5-HT5受体又分为5-HT5A及5-HT5B两种亚型受体,5-HT5A受体主要存在于小脑,大脑皮层,海马等脑区,5-HT5B受体仅在海马CA1区,缰核,背侧缝际核发现,该受体与麦角胺(Ergo-tamine),5-羧基酰氨基色胺(5-Carboxamidotryptamine,5-CT),5-HT,美西麦角(Methysergide)等配体亲和力较大,可能与5-HT 1D受体有某种功能联系,5-HT6受体主要在纹状体,边缘脑和大脑皮层等脑区表达,这类受体与三环类抗精神病药及抗抑郁药有高亲和力反应,提示它参与某些精神疾病的发生与治疗。  相似文献

To predict outbreaks of infectious disease and to prevent epidemics, it is essential not only to conduct pathological studies but also to understand the interactions between the environment, pathogen, host and humans that cause and spread infectious diseases. Outbreaks of mass mortality in carp caused by Cyprinid herpesvirus 3 (CyHV-3), formerly known as koi herpesvirus (KHV), disease have occurred worldwide since the late 1990s. We proposed an environment?CKHV?Ccarp?Chuman linkage as a conceptual model for ??environmental diseases?? and specify research subjects that might be necessary to construct and shape this linkage.  相似文献

This case report shows a genealogical study where a woman has limb hypertrophy and her son has an association of Sturge–Weber syndrome with Klippel–Trenaunay–Weber syndrome. The Sturge–Weber and Klippel–Trenaunay–Weber syndromes appear to be different manifestations of the same affliction. Familial aggregation exists and transmission may be almost imperceptible between generations. Identification of minor manifestations may prove to be a valuable contribution to genetic counseling of families and the prevention of new cases.  相似文献

Both the 5α, 6α- and 5β, 6β-dichloromethylene adducts (2a and 2b) of 3β-acetoxy-5-androsten-17-one (1) are produced when the latter is exposed to dichlorocarbene generated from chloroform and base by Phase Transfer Catalysis using ultrasound as a means of agitation. The 1H NMR substituent effects of 5α, 6α- and 5β, 6β-dichloromethylene on the angular methyl groups (Zürcher values) are given. 摘要: 通过对5个拟南芥(Arabidopsis thaliana L.)预测性多肽进行RT-PCR分析, 在mRNA水平证实了Peptide5和Peptide6预测性多肽的真实性。表达谱分析表明: 两基因在不同的发育期和不同的组织普遍表达, 为组成型基因; 对NaCl、聚乙二醇4000(PEG4000)、茉莉酸甲酯(MeJA)、水杨酸(SA)、机械损伤和冷害做出基因转录水平的响应。启动子顺式作用元件分析提示, 拟南芥Peptide5基因可能参与了次生木质部的形成。  相似文献

Electrospun nanofibrous scaffolds varying different materials are fabricated for tissue engineering. PLGA, silk fibroin, and collagen-derived scaffolds have been proved on good biocompatibility with neurons. However, no systematic studies have been performed to examine the PLGA-silk fibroin-collagen (PLGA-SF-COL) biocomposite fiber matrices for nerve tissue engineering. In this study, different weight ratio PLGA-SF-COL (50:25:25, 30:35:35) scaffolds were produced via electrospinning. The physical and mechanical properties were tested. The average fiber diameter ranged from 280 + 26 to 168 + 21 nm with high porosity and hydrophilicity; the tensile strength was 1.76 ± 0.32 and 1.25 ± 0.20 Mpa, respectively. The results demonstrated that electrospinning polymer blending is a simple and effective approach for fabricating novel biocomposite nanofibrous scaffolds. The properties of the scaffolds can be strongly influenced by the concentration of collagen and silk fibroin in the biocomposite. To assay the cytocompatibility, Schwann cells were seeded on the scaffolds; cell attachment, growth morphology, and proliferation were studied. SEM and MTT results confirmed that PLGA-SF-COL scaffolds particularly the one that contains 50% PLGA, 25% silk fibroin, and 25% collagen is more suitable for nerve tissue engineering compared to PLGA nanofibrous scaffolds.  相似文献

Incubation of o-nitrophenyl 6-deoxy-β-d-xylo-hex-5-enopyranoside (1) with emulin β-d-glucosidase gave, instead of the expected 6-deoxy-d-xylo-hexos-5-ulose (3), o-nitrophenyl 6-deoxy-3-O-(6-deoxy-β-d-xylo-hex-5-enopyranosyl)-β-d-xylo-hex-5-enopyranoside (2) in high yield (≈90% under optimal conditions). The structure of 2 was established from spectroscopic data and by correlation with compounds synthesised definitively. The specificity of the transfer reaction is discussed as an argument for an acceptor or aglycon binding-site.  相似文献

The maternal separation (MS) animal model has been widely used to study early life stress and several psychiatric conditions such as depression and anxiety. In this study, we investigated the effect of acupuncture on anxiety-related behaviors and hypothalamic-pituitary-adrenal (HPA) system in MS-induced early life stress of Sprague-Dawley rat pups (14-21 postnatal days). For determining anxiety-related behaviors, the elevated plus-maze test was performed. The effects of acupuncture on the activation of stress were measured by assessing plasma levels of corticosterone (CORT) and adrenocorticotropin hormone (ACTH). The hypothalamic immunoreactivity (IR) of arginine vasopressin (AVP) was also examined. Acupuncture was conducted at acupoint HT7, which is used to treat mental disorders in Oriental medicine, for seven consecutive days. Acupuncture significantly decreased the frequencies of open arm entries and the amount of time spent in the open arms in MS rats. In addition, acupuncture reduced CORT and ACTH levels in plasma of MS rats, and AVP-IR in the hypothalamic paraventricular nucleus of MS rats. In conclusion, acupuncture reduced anxiety-related behaviors and modulated the HPA system. These findings suggest that acupuncture at HT7 may be useful as a therapeutic treatment in MS-induced early life stress.  相似文献

Somatostatin mediates inhibitory functions through five G protein–coupled somatostatin receptors (sst1–5). We used immunohistochemistry, immunofluorescence, and RT-PCR to determine the presence of somatostatin receptors sst1, sst2A, sst2B, sst3, sst4, and sst5 in normal and IgA nephropathy human kidney. All somatostatin receptors were detected in the thin tubules (distal convoluted tubules and loops of Henle) and thick tubules (proximal convoluted tubules) in the tissue sections from nephrectomy and biopsy samples. Immunopositive sst1 and sst4 staining was more condensed in the cytoplasm of tubular epithelial cells. In normal kidney tissue sections, podocytes and mesangial cells in the glomeruli stained for sst1, sst2B, sst4 and sst5, and stained weakly for sst3. In IgA kidney tissue, the expression of somatostatin receptors was significantly increased with particular immmunopositive staining for sst1, sst2B, sst4, and sst5 within glomeruli. In the epithelial cells, the staining for sst2B and sst4 in proximal tubules and sst1, sst2B, and sst5 in distal tubules was increased. The mRNA expression of sst1–5 was also detected by RT-PCR. Somatostatin and all five receptor subtypes were ubiquitously distributed in normal kidney and IgA nephropathy. The increased expression of somatostatin receptors in IgA nephropathy kidney might be the potential pathogenesis of inflammatory renal disease. (J Histochem Cytochem 56:733–743, 2008)  相似文献

Lipid bilayer membrane is the main site where Bcl-xL executes its anti-apoptotic function. Here we used site-directed mutagenesis and cysteine-directed cross-linking to trap the structure of Bcl-xL upon membrane insertion. Cys151 on α5-helix and Asn185 on α6-helix of two neighboring Bcl-xL are found in close positions, respectively. The FRET based binding assay indicated that the BH3-peptide binding pocket in Bcl-xL is disrupted after its membrane insertion. Co-immunoprecipitation experiments showed that the membrane-bound Bcl-xL sequestered tBid by direct interaction at physiological pH. If Bcl-xL behaves similarly at low pH as it does at physiological pH, the membrane-bound Bcl-xL should bind to tBid through protein regions other than the BH3 domain of tBid and the hydrophobic pocket of Bcl-xL. Previously, a crystallography study demonstrated that Bcl-xL formed homodimers through domain swapping in water, where Cys151 and Asn185 of two monomeric subunits are far apart from each other and the BH3-peptide binding pocket is intact. Our results indicated that Bcl-xL dimer trapped by cross-linking in lipids is distinct from the domain swapped dimer, suggesting that Bcl-xL transits through a structural change from the water-soluble state to the membrane-bound state and there are multiple possibilities for structural reorganization of Bcl-xL protein.  相似文献

Summary Ecto-5-nucleotidase activity (5NT) was measured on whole cells of 26 human x Chinese hamster hybrids. Concordance analysis showed 100% correlation between enzyme activity and inheritance of human chromosome 6. This observation was confirmed by a segregation analysis in which cells of a hybrid containing chromosome 6 were stained by indirect immunofluorescence for HLA Class 1 antigen and sorted by a fluorescence-activated cell sorter (FACS). Cells in the HLA- compartment were cloned and expression of HLA and 5NT was determined. Of nine clones, three were HLA-, 5NT- and six were HLA+, 5NT+, supporting the linkage of 5NT to chromosome 6.  相似文献

Polarographic half-wave potentials for two reduction steps of 49 cyclopentadienyliron complexes of substituted arenes or heterocycles in dimethylformamide were determined. A fast cyclic voltammetry (10–40 V/s) was used to study the electron transfer kinetics of some of these complexes. After correction for the double layer effects, the rate constants of all the complexes studied show that the transfer of electrons in the second reduction step occurs significantly faster than in the first one. This was interpreted as a result of greater delocalization of electrons in the 20-electron complexes of iron compared to the 19-electron complexes.  相似文献

Human ribosomal protein S6 kinase includes two protein families: P90RSK and P70S6K, they participate in two different signaling pathways. When the two kinases were inhibited by their antibodies or rapamycin, the proliferation of cells was arrested. However, their analog, the immunosupressant FK-506, can inhibit the proliferation of fibroblast PBL1 without interfering with the activities of P90RSK, P70S6K and MAPK. We take the tactics of "homolog screening" to demonstrate whether there are some novel proteins which can substitute for the known P90RSK and P70S6K or other pathways without interfering with the known P90RSK and P70S6K. With the conserved sequence of mouse p90RSK as a probe, we screened the homologous sequence in NCBI EST database and got three human EST fragments. With the assembled contig as a probe to screen human brain cDNA library, a full-length cDNA of 3833 bp was attained. It contains a completed open reading frame from 165 bp to 2570 bp encoding 802 amino acids. The putative protein has higher homology with other members of p90RSK family. The gene was named RPS6KA5, the accession number in GenBank is AF090421. Northern hybridization showed the gene expressed in 16 human tissues tested, and the gene was localized in 14q31-32.1 by RH mapping. Another novel P70S6K gene has also been cloned. Thus, our initial presumption that there is an analog of known P90RSK and P70S6K in human beings was proved.  相似文献

人类核糖体S6激酶包括两个蛋白家族P90RSK和P70S6K,它们分别介导着两条细胞信号传导通路。当这些激酶活性被它们的抗体或纳巴霉素抑制时,细胞的增殖随之停止。但当纳巴霉素的结构类似物-免疫抑制剂FK-506作用于细胞时,虽可抑制成纤维细胞PBLl的增殖,但却不能抑制P90RSK、P70S6K和MAPK的活性。这提示体内还存在着已知P90RSK和P70S6K蛋白的替代者或还存在着不涉及已知P90RSK和70S6K的信号通路。为此,本文采用“同源筛选”策略,试图证实上述推测。我们以小鼠P90RSK基因的保守性序列为探针,在NCBIEST数据库中进行同源筛选,得到三个人体同源EST片段。以EST片段的整合序列为探针,在人脑组织cDNA文库中进行杂交筛选,最终获得3833bp的全长cDNA序列,其中第165-2570bp为一完整的开放阅读框,编码了802个氨基酸。这个推导蛋白质与人P90RSK家族成员具有较高氨基酸同源性,并被命名为RPS6KA5,它在国际GenBank的登录号为AF090421,Northern杂交显示该基因在人各组织中广泛表达,RH定位将该基因定于14号染色体长臂31-32.1的范围内,另一新的P70S6K基因(GenBank注册登记号为AF037447)也已被克隆,从而证实了人体内存在着已知P90RSK及P70S6K家族基因替代者的最初设想。  相似文献
