The purpose of this research was to mask the intensely bitter taste of ondansetron HCl and to formulate a rapiddisintegrating tablet (RDT) of the taste-maske drug. Taste masking was done by complexing ondansetron HCl with aminoalkyl methacrylate copolymer (Eudragit EPO) in different ratios by the precipitation method. Drug-polymer complexes (DPCs) were tested for drug content, in vitro taste in simulated salivary fluid (SSF) of pH 6.2, and molecular property. Complex that did not release drug in SSF was considered taste-masked and selected for formulation RDTs. The complex with drug-polymer ratio of 8∶2 did not show drug release in SSF; therefore, it was selected. The properties of tablets such as tensile strength, wetting time, water absorption ratio, in vitro disintegration time, and disintegration in the oral cavity were investigated to elucidate the wetting and disintegration characteristics of tablets. Polyplasdone XL-10 7% wt/wt gave the minimum disintegration time. Tablets of batch F4 containing spray-dried mannitol and microcrystalline cellulose in the ratio 1∶1 and 7% wt/wt Polyplasdone XL-10 showed faster disintegration, within 12.5 seconds, than the marketed tablet (112 seconds). Good correlation between in vitro disintegration behavior and in the oral cavity was recognized. Taste evaluation of RDT in human volunteers revealed considerable taste masking with the degree of bitterness below threshold value (0.5) ultimately reaching to 0 within 15 minutes, whereas ondansetron HCl was rated intensely bitter with a score of 3 for 10 minutes. Tablets of batch F4 also revealed rapid drug release (t90, 60 seconds) in SGF compared with marketed formulation (t90, 240 seconds;P<.01). This study compares the effects of rest intervals on isokinetic muscle torque recovery between sets of a knee extensor and flexor exercise protocol in physically active younger and older women. Twenty young (22.4 +/- 1.7 years) and 16 older (70.7 +/- 4.3 years) women performed three sets of eight maximum repetitions of knee extension/flexion at 60 degrees x s(-1). The rest interval between sets was 15, 30, and 60 seconds and was randomly assigned across three testing days. No significant interaction of rest by set by age group was observed. There was a significant decline in mean knee extensor torque when 15- and 30-second rest intervals were used between sets, but not when a 60-second rest interval was applied for both the young and the old women. No significant decline for mean knee flexor torque was observed in the older women when a 30-second rest interval was used, whereas a longer 60-second rest interval was required in younger women. Active younger and older women require similar rest intervals between sets of a knee extensor exercise (60 seconds) for complete recovery. However, older women recovered faster (30 seconds) than younger women (60 seconds) between sets of a knee flexor exercise. The exercise-to-rest ratio for knee extensors was similar for young and old women (1:2). Old women required only a 1:1 exercise-to-rest ratio for knee flexor recovery, whereas younger women required a longer 1:2 exercise-to-rest ratio. The results of the present study are specific to isokinetic testing and training and are more applicable in rehabilitation and research settings. Practitioners should consider age and gender when prescribing rest intervals between sets.  相似文献

Three experiments were conducted to determine the effect of ultraviolet (UV) radiation on poliovirus-contaminated seawater. In two of the experiments, the effectiveness of the Kelly-Purdy UV Seawater Treatment Unit to inactivate poliovirus type 1 (T(1)) suspended in continuously flowing seawater was determined. In experiment 1, the observed survival ratio of poliovirus T(1) was 2.3 x 10(-4) (99.98% reduction) in 15.7 sec. No virus was detected (<0.2 plaque-forming unit/ml) in 20.6 seconds. The calculated half-life value was 1.29 sec. In experiment 2, the observed survival ratio of poliovirus T(1) was 5.9 x 10(-4) (99.94% reduction) in 11.7 sec. No virus was detected in 15.7 sec. The calculated half-life value was 1.37 sec. In experiment 3, a laboratory-controlled UV experiment designed to closely simulate the geometry of the continuously flowing seawater system, the observed survival ratios of poliovirus T(1) were 9.7 x 10(-3) (99.03% reduction) and 3.6 x 10(-4) (99.96% reduction) in 15 and 30 sec, respectively; the calculated half-life value was 2.38 sec. A statistically significant difference was found between the inactivation rates of poliovirus T(1) in the two test systems. This rate difference was attributed primarily to UV dosage and stirring effects. The data indicated that UV radiation effectively inactivated poliovirus T(1) in flowing seawater. These results validate the efficacy of the Kelly-Purdy UV Seawater Treatment Unit for use in commercial depuration systems.  相似文献

Summary Mechanical and visual stimulation lasting for fractions of seconds to few seconds induce inTenebrio molitor at larval stage coordinate motor responses. Such movements are oscillatory in nature, their frequency is about 0.1 Hz and this value is largely independent from the intensity of the stimulus, their amplitude is proportional to it.About 50s after the cessation of the stimulus the oscillations tend to fade away. During the oscillatory behaviour the thorax plays an active role in contrast with the more passive abdomen.These motor responses can be elicited by different stimulation; visual and mechanical, only visual, only mechanical, and are still present in an experimental contest in which both light and mechanical stimuli are absent. The ability of the larva ofTenebrio to play a coordinate motor program lasting for a noticeable time enables it in maintaining a coherent exploratory strategy in total absence of visual and mechanical inputs.The importance of such oscillatory responses has been analyzed in the general contest of the correcting behaviour. Its biological significance resides presumably in a deep cooperation between the central nervous system and the motor machinery in setting up a memory process or in recalling recent experiences.  相似文献

Host-size related feeding and oviposition behaviour, and allocation of progeny sex by Anisopteromalus calandrae (Howard) were tested on Sitophilus oryzae L. The parasitoid showed a host-size-dependent partition of feeding and oviposition behaviour, preferring small hosts for feeding, but large hosts for oviposition. Neither the mutual interference nor the host density showed any effect on the behaviour of the parasitoid. Allocation of progeny sex by the female parasitoid appeared to be based more likely on absolute than on relative host size encountered. A model for the progeny sex ratio was constructed based on: (1) ovipositional preference of the parasitoid on large hosts; (2) feeding preferentially on small hosts; and (3) host-size-related regulation of progeny sex ratio. The progeny sex ratio of the parasitoid predicted by the model was in close agreement with the observed value.  相似文献

Previous research has indicated that short-duration, high-intensity work intervals performed at velocities associated with maximal oxygen uptake (vVO2max) combined with active recovery intervals may be effective in eliciting improvements in endurance performance. This study was designed to characterize selected physiological responses to short-duration (< or = 60 seconds) interval work performed at velocities corresponding to 100% of vVO2max. Twelve men participated in 3 randomized trials consisting of treadmill running using work (W)/recovery (R) intervals of 15 seconds W/15 seconds R (15/15); 30 seconds W/15 seconds R (30/15); and 60 seconds W/15 seconds R (60/15). Work intervals were performed at 100% of vVO2max, whereas R intervals were performed at 50% of vVO2max. A fourth trial consisting of continuous work (C) at 100% of vVO2max was also performed. All subjects completed the 15/15 and 30/15 trials; however, only 5 of the 12 completed the 60/15 trial. The percentage of VO2max (mean +/- SD) during 15/15 (71.6 +/- 4.2%) was significantly lower (p < or = 0.05) than the percentages during 30/15 (84.6 +/- 4.0%), 60/15 (89.2 +/- 4.2%), or C (87.9 +/- 5.0%). Similar results were found for heart rate and perceived exertion. Blood lactate concentrations following exercise were significantly lower (p < or = 0.05) in 15/15 (7.3 +/- 2.4 mmol x L(-1)) than in the other trials. No significant differences (p > 0.05) existed among 30/15 (11.5 +/- 1.8 mmol x L(-1)), 60/15 (12.5 +/- 1.8 mmol x L(-1)) or C (12.1 +/- 1.8 mmol x L(-1)). High intensity, short-duration 2:1 W/R intervals appear to produce responses that may benefit both aerobic and anaerobic energy system development. A 4:1 W/R ratio may be an upper limit for individuals in the initial phases of interval training.  相似文献

The ability of ten baker's yeast strains to synthesize sterols was checked. Ergosterol/24(28)-dehydroergosterol (E/D) ratio had the value of 0.8 to 1 in most strains grown in a medium containing molasses as the carbon source. The ratio values were significantly increased in the cultures grown in a glucose medium. Both a decrease in the content of 24(28)-dehydroergosterol and a slight increase of the ergosterol content were found to be responsible for the high values of E/D in the glucose-grown cells. The strains examined could be divided into three groups on the basis of their behaviour towards glucose. The effect of the type of cultivation on sterol accumulation is demonstrated by comparing the sterol content of the representatives of the three groups of baker's yeasts grown in a fermenter or in shaken flasks.  相似文献

The purpose of this study was to determine the optimal 1.63-km (1-mile) pacing strategy for 5-km running performance in moderately trained women distance runners. Eleven women distance runners (20.7 +/- 0.8 years, 163.8 +/- 2.0 cm, 57.0 +/- 2.2 kg, 51.7 +/- 1.0 ml.kg(-1).min(-1), 18.9 +/- 0.8% fat, 78.1 +/- 1.4% VO(2)max at lactate threshold) performed 2 preliminary 5-km time trials on a treadmill to establish baseline 5-km times. The average 1.63-km split pace of the fastest preliminary trial was manipulated for the first 1.63 km of the experimental trials and run either equal to (EVEN), 3% faster than (3%), or 6% faster than (6%) the current baseline average 1.63-km pace for each subject. Ventilation (V(E)), oxygen consumption VO(2)max )), respiratory exchange ratio, and heart rate were measured continuously. Overall 5-km times were not different (p > 0.05) for the EVEN, 3% and 6% trials finishing in 21:11 (minutes/seconds) +/- 29 seconds, 20:52 +/- 36 seconds and 20:39 +/- 29 seconds, respectively. The fastest time for 8 subjects resulted from the 6% trial and the other 3 subjects' fastest times resulted from the 3% trial. The overall exercise intensity (%VO(2)max , %VO(2)max above lactate threshold, V(E), and respiratory exchange ratio) of the first 1.63-km split was not different between the 3 and 6% trials, despite the 6% trial being 13 seconds faster than the 3% trial. Based on these findings, initial 1.63-km starting paces of a 5-km race can be 3 to 6% greater than current average race pace without negatively impacting performance. In order to optimize 5-km performance, runners should start the initial 1.63 km of a 5-km race at paces 3-6% greater than their current average race pace.  相似文献

BackgroundThe six-second spirometry has been proposed as an alternative to diagnose airflow limitation, although its prognostic value in patients with chronic obstructive pulmonary disease (COPD) remains unknown. The purpose of this study was to determine the prognostic value of the postbronchodilator forced expiratory volume in 1 second (FEV1)/forced expiratory volume in 6 seconds (FEV6) ratio and FEV6 in COPD patients.ConclusionsIn a general COPD outpatient population, airflow obstruction assessed by the FEV1/FEV6 is an independent risk factor for both death and hospitalization.  相似文献

The effect of 2,4-dichlorophenol (DCP) was studied on the fully hydrated 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC)--water liposomes. The structure and the thermotropic phase behaviour of the liposomes was examined in the presence of DCP (DCP/DPPC molar ratio, varied from 2x10(-2) up to 1) using small- and wide-angle X-ray scattering (SAXS, WAXS) and freeze-fracture electron microscopy. The structural behaviour of the DPPC/DCP/water system was strongly dependent on the concentration of the DCP. In the pretransition range the DCP molecules (at 2x10(-2) DCP/DPPC molar ratio) induced the interdigitated phase beside the parent (gel and rippled gel) phases, locally which can be form at higher DCP concentration. When the DCP/DPPC molar ratio was increased the pretransition disappeared and the main transition was shifted to lower temperatures. In the molar ratio range from 2x10(-1) up to 5x10(-1), a coexistence of different phases was observed in the wide temperature range from 20 up to 40 degrees C. With a further increase of the DCP/DPPC molar ratio (6x10(-1) to 1) only the interdigitated gel phase occurred below 25 degrees C. A schematic phase diagram of DPPC/DCP/water system was constructed to summarise the results.  相似文献

1. Vasodilatation induced by venous blood infusion and reactive hyperaemia following the reduction of perfusion pressure for different lengths of time (1 to 128 sec) were studied in the haemodynamically isolated and denervated vascular bed of the dog gracilis muscle. The two local regulatory situations differed chiefly in respect to the oxygen concentration in the precapillary part of the blood bed (51.4% and 97.6% respectively) and in the size of the blood flow (100% and 48.5% of the resting value). 2. The interval between the time when venous blood entered the resistant precapillary vessels and the onset of the local regulatory response was 25.1 +/- 1.9 seconds. In prolongation of the duration of reduced perfusion pressure, the continuous increase in the maximum reactive hyperaemia value was interrupted, on reducing it for 16-32 seconds, by a significant (P less than 0.001) abrupt increase in this value. 3. We conclude from the good agreement of these two time values that: 1. reduction of the blood oxygen concentration to 51.4% does not directly affect the smooth muscle cells in the wall of resistant vessels; 2. an interval of 25.1 +/- 1.9 sec (16-32 seconds) is needed for interference with the metabolism of skeletal muscle to attain a critical value and for the relevant chemical signal to produce a dilatation response in the arterioles.  相似文献

The tumbling dynamics of a 20-mer HIV-1 RNA stem loop 3 spin-labeled at the 5' position were probed in the nanosecond time range. This RNA interacted with the HIV-1 nucleocapsid Zn-finger protein, 1-55 NCp7, and specialized stopped-flow EPR revealed concomitant kinetics of probe immobilization from milliseconds to seconds. RNA stem loop 3 is highly conserved in HIV, while NCp7 is critical to HIV-RNA packaging and annealing. The 5' probe did not perturb RNA melting or the NCp7/RNA interaction monitored by gel shift and fluorescence. The 5'-labeled RNA tumbled with a subnanosecond isotropic correlation time (approximately 0.60 ns at room temperature) reflecting both local viscosity-independent bond rotation of the probe and viscosity-dependent diffusion of 40-60% of the RNA. The binding of NCp7 to spin-labeled RNA stem loop 3 in a 1:1 ratio increased the spin-labeled tumbling time by about 40%. At low ionic strength with a ratio of NCp7 to RNA >or=3 (i.e., an NCp7 to nucleotide ratio or=3:1 complex also required intact Zn fingers. Stopped-flow EPR kinetics with NCP7/RNA mixed at a 4:1 ratio showed the major phase of NCp7 interaction with RNA stem loop 3 occurred within 4 ms, a second phase occurred with a time constant of approximately 30 ms, and a slower immobilization, possibly concomitant with large complex formation, proceeded over seconds. This work points the way for spin-labeling to investigate oligonucleotide-protein complexes, notably those lacking precise stoichiometry, that are requisite for viral packaging and genome fabrication.  相似文献

This experiment aimed to identify how a pig's age affected the extent and synchrony of use of different environmental enrichment materials, and how this use changed over time. Ten diverse novel objects were each presented to three replicate litters of 3 weeks of age (sucklers) and three replicate groups of three animals of 5 (weaners) weeks and 13 (growers) weeks of age. Video recordings were made of the pigs’ behaviour over a period of 5 days and subsequently analysed for activity, inactivity and object directed behaviour of three animals per group on days 1 and 5. The observed performance of any given behaviour, when at least one other member of the group was also performing that behaviour, was compared with the probability that such concurrence occurred by chance and these results were used to calculate the degree of synchronisation. Gender had no effect on the duration of object use or approach latency. Growers displayed a shorter latency to approach the objects initially compared to sucklers and weaners (mean seconds; 2153 versus 2660 versus 980 for sucklers, weaners and growers, respectively, S.E.D. 542, p = 0.007). Sucklers used the objects to a much lesser extent than either the weaners or growers (mean duration object interaction (s); 129 versus 1253 versus 1412 for sucklers, weaners and growers, respectively, S.E.D. 93, p < 0.001). Overall object use decreased between days 1 and 5 (mean duration object interaction (s); 1326 versus 536 for days 1 and 5, respectively, S.E.D. 76, p < 0.001). All of the age groups synchronised their behaviour to a much greater extent than expected by chance. The sucklers showed a higher degree of synchrony of activity (K; 0.890 versus 0.776 versus 0.682 for sucklers, weaners and growers respectively, S.E.D. 0.013, p < 0.001) and inactivity (K; 0.963 versus 0.905 versus 0.909 respectively, S.E.D. 0.007, p < 0.001), but lower degree of synchrony for object directed behaviour (K; 0.149 versus 0.375 versus 0.370 respectively, S.E.D. 0.025, p < 0.001), than the weaners and growers. The degree of synchronisation of object directed behaviour decreased over the 5-day period, irrespective of age (K; 0.382 versus 0.214 for days 1 and 5 respectively, S.E.D. 0.020, p < 0.001). Significant correlations (r = 0.678–0.879) were found between the degree of synchrony and extent of object interaction only for the sucklers. Since pigs showed behavioural synchronisation, object availability should be considered when providing desirable enrichment in order to avoid excessive competition in larger commercial groups.  相似文献

The activity profile of men's competitive surfing was investigated during an international contest. Forty-two male surfers were filmed during 42 elimination heats. Surfers' activities were classified into 1 of 4 distinct categories (paddling, wave riding, stationary, and miscellaneous). The surfers were paddling and stationary 51% (25-70%) and 42% (23-72%) of the total time, respectively. Wave riding accounted for 4% (2-7%) of the total time, whereas miscellaneous activity accounted for the remaining 2% (0.1-6%) of the total time. The frequency (average number of motions in a heat) of each activity was 26 for paddling (15-37); 17 for stationary (10-26); 5 for wave riding (2-8); and 6 for miscellaneous activity (1-13). The mean duration lengths of the activities were 30.1 seconds (1-286 seconds), 37.7 seconds (1-413 seconds), 11.6 seconds (1-44 seconds), and 5 seconds (1- 31 seconds) for paddling, stationary, wave riding, and miscellaneous activities, respectively. Most of the paddling bouts ( approximately 60%) were performed at time intervals of between 1 and 20 seconds. Approximately 50% (50.9%) of the rest intervals were between 1 and 20 seconds in duration. The results showed that surfing is an intermittent activity characterized by a large variability and random distribution of each parameter analyzed. This information can be used to assist in the development of fitness training programs and physiological testing for competitive surfers.  相似文献

In 1993 we proposed an empirical formula for describing the relaxation modulus of cortical bone based on the results of stress relaxation experiments performed for 1 x 10(5) sec: [E(t) = E0{A exp[ -(t/tau1)beta] + (1 - A) exp(-t/tau2)}, (0 < A, beta <1 and tau1 < tau2) where E0 is the initial value of the relaxation modulus, A is the portion of the first term, tau1 and tau2 are characteristic relaxation times, and beta is a shape factor [Sasaki et al., J. Biomechanics 26 (1993), 1369]. Although the relaxation properties of bone under various external conditions were described well by the above equation, recent experimental results have indicated some limitations in its application. In order to construct an empirical formula for the relaxation modulus of cortical bone that has a high degree of completeness, stress relaxation experiments were performed for 6 x 10(5) seconds. The second term in the equation was determined as an apparently linear portion in a log E(t) vs t plot at t>1 x 10(4) sec. The same plot for experiments performed for 6 x 10(5) seconds revealed that the linear portion corresponding to the second term was in fact a curve with a large radius of curvature. Cervical and uterine electromyograms (EMG) have been recorded from ovariectomized non-pregnant ewes. When the animals were infused with saline, the frequency of EMG events lasting less than 180 seconds was not different from those lasting more than 180 seconds. During infusion of estradiol at 100 micrograms X 24 hours-1 into the maternal jugular, the frequency of events less than 180 seconds increased significantly in the myometrium and in the cervix. Contracture activity (events lasting more than 180 seconds) was not significantly different in the myometrium compared to the cervix before estradiol administration. During estradiol infusion, the contracture activity remained unchanged. During 4-amino-antipyrine (4AA) administration, the contracture activity decreased significantly in the myometrium, while an insignificant change occurred in the cervix. This state was associated with a decrease in the venous PGFM:6-keto F1 alpha plasma ratio. Infusion of PGF2 alpha (.5 micrograms min-1 and 1.0 microgram X min-1 for ten minutes) into the femoral artery resulted in a significant increase in the frequency of events less than 180 seconds in both the myometrium and cervix. For the duration of the ten-minute infusion, the activity was contracture-like. These findings suggest that the cervix may not only be influenced by mechanical properties (stretch) and local paracrine factors but also by various stimulators and inhibitors irrespective of the myometrium.  相似文献

The effect of 1,25(OH)(2)D(3) on the intracellular calcium, (Ca(+2))i, in both cultured human keratinocytes and in cultured human dermal fibroblasts was investigated. When the intracellular calcium (Ca(+2))i in cultured human keratinocytes, grown in a serum-free medium containing 1.8 mM calcium, was measured by the fluorescent calcium-indicator, Furu-2, the (Ca(+2)i increased 154%, 202%, and 409% over the control value after incubation with 1,25(OH)(2)D(3) at 10(-10) m, 10(-8) m, and 10(-6) m, respectively. This response was immediate (15 seconds), specific (no effect with either 25(OH)D(3) at 10(-8) m or vitamin D(3) at 10(-8) m), and occurred with or without EGTA in the medium. In contrast, 1,25(OH)(2)D(3) did not increase the (Ca(2+))i in either cultured human keratinocytes that were grown in low calcium (0.05 mm), serum-free medium or in cultured human dermal fibroblasts that were grown in medium containing 0.05 mm calcium and 1% serum. The effect of 1,25(OH)(2)D(3) on the the turnover of phosphatidylinositol was investigated as a possible cause for the observed increase in (Ca(+2)i. Cultured human keratinocytes that were incubated with (3)H-inositol demonstrated a 50 % +/- 10% increase in the triphosphated, plasma membrane-bound metabolite of phosphatidylinositol, PIP(2), by 15 seconds, followed by a rapid decrease at 30 seconds, then a return toward basal levels by 1 minute. Lysophosphatidylinositol, which results from the sn-2 deacylation of phosphatidylinositol by phospholipase A(2), decreased 20% +/- 8% within 30 seconds, then increased to 200% +/- 10% of the control value by 5 minutes. The accumulation of IP(3) was increased 50% to 100% above the control value within 30 seconds and this increase was substained during the 5-minute incubation period. Stimulation of phosphatidylinositol turnover by 1,25(OH)(2)D(3) was not detected in either cultured human keratinocytes that were grown in serum-free, low calcium medium or in cultured human dermal fibroblasts that were grown in 1% serum.  相似文献

The effects of molar NaCl concentrations on the phase behaviour of the total lipid extracts and binary mixtures of the major phospholipids, namely phosphatidylethanolamine (PE) and phosphatidylglycerol (PG), isolated from the moderately halophilic eubacterium, Vibrio costicola, grown in 1 M and 3 M NaCl containing media have been studied using X-ray diffraction and freeze-fracture electron microscopy. The effect of both the PE/PG ratio and alterations in fatty acid composition were examined by using binary mixtures which mimicked the PE/PG ratio found in the native bacterial membranes. We show that the samples exhibited complex phase behaviour, including the formation of non-bilayer phases, which depend upon the salinity of both the bacterial culture medium and the suspending solution. The total lipid from bacteria cultured in 1 M NaCl-containing medium and dispersed in 1 M NaCl exhibited a mixture of L alpha and hexagonal-II phases at the optimum growth temperature of the organism (i.e., 30 degrees C), whereas the same lipid dispersed in 3 M NaCl showed only a hexagonal-II phase down to a temperature of +3 degrees C. The total lipid extracted from 3 M NaCl cultures showed only lamellar phases over the temperature range studied (+50 degrees C to -50 degrees C), but the phase transition temperatures of the various lamellar phases were generally higher when the lipid was dispersed in 3 M compared with 1 M NaCl. The phase behaviour of the binary mixtures was similar but not identical to that of the corresponding total lipid extracts and it is suggested that the minor lipid components (diphosphatidylglycerol, lysophosphatidylethanolamine and lysophosphatidylglycerol) play a part in determining the phase behaviour of the native membranes. These results show that the PE/PG ratio and fatty acid composition of the individual phospholipids, which are normally regulated by Vibrio costicola in vivo in response to culture medium salinity, are both important in maintaining a stable bilayer structure within the membrane.  相似文献

Two new methods applicable to the staining of fixed and fresh frozen tissue sections are presented herein. In addition certain improvements are suggested for the technic reported by Geschickter, Walker, Hjort and Moulton (1931). In brief the procedures are as follows:
  1. The thionin eosinate method of Geschickter et al (1931). This procedure has been modified as follows:
    1. A mixture of diethylene glycol, 40 parts, ethylene glycol, 40 parts, and grain alcohol, 20 parts is superior to ethylene glycol, 80 parts, and ethyl alcohol, 20 parts, as a solvent for the compound stain in that the staining is intensified.
    2. Ethylene glycol monobutyl ether supplants diethylene glycol monobutyl ether because of its lower viscosity.
    3. Ethyl phthalate replaces butyl phthalate on account of a more satisfactory viscosity.
  2. The methyl green eosinate procedure is the same as the modified thionin eosinate method except for the following variations:
    1. The staining time is increased to one minute.
    2. Decolorization and washing are reduced to about 15 seconds.
  3. The hematoxylin-eosin method. After cutting, the tissue sections are carried thru the following steps:


Unfold in water; transfer to formalin (4 to 40%) for at least 30 seconds; stain in hematoxylin (Harris) for 30 to 60 seconds; wash in water, 5 seconds; decolorize in 0.1% HC1 or saturated aqueous picric acid, 5 seconds; wash in water, S seconds; float in 0.5% ammonia, 5 to 10 seconds; wash in water, 5 seconds; stain in 5% aqueous eosin, 15 seconds1; wash in water, 5 to 10 seconds; dehydrate in a mixture of diethylene glycol, 30 parts, and ethyl alcohol, 70 parts, 5 to 10 seconds; dehydrate in ethylene glycol monobutyl ether, 5 to 10 seconds; clear in ethyl phthalate, 5 to 10 seconds; float on a glass slide, blot with photographic lintless blotter, place a drop of neutral gum damar on the section, and cover with glass cover slip.  相似文献

It is unclear if a constant sprint-to-rest ratio allows full performance recovery between repeated sprints over different distances. This is important for the development of sprint-training programs. Additionally, there is conflicting evidence on whether active recovery enhances sprint performance. Three repeated sprint protocols were used (22 × 15, 13 × 30, and 8 × 50 m), with each having an active and passive recovery. Each trial was conducted with an initial sprint-to-rest ratio of 1:10. Repeated sprints were analyzed by comparing the first sprint to the last sprint. For the 15-m trials, there were no significant main effects for recovery or time and no significant interaction. For the 30-m trials, there was no main effect for recovery, but a main effect for time (F[1,10] = 15.995, p = 0.003; mean difference = 0.20 seconds, 95% confidence interval [CI] = 0.09-0.31 seconds, d = 1.4 [large effect]). There was no interaction of recovery and time in the 30-m trials. For the 50-m trials, there was no main effect for recovery, but a main effect for time (F[1,10] = 34.225, p = 0.0002; mean difference = 0.39 seconds, 95% CI = 0.24-0.55 seconds, d = 1.3 [large effect]). There was no interaction of recovery and time in the 50-m trials. The results demonstrate that a 1:10 sprint-to-rest ratio allows full performance recovery between 15-m sprints, but not between sprints of 30 or 50 m, and that recovery mode did not influence repeated sprint performance.  相似文献
