性,假阳性率为7.7%;正常体检样本248例,检测结果均为阴性。结论:喇曼光谱技术适用于肿瘤初筛、普查及早期诊断,有推广应用前途。  相似文献

用改进的固相磷酰三酯法合成了oligo-d(G-C)_3。以氩离子激光为激发光源,波长488nm.,在室温条件下,分别测定了纯化后的oligo-d(G-C)_3和其组分单体5’-dGMP和5’-dCMP的激光喇曼谱。观察到被测定的物质在300-2500cm~(-1)频率区间,各自都有其特征的谱形和喇曼峰。5’-dGMP和5’-dCMP谱中大多数特征峰在寡聚体的谱中消失,而在oligo-d(G-C)_3谱中出现了几处新的喇曼峰。经查证,峰832,851和899cm~(-1)系糖-磷酸主链的特征喇曼峰,另外几处峰与DNA的构象有关。实验结果表明oligo-d(G-C)_3在水溶液中(室温)主要以B-构象存在。  相似文献

本文报导水稻簇矮病毒(RBSV),水稻矮缩病毒(RDV),水稻东格鲁病毒(RTV),水稻齿矮病毒(RRSV)和水稻草矮病毒(RGSV)等的吸收光谱和荧光光谱.结果表明:不同病毒有不同的吸收光谱带和荧光光谱带.它包括主峰,各次峰及其轮廓.通过不同相对强度的光谱,可以判断其浓度.这些光谱可以反映病毒的某些结构信息.同时通过光谱也可以鉴别水稻病毒的类型.  相似文献

对提纯的细胞色素bc1复合物的氧化态和底物琥珀酸还原态两个样品进行了共振喇曼和富立叶喇曼光谱测定和比较。琥珀酸还原态与氧化态的共振谱比较明显有变化,而富立叶红外谱没有什么差别。说明呼吸链的电子传递体在氧化态与还原态交替变化进行电子传递时,蛋白总体构象不发生大的改变,而活性中心血红素辅基局部构象变化很大。  相似文献

Laser scanning confocal microscopy is a powerful technique that can be applied to study the localisation and behaviour of proteins and nucleic acids in many experimental situations. It is a particularly useful technique for the study of virus infections because of the changes that occur in the distribution and amounts of both viral and cellular proteins as infection develops. These changes reflect key stages and important regulatory events that govern the efficiency of infection. Using herpes simplex virus type 1 infected cells as an experimental model, this article provides guidance for users new to confocal microscopy on basic principles and techniques. Using functional and proteomic screens of proteins that regulate the Cdc42 GTPase, we have identified a network of protein interactions that center around the Cdc42 RhoGAP Rich1 and organize apical polarity in MDCK epithelial cells. Rich1 binds the scaffolding protein angiomotin (Amot) and is thereby targeted to a protein complex at tight junctions (TJs) containing the PDZ-domain proteins Pals1, Patj, and Par-3. Regulation of Cdc42 by Rich1 is necessary for maintenance of TJs, and Rich1 is therefore an important mediator of this polarity complex. Furthermore, the coiled-coil domain of Amot, with which it binds Rich1, is necessary for localization to apical membranes and is required for Amot to relocalize Pals1 and Par-3 to internal puncta. We propose that Rich1 and Amot maintain TJ integrity by the coordinate regulation of Cdc42 and by linking specific components of the TJ to intracellular protein trafficking.  相似文献

The involvement of wild birds in western equine encephalitis (WEE) and St. Louis encephalitis (SLE) virus activity in the Red River valley area of North Dakota (USA) during a WEE epidemic was investigated in August 1975. Free-ranging birds were captured with mist nets and nestlings by hand. Virologic and serologic results indicated that a similar rate of WEE virus activity occurred throughout Richland County and between permanent and summer resident birds. The rate of SLE virus activity in the birds of Richland County was lower than for WEE virus, but the SLE antibody prevalence was greater in rural areas than within urban locations. Seven of the nine WEE virus isolations were from nestling birds of four different species; the remaining two from adults of two different species. Overall prevalence of neutralizing (N) antibody against WEE virus was 5% in nestling and 14% in adult birds but was the opposite for N antibody against SLE virus, 17% in nestling and 5% in adult birds. Differences between the two viruses in the presence and persistence of maternal N antibody or differential mortality in nestling birds may have caused the disparity in antibody prevalences.  相似文献

Development of dendritic spines is important for synaptic function, and alteration in spine morphogenesis is often associated with mental disorders. Rich2 was an uncharacterized Rho-GAP protein. Here we searched for a role of this protein in spine morphogenesis. We found that it is enriched in dendritic spines of cultured hippocampal pyramidal neurons during early stages of development. Rich2 specifically stimulated the Rac1 GTPase in these neurons. Inhibition of Rac1 by EHT 1864 increased the size and decreased the density of dendritic spines. Similarly, Rich2 overexpression increased the size and decreased the density of dendritic spines, whereas knock-down of the protein by specific si-RNA decreased both size and density of spines. The morphological changes were reflected by the increased amplitude and decreased frequency of miniature EPSCs induced by Rich2 overexpression, while si-RNA treatment decreased both amplitude and frequency of these events. Finally, treatment of neurons with EHT 1864 rescued the phenotype induced by Rich2 knock-down. These results suggested that Rich2 controls dendritic spine morphogenesis and function via inhibition of Rac1.  相似文献

The secreted alphaherpesvirus glycoprotein G (gG) works differently from other proteins. Analysis of the role of ILTV gG in virus attachment, penetration, direct cell-to-cell spread (CTCS) and the growth curve showed that gG or its antibody had no effect on ILTV attachment and penetration and that the gG antibody reduced the virus plaque size and the one-step growth curve on chicken embryo liver (CEL) cells, but gG did not affect the virus plaque size or the one-step growth curve on CEL cells. Laser scanning confocal microscopy (LSCM) detection showed that ILTV gG is located in the perinuclear region and the membrane of the CEL cells. These results suggested that ILTV gG might contribute to direct cell-to-cell transmission.  相似文献
