Ligularia , a highly diversified genus in the eastern Qinghai–Tibet Plateau and adjacent areas, was chosen as a suitable subject in which to study speciation patterns in this 'hot spot' area at the chromosomal level. Chromosome numbers and karyotypes were studied in 23 populations of 14 species, most of which are endemic to this area. The basic number x  = 29 was confirmed for all species. Ligularia virgaurea was found to have diploid and triploid cytotypes, 2 n  = 58 and 87. Other species are only diploid, with 2 n  = 58. The karyotypes of all populations within any species, and all species spanning most sections and covering most of the morphological range in Ligularia , are very similar to each other, belonging to type 2A according to Stebbin's classification. This karyotype was also found in its close allies, e.g. Cremanthodium , Ligulariopsis , Parasenecio , and Sinacalia . Aneuploid reduction of chromosome number from 2 n  = 60 to 58 and karyotypic variation was found in Ligularia and its allies. Such a chromosomal pattern with few polyploids infers that variation of karyotype structure at the diploid level seems to be the predominant feature of chromosomal evolution in this group and sympatric speciation via hybridization and polyploidization has played a minor role in its species diversity.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 144 , 329–342.  相似文献

Chromosomes of four species in three genera of Commelinaceae from China were examined. Spatholirion longifolium had 2 n  = 20. This small genus is considered to be of a possible polyploid origin based on x  = 5, a basic number shared by its closest ally, the monospecific genus Streptolirion , with its only species having 2 n  = 10. The two genera were found to be very different from each other in their karyotypic constitution. The differences in both chromosome number (2 n  = 20 vs. 10) and karyotypic constitution support their separation as two independent genera. The remaining three species, Amischotolype hispida , Porandra ramosa and P. scandens , all had 2 n  = 36. In view of the occasional occurrence of 2 n  = 18 in the African Coleotrype , a genus very close to Amischotolype and Porandra in gross morphology, these three species are all very probably polyploid based on x  = 9. Molecular study of mitochondrial and nuclear genes and cytogenetic analysis were performed to examine possible patterns of speciation in the diverse Lophuromys flavopunctatus species complex of Ethiopia. Phylogenetic analysis of mtDNA data resulted in an unresolved bush of ten deeply diverged haplotype groups corresponding to potential species either well supported by various types of character or 'cryptic'. The cytogenetic analysis showed representatives of five of these mtDNA lineages to share an identical karyotype (2 n  = 70, NFa = 84), that has not been found previously in Ethiopia. One of them, L.  cf.  sikapusi , being a member of the L. flavopunctatus species complex, demonstrates remarkable morphological similarity to representatives of another species complex, L. sikapusi s.l ., which might be considered as a result of convergent evolution in analogous environments. Analysis of RAPD data suggests that at least two mtDNA types might have been subject to interspecific transfer due to hybridization. In the case of two sympatric haplotypes of L. brunneus we may assume that the contemporary pattern of variation between them can be explained by relatively recent hybridization with another distinct species, L. flavopunctatus . Somatic chromosomes of four species of Ceratozamia , C. hildae , C. kuesteriana , C. mexicana and C. norstogii , and Stangeria eriopus , were observed and compared by the fluorescence in situ hybridization method using 5S ribosomal (rDNA) probes. The four Ceratozamia species and S. eriopus showed the same chromosome number of 2 n  = 16, and had similar karyotypes, comprising 12 metacentric (m), two submetacentric (sm) chromosomes and two telocentric (t) chromosomes. The four Ceratozamia species exhibited a proximal 5S rDNA site in the interstitial region of two m chromosomes. The genetic diversity, structure, and relationships of 12 populations of blind subterranean mole rats belonging to four species of the Spalax ehrenbergi superspecies were examined. Diversity levels and divergence of 19 nuclear microsatellite markers were analysed. Results indicate that the 12 populations fall into two major clusters representing the species pairs Spalax galili (2 n  = 52) and S. golani (2 n  = 54) vs. S. carmeli (2 n  = 58) and S. judaei (2 n  = 60). There is very little gene flow between the species pairs, except for one population (Afiq) of S. carmeli (2 n  = 58), which is located near the hybrid zone between S. golani (2 n  = 54) and S. carmeli (2 n  = 58). Higher values of the mean for alleles per locus, gene diversity, and number of unique alleles were observed for the southern species pair, S. carmeli and S. judaei, than for the northern species pair, S. galili and S. golani . Microsatellite diversity increases southward toward the Negev Desert and is positively correlated with aridity stress. Climatic factors of water availability coupled with temperature account for 76.7% of microsatellite variance. Somatic chromosome numbers, conventional karyotype features and idiograms are reported for 27 Limonium species inhabiting the Western Mediterranean basin (Iberian Peninsula and the Balearic Islands). The chromosome numbers of Limonium barceloi (2 n  = 36), L. ejulabilis (2 n  = 24), L. inexpectans (2 n  = 26), L. interjectum (2 n  = 24), and L. scopulorum (2 n  = 25) were determined for the first time. In addition, new aneuploid and/or polyploid cytotypes are reported in L. alcudianum (2 n  = 26), L. bonafei (2 n  = 26), L. camposanum (2 n  = 26) , L. companyonis (2 n  = 26), L. dufourii (2 n  = 26), L. geronense (2 n  = 36), L. marisolii (2n = 54), L. migjornense (2 n  = 50), and L. pseudodictyocladon (2 n  = 16). A group of polyploid species showed karyotypes comprising homologous chromosomes in groups of three ( L. antonii-llorensii, L. ejulabilis, L. interjectum, L. virgatum, and L. wiedmanii ) , four ( L. geronense ), or six ( L. marisolii ), which suggests an autopolyploid origin. Other polyploid species were characterized by the presence of two different chromosome sets ( x  = 8 and x  = 9) in the genome. Ploidal level information is of particular importance in intricate polyploid complexes such as in arctic-alpine Draba . Relative DNA content is reported for the tetra- and hexaploid D. lactea and seven of its low-ploid relatives. Flow cytometry was used to study 200 plants from 93 populations, the screening based on relative fluorescence. Absolute DNA content was determined by Feulgen densitometry for 13 plants from seven species, and reference chromosome numbers were determined in 12 plants (1–3 per species) representing six species. The plants grouped into diploids (2 n  = 16), tetraploids (2 n  = 32), hexaploids (2 n  = 48), and two triploids. Each ploidal level showed a linear increase in relative DNA content, pointing to a relatively recent polyploid origin. The diploid level was confirmed in D. nivalis, D. subcapitata, D. fladnizensis , and D. lonchocarpa. Draba palanderiana , reported previously as di-, tetra- and octoploid, was diploid in all investigated accessions. Hexa- and tetraploids were observed in D. lactea , in approximately the same ratio (8 : 1) as reported previously. There are 20 Bellevalia species in Turkey, half of which are endemic. Chromosome numbers are known for 15 species. A chromosome survey of 145 Bellevalia individuals showed that the karyotype is remarkably stable. All are based on x  = 4. The majority are diploid with 2 n  = 8, but there is also a polyploid series of 2 n  = 16, 24 and 32. Aneuploidy occurs only at the octoploid level. Eleven individuals had metacentric B chromosomes, one had acrocentric Bs and one had telocentric Bs. The karyotypes of six species of Solms-laubachia and one each of Parrya and Desideria (Brassicaceae) from the Himalayan-Hengduan Mountains (Tibet, Sichuan and Yunnan provinces) were investigated. Chromosome counts of S. xerophyta , S. lanata, S. platycarpa and D. baiogoinensis are reported for the first time and found to be diploid with 2 n  = 14. Diploid counts were also made of S. retropilosa , a species previously known to be a tetraploid. The karyotype formulae of D. baiogoinensis , S. eurycarpa , S. pulcherrima , S. retropilosa , S. xerophyta and S. platycarpa are 2 n  = 14 = 6 m + 6sm + 2st, whereas those of S. lanata and P. nudicaulis are 2 n  = 14 = 6 m + 2sm + 6st and 2 n  = 14 = 8 m + 4sm + 2st, respectively. Karyotypes and counts of S. eurycarpa and S. pulcherrima are in agreement with prior studies. The relationships within Solms-laubachia and among the three genera are discussed. The discovery of tetraploidy in the red viscacha rat, Tympanoctomys barrerae (4 n  = 102) has emphasized the evolutionary role of genome duplication in mammals. The tetraploid status of this species is corroborated here by in situ PCR and Southern blot analysis of a single-copy gene. The species meiotic configuration strongly suggests a hybrid derivation. To investigate the origin of T. barrerae further, the recently described Pipanacoctomys aureus was studied. This 92-chromosome species also has a duplicated genome size, redundant gene copy number and diploid-like meiotic pairing, consistent with an event of allotetraploidization. Phylogenetic analysis of mitochondrial sequences indicates sister-group relationships between these two tetraploid rodents. The new karyotypic data and the phylogenetic relationships suggest the participation of the ancestral lineages of Octomys mimax in the genesis of P. aureus . Meiotic chromosome counts, chromosomal behaviour and meiotic configurations of ten taxa of Senecio from Argentina were examined. Most counts are original: S. crepidifolius DC., S. francisci Phil. and S. octolepis Griseb. var. saltensis (Hicken) Cabrera & Zardini have 2 n  = 40, and S. chrysolepis Phil., 2 n  = 80 + 8B. We confirmed previous reports for S. deferens Griseb. (2 n  = 40 + 4B), S. filaginoides DC. var. filaginoides (2 n  = 40), S. hieronymi Griseb. (2 n  = 40 + 7B), S. pampeanus Cabrera (2 n  = 40) and S. rudbeckiifolius Meyen & Walp. (2 n  = 40 + 7B). In S. bracteolatus Hook. & Arn. var. bracteolatus , we found a new number (2 n  = 40) that differs from the one cited previously. In four species, numerical polymorphisms for B-chromosomes were observed. Several of the analysed species presented secondary bivalent association. This phenomenon, together with other evidence, supports x  = 5 as the basic chromosome number. The number of chiasmata and their positions were also surveyed, with the finding that open bivalents were the most frequent meiotic figures and terminal chiasmata the preferential position. These features are related to recombination rate, and the success and persistence of these polyploids. We review and extend data showing concerted evolution of parental 18–5.8–26S nuclear ribosomal DNA (18–26S rDNA) gene families in three natural Nicotiana allotetraploids ( N. tabacum , N. rustica and N. arentsii , each 2 n  = 4 x  = 48) and one synthetic N. tabacum line (Th37, ♀ N. sylvestris (2 n  = 24) × ♂ N. tomentosiformis (2 n  = 24)). The origin of the gene families was analysed by sequence polymorphisms in the intergenic spacer (IGS) region and the number of chromosomal loci by fluorescence in situ hybridization (FISH). FISH revealed that the number and locations of 18–26S rDNA in the natural allopolyploids was the sum of those found in the diploid progenitors. However, the rDNA restriction patterns showed polymorphisms in the IGS that were not additive, suggesting that parental rDNA clusters were partially ( N. tabacum, N. rustica ) or completely ( N. arentsii ) overwritten by hybrid-specific units. Thus the Nicotiana allotetraploids show evidence of concerted evolution, including both intralocus and interlocus gene conversion. A feral N. tabacum collected in Bolivia had a higher proportion of unconverted parental rDNA units than cultivated tobacco varieties, suggesting either that rDNA homogenization is accelerated by inbreeding or multiple origins of tobacco. There is no evidence for the elimination of N. sylvestris- derived rDNA units in the synthetic Th37 tobacco line as occurred in natural tobacco, although several novel rDNA unit variants were found in most but not all the hybrid plants. Cyrtomium (Dryopteridaceae) is an Asiatic genus comprising about 40 species characterized by having anastomosing veins that form areolae with included veinlets. Twelve chromosome counts representing 11 taxa were made in the present study. The chromosome numbers of six species endemic to China are reported for the first time: C. uniseriale , 2 n  = 164; C. guizhouense , 2 n  = 82; C. shingianum , n  = 82, 2 n  =  c .164; C. chingianum , 2 n  = 164; C. urophyllum , 2 n  = 82 and C. aequibasis , 2 n  = 123. The results were used to show that (1) C. uniseriale should be reduced to varietal status under C. balansae ; (2) subseries Balansana is not closely related to any other Cyrtomium species and should be separated from Cyrtomium ; and (3) C. guizhouense and C. lonchitoides are basal groups when subseries Balansana is not considered. These results were also supported by morphology. A high level of differentiation is a distinct characteristic in Cyrtomium . The genus has two reproductive types: sexual and apogamous. Sexual species are distributed mostly in China. Southwestern China is a modern diversity centre of Cyrtomium , with 80.6% of known species occurring there. Many species are endemic, most of which are sexual tetraploids or apogamous triploids. Polyploidy and the sexual system: what can we learn from Mercurialis annua?   总被引:1,自引:0,他引:1  
The evolutionary success of polyploidy most directly requires the ability of polyploid individuals to reproduce and transmit their genes to subsequent generations. As a result, the sexual system (i.e. the mating system and the sex allocation of a species) will necessarily play a key role in determining the fate of a new polyploid lineage. The effects of the sexual system on the evolution of polyploidy are complex and interactive. They include both aspects of the genetic system, the genetic load maintained in a population and the ecological context in which selection takes place. Here, we explore these complexities and review the empirical evidence for several potentially important genetic and ecological interactions between ploidy and the sexual system in plants. We place particular emphasis on work in our laboratory on the European annual plant Mercurialis annua , which offers promising scope for detailed investigations on this topic. Chromosome numbers and morphology in 22 populations belonging to 11 taxa of Aconitum subgenus Lycoctonum (Ranunculaceae) from China were studied. Some taxa were diploid, with 2 n  = 16, but four species and two varieties were found to be tetraploid, with 2 n  = 32. They are concentrated in the Hengduan Mountains region in south-west China, indicating that polyploidy could have played an important role in the speciation of the subgenus in this region, one of the areas of the world with a high concentration of endemic species. Senecio cambrensis (Welsh groundsel) is a new allohexaploid species, which originated in Wales, UK, in the early part of the 20th century following hybridization between the native tetraploid groundsel (Senecio vulgaris) and the introduced diploid Oxford ragwort (Senecio squalidus). A survey of the number of populations and flowering individuals per population of S. cambrensis in Wales was conducted at peak flowering time in June 2002, 2003 and 2004. The results show a dramatic decrease in both population number and population size of the species since the 1980s when the last population census was conducted. A survey of amplified fragment length polymorphism (AFLP) variation showed that this decline has occurred despite the fact that S. cambrensis contains a high level of genetic diversity with each individual screened possessing a unique multilocus phenotype. The level of variance within the species was similar to that found in one parent (S. vulgaris) and slightly greater than that among samples of the other parent (S. squalidus). Only a small proportion (5%) of AFLP diversity was partitioned among populations indicating a lack of population structure and possibly high levels of gene flow via seed dispersal in what is predominantly a selfing species. Senecio cambrensis showed closer similarity in AFLP phenotype to S. vulgaris than to S. squalidus. Possible causes of this and also the high level of AFLP diversity found in S. cambrensis are discussed. It is suggested that intergenomic recombination following occasional multivalent formation during meiosis in S. cambrensis is likely to be an important cause of both phenomena, although other causes are not ruled out.  相似文献

Sequence analysis of the internal transcribed spacer (ITS) of the 18S(ITS1)-5.8S-26S(ITS2) rDNA region was performed in order to analyse the phylogenetic relationships between 13 Patagonian species of the genus Berberis (Berberidaceae). The divergence values between the pairwise sequence in the studied Patagonian species were in the range 2.9–22.9%. The lengths of the ITS1 and ITS2 sequences were in the range 227–231 bp and 220–224 bp, respectively, and the 5.8S sequence was 159 bp throughout all species . B. microphylla sensu Landrum does not appear to be monophyletic based on current sampling. Indeed, we suggest that B. microphylla should be distinguished from B. buxifolia , B. parodii , and B. heterophylla . Spartina anglica arose during the end of the 19th century in England by hybridization between the indigenous Spartina maritima and the introduced East American Spartina alterniflora and following genome duplication of the hybrid ( S.  ×  townsendii ). This system allows investigations of the early evolutionary changes that accompany stabilization of a new allopolyploid species in natural populations. Various molecular data indicate that S. anglica has resulted from a unique parental genotype. This young species contains two distinctly divergent homoeologous genomes that have not undergone extensive change since their reunion. No burst of retroelements has been encountered in the F1 hybrid or in the allopolyploid, suggesting a 'structural genomic stasis' rather than 'rapid genomic changes'. A new species of Holcus L. (Poaceae), endemic to the Azores archipelago (Portugal), is described. The vegetative and reproductive structures were studied. The diagnostic characters were compared with those of closely related species. New chromosome counts in Holcus azoricus M. Seq. & Castrov. (2 n  = 35) and H. rigidus Hochst. (2 n  = 28) are included. Eleocharis subarticulata is recorded as the third species of Cyperaceae with a reduced chromosome number ( n  = 3), following reports on Rhynchospora tenuis ( n  = 2) and Fimbristylis umbellaris ( n  = 3). For Eleocharis, the numbers recorded to date vary from 2 n  = 10 to 2 n  = c. 196, with x  = 5 as the possible basic number. The karyotype of E. subarticulata was studied using conventional staining (mitosis and meiosis), C-CMA3/DAPI banding, and FISH with 45S rDNA and telomere probes. The chromosomes showed no primary constrictions, as expected in the holocentric chromosomes of Cyperaceae. The meiotic behaviour was abnormal, with a single multivalent ring of six chromosomes at metaphase I, resulting from multiple translocations. At anaphase I six chromatids migrated to each pole, evidencing the inverted meiosis, and these groups were also visible at metaphase II. The C-CMA3/DAPI banding technique showed only four terminal GC-rich blocks. FISH with 45S rDNA probes revealed four terminal signals, probably associated with GC-rich blocks. The telomeric probe located terminal signals in all the chromosomes, besides a hybridization site in the middle of the large pair. The occurrence of ectopic telomeric sites has not been described previously for plants with holokinetic karyotypes and with reduced chromosome numbers.
