To assess whether long-term faunal changes have occurred on intertidal flats in the Danish Wadden Sea, results of faunal surveys in the Skallingen area during the 1930s, 1940s and 1980s were compared. Since the earlier studies, two species have disappeared (Scrobicularia plana andPetricola pholadiformis) and two have invaded the area (Tharyx killariensis andEnsis americanus). This is, however, not a local event as species changes have occurred on a larger scale (Wadden Sea region). Comparison of abundance data did not provide evidence of changes from the 1930s until the 1980s. Spatio-temporal fluctuations in two dominant species, the mudsnailHydrobia ulvae and the cockleCerastoderma edule chosen for closer examination, could be explained by natural processes. Growth data on cockles from the 1930s and the 1980s matched perfectly. During both periods, cockles showed a much lower growth rate than generally reported from the Wadden Sea, while at the same time they occurred at high densities (>2000 ind·m−2). Intraspecific competition among cockles is suspected as being a major cause of the low growth rates. Estimates of secondary production and P/B-ratios of cockles were also in general agreement during the 1930s and the 1980s when corrected for differences in the age structure of the cockle populations. With the possible exception ofMytilus edulis, which according to some observations has extended its range along the low-water line, the present comparison failed to demonstrate long-term faunal changes caused by increased eutrophication. This results is expected to be representative for intertidal flats not exposed to direct impacts from terrestrial run-offs and waste-water discharges.  相似文献

Mycoplasma arthritidis-derived mitogen (MAM) is a superantigen that can activate large fractions of T cells bearing particular Vβ elements of T cell receptor. Here, we report the crystal structure of a MAM mutant K201A in apo form (unliganded) at 2.8-Å resolutions. We also partially refined the crystal structures of the MAM wild type and another MAM mutant L50A in apo forms at low resolutions. Unexpectedly, the structures of these apo MAM molecules display a three-dimensional domain-swapped dimer. The entire C-terminal domains of these MAM molecules are involved in the domain swapping. Functional analyses demonstrated that the K201A and L50A mutants do not show altered ability to bind to their host receptors and that they stimulate the activation of T cells as efficiently as does the wild type. Structural comparisons indicated that the “reconstituted” MAM monomer from the domain-swapped dimer displays large differences at the hinge regions from the MAMwt molecule in the receptor-bound form. Further comparison indicated that MAM has a flexible N-terminal loop, implying that conformational changes could occur upon receptor binding.  相似文献

Intracellular β-amyloid (Aβ) accumulation is an early event in Alzheimer’s disease (AD) progression. Recently, it has been uncovered that presenilins (PSs), the key components of the amyloid precursor protein (APP) processing and the β-amyloid producing γ-secretase complex, are highly enriched in a special sub-compartment of the endoplasmic reticulum (ER) functionally connected to mitochondria, called mitochondria-associated ER membrane (MAM). A current hypothesis of pathogenesis of Alzheimer’s diseases (AD) suggests that MAM is involved in the initial phase of AD. Since MAM supplies mitochondria with essential proteins, the increasing level of PSs and β-amyloid could lead to metabolic dysfunction because of the impairment of ER-mitochondrion crosstalk. To reveal the early molecular changes of this subcellular compartment in AD development MAM fraction was isolated from the cerebral cortex of 3 months old APP/PS1 mouse model of AD and age-matched C57BL/6 control mice, then mass spectrometry-based quantitative proteome analysis was performed. The enrichment and purity of MAM preparations were validated with EM, LC-MS/MS and protein enrichment analysis. Label-free LC-MS/MS was used to reveal the differences between the proteome of the transgenic and control mice. We obtained 77 increased and 49 decreased protein level changes in the range of ??6.365 to +?2.988, which have mitochondrial, ER or ribosomal localization according to Gene Ontology database. The highest degree of difference between the two groups was shown by the ATP-binding cassette G1 (Abcg1) which plays a crucial role in cholesterol metabolism and suppresses Aβ accumulation. Most of the other protein changes were associated with increased protein synthesis, endoplasmic-reticulum-associated protein degradation (ERAD), oxidative stress response, decreased mitochondrial protein transport and ATP production. The interaction network analysis revealed a strong relationship between the detected MAM protein changes and AD. Moreover, it explored several MAM proteins with hub position suggesting their importance in Aβ induced early MAM dysregulation. Our identified MAM protein changes precede the onset of dementia-like symptoms in the APP/PS1 model, suggesting their importance in the development of AD.  相似文献

Based on fieldwork among the Sa'dan Toraja of Indonesia, this article examines a specific relationship between the living and the dead. It is a relationship which is usually described in terms of intimacy. Within this intimacy, the living and the dead are described as sharing the same desires. Furthermore, as far as the living are concerned, these desires have to be expressed in words and acted out in the form of mortuary sacrifices. In this way, the living are said to remember the dead. A natural experiment is employed to analyze the relationship between living standards, diabetes, and cardiovascular disease. Results show that shocks generated by two powerful tropical storms striking Puerto Rico during the late 1920s and early 1930s had long-term consequences consistent with the fetal origins hypothesis. Individuals in the womb or early infancy in the aftermath of the storms are more likely to report a diagnosis of hypertension, high cholesterol, diabetes, and are considerably more likely to have no formal schooling.  相似文献

One mechanism by which communication between the endoplasmic reticulum (ER) and mitochondria is achieved is by close juxtaposition between these organelles via mitochondria-associated membranes (MAM). The MAM consist of a region of the ER that is enriched in several lipid biosynthetic enzyme activities and becomes reversibly tethered to mitochondria. Specific proteins are localized, sometimes transiently, in the MAM. Several of these proteins have been implicated in tethering the MAM to mitochondria. In mammalian cells, formation of these contact sites between MAM and mitochondria appears to be required for key cellular events including the transport of calcium from the ER to mitochondria, the import of phosphatidylserine into mitochondria from the ER for decarboxylation to phosphatidylethanolamine, the formation of autophagosomes, regulation of the morphology, dynamics and functions of mitochondria, and cell survival. This review focuses on the functions proposed for MAM in mediating these events in mammalian cells. In light of the apparent involvement of MAM in multiple fundamental cellular processes, recent studies indicate that impaired contact between MAM and mitochondria might underlie the pathology of several human neurodegenerative diseases, including Alzheimer's disease. Moreover, MAM has been implicated in modulating glucose homeostasis and insulin resistance, as well as in some viral infections.  相似文献

Question: What was the role of fire in montane pine‐oak (Pinus‐Quercus) stands under changing human land uses on a temperate forest landscape in eastern North America? Location: Mill Mountain in the central Appalachian Mountains, Virginia, US. Methods: A dendroecological reconstruction of fire history was generated for four stands dominated by xerophytic pine and oak species. The fire chronology began under presettlement conditions following aboriginal depopulation. Subsequent land uses included European settlement, iron mining, logging, and US Forest Service acquisition and fire protection. Results: Fires occurred approximately every 5 years until 1930 without any evidence of a temporal trend in fire frequency. Burning ceased after 1930. Area‐wide fires affecting multiple pine stands were common, occurring at intervals of approximately 16 years. Most living pines became established during the late 1800s and early 1900s. Dead pines indicated that an older cohort established ca. 1730. Most hardwoods were established between the 1920s and 1940s. Conclusions: Except for fire protection, changes in land use had no discernible influence on fire frequency. Lightning ignitions and/or large fire extent may have been important for maintaining frequent burning in the 1700s, while fuel recovery may have constrained fire frequency during later periods. The disturbance regime appears to be characterized by frequent surface fires and occasional severe fires, insect outbreaks or other disturbances followed by pine recruitment episodes. Industrial disturbances appear to have had little influence on the pine stands. The greatest impact of industrial society is fire exclusion, which permitted hardwood establishment.  相似文献

The invasive American mink has been a component of Iceland's fauna since the 1930s. Hunting statistics indicate that until 2003 the population size was increasing, but thereafter decreased rapidly. The Icelandic marine environment has experienced various changes in recent years, including rising sea temperature and sand-eel collapse followed by seabird recruitment failure and population declines. Furthermore the arctic fox population has increased at least six-fold in the last three decades. Mink stomach content analysis in the period 2001–2009 revealed diet changes, and signs of reduced prey availability for this generalist predator, that were most significant in males. The most marked shift in composition was a decrease in consumption of birds. Our findings suggest that climate events, together with competition with increasing numbers of arctic foxes over terrestrial food, contributed to the sharp reduction in the mink population from 2004 and onwards. Despite their generalist behaviour, mink have apparently failed to respond fully to these environmental changes, and this susceptibility may benefit attempts to control their numbers. The results are relevant to the ability of top predators in general to cope with diverse ecosystem alterations triggered by climate change.  相似文献

As late as the 1930s, approaches to biochemical research not only were rather primitive, but a certain amount of mysticism still surrounded the biochemical events that occur in the living cell. To a great extent, this was due to the lack of techniques needed to uncover the subtle reactions in the living cell. In the early 1930s, an accidental meeting of two scientists revolutionized approaches in biochemical studies and led to the scientific explosion in molecular biology that has occurred during the last few decades. The dark political storm in Germany deposited Dr. Rudolf Schoenheimer on the New York shore, where he met Professor Urey, who recently had discovered "heavy" hydrogen. Schoenheimer suggested that biological compounds tagged with heavy atoms of hydrogen would enable investigators to follow their metabolic pathways. This intellectual leap revolutionized the thinking and design of experiments and made it possible to uncover the myriad reactions that occur in the living cell.  相似文献

Children with malformations of cortical development (MCD) frequently have associated cognitive impairments which reduce quality of life. We hypothesized that cognitive deficits associated with MCD can be improved with environmental manipulation or additional training. The E17 methylazoxymethanol acetate (MAM) exposure model bears many anatomical hallmarks seen in human MCDs as well as similar behavioral and cognitive deficits. We divided control and MAM exposed Sprague-Dawley rats into enriched and non-enriched groups and tested performance in the Morris water maze. Another group similarly divided underwent sociability testing and also underwent Magnetic Resonance Imaging (MRI) scans pre and post enrichment. A third group of control and MAM rats without enrichment were trained until they reached criterion on the place avoidance task. MAM rats had impaired performance on spatial tasks and enrichment improved performance of both control and MAM animals. Although MAM rats did not have a deficit in sociability they showed similar improvement with enrichment as controls. MRI revealed a whole brain volume decrease with MAM exposure, and an increase in both MAM and control enriched volumes in comparison to non-enriched animals. In the place avoidance task, MAM rats required approximately 3 times as long to reach criterion as control animals, but with additional training were able to reach control performance. Environmental manipulation and additional training can improve cognition in a rodent MCD model. We therefore suggest that patients with MCD may benefit from appropriate alterations in educational strategies, social interaction and environment. These factors should be considered in therapeutic strategies.  相似文献

Studies on ancient coral communities living in marginal conditions, including low light, high turbidity, extreme temperatures, or high nutrients, are important to understand the current structure of reefs and how they could potentially respond to global changes. The main goal of this study was to document the rich and well-preserved fossil coral fauna preserved in Miocene exposures of the Kutai Basin in East Kalimantan, Indonesia. Our collections include almost forty thousand specimens collected from 47 outcrops. Seventy-nine genera and 234 species have been identified. Three different coral assemblages were found corresponding to small patch reefs that developed under the influence of high siliciclastic inputs from the Mahakam Delta. Coral assemblages vary in richness, structure, and composition. Platy coral assemblages were common until the Serravallian (Middle Miocene), while branching coral assemblages became dominant in the Tortonian (Late Miocene). By the late Tortonian massive coral assemblages dominated, similar to modern-style coral framework. Our results suggest that challenging habitats, such as the Miocene turbid habitats of East Kalimantan, might have played an important role during the early diversification of the Coral Triangle by hosting a pool of resilient species more likely to survive the environmental changes that have affected this region since the Cenozoic. Further research that integrates fossil and recent turbid habitats may provide a glimpse into the dynamics and future of coral reefs as “typical” clear-water reefs continue to decline in most regions.  相似文献

The mitochondria-associated membrane (MAM) is a domain of the endoplasmic reticulum (ER) that mediates the exchange of ions, lipids and metabolites between the ER and mitochondria. ER chaperones and oxidoreductases are critical components of the MAM. However, the localization motifs and mechanisms for most MAM proteins have remained elusive. Using two highly related ER oxidoreductases as a model system, we now show that palmitoylation enriches ER-localized proteins on the MAM. We demonstrate that palmitoylation of cysteine residue(s) adjacent to the membrane-spanning domain promotes MAM enrichment of the transmembrane thioredoxin family protein TMX. In addition to TMX, our results also show that calnexin shuttles between the rough ER and the MAM depending on its palmitoylation status. Mutation of the TMX and calnexin palmitoylation sites and chemical interference with palmitoylation disrupt their MAM enrichment. Since ER-localized heme oxygenase-1, but not cytosolic GRP75 require palmitoylation to reside on the MAM, our findings identify palmitoylation as key for MAM enrichment of ER membrane proteins.  相似文献

Paleolimnological techniques can reveal long-term perturbations and associated stable state transitions of lake ecosystems. However, such transitions are difficult to predict since changes to lake ecosystems can be abrupt or gradual. This study examined whether there were past transitions in the ecological regime of Kings Billabong, a shallow River Murray wetland in southeast Australia. A 94-cm-long core, covering c. 90 years of age, was analysed at 1 cm resolution for subfossil cladocerans, diatoms and other proxies. Prior to river regulation (c. 1930), the littoral to planktonic ratios of cladocerans and diatoms, and bulk sediment δ13C values were high, while the period from c. 1930 to c. 1970 experienced considerable changes to the wetland ecosystem. The abrupt nature of changes of planktonic cladocerans and diatoms, particularly after the onset of river regulation (1930s), was triggered by inundation, high rates of sedimentation and shifts in bulk sediment δ15N values. However, the transition of a once littoral-dominated community, to one favouring an increasingly turbid, plankton-dominated trophic condition following river regulation was relatively slow and lasted for decades. The progression to a new regime was likely delayed by the partial recovery of submerged plant communities and related internal dynamics.  相似文献

BackgroundChildren with moderate acute malnutrition (MAM) have an increased risk of mortality, infections and impaired physical and cognitive development compared to well-nourished children. In parts of Ethiopia not considered chronically food insecure there are no supplementary feeding programmes (SFPs) for treating MAM. The short-term outcomes of children who have MAM in such areas are not currently described, and there remains an urgent need for evidence-based policy recommendations.MethodsWe defined MAM as mid-upper arm circumference (MUAC) of ≥11.0cm and <12.5cm with no bilateral pitting oedema to include Ethiopian government and World Health Organisation cut-offs. We prospectively surveyed 884 children aged 6–59 months living with MAM in a rural area of Ethiopia not eligible for a supplementary feeding programme. Weekly home visits were made for seven months (28 weeks), covering the end of peak malnutrition through to the post-harvest period (the most food secure window), collecting anthropometric, socio-demographic and food security data.ResultsBy the end of the study follow up, 32.5% (287/884) remained with MAM, 9.3% (82/884) experienced at least one episode of SAM (MUAC <11cm and/or bilateral pitting oedema), and 0.9% (8/884) died. Only 54.2% of the children recovered with no episode of SAM by the end of the study. Of those who developed SAM half still had MAM at the end of the follow up period. The median (interquartile range) time to recovery was 9 (4–15) weeks. Children with the lowest MUAC at enrolment had a significantly higher risk of remaining with MAM and a lower chance of recovering.ConclusionsChildren with MAM during the post-harvest season in an area not eligible for SFP experience an extremely high incidence of SAM and a low recovery rate. Not having a targeted nutrition-specific intervention to address MAM in this context places children with MAM at excessive risk of adverse outcomes. Further preventive and curative approaches should urgently be considered.  相似文献

Abstract We used the cytotoxic properties of methylazoxymethanol acetate (MAM), which ablates mitotically active neuroblasts, to eliminate neurons in the fetal striatum to define the factors that regulate the development of the synaptic circuitry of this region. Adult rats whose mothers received a single intraperitoneal injection of 20 mg/kg of MAM on gestational days (DG) 14-17 were used in this study. MAM treatment at 14 DG caused a 49% decrease in striatal mass whereas treatment at 17 DG reduced the striatal weight by only 16%; MAM treatment on 15 or 16 DG gave intermediate results. Histologic analysis of Nissl-stained sections did not reveal an obvious disruption of striatal organization, although the region was clearly hypoplastic. The hypoplasia was associated with significant increases in the specific activities of choline acetyltransferase and tyrosine hydroxylase, although total activities of these enzymes per striatum were significantly depressed with the 14 or 15 DG treatments. In contrast, the specific activity of glutamate decarboxylase was unaffected by MAM treatment whereas the total activity of this enzyme was reduced commensurate with the degree of striatal hypoplasia. In rats lesioned at 15 DG, there was a similar 30% increase in the specific activities of all presynaptic dopaminergic markers studied. In contrast, the specific activity of the synaptosomal uptake process for [3H]choline was elevated by 60%, the specific activity of choline acetyltransferase was increased by only 30%, and the concentration of acetylcholine in the striatum was unchanged. Whereas the specific activities of glutamate decarboxylase and of the synaptosomal uptake process for [3H]γ-aminobutyric acid ([3H]GABA) were unaffected by the 15 DG MAM treatment, the concentration of GABA was increased significantly by 20%. The specific binding of [3H]spiroperidol, [3H]quinuclidinyl benzilate ([3H]QNB). and [3H] muscimol to, respectively, dopamine, muscarinic, and GABA receptors was unchanged by the 15 DG MAM lesion. The nigral dopaminergic perikarya appeared unaffected by the 15 DG MAM lesion in that the tyrosine hydroxylase activity remained normal. Consistent with the loss of striatal GABAergic perikarya, the specific activities of glutamate decarboxylase and of the synaptosomal uptake process for [3H]GABA were significantly reduced in the substantia nigra; however, the concentration of endogenous GABA was twofold greater than in control in this terminal region. The results of these studies indicate that the nigro-striatal dopaminergic pathway only partially compensates for the loss of neurons in its terminal field within the hypoplastic striatum. Striatal cholinergic and GABAergic neurons differ considerably in their responses to the MAM lesion, suggesting that they are derived from different neuroblast pools. Finally, the altered synaptic relationships induced by the fetal lesion may affect neurotransmitter turnover as evidenced by disparities in GABA and acetylcholine levels when compared with other presynaptic markers for the GABAergic and cholinergic neurons.  相似文献

Abstract. Changes in the tree layer (> 1.3m) and sapling layer (< 1.3m, including seedlings) of a Swedish boreal old-growth Picea abies (Norway spruce) forest from the 1930s to the 1980s were studied in permanent plots. The plots were established in 1938–1939 and re-analysed in 1983–1988. Regeneration, mortality, turnover rate in the tree layer and amount of decomposing logs as well as the time required for complete decomposition of logs were investigated using the detailed data from the 1930s. Ca. 25 % of the trees present during the first analysis were no longer alive. This mortality was balanced by recruitment from the sapling layer. The rate of mortality suggests a turnover time for the tree layer of ca. 200 yr. The number of spruces in the sapling layer has increased by ca. 85 %, hypothetically in response to an increase in amount of decomposing wood that can serve as nurse logs and stumps. The mean time for total decomposition was calculated as ca. 200 yr. Spruce regeneration on logs does not occur until the log is at least ca. 50 yr old. The survival pattern in the sapling layer suggests a high mortality rate at the seedling stage (≤ 1 yr) and a low mortality rate at the sapling stage. In conclusion, it is suggested that the amount of coarse woody debris available for regeneration, the occurrence of seedlings, and seedling mortality constitute concurrent factors through which climatic fluctuations, in a long-term perspective, direct stand recruitment and density. As a consequence, these boreal forests will be kept in a dynamical equilibrium.  相似文献

Methylthioalkylmalate synthase (MAM) catalyzes the committed step in the side chain elongation of Met, yielding important precursors for glucosinolate biosynthesis in Arabidopsis thaliana and other Brassicaceae species. MAM is believed to have evolved from isopropylmalate synthase (IPMS), an enzyme involved in Leu biosynthesis, based on phylogenetic analyses and an overlap of catalytic abilities. Here, we investigated the changes in protein structure that have occurred during the recruitment of IPMS from amino acid to glucosinolate metabolism. The major sequence difference between IPMS and MAM is the absence of 120 amino acids at the C-terminal end of MAM that constitute a regulatory domain for Leu-mediated feedback inhibition. Truncation of this domain in Arabidopsis IPMS2 results in loss of Leu feedback inhibition and quaternary structure, two features common to MAM enzymes, plus an 8.4-fold increase in the k(cat)/K(m) for a MAM substrate. Additional exchange of two amino acids in the active site resulted in a MAM-like enzyme that had little residual IPMS activity. Hence, combination of the loss of the regulatory domain and a few additional amino acid exchanges can explain the evolution of MAM from IPMS during its recruitment from primary to secondary metabolism.  相似文献

Notch signaling releases the Notch receptor intracellular domain (ICD), which complexes with CBF1 and Mastermind (MAM) to activate responsive genes. We previously reported that MAM interacts with CBP/p300 and promotes hyperphosphorylation and degradation of the Notch ICD in vivo. Here we show that CycC:CDK8 and CycT1:CDK9/P-TEFb are recruited with Notch and associated coactivators (MAM, SKIP) to the HES1 promoter in signaling cells. MAM interacts directly with CDK8 and can cause it to localize to subnuclear foci. Purified recombinant CycC:CDK8 phosphorylates the Notch ICD within the TAD and PEST domains, and expression of CycC:CDK8 strongly enhances Notch ICD hyperphosphorylation and PEST-dependent degradation by the Fbw7/Sel10 ubiquitin ligase in vivo. Point mutations affecting conserved Ser residues within the ICD PEST motif prevent hyperphosphorylation by CycC:CDK8 and stabilize the ICD in vivo. These findings suggest a role for MAM and CycC:CDK8 in the turnover of the Notch enhancer complex at target genes.  相似文献
