An 1H NMR (nuclear magnetic resonance) spectroscopic structural analysis of Cd2+ complexes formed with the pentapeptide phytochelatin, (NH3)+−(ψ-Glu-Cys)2−Gly−COO−(PC2), at a pH of 7.5 showed that the two thiol groups of the Cys residues and either the carbonyl or amide group of the peptide bond between Glu1 and Cys1 act as possible donor groups in the complexes at Cd2+/PC2 ratios up to 0.4. As the ratio increases, the carboxylate group of Glu2 and either the carbonyl or amide group of the peptide bond between Cys1 and Glu2 comes to serve as a donor group. The manner in which Cd2+ forms complexes with PC2 is distinctly different from Zn2+ and might account for the role of phytochelatin in Cd2+ detoxification. Electron absorption spectrometry demonstrated that the Cd2+ complexes are coordinated in a tetrahedral fashion by four thiol groups and that several sulfur atoms might bridge Cd2+ ions, resulting in the formation of polynuclear complexes. This contrasts with Zn2+ complex formation, which consists exclusively of a 1:1 complex.  相似文献

Previous work in our laboratory led to the isolation of a cadmium (Cd)-resistant variant (Cdr2C10) of the line CHO Chinese Hamster cell having a 10-fold greater resistance to the cytotoxic action of Cd2+ compared with the CHO cell. This resistance was attributed to an increased capacity of the Cd2+-resistant Cdr2C10 subline to induce synthesis of the Cd2+- and Zn2+-binding protein(s), metallothionein(s) (MT). Evidence that Cd2+ behaves as an analog of the essential trace metal, Zn2+, especially as an inducer of MT synthesis, suggested that the Cdr and CHO cell types could be employed to investigate cellular Zn2+ metabolism. In the present study, measurements were made to compare CHO and Cdr cell types for (a) growth as a function of the level of ZnCl2 added to the culture medium, (b) uptake and subcellular distribution of Zn2+, and (c) capacity to induce MT synthesis. The results of these measurements indicated that (a) both CHO and Cdr cell types grew normally (T d≊16–18 h) during exposures to Zn2+ at levels up to 100 μM added to the growth medium, but displayed abrupt growth inhibition at higher Zn2+ levels, (b) Cdr cells incorporate fourfold more Zn2+ during a 24-h exposure to the maximal subtoxic level of Zn2+ and (c) the CHO cell lacks the capacity to induce MT synethesis while the Cdr cell is proficient in this response during exposure to the maximal subtoxic Zn2+ level. These findings suggest that (a) the CHO and Cdr cell systems will be useful in further studies of cellular Zn2+ metabolism, especially in comparisons of Zn2+ metabolism in the presence and absence of induction of the Zn2+-sequestering MT and (b) a relationship exists between cellular capacity to induce MT synthesis and capacity for cellular Zn2+ uptake.  相似文献

Through subcultivations of Thiobacillus thiooxidans WU-79A in autotrophic media in which the concentrations of Cd2+ and Zn2+ were increased successively, Cd2+-resistant (CDR) and Zn2+-resistant strains (ZNR) were obtained. The growth of WU-79A was inhibited by the addition of 25 mM Cd2+ as well as Zn2+. However, CDR and ZNR could grow without any lag phase in media containing 200 mM Cd2+ and 250 mM Zn2+, respectively. CDR and ZNR were able to grow even in media containing up to 400 mM Cd2+ and 600 mM Zn2+, respectively, although they exhibited lag phases. CDR could grow in medium containing up to 250 mM Zn2+, as could ZNR in medium containing up to 200 mM Cd2+. Cd2+-binding and Zn2+-binding proteins were isolated from CDR and ZNR, respectively, by gel filtration and ion exchange chromatography. The molecular weights of both proteins were estimated to be approximately 13,000 by gel filtration. The fact that there was no strong absorption at 280 nm of the proteins suggested that they had few aromatic amino acids. Broad absorption bands which are typical of mercaptide (metal thiolate) complexes were detected. The properties of the proteins were spectrophotometrically similar to those of metallothionein.  相似文献

Bis(ZnII-cyclen)-azobenzene derivative, which has two ZnII-macrocyclic tetraamine complexes connected through azobenzene spacer, has been synthesized as a cross-linking agent for double stranded DNA in aqueous solution. The ZnII-cyclen derivative selectively binds to A-T base pairs producing complexes between the ZnII-cyclen moiety and the imide-deprotonated thymine with breaking A-T base pairs. The azobenzene spacer undergoes cis/trans photoisomerization in the complex between the ZnII-cyclen derivative and the DNA duplex. The conformation of the DNA remarkably changed by photoisomerization of the azobenzene linker, when the ZnII-cyclen derivative binds to the DNA duplex with an interstrand cross-linking manner.  相似文献

The magnetic circular dichroism (MCD) spectra of metal complexes of tetrakis(thiadiazole)porphyrazines ([TTDPzM] with M = 2HI, ZnII, MgII(H2O), and CdII) have been recorded in dimethyl formamide solution. Together with the UV–Vis spectra, the MCD spectra provide useful information about the structure and electronic properties of the complexes. The experimental UV–Vis and MCD spectra compare pretty well with DFT calculations of two sorts, based either on the sum-over-states (SOS) approach or on the complex polarization propagator approach. They further corroborate the findings and interpretation of MCD spectra of porphyrazines based on the model of Michl for peripheral molecular orbitals. Magnetic circularly polarized luminescence (MCPL) spectra, quite uncommon in the literature, have been recorded for [TTDPzM] (M = 2HI, ZnII, MgII(H2O)).  相似文献

Metallothioneins are small cysteine-rich proteins believed to play a role, among others, in the homeostasis of essential metal ions such as ZnII and CuI. Recently, we could show that wheat Ec-1 is coordinating its six ZnII ions in form of metal-thiolate clusters analogously to the vertebrate metallothioneins. Specifically, two ZnII ions are bound in the N-terminal and four in the C-terminal domain. In the following, we will present evidence for the relative independence of the two domains from each other with respect to their metal ion binding abilities, and uncover three intriguing peculiarities of the protein. Firstly, one ZnII ion of the N-terminal domain is relative resistant to complete replacement with CdII indicating the presence of a ZnII-binding site with increased stability. Secondly, the C-terminal domain is able to coordinate an additional fifth metal ion, though with reduced affinity, which went undetected so far. Finally, reconstitution of apoEc-1 with an excess of ZnII shows a certain amount of sub-stoichiometrically metal-loaded species. The possible relevance of these finding for the proposed biological functions of wheat Ec-1 will be discussed. In addition, extended X-ray absorption fine structure (EXAFS) measurements on both, the full-length and the truncated protein, provide final evidence for His participation in metal ion binding.  相似文献

Metallothioneins (MTs) are ubiquitous, low molecular mass and cysteine-rich proteins that play important roles in maintaining intracellular metal homeostasis, eliminating metal toxification and protecting the cells against oxidative damages. MTs are able to bind metal ions through the thiol groups of their cysteine residues. Plants have several MT isoforms which are classified into four types based on the arrangement of cysteine residues. In the present study, a rice (Oryza sativa) gene encoding type 1 MT isoform, OsMTI-1b, was inserted in vector pET41a and overexpressed in Escherichia coli as carboxy-terminal extensions of glutathione-S-transferase (GST). The recombinant protein GST-OsMTI-1b was purified using affinity chromatography and its ability to bind with Ni2+, Cd2+, Zn2+ and Cu2+ ions was analyzed. The results demonstrated that this isoform has ability to bind Ni2+, Cd2+ and Zn2+ ions in vitro, whereas it has no substantial ability to bind Cu2+ ions. The uptake of cadmium and its effect on the growth of potato shoot tips grownin vitro were followed in dependence on cadmium concentration in nutrient medium. Concentration of 10 −6 M Cd 2+ did not substantially affect potato plantlet growth dynamics; but the concentration of 10−3 M Cd2+ showed a strong growth inhibitory effect accompanied with increased cadmium accumulation in both root and shoot tissues.  相似文献

The effects of some metal ions on amidolytic and fibrinogenolytic activities of highly purified human plasmin were investigated in vitro. In the presence of Zn2+, Cu2+, Cd2+, and Au+ in the incubation mixture at the concentrations of 1×10−5−1×10−3 M, the anidolytic plasmin activity was strongly inhibited, whereas Ca2+ and Mg2+ at the same concentrations were not effective. The analysis of the kinetic study has shown that Zn2+ or Cu2+ acts as mixed-type inhibitors of plasmin activity. The inhibition of amidolytic plasmin activity by Zn2+ and Cu2+ was reduced in the presence of EDTA, histidine, or albumin. Incubation of plasmin with Zn2+ or Cu2+ (at the concentration of 5×10−4 M) resulted in complete loss of its proteolytic action on fibrinogen, whereas Cd2+ and Au+ under the same conditions only partially inhibited this process.  相似文献

ZnuA is the periplasmic Zn2+-binding protein associated with the high-affinity ATP-binding cassette ZnuABC transporter from Escherichia coli. Although several structures of ZnuA and its homologs have been determined, details regarding metal ion stoichiometry, affinity, and specificity as well as the mechanism of metal uptake and transfer remain unclear. The crystal structures of E. coli ZnuA (Eco-ZnuA) in the apo, Zn2+-bound, and Co2+-bound forms have been determined. ZnZnuA binds at least two metal ions. The first, observed previously in other structures, is coordinated tetrahedrally by Glu59, His60, His143, and His207. Replacement of Zn2+ with Co2+ results in almost identical coordination geometry at this site. The second metal binding site involves His224 and several yet to be identified residues from the His-rich loop that is unique to Zn2+ periplasmic metal binding receptors. Electron paramagnetic resonance and X-ray absorption spectroscopic data on CoZnuA provide additional insight into possible residues involved in this second site. The second site is also detected by metal analysis and circular dichroism (CD) titrations. Eco-ZnuA binds Zn2+ (estimated K d < 20 nM), Co2+, Ni2+, Cu2+, Cu+, and Cd2+, but not Mn2+. Finally, conformational changes upon metal binding observed in the crystal structures together with fluorescence and CD data indicate that only Zn2+ substantially stabilizes ZnuA and might facilitate recognition of ZnuB and subsequent metal transfer. Heavy metal pollutants such as Cd, Hg, Pb, As, and Se are considered as both a global problem and a growing threat to the humanity. Being strongly poisonous to the metal-sensitive enzymes and leading to the growth inhibition and death of organisms, these metals have a toxic impact on the plants and animals. Inducing the metal-binding cysteine-rich peptides such as metallothioneins, phytochelatins, and defensins, higher organisms like plants and animals usually react to the heavy metal stress. In this study, a recombinant defensin protein was expressed in bean and its ability in the cadmium absorption was determined. Experimental studies revealed that this protein was able to absorb cadmium ions in reduced form more than oxide one. Molecular dynamics simulations were carried out in order to evaluation of experimental studies, using a model of Cd2+ or Na+ and Cl ions enclosed in a fully hydrated simulation box with the recombinant defensin. The theoretical results also suggested that the reduced recombinant defensin was more powerful in the absorption of Cd2+ than its oxide form. The present study is the first report of Cd2+ absorption potential of this new reduced recombinant defensin. The results unraveled that this recombinant defensin can be adopted as a molecular switch in the cadmium pollution of the environment and also the important role of sulfur groups in the absorption of cadmium ions.  相似文献

Three new supramolecular complexes, [Cu(L1)H2O]n (1), [Zn(L2)(H2O)2]n (2), and [Cd(L2)(H2O)2]n (3), have been synthesized and characterized by FT-IR spectra, fluorescence spectra, and thermal analyses. And the structures of complexes 1-3 have been elucidated by X-ray analyses. Complex 1 is square pyramidal geometry with an unusually long bond (2.262 Å) from penta-coodinated CuII center to the oxygen atom of the apical coordinated water molecule. Molecules are linked by hydrogen bonding between the coordinated water and the phenolic oxygen atoms of adjacent molecules, thus formed a self-assembling continual zigzag chain supramolecular structure. The crystal structure of complex 2 (or 3) has indicated that the complex consists of one ZnII (or CdII) atom, one L2− unit and two coordinated water molecules, the coordination number of the ZnII (or CdII) atom is six, and formed an infinite metal-water chain supramolecular structure by intermolecular hydrogen bonds and π-π stacking of neighboring benzene rings. Meanwhile, the thermal and photophysical properties of the resulted complexes have also been discussed.  相似文献

A metallothionein-like gene, ThMT3, encoding a type 3 metallothionein, was isolated from a Tamarix hispida leaf cDNA library. Expression analysis revealed that mRNA of ThMT3 was upregulated by high salinity as well as by heavy metal ions, and that ThMT3 was predominantly expressed in the leaf. Transgenic yeast (Saccharomyces cerevisiae) expressing ThMT3 showed increased tolerance to Cd2+, Zn2+, Cu2+, and NaCl stress. Transgenic yeast also accumulated more Cd2+, Zn2+, and NaCl, but not Cu2+. Analysis of the expression of four genes (GLR1, GTT2, GSH1, and YCF1) that aid in transporting heavy metal (Cd2+) from the cytoplasm to the vacuole demonstrated that none of these genes were induced under Cd2+, Zn2+, Cu2+, and NaCl stress in ThMT3-transgenic yeast. H2O2 levels in transgenic yeast under such stress conditions were less than half those in control yeast under the same conditions. Three antioxidant genes (SOD1, CAT1, and GPX1) were specifically expressed under Cd2+, Zn2+, Cu2+, and NaCl stress in the transgenic yeast. Cd2+, Zn2+, and Cu2+ increased the expression levels of SOD1, CAT1, and GPX1, respectively, whereas NaCl induced the expression of SOD1 and GPX1.  相似文献

The influence of cadmium, zinc and lead on fungal emulsifier synthesis and on the growth of filamentous fungus Curvularia lunata has been studied. Tolerance to heavy metals established for C. lunata was additionally compared with the sensitivity exhibited by strains of Curvularia tuberculata and Paecilomyces marquandii—fungi which do not secrete compounds of emulsifying activity. Although C. lunata, as the only one out of all studied fungi, exhibited the lowest tolerance to heavy metals when grown on a solid medium (in conditions preventing emulsifier synthesis), it manifested the highest tolerance in liquid culture - in conditions allowing exopolymer production. Cadmium, zinc and lead presented in liquid medium up to a concentration of 15 mM had no negative effect on C. lunata growth and stimulated emulsifier synthesis. In the presence of 15 mM of heavy metals, both the emulsifier and 24-h-old growing mycelium exhibited maximum sorption capacities, which were determined as 18.2 ± 2.67, 156.1 ± 10.32 mg g−1 for Cd2+, 22.2 ± 3.40, 95.2 ± 14.21 mg g−1 for Zn2+ and 51.1 ± 1.85, 230.0 ± 28.47 mg g−1 for Pb2+ respectively. The results obtained by us in this work indicate that the emulsifier acts as a protective compound increasing the ability of C. lunata to survive in heavy metal polluted environment. Enhancement of exopolymer synthesis in the presence of Cd2+, Zn2+ and Pb2+ may also suggest, at least to some extent, a metal-specific nature of emulsifier production in C. lunata. Due to accumulation capability and tolerance to heavy metals, C. lunata mycelium surrounded by the emulsifier could be applied for toxic metal removal.  相似文献

Acutolysin D, isolated from the venom of Agkistrodon acutus, possesses marked haemorrhagic and proteolytic activities. The molecular weight and the absorption coefficients (A 1% 280) of acutolyisn D have been determined to be 47,850 ± 8 amu and 9.3 by mass spectrometer and UV spectrum, respectively. The effects of metal ions on the conformation and activity of acutolysin D have been studied by following fluorescence, circular dichroism and biological activity measurements. Acutolysin D contains two Ca2+-binding sites and two Zn2+-binding sites determined by atomic absorption spectrophotometer. Zn2+ is essential for the enzyme activities of acutolysin D, however, the presence of 1 mM Zn2+ significantly decreases its caseinolytic activity and intrinsic fluorescence intensity at pH 9.0 due to Zn(OH)2 precipitate formation. Ca2+ is important for the structural integrity of acutolysin D, and the presence of 1 mM Ca2+ markedly enhances its caseinolytic activity. Interestingly, the caseinolytic activity which is inhibited partly by Cu2+, Co2+, Mn2+ or Tb3+ and inhibited completely by Cd2+, is enhanced by Mg2+. The fluorescence intensity of the protein decreases in the presence of Cu2+, Co2+, Cd2+ or Mn2+, but neither for Ca2+, Mg2+ nor for Tb3+. Zn2+, Ca2+, Mg2+, Cu2+, Mn2+, Co2+ and Tb3+ have slight effects on its secondary structure contents. In addition, Cd2+ causes a marked increase of antiparallel β-sheet content from 45.5% to 60.2%.  相似文献

Metallothioneins (MTs) are ubiquitous cysteine-rich proteins with a high affinity for divalent metal ions such as ZnII, CuI, and CdII that are involved in metal ion homeostasis and detoxification, as well as protection against reactive oxygen species. Here we show the NMR solution structure of the βE-domain of the early cysteine-labeled protein (Ec-1) from wheat (βE-Ec-1), which represents the first three-dimensional structure of a plant MT. The βE-domain comprises the 51 C-terminal residues of Ec-1 and exhibits a distinctive unprecedented structure with two separate metal-binding centers, a mononuclear ZnII binding site constituted by two cysteine and two highly conserved histidine residues as found in certain zinc-finger motifs, and a cluster formed by three ZnII ions coordinated by nine Cys residues that resembles the cluster in the β-domain of vertebrate MTs. Cys-metal ion connectivities were determined by exhaustive structure calculations for all 7560 possible configurations of the three-metal cluster. Backbone dynamics investigated by 15N relaxation experiments support the results of the structure determination in that βE-Ec-1 is a rigidly folded polypeptide. To further investigate the influence of metal ion binding on the stability of the structure, we replaced ZnII with CdII ions and examined the effects of metal ion release on incubation with a metal ion chelator.  相似文献

Suspension cultures of Lycopersicon peruvianum were selected for resistance to cadmium by stepwise exposure to increasing concentrations of cadmium sulfate. Resistant cells grow in 1500 micromolar Cd++. This resistance was retained for thirty generations without selection. Both resistant and parental sensitive cultures take up Cd++ at similar rates and to the same final levels. Exposure of sensitive or resistant cultures to Cd++, Cu++, or Zn++ leads to the intracellular accumulation of a low molecular weight, cysteine-rich, cadmium-binding protein. This report discusses the identification of a Zn2+- and Cd2+-binding protein of Xenopus laevis that is abundant in vitellogenic oocytes and in embryos from fertilization to stage 46. Oocyte or embryo homogenates were fractionated by SDS-PAGE, blotted onto nitrocellulose, and probed with 65Zn2+ or 109Cd2+. The resulting autoradiograms showed binding of both radio-nuclides to a protein, designated pCdZn. Freon extraction of oocyte and embryo homogenates showed pCdZn to be a yolk protein. When pCdZn was isolated from oocyte homogenates by ammonium sulfate precipitation, delipidation, and chromatography, it co-purified with lipovitellin 1. The amino acid composition of pCdZn closely resembled the reported composition of lipovitellin 1 and the molecular weight of purified pCdZn (~115 kD) corresponded to reported values for lipovitellin 1 (111–121 kD). Amino acid sequence analyses of five peptides derived from pCdZn yielded 94% identity to the reported sequence of lipovitellin 1, deduced from the DNA sequence of the Xenopus vitellogenin A2 precursor gene. Based on these findings, pCdZn was identified as lipovitellin 1. This study suggests that lipovitellin 1 is the major storage protein for zinc in mature oocytes and developing embryos of Xenopus laevis. © 1995 wiley-Liss, Inc.  相似文献

The purpose of the present study was to investigate the in vitro and the in vivo effects of cadmium, zinc, mercury and lead on -aminolevulinic acid dehydratase (ALA-D) activity from radish leaves. The in vivo effect of these metals on growth, DNA and protein content was also evaluated. The results demonstrated that among the elements studied Cd2+ presented the highest toxicity for radish. 50% inhibition of ALA-D activity (IC50) in vitro was at 0.39, 2.39, 2.29, and 1.38 mM Cd2+, Zn2+, Hg2+ and Pb2+, respectively. After in vivo exposure Cd2+, Zn2+, Hg2+ and Pb2+ inhibited ALA-D by about 40, 26, 34 and 15%, respectively. Growth was inhibited by about 40, 10, 25, and 5% by Cd2+, Zn2+, Hg2+, and Pb2+, respectively. DNA content was reduced about 35, 30, 20, and 10% for Cd2+, Zn2+, Hg2+, and Pb2+, respectively. The metal concentration in radish leaves exposed to Cd2+, Zn2+, Hg2+, and Pb2+ was 18, 13, 6, and 7 mol g–1, respectively. The marked ability of radish to accumulate Cd2+ and Zn2+ raises the possibility of using this vegetable as a biomonitor of environmental contamination by these metals.  相似文献
