Effects of sublethal azadirachtin exposure to the biological performance of Tetranychus urticae Koch was studied under laboratory conditions. Bioassay was used to asses the effect of different concentrations of azadirachtin on longevity, fecundity, fertility, and offspring development. Azadirachtin (64 and 128 ppm) affected fecundity and mortality but had no effect on fertility and offspring development. A subsequent life-table study with 80 ppm of azadirachtin found that the compound caused a reduction of 50% in survival to adult stage. The peak of reproduction was reached at 5 days causing a decrease in the mean fecundity to almost eight times than of untreated females. The net reproductive rate (R
0), the intrinsic rate of increase (r
m), and the finite rate of increase () of treated females were lower. Treatment showed a negative value of r

Tetranychus evansi Baker and Pritchard and Tetranychus urticae Koch (Acari: Tetranychidae) are important pests of Solanaceae in many countries. Several studies have demonstrated that T. urticae is an acceptable prey to many predatory mites, although the suitability of this prey depends on the host plant. T. evansi, has been shown to be an unfavorable prey to most predatory mites that have been tested against it. The predator Phytoseiulus fragariae Denmark and Schicha (Acari: Phytoseiidae) has been found in association with the two species in Brazil. The objective of
this work was to compare biological parameters of P. fragariae on T. evansi and on T. urticae as prey. The study was conducted under laboratory conditions at 10, 15, 20, 25 and 30°C. At all temperatures, survivorship
was lower on T. evansi than on T. urticae. No predator reached adulthood at 10°C on the former species; even on the latter species, only about 36% of the predators
reached adulthood at 10°C. For both prey, in general, duration of each life stage was shorter, total fecundity was lower and
intrinsic rate of population increase (r
m
) was higher with increasing temperatures. The slower rate of development of P. fragariae on T. evansi resulted in a slightly higher thermal requirement (103.9 degree-days) on that prey than on T. urticae (97.1 degree-days). The values of net reproduction rate (R
0), intrinsic rate of increase (r

) and finite rate of increase (λ) were significantly higher on T. urticae, indicating faster population increase of the predator on this prey species. The highest value of r

The effect of fungal infection on the reproductive potential of two-spotted spider mite, Tetranychus urticae, was evaluated as part of the full biocontrol potential of three entomopathogenic fungi by modeling of fecundity probability.
Female mites (≤2-day-old) on leaves were exposed to the sprays of Beauveria bassiana, Paecilomyces fumosoroseus and Metarhizium anisopliae at the concentrations of 1.13 × 103, 1.55 × 103 and 0.95 × 103 deposited conidia mm−2 and then individually reared at 25°C and 12:12 L:D for oviposition. Mite mortalities 10 days after spraying were 73.1, 75.4
and 67.9% in the fungal treatments versus 15.5% in control. On average, females infected by the three fungal species survived
5.8, 6.2 and 6.3 days, and laid 3.1, 4.0 and 4.0 eggs per capita, respectively. These were 3–4 fold lower than the control
fecundity at 12.3. The cumulative probabilities [P(m ≤ N)] for the counts of infected and non-infected (control) females laying m eggs per capita (m ≤ N) during 10 days fit very well the equation P(m ≤ N) = 1/[1 + exp(a + bm)] (r
2 ≥ 0.98), yielding a solution to the probability for the female mites to achieve a specific fecundity {P(m ≤ N)−P[m ≤ (N − 1)]}. Consequently, the infected mites had 71–78% chance to lay ≤5 eggs per capita but only 5–8% to deposit >10 eggs despite
some variation among the tested fungi. In contrast, the chances for the non-infected mites to achieve the low and high fecundities
were 23 and 55%. The fitted probabilities provide a full coverage of the fecundity potential of infected versus non-infected

In this study the reproductive capacity of a laboratory-selected spirodiclofen resistant strain was investigated after treatment
with spirodiclofen. Firstly, females were exposed to different concentrations of spirodiclofen (200 and 1,000 mg/l) during
6, 12 or 24 h. In contrast to the susceptible parental strain, the fecundity and fertility of resistant mites was not affected
by treatment with these concentrations after any time of exposure tested. Secondly, pre-treatment of the resistant females
with the synergists PBO or DEF could increase the inhibitory effect of spirodiclofen on reproduction, demonstrating the possible
involvement of monooxygenases and esterases in metabolic detoxification of the acaricide. Because spirodiclofen interferes
with lipid biosynthesis, total lipid content was measured in female adults. There were no significant differences between
treated and non-treated female adults, both in the susceptible and resistant strain. However, the total lipid content in the
resistant females was significantly higher than in susceptible females. Our data shows that the detection of spirodiclofen
resistance should not be limited to mortality bioassays with eggs or larvae, but should be combined with inhibitory studies

Laboratory bioassays were conducted to evaluate the effects of spirodiclofen on life history and life-table parameters of
two-spotted spider mite (Tetranychus urticae Koch) females treated at pre-ovipositional period with a series of acaricide concentrations starting with the concentration
discriminative for eggs and immatures i.e. the lowest concentration that causes 100% mortality of those stages. After a 24 h
exposure, the proportion of females that survived treatments was 0.71 (6 mg/l), 0.51 (12 mg/l), 0.41 (24 mg/l), 0.30 (48 mg/l)
and 0.25 (96 mg/l). At the end of the trial, the survival rate of females treated with the lowest concentration was significantly
lower than the survival rate of untreated females but it remained above that of females treated with higher concentrations.
Total fecundity/fertility significantly decreased as concentrations of spirodiclofen increased. Viable eggs were laid by females
treated with 6, 12 and 24 mg/l, and total fertility was reduced by 42, 84 and 97%, respectively. Compared with control, the
gross fecundity/fertility of the treated females was significantly reduced throughout the trial, except in females treated
with 6 mg/l. All concentrations caused a significant reduction in the net fecundity/fertility throughout the trial. The females
treated with 12 mg/l had significantly reduced net reproductive rate (R
0 = 6.45), compared to females treated with 6 mg/l (R
0 = 23.35) and to untreated females (R
0 = 28.92); there was no significant difference between the last two treatments. The intrinsic rate of increase (r

) and finite rate of increase (λ) were significantly reduced in treated females (r

Twenty-three isolates of Metarhizium anisopliae (Metschnikoff) Sokorin and three isolates of Beauveria bassiana (Balsamo) Vuillemin (Ascomycota: Hypocreales: Clavicipitaceae) were assessed for their virulence against the two-spotted
spider mite, Tetranychus urticae Koch (Acari: Tetranychidae). Based on the screening results, nine isolates of M. anisopliae and two isolates of B. bassiana were tested for their virulence against young adult (1- to 2-day-old) female T. urticae at constant temperatures of 20, 25, 30 and 35°C. At all temperatures tested, all the fungal isolates were pathogenic to T. urticae but mortality varied with isolates and temperatures. Fungal isolates were more virulent at 25, 30 and 35°C than at 20°C.
The lethal time to 50% mortality (LT50) and lethal time to 90% mortality (LT90) values decreased with increased temperature. There were no significant differences in virulence between fungal isolates

The purpose of this study was to evaluate the performance of Neoseiulus californicus (McGregor) for the control of Tetranychus urticae Koch in commercial strawberry fields, under greenhouse conditions, in association or not with the use of acaricides. The N. californicus strain used in this study was tolerant or resistant to several pesticides. Three experiments were carried out in the State of São Paulo, Brazil. For the first experiment, the initial infestation of T. urticae was 87.1 active stages per leaflet. Two applications of propargite were made on the first and the 14th day of the experiment. Approximately 2 h after each propargite application, N. californicus was released at a rate of 3.0 and 1.9 adult mites per plant, respectively, for each application. The population of T. urticae decreased from 87.1 to 2.8 mites per leaflet in the first three weeks. After this period, the population of T. urticae was maintained at low levels (≤1.5 mites/leaflet) until the end of the experiment (10th week). Propargite and dimethoate sprayed on the strawberry field did not affect significantly the population of this predaceous mite. For the second experiment, the infestation of T. urticae was 29.1 mites per leaflet, when the acaricide chorfenapyr was applied on the strawberry field. The release of N. californicus (2 mites per plant) was made 2 weeks after spraying the acaricide. The population of T. urticae was maintained at low levels (≤2.8 mites/leaflet) for 8 weeks (evaluation period). The T. urticae infestations in plots with N. californicus were significantly lower than in non-release plots, for the experiments 1 and 2. In the third experiment, the initial infestation of T. urticae was 40.5 mites per leaflet (55.5 active stages/leaflet on release plants; 25.5 active stages/leaflet on non-release plants). Three releases of N. californicus (average rate of 3.0 adult mites/plant), without any acaricide application, were not sufficient to reduce significantly the T. urticae population in release plots (release plants + non-release plants) in 6 weeks from the first release, however, the spider mite population decreased from 55.5 to 7.8 active stages per leaflet on release plants, during this period. Interplant dispersal of N. californicus was low in this strawberry field with high infestation of T. urticae. The studies indicate the viability of the use of this strain of N. californicus for the control of T. urticae in strawberry fields under greenhouse conditions, especially in association with selective acaricides. 相似文献

Sublethal effects of the mitochondrial electron transport inhibitor (METI) tebufenpyrad on Tetranychus urticae Koch females surviving treatment as eggs or immatures (≥90% mortality) were investigated in life-table assay. The developmental time of females that had survived treatment as eggs (2 mg/l) or larvae (2.5 mg/l) was 1 day longer, while that of protonymphs (2.5 mg/l) or deutonymphs (4 mg/l) was 2 days longer, in addition to reduced longevity and fertility, compared to control. The treatment significantly reduced the intrinsic rate of increase in female survivors: corresponding values were 0.258, 0.278, 0.207 and 0.209, respectively (0.307 in untreated females). The offspring age distribution was significantly affected by the treatment. Sublethal effect of tebufenpyrad and its impact on T. urticae management are discussed. 相似文献

Sex ratio distorting endosymbionts induce reproductive anomalies in their arthropod hosts. They have recently been paid much
attention as firstly texts of evolution of host-symbiont relationships and secondly potential biological control agents to
control arthropod pests. Among such organisms, Wolbachia and Cardinium bacteria are well characterized. This study aims at probing such bacteria in the Osmia community to evaluate their potential utilization to control arthropod pests. Among 17 PCR tested species, Osmia cornifrons and a parasitic fly are infected with Wolbachia and a mite species is infected with Cardinium. Phylogenetic tree analyses suggest that horizontal transfer of the bacteria occurred between phylogenetically distant hosts. 相似文献

We sampled mites in three apple orchards in Nova Scotia, Canada, that had been inoculated with pyrethroid-resistant Typhlodromus pyri and had a history of Tetranychus urticae outbreaks. The objective of this study was to monitor populations of T. urticae and phytoseiid predators on the ground and in trees and to track dispersal between the two habitats. Pesticides were the
chief cause of differences in mite dynamics between orchards. In two orchards, application of favourably selective acaricides
(abamectin, clofentezine) in 2002, coupled with predation by T. pyri in trees and Neoseiulus fallacis in ground cover, decreased high T. urticae counts and suppressed Panonychus ulmi. By 2003 phytoseiids kept the tetranychids at low levels. In a third orchard, application of pyrethroids (cypermethrin, lambda-cyhalothrin),
plus an unfavourably selective acaricide (pyridaben) in 2003, suppressed phytoseiids, allowing exponential increases of T. urticae in the ground cover and in tree canopies. By 2004 however, increasing numbers of T. pyri and application of clofentezine strongly reduced densities of T. urticae in tree canopies despite high numbers crawling up from the ground cover. Another influence on T. urticae dynamics was the distribution of the phytoseiids, T. pyri and N. fallacis. When harsh pesticides were avoided, T. pyri were numerous in tree canopies. Conversely, only a few N. fallacis were found there, even when they were present in the ground cover and on tree trunks. Low numbers were sometimes due to pyrethroid
applications or to scarcity of prey. Another factor was likely the abundance of T. pyri, which not only competes with N. fallacis, but also feeds on its larvae and nymphs. The scarcity of a specialist predator of spider mites in trees means that control
of T. urticae largely depends on T. pyri, a generalist predator that is not particularly effective in regulating T. urticae.

The cytoplasmically inherited bacterium Wolbachia is widespread in arthropod species and has been repeatedly detected in the predaceous mite Phytoseiulus persimilis. Our original goal was to assess the prevalence of Wolbachia infection in P. persimilis and the potential fitness consequences for this host. To accomplish that goal, seven P. persimilis strains were obtained from Europe, Africa and the USA and reared on the phytophagous mite Tetranychus urticae as prey. After preliminary results showed that the T. urticae used was infected with Wolbachia, the minimum starvation time of the predators to prevent false positive results from undigested prey was determined. We tested DNA samples by PCR (polymerase chain reaction) after starving the predators or feeding them Wolbachia-free T. urticae for various periods. Those experiments showed that Wolbachia could not be detected after 16 h at 25 °C and 48 h at 20 °C. To verify the results of the PCR analyses, we furthermore conducted crossing experiments with antibiotic-treated and untreated individuals. No indications of Wolbachia effects were recorded. Additionally, we screened live eggs of four of the seven strains reared in our laboratory and alcohol samples of 10 other P. persimilis strains for the occurrence of Wolbachia by PCR, none of which tested positive. Synthesis of our study and previous reports suggests that infection of P. persimilis with Wolbachia is extremely rare and of minor importance. We discuss the significance of our findings for future studies on the presence of Wolbachia in predaceous arthropods. 相似文献

We experimentally tested the indirect and direct effects of Amblyseius womersleyi on Tetranychus kanzawai. The presence of A. womersleyi indirectly reduced egg production of T. kanzawai by 25.9%, although this effect had less impact than direct egg predation. The mechanism of this indirect effect could be explained by behavioral changes in T. kanzawai females; in the presence of A. womersleyi, T. kanzawai females allocated more time to seeking refuge on webs at the expense of feeding on leaves. 相似文献

Artificial lighting is a merit of a ‘plant factory’, which might be utilized to suppress an increase in pest population. We
investigated the effects of extending the light phase on diapause induction in the two-spotted spider mite (TSSM), Tetranychus urticae. TSSM were reared at 18°C under light phases ranging from 2 to 64 h combined with a constant dark phase of 16 h in aluminum
bottles, with white light emitting diodes attached inside to minimize fluctuations in air temperature between the light and
dark phases. Diapause was induced in adult TSSM females when the light phase was 24 h or shorter, and diapause induction was
inhibited when the light phase extended over 32 h. The development of deutonymphs was delayed under a diapause-inducing photoperiod.
Diapause inducing photoperiods may suppress an increase in the TSSM population, by slowing down development and reproduction. 相似文献

Electron microscopy and morphometric analysis have shown that the symbiotic bacteria Wolbachia occur the testis cells D. simulans during spermatogenesis and are absent in mature spermatids. Bacteria did not affect the structural organization of testis cells, which have a typical morphology during morphogenesis. Bacteria were distributed along the meiotic spindle microtubules near the mitochondria. They increased in number in spermatids at the stage of elongation. Endosymbionts aggregated at the spermatid distal end and contained many vacuoles but were absent at the spermatid proximal end near the nuclei. It was shown for the first time that the diameter of spermatids in a strongly infected line was two of three times that in a noninfected line. Seventeen isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and two isolates of Beauveria bassiana (Balsamo) Vuillemin were evaluated for their pathogenicity against the tobacco spider mite, Tetranychus evansi Baker & Pritchard. In the laboratory all the fungal isolates were pathogenic to the adult female mites, causing mortality between 22.1 and 82.6%. Isolates causing more than 70% mortality were subjected to dose–response mortality bioassays. The lethal concentration causing 50% mortality (LC50) values ranged between 0.7×107 and 2.5×107 conidia ml−1. The lethal time to 50% mortality (LT50) values of the most active isolates of B. bassiana and M. anisopliae strains varied between 4.6 and 5.8 days. Potted tomato plants were artificially infested with T. evansi and treated with B. bassiana isolate GPK and M. anisopliae isolate ICIPE78. Both fungal isolates reduced the population density of mites as compared to untreated controls. However, conidia formulated in oil outperformed the ones formulated in water. Somatic hybrids were produced by protoplast fusion between Arabidopsis thaliana ecotype Columbia and a male-sterile radish line MS-Gensuke (Raphanus sativus) with the Ogura cytoplasm. Forty-one shoots were differentiated from the regenerated calli and established as shoot cultures in vitro. About 20 of these shoots were judged to be hybrids based on growth characteristics and morphology. Molecular analyses of 11 shoots were performed, confirming the hybrid features. Of these 11 shoots, eight were established as rooted plants in the greenhouse. Polymerase chain reaction and randomly amplified polymorphic DNA analyses of the nuclear genomes of all analyzed shoots and plants confirmed that they contained hybrid DNA patterns. Their chromosome numbers also supported the hybrid nature of the plants. Investigations of the organelles in the hybrids revealed that the chloroplast (cp) genome was exclusively represented by radish cpDNA, while the mitochondrial DNA configuration showed a combination of both parental genomes as well as fragments unique to the hybrids. Occasional pesticide application in integrated pest management to at least part of a crop requires that any biological control
agents must re-invade previously sprayed areas in order that resurgent pests can be constrained. The ability of the phytoseiid
predatory mite Phytoseiulus persimilis to feed on adult two-spotted spider mite (TSSM) Tetranychus urticae on excised leaf discs in both control conditions and in a treatment with a sub lethal residue of agricultural mineral oil
(AMO) was assessed. The predator exhibited a Type II functional response with the asymptote significantly higher in the AMO
conditions due to the fact that the prey grew slower and reached a smaller size in this treatment. In terms of prey volume
eaten, the satiation level of the predator was unchanged by the AMO deposits. The numbers of eggs produced by adult P. persimilis females at densities of 4, 8 and 16 TSSM adult females/disc in the control were significantly higher than those in the AMO
treatment, but were similar for the higher density levels, 32 and 64 prey per disc. Thus the functional response in terms
of volume of prey eaten explained the numerical response in terms of predator eggs produced. The presence of AMO deposits
when the prey were at high density had no effect on predator efficiency (volume eaten) but resulted in a lower intake than

Predicting the potential geographical distribution of a species is particularly important for pests with strong invasive abilities.
Tetranychus evansi Baker & Pritchard, possibly native to South America, is a spider mite pest of solanaceous crops. This mite is considered
an invasive species in Africa and Europe. A CLIMEX model was developed to predict its global distribution. The model results
fitted the known records of T. evansi except for some records in dry locations. Dryness as well as excess moisture stresses play important roles in limiting the
spread of the mite in the tropics. In North America and Eurasia its potential distribution appears to be essentially limited
by cold stress. Detailed potential distribution maps are provided for T. evansi in the Mediterranean Basin and in Japan. These two regions correspond to climatic borders for the species. Mite establishment
in these areas can be explained by their relatively mild winters. The Mediterranean region is also the main area where tomato
is grown in open fields in Europe and where the pest represents a threat. According to the model, the whole Mediterranean
region has the potential to be extensively colonized by the mite. Wide expansion of the mite to new areas in Africa is also
predicted. Agricultural issues highlighted by the modelled distribution of the pest are discussed.

<Emphasis Type="Italic">Wolbachia</Emphasis> Are Present in Southern African Scorpions and Cluster with Supergroup F 总被引:1,自引:0,他引:1
The presence and distribution of the intracellular bacteria Wolbachia in the arthropod subphylum Chelicerata (including class Arachnida) has not been extensively explored. Here we report the discovery of Wolbachia in scorpions. Five strains found in host species of the genus Opistophthalmus (Southern African burrowing scorpions) have been characterized by Multilocus Sequence Typing and by Wolbachia Surface Protein. Phylogenetic analyses indicate clustering in the supergroup F and a high genetic relatedness among all scorpion strains as a result of a potential transmission within the host genus. The F-group is an uncommon lineage compared to the A and B supergroups, although it is present in a broad range of hosts (including insects, filarial nematodes, and now arachnids) and across a large geographical area (e.g., North America, Africa, Europe, and Australia). It also shows no evidence of recombination and has a significantly higher genetic diversity than supergroup A and B. Overall, this pattern suggests an older radiation of F-strains with respect to A and B-strains, followed by limited horizontal transmission across host genera and reduced genetic flux among strains. A more extensive sampling of supergroup F-strains is required to confirm this scenario. 相似文献

Wolbachia harbored by most filarial parasites, is critical to both embryogenesis and microfilarial development, and may lead to inflammation and pathogenesis in infected hosts. Based on alignment of the sequences from the wsp, ftsZ, and 16S rRNA genes, Wolbachia was demonstrated to exist in Angiostrongylus cantonensis, a non-filaroid nematode. Although the wsp gene may not be the best candidate for evolutionary analysis of Wolbachia, this gene has been sequenced from a broader coverage of the host species, making it feasible to be used for phylogenetic analysis in this study. The results from both Neighbor-joining and Maximum parsimony methods showed that this novel Wolbachia does not belong to any of the known groups (C or D) of nematode-derived Wolbachia. In addition, the wsp gene sequence of this newly identified endosymbiont revealed a high degree of identity (98%) with that from Diaea circumlita c2, tentatively classified into the putative group G. This suggests that Wolbachia from A. cantonensis could represent a deeply branched lineage in Wolbachia evolution or the occurrence of horizontal transfer between infected hosts.
