The metabolism of iron (Fe) has been shown to interact with that of aluminum (Al) in relation to intestinal absorption, transport in the blood plasma, and the induction of lipid peroxidation and cellular damage. Also, dietary supplementation with citrate has been shown to increase the absorption of both metals and, in the presence of high intakes of Fe and Al, leads to excessive accumulation of both metals in the body. In this study, the likely interaction between Al and internal Fe metabolism was investigated using rats fed diets that were either deficient, sufficient, or loaded with Fe, with or without the addition of Al and sodium citrate. These diets commenced when the rats were 4 wk old and were continued for 9–11 wk. At that time, Fe metabolism as assessed by measurement of organ uptake of59Fe and125I-transferrin, after iv injection of transferrin labeled with both isotopes, plus measurement of tissue concentrations of nonheme Fe and Al. The Fedeficient diet and Fe-loaded diet led to states of Fe deficiency and Fe overload in the rats, and supplementation of the diet with Al increased Al levels in the kidneys, liver, and femurs, but, generally, only when the diet also contained citrate. Neither Al nor citrate supplementation of the diet had any effect on nonheme Fe concentrations in the liver, kidney, or brain, or on the uptake of59Fe or125I-transferrin by liver, kidney, brain, or spleen. Only with the femurs was a significant effect observed: increased59Fe uptake in association with increased Al intake. Therefore, using this animal model, there was little evidence for interaction between Fe and Al metabolism, and no support was obtained for the hypothesis that dietary supplementation with Fe and citrate can lead to excessive Fe absorption and deposition in the tissues.  相似文献

Homozygous Belgrade rats have a hypochromic anaemia due to impaired iron transport across the cell membrane of immature erythroid cells. This study aimed at investigating whether there are also abnormalities of Mn metabolism in erythroid and other types of cells. The experiments were performed with homozygous (b/b) and heterozygous (+/b) Belgrade rats and Wistar rats and included measurements of Mn uptake by reticulocytes in vitro, Mn absorption from in situ closed loops of the duodenum, and plasma clearance and uptake by several organs after intravenous injection of radioactive Mn bound to transferrin (Tf ) or mixed with serum. Similar measurements were made with 59Fe-labelled Fe in several of the experiments. Mn uptake by reticulocytes and absorption from the duodenum was impaired in b/b rats compared with +/b or Wistar rats. The plasma clearance of Mn-Tf was much slower than Mn-serum, but both were faster than the clearance of Fe-Tf. Uptake of 54Mn by the kidneys, brain and femurs was less in b/b than Wistar or +/b rats, but uptake by the liver was greater in b/b rats. Similar differences were found for 59Fe uptake by kidneys, brain and femurs but 59Fe uptake by the liver was also impaired in the liver. It is concluded that the genetic abnormality present in b/b rats affects Mn metabolism as well as Fe metabolism and that Mn and Fe share similar transport mechanisms in the cells of erythroid tissue, duodenal mucosa, kidney and blood-brain barrier. Environmental lead intoxication, which frequently causes neurological disturbances, and iron deficiency are clinical problems commonly found in children. Also, iron deficiency has been shown to augment lead absorption from the intestine. Hence, there is evidence for an interaction between lead and iron metabolism which could produce changes in lead and iron uptake by the brain and other tissues. These possibilities were investigated using 15-, 21-, and 63-old rats with varying nutritional iron and lead status. Dams were fed diets containing 0 or 3% lead-acetate and 0.2% lead-acetate in the drinking water. After weaning, 0.2% lead-acetate in the drinking water became the sole source of dietary lead. Measurements were made of tissue lead and nonheme iron levels and the uptake of59Fe after intravenous injection of transferrin-bound59Fe. Iron deficiency was associated with increased intestinal absorption of lead as indicated by blood and kidney lead levels in rats exposed to dietary lead. However, iron deficiency did not increase lead deposition in the brain, and in all rats brain lead levels were relatively low (<0.1 μg/g). Lead concentrations in the liver were below 2 μg/g, whereas kidneys had almost 20 times this concentration. Animals with iron deficiency had lower liver iron levels and had increased brain59Fe uptake in comparison to control rats. However, iron levels in brain and kidneys were unaffected by lead intoxication regardless of the animal's iron status.59Fe uptake rates were also unaffected by lead, but increased rates of uptake were apparent in iron-deficient rats. Lead did increase liver iron levels in all iron-adequate rats, but iron deficiency had little effect. It is concluded that, compared with other tissues, the blood-brain barrier largely restricts lead uptake by the brain and that the uptake that does occur is unrelated to the iron status of the animal. Also, the level of lead intoxication produced in this investigation did not influence iron uptake by the brain and kidneys, but liver iron stores could be incresed if iron levels were already adequate.  相似文献

The iron chelators desferrioxamine (DFO), pyridoxal isonicotinoyl hydrazone (PIH), 2,2-bipyridine, diethylenetriamine penta-acetic acid (DTPA) and 1,2 dimethyl-3-hydroxy pyrid-4-one (CP20) were analysed for their ability to change59Fe uptake and release from the brain of 15- and 63-day rats either during or after intravenous injection of59Fe-125I-transferrin. DTPA was the only chelator unable to significantly reduce iron uptake into the brain of 15-day rats. This indicates that iron is not released from transferrin at the luminal surface of brain capillary endothelial cells. CP20 was able to reduce iron uptake in the brain by 85% compared to 28% with DFO. Only CP20 was able to significantly reduce brain iron uptake in 63 day rats. Once59Fe had entered the brain no chelator used was able to mediate its release. All of the chelators except CP20 had similar effects on femur iron uptake as they did on brain uptake, suggesting similar iron uptake mechanisms. It is concluded that during the passage of transferrin-bound iron into the brain the iron is released from transferrin within endothelial cells after endocytosis of transferrin.  相似文献

Hereditary hemochromatosis type 3 is an iron (Fe)-overload disorder caused by mutations in transferrin receptor 2 (TfR2). TfR2 is expressed highly in the liver and regulates Fe metabolism. The aim of this study was to investigate duodenal Fe absorption and hepatic Fe uptake in a TfR2 (Y245X) mutant mouse model of hereditary hemochromatosis type 3. Duodenal Fe absorption and hepatic Fe uptake were measured in vivo by 59Fe-labeled ascorbate in TfR2 mutant mice, wild-type mice, and Fe-loaded wild-type mice (2% dietary carbonyl Fe). Gene expression was measured by real-time RT-PCR. Liver nonheme Fe concentration increased progressively with age in TfR2 mutant mice compared with wild-type mice. Fe absorption (both duodenal Fe uptake and transfer) was increased in TfR2 mutant mice compared with wild-type mice. Likewise, expression of genes participating in duodenal Fe uptake (Dcytb, DMT1) and transfer (ferroportin) were increased in TfR2 mutant mice. Nearly all of the absorbed Fe was taken up rapidly by the liver. Despite hepatic Fe loading, hepcidin expression was decreased in TfR2 mutant mice compared with wild-type mice. Even when compared with Fe-loaded wild-type mice, TfR2 mutant mice had increased Fe absorption, increased duodenal Fe transport gene expression, increased liver Fe uptake, and decreased liver hepcidin expression. In conclusion, despite systemic Fe loading, Fe absorption and liver Fe uptake were increased in TfR2 mutant mice in association with decreased expression of hepcidin. These findings support a model in which TfR2 is a sensor of Fe status and regulates duodenal Fe absorption and liver Fe uptake.  相似文献

Transferrin and Iron Uptake by the Brain: Effects of Altered Iron Status   总被引:7,自引:3,他引:4  
Transferrin (Tf) and iron uptake by the brain were measured in rats using 59Fe-125I-Tf and 131I-albumin (to correct for the plasma content of 59Fe and 125I-Tf in the organs). The rats were aged from 15 to 63 days and were fed (a) a low-iron diet (iron-deficient) or, as control, the same diet supplemented with iron, or (b) a chow diet with added carbonyl iron (iron overload), the chow diet alone acting as its control. Iron deficiency was associated with a significant decrease and iron overload with a significant increase in brain nonheme iron concentration relative to the controls. In each dietary treatment group, the uptake of Tf and iron by the brain decreased as the rats aged from 15 to 63 days. Both Tf and iron uptake were significantly greater in the iron-deficient rats than in their controls and lower in the iron-loaded rats than in the corresponding controls. Overall, iron deficiency produced about a doubling and iron overload a halving of the uptake values compared with the controls. In contrast to that in the brain, iron uptake by the femurs did not decrease with age and there was relatively little difference between the different dietary groups. 125I-Tf uptake by the brains of the iron-deficient rats increased very rapidly after injection of the labelled proteins, within 15 min reaching a plateau level which was maintained for at least 6 h. The uptake of 59Fe, however, increased rapidly for 1 h and then more slowly, and in terms of percentage of injected dose reached much higher values than did 125I-Tf uptake.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Concentrations of intracellular, low molecular weight (LMW) and desferrioxamine B (DF) chelatable Fe, in tissues of normal, Fe-deficient and Fe-loaded female rats, were determined. Ice cold, high speed supernatants were rapidly fractionated on Ultrogel AcA202 or by filter centrifugation. After correction for residual blood and DF effects on Fe proteins, liver, kidney, heart and spleen contained 3-8 micrograms/g LMW Fe, brain 20 micrograms/g, with DF; two-thirds of this was detected without DF. There was little variation with Fe status. MW standardization and fractionation on Sephadex G-25 indicated components of apparent MW 13,000, 1400 and 350; the latter two were rapidly labeled with in vivo 59Fe.  相似文献

The capability of cucumber (Cucumis sativus L., cv. Serpente cinese), a Strategy I plant and barley (Hordeum vulgaris L., cv. Europa), a Strategy II plant to use Fe complexed by a water-soluble humic fraction (WEHS) extracted from a peat, was studied. Uptake of 59Fe from 59Fe-WEHS by cucumber plants was higher at pH 6.0 than at pH 7.5. Roots of intact cucumber plants were able to reduce the FeIII-WEHS complex either at pH 6.0 or 7.5, rates being higher in the assay medium buffered at pH 6.0. After supply of 59Fe-WEHS, a large pool of root extraplasmatic 59Fe was formed, which could be used to a large extent by Fe-deficient plants, particularly under acidic conditions. Uptake of 59Fe from 59Fe-WEHS by Fe-sufficient and Fe-deficient barley plants was examined during periods of high (morning) and low (evening) PS release. Uptake paralleled the diurnal rhythm of PS release. Furthermore, 59Fe uptake was strongly enhanced by addition of PS to the uptake solution in both Fe-sufficient and Fe-deficient plants. High amount of root extraplasmatic 59Fe was formed upon supply of Fe-WEHS, particularly in the evening experiment. Fe-deficient barley plants were able to utilize Fe from the root extraplasmatic pool, conceivably as a result of high rates of PS release. The results of the present work together with previous observations indicate that cucumber plants (Strategy I) utilize Fe complexed to WEHS, presumably via reduction of FeIII-WEHS by the plasma membrane-bound reductase, while barley plants (Strategy II) use an indirect mechanism involving ligand exchange between WEHS and PS.  相似文献

Melanotransferrin (MTf) or melanoma tumor antigen p97 is a membrane-bound transferrin (Tf) homologue that binds iron (Fe). This protein is also found as a soluble form in the plasma (sMTf) and was suggested to be an Alzheimer's disease marker. In addition, sMTf has been recently suggested to cross the blood-brain barrier (BBB) and accumulate in the brain of the mouse following intravenous infusion. Considering the importance of this observation to the physiology and pathophysiology of the BBB and the function of sMTf in vivo, we investigated the uptake and distribution of 59Fe-125I-sMTf and compared it to 59Fe-125I-Tf that were injected intravenously in rats. Studies were also performed to measure 59Fe and 125I-protein uptake by reticulocytes using these radiolabelled proteins. The results showed that sMTf was rapidly catabolized, mainly in the liver and to a lesser extent by the kidneys. The 59Fe was largely retained by these organs but the 125I was released into the plasma. Only a small amount of 125I-sMTf or its bound 59Fe was taken up by the brain, less than that from 59Fe-125I-Tf. There was much less 59Fe uptake by erythropoietic organs (spleen and femurs) from 59Fe-sMTf than from 59Fe-Tf, and no evidence of receptor-mediated uptake of sMTf was obtained using reticulocytes. It is concluded that compared to Tf, sMTf plays little or no role in Fe supply to the brain and erythropoietic tissue. However, a small amount of sMTf was taken up from the plasma by the brain and a far greater amount by the liver.  相似文献

A study has been made on the effect of primary leaves on iron (Fe) distribution in the shoot. Bean (Phaseolus vulgaris L.) seedlings were precultured in nutrient solution with 8×10-5 M FeEDTA for 4 days, and then grown further with either 8×10-5 M FeEDTA (+Fe) or without Fe supply (-Fe) for another 5 days. Thereafter, both +Fe and -Fe plants were treated in three different ways: undisturbed; one primary leaf removed; or one primary leaf shaded, starting two hours before supply 59FeEDTA to the roots. The +Fe plants were supplied with 8×10-5 M 59FeEDTA, and the -Fe plants with only 1×10-6 M 59FeEDTA. After 1 to 8 hour uptake periods, plants were harvested and 59Fe in different organs was determined. Removal or shading of one primary leaf did not affect 59Fe uptake by roots and 59Fe translocation to the shoot in +Fe plants. In the -Fe plants, however, removal of one primary leaf decreased 59Fe uptake by roots, whereas shading of one primary leaf had no effect on 59Fe uptake but slightly enhanced 59Fe translocation from roots to the shoot. The quantity of 59Fe in primary leaves was positively correlated with quantity of 59Fe in the stem in the -Fepplants, but not in the +Fe plants. In both, the +Fe and -Fe plants, the quantity of 59Fe in the shoot apex was positively correlated with 59Fe in primary leaves. The results suggest that irrespective of the Fe nutritional status of plants, the source of Fe for the shoot apex is Fe retranslocated from primary leaves.  相似文献

Iron-deficiency anemia leads directly to both reduced hemoglobin levels and work performance in humans and experimental animals. In an attempt to observe a direct link between work performance and insufficient iron at the cellular level, we produced severe iron deficiency in female weanling Sprague-Dawley rats following five weeks on a low-iron diet. Deficient rats were compared with normal animals to observe major changes in hematological parameters, body weight, and growth of certain organs and tissues. The overall growth of iron-deficient animals was approximately 50% of normal. The ratio of organ weight: body weight increased in heart, liver, spleen, kidney, brain, and soleus muscle in response to iron deficiency. Further, mitochondria from heart and red muscle retained their iron more effectively under the stress of iron deficiency than mitochondria from liver and spleen. Metabolism of iron in normal and depleted tissue was measured using tracer amounts of59Fe administered orally. As expected, there was greater uptake of tracer iron by iron-deficient animals. The major organ of iron accumulation was the spleen, but significant amounts of isotope were also localized in heart and brain. In all muscle tissue examined the59Fe preferentially entered the mitochondria. Enhanced mitochondrial uptake of iron prior to any detectable change in the hemoglobin level in experimental animals may be indicative of nonhemoglobin related biochemical changes and/or decrements in work capacity.  相似文献

This solution culture study examined the effect of the deposition of iron plaque on zinc uptake by Fe-deficient rice plants. Different amounts of iron plaque were induced by adding Fe(OH)3 at 0, 10, 20, 30, and 50 mg Fe/L in the nutrient solution. After 24 h of growth, the amount of iron plaque was correlated positively with the Fe(OH)3 addition to the nutrient solution. Increasing iron plaque up to 12.1 g/kg root dry weight increased zinc concentration in shoots by 42% compared to that at 0.16 g/kg root dry weight. Increasing the amount of iron plaque further decreased zinc concentration. When the amounts of iron plaque reached 24.9 g/kg root dry weight, zinc concentration in shoots was lower than that in shoots without iron plaque, implying that the plaque became a barrier for zinc uptake. While rice plants were pre-cultured in –Fe and +Fe nutrient solution in order to produce the Fe-deficient and Fe-sufficient plants and then Fe(OH)3 was added at 20, 30, and 50 mg Fe/L in nutrient solution, zinc concentrations in shoots of Fe-deficient plants were 54, 48, and 43 mg/kg, respectively, in contrast to 32, 35, and 40 mg/kg zinc in shoots of Fe-sufficient rice plants. Furthermore, Fe(OH)3 addition at 20 mg Fe/L and increasing zinc concentration from 0.065 to 0.65 mg Zn/L in nutrient solution increased zinc uptake more in Fe-deficient plants than in Fe-sufficient plant. The results suggested that root exudates of Fe-deficient plants, especially phytosiderophores, could enhance zinc uptake by rice plants with iron plaque up to a particular amount of Fe.  相似文献

Chlorophyll (Chl), phycoerythrin (PE), total nitrogen (TN% dw) and Fein tissues were measured in Fe-deficient cultures of Gracilariatenuistipitata var. liui over a period of 60 days. 55Fe uptakeand photosynthetic carbon fixation (NaH14CO3) werecompared in Fe-rich and Fe-deficient cultures and analyzed the effects ofFe-deficiency on the ultrastructure. The maximum carbon fixationdecreased significantly (p < 0.01) under Fe-deficiency. Thechlorophyll and phycoerythrin contents also declined with decreasing tissueiron content, falling, respectively, to 7.9 and 33.8% of their originallevel. Photosynthesis in Fe-deficient cells became light-saturated at lowerirradiance than the control. Total N in tissue decreased from 3.65 to2.49%. 55Fe uptake rate for cultures grown on NO3 -was measured following resuspension in either NH4 + orNO3 - as N source. Enhanced Fe uptake developedunder Fe stress, especially with cells resuspended in NH+ 4-N medium. The Vmaxfor Fe uptake was higher with NH4 + thanNO3 - (62.8 versus 12.1 pmol mg dw-1 h-1). The requirement for N accelerates further Fe uptake. Ultrastructuralobservations of Fe-deficient cells showed reductions in chloroplast number,degeneration of lamellar organization, decrease in mitochondrial matrixdensity and variation in accumulation body number and morphology. During Fe-deficiency, the growth rate continued to decline and after 40days of iron deficiency, no further growth was detectable, and eventuallyiron deficiency resulted in chlorosis. The results suggest that the lowergrowth rate of Gracilaria tenuistipitata var. liui underFe-deficiency may result from largely from inhibition of photosynthesis andnitrogen utilization.  相似文献

By using two tomato genotypes line 227/1 (Fe chlorosis susceptible) and Roza (Fe chlorosis tolerant) and their reciprocal F1hybrid, some root morphological changes, pH changes of nutrient solution, reduction capacity of FeIII and uptake and root-to-shoot translocation of 59Fe were studied under controlled environmental conditions in nutrient solution with 3 different Fe supplies as Fe EDDHA (i.e., 10–7 M, severe Fe deficiency; 10–6 M, intermediate Fe deficiency; 10–4 M, adequate Fe supply). Tolerant parent `Roza' was less affected by low Fe supply than susceptible parent `line 227/1' as judged from the severity of leaf chlorosis. Under both Fe deficient conditions there were no differences between the reciprocal hybrids concerning the appearance of chlorosis. Under intermediate Fe deficiency, reciprocal F1 hybrids (`line 227/1 × Roza' and `Roza × line' 227/1) showed an intermediate chlorosis between tolerant and susceptible parents. However, under severe Fe deficiency the reciprocal hybrids were more chlorotic than the tolerant parent irrespective of which parent was the cytoplasm contributor. A decreased Fe supply during preculture enhanced FeIII reduction capacities of the parents and reciprocal hybrids. Differences in the tolerance to Fe deficiency always were better correlated with FeIII reduction capacity of the genotypes than the Fe deficiency-induced release of H+ ions. Under both Fe deficient conditions the tolerant parent Roza had a much higher FeIII reduction capacity than the susceptible parent line 227/1. The reduction capacity of the hybrids `Roza × line 227/1' was very similar to the capacity of the parent Roza, but higher than the capacity of the hybrids `line 227/1×Roza' at both Fe-deficient conditions. Under both Fe deficient conditions tolerant parent had higher number of lateral roots than the susceptible parent. Among the reciprocal hybrids `Roza × line 227/1' possessed more lateral roots than the `line 227/1 × Roza' under both Fe deficient conditions. Low Fe nutritional status resulted in marked increase in root uptake of 59Fe. At adequate Fe supply, reciprocal hybrids and their parents did not differ in uptake and root-to-shoot translocation of Fe. However, under Fe-deficient conditions uptake and root-to-shoot translocation of 59Fe were significantly higher in the Fe chlorosis tolerant than the susceptible parent. Based on the reduction capacity of FeIII and uptake and root-to-shoot translocation of Fe, the F1 hybrids obtained from the cross in which the maternal genotype was Roza appeared to be more tolerant than when the maternal genotype was the susceptible line 227/1. Uptake and translocation ratio of the F1 hybrids obtained from `Roza × line 227/1' were similar to those of the parent Roza, but higher than the F1 hybrids obtained from `line 227/1 × Roza', particularly under intermediate Fe deficiency. The results indicate that FeIII reduction show a better relationship to Fe efficiency than Fe deficiency induced release of H+ ions. The inheritance of Fe deficiency tolerance of Roza seems not to be simple monogenic. It might be characterised by both, nuclear and extranuclear heredity. The intermediate responses of the reciprocal hybrids of the `line 227/1 × Roza' indicates that the Fe deficiency tolerance character of Roza is transferable by nuclear heredity. The better responses of the hybrids of `Roza × line 227/1' than the hybrids of `line 227/1 × Roza' may be due to maternal transmission from the parent Roza besides the nuclear transmission.  相似文献

Comparative studies on the effect of nitrogen (N) form on iron (Fe) uptake and distribution in maize (Zea mays L. cv Yellow 417) were carried out through three related experiments with different pretreatments. Experiment 1: plants were precultured in nutrient solution with 1.0×10–4 M FeEDTA for 6 d and then exposed to NO3–N or NH4–N solution with 1.0×10–4 M FeEDTA or without for 7 d. Experiment 2: plants were precultured with 59FeEDTA for 6 d and were then transferred to the solution with different N forms, and 0 and 1.0×10–4 M FeEDTA for 8 d. Experiment 3: half of roots were supplied with 59FeEDTA for 5 d and then cut off, with further culturing in treatment concentrations for 7 d. In comparison to the NH4-fed plants, young leaves of the NO3-fed plants showed severe chlorosis under Fe deficiency. Nitrate supply caused Fe accumulation in roots, while NH4–N supply resulted in a higher Fe concentration in young leaves and a lower Fe concentration in roots. HCl-extractable (active) Fe was a good indicator reflecting Fe nutrition status in maize plants. Compared with NO3-fed plants, a higher proportion of 59Fe was observed in young leaves of the Fe-deficient plants fed with NH4–N. Ammonium supply greatly improved 59Fe retranslocation from primary leaves and stem to young leaves. Under Fe deficiency, about 25% of Fe in primary leaves of the NH4-fed plants was mobilized and retranslocated to young leaves. Exogenous Fe supply decreased the efficiency of such 59Fe retranslocation. The results suggest that Fe can be remobilized from old to young tissues in maize plants but the remobilization depends on the form of N supply as well as supply of exogenous Fe.  相似文献

To further examine the interrelationships between manganese and iron absorption, the mucosal uptake, initial rate of loss, wholebody retention, and tissue distribution of an orally administered 54Mn radiotracer were compared between normal and β2-microglobulin knockout [β2m(-/-)] mice. These mutant mice are commonly used as a model for the study of human hemochromatosis, a hereditary ironoverload disease. Initial uptake of 54Mn by the intestinal mucosa, the liver, and the brain was not different between the two strains. The mutant mice had much higher concentrations of nonheme and total iron in the liver, but hepatic manganese, copper, magnesium, and zinc concentrations were similar between the two strains. Summary The role of specific transferrin (Tf) and Tf receptor interaction on brain capillary endothelial cells in iron transport from the plasma to the brain was investigated by using Tf from several species of animals labeled with 59Fe and 125I, and 15-day and adult rats. The rate of iron transfer was much greater in the 15-day rats. It was greatest with Tf from the mammals, rat, rabbit and human, but much lower with chicken ovotransferrin and quokka (a marsupial), toad, lizard, crocodile, and fish Tf. The uptake of Tf by the brain showed a similar pattern, except for a very high uptake of ovotransferrin (ovo Tf). Iron uptake by the femurs (a source of bone marrow) was also high with Tf from the mammalian species and low with the other types of Tf, but showed little change with aging of the animals. It is concluded that iron transport into the brain is dependent on the function of Tf receptors, probably on capillary endothelial cells, and that these receptors show the same type of species specificity as the receptors on immature erythroid cells. Iron uptake from ferrated (59Fe) pseudobactin (PSB), a Pseudomonas putida siderophore, by various plant species was studied in nutrient solution culture under short term (10 h) and long term (3 weeks) conditions. In the short term experiments, 59Fe uptake rate from 59FePSB by dicots (peanuts, cotton and sunflower) was relatively low when compared with 59Fe uptake rate from 59FeEDDHA. Iron uptake rate from 59FePSB was pH and concentration dependent, as was the Fe uptake rate from 59FeEDDHA. The rate was about 10 times lower than that of Fe uptake from the synthetic chelate. Results were similar for long term experiments.Monocots (sorghum) in short term experiments exhibited significantly higher uptake rate of Fe from FePSB than from FeEDDHA. In long term experiments, FePSB was less efficient than FeEDDHA as an Fe source for sorghum at pH 6, but the same levels of leaf chlorophyll concentration were obtained at pH 7.3.Fe uptake rates by dicots from the siderophore and FeEDDHA were found to correlate with Fe reduction rates and reduction potentials (E0) of both chelates. Therefore, it is suggested that the reduction mechanism governs the Fe uptake process from PSB by dicots. Further studies will be conducted to determine the role of pH in Fe aquisition from PSB by monocots.  相似文献

ObjectiveThe impact of multistrain probiotics on iron (Fe) metabolism under Fe-deficient diet conditions remains unknown. The study aimed to compare the effect of 6 weeks simultaneous and exclusive oral multistrain probiotic and iron supplementation on selected parameters of Fe metabolism in rats on an Fe-deficient diet.MethodsForty rats were assigned to five groups, with eight animals in each, and for 6 weeks received: the CC group- a standard diet, the DD group- an Fe-deficient diet, the DPB group- an Fe-deficient with a multispecies probiotic, the DFE group- an Fe-deficient diet supplemented with iron, the DPBFE group- an Fe-deficient diet with iron and a multispecies probiotic. The Fe content in blood and tissues; serum concentration of erythroferrone, ferritin (Ft), homocysteine, hepcidin (HEPC) and lactoferrin; liver content of divalent metal transporter 1 (DMT1), transferrin receptor protein 1 (TfR1) and 2 (TfR2) and ZRT/IRT-like protein 14 (ZIP14) and faecal microbiota were assessed.ResultsIn DPBFE group, unlike in DPB and DFE groups, duodenal Fe content was higher compared to DD group. Similarly, serum Ft level was higher in DPBFE group, but not in DPB and DFE groups, compared to DD group.ConclusionsSix weeks simultaneous oral multistrain probiotic and Fe supplementation, but not exclusive probiotic or Fe intake, increases duodenal Fe absorption in rats and presents higher effectiveness in increasing tissue Fe stores.  相似文献

Under anaerobic conditions, ferric hydroxide deposits on the surface of rice roots have been shown to affect the uptake of some nutrients. In the present experiment, different amount of this iron plaque were induced on the roots of rice (Oryza sativa L. cv. TZ88-145) by supplying different Fe(OH)3 concentrations in nutrient solutions, and the effect of the iron plaque on phosphorus uptake was investigated. Results showed that 1) iron plaque adsorbed phosphorus from the growth medium, and that the amount of phosphorus adsorbed by the plaque was correlated with the amount of plaque; 2) the phosphorus concentration in the shoot increased by up to 72% after 72 h at concentration of Fe(OH)3 in the nutrient solution from 0 to 30 mg Fe/L, corresponding with amounts of iron plaque from 0.2 to 24.5 mg g-1 (root d. wt); 3) the phosphorus concentration in the shoots of rice with iron plaque was higher than that without iron plaque though the concentration in the shoot decreased when Fe(OH)3 was added at 50 mg Fe/L producing 28.3 mg g-1 (root d. wt) of plaque; and 4) the phosphorus concentrations in Fe-deficient and Fe-sufficient rice plants with iron plaque were the same, although phytosiderophores were released from the Fe-deficient roots. The phytosiderophores evidently did not mobilise phosphorus adsorbed on plaque. The results suggest that iron plaque on rice plant roots might be considered a phosphorus reservoir.
