Separation of different molecular species of hemoglobin from developing chickens by starch gel electrophoresis has revealed the appearance of early embryonic (embryonic), late embryonic (fetal) and adult hemoglobin (Hb) type during development. In 5-day embryos, there are 3 or 4 forms of embryonic Hb type. They begin to decrease in 6-day embryos and cannot be detected in embryos after 10 days of incubation. In 6-day embryos, two forms of adult Hb type appear, and one of them, which is a major form in adults, becomes t o be a major one in 7-day embryos. One or two forms of fetal Hb type first appear in 10-day embryos and are still present in 5-day posthatching chickens.
Ultracentrifugation of carbonmonoxyhemoglobins from embryos at early and at later stages (fetuses), from newly hatched and from adult chickens has shown that they have a single monodisperse peak. Some heterogeneity, however, has been detected after starch gel electrophoresis, probably owing to aggregation or polymerization.
Subunit analysis of embryonic, fetal and adult Hb type by starch gel electrophoresis in formate buffer at pH 1.9 has indicated that embryonic Hb type contains total 5 subunits, C, D, E, F and G; fetal Hb type, total 2, A and H; and adult Hb type, total 3, B, F and H.  相似文献

By applying the double diffusion technique of Ouchterlony and the immunoelectrophoresis, sequence in the appearance of antigens reactive with antisera against HbCOs from 5-day embryos and adult chickens, and the major component of adult HbCO in the course of chicken development has been studied.
Antigenic components reacting to antiserum against HbCO from 5-day embryos have been detected throughout development. Three globin-like components specific to early embryos are detectable in embryos to 2 days of incubation. Two Hb components are detectable in embryos from 3 to 5 days of incubation; one is apparently specific to embryos, while the other seems to be somewhat different from adult Hb. After 6 days of incubation only one adult Hb component is detectable.
Antigenic components reacting to antiserum against HbCO from adults have also been detectable throughout embryonic life. One globin-like component specific to early embryos can be found in embryos to 2 days of incubation. One or two Hb components which are probably specific to embryos can be detected in embryos from 3 to 5 days of incubation. After 6 days of incubation one adult Hb component is detectable, while one globin-like component specific to adults can be found after 15 days of incubation. Further, the other globin-like component is detectable after 3 days of incubation.
Antigenic components reacting to antiserum against the major component of adult HbCO have been detected throughout development; one which seems to be somewhat different from either adult Hb components can be found in embryos from 2 to 5 days of incubation, while the other which is identical with the major component of adult Hb is detectable after 6 days of incubation.  相似文献

Total hemolysates of Synbranchus marmoratus Bloch, 1795, captured in Vitoriana, district of Botucatu, State of S?o Paulo, Brazil, were submitted to agar-starch gel electrophoresis on glass slides using 42 mM-Tris 1.7 mM EDTA-6.1 mM borate buffer, pH 8.8, for the gel and 10 mM borate-1.7 mM NaOH buffer, pH 8.6, for the cuvette. Three distinct hemoglobin bands were detected, with Hb I being of the cathodic type. Cellulose acetate electrophoresis in 800 mM Tris-2.1 mM EDTA buffer, pH 8.9, containing 6 M urea and 2.25 mM beta-mercaptoethanol indicated the presence of four globin chains denoted alpha 1, alpha 2, beta and gamma. It is suggested that the probable tetrameric constitution of the hemoglobin of Synbranchus marmoratus Bloch, 1795 is Hb I (alpha (2)2 gamma 2), Hb II (alpha (1)2 gamma 2) and Hb III (alpha (1)2 beta 2).  相似文献

When adding alpha-ketoglutarate and glutamate the intensity of respiration by the myocardium mitochondria increases gradually from the 15th day of embryonic development till the chicken hatching out. In the presence of succinate respiration of mitochondria of 15- and 20-day embryos and 5-day chickens is almost the same and decreases noticeably in adult chickens. When the above-mentioned substrates are added the value of P/O gradually decreases during the chicken development.  相似文献

It has been suggested that the switch in hemoglobin (Hb) types (from embryonic to adult) during chicken embryonic development is associated with the substitution of one erythroid cell line (“primitive”) for another (“definitive”). For the detection of two Hb types inside single erythroid cells, rabbit antibodies specific for embryonic and adult Hbs were prepared. Rabbit antibody specific for embryonic Hb cross-reacted only with embryonic major Hb components, while antibody specific for adult Hb did solely with adult minor Hb component. The antibodies were conjugated with fluorescein isothiocyanate. The conjugated antibodies were used for the fluorescent staining of blood smears of developing chicken embryos at different ages. Direct fluorescent antibody technique demonstrated that the major components of embryonic Hb and the minor component of adult Hb were not present within the same erythrocyte during chicken ontogenesis. It strongly suggested that embryonic-type Hb and adult-type Hb do not coexist within the same cell.  相似文献

The AMP deaminase activity measured in crude chicken liver extract did not change significantly during development. The livers of 10- and 14-day chick embryos, 1-day, 5-, 10- and 16-week-old chickens and adult hens were examined for the existence of multiple forms of AMP deaminase. Phosphocellulose column chromatography revealed the existence of two peaks of enzyme activity in the liver of 10- and 16-week-old chickens and adult hens. Kinetic studies with the preparations of AMP deaminase revealed sigmoid-shaped substrate-saturation curves at all developmental stages and hyperbolic-shaped saturation curves for the enzyme form appearing in 10-week-old chickens. All AMP deaminases investigated were susceptible to activation by ATP and inhibition by Pi. Kinetic and regulatory properties as well as pH optima of all the enzyme preparations tested indicate that AMP deaminase isolated from the embryos and from 1-day-old chicks was similar to the form I isolated from adult hens and differed significantly from the form II of this enzyme.  相似文献

The chemical composition of chromatin from the livers of 12-, 15- and 19-day-old embryos, of 1-day-old chicks and of adult chickens was analysed. The process of embryonic development is accompanied by an increase in non-histone chromatin proteins and chromatin RNA, as well as in the phosphorus content of chromatin phosphoproteins. The amount of these components decreases in the livers of 1-day-old chicks and adults. Two-dimensional polyacrylamide gel electrophoresis of acid-soluble chromatin proteins showed an increase in the amount of the H1 histone in 19-day-old embryos and adult chickens. Non-histone proteins of embryo liver chromatin showed a high content of the fraction of Mr of about 40 000; this was not the case for adult chickens. The non-histone protein fraction of Mr of about 120 000, characteristic of adult chicken liver proteins, was not found in the livers of 12- and 15-day-old embryos. Non-histone chromatin proteins isolated from the livers of animals of different age exhibited also quantitative differences.  相似文献

(1) 2,3-Diphosphoglyceric acid (2,3-DPG) is present in the erythrocytes (RBC) of the 68-day loggerhead turtle embryo and 44-day green sea turtle embryo at levels of 7.4 and 5.5 μmoles/ml of RBC, representing the major organic phosphate during the latter period of embryonic development. (2) Inositol pentaphosphate (IPP) is absent in the red blood cells of the embryos of both the loggerhead and green sea turtle. (3) Near equimolar amounts of 2,3-DPG and IPP are present in the erythrocytes of the adult loggerhead and green sea turtle. The total concentration of these two organic phosphates is approximately 0.75 μmoles/ml of RBC in the adult of both species. (4) There is a switch from embryonic to adult hemoglobin during development of these two species of turtles; the two embryonic bands have identical electrophoretic mobilities, whereas the two adult bands migrate differently on cellulose acetate at pH 8.6. (5) The whole blood oxygen affinity of the adult loggerhead and green sea turtle is 60.3 and 32.6 Torr, respectively. (6) The stripped adult hemoglobins in these two species of turtles show no change in oxygen affinity upon addition of 2,3-DPG, ATP, or IPP. (7) It therefore appears unlikely that whole blood oxygen affinity is controlled by organic phosphate modulation of hemoglobin function in these species of turtles.  相似文献

啮齿动物种群数量波动的研究,国内外部已有不少报道(夏武平1958,1961,1963;Evans等,1944;Delong,1967)。种群数量随季节的一般性波动以及年间的一般性差异是常见的(孙儒泳等1962;Evans,1949;Krebs等,1973),但极大的波动并不多见。一个种在某一年间种群数量发生的极大波动的现象,现在习惯地称作大发生或大暴发(Pearson,1963;Plomley,1972),我国新疆北部农业带分布的小型啮齿类就有这种大暴发的历史(青海省生物研究所新疆鼠害研究组,1975)。  相似文献

The authors have morphometrically studied the differentiation of the myocardium in dynamic phases of the embryonic and postnatal development in chickens and Syrian Hamsters. Moreover, they investigated the action of the beta-adrenalytic substances Practolol and Trimepranol on ultrastructure of the cardiac muscle in adult animals. The volume of mitochondria in myocardial cells in 6-day old chicken embryos amounts to 5.65% of the total cell volume, in 12-day old embryos 14.35%, in 18-day old embryos 19.60%, in 1-day old chickens 23.24% which is nearly as much as in adult animals. The volume of myofibrils in 6-day old embryos is about 3.2%, in 12-day old embryos about 7.4%, in 18-day old embryos about 16.4% and in 1-day old chickens about 21.2%. The differences between individual groups are statistically significant. The dynamics of differentiation of the myocardium in Syrian Hamsters was studied in 5 phases, namely in 14-day old embryos and in postnatal phases on the 2nd, 5th, 14th and 21st days after birth. Most cells in 14-day old embryos are rather immature. Participation of the volume of mitochondria, myofibrils, equipment of mitochondria with cristae etc. considerably increase in postnatal phases. These findings suggest that the heart of mammals is rather immature at birth and will differentiate mainly in the postnatal developmental phases. Many morphometric findings, as regards the action of beta-adrenalytic drugs on the ultrastructure of the myocardium in adult rabbits, point to the fact that application of these substances will give rise to degenerative alterations in approximately 10% of myocardial cells. Theoretical explantation of these mechanisms is being discussed.  相似文献

Line-restricted hemoglobin synthesis in chick embryonic erythrocytes   总被引:1,自引:0,他引:1  
The presence of embryonic hemoglobin in early definitive erythrocytes was checked by indirect immunofluorescence assay, using specific antibodies raised against embryonic Hb P. As positive control we used anti-Hb A which reacted with the alpha A chain shared by the minor embryonic Hb E and the adult Hb A. The assay was performed using blood smears from embryos between 6 and 15 days of incubation and yolk sac sections from embryos between 4 and 6 days. Hb P was never detected in the definitive line in circulating erythrocytes or in maturing erythroblasts still sequestered in the blood islands of the yolk sac. The expression of the 'specific' embryonic genes is thus restricted to the primitive line (as the 'specific' adult beta gene is restricted to the definitive line), and the hemoglobin switch is the result of the progressive substitution of the primitive line by the definitive one.  相似文献

Myosins from the following sources were purified by diethylaminoethyl-Sephadex chromatography: moytubes grown in vitro for 7–8 days, prepared from pectoralis muscles of 10-day old embryos, and breast and leg muscles from 16-day old embryos. The adenosine triphosphatase activities of these myosins were close to that of adult m. pectoralis myosin. The light chains of the embryonic myosins had the same mobilities in sodium dodecyl sulfate electrophoresis as those in adult pectoralis muscle myosin and were clearly distinguishable from those in myosin from tonic muscle m. latissimus dorsi anterior. The fastest light chain in embryonic muscle myosin—apparent mol wt 16,000—was present in smaller amounts than in adult myosin. The negative staining pattern of paracrystals of embryonic light meromyosin (LMM) was indistinguishable from that of adult fast muscle LMM. The significance of these results for differentiation of various muscle types has been discussed.  相似文献

The erythrocytic stages of Plasmdoium gallinaceum in chicken embryos injected with parasited blood either from a syringe-passaged infection in chickens or from a chicken infected with sporozoites were characterized by abnormal structure. Particularly evident were large, unstained vacuoles within the cytoplasm; these occurred with greatest frequency in schizonts. The presence of myelin bodies within these vacuoles was revealed by transmission electron microscopy; abnormal cytokinesis and aberrant merozoites provided additional evidence of the parasite's inability to develop naturally within the milieu of the embryonic erythrocytes. Fifty-five passages were necessary to restore normal structure of the parasites in embryos, while only 5 passages were required for such restoration in neonate chicks. The probable adaptation of the parasite to the proportions of hemoglobin of the adult chicken may be responsible for the abnormal growth in the immature host.  相似文献

The adult Stump-Tail Monkey (Macaca speciosa) was found to have two major hemoglobin components (Hb 1 and Hb 2) which were separated by carboxymethyl cellulose column chromatography. The tryptic peptides of the alpha and beta chains from the two components were isolated and sequenced. The peptides were aligned based on the homology of their sequences with that of human adult hemoglobin. Only one amino-acid difference was found between the alpha chains from Hb 1 and Hb 2 at the 15th position from the N-terminus. On the other hand, the beta chains from the two hemoglobin components were considered to be identical.  相似文献

1. Adult chicken hemoglobins Hb A and Hb D interact with glutathione disulfide, GSSG. The major hemoglobin, Hb A, forms at least two new components, termed GHb AI and GHb AII, and Hb D forms at least one, GHb DI. 2. At pH 8.0 and 5 degrees C, glutathione disulfide (GSSG) in a molar excess of 50 x took 6 days to complete the reaction, although at pH 8.6 and 41 degrees C only 1 hr was needed, where the hemoglobins Hb A and Hb D were converted to their most mobile forms GHb AII and GHb DI. 3. Slight molar excess (2.7 GSSG/Hb, pH 7.4, 41 degrees C), reacting for 1 hr, showed extensive formation of GHb AI and some GHb AII. 4. Electrophoretic patterns, from the reaction products of 54 GSSG/Hb excess at different times, showed a marked pH dependence. 5. Titration with pCMB (p-chloromercuribezoic acid) of DTE (dithioerythrytol)-reduced samples showed 8.0 +/- 0.4 (N = 5) -SH (sulfhydryl) per tetramer. In hemolysates not reacted with DTE, 6.0 +/- 0.4 (N = 3) -SH were detected. 6. DTE-reduced and GSSG-reacted hemoglobins showed 4.6 +/- 0.5 (N = 7) -SH and 1.5 +/- 0.4 (N = 6) -SH, respectively, as titrated by DTNB, pH 8.0. DTE-reduced hemoglobins showed four fast-reacting -SH groups, no longer present in GSSG-reacted hemoglobins. 7. Lens capsule collagen synthesis by epithelial and fiber cells was examined by immunoprecipitation and collagenase digestion in embryonic and posthatch chicken eye lens. Epithelial cells and lens fibers in the process of terminal differentiation produce alpha 1 and alpha 2 type IV collagen chains. At 6 days of embryonic development in addition to the alpha 1 (IV) and alpha 2 (IV) collagen chains, lens cells produce high molecular weight collagenase-sensitive proteins not immunologically related to type IV collagen. Lens capsule collagen components have been identified in central and outer fibers isolated from 18-day embryos and from 10-day posthatch chicken eyes. At these stages, fibers which have an increasing number of picnotic nuclei still show collagen synthesis due to long-lived mRNA. Analysis of collagen synthesis by lens cells incubated with actinomycin D suggests that stabilization of collagen mRNA occurs in lens fiber cells and to a lesser extent in epithelial cells as early as 6 days of embryonic development.  相似文献

A hitherto unknown inherited hemoglobin variant, Hb Tacoma, was discovered in three healthy members of a family of European extraction.Hybridization experiments with canine hemoglobin indicated a structural abnormality in the -chain. The variant was therefore designated as Hb 2 2 Tacoma. Separation of Hb Tacoma from Hb A could only be clearly achieved by starch grain electrophoresis in TEB buffer at pH 8.6–9.0, where Hb Tacoma moved more rapidly towards the anode than Hb A; gradient column chromatography on DEAE cellulose and CM-Sephadex achieved partial separation. The proportion of abnormal hemoglobin in the heterozygote amounted to 43 per cent of the total hemoglobin. Hb Tacoma was less heat resistant and became more rapidly denatured in 8 M urea solution than Hb A, Hb C, or Hb S.On thin-layer starch gel electrophoresis in TCB buffer at pH 8.6 Hb Tacoma was associated with an electrophoretically slow, benzidine-positive component, Hb Tacoma-slow. The exact biochemical nature of this minor component could not be determined, although a polymeric product of Hb Tacoma is suspected.Heterozygosity for Hb Tacoma is associated with a raised Hb A2 level without evidence of -thalassemia.A preliminary report of Hb Tacoma was presented at the Annual Meeting of the American Society of Human Genetics, Boulder, Colorado, August, 1964, and at the Seventh Annual Meeting of the American Society of Hematology, November, 1964, Seattle, Washington (Blood, 24, 848 (1964)).  相似文献

Vertical starch-gel electrophoresis at pH 8.6 revealed extensive hemoglobin multiplicity with several distinct cathodal and anodal hemoglobin components. Anodal hemoglobin components are present throughout the life cycle of the king salmon. Additional cathodal components are found in the adult fish. Cathodal hemoglobin components exhibited a higher oxygen affinity (P50 = 10.2 mm at 13 degrees C, pH 7.3) than the anodal hemoglobin components (P50 = 21.8 mmHg at 13 degrees C). Oxygen binding of the anodal hemoglobins are sensitive to pH, temperature, organic phosphates (ATP and GTP), as well as, ionic strength; binding of oxygen to the cathodal hemoglobins is independent of pH and not affected by organic phosphates. Anodal hemoglobin components are less resistant to thermal denaturation over the pH 6.0 to 8.0 range. Isothermal urea denaturation of separated anodal and cathodal hemoglobin fractions of the king salmon indicate inherent differences in the stabilization energies of these hemoglobins. Autoxidation of these hemoglobins occurs around pH 7.0 and below, as well as, in the presence of increasing Cl- concentrations.  相似文献
