Studies of variability in Holm oak (Quercus ilex subsp. ballota [Desf.] Samp.), the dominant tree species in the typical Mediterranean forest, have been carried out by using electrophoresis-based proteomic analysis of acorns. Ten populations distributed throughout the Andalusia region have been surveyed. Acorns were sampled from individual trees and proteins extracted from seed flour by using the TCA-acetone precipitation protocol. Extracts were subjected to SDS-PAGE and 2-DE for protein separation, gel images captured, spot or bands quantified, and subjected to statistical analysis (ANOVA, SOM and clustering). Variable bands or spots among populations were subjected to MALDI-TOF/TOF and LC-MS/MS for identification. The protein yield of the used protocol varied among populations, and it was in the 2.92-5.92 mg/g dry weight range. A total of 23 bands were resolved by SDS-PAGE in the 3-35 kDa Mr range, with 8 and 12, out of the total, showing respectively qualitative and quantitative statistically significant differences among populations. Data allowed grouping populations, with groups being correlated according to geographical location and climate conditions, to northern and southern, as well as the discrimination of both mesic and xeric groups. Acorn flour extracts from the most distant populations were analyzed by 2-DE, and 56 differential spots were proposed as markers of variability. Identified proteins were classified into two principal categories; storage and stress/defense protein. Besides providing the first reference map of mature acorn seeds, the use of SDS-PAGE and proteomics in characterizing natural biodiversity in forest trees will be discussed. 相似文献

Nursery researchers tend to study seedling quality instead of other sylvicultural practices such as soil preparation and planting date. The aim of this study was to determine the effects and interactions of site preparation and stock quality on the survival and growth of 1-year-old Quercus ilex L. seedlings planted on different dates. Based on the hypothesis that soil preparation affects out-planting performance more than stock quality does in Mediterranean areas, two different site preparations (subsoiling and manual holing) and three planting dates were studied. Two years after planting, high-quality seedlings planted on an early date over a subsoiling preparation showed the best survival rates (61%), followed by the same quality plant and soil preparation treatments on a mid-season planting date (40%). After two growing seasons, planting date and site preparation affected height growth rate positively, whereas relative diameter growth rate of surviving seedlings was affected by planting date only. A correct selection of the planting date and soil treatment plays an important role in the expression of seedling quality in terms of survival and growth. 相似文献

Variation in the lengths of restriction fragments (RFLPs) of the whole chloroplast DNA molecule was studied in 174 populations of Quercus ilex L. sampled over the entire distribution of this evergreen and mainly Mediterranean oak species. By using five endonucleases, 323 distinct fragments were obtained. From the 29 and 17 cpDNA changes identified as site and length mutations, respectively, 25 distinct chlorotypes were distinguished, mapped and treated cladistically with a parsimony analysis, using as an outgroup Q. alnifolia Poech, a closely related evergreen oak species endemic to Cyprus where Q. ilex does not grow. The predominant role of Q. ilex as maternal parent in hybridization with other species was reflected by the occurrence of a single very specific lineage of related chlorotypes, the most ancestral and recent ones being located in the southeastern and in the northwestern parts of the species’ geographical distribution, respectively. The lineage was constituted of two clusters of chlorotypes observed in the ‘ilex’ morphotyped populations of the Balkan and Italian Peninsulas (including the contiguous French Riviera), respectively. A third cluster was divided into two subclusters identified in the ‘rotundifolia’ morphotyped populations of North Africa, and of Iberia and the adjacent French regions, respectively. Postglacial colonization probably started from three distinct southerly refugia located in each of the three European peninsulas, and a contact area between the Italian and the Iberian migration routes was identified in the Rhône valley (France). Chlorotypes identical or related to those of the Iberian cluster were identified in the populations from Catalonia and the French Languedoc region, which showed intermediate morphotypes, and in the French Atlantic populations which possessed the ‘ilex’ morphotype, suggesting the occurrence of adaptive morphological changes in the northern part of the species’ distribution. 相似文献

Cotyledon explants of Panax ginseng produced somatic embryos directly on solid hormone-free MS medium containing 3% (w/v) sucrose while high concentration of NH4NO3 (60 mM) induced embryogenic callus. Ten subcultures of the embryogenic callus on hormone-free MS medium with 40 mM NH4NO3 gave hormone-independent proliferation of callus, which exhibited proliferation potential even on MS medium with a standard level of NH4NO3 (20 mM). Pulse treatment of callus with exogenous auxin or cytokinin (1.0 mg 1–1 2,4-D, 1.0 mg 1–1 kinetin) resulted in the loss of the hormone-independent characteristic and caused the callus to brown. For the suspension culture, embryogenic callus was transferred to MS liquid medium containing 3% (w/v) sucrose in an 500 ml Erlenmyer flask. Embryogenic cell clumps in full-strength MS liquid medium discharged toxic substances, resulting in strong suppression of cell growth. In 1/3-strength MS medium, exudation of toxic material did not occur. Embryogenic cell clumps were mass-grown on a large-scale in a bioreactor (20-1), showing a 7.1 increase of fresh weight in 1/3-strength MS medium with 3% (w/ v) sucrose after 5 weeks of culture. Total ginsenoside content of cultured embryogenic cell clumps was low and 6 times below naturally-cultivated ginseng roots. 相似文献

Holm oak (Quercus ilex L.) forest is one of the most widespread biocoenoses in the southern part of France. Until recently, clearcutting of wood for domestic use was carried out every 20 years or so in these coppice stands. In order to study coppice stand dynamics after cutting this paper presents results of observations of some items describing the vegetation structure, the floral composition, and the life cycles in a holm oak coppice stand during a six year period after clearcutting. One of the questions addressed in this study was whether these coppice stands reach a steady state based on auto-succession, or if they continue to change. The results of the floristic changes after cutting suggest that the model of auto-succession best fits with the vegetation dynamics, even if changes in the density and life cycles of herbaceous species still exist six years after clearcutting. 相似文献

The nuclear genetic variation within and between four sessile ( Q. petraea) and six pedunculate ( Q. robur) autochthonous Flemish oak populations was investigated with AFLP markers. One sessile and one pedunculate oak population were additionally screened for detailed leaf characteristics using an image analysis system. Principal coordinate analysis on the AFLP data classified the oaks in two main groups, according to their taxonomic status. No species-specific AFLP markers were found using four primer combinations, but marker frequency differences up to 71% were recorded between both species. Analysis of the genetic structure showed that the divergence between species, as observed by ordination, was significant. Both species revealed similar diversity levels. A smaller though significant differentiation was also revealed for both species among populations within species. Molecular and morphology based approaches showed a high degree of consistency. Screening of 60 AFLP primer combinations using a bulking strategy did not allow identifying species-specific markers, which supports the conclusions reached in previous studies. The distribution of genetic variability at the species and at the population level is discussed. 相似文献

We investigated the effects of a short period of water stress on monoterpene emissions from Quercus ilex, a common oak species of the Mediterranean vegetation and a strong emitter of monoterpenes. The experiment was carried out on two young saplings with a branch enclosure system under semi-controlled conditions. Under unstressed conditions, small qualitative (cis--ocimene, trans--ocimene, -caryophyllene and 1,8-cineol) and large quantitative (as much as 40% for the main compounds emitted) differences were observed between the two apparently similar trees. Nevertheless these differences did not affect the short-and long-term responses to temperature and water stress. Daily courses of emissions and gas exchanges were similar before and after the stress. During the most severe stress, emissions were reduced by a factor of two orders of magnitude and the log-linear relationship between emissions and temperature no longer existed. Photosynthesis and transpiration rates decreased as soon as the soil started to dry, whereas monoterpene emissions slightly increased for few days and then dropped when the daily CO2 balance approached zero. We concluded that under water stress monoterpene emissions were highly limited by monoterpene synthesis resulting from a lack of carbon substrate and/or ATP. After rewatering, both emissions and gas exchanges recovered immediately, but to a level lower than the pre-stress level. These results have many implications for monoterpene emission modelling in the Mediterranean area, since the dry period generally extends from May to August. If our results are confirmed by field experiments, water stress could lead to a large overestimation of the emissions under summer conditions, when the algorithms based on light and temperature would give high emission rates. 相似文献

At three sites in northwestern Germany, which represent the centres of the present oak damage, root distribution and biomass beneath healthy and damaged trees of mature pedunculate oak (Quercus robur L.; Neuenburg site) and sessile oak (Q. petraea [Matt.] Liebl.; Lappwald and Sprakensehl sites) were investigated, and soil texture, bulk density, duration of waterlogging periods and the water available in the mineral soil were determined. For Neuenburg and Sprakensehl, the available soil water was related to leaf water parameters determined in a separate investigation. At the clayey and hydromorphic sites of Neuenburg and Lappwald, the measurements were performed in each one healthy and one damaged part of the site, which differed in the number of oaks with crown damage. In the damaged stand of Neuenburg, the clay content of the subsoil was higher than in the healthy stand, and the soil water availability was reduced especially in dry periods. Compared to healthy oaks of the healthy stand, the density of finest plus fine roots as well as the biomasses of finest roots were lower beneath damaged oaks of the damaged stand. With decreasing relative available soil water (actually available water in relation to water available at the saturation state), the relative leaf water content decreased in damaged, but not in healthy oaks. At Lappwald, similar differences in soil water availability between the healthy and the damaged stand were found, but had no effect on the distribution or biomass of the roots. At the sandy site (Sprakensehl), the available soil water decreased drastically during a dry period, and predawn leaf water potentials of both healthy and damaged oaks declined with decreasing relative available soil water. However, the damaged oaks were not inferior to the healthy ones with respect to root density and biomass. It is concluded that, in the damaged stand of Neuenburg, the high clay content of the subsoil, which results in prolonged periods of waterlogging, in sharp changes from waterlogging to drought and decreased water availability in dry periods, is the reason for the reduced biomass and density of roots of the pedunculate oak. Thus, in northwestern Germany, unfavourable soil water relations are considered as a factor contributing to crown damage of pedunculate oak at hydromorphic sites, but not to damage of sessile oak. 相似文献

Holm oak (Quercus ilex ssp. ballota [Desf.] Samp) is one of the most important species in forest communities of the western Mediterranean Basin, but is very
vulnerable to environmental stress during the first years of its life. In particular, summer drought and winter frosts limit
the distribution, survival, and growth of Holm oak. These two stress factors can lead to plasmolysis and/or oxidative stress.
We hypothesized that autumn fertilization with nitrogen (N) can improve plant tolerance to these abiotic stress factors and
improve plant quality and therefore reforestation success. A 12-week autumn application of 30 or 60 mg N (plus 70 mg N applied
to both treatments during the previous 28 weeks in the nursery, i.e. 100 and 130 mg N in total, respectively) improved overall
growth, root growth capacity, frost tolerance, and nutritional status of plants relative to plants given 0.0 and 1.5 mg autumnal
N, and had no negative effect on seedling response to water stress. A very small increment in N doses during the autumn (1.5
vs. 0.0 mg N) improved some morphological parameters, such as stem diameter (D) and shoot dry weight, and physiological parameters, such as total antioxidant activity. The highest autumnal N dose (60 mg)
increased leaf nutrient content without decreasing the concentration, but had a tendency to reduce frost tolerance relative

Organic matter distribution and fluxes within a holm oak (Quercus ilex L.) stand in the Etna volcano
Located at 1100 m above sea level, on the western site of the Etna volcano, the ecophysiology of the Mount Minardo holm oak coppice has been investigated for more then twenty years. In this stand, now 31 years old, the above ground biomass amounts to 15000 g m-2 of organic material, including leaves and perennial woody material. During these 31 years, the mean annual production has been around 775 g m-2. The yearly mean litterfall amounts to 310 g m-2, including 200 g m-2 of leaves, mostly two years old. The soil surface is covered by a litter layer amounting to 3150 g m-2. Each year, following Jenny's decomposition rate and field measurements, 290 g m-2 of the litter turns into CO2, or becomes incorporated in the soil organic matter. 相似文献

The effect of clearcutting on biomass production and nutrient consumption was examined during the two years in a clearcut and a mature Quercus ilex L. coppice. The comparison was limited to the leaf compartment and to the annual shoots, which were the only comparable compartments in the two situations. At the same time, the dynamics of the nutrient pool (N, P, K, Ca, Mg), monthly uptake and recycling from the foliage, were studied.The second year after cutting, the leaf production was roughly the same at the two sites. However, the photosynthates appear to be used differently: in the young stand the perennial wood production was greater than in the mature stand, where the maintenance costs are higher. This article studies the influence of cleaning or thinning intensity and of coppice age at the time of silvicultural treatment, on the growth of shoots remaining on the stool, in Quercus ilex coppices.Two experimental designs were used to answer these questions. The first studied the effect of two cleaning intensities on 4, 8, 15, 20, and 25 year old coppices. The second studied the effect of 4 thinning intensities (26%, 42%, 58%, or 78% removal of the initial basal area) on 43 and 57 year old coppices. In both cases, control plots undergoing no silvicultural treatment were maintained for each age class.The results, 4 and 5 years after silvicultural treatment, concerned girth increment and height growth of coppice shoots.Cleaning or thinning consistently had a positive influence on girth increment. In young coppices, less than 25 years old, mean annual girth increment was approximately 6 mm in the controls and double that under heavy cleaning. In older coppices, girth increment was lower in controls (3 mm per year) but the effect of thinning was very positive and, under very heavy thinning, growth was more than 11 mm per year.Cleaning and thinning often modify height growth. This effect was found to be either positive or negative and no general trend could be defined which was valid for all the age classes.Silvicultural treatments also had a positive effect on the appearance and growth of epicormic shoots on the holm oak trunks.The results were also analysed at the stool level. The number or percentage of shoots removed had a positive effect on the individual growth of selected shoots. 相似文献

In a laboratory study, we investigated the monoterpene emissions from Quercus ilex, an evergreen sclerophyllous Mediterranean oak species whose emissions are light dependent. We examined the light and temperature responses of individual monoterpenes emitted from leaves under various conditions, the effect of heat stress on emissions, and the emission-onset during leaf development. Emission rate increased 10-fold during leaf growth, with slight changes in the composition. At 30 °C and saturating light, the monoterpene emission rate from mature leaves averaged 4·1 nmol m–2 s–1, of which α-pinene, sabinene and β-pinene accounted for 85%. The light dependence of emission was similar for all monoterpenes: it resembled the light saturation curve of CO2 assimilation, although monoterpene emission continued in the dark. Temperature dependence differed among emitted compounds: most of them exhibited an exponential increase up to 35 °C, a maximum at 42 °C, and a slight decline at higher temperatures. However, the two acyclic isomers cis-β-ocimene and trans-β-ocimene were hardly detected below 35 °C, but their emission rates increased above this temperature as the emission rates of other compounds fell, so that total emission of monoterpenes exponentially increased from 5 to 45 °C. The ratio between ocimene isomers and other compounds increased with both absolute temperature and time of heat exposure. The light dependence of emission was insensitive to the temperature at which it was measured, and vice versa the temperature dependence was insensitive to the light regime. The results demonstrated that none of the models currently applied to simulate isoprene or monoterpene emissions correctly predicts the short-term effects of light and temperature on Q. ilex emissions. The percentage of fixed carbon lost immediately as monoterpenes ranged between 0·1 and 6·0% depending on temperature, but rose up to 20% when leaves were continuously exposed to temperatures between 40 and 45 °C. 相似文献

An 859-bp cDNA segment of a terpene synthase gene was amplified by PCR from the evergreen sclerophyllous holm oak (Quercus ilex L.) using heterologous primers for conserved regions of terpene synthase genes (TPS) in dicotyledonous plants. Based on the sequence of this segment, homologous primers were designed for amplification by RACE-PCR of a cDNA segment carrying the monoterpene synthase gene myrS. The gene encodes a protein of 597 amino acids including an N-terminal putative plastid transit peptide. The gene without the segment encoding the transit peptide was cloned by PCR into a bacterial expression vector. Expression in Escherichia coli yielded an active monoterpene synthase, which converted geranyl diphosphate (GDP) predominantly into the acyclic monoterpene myrcene and to a very small extent into cyclic monoterpenes. Sequence comparison with previously cloned monoterpene synthases revealed that the myrcene synthase from Q. ilex belongs to the TPSb subfamily. 相似文献

Nutrient availability is a key factor in Mediterranean ecosystems that affects the primary productivity and the community structure. The great variability of its natural availability is now increasing due to frequent fires, pollution events and changes in rainfall regime associated to climate change. Quercus ilex ssp. ballota and Pinus halepensis are the most abundant tree species in the NW Mediterranean basin. They frequently compete in the early and middle successional stages. We investigated the effects of N and P pulse supplies on nutrient uptake capacity in these two species in an after-fire field area and in nursery conditions on different soil types and competing conditions. In the field, N fertilisation had weak effects on nutrient concentration and mineralomass likely as a consequence of this nutrient not being limiting in this field site whereas P fertilisation increased the P mineralomass and the Mg, S, Fe, K and Ca concentrations and mineralomass in the different biomass fractions of both species 1 and 3 years after fertilisation application. In the nursery experiments, P fertilisation increased the mineralomass and concentrations of P, Mg, S, Fe, K and Ca in all biomass fractions including the roots in both species and in different soils and competition conditions. The increment of nutrient mineralomass was due to both the increase of growth and of nutrient concentrations. Both species were able to absorb significant amounts of the P applied by fertilisation (between 5 and 20%) in short time (18 months). Competing vegetation decreased the positive effects of P fertilisation, and in many cases the negative effect of competing vegetation on nutrient mineralomass was stronger when P availability was increased by fertilisation. Q. ilex subsp ballota showed a greater competitive ability for P than the more pioneer species Pinus halepensis in the field but not in the nursery conditions. Pinus halepensis had greater nutrient mineralomass in calcareous than in siliceous soils. Q. ilex subsp. ballota had a higher root biomass allocation and root nutrient allocation than P. halepensis, but both species showed a high capacity to increase their nutrient uptake when its availability increased by fertilisation, thus assuring a great nutrient reserve for future growth periods and contributing to retain nutrients in the ecosystem. 相似文献

We investigated the response of the photosynthetic apparatus during an episode of extreme low winter temperature in Quercus ilex subsp. ballota (Desf.) Samp., a typical Mediterranean evergreen species in the Iberian peninsula. Both plants in a woodland located at high altitude (1,177 m. a.s.l.) and potted plants obtained from acorns of the same populations grown at low altitude (225 m. a.s.l.) were analyzed. Net CO2 assimilation rate was negative and there was a marked decrease in photosystem II (PSII) efficiency during winter in leaves of the woodland population (high altitude individuals). These processes were accompanied by increases in non-photochemical quenching (NPQ) and in the de-epoxidated carotenoids within the xanthophyll cycle, mechanisms aimed to dissipate excess energy. In addition, these de-epoxidated carotenoids were largely preserved during the night. There was no chlorophyll bleaching during the winter, which suggests that leaves were not experiencing photoinhibitory damage. In fact, the net photosynthetic rate and the PSII efficiency recovered in spring. These changes were not observed, or were much more reduced, in individuals located at lower altitude after a few frosts. When the response to rapid temperature changes (from 20°C to –5°C and from –5°C to 20°C) was studied, it was found that the maximum potential PSII efficiency was fairly stable, ranging from 0.70 to 0.75. The rest of the photosynthetic parameters measured, actual and intrinsic PSII efficiency, photochemical and NPQ, responded immediately to the changes in temperature and, also, the recovery after cold events was practically immediate. 相似文献

Sun and shade leaves of two Mediterranean Quercus species, Quercus ilex subsp. ballota (Desf.) Samp. and Quercus coccifera L., were compared by measuring leaf optical properties, photosynthetic pigment composition and photosystem II efficiency. The presence of trichomes in the adaxial (upper) leaf surface of Q. ilex subsp. ballota seems to constitute an important morphological mechanism that allows this species to maintain a good photosystem II efficiency during the summer. Q. coccifera has almost no trichomes and seems instead to develop other physiological responses, including a smaller light-harvesting antenna size, higher concentrations of violaxanthin cycle pigments and a higher (zeaxanthin + antheraxanthin)/(violaxanthin + antheraxanthin + zeaxanthin) ratio. Q. coccifera was not able to maintain a good photosystem II efficiency up to the end of the summer. In Q. ilex subsp. ballota leaves, natural loss or mechanical removal of adaxial-face leaf trichomes induced short-term decreases in photosystem II efficiency. These changes were accompanied by de-epoxidation of violaxanthin cycle pigments, suggesting that the absence of trichomes would trigger physiological responses in this species. Our data have revealed different patterns of response of Q. ilex subsp. ballota and Q. coccifera facing the stress conditions prevailing in the Mediterranean area. 相似文献

An increase in the frequency and intensity of extreme climatic conditions is expected for the Mediterranean area as a response to climate change. As a consequence, the ability of Mediterranean plant species to adapt to complex and stressful environmental conditions plays an important role in driving their future distribution. The adaption of plant species may be expressed by ecotypes already adapted to local climate. Our goal was to analyse the seasonal physiological behaviour of five Quercus ilex ecotypes coming from different Italian geographical areas (from the north to the south) in order to test if ecotypes maintained their physiological traits when grown in the same environmental conditions. Measurements of gas exchange, biochemistry and chlorophyll fluorescence carried out during winter, spring and summer underlined that the response of the considered ecotypes reflected the climate of the original provenances, particularly under suboptimal conditions. The ecotypes from the northernmost and the southernmost limits were the most sensitive to high and low temperatures, respectively. Our results can be used to advance hypotheses about the respone of Q. ilex to climate change. 相似文献
