The systematic position of the genus Cryothecomonas has been determined from an analysis of the nuclear-encoded small subunit ribosomal RNA gene of Cryothecomonas longipes and two strains of Cryothecomonas aestivalis. Our phylogenetic trees inferred from maximum likelihood, distance and maximum parsimony methods robustly show that the genus Cryothecomonas clusters within the phylum Cercozoa, and is related to the sarcomonad flagellate Heteromita globosa. Morphological data supporting the taxonomic placement of Cryothecomonas near the sarcomonad flagellates has been compiled from the literature. The high number of nucleotide substitutions found between two morphologically indistinguishable strains of Cryothecomonas aestivalis suggests the possibility of cryptic species within Cryothecomonas aestivalis.  相似文献

The genus Chlamydomonas (including Chloromonas) is one of the largest green algal genera comprising more than 600 species. To initiate a comprehensive analysis of the phylogeny and systematics of the genus, we determined nuclear-encoded SSU rRNA sequences from 32 strains of Chlamydomonas, Chloromonas and Chlorogonium with emphasis on oogamous taxa and related strains, and incorporated these into global molecular phylogenetic analyses of 132 strains of Chlorophyceae. In addition, we studied the morphology and reproduction of oogamous and related strains by light microscopy. We recognize and designate 18 monophyletic lineages (clades) within the Chlorophyceae, 11 of which are confined to the CW (basal bodies displaced clockwise) subgroup. The majority of clades recognized within the Chlorophyceae do not correspond to any of the traditional classification systems, which are still largely based on the organization level. Strains assigned to Chlamydomonas and Chloromonas were found in seven different clades confirming the polyphyly of the two genera as presently conceived. To initiate the taxonomic revision of Chlamydomonas, C. reinhardtii is proposed as the conserved type of the genus. In consequence, species in clades other than the clade containing C. reinhardtii must be transferred to other genera, a process initiated in this contribution. The oogamous strains studied represent a monophyletic lineage, which is described as Oogamochlamys gen. nov. comprising three species (O. gigantea, O. zimbabwiensis and O. ettlii spec. nov.). The sister clade to Oogamochlamys consists of isogamous strains characterized by chloroplasts with incisions and is described as Lobochlamys gen. nov. with two species (L. culleus and L. segnis). Another clade is characterized by asteroid or perforated, parietal chloroplasts and contains the type species of Chloromonas (C. reticulata). Thus, the polyphyletic Chloromonas (traditionally defined as “Chlamydomonas without pyrenoids”) can be legitimized as a monophyletic genus by restriction to this clade and is here emended on the basis of chloroplast characters (the clade contains strains with or without pyrenoids thus rejecting the character “absence of pyrenoids”).  相似文献

红曲霉属Monascus具有重要的经济和生态价值,但对该属的物种识别和系统发育学研究有限。本研究首先对红曲霉属红色组内物种开展ITS和LSU序列的测定并综合分析,结合GenBank中相关物种序列及菌株的形态学特征,探讨红曲霉属的系统发育关系,鉴定出红色组3个种,弗罗里达组7个种。其次采用红曲霉色素表型控制基因簇部分基因pksKS序列进行分析,以期解决ITS和LSU等基因序列分析无法有效区分红色红曲与紫色红曲不同形态种的问题。通过分子克隆测序与直接测序结果的比较,以及对3个血红红曲菌株单核苷酸多态性的分析,解析了血红红曲种内遗传差异,首次在红曲霉属真菌中发现疑似红色红曲血红自然杂交种。最后综合ITS、LSU、pksKS序列和形态学分析的结果,统一了红曲霉属内物种的系统发育关系,确认红色组包括3个种:红色红曲M. ruber、紫色红曲M. purpureus、血红红曲M. sanguineus;发现3个变种:红色红曲发白变种M. ruber var. albidulus、紫色红曲橙色变种M. purpureus var. aurantiacus及紫色红曲火红色变种M. purpureus var. rutilus;弗罗里达组包括7个种:弗罗里达红曲M. floridanus、苍白红曲M. pallens、新月红曲M. lunisporas、阿根廷红曲M. argentinensis、累西腓红曲M. recifensis、黄色红曲M. flavipigmentosum、蜂蜜红曲M. mellicola。另外,发现1个疑似杂交种:红色红曲血红杂种M. ruber × sanguineus。结果表明,红曲霉属具有丰富的物种多样性,而且通过多基因和形态学分析可以将相近种区分开来。  相似文献

Five indigenous fungal strains with characteristics of the genus Penicillium capable of degrading and utilizing pyrene, as sole carbon source were isolated from soil of a former gas work site. Two strains were identified as Penicillium ochrochloron. One of the strains was able to degrade a maximum of 75% of 50 mg l−1 pyrene at 22 °C during 28 days of incubation. The presence of pyrene in the medium resulted in an aggregation of hyphae into pellets by the two Penicillium ochrochloron strains. Formation of pellets was observed after 48 h of incubation with difference in size and texture between the two strains. This indicated the individual variation within the same genus of fungi. However, remaining strains did not show this behavior even though they were capable of utilizing pyrene as sole carbon source. The macro- and microscopic morphology of fungal pellets was studied using scanning electron microscopy. It was found that the addition of varying concentration of pyrene ranging from 10 to 50 mg l−1 in the medium influenced shape and structure of the mycelial pellets. A two-fold increase in hyphal branching (with concomitant decrease in the average hyphal growth unit) was observed at a concentration of 10 mg l−1. The relevance of fungal growth and morphology for bioremediation of polycyclic aromatic hydrocarbons (PAHs) contaminated sites are discussed.  相似文献

Torreya Arn., a small genus of Taxaceae, consists of six species occurring in North America and eastern Asia. Several phylogenetic studies have previously been undertaken to reveal relationships within this genus, although only a few DNA segments or species were used. In the present study, we sequenced five Torreya plastomes and combined these with two existing plastomes from the genus to investigate plastome evolution and phylogenetic relationships within Torreya. All sequenced Torreya plastomes shared the same complement of 82 protein‐coding genes, 4 ribosomal RNA genes, and 31 transfer RNA genes. Phylogenetic inference using a maximum likelihood framework consisted of an 82‐gene, 17‐taxon dataset, including all species of Torreya, resolved Torreya as a monophyletic clade. Strongly supported relationships within the genus include the position of the early diverging T. jackii Chun, the two sister pairs T. fargesii Franch.–T. nucifera (L.) Siebold & Zucc. and T. grandis Fortune ex Lindl.–T. californica Torr., and the monophyly of the clade including T. fargesii var. yunnanensis, T. fargesii, and T. nucifera. In addition to the inference of species relationships, divergence time estimation and biogeographical analysis were carried out. The diversification of Torreya was estimated to be approximately 8.9 Ma. Ancestral state reconstruction of the geographical area suggested China/eastern North America as the most likely ancestral region for the six extant Torreya species.  相似文献

Two species of the genus Antechiniscus have been found in Nothofagus forests in Australia. The new species A. moscali sp. n. is described. The New Zealand species A. parvisentus (Horning & Schuster, 1983) is redescribed from type material and from the new material from Australia. Nine psychrotolerant bacteria were isolated from fast ice in the middle of Geologie Archipelago, Adelie Land, Antarctica and were categorized into two groups, based on their SDS-PAGE profiles. Representatives from each of the two groups, namely strains DD 48T and SJ 14T exhibited phenotypic and chemotaxonomic characteristics confirming to the genus Psychrobacter. The 16S rRNA gene sequence indicated that the two isolates are closely related to each other and to the already reported fifteen species of Psychrobacter. Detailed studies on the phenotypic characteristics, chemotaxonomic properties and phylogenetic analysis of strains DD 48T and SJ 14T indicated that they are distinctly different from each other and the reported species of Psychrobacter. At the DNA-DNA hybridisation level, the two species exhibit less than 70% similarity. Thus, strains DD 48T and SJ 14T are identified as new species of the genus Psychrobacter for which the names Psychrobacter salsus sp. nov. and Psychrobacter adeliensis sp. nov. respectively are proposed.  相似文献

Seventy-three strains of cryptophytes assigned to the genera Cryptomonas, Campylomonas or Chilomonas were studied by light microscopy, spectrophotometry and whole-mount electron microscopy. Twelve groups of strains were distinguished by light and whole mount electron microscopy using a combination of characters, mainly cell size, type of periplast and presence/absence and number of pyrenoids. However, characters previously used to distinguish Cryptomonas from Campylomonas (e.g. the type of periplast: polygonal periplast plates vs. a continuous periplast sheet) were found to occur together in dimorphic strains, indicating that periplast types relate to different life-history stages of a single taxon. To evaluate the taxonomic significance of the type of periplast and other characters previously used to distinguish genera and species, representatives of each strain group were subjected to molecular phylogenetic analyses using two nuclear ribosomal DNA regions (ITS2, partial LSU rDNA) and a nucleomorph ribosomal gene (SSU rDNA). The results of the phylogenetic study provide molecular evidence for a life history-dependent dimorphism in the genus Cryptomonas: the genus Campylomonas represents the alternate morph of Cryptomonas. Campylomonas and Chilomonas are reduced to synonyms of Cryptomonas, the genus Cryptomonas is revised and typified, two new species are described and six species are emended.  相似文献

Rapid and reliable identification of Staphylococcus (S.) equorum was achieved by species-specific PCR assays. A set of primers targeting the manganese-dependent superoxide dismutase (sodA) gene of S.equorum was designed. Species-specificity of the primer set was evaluated by using a total of 112 strains (including 27 reference strains of the DSM collection), representing 26 different species of the genus Staphylococcus, 3 species of the genus Kocuria, and different strains of Macrococcus caseolyticus. By using primers SdAEqF and SdAEqR the expected PCR fragment was obtained only when DNA from S. equorum strains was used as template. The rapidity (about 4 h from DNA isolation to results) and reliability of the PCR procedures established suggests that the method may be profitably applied for specific detection and identification of S. equorum strains.  相似文献

We have isolated and identified 12 previously unreported strains of polycyclic aromatic hydrocarbon (PAH)-degrading bacteria. Three of these isolates belong to the genus Burkholderia, with the remainder comprised of two Pseudomonas species, and seven strains from the genus Sphingomonas. These isolates were examined for their ability to utilize a variety of three- to five-ring PAHs as sole sources of carbon and energy. All were also checked for the ability to grow on salicylic acid and phthalic acid, two compounds that are key intermediates in almost all published PAH degradative pathways. Only 3 of the 12 strains were able to grow on both of these monoaromatic acids, while three others (all Sphingomonas) could not grow on either. The implications of these findings on the universal applicability of known PAH catabolic pathways, and on possible induction of PAH degradation by pathway intermediates, are discussed.  相似文献

Two strains designated strains L-1T and L-9T were isolated from activated sludge of a treatment plant that receives wastewater from the tannery industry contaminated with chromium. Phylogenetic analysis showed that the organisms represented two new species of the genus Leucobacter. Strains L-1T and L-9T could be distinguished from the type strain of L. komagatae and from the type strain of “L. albus” by the B-type peptidoglycan composition, fatty acid composition, several phenotypic and physiological characteristics. The major fatty acids of the organisms were iso- and anteiso-branched C15:0 and C17:0, straight-chain C16:0 was also found in relatively high proportions. The organisms were halotolerant, grew in medium containing 9% NaCl, and all strains, including the type strain of L. komagatae grew in medium containing 5 mM Cr(VI). On the basis of the distinct peptidoglycan composition, 16S ribosomal DNA sequence analysis, percentage of DNA-DNA reassociation values, and phenotypic characteristics we are of the opinion that strain L-1T represents a new species of the genus Leucobacter for which we propose the name Leucobacter chromiireducens and that strain L-9T represents an additional new species of the same genus for which we propose the name Leucobacter aridicollis.  相似文献

tDNA intergenic spacer PCR (tDNA-PCR) using consensus primers complementary to the conserved edges of the tRNA genes can amplify the intergenic spacers. Separation of the PCR products with capillary electrophoresis enables discrimination between fragments differing only one basepair in length. This method was applied to a collection of 82 Lactobacillus strains belonging to 37 species in order to evaluate the discriminatory power of this technique within this genus. Twenty-one species could be distinguished to species level on the basis of a unique tDNA fingerprint pattern. The other species grouped by two (e.g. L. fermentum and L. cellobiosus) or three (L. acidophilus, L. gallinarum and L. helveticus). Inclusion of the resulting fingerprints in a numerical database containing fingerprints of numerous other Gram-positive and Gram-negative species makes the identification of unknown strains possible.  相似文献

The genus Cayratia Juss. in the traditional sense (i.e., Cayratia s.l.) of the grape family has been shown to be non‐monophyletic. Previous studies supported the splitting of Cayratia s.l. into three genera, that is, Cayratia s.s., Causonis Raf., and a new genus representing the African Cayratia clade. However, the morphology of the African Cayratia clade has not been studied carefully and its phylogenetic position within Vitaceae remains unclear. Our study integrates molecular, distributional, and morphological data and supports the recognition of the new genus Afrocayratia from continental Africa and Madagascar. Phylogenetic analyses strongly support the monophyly of Afrocayratia and resolve it as a sister of Cayratia s.s. based on the chloroplast data, but it is placed sister to Cyphostemma based on the internal transcribed spacer dataset. Molecular dating suggests that Afrocayratia split with Cayratia s.s. during the Paleocene, but that the extant species of Afrocayratia did not diversify until the early Miocene. Afrocayratia differs from its allied genera in having short stigmas and seeds with subcircular ventral infold cavities in cross‐section. Three clades are detected within Afrocayratia, with A. debilis (Baker) J.Wen & L.M.Lu as the first diverged lineage. The second diverged lineage includes A. delicatula (Willems) J.Wen & Z.D.Chen and A. gracilis (Guill. & Perr.) J.Wen & Z.D.Chen. The third diverged lineage includes A. imerinensis (Baker) J.Wen & L.M.Lu, A. longiflora (Desc.) J.Wen & Rabarijaona, and A. triternata (Baker) J.Wen & Rabarijaona from Madagascar, which form a monophyletic group that diverged from the second lineage in the middle Miocene. Combining the morphological and molecular evidence, we formally describe the new genus Afrocayratia, make seven new combinations, and provide a key to species of the genus.  相似文献

高秆野生稻(Oryza alta)是一种重要的种质资源, 其组织内也蕴藏着非常宝贵的功能微生物资源。本实验采用无氮培养基, 从高秆野生稻中分离到43株内生固氮菌, 结合乙炔还原法测定其固氮酶活性。经固氮酶基因(nifH)的PCR扩增检测, 43株内生固氮菌代表菌株均能扩增出固氮酶基因片段。利用IS-PCR DNA指纹图谱和SDS-PAGE全细胞蛋白电泳图谱将获得的菌株聚类为6个类群(I、II、III、IV、V、VI)。对各个类群的代表菌株(ZF3, ZF8, ZF13, ZF15, ZF24, ZF43)进行16S rRNA基因序列测定, 结果表明, 类群I属于土生拉乌尔菌(Raoultella terrigena), 类群II属于类肺炎克雷伯氏菌类肺炎亚种(Klebsiella quasipnmoniae subsp. quasipneumoniae), 类群III属于越南伯克氏菌(Burkholderia vietnamiensis), 类群IV的菌株代表肠秆菌属的一个类群(Enterobacter sp.), 类群V的菌株属于门多萨假单胞菌(Pseudomonas mendocina), 类群VI归属于固氮植物菌(Phytobacter diazotrophicus)。Biolog聚类结果与IS-PCR指纹图谱类型及SDS-PAGE全细胞蛋白聚类结果一致。Biolog板测定结果显示, 来自不同类群的代表菌株对碳源的利用差异显著, 说明野生稻内多样的内生固氮菌从环境中获取碳源和氮素的适应能力较强。  相似文献

Legumes from the genus Pachyrhizus, commonly known as yam bean, are cultivated in several countries from the American continent and constitute an alternative source for sustainable starch, oil and protein production. The endosymbionts of these legumes have been poorly studied although it is known that this legume is nodulated by fast and slow growing rhizobia. In this study we have analyzed a collection of strains isolated in several countries using different phenotypic and molecular methods. The results obtained by SDS-PAGE analysis, LPS profiling and TP-RAPD fingerprinting showed the high diversity of the strains analyzed, although all of them presented slow growth in yeast mannitol agar (YMA) medium. These results were confirmed using 16S-23S internal transcribed spacer (ITS) region and complete sequencing of the 16S rRNA gene, showing that most strains analyzed belong to different species of genus Bradyrhizobium. Three strains were closely related to B. elkanii and the rest of the strains were related to the phylogenetic group constituted by B. japonicum, B. liaoningense, B. yuanmingense and B. betae. These results support that the study of rhizobia nodulating unexplored legumes in different geographical locations will allow the discovery of new species able to establish legume symbioses.  相似文献

本文叙述了杭子梢属(gen.Campylotropis)植物的研究历史,说明在中国一共记录过59种杭子梢属植物,经过著者的研究整理,确定并提出了14种新的异名名称,经过详细考订,本属植物在中国已确知为29种、6变种、6变型。其中包括经发现和研究整理的新种4、新变种3、新变型3、新组合变种2、新组合变型3。此外尚有2个存疑种有待今后采集调查解决。本文历述了关于本属是否应该并入胡枝子属(gen.Lespedeza)这一问题所存在的长期争议之后,详细论述了本属与胡枝子属在特征上的异同诸点、进行了比较和分析,据此,明确地肯定本属与胡枝子属亲缘关系极为相近,但特征的分化已经必须确认本属应是与胡枝子属有明确区别的独立的属。此外,从特征分析中也可得出本属应是由胡枝子属大胡枝子组(sect.Macrolespedeza)演化发展而来的。本文在讨论了属内主要特征的演化趋势与种群关系之后,论述了本属在世界主要分布地区的种数和分布概况,同时将此与胡枝子属大胡枝子组的分布概况作了比较和分析,明确地认定我国机子梢属植物的大部分种类是本国西南地区发生的(中国有20个特有种),我国的西南地区是本属植物的分布中心,也是本属植物从较原始特征开始分化的一个特征分化多样化中心,并且此地区正是胡枝子属大胡枝子组与本属多数种类包括较原始种类分布相交叉的地区,因而我国的西南地区有可能即是本属植物的起源中心,并且这一起源应是与胡枝子属大胡枝子组植物分布到中国西南地区以后的分化发展想联系的。  相似文献

在前人对列当科系统发育研究的基础上,追加了肉苁蓉属(Cistanche)的基因序列数据,运用最大简约法、最大似然法和贝叶斯推断方法探讨了其在列当科中的系统位置及列当科中属间关系。基于rps16基因序列及rps16+ITS联合序列建立了列当科系统发育树,结果显示,肉苁蓉属、列当属(Orobanche)以及草苁蓉属(Boschniakia)聚在同一进化枝内,肉苁蓉属和列当属表现出最近亲缘关系;列当科中的全寄生类群、半寄生类群和非寄生类群分属在3个不同分支中。  相似文献

Although phylogenetic studies have revealed major clades, the deepest relationships in Isoetes remain unresolved. The use of next-generation sequencing provides enormous amounts of gene sequences, which allows not only clarification of the basal relationships but also rapid radiations. Plastomes of six key Isoetes species were annotated, revealing a total of 129 or 130 genes, depending on the species. Our phylogenomic analyses comprising representatives of all major clades yielded well-supported nodes and identical topologies using maximum likelihood and Bayesian inference. The phylogenetic reconstructions detangled the deep relationships in Isoetes and illuminated the more recent radiations in the genus. A basal dichotomy was found that grouped Isoetes spp. from Brazil and South Africa into a clade sister to the remaining Isoetes groups. Interestingly, I. andicola was found to be sister to the North American species complex. Genomic trait mapping analysis showed that the missing introns in the atpF and clpP genes were well conserved in two major clades. The absence of trnK-UUU was observed in the Brazilian tropical species and in I. velata. Among lycophytes, the gene trnR-CCG was missing only in I. eludens. In general, genomic traits such as the presence or absence of internal stop codons, a tRNA, and an intron were revealed to be conserved within groups, suggesting that these genomic traits might reveal vital information about the evolution of the genus. This study will contribute to understanding the diversification of Isoetes and the establishment of a better framework to address the evolutionary history of the genus.  相似文献
