The cytology of phytophthora infestans

Summary Phytophthora infestans has three kinds of somatic nuclei: an oval shaped nucleus (approx. 3.1×2.7 ) which stains diffusely except for a crescent shaped Feulgen positive cap which stains intensely; a granular nucleus whose contents are organized into a fairly constant number of stained bodies, and, a deeply staining condensed nucleus. The capped nucleus is thought to be metabolic or resting and the granular nucleus is thought to be dividing as it is most commonly found in hyphal tips. Attenuated forms of all three kinds of nuclei are found.Nuclear division is mitotic and intranuclear. Eight—ten chromosomes are seen at metaphase.Sporangia have a mean of 6.3 nuclei which is constant for age and strain of culture. Sporangia become multinucleate as a result of nuclear migration and not by division in the developing sporangium. Zoospores are usually uninucleate.The nuclear cap is persistent throughout nuclear division when it also divides. It is associated with flagella production and nuclear migration and has some of the properties of a blepharoplast.     

Das organische Gerüstwerk der Kalkschale des Schwanen-Eies

Zusammenfassung Kegel und Säulen der Schwanen-Eischale hinterlassen am Querschliff nach Entkalkung mit EDTA organisches (mucoproteides) Material als ein zusammenhängendes Gerüst, das sich mit Thionin metachromatisch färbt; ohne Demineralisierung oder wenigstens Anätzung bleibt Thionin an Schliffen und Bruchkanten der Schale wirkungslos. Das Lichtmikroskop zeigt an Schliffen nichts von dem organischen Material, es wurde während des Kristallwachstums fein zerteilt in Gitterlücken des Schalencalcits eingeschlossen. Es findet sich am stärksten angehäuft an den äueren und inneren Oberflächen der Kristall-individuen. In den Kegeln ist das Gerüst radial ausgebildet als die Loculi der Keile, und konzentrisch geschichtet, entsprechend den Lagen der Globularinklusionen, um deren jede herum Verdichtung der organischen Substanz statthat. In den inneren Säulen folgt das organische Gerüst dem Rhombenmuster; die äueren Säulen sind arm an organischer Substanz, hier verbleibt nach der Entkalkung eine dünne laterale Oberflächenschicht.

Summary The cones and columns of the swans egg shell leave behind after decalcification with EDTA an organic (mucoproteid) material in form of a continuous frame work stainable metachromatically with thionine. Without demineralisation or at least etching, thionine proves ineffectual in ground sections or breaking edges of the shell. In ground sections the light microscope demonstrates nothing of the organic material: it was inclosed during the crystal growth in submicroscopical lattice gaps of the calcite individuals. The organic material is chiefly accumulated in the outer and inner surfaces of the crystals. In the cones the organic frame work is developed radially as the loculi of the wedges and concentrically layered corresponding with the globular inclusions, concentrated in the circumference of each. In the inner columns the organic material follows to the rhomb pattern. The outer columns after decalcification only leave behind a thin lateral organic sheath.

The peritrophic membranes, which is several groups of animals are produced by midgut epithelia, have been investigated most thoroughly in insects. The following results concern Crustacea: In the peritrophic membranes chitin containing microfibrils are embedded in the ground substance consisting of proteins and mucopolysaccharides. In addition to a felt-like texture (Streuungstextur), microfibrils are arranged in textures of higher order (orthogonal texture — Gittertexture; hexagonal texture — Wabentexture).

Zur Frage des Zeipunktes einer Pr?gung auf die Heimatregion beim Halsbandschn?pper(Ficedula albicollis)

Zusammenfassung Junge Halsbandschnäpper wurden handaufgezogen, flogen im Flugkäfig aus und wurden dort selbständig. Darauf wurden sie 90 km nach Süden verfrachtet und in einem von dieser Art unbewohnten Gebiet freigelassen. Im nächsten Frühjahr siedelten sich mindestens 9 dort an, was 19% Rückkehrern entspricht, wenn die Hälfte der Vögel waren. kehrten in geringerer Zahl zurück und wurden nicht restlos erfaßt.Eine weitere Gruppe wurde erst vor Ende der Jugendmauser verfrachtet. Auch davon kehrten 18-19% der zurück. Ein Zeitraum von rund 2 Wochen vor dem Wegzug reichte also zur Prägung auf ein Gebiet als Heimat aus.Von einer dritten Gruppe von insgesamt 68 Schnäppern (= ca. 34 ), die erst nach Ende der Jugendmauser zur Wegzugzeit aufgelassen wurde, konnte später keiner nachgewiesen werden, auch nicht am Aufzuchtsort. Letzteres könnte an der Ungunst der örtlichen Verhältnisse liegen.Mit Unterstützung der Deutschen Forschungsgemeinschaft.     

Role of plastidial acyl-acyl carrier protein: Glycerol 3-phosphate acyltransferase and acyl-acyl carrier protein hydrolase in channelling the acyl flux through the prokaryotic and eukaryotic pathway

Monoclonal antibodies raised against extracts of the rachis abscission zone of Sambucus nigra L. were selected for high reactivity towards abscission-zone proteins. One antibody (YZ1/2.23) has been shown to cross-react, by both indirect and competition enzyme-linked immunosorbent assay and by Western blotting, with a number of plant enzymes including horseradish peroxidase, rice -glucosidase, almond -glucosidase and the lectins from Phaseolus vulgaris and Erythrina cristagalli.The major N-linked oligosaccharide isolated from horseradish peroxidase has the sequence Man 3(Man6)(Xyl2)Man4GlcNAc4(Fuc3) GlcNAc. This oligosaccharide was found to be a potent inhibitor of the binding of YZ1/2.23 to the intact glycoprotein. Die Aktivit?tsperiodik bei V?geln mit und ohne dunklem Schlafkasten

Zusammenfassung In 2 Versuchsserien wurden Kohlmeisen(Parus major) und japanische Möwchen(Lonchura striata var.domestica) einzeln und schallisoliert gehalten. In der ersten Versuchsserie, in der alle Vögel einen dunklen Schlafkasten hatten, wurde der Einfluß der Beleuchtungsstärke auf die Periode () der Hüpfaktivität und auf das Verhältnis von Aktivitätszeit zu Ruhezeit ( : -Verhältnis) untersucht. Sowhol Kohlmeisen als auch japanische Möwchen folgen der Regel, daß mit wachsender Beleuchtungsstärke die Periode kürzer und das : -Verhältnis größer wird.In der 2. Serie wurde der Einfluß Ruhe im dunklen Schlafkasten auf die Periodenlänge und auf das : -Verhältnis untersucht. Es wurden die Messungen aus Bedingung 1 (der Vogel hat einen dunklen Schlafkasten zur Verfügung) mit den Messungen aus Bedingung 2 (der Vogel hat keinen oder einen hellen Schlafkasten zur Verfügung) verglichen. Das Ergebnis bei Kohlmeisen entspricht den Befunden bei konstantem Licht verschiedener Intensität. Unter Bedingung 1 ist länger und das : -Verhältnis kleiner als in Bedingung 2. Das Ausmaß der Änderung von nach Fortnahme des dunklen Kastens ist unabhängig von der Periodenlänge in Bedingung 1. Das Ausmaß der änderung von : ist unabhängig von a : in Bedingung 1, jedoch schwach negativ korreliert mit der Periodenlänge in Bedingung 1.Bei japanischen Möwchen entsprechen die Ergebnisse dieser Versuchsserie nicht der Regel für tagaktive Vögel. Mit Benützen des dunklen Schlafkastens ist kürzer als ohne den Schlafkasten. Ohne den Schlafkasten ist etwa 24 Std. Das : -Verhältnis ist in Bedingung 1 unter bestimmten Voraussetzungen kleiner als in Bedingung 2. Das Ausmaß der Änderung von nach Fortnahme des Kastens ist mit der dazugehörigen Periode in Bedingung 1 hochsignifikant korreliert (Regressionskoeffizient b=-1.01, Korrelationskoeffizient r=0.89). Ebenfalls ist das Ausmaß der Änderung von : nach Fortnahme des Kastens mit : aus Bedingung 1 korreliert; es scheint, als würde ein bevorzugtes : -Verhältnis von etwa 2.0 eingeregelt.Die Ergebnisse werden im Hinblick auf 4 Punkte diskutiert: 1) Das circadiane System arbeitet innerhalb eines engen Bereiches von - und : -Werten optimal. 2) Der Optimalbereich wird bevorzugt unter ungünstigen Bedingungen angestrebt. 3) Der Entzug des dunklen Schlafkastens belastet japanische Möwchen mehr als Kohlmeisen. 4) Bei japanischen Möwchen wird in Bedingung 1 durch fortplfanzungsphysiologischen Einfluß verkürzt.

Circadian activity rhythms of birds with and without a dark nest box

Summary Perch-hopping activity of Great tits(Parus major) and Bengales finches(Lonchura striata domestica), housed individually in soundproof boxes, was studied in two series of experiments. In the first series all birds had access to a dark nest box, in which they retired during their subjective night. In this experiment the effect of light intensity on the freerunning circadian activity rhythm was investigated. Both Great tits and Bengalese finches obey the circadian rule by responding to an increase in light intensity with shortening the circadian period () and with an increase of the ratio of activity time and rest time ( : ).In the second series of experiments the influence of sleeping in the dark nest box on both circadian period and : -ratio was studied. The results of two experimental conditions — without and with access to a dark nest box — were compared. In the Great tits, the results are in agreement with the effect of light intensity: when a dark nestbox is available, is longer and the : -ratio is smaller than in the absence of a nest box. The magnitude of the change in free-running period after removal of the nest box is independent of the original value of ; the amount of change : -ratio is likewise independent of the original : -ratio, but is weakly correlated to the original .InLonchura striata var. domestica, removal of the dark nest box leads to a lenghtening of the free-running period to about 24 hours; the : -ratio is smaller in the presence of a dark nestbox, if certain other conditions are fulfilled. The magnitude of the change in after removal of the nest box is highly correlated to the original free-running period (r=-0.89) in such a way that, without nest box, the period approaches a value of 24 hours. Also, the amount of change in : -ratio due to nest box removal is negatively correlated to the original : -ratio. A probably preferred : -ratio of 2.0 is adopted.These results are discussed in the view of 4 points: 1) The circadian system operates at its optimum within a narrow range of - and : -values. 2) This optimal range is especially adopted when conditions become adverse. 3) Removal of the dark nest box results in a more stressful situation for Bengalese finches than for Great tits. 4) In the Bengalese finches, is shortened in the presence of a nest box due to effects on reproductive physiology.

Intercellular junctions in nerve-free hydra

Summary Epithelial cells of nerve-free hydra contain septate and gap junctions. In thin sections the gap junctions are characterized by a gap of 3–4 nm. Freeze-fracture demonstrates the presence of septate junctions and two further types of structures: (i) the E-type or inverted gap junctions with particles in an en plaque conformation appearing as a raised plateau on the E-face or as a depression on the P-face; (ii) structures morphologically similar to gap junctions in rat liver, containing particles on the P-face and corresponding pits on the E-face, both having hexagonal packing with a lattice constant of 8 nm. We propose that these structures are also gap junctions.     

Contextualizing Images in Dreams and Daydreams

A contextualizing image (CI) is a powerful central image of a dream which appears to contextualize (provide a picture-context for) the dreamer's emotion. For instance, dreamers who have experienced any serious traumatic event sometimes dream, I was overwhelmed by a tidal wave. This appears to picture their feeling of terror and/or vulnerability.A scoring system for CIs is examined here and is applied to dreams and daydreams supplied by 40 students. Two raters scoring dreams on a blind basis showed good inter-rater reliability. Recent dreams were shown to have more as well as more intense CIs than recent daydreams; likewise, dreams that stand out had more intense CIs than daydreams that stand out. Students with thin boundaries had more and more intense CIs than students with thick boundaries in their recent dreams and nightmare, but not so clearly in dreams and nightmares that stand out. The emotions judged as contextualized by the powerful images tended towards fear/terror and helplessness/vulnerability in dreams (especially in dreams that stand out) whereas emotions contextualized by images in daydreams showed a wide range with no clusters.     

Over‐expression of the murine polymeric immunoglobulin receptor gene in the mammary gland of transgenic mice

The polymeric immunoglobulin receptor (pIgR), a transmembrane protein, transports dimeric IgA (dIgA) across the epithelial cells of the mucosal surfaces into the external secretions, for example milk from the mammary glands. The pIgR is consumed during the transcytosis of dIgA and is cleaved at the apical side of the epithelial cells, regardless of the binding to its ligand (dIgA), to form secretory component (SC). We hypothesize that the expression level of the endogenous murine pIgR gene in the epithelial cells is ratelimiting for the transport of dIgA across the epithelial cells into the secretions. We address this key issue by generating transgenic mice overexpressing the pIgR gene in their mammary glands in order to examine the effect on dIgA levels in the milk. Here we report on the generation of transgenic mice and analysis of the expression level of pIgR in their mammary glands. We cloned and characterized the murine pIgR gene and constructed an expression cassette bearing the pIgR gene under the control of the regulatory sequences of the bovine _s1casein gene. Four transgenic lines were made, expressing the pIgR construct at RNA and protein level only in their mammary glands. The levels of the SC protein in the milk ranged from 0.1 to 2.7mg/ml during midlactation. These levels are 10–270 times higher than wildtype SC levels (0.01mg/ml).     

Elektronenmikroskopische Untersuchungen über Bildung und Struktur der Zygotenwand beiMicrasterias papillifera (Desmidiaceae)

Zusammenfassung Die Feinstruktur von Mesospor und Endospor reifer Zygoten vonMicrasterias papillifera wurde untersucht. Das für die Resistenz der Zygoten gegenüber ungünstigen Umweltbedingungen wichtige Mesospor besteht aus vier Schichten von unterschiedlicher Elektronendichte. Es ist insgesamt 460–500 nm dick. Die Schichten Mes a und Mes c bestehen aus akkrustierten, dicht gepackten globulären Elementen eines Stoffes, der dem Sporopollenin ähnlich ist. Die Schicht Mes b zeigt ein fibrilläres Grundgerüst, wahrscheinlich aus Zellulose, in das Stoffe inkrustiert sind, die nicht mit denen der Schichten Mes a und Mes c identisch, aber gegen Abbauversuche ähnlich résistent sind.Die Schicht Mes d ist eine Übergangsschicht zum Endospor. Zwischen die Zellulose-Mikrofibrillen in Streutextur sind isolierte Partikel des Materials der Schicht Mes c eingestreut. Das etwa 650 nm dicke Endospor ist eine Zelluloseschicht mit Streutextur. Es wird als Wand der sogenannten Keimblase bei der Zygotenkeimung nach Sprengung von Exospor und Mesospor stark gedehnt.

Electron microscopical investigations on the structure and formation of the zygote wall inMicrasterias papillifera (Desmidiaceae) II. The structure of the mesospore and the endospore

Summary The mesospore (460–500 nm thick) which is responsible for the resistance of the zygote against desiccation during its resting period, consists of four layers of different electron density. The layers Mes a and Mes c are composed of densely packed amorphous globular elements of a substance resembling sporopollenine. The layer Mes b has a fibrillar, probably cellulosic frame. It is incrusted by a substance which is not identical with that of Mes a or Mes c but which shows a comparable resistance against degradation.The layer Mes d contains isolated particles such as in Mes c and cellulose microfibrils of the endospore. The endospore (650 nm thick) has foliate texture. This layer surrounds the protoplast after it has escaped from the ruptured exospore and mesospore during zygospore germination.

The rotifer trap of Zoophagus

The predacious watermold Zoophagus insidians traps loricate rotifers on short, lateral branches of the main, hyphal axis. These branches or traps are packed at their distal ends with a number of vesicles filled with an electron-dense matrix. Electron micrographs of the mycelium disclose a two-layered wall; the outer layer is electron dense and the inner, electron transparent. The outer dense layer on the tip of the trap is organized into a number of fine ridges and occasional discontinuities. Thin sections through recently trapped rotifers indicate that the cilia of the animals are stuck to the trap by a glue. This adhesive is derived from secretion of the matrix of the vesicles aggregated in the tip of the trap. The secretion mechanism is triggered by the animal and is accompanied by: 1. The separation of the two layers of the wall, 2. fusion of the vesicles with the cell membrane and 3. extrusion of the glue through pits in the tip of the inner wall of the trap. After snaring a rotifer, the previously arrested branch grows as a haustorium into the body cavity of the animal. The host tissues disintegrate within a few hours and appear to be the main nutrient source of the fungus.

Sie zeigen stellenweise einen mäanderähnlichen Verlauf und Knäuel-Bildungen. Entsprechende Arterien finden sich auch in der Dura von Huhn und Kaninchen.Mögliche Entstehungsursache, charakteristische Verteilung und funktionelle Bedeutung der Spiralarterien der menschlichen Pachymeninx werden diskutiert.Wiss. Assistent am planmä¿igen Extraordinariat für Anatomie.     

A comparison and evaluation of some phytosociological techniques

Summary In three areas of vegetation (dune, mountain heath and salt marsh) the following phytosociological techniques have been tested and compared, using the same data: the Braun-Blanquet method; association and inverse analysis ofWilliams &Lambert; cluster analysis (agglomerative classification) based on different coefficients of similarity; and ordination (principal components analysis performed on matrices of different coefficients).The Braun-Blanquet method is considered to combine several advantages of the other methods and to be most economical in terms of efficiency (ratio of time input to information emerging).

Zusammenfassung Auf drei verschiedenen Vegetationsflächen (Bergheide, Küstendünen und Salzwiesen) sind die folgenden pflanzensoziologischen Methoden geprüft und verglichen worden: die Methode von Braun-Blanquet; association analysis vonWilliams &Lambert (1959, 1961); Ordination (principal components analysis); und cluster analysis (Sokal &Sneath, 1963). Die letzteren beiden wurden mit verschiedenen Ähnlichkeitskoeffizienten geprüft.Auf Grund solchen Erfahrungen, zeigte sich die Braun-Blanquetische Methode leistungsfähiger als die anderen Methoden (d.h. optimale Einsicht in der Vegetation pro Arbeitsstunde). Sie vereinigte viele Vorteile der anderen Methoden.
