Temperature dependencies were established for the egg-to-mummy and mummy-to-adult phases, for mummy mortality, and for parasitism of Aphidius ervi Haliday, Aphidius rhopalosiphi De Stefani-Perez, and Praon volucre (Haliday) (Hymenoptera, Aphidiidae), three parasitoids of Sitobion avenae (Fabricius) (Homoptera, Aphididae), at 8°C, 12°C, 16°C, 20°C, and 25°C on winter wheat (cv. Haven). A physiological model described temperature-dependent development over the full temperature range, whereas a linear model was fitted for data above 8°C and used to estimate the lower temperature thresholds and day-degrees (° D) required for development. The thresholds for A. ervi were 2.2°C for egg-mummy development and 6.6°C for mummy-adult development, those for A. rhopalosiphi were 4.5°C and 7.2°C, and those for P. volucre were 3.8°C and 5.5°C. The time to develop into mummies and adults differed significantly between the three species: A. ervi development into mummies required an average of 159 ° D, while development into adults took an average of 73 ° D. The corresponding average times required for A. rhopalosiphi and P. volucre to develop mummies were 124° D and 126° D, while their development into adults required an average of 70° D and 150° D, respectively. Mummy mortality was 25–35% at 8°C and less at the higher temperatures tested, but began to increase again at 25°C, showing a quadratic relationship between mortality and temperature. Parasitization was very low or, in the case of P. volucre, absent up to 12°C and thereafter increased with increasing temperature. The relationship between parasitization, recorded as percent aphids mummified, and temperature was linear at the temperatures tested and depended on species. A. ervisuperparasitized 11.1% aphids at 20°C and 16.6% aphids at 25°C, whereas superparasitism was low in A. rhopalosiphi and absent in P. volucre. From 16°C to 25°C the P. volucre sex ratio increased. For A. ervi and A. rhopalosiphi there was no trend with temperature, but at 20°C and 25°C it was close to even. Field data for 1996 and 1997 allowed for a comparison of actual and expected emergence of overwintering mummies. In both years, parasitoids were predicted to have emerged from overwintering mummies well in advance of the onset of aphid infestation, and more than a month earlier than the first parasitized aphids were found in winter wheat. Observations from trap plants in other crops supported the predictions of the models. Other factors that can affect biological control by cereal aphid parasitoids are discussed.  相似文献

Hormone releasing properties from an emulsion prepared with lipophilized gelatin (LG emulsion) were investigated on salmon gonadotropin (sGTH) as a peptide hormone and 17-hydroxyprogesterone (17P) as a steroid at 10, 20 and 30°C by monitoring plasma profiles of these hormones after administration in the Japanese eel. Release of these hormones from the LG emulsion were slow and not largely influenced by water temperature, whereas release from saline solution (sGTH) or cottonseed oil (17P) were rapid and increased with the elevation of temperature. © Rapid Science Ltd. 1998  相似文献

Laboratory studies were performed to assess the importance of temperature on sporulation and infection by the aphid-pathogenic fungus Pandora neoaphidis (Remaudière and Hennebert) Humber. Numbers of primary conidia discharged from mycelium formulated as alginate granules and unformulated mycelial mats were assessed, as well as infection of the potato aphid, Macrosiphum euphorbiae (Thomas) (Homoptera, Hemiptera, Aphididae), using culture plugs as inoculum sources. Sporulation from experiments at constant temperatures indicated the optimum temperature range was 10–20°C for both mycelial preparations and there was no or very little sporulation at 30°C. Infection of aphids kept at 15°C was 34–50%, while infection at 25°C was 11–44%. At 20°C, 77–79% of aphids were infected. Under fluctuating temperature cycles, conidia numbers did not differ when mycelial preparations were maintained at 18–25°C compared with 18–20°C, but fewer conidia were recorded when preparations were exposed continuously to 18–30°C. Infections of inoculated aphids kept for varying numbers of days at 18–25°C varied between 24–47%, but only 3–32% of aphids were infected when exposed to a cycle of 18–30°C for various times. Unformulated mycelial mats of P. neoaphidis appear to be superior to forumlated alginate granules for use in experimental greenhouse and field trials, since temperature stability is similar for both materials but mycelial mats are much easier to produce.  相似文献

Summary Eel atrial natriuretic peptide inhibited the serosa-negative transepithelial potential difference and short-circuit current, accompanied by a decrease in NaCl and water absorption across the seawater eel intestine. Similar effects were obtained after treatment with N-terminally truncated eel atrial natriuretic peptide (5–27), indicating that N-terminal amino acids are not essential for the action of eel atrial natriuretic peptide. Although mammalian atrial natriuretic peptides also inhibited the short-circuit current, a 100-fold higher concentration was reuired to obtain the same effect as with eel atrial natriuretic peptide, indicating that eel atrial natriuretic peptide is 100 times as potent in eel intestine as the mammalian atrial natriuretic peptides. Similarly, in mammalian atrial natriuretic peptide, the four N-terminal amino acids had no significant effects. However, when the C-terminal tyrosine was removed, the potency of rat atrial natriuretic peptide was lowered. Compared with the effects of acetylcholine, serotonin and histamine, eel atrial natriuretic peptide was the most potent inhibitor, with 100% inhibition at 10-7 M; 50% inhibition was obtained at 10-2 M in acetylcholine, and 30% inhibition in serotonin (10-5 M) and histamine (10-3 M). These inhibitory effects of eel atrial natriuretic peptide were not diminished even in the presence of tetradoxin, and were mimicked by 8-bromoguanosine 3,5-cyclic monophosphate. Bragg and Cobb soybean [Glycine max (L.) Merr.] seeds were germinated in sand at temperatures ranging from 25 to 40°C. Emergence decreased with increasing temperature above 37°C, with virtually no emergence at 40°C. Emergence of 12 other cultivars at 38°C ranged from 25 to 95%. Foster and Coker 338 were more sensitive to high temperature than the other cultivars.  相似文献

On the basis of the filament rotation model that was elaborated for interpretations in cell motility, the lateral hydrodynamic effects of rotating filaments have been investigated by large-scale model experiments. Helices were rotated by small electric motors in a medium of high viscosity (honey or polyethyleneglycol). The observed effects, hitherto not investigated in detail by hydrodynamics, show some features that were attributed to the indefinable formative power or vital force of the past. The main effects generated by the rotating filaments are (1) flows and flow patterns with impact zones where flows collide, (2) regions of excessive pressure and negative pressure (corner effect) along a wall, (3) grooves and smoothly shaped ridges on a free fluid surface, and (4) rolling motions of freely hanging filaments. All effects and flow patterns depend on the appropriate distribution of rotating and counterrotating filaments. Each change of the rotational direction means a dramatic alteration. The application of the observed effects explains largely the function of the microtubule/microfilament hoops or helices during the cytokinesis of a plant cell. Interpretations or simulations are described for events as the formation of secondary wall thickenings, the orientation of their microfibrils, the motion of the preprophase band microtubules, the formation of the phragmosome, the migration, stationary position and shape of the preprophase nucleus, the girdle-, septum- and H-piece formation of cell walls in algae and some events of morphogenesis inMicrasterias. Further interpretations are related to the lateral flows and to invaginations of free cell membranes, to lateral filament motions, to the right-left problem, to the selfintertwining of filaments, to the rotation of a cell body by its flagellum, to the repulsion of chromatids during meiosis and to the tetragonal and hexagonal arrangement of filaments.Dedicated to Prof. DrLothar Geitler on the occasion of the 90th anniversary of his birthday.  相似文献

Development, survival and reproduction of Euseius finlandicus Oudemans were studied at seven constant temperatures (15, 20, 25, 27, 30, 32 and 34°C) in the laboratory. Within the temperature range tested, developmental period from egg to adult varied from 148 to 360.5h and 133.7 to 336.5h for females and males, respectively. The lower thermal threshold for immature development for females and males was 8.9 and 6.4°C, respectively. Survival during immature development exceeded 90% at all the temperatures from 15 to 32°C, but at 34°C an abrupt decline was recorded. Female longevity decreased gradually from 82.7d at 15°C to 12.2 d at 34°C. The mean generation time ranged from 44.3d at 15°C to 15.9d at 32°C. The highest r m value (0.2817) was obtained at 30°C and the lowest at 15°C (0.0976). Temperatures above 30°C had an adverse effect on population increase.  相似文献

Changes in carbon isotope composition(13C) and leaf morphology associated withvegetative phase change were monitored in Metrosiderosexcelsa Sol. ex Gaertn. (family Myrtaceae). Plants of threeontogenetic states were used: juvenile seedlings, micropropagated plants in arejuvenated state, and reproductively mature plants bearing leaves with adultcharacteristics. The effects of temperature regime (32/24 °C,24/16 °C, and 16/8 °C day/night) and plantarchitecture (branched and single-stemmed plants) were studied in two separateexperiments. Although both juvenile and rejuvenated plants exhibited juvenileleaf morphology at the start of the experiments, there was no differencebetweenleaf 13C in these plants and that in adultplantsat this time (mean ca. –27%). Vegetative phase change occurred injuvenileand rejuvenated plants grown at 24/16 °C, and there was acorresponding increase in leaf 13C (from ca.–27% to –23%) in these two groups of plants. Leaf13C in adult plants remained relatively constant(ca. –26%) at 24/16 °C. There was little change in leaf13C in all plant states maintained at 32/24°C or 16/8 °C, and vegetative phase change didnot occur in juvenile and rejuvenated plants grown under these two temperatureregimes. Rejuvenated plants grown in a greenhouse also exhibited a progressivedevelopment of adult leaf morphology, accompanied by an increase in leaf13C, an effect that was more pronounced insingle-stemmed (from –26.4% to ca. –24%) than in branched plants. Itis suggested that increasing 13C in juvenile andrejuvenated plants undergoing phase change is a result of reduced sink strengthin single-stemmed plants, and to a lesser extent within each branch of branchedplants, causing reduced stomatal conductance and photosynthesis.  相似文献

In the present study we investigated the response to iron (Fe) deficiency in two cultivars of Festuca rubra L. (Rubina and Barnica) used in correction of chlorosis of fruit trees cultivated on calcareous soils. We found that a Fe-chelating compound, identified as 2-deoxymugineic acid (DMA), was secreted from the roots in response to Fe-deficiency in both cultivars. The amount of DMA secreted into solution increased with the development of Fe-deficiency. The secretion showed a distinct diurnal rhythm characterized by a secretion peak at between 2 and 5 hours after sunrise at 20°C. However, this secretion peak was delayed by 3 hour at low temperature (<10°C) and occurred 3 h earlier at high temperature (30°C). When water used for the collection of root exudates was pre-warmed (25°C) or pre-cooled (10°C), this led to an earlier or a delayed secretion compared to control (15°C) under the same air temperature, respectively. Short-term shading treatment did not affect the secretion pattern of DMA. These results demonstrate that the secretion time of DMA from the roots is, at least partly controlled by the temperature in the root environment. Overall, these findings suggest that the ability of Festuca rubra to prevent Fe chlorosis symptoms (`re-greening effect') of associated fruit trees is partially related to the secretion of DMA which increase Fe availability in calcareous soils.  相似文献

Delicious apple (Malus domestica Borkh.) and several of its strains, which have been difficult to root in vitro, were successfully propagated with rooting percentages up to 100%. The combination of treatments used to achieve this result included placing the shoots on rooting medium in the dark at 30°C for the first week of the rooting stage, then moving them to a regime of 16 hr light-8 hr dark at 25°C. The rooting medium contained half strength Murashige and Skoog salts plus 1.2 M thiamine HCl, 0.56 mM myo-inositol, 1 mM phloroglucinol (PG), 1.4 M indolebutyric acid (IBA), 1.3 M gibberellic acid (GA3), 87.6 mM sucrose, and 7 g l–1 Difco Bacto agar. Dark treatment applied during the proliferation stage (etiolation) was less effective than one applied at the beginning of the rooting stage. The optimum length of dark treatment during rooting was 4 to 7 days. Increasing the temperature from 25°C to 30°C improved rooting of Delicious, Royal Red Delicious, and Vermont Spur Delicious in the absence of PG but generally had less effect in the presence of PG. Further increase in temperature to 35°C stimulated rooting of Royal Red Delicious but reduced rooting of Vermont Spur Delicious. Transfer of the cuttings to auxin-free medium after 1 week had no effect on percentage rooting and increased the number of roots per cutting for only 1 of 4 cultivars tested and then only in the presence of PG. In general PG stimulated rooting of Delicious and its strains, but had no effect on Golden Delicious.  相似文献

Cryopreservation of callus tissue of Artimisia annua L. was optimized. Two lines of calli were precultured on MS medium with 5% (v/v) dimethyl sulfoxide, and protected by a cryoprotectant containing 15% (v/v) ethylene glycol, 15% (v/v) dimethyl sulfoxide, 30% (v/v) glycerol and 13.6% (w/v) sucrose. The highest survival rate of callus A201 reached 87% after it was pretreated at 25°C, cryopreserved by liquid nitrogen, recovered in water bath at 25°C and reloaded at 25°C with 34% (w/v) sucrose solution, and that of callus A202 reached 78% after it was treated as callus A201, except pretreated at 35°C, recovered at 35°C and reloaded with 47.8% (w/v) sucrose solution.  相似文献

A new cDNA (Osfruct2) encoding an acid –d-fructofuranosidase from rice has been cloned, sequenced and expressed in Pichia pastoris. The full-length cDNA is 2453 base pairs long and encodes a pre-pro-protein of 682 amino acids. The cDNA fragment coding for mature enzyme was sub-cloned into vector pPICZA for extracellular expression in the methylotrophic yeast. The recombinant product was purified by Ni2+-nitrilotriacetic acid agarose column and biochemically characterized. The enzyme could hydrolyze sucrose and raffinose. Molecular mass is 66 kDa. The activity optimum was at pH 4.8 and 40 °C.  相似文献

The response of effective quantum yield of photosystem 2 (F/Fm) to temperature was investigated under field conditions (1 950 m a.s.l.) in three alpine plant species with contrasting leaf temperature climates. The in situ temperature response did not follow an optimum curve but under saturating irradiances [PPFD >800 µìmol(photon) m–2s–1] highest F/Fm occurred at leaf temperatures below 10°C. This was comparable to the temperature response of antarctic vascular plants. Leaf temperatures between 0 and 15°C were the most frequently (41 to 56%) experienced by the investigated species. At these temperatures, F/Fm was highest in all species (data from all irradiation classes included) but the species differed in the temperature at which F/Fm dropped below 50% (Soldanella pusilla >20°C, Loiseleuria procumbens >25°C, and Saxifraga paniculata >40°C). The in situ response of F/Fm showed significantly higher F/Fm values at saturating PPFD for the species growing in full sunlight (S. paniculata and L. procumbens) than for S. pusilla growing under more moderate PPFD. The effect of increasing PPFD on F/Fm, for a given leaf temperature, was most pronounced in S. pusilla. Despite the broad diurnal leaf temperature amplitude of alpine environments, only in S. paniculata did saturating PPFD occur over a broad range of leaf temperatures (43 K). In the other two species it was half of that (around 20 K). This indicates that the setting of environmental scenarios (leaf temperature×PPFD) in laboratory experiments often likely exceeds the actual environmental demand in the field.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

Temperature dependence (25–50 °C) of chlorophyll (Chl) fluorescence induction, far-red radiation (FR)-induced relaxation of the post-irradiation transient increase in apparent F0, and the trans-thylakoid proton gradients (pH) was examined in maize leaves. Temperatures above 30 °C caused an elevation of F0 level and an enhancement of F0 quenching during actinic irradiation. Millisecond delayed light emission (ms-DLE), which reflects the magnitude of pH, decreased strikingly above 35 °C, and almost disappeared at 50 °C. It indicates that the heat-enhanced quenching of F0 under actinic irradiation could not be attributed mainly to the mechanism of pH-dependent quenching. The relaxation of the post-irradiation transient increase in apparent F0 upon FR irradiation could be decomposed into two exponential components (1 = 0.7–1.8 s, 2 = 2.0–9.9 s). Decay times of both components increased with temperature increasing from 25 to 40–45 °C. The bi-phasic kinetics of FR-induced relaxation of the post-irradiation transient increase in apparent F0 and its temperature dependence may be related to plastoquinone (PQ) compartmentation in the thylakoid membranes and its re-organisation at elevated temperature.  相似文献

A 1.2 kb DNA fragment coding for the pro-peptide and mature keratinase from Bacillus licheniformis PWD-1 (kerA) was cloned into vectors pPICZA and pGAPZA for extracellular expression in the methylotrophic yeast, Pichia pastoris. Recombinant keratinase was secreted by the pPICZA-kerA transformants 24 h after methanol induction of shake-flask cultures, and reached a final yield of 124 mg l–1 (285 U ml–1) 144 h after the induction. The recombinant keratinase was glycosylated ( 39 kDa), and was optimal between pH 8.5–9.5 and between 55°C –60°C using azokeratin as substrate. The enzyme degraded bovine serum albumin, collagen, and soy protein concentrate. In conclusion, P. pastoris can be used as an efficient host to express keratinase for nutritional and environmental applications.  相似文献

Summary In barley (Hordeum vulgare L.) seedlings, a temperature step-down from 24 °C to 6°C (cold shock) determined a reduction in the incorporation of labeled aminoacids and modified the electrophoretic pattern of total proteins. At 6 °C some new proteins appeared and others were intensified (cold shock-induced proteins= CSPs); meantime, few proteins disappeared or were curtailed (cold-repressed proteins=CRPs). The majority of the proteins of the seedlings were labeled at about the same rate both at 6 °C and 24 °C, whereas at 0 °C only the cold shock proteins and a few others were detectable. The cold shock-induced variations of the protein profile differed in roots and in seed remnants which showed only some of the CSPs detected in roots. Total protein synthesis of barley genotypes Onice and Georgie, which have respectively a winter and spring growth habit, were similarly inhibited by a temperature drop. The two genotypes, however, showed some differences in the CSPs and CRPs pattern. Because Onice and Georgie have also a different thermotolerance, the hypothesis can be made that in barley specific CSPs are involved in conferring various degrees of cold resistance.  相似文献

Morphometric variation in three clones of Brachionus plicatilis cultured at 20°C, 25°C and 30°C, and 9, 12 and 24 salinity was analysed. Size appeared to be largely under genetic control and this defined the narrow limits within which variation due to abiotic factors could occur. Temperature had a significant effect on size, but affected the three clones differently. The most general effect of temperature was a reduction in size which levelled off as the temperature rose. Of the measurements taken, only distance between the median spines was affected by salinity. An important inter-relationship between the effects of temperature and salinity was also detected.  相似文献

Effects of the combination of low temperature and silicone treatment on the storage of grape callus (Vitis vinifera L. x V. labrusca L. cv. Kyoho; V. vinifera L. cv. Koshusanjaku) were examined. In Kyoho, the calli were stored at 10°C successfully for up to 360 days. Embryogenic calli of Koshusanjaku stored at 10°C retained the ability of embryogenesis after 360 days of storage. However, the color of both calli became brownish. This was improved by the combination of low temperature and silicone treatment. The calli of Kyoho survived by the storage under the combination of 15°C and silicone. Embryogenic calli stored at 10 and 15°C in combination with silicone survived for 360 days, and regenerated only after transfer onto a regeneration medium. Thus the combination of low temperature and silicone affects the longevity of the grape callus.  相似文献

The critical swimming velocity (Ucrit) of four California stream fishes, hardhead, Mylopharodon conocephalus, hitch, Lavinia exilicauda, Sacramento pikeminnow, Ptychocheilus grandis, and Sacramento sucker, Catostomus occidentalis was measured at 10, 15, and 20°C. Hardhead, Sacramento sucker, and Sacramento pikeminnow swimming performances tended to be lowest at 10°C, higher at 15°C, and then decreased or remained constant at 20°C. Hitch swimming performance was lower at 10°C than at 20°C. There were no significant differences among species at 10 or 15°C, although pikeminnow and hitch were ca. 20% slower than hardhead or sucker. At 20°C hardhead, Sacramento sucker, and Sacramento pikeminnow had remarkably similar Ucrit but hitch were significantly (by 11%) faster. We recommend that water diversion approach velocities should not exceed 0.3ms–1 for hitch (20–30cm total length) and 0.4ms–1 for hardhead, Sacramento pikeminnow, and Sacramento sucker (20–30cm TL).  相似文献
