Summary AMycobacterium sp., which was previously isolated from oil-contaminated estuarine sediments, mineralized the polycyclic aromatic hydrocarbon fluoranthene. When supplemented with an alternative carbon source, the organism was able to mineralize up to 78% of the added [3-14C]fluoranthene to14CO2 after 5 days of incubation, with relatively little accumulation of intermediate metabolites. The distribution of the C-14 label was monitored throughout the mineralization process. TheMycobacterium degraded in excess of 95% fluoranthene within a 24 hour period following an initial 6–12h lag phase. At that point approximately 53% of the radioactivity was located in the ethyl acetate extractable fraction, 31.8% in CO2, and 14.7% in the aqueous phase. Incubation of theMycobacterium sp. with soil and river water, in the presence of fluoranthene, enhanced mineralization of fluoranthene by 92.7% over the indigenous biota. These results, in conjunction with previously reported studies, suggest the potential application of thisMycobacterium sp. for the bioremediation of polycyclic aromatic hydrocarbon contaminated wastes in the environment.  相似文献

Pyrene and fluoranthene, when supplied as the sole carbon source, were not degraded by Burkholderia sp. VUN10013. However, when added in a mixture with phenanthrene, both pyrene and fluoranthene were degraded in liquid broth and soil. The amounts of pyrene and fluoranthene in liquid media (initial concentrations of 50 mg l−1 each) decreased to 42.1% and 41.1%, respectively, after 21 days. The amounts of pyrene and fluoranthene in soil (initial concentrations of 75 mg kg−1 dry soil each) decreased to 25.8% and 12.1%, respectively, after 60 days. None of the high molecular weight (HMW) polycylic aromatic hydrocarbons (PAHs) tested adversely affected phenanthrene degradation by this bacterial strain and the amount of phenanthrene decreased rapidly within 3 and 15 days of incubation in liquid broth and soil, respectively. Anthracene also stimulated the degradation of pyrene or fluoranthene by Burkholderia sp. VUN10013, but to a lesser extent than phenanthrene. The extent of anthracene degradation decreased in the presence of these HMW PAHs.  相似文献

A novel indole dioxygenase (idoA) gene has been cloned from Pseudomonas alcaligenes PA-10, based on its ability to convert indole to indigo. The chromosomally encoded idoA gene exhibits no similarity to previously cloned naphthalene dioxygenases or to aromatic oxygenases from other species at the nucleotide level. Phylogenetic analysis indicates that the idoA gene product is most similar to an acyl-CoA dehydrogenase from Novosphingobium aromaticivorans. The enzyme encoded by the idoA gene is essential for the metabolism of fluoranthene, since a mutant in which the idoA gene has been disrupted looses the ability to degrade this compound. The idoA gene appears to be constitutively expressed in PA-10, but its expression is also subject to regulation following prior exposure to salicylate and to fluoranthene degradative intermediates.  相似文献

Two surfactants, Tween 80 and JBR, were investigated for their effect on fluoranthene degradation by a Pseudomonad. Both surfactants enhanced fluoranthene degradation by Pseudomonas alcaligenes PA-10 in shake flask culture. This bacterium was capable of utilising the synthetic surfactant and the biosurfactant as growth substrates and the critical micelle concentration of neither compound inhibited bacterial growth. The biosurfactant JBR significantly increased polycyclic aromatic hydrocarbon (PAH) desorption from soil. Inoculation of fluoranthene-contaminated soil microcosms with P. alcaligenes PA-10 resulted in the removal of significant amounts (45 ± 5%) of the PAH after 28 days compared to an uninoculated control. Addition of the biosurfactant increased the initial rate of fluoranthene degradation in the inoculated microcosm. The presence of a lower molecular weight PAH, phenanthrene, had a similar effect on the rate of fluoranthene removal.  相似文献

Two bacterial strains,Pyl and Py4,have been tamed and isolated through long cultivation with polycyclic aromatic hydrocarbon-pyrene as the single carbon source.It has been proven that they are both highly-efficient pyrene degrading bacteria and both Bacillus sp..The pyrene degradation ability of separated Pyl,Py4 and the consortium of equal Pyl and Py4 was studied in this project.It is shown that pyrene degradation rates were 88% in 10hr by Py1,84% in 14hr by Py4,and 88% in 8hr by the consortium.It was also determined that the best degradation temperatures were 37℃ and pH 7.0 respectively.The influence of different nutrient substrates added in the degradation experiments was also studied.It was shown that sodium salicylate,sodium acetate and yeast exuact had obvious simulative effect,but glucose had no obvious effect.  相似文献

An investigation was carried out for in vitro degradation of fluoranthene by four bacterial strains (PSM6, PSM7, PSM10 and PSM11) isolated from the petroleum sludge. Although all the strains registered their growth in MSM with 100 ppm fluoranthene, PSM11 growth was better than other strains. Growth of bacterial strains invariably corresponded to their degradation potential of fluoranthene. After 168 h of incubation, 61% fluoranthene was degraded by PSM11, followed by PSM10 (48%) and PSM6 (42%) and the least was recorded in PSM7 (41%). Besides, 11% loss in fluoranthene was attributed to abiotic factors. Thirty-eight times more activity of catechol 2,3-dioxygenase than catechol 1,2-dioxygenase showed that it played a significant role in fluoranthene degradation. Molecular weight of catechol 2,3-dioxygenase isolated from PSM11 was determined as ∼136 kDa by size exclusion chromatography and 34 kDa on denaturing SDS-PAGE, indicating tetrameric nature of the enzyme.  相似文献

Biodegradation of a mixture of PAHs was assessed in forest soil microcosms performed either without or with bioaugmentation using individual fungi and bacterial and a fungal consortia. Respiratory activity, metabolic intermediates and extent of PAH degradation were determined. In all microcosms the low molecular weight PAH’s naphthalene, phenanthrene and anthracene, showed a rapid initial rate of removal. However, bioaugmentation did not significantly affect the biodegradation efficiency for these compounds. Significantly slower degradation rates were demonstrated for the high molecular weight PAH’s pyrene, benz[a]anthracene and benz[a]pyrene. Bioaugmentation did not improve the rate or extent of PAH degradation, except in the case of Aspergillus sp. Respiratory activity was determined by CO2 evolution and correlated roughly with the rate and timing of PAH removal. This indicated that the PAHs were being used as an energy source. The native microbiota responded rapidly to the addition of the PAHs and demonstrated the ability to degrade all of the PAHs added to the soil, indicating their ability to remediate PAH-contaminated soils.  相似文献

Twenty different strains of Pseudomonas, Mycobacterium, Gordona, Sphingomonas, Rhodococcus and Xanthomonas which degrade polycyclic aromatic hydrocarbons (PAH) were characterized in respect to genes encoding degradation enzymes for PAH. Genomic DNA from these strains was hybridized with a fragment of ndoB, coding for the large iron sulfur protein (ISP alpha) of the naphthalene dioxygenase from Pseudomonas putida PaW736 (NCIB 9816). A group of seven naphthalene-degrading Pseudomonas strains showed strong hybridization with the ndoB probe, and five Gordona, Mycobacterium, Rhodococcus and Pseudomonas strains able to degrade higher molecular weight PAH showed weaker hybridization signals. Using a polymerase chain reaction (PCR) approach, seven naphthalene-degrading Pseudomonas strains showed a PCR fragment of the expected size with ndoB-specific primers and additionally ten strains of Gordona, Mycobacterium, Pseudomonas, Sphingomonas and Xanthomonas able to degrade higher molecular weight PAH were detected with degenerate primer-pools specific for the ISP alpha [2Fe-2S]-Rieske center of diverse aromatic hydrocarbon dioxygenases. This suggests a molecular relationship between genes coding for PAH catabolism in various PAH-degrading bacterial taxa, which could be used to evaluate the PAH-degradation potential of mixed populations.  相似文献

选用模式植物拟南芥为材料,以四环的多环芳烃(PAHs)荧蒽为研究对象,从植物对非生物胁迫响应紧密相关的抗氧化酶及膜保护系统的变化入手,研究了植物对多环芳烃胁迫的生理响应.结果表明:荧蒽胁迫下拟南芥经历了氧化胁迫和膜脂过氧化过程.0.75 mmol·L-1的荧蒽使拟南芥光合作用过程受到抑制;1.00 mmol·L-1的荧蒽使拟南芥丙二醛(MDA)含量极显著增加, 抗坏血酸过氧化物酶(APX)活性极显著下降, 膜脂过氧化作用加剧,1.25 mmol·L-1的荧蒽使拟南芥过氧化物酶(POD)活性极显著下降,H2O2在细胞内累积,拟南芥明显受害.  相似文献

The release of 14CO2 from 9-[14C]phenanthrene, 4,5,9,10-[14C]pyrene and 7-[14C]benzo[a]pyrene, added to Brent/Fortes crude oil and mixed into a pristine sand soil (0.40% organic C) and a pristine organic soil (22.9% organic C), was determined. After 244 days at 25 degrees C, 11.1 +/- 3.5% (sand) and 17.1 +/- 0.30% (organic) phenanthrene-14C and 9.77 +/- 2.8% (sand) and 5.86 +/- 1.4% (organic) benzo[a]pyrene-14C was released. After 210 days, 3.65 +/- 0.5% (sand) and 4.43 +/- 0.33% (organic) pyrene-14C was released. Inoculation of these two soils with DC1 and PD2 (bacteria capable of accelerating the phenanthrene and pyrene mineralisation in soil in the absence of crude oil) either at day 0 or after release as 14CO2 by indigenous degraders had ceased, failed to increase or initiate further mineralisation. Thus, aged PAH residues were non-bioavailable to these metabolically competent degrading microorganisms. At the end of the first period of incubation (210 days or 244 days), the total aromatic hydrocarbons recovered using Soxhlet extraction was 0.18% (sand) and 42.8% (organic) compared with approximately 100% from bio-inhibited soils. This confirmed that the indigenous microbiological activity not only caused a limited amount of PAH mineralisation but also reduced the extractability of residues, possibly due to the generation of metabolites which were chemisorbed and bound (and non extractable) in 'aged' soils.  相似文献

The fluorescence in situ hybridization (FISH) technique with whole chromosome painting for chromosomes #1 and #4 was used to study the impact of air pollution containing higher concentrations of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs) in three European cities, Prague (Czech Republic), Kosice (Slovakia) and Sofia (Bulgaria). In each site were followed an exposed group, who were police officers or bus drivers who work usually through busy streets for at least 8 h, and a reference group, who spent more than 90% of their daily time indoors.

In Prague, a significant increase was observed in percentage of aberrant cells (% AB.C.) in the police officers compared to the reference group (0.33 ± 0.25 versus 0.24 ± 0.18, p < 0.05). In Kosice, the exposed group differed from reference in the endpoints FG/100 1.52 ± 1.18 versus 1.12 ± 1.30, p < 0.05; % AB.C. 0.30 ± 0.19 versus 0.21 ± 0.20, p < 0.05; t/1000 3.91 ± 3.18 versus 2.84 ± 3.10, p < 0.05. In Sofia were followed two exposed groups: police officers and bus drivers. All FISH endpoints were significantly higher in police officers compared to reference group (FG/100 1.60 ± 0.99 versus 0.82 ± 0.79, p < 0.01; % AB.C. 0.25 ± 0.14 versus 0.13 ± 0.13, p < 0.01; t/1000 4.19 ± 2.65 versus 2.13 ± 2.05, p < 0.05; rcp 1.46 ± 1.07 versus 0.70 ± 0.76, p < 0.05). In bus drivers compared to reference there was an increase in % AB.C. (0.25 ± 0.18 versus 0.13 ± 0.13, p < 0.05).

This is the first study when FISH method was used to analyze the impact of environmental air pollution. According to the original hypothesis it is expected that the most important group of chemicals responsible for the biological activity of air pollution represent c-PAHs.  相似文献
